CO CV CM CI Ryugu A Ryugu C 0.0 0.5 1.0 1.5 44/40CaSRM915a (age corrected) The Hayabusa2 spacecraft has returned samples from the Cb-type asteroid (162173) Ryugu to Earth. Previous petrological and chemical analyses support a close link between Ryugu and CI chondrites that are presumed to be chemically the most primitive meteorites with a solar-like composition. However, Ryugu samples are highly enriched in Ca compared to typical CI chondrites. To identify the cause of this discrepancy, here we report stable Ca isotopic data (expressed as delta 44/40CaSRM915a) for returned Ryugu samples collected from two sites. We found that samples from both sites have similar delta 44/40CaSRM915a (0.58 +/- 0.03 parts per thousand and 0.55 +/- 0.08 parts per thousand, 2 s.d.) that fall within the range defined by CIs. This isotopic similarity suggests that the Ca budget of CIs and Ryugu samples is dominated by carbonates, and the variably higher Ca contents in Ryugu samples are due to the abundant carbonates. Precipitation of carbonates on Ryugu likely coincided with a major episode of aqueous activity dated to have occurred similar to 5 Myr after Solar System formation. Based on the pristine Ryugu samples, the average delta 44/40CaSRM915a of the Solar System is defined to be 0.57 +/- 0.04 parts per thousand (2 s.d.).

Context. JAXA’s Hayabusa2 mission rendezvoused the Ryugu asteroid for 1.5 years to clarify the carbonaceous asteroids’ record for Solar System origin and evolution. Aims. We studied the photometric behavior of the spectral parameters characterizing the near-infrared (NIR) spectra of Ryugu provided by the Hayabusa2/NIRS3 instrument, that is to say 1.9 µm reflectance, 2.7 and 2.8 µm band depths (ascribed to phyllosilicates), and NIR slope. Methods. For each parameter, we applied the following empirical approach: (1) retrieval of the equigonal albedo by applying the Akimov disk function (this step was only performed for the reflectance photometric correction); (2) retrieval of the median spectral parameter value at each phase angle bin; and (3) retrieval of the phase function by a linear fit. Results. Ryugu’s phase function shows a steepness similar to Ceres, according to the same taxonomy of the two asteroids. Band depths decrease with increasing phase angle: this trend is opposite to that observed on other asteroids explored by space missions and is ascribed to the very dark albedo. NIR and visible phase reddening are similar, contrary to other asteroids, where visible phase reddening is larger: this could be due to surface darkness or to particle smoothness. Albedo and band depths are globally uncorrelated, but locally anticorrelated. A correlation between darkening and reddening is observed.

A new event reconstruction algorithm based on a maximum likelihood method has been developed for Super-Kamiokande. Its improved kinematic and particle identification capabilities enable the analysis of atmospheric neutrino data in a detector volume 32% larger than previous analyses and increases sensitivity to the neutrino mass hierarchy. Analysis of a 253.9 kton-year exposure of the Super-Kamiokande IV atmospheric neutrino data has yielded a weak preference for the normal hierarchy, disfavoring the inverted hierarchy at 74% assuming oscillations at the best fit of the analysis., 42 pages, 28 figures, submitted to PTEP

Hayabusa2 is the Japanese Asteroid Return Mission and targeted the carbonaceous asteroid Ryugu, conducted by the Japan Aerospace Exploration Agency (JAXA). The goal of this mission was to conduct proximity operations including remote sensing observations, material sampling, and a Small Carry-On Impact experiment, as well as sample analyses. As of September 2020, the spacecraft is on the way back to Earth with samples from Ryugu with no critical issues after the successful departure in November 2019. Here, we propose an extended mission in which the spacecraft will rendezvous with a small asteroid with ~30 m - ~40 m in diameter that is rotating at a spin period of ~10 min after an additional ~10-year cruise phase. We introduce that two scenarios are suitable for the extended mission. In the first scenario, the spacecraft will perform swing-by maneuvers at Venus once and Earth twice to arrive at asteroid 2001 AV43. In the second scenario, it will perform swing-by maneuvers at Earth twice to reach asteroid 1998 KY26. In both scenarios, the mission will continue until the early 2030s. JAXA recently released the decision that the spacecraft will rendezvous with 1998 KY26. This paper focuses on our scientific assessments of the two scenarios but leaves the decision process to go to 1998 KY26 for future reports. Rendezvous operations will be planned to detail the physical properties and surrounding environments of the target, one of the smallest elements of small planetary bodies. By achieving the planned operations, the mission will provide critical hints on the violent histories of collisions and accumulations of small bodies in the solar system. Crystal and magnetic structures of the mineral centennialite CaCu$_3$(OH)$_6$Cl$_2\cdot0.6$H$_2$O are investigated by means of synchrotron x-ray diffraction and neutron diffraction measurements complemented by density functional theory (DFT) and pseudofermion functional renormalization group (PFFRG) calculations. CaCu$_3$(OH)$_6$Cl$_2\cdot0.6$H$_2$O crystallizes in the $P\bar{3}m1$ space group and Cu$^{2+}$ ions form a geometrically perfect kagome network with antiferromagnetic $J_1$. No intersite disorder between Cu$^{2+}$ and Ca$^{2+}$ ions is detected. CaCu$_3$(OH)$_6$Cl$_2\cdot0.6$H$_2$O enters a magnetic long-range ordered state below $T_\text{N}=7.2$~K, and the $\mathbf{q}=\mathbf{0}$ magnetic structure with negative vector spin chirality is obtained. The ordered moment at 0.3~K is suppressed to $0.58(2)\mu_\text{B}$. Our DFT calculations indicate the presence of antiferromagnetic $J_2$ and ferromagnetic $J_d$ superexchange couplings of a strength which places the system at the crossroads of three magnetic orders (at the classical level) and a spin-$\frac{1}{2}$ PFFRG analysis shows a dominance of $\mathbf{q}=\mathbf{0}$ type magnetic correlations, consistent with and indicating proximity to the observed $\mathbf{q}=\mathbf{0}$ spin structure. The results suggest that this material is located close to a quantum critical point and is a good realization of a $J_1$-$J_2$-$J_d$ kagome antiferromagnet., Comment: 4 figures and supplemental materials

We have searched for proton decay via $p\to e^+��^0$ and $p\to ��^+��^0$ modes with the enlarged fiducial volume data of Super-Kamiokande from April 1996 to May 2018, which corresponds to 450 kton$\cdot$years exposure. We have accumulated about 25% more livetime and enlarged the fiducial volume of the Super-Kamiokande detector from 22.5 kton to 27.2 kton for this analysis, so that 144 kton$\cdot$years of data, including 78 kton$\cdot$years of additional fiducial volume data, has been newly analyzed. No candidates have been found for $p\to e^+��^0$ and one candidate remains for $p\to ��^+��^0$ in the conventional 22.5 kton fiducial volume and it is consistent with the atmospheric neutrino background prediction. We set lower limits on the partial lifetime for each of these modes: $��/B(p\to e^+��^0) > 2.4 \times 10^{34}$ years and $��/B(p\to ��^+��^0) > 1.6 \times 10^{34}$ years at 90% confidence level., 16 pages, 10 figures

BACKGROUND AND PURPOSE: A thorough knowledge of fetal growth and development is key to understanding both the normal and abnormal fetal MR imaging findings. We investigated the size and signal intensity of the normal pituitary gland and the intrasphenoidal ossification around the Rathke pouch in formalin-fixed fetuses on MR imaging. MATERIALS AND METHODS: Thirty-two fetuses with undamaged brains were included in this study (mean age, 19.93 weeks; age range, 12–31 weeks). Visual inspection of the pituitary and ossification around the Rathke pouch in the sphenoid bone or the postsphenoid ossification was conducted. The extent of pituitary and postsphenoid ossification, pituitary/pons signal ratio, and postsphenoidal ossification/sphenoid bone signal ratio was compared according to gestational age. RESULTS: The pituitary gland was identified as a hyperintense intrasellar structure in all cases, and postsphenoid ossification was identified as an intrasphenoidal hyperintense area in 27 of the 32 cases (84%). The mean pituitary/pons signal ratio was 1.13 ± 0.18 and correlated weakly with gestational age (R(2) = 0.243), while the mean postsphenoid ossification/sphenoid bone signal ratio was 2.14 ± 0.56 and did not show any increase with gestational age (R(2) = 0.05). No apparent change in the size of pituitary hyperintensity was seen with gestational age (R(2) = 0.001). Postsphenoid ossification showed an increase in size with gestational age (R(2) = 0.307). CONCLUSIONS: The fetal pituitary gland was hyperintense on T1-weighted images and the pituitary/pons ratio and extent of postsphenoid ossification correlated weakly with gestational age.

Background: Individuals with type 2 diabetes mellitus are at increased risk for heart failure (HF), particularly those with coexisting atherosclerotic cardiovascular disease and/or kidney disease. Some but not all dipeptidyl peptidase-4 inhibitors have been associated with increased HF risk. We performed secondary analyses of HF and related outcomes with the dipeptidyl peptidase-4 inhibitor linagliptin versus placebo in CARMELINA (The Cardiovascular and Renal Microvascular Outcome Study With Linagliptin), a cardiovascular outcomes trial that enrolled participants with type 2 diabetes mellitus and atherosclerotic cardiovascular disease and/or kidney disease. Methods: Participants in 27 countries with type 2 diabetes mellitus and concomitant atherosclerotic cardiovascular disease and/or kidney disease were randomized 1:1 to receive once daily oral linagliptin 5 mg or placebo, on top of standard of care. All hospitalization for HF (hHF), cardiovascular outcomes, and deaths were prospectively captured and centrally adjudicated. In prespecified and post hoc analyses of HF and related events, Cox proportional hazards models adjusting for region and baseline history of HF were used. Recurrent hHF events were analyzed using a negative binomial model. In a subset of participants with left ventricular ejection fraction captured within the year before randomization, HF-related outcomes were assessed in subgroups stratified by left ventricular ejection fraction > or 50%. Results: CARMELINA enrolled 6979 participants (mean age, 65.9 years; estimated glomerular filtration rate, mL/min per 1.73m(2); hemoglobin A1c, 8.0%; 62.9% men; diabetes mellitus duration, 14.8 years), including 1873 (26.8%) with a history of HF at baseline. Median follow-up was 2.2 years. Linagliptin versus placebo did not affect the incidence of hHF (209/3494 [6.0%] versus 226/3485 [6.5%], respectively; hazard ratio [HR], 0.90; 95% CI, 0.74-1.08), the composite of cardiovascular death/hHF (HR, 0.94; 95% CI, 0.82-1.08), or risk for recurrent hHF events (326 versus 359 events, respectively; rate ratio, 0.94; 95% CI, 0.75-1.20). There was no heterogeneity of linagliptin effects on hHF by history of HF at baseline, baseline estimated glomerular filtration rate or urine albumin-creatinine ratio, or prerandomization left ventricular ejection fraction. Conclusions: In a large, international cardiovascular outcome trial in participants with type 2 diabetes mellitus and concomitant atherosclerotic cardiovascular disease and/or kidney disease, linagliptin did not affect the risk of hHF or other selected HF-related outcomes, including among participants with and without a history of HF, across the spectrum of kidney disease, and independent of previous left ventricular ejection fraction. - Abstract
IMPORTANCE Type 2 diabetes is associated with increased cardiovascular (CV) risk. Prior trials have demonstrated CV safety of 3 dipeptidyl peptidase 4 (DPP-4) inhibitors but have included limited numbers of patients with high CV risk and chronic kidney disease. OBJECTIVE To evaluate the effect of linagliptin, a selective DPP-4 inhibitor, on CV outcomes and kidney outcomes in patients with type 2 diabetes at high risk of CV and kidney events. DESIGN, SETTING, AND PARTICIPANTS Randomized, placebo-controlled, multicenter noninferiority trial conducted from August 2013 to August 2016 at 605 clinic sites in 27 countries among adults with type 2 diabetes, hemoglobin A(1c) of 6.5% to 10.0%, high CV risk (history of vascular disease and urine-albumin creatinine ratio [UACR] > 200mg/g), and high renal risk (reduced eGFR and micro-or macroalbuminuria). Participants with end-stage renal disease (ESRD) were excluded. Final follow-up occurred on January 18, 2018. INTERVENTIONS Patients were randomized to receive linagliptin, 5 mg once daily (n = 3494), or placebo once daily (n = 3485) added to usual care. Other glucose-lowering medications or insulin could be added based on clinical need and local clinical guidelines. MAIN OUTCOMES AND MEASURES Primary outcomewas time to first occurrence of the composite of CV death, nonfatalmyocardial infarction, or nonfatal stroke. Criteria for noninferiority of linagliptin vs placebo was defined by the upper limit of the 2-sided 95% CI for the hazard ratio (HR) of linagliptin relative to placebo being less than 1.3. Secondary outcome was time to first occurrence of adjudicated death due to renal failure, ESRD, or sustained 40% or higher decrease in eGFR from baseline. RESULTS Of 6991 enrollees, 6979 (mean age, 65.9 years; eGFR, 54.6 mL/min/1.73m2; 80.1% with UACR > 30mg/g) received at least 1 dose of study medication and 98.7% completed the study. During a median follow-up of 2.2 years, the primary outcome occurred in 434 of 3494 (12.4%) and 420 of 3485 (12.1%) in the linagliptin and placebo groups, respectively, (absolute incidence rate difference, 0.13 [95% CI,-0.63 to 0.90] per 100 person-years) (HR, 1.02; 95% CI, 0.89-1.17; P

We review the evolution of late Cenozoic magmatism in the NE Japan arc, and examine the relationship between the magmatism and the crust–mantle structure. Recent studies reveal secular changes in the mode of magmatic activity, the magma plumbing system, erupted volumes and magmatic composition associated with the evolution of crust–mantle structures related to the tectonic evolution of the arc. The evolution of Cenozoic magmatism in the arc can be divided into three periods: the continental margin (66–21 Ma), the back-arc basin (21– 13.5 Ma) and the island-arc period (13.5–0 Ma). Magmatic evolution in the back-arc basin and the island-arc periods appears to be related to the 2D to 3D change in the convection pattern of the mantle wedge related to the asthenosphere upwelling and subsequent cooling of the mantle. Geodynamic changes in the mantle caused back-arc basin basalt eruptions during the back-arc basin opening (basalt phase) followed by crustal heating and re-melting, which generated many felsic plutons and calderas (rhyolite/granite phase) in the early stage of the island-arc period. This was followed by crustal cooling and strong compression, which ensured vent connections and mixing between deeper mafic and shallower felsic magmas, erupting large volumes of Quaternary andesites (andesite phase). The evolution of late Cenozoic magmatism in the NE Japan arc can be divided into three major periods – (1) the continental margin (66–21 Ma), (2) the back-arc basin (21–13.5 Ma) and (3) the island-arc (13.5–0 Ma) periods (Ohguchi et al. 1989; Yoshida et al. 1995; Yoshida 2001) – all of which are related to the development of magmatism in the subduction zone, including the back-arc basin opening (Sato 1994; Yoshida et al. 1995; Yamada et al. 2012). The continental margin period is characterized by mafic to felsic subaerial volcanism, with thick ignimbrites in the continental rift systems. The back-arc basin period is characterized by submarine bimodal volcanism under extensional tectonics, and the island-arc period is characterized by felsic caldera-forming events followed by andesitic stratovolcano-forming volcanism under neutral to compressional stress fields (Ohguchi et al. 1989; Sato & Yoshida 1993; Sato 1994; Yoshida 2001). The characteristic magma of subduction zones is andesite, although it is usually associated with basaltic to rhyolitic magmas (Gill 1981). Calcalkaline andesites are typical, particularly in mature arcs constructed on a thick crust (Kuno 1968, and references therein). The mafic magmas derived from the mantle wedge may be underplated and stagnating around the Moho beneath the thick crust, especially under horizontal compression. Differentiated magmas can stagnate at levels of neutral buoyancy in the crust (Ryan 1987; Takada 1989, and references therein). There are many models for andesite genesis in the subduction zone of Japan, including differentiation by fractional crystallization of mafic magmas (low-K tholeiite from Adatara volcano: Fujinawa 1988), magma mixing/ co-mingling between mafic and felsic end-members (low-K andesite: Toya et al. 2005; medium-K andesite: Ban & Yamamoto 2002; Hirotani & Ban 2006; high-K andesite: Inoue & Ban 1996), assimilation and fractional crystallization (DePaolo 1985) or crustal contamination (Kobayashi & Nakamura 2001; Hirotani et al. 2009), re-melting of lowercrustal hydrous mafic rocks (Takahashi 1986; Kimura et al. 2001, 2002) and melting of the mantle wedge or slab (high-magnesian andesite and adakite: Hanyu et al. 2006; Yamamoto & Hoang 2009; Hoang et al. 2009). Possible processes which could produce the felsic end-member of mixed magmas are: the fractional crystallization from mafic end-member magmas (Kanisawa & Yoshida 1989; Miyagi et al. 2012); the melt extraction or re-melting of partially or fully crystallized precursory mafic magmas in the upper to middle crust (Hildreth & Wilson 2007; Hirotani et al. 2009; Miyagi et al. 2012); the partial re-melting of mafic end-member material solidified to become hornblende gabbro or amphibolite (Feeley et al. 1998; Hansen et al. 2002; Toya et al. 2005); and the partial re-melting of crustal basement rocks (Shuto et al. 2006, 2008). All of these crustal processes forming calc-alkaline andesites work effectively in mature arcs on thick crust. To understand the origin of island-arc andesites, it is very important to know in detail the structure of the magma plumbing system from the upper mantle through the entire crust. Moreover, the observation that andesite-dominated volcanism may occur only at specific stages of the evolution of a subduction zone justifies a comprehensive study of the evolutional history of the NE Japan arc and the crust– mantle structures throughout its evolution. The purpose of this paper is to review the evolutionary history of late Cenozoic magmatism in the arc, including its thermal and tectonic evolution, and to examine the relationship between magmatism and the crust–mantle structures at each evolutionary stage. Using 0.37 megaton$\cdot$years of exposure from the Super-Kamiokande detector, we search for 10 dinucleon and nucleon decay modes that have a two-body final state with no hadrons. These baryon and lepton number violating modes have the potential to probe theories of unification and baryogenesis. For five of these modes the searches are novel, and for the other five modes we improve the limits by more than one order of magnitude. No significant evidence for dinucleon or nucleon decay is observed and we set lower limits on the partial lifetime of oxygen nuclei and on the nucleon partial lifetime that are above $4\times 10^{33}$ years for oxygen via the dinucleon decay modes and up to about $4 \times 10^{34}$ years for nucleons via the single nucleon decay modes.

International audience; Regulation of gene expression is necessary to avoid possible adverse effects of gene therapy due to excess synthesis of transgene products. To reduce transgene expression, we developed a viral vector-mediated somatic regulation system using inducible Cre recombinase. A recombinant adeno-associated virus (AAV) vector expressing Cre recombinase fused to a mutated ligand-binding domain of the estrogen receptor alpha (CreER(T2)) was delivered along with AAV vectors expressing dopamine-synthesizing enzymes to rats of a Parkinson disease model. Treatment with 4-hydroxytamoxifen, a synthetic estrogen receptor modulator, activated Cre recombinase within the transduced neurons and induced selective excision of the tyrosine hydroxylase (TH) coding sequence flanked by loxP sites, leading to a reduction in transgene-mediated dopamine synthesis. Using this strategy, aromatic L-amino acid decarboxylase (AADC) activity was retained so that l-3,4-dihydroxyphenylalanine (L-dopa), a substrate for AADC, could be converted to dopamine in the striatum and the therapeutic effects of L-dopa preserved, even after reduction of TH expression in the case of dopamine overproduction. Our data demonstrate that viral vector-mediated inducible Cre recombinase can serve as an in vivo molecular switch, allowing spatial and temporal control of transgene expression, thereby potentially increasing the safety of gene therapy.

Eukaryotic cells control the levels of molecular chaperones and folding enzymes in the endoplasmic reticulum (ER) by a transcriptional induction process termed the unfolded protein response (UPR). The mammalian UPR is mediated by the cis-acting ER stress response element consisting of 19nt (CCAATN9CCACG), the CCACG part of which is considered to provide specificity. We recently identified the basic leucine zipper (bZIP) protein ATF6 as a mammalian UPR-specific transcription factor; ATF6 is activated by ER stress-induced proteolysis and binds directly to CCACG. Here we report that eukaryotic cells express another bZIP protein closely related to ATF6 in both structure and function. This protein encoded by the G13 (cAMP response element binding protein-related protein) gene is constitutively synthesized as a type II transmembrane glycoprotein anchored in the ER membrane and processed into a soluble form upon ER stress as occurs with ATF6. The proteolytic processing of ATF6 and the G13 gene product is accompanied by their relocation from the ER to the nucleus; their basic regions seem to function as a nuclear localization signal. Overexpression of the soluble form of the G13 product constitutively activates the UPR, whereas overexpression of a mutant lacking the activation domain exhibits a strong dominant-negative effect. Furthermore, the soluble forms of ATF6 and the G13 gene product are unable to bind to several point mutants of the cis-acting ER stress response element in vitro that hardly respond to ER stress in vivo. We thus concluded that the two related bZIP proteins are crucial transcriptional regulators of the mammalian UPR, and propose calling the ATF6 gene product ATF6α and the G13 gene product ATF6β.

Infective endocarditis caused by Kingella denitrificans occurs rarely. A review of the literature reveals only 6 cases of endocarditis caused by the bacillus. K. denitrificans is normally a commensal of the upper respiratory airways, may exceptionally be responsible for endocarditis. A case of possible prosthetic endocarditis caused by K. denitrificans is presented. A 78-year-old male with Type II diabetes was admitted to the hospital complaining of fever, a sore throat and arthralgia. He underwent replacement surgery of a St. Jude medical prosthesis for aortic stenosis at the age of 75. The only physical findings at admission were a temperature of 38.2 degrees C and murmurs of mild mitral regurgitation. The liver and spleen were not palpable, and there were no skin or eye lesions. Laboratory findings were as follows: white blood cell count 9500/microliters with 77% neutrophils, erythrocyte sedimentation rate 71 mm/h (Westergren), blood urea nitrogen 50.2 mg/dl, serum creatinine 1.7 mg/dl and C-reactive protein 22.2 mg/dl. The Gram-negative bacillus isolated from the blood was identified as K. denitrificans by the identification system, namely ID test.FN-20 rapid (Nissui, Japan). Although an echocardiogram detected no vegetation, infective endocarditis was diagnosed because the same bacillus was detected by separate blood cultures and an obvious source of infection was not found other than the prosthetic valve. Initial treatment was flomoxef, which was changed to Ampicillin 2 g/day after K. denitrificans was identified. Ampicillin continued for 6 weeks. The clinical course was good and he did not require further surgery. He has been afebrile for 2 years after completing treatment. This case represents the first report of prosthetic valve endocarditis caused by K. denitrificans in Japan.

A 25-year-old female presented with amenorrhea and bitemporal hemianopsia without diabetes insipidus due to an intra and suprasellar dumbbell tumor. The preoperative and operative diagnosis was pituitary adenoma, but the histological diagnosis was germinoma. Differential diagnosis between germinoma and pituitary adenoma based on computed tomograms and magnetic resonance (MR) imaging features and signal intensity characteristics was difficult in this patient. An enhanced mass posterior to the normal pituitary gland on the mid-sagittal MR image normally indicates an intrasellar germinoma rather than a pituitary adenoma, but this finding was equivocal in our patient.

To clarify the relationship between cognitive function and CBZ therapy, auditory event-related potentials (P300) were examined in 23 patients with Benign Childhood Epilepsy with Centrotemporal Spike (BCECT) compared with 54 normal controls. The results were 1) the mean P300 latency in BCECT (368 +/- 29 msec) was significantly prolonged compared with that in normal controls (349 +/- 30 msec), but most individual patients showed normal values. 2) The prolongation of P300 latency was greatest during the course of therapy. 3) On repeated examination of P300, P300 latency was found to gradually become shorter with age in spite of continuous CBZ therapy. At initiation of CBZ therapy, the P300 latency became shorter; on the other hand, P300 latency became shorter with the discontinuation of CBZ. 4) The age-corrected P300 latency showed a significant positive correlation with the serum concentration of CBZ. Our results suggest that CBZ therapy has both an undesirable effect (chronic impairment) and a desirable effect (improvement of underlying dysfunction caused by epileptogenesis) on cognitive function.

Lumbar disc degeneration is frequently seen in athletes. Lumbar disc diseases include a spectrum of diseases and/or symptoms, including lumbar disc degeneration. Some reports suggest an association between lumbar disc diseases and a functional single-nucleotide polymorphism (SNP;1184T/C, rs 2073711) of the cartilage intermediate layer protein ( CILP) gene. We hypothesized that lumbar disc degeneration occurrence may be significantly associated with SNP variants at the CILP gene in Japanese collegiate judo athletes. Eighty-nine Japanese judo athletes participated in this study. T2-weighted magnetic resonance imaging was used to define lumbar disc degeneration. Genotyping of the CILP gene (1184T/C) was performed using DNA sequencing. By using logistic regression analysis, significant associations of lumbar disc degeneration with the CILP C allele (odds ratio=4.1, 95% confidence interval: 1.57-10.71) and body weight (odds ratio=1.06, 95% confidence interval: 1.02-1.09) were observed. We conclude that the CILP gene 1184T/C polymorphism is a significant risk factor for lumbar disc degeneration occurrence in Japanese collegiate judo athletes.

Our objective was to investigate the influence of molecular charge on transperitoneal solute movement in continuous ambulatory peritoneal dialysis (CAPD). Tests of peritoneal equilibration were performed. Two liters of 2.27% or 2.5% glucose CAPD dialysate were infused and the dialysate samples were taken after 2 hr and 4 hr, and blood samples were obtained after 4-hr dwell time. Dialysate-to-plasma concentrations ratios (DIP) were calculated for creatinine (Cr) and three amino acids with almost the same molecular weight but quite different charges: glutamic acids (Glu: negatively charged), glutamine (Gin: near neutrally charged), and lysine (Lys: positively charged). The setting was a university hospital. There were 23 stable CAPD patients with a mean age of 56.5±9.5 years and a mean CAPD duration of 15.2±19.4 months. DIP ratio of Glu was much lower than those of Gin, Lys and Cr at both 2 hr and 4 hr (p < 0.01), and DIP of Lys was significantly lower than that of Gin (p < 0.01). There was no significant difference of DIP between Gin and Cr. The order of transperitoneal mobility among the three amino acids was Gin > Lys > Glu. Transperitoneal movement of solutes in CAPD is influenced by molecular charge, the movement of negatively charged solutes is most remarkably retarded in cases of amino acids.

Glial cell line-derived neurotrophic factor (GDNF) is a strong candidate agent in the neuroprotective treatment of Parkinson's disease (PD). We investigated whether adeno-associated viral (AAV) vector-mediated delivery of a GDNF gene in a delayed manner could prevent progressive degeneration of dopaminergic (DA) neurons, while preserving a functional nigrostriatal pathway. Four weeks after a unilateral intrastriatal injection of 6-hydroxydopamine (6-OHDA), rats received injection of AAV vectors expressing GDNF tagged with FLAG peptide (AAV-GDNFflag) or beta-galactosidase (AAV-LacZ) into the lesioned striatum. Immunostaining for FLAG demonstrated retrograde transport of GDNFflag to the substantia nigra (SN). The density of tyrosine hydroxylase (TH)-positive DA fibers in the striatum and the number of TH-positive or cholera toxin subunit B (CTB, neuronal tracer)-labeled neurons in the SN were significantly greater in the AAV-GDNFflag group than in the AAV-LacZ group. Dopamine levels and those of its metabolites in the striatum were remarkably higher in the AAV-GDNFflag group compared with the control group. Consistent with anatomical and biochemical changes, significant behavioral recovery was observed from 4-20 weeks following AAV-GDNFflag injection. These data indicate that a delayed delivery of GDNF gene using AAV vector is efficacious even 4 weeks after the onset of progressive degeneration in a rat model of PD.

A novel experimental investigation is presented on the connection between discontinuous transitions of macroscopic structures of plasma and self-induced chaotic oscillations characterized by the positive Lyapunov exponents lambda(L) through the period-doubling route in a dc hollow cathode discharge, which has features of nonlinear open systems. We have clarified experimentally that there appear different discharge modes accompanying the discontinuous transitions, and detailed qualitative explanations are presented about the mechanism of those transitions. It is shown that fundamental frequencies of the self-induced periodic oscillations with nonpositive lambda(L) change with the changes of discharge current, and the amplitude of chaotic oscillations with the positive lambda(L) jumps up almost one order higher than that of nonchaotic ones with the nonpositive lambda(L). The self-induced chaotic oscillations with the positive lambda(L) have been observed near two edges of discontinuous transitions of plasma structures, suggesting that the chaotic mode is associated with the discontinuous transition of macroscopic structures in some nonlinear open systems.

Rebamipide, an antiulcer agent, has been shown to be able to prevent gastric mucosal injury resulting in part from activation of neutrophils. The mechanism of its suppressive action, however, remains to be established. The present study aimed to determine the effect of rebamipide on activation of isolated human neutrophils and to identify the signal transduction pathway involved in its regulation. In unstimulated cells, alkaline phosphatase activity was found residing in short rod-shaped intracellular granules. Upon stimulation with a chemotactic peptide formyl-methionyl-leucylphenylalanine, the granules fused to form elongated tubular structures and spherical vacuoles. Rebamipide inhibited reorganization of alkaline phosphatasecontaining granules along with upregulation of alkaline phosphatase activity and CD16, a marker of the granules. It also suppressed chemotaxis, an increase in intracellular calcium ion concentration, and NADPH oxidase activation in cells stimulated with formylmethionyl- leucyl-phenylalanine. In contrast, the drug showed no inhibitory action toward upregulation of alkaline phosphatase activity and CD16, and activation of NADPH oxidase in cells stimulated with phorbol myristate acetate, an activator of protein kinase C. These findings demonstrate that rebamipide exerts a broad spectrum of suppressive actions toward biological functions of human neutrophils stimulated with formylmethionyl- leucyl-phenylalanine, but not with phorbol myristate acetate, and suggest that the upstream point of protein kinase C is the signal transduction pathway involved in its regulation.

To clarify the relationship between the cognitive function and EEG activity, auditory event-related potentials (P300) were examined in 72 patients with partial epilepsy. Twenty-six patients (67 trials) had idiopathic partial epilepsies (IPE), and 46 (118 trials) symptomatic or cryptogenic partial epilepsies (SPE). For this study, patients undergoing carbamazepine monotherapy with a dose of less than 16 mg/kg/day were selected to rule out the effects of anti-epileptic drugs. The results were as follows: (1) The P300 latency tended to be prolonged in association with the EEG slowing in both epileptic groups. (2) There was no clear relationship between the frequency of paroxysmal discharges and the P300 latency. (3) The P300 latency was slightly prolonged in the patients with temporal foci compared with that in ones with extra-temporal foci. (4) There was no significant relationship between the generalization of focal paroxysmal discharges and the P300 latency. These results suggested that the influence of EEG abnormalities (particularly paroxysmal discharges) on the P300 latency is relatively little, and the cognitive dysfunction in partial epilepsies mainly originates from other factors such as the epileptogenic lesion itself.

The effects of antiepileptic drugs (AED) on EEG background activity were evaluated in 37 newly treated children with epilepsy, compared with 46 age-matched healthy controls. Before AED therapy, the children with epilepsy, both partial (treated with carbamazepine, CBZ group) and generalized seizures (treated with valproic acid, VPA group), already exhibited significant slowing of EEG with increased delta and decreased alpha power. Following 3 to 6 months of AED therapy, this EEG slowing was enhanced in the CBZ group and reduced in the VPA group. Following 1 year of AED therapy, an increase in frequency was recognized in the CBZ group. These results suggest that 1) most children with epilepsy already exhibit slowing of the EEG at the onset of seizures, which may reflect CNS developmental deficit, 2) the short-term effects on EEG are different between CBZ and VPA, and 3) EEG development with age continues under continuous AED administration. The EEG background activity in children with epilepsy is affected by many factors, which include the underlying CNS dysfunction of the epilepsy itself and also AED therapy (type of AEDs, duration of therapy, etc).

Eight cases of clinically diagnosed sleep apnea syndrome (SAS) and two normal volunteers were studied with low field MR fluoroscopy in order to monitor the waking and sleeping status of the upper airway. MR fluoroscopy revealed that only the sleeping patients showed occlusions of the upper airway. This technique provided us with useful information about the level, frequency and duration of occlusion in each case. Four of the eight patients demonstrated simple retropalatal occlusion, whereas the other four demonstrated mixed retropalatal and retropalato-retroglossal occlusion. Thus long-time monitoring, which is only possible with MR fluoroscopy, is needed to appreciate the complex nature of the disease. In addition, the comfortable surroundings and low noise level provided by the low field enabled physiological study to be performed without any tranquilizers in most of the patients, which is again only possible with MR fluoroscopy. MR fluoroscopy may become a tool of great clinical value, providing much important information for disease evaluation and treatment selection.
