Background/Aims: Daratumumab has shown an encouraging antitumor effect in patients with multiple myeloma (MM), and was known to alter the immune properties by off-targeting immunosuppressive cells. Here, we aimed to evaluate the change in absolute lymphocyte count (ALC) as a surrogate marker for predicting survival outcomes of patients treated with daratumumab.Methods: Between 2018 and 2021, the medical records of patients with relapsed/refractory MM (RRMM) treated with daratumumab monotherapy at 10 centers in South Korea were reviewed. We collected the ALC data at pre-infusion (D0), day 2 after the first infusion (D2), and prior to the third cycle of daratumumab therapy (D56).Results: Fifty patients who were administered at least two cycles of daratumumab were included. Overall response rate was 54.0% after two cycles of daratumumab treatment. On D2, almost all patients experienced a marked reduction in ALC. However, an increase in ALC on D56 (ALCD56) was observed in patients with non-progressive disease, whereas failure of ALC recovery was noted in those with progressive disease. Patients with ALCD56 > 700/μL (n = 39, 78.0%) had prolonged progression- free survival (PFS) and overall survival (OS) than those with ALCD56 ≤ 700/μL (median PFS: 5.8 months vs. 2.6 months, p = 0.025; median OS: 24.1 months vs. 6.1 months, p = 0.004). In addition, ALCD56 >700/μL was a significant favorable prognostic factor for PFS (hazard ratio [HR], 0.22; p = 0.003) and OS (HR, 0.23; p = 0.012).Conclusions: Increase in ALC during daratumumab treatment was significantly associated with prolonged survival outcomes in patients with RRMM. The ALC value can predict clinical outcomes in patients treated with daratumumab.

Autoimmune limbic encephalitis (LE) is a rare, but devastating complication of allogeneic hem-atopoietic stem cell transplantation (HSCT). There is currently limited evidence describing the risk factors, laboratory features, and underlying mechanisms of this neurologic adverse event. We retrospectively reviewed available clinical, imaging, and laboratory data from adult patients with hematological malignancies who underwent haploidentical HSCT with cyclophosphamide (PTCy) at Chungnam National University Hospital from June 2016 to May 2020. Patients who developed LE were compared to those who did not based on clinical assessment, serum inflam-matory biomarkers, and reconstitution of various T cell populations. Of 35 patients, four devel-oped LE. There were no differences in patient demographics, donor demographics, or treatment conditions between patients that did and did not develop LE. Overall, patients with LE had worse clinical outcomes and overall survival than those without. In addition, they tended to have higher markers of systemic inflammation in the early post-transplant period, including fever, C-reactive protein (CRP), and cytokines. Remarkably, baseline interleukin-6 levels before HSCT were found to be higher in patients who developed LE than those who did not. In addition, analysis of T cell subsets showed impaired expansion of CD25+FOXP3+ regulatory T (Treg) cells in LE compared to non-LE patients despite appropriate reconstitution of the total CD4+ T cell population. Patients that developed LE within the first 30 days of HSCT were likely to have high serum IL-6 among other inflammatory cytokines coupled with suppression of regulatory T cell differentiation. Further work is needed on the mechanisms underlying impaired Treg expansion following HSCT and potential therapies.

Purpose Fluoropyrimidine (FP) and oxaliplatin-based chemotherapy is the standard first-line treatment for metastatic colorectal cancer (mCRC); however, oxaliplatin-induced neuropathy critically affects the quality of life of patients. Maintenance strategies with FP plus bevacizumab have been well established; nonetheless, the real-world outcomes of maintenance therapy with FP and cetuximab are unclear. We investigated the clinical outcomes of patients who underwent maintenance therapy with cetuximab. Methods We retrospectively identified and analyzed patients with mCRC who were treated between 2012 and 2021 with first-line oxaliplatin-based induction chemotherapy (IC) and patients treated with maintenance therapy after IC. Results A total of 112 patients were treated with IC. Among them, 48 patients underwent maintenance therapy (same IC regimen without oxaliplatin). The most common reason for the discontinuation of IC was peripheral neuropathy (n = 40, 83%). For the IC of the maintenance group, 3, 26, and 19 patients were treated with CAPOX (capecitabine/oxaliplatin), mFOLFOX6 (FP/leucovorin/oxaliplatin) plus bevacizumab, and mFOLFOX6 plus cetuximab. The best overall response of maintenance therapy was a partial response in 7 patients and stable disease in 30 patients The median progression-free survival (PFS) of maintenance therapy and overall survival (OS) was 4.83 months and 25.6 months in the bevacizumab group, and 5.98 months and 32.4 months in the cetuximab group, respectively. Conclusions Maintenance therapy with FP and cetuximab is a feasible strategy for appropriate mCRC patients. Further large-scale randomized studies are needed to validate the efficacy of anti-epidermal growth factor receptor-based maintenance therapy.

Restoring the microbiota via fecal microbiota transplantation (FMT) can be an effective treatment for steroid-refractory acute graft-versus-host disease (GVHD) of the gut. Here, we report two adult patients who underwent FMT to treat steroid-refractory acute GVHD of the gut. The first patient was a 43-year-old man who underwent allogeneic hematopoietic stem cell transplantation (HSCT) with cells from a matched sibling donor. The second patient was a 70-year-old woman who underwent haplo-identical HSCT with cells from her son. Gut GVHD developed at 7 and 4 weeks after HSCT, respectively. After undergoing FMT, the clinical symptoms improved; the first patient had a complete response and the second patient had a partial response. Microbial analyses using RNA gene sequencing showed that a diverse fecal microbiome was recovered by 4 weeks after FMT. FMT should be considered an effective therapeutic option for managing steroid-refractory acute GVHD of the gut.

In this article, we designed and synthesized the thionated NpImidazole derivatives BS and NS, new heavy-atom-free photosensitizers, which efficiently generate a triplet excited state with high singlet oxygen quantum yield. The introduction of the C═S bond to the NpImidazole core is essential for increasing spin-orbit coupling (SOC). The fluorescence emission of BS and NS was quenched at standard ambient temperature, accompanied with the increase in the ISC process from the singlet states to triplet excited states via thionation. BS and NS showed negligible dark cytotoxicity against HeLa cells in working concentration. In contrast, BS and NS rapidly induced cell death under blue light irradiation both under normoxia and hypoxia conditions. Our current study demonstrates that the C═S group can play an important role in type I ROS generation of PSs, which are unprecedented in the previous reports. Finally, the photophysical changes were assigned to the oxidative desulfurization of the C═S group of BS and NS to the C═O group of the corresponding BO and NO via hypochlorite. The combined results demonstrated the dual function of BS and NS as a fluorescent imaging agent for ClO- and an anti-cancer therapeutic by PDT that showed the potential strategy for "one-for-all" and multifunctional agents.

Non-palpable, volumetric splenomegaly at diagnosis was evaluated using computed tomography in patients with essential thrombocythemia (ET) and prefibrotic/early primary myelofibrosis (pre-PMF) based on 2016 World Health Organization guidelines. Each patient’s spleen volume was adjusted for their age and body surface area. The degree of splenomegaly was classified as no, borderline volumetric, overt volumetric, or palpable splenomegaly. Seventy-six patients with ET (median age, 62.5 years) and 19 patients with pre-PMF (median age, 65 years) were followed up for a median of 2.4 years (range 0.1–17.6 years) and 4.2 years (range 0.2–19.6 years), respectively. Changes in fecal microbiota affect the incidence and extent of graft-versus-host disease (GVHD) after allogeneic hematopoietic stem cell transplantation (HSCT). Most patients with hematological malignancies receive antibiotics for the treatment of febrile neutropenia prior to allogeneic HSCT, and pre-transplant use of antibiotics may influence the fecal microbiota and GVHD.We retrospectively analysed consecutive adult patients with hematological malignancies who received allogeneic HSCT at Chungnam National University Hospital between 2007 and 2018. Pre-transplant use of antibiotics was defined as the use of antibiotics before conditioning chemotherapy.This study included 131 patients with a median age of 46 (range, 18-71) years: 76 (58%) patients were AML, 28 (21.4%) with ALL, 23 (17.6%) with MDS, and 4 (3.1%) with CML. All patients received calcineurin inhibitors with short-course methotrexate for GVHD prophylaxis. A total of 31 (23.7%) patients received anti-thymocyte globulin. All patients received antibiotics prior to HSCT: 70 (53.4%) patients received glycopeptide, 114 (87.0%) received cefepime, 87 (66.4%) received piperacillin/tazobactam, and 51 (38.9%) received carbapenem. Patients who received glycopeptide had more frequently extensive chronic GVHD (cGVHD) than those who did not (51.1% vs. 28.1% at 5 years) and had more frequently cGVHD of the lung (34.8% vs. 15.8% at 5 years). Pre-transplant use of glycopeptide did not affect the overall survival (OS) or GVHD- and relapse-free survival (GRFS) (median OS; 49 months in glycopeptide group vs. not reached in non-glycopeptide group,Pre-transplant use of glycopeptide tends to increase the incidence of extensive cGVHD.

We prepared the monomer PCDA-HP composed of 5-hydroxy-N 1,N 3-bis(pyridin-2-ylmethyl)isophthalamide (HP) as a cadmium ion tweezer and then polymerized them to form a polydiacetylene (PDA)-based sensor (PDA-HP), which displayed selective and sensitive colorimetric and fluorometric change upon addition of a cadmium ion (Cd2+) at both pH 7.4 and 6.8. The PDA-HP polymer was highly selective for Cd2+ over other metal ions with colorimetric change. In addition, the PDA-HP chemosensor also showed a red fluorescence change in the presence of Cd2+ at both pH 7.4 and 6.8. Naked-eye detection of Cd2+ was accomplished in an aqueous solution through a PDA-based sensor system. Finally, the lowest energy structure of an HP chelator was obtained by the crystal structure and density functional theory (DFT) calculations.

Due to the concern for COVID-19, universities in Korea switched to online teaching in most chemistry courses in the spring semester, 2020. While online lectures have been available for a small number of courses in the previous semesters, it is unprecedented that most lectures should be given online. In this communication, we describe how higher education in chemistry has been made in the spring semester, 2020, mainly based on the experience of the author and colleagues, and address the insights gained that we would like to share. Admittedly, the experience is limited, and there are still unresolved challenges.

The different temperature drying processes were carried out at −60 °C, 15 °C, and 60 °C to control the nanosheet morphology of MXene matrix. The MXene electrode prepared at −60 °C (denoted as LT-Ti3C2Tx) produced more nanosheet arrays than those with drying temperatures of 15 °C and 60 °C (denoted as RT-Ti3C2Tx and HT-Ti3C2Tx MXene electrodes, respectively). The results of BET and electrochemical measurements show that the specific surface area and capacitance initially decrease and then increase with the change in nanosheet layers in MXene. Among these prepared electrodes, LT-Ti3C2Tx, with a well-controlled nanosheet array, showed outstanding specific capacitance (Cs) of 467.4 F g−1 at current density of 0.5 A g−1, and 98.13% stability after 5000 cycles. Furthermore, an LT-Ti3C2Tx // LT-Ti3C2Tx symmetric supercapacitor device (SSD) was assembled, employing the LT-Ti3C2Tx with a well-controlled nanosheet acting as both anode and cathode. The SSD exhibited high energy density of 5.67 Wh kg−1 at power density of 589.09 W kg−1, and long cycle life electrochemical stability of 99.9% after 5000 cycles. These promising results show that MXene electrodes prepared by low-temperature drying (i.e. –60 °C) may be useful for supercapacitor applications.

Non-palpable splenomegaly in patients with polycythemia vera (PV) has seldom been addressed. In this retrospective study, we evaluated non-palpable, volumetric splenomegaly defined based on age- and body surface area (BSA)-matched criteria in patients with PV diagnosed according to the 2016 World Health Organization diagnostic criteria.Patients with PV who underwent abdominal computed tomography (CT) and who had palpable splenomegaly at diagnosis from January 1991 to December 2020 at Chungnam National University Hospital were enrolled. The spleen volume of each patient was determined by volumetric analysis of abdominal CT and adjusted for the patient's age and BSA. Then the degree of splenomegaly was classified as no splenomegaly, borderline volumetric splenomegaly, overt volumetric splenomegaly, or palpable splenomegaly.Of the 87 PV patients enrolled, 15 (17.2%) had no splenomegaly, whereas 17 (19.5%), 45 (51.7%), and 10 (11.5%) had borderline volumetric, overt volumetric, and palpable splenomegaly, respectively. The degree of splenomegaly did not affect the cumulative incidence of thrombotic vascular events (10-year incidence: 7.7%, 0%, 22.3%, and 50.7%, respectively,The degree of splenomegaly, including volumetric splenomegaly, based on age- and BSA-matched reference spleen volumes at diagnosis reflects disease progression in PV patients. Therefore, volumetric splenomegaly should be evaluated at the time of diagnosis and taken into consideration when predicting the prognosis of patients with PV.

Purpose: To evaluate the role of the cardiopulmonary exercise test (CPET) as a predictor of overall survival (OS) and non-relapse mortality (NRM) in patients with hematological malignancies who undergo allogeneic hematopoietic stem cell transplantation (HSCT).Methods: We retrospectively analyzed consecutive adult patients with hematological malignancies who underwent HLA-matched donor-HSCT at Chungnam National University Hospital (Daejeon, South Korea) between January 2014 and December 2020. Peak oxygen consumption (VO2max) was classified using the recommendations of the Mayo Clinic database.Results: Of 72 patients, 38 (52.8%) had VO2max values lower than the 25th percentile (VO2max < 25th) of an age- and sex-matched normal population. Patients with VO2max < 25th exhibited slightly lower OS and higher NRM (30-month OS 29.8% vs. 41%, p = 0.328; and 30-month NRM 16% vs. 3.3%, p = 0.222), compared with other patients. VO2max ≤ 25th was assigned a weight of 1 when added to the Hematopoietic Cell Transplantation-specific Comorbidity Index (HCT-CI) to form a composite comorbidity/CPET index (HCT-CI/CPET). Patients with HCT-CI/CPET scores of 0–1 demonstrated significantly better OS and NRM than did patients with HCT-CI/CPET scores ≥ 2 [median OS not reached vs. 6 months, p < 0.001 and 30-month NRM 7.4% vs. 33.3%, p = 0.006]. An HCT-CI/CPET score ≥ 2 was the only adverse risk factor for NRM on multivariate analysis [hazard ratio (HR) of NRM 10.36 (95% CI 1.486-2.25, p = 0.018)].Conclusion: The CPET may predict the survival and mortality of patients with hematological malignancies who undergo allogeneic HSCT.

The ability to detect hypochlorite (HOCl/ClO−) in vivo is of great importance to identify and visualize infection. Here, we report the use of imidazoline-2-thione (R1SR2) probes, which act to both sense ClO− and kill bacteria. The N2C=S moieties can recognize ClO− among various typical reactive oxygen species (ROS) and turn into imidazolium moieties (R1IR2) via desulfurization. This was observed through UV–vis absorption and fluorescence emission spectroscopy, with a high fluorescence emission quantum yield (ՓF = 43–99%) and large Stokes shift (∆v∼115 nm). Furthermore, the DIM probe, which was prepared by treating the DSM probe with ClO−, also displayed antibacterial efficacy toward not only Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) but also methicillin-resistant Staphylococcus aureus (MRSA) and extended-spectrum ß-lactamase–producing Escherichia coli (ESBL-EC), that is, antibiotic-resistant bacteria. These results suggest that the DSM probe has great potential to carry out the dual roles of a fluorogenic probe and killer of bacteria.

A novel chiral resolution method for racemic 1-(pyridin-2-yl)ethylamine (PEA) was developed in this study. Sequential conversion of 1-(pyridin-2-yl)ethylamine to 1-(pyridin-2-yl)-N-((4R)-1,7,7-trimethylbicyclo[2.2.1]heptan-2-ylidene)ethylamine (PIC) and to dichloro{(E)-1-(pyridin-2-yl)-N-(1,7,7-trimethylbicyclo[2.2.1]heptan-2-ylidene)ethylamine}zinc (PIC-Zn) allowed for the spontaneous separation of dichloro{(1R,E)-1-(pyridin-2-yl)-N-(1,7,7-trimethylbicyclo[2.2.1]heptan-2-ylidene)ethylamine}zinc (RE-PIC-Zn) and dichloro{(1S,E)-1-(pyridin-2-yl)-N-(1,7,7-trimethylbicyclo[2.2.1]heptan-2-ylidene)ethylamine}zinc (SE-PIC-Zn) by fractional crystallization. The overall yields of both (S)- and (R)-PEA were 24.8% with estimated ee values of 98 and 99%, respectively. The preferred formation of diastereopure crystals over that of diastereomeric mixture crystals was interrogated by comparing the cohesive energies (Ecoh) of the respective crystals, as obtained via DFT calculations under periodic boundary conditions with a plane wave basis set based on the experimentally determined X-ray crystallographic structure. The Ecoh values of diastereopure SE-PIC-Zn and RE-PIC-Zn were −45.8 and −44.4 kcal mol−1, respectively. The Ecoh values of the diastereomeric mixture crystal (RE-PIC-Zn·SE-PIC-Zn·2CHCl3) and its virtual counterpart lacking CHCl3 (RE-PIC-Zn·SE-PIC-Zn) were −56.0 and −37.2 kcal mol−1, respectively. These data provided the background underlying the spontaneous segregation of the diastereopure crystals where the energy preference for forming diastereopure crystals over their corresponding diastereomeric mixture crystals is ca. 8 kcal mol−1.

Inhibitors of sodium-glucose cotransporters type 2 (SGLT2) are proposed as a novel approach for the management of type 2 diabetes mellitus. SGLT2 cotransporters are responsible for reabsorption of 90 % of the glucose filtered by the kidney. The glucuretic effect resulting from SGLT2 inhibition contributes to reduce hyperglycaemia and also assists weight loss and blood pressure reduction. In this study, we presented the case of a 59-year-old male who developed hyperosmolar hyperglycemic state (HHS), possibly caused by a sodium-glucose cotransporter 2 (SGLT2) inhibitor, a novel class of antihyperglycemic agents. This case highlights that HHS can develop in patients with diabetes treated with SGLT2 inhibitors

Simple Summary Microphthalmia-associated transcription factor (MITF) has been reported to play a role in the progression of melanoma and other cancer types. However, the biological role of MITF in clear cell renal cell carcinoma (ccRCC) is largely unknown. In this study, we elucidate the role of MITF in the progression of ccRCC. MITF- and MITF-mediated signaling pathways were investigated in ccRCC cell through MITF knockdown as well as overexpression of MITF in vitro and in vivo. MITF contributed to cell proliferation, migration, invasion and tumor growth in ccRCC through activation of the RhoA/YAP signaling pathways. This study suggests that MITF has potential as a therapeutic target in ccRCC. Abstract Microphthalmia-associated transcription factor (MITF) is a basic helix-loop-helix leucine zipper transcription factor involved in the lineage-specific regulation of melanocytes, osteoclasts and mast cells. MITF is also involved in the progression of melanomas and other carcinomas, including the liver, pancreas and lung. However, the role of MITF in clear cell renal cell carcinoma (ccRCC) is largely unknown. This study investigates the functional role of MITF in cancer and the molecular mechanism underlying disease progression in ccRCC. MITF knockdown inhibited cell proliferation and shifted the cell cycle in ccRCC cells. In addition, MITF knockdown reduced wound healing, cell migration and invasion compared with the controls. Conversely, MITF overexpression in SN12C and SNU482 cells increased cell migration and invasion. Overexpression of MITF activated the RhoA/YAP signaling pathway, which regulates cell proliferation and invasion, and increased YAP signaling promoted cell cycle-related protein expression. Additionally, tumor formation was impaired by MITF knockdown and enhanced by MITF overexpression in vivo. In summary, MITF expression was associated with aggressive tumor behavior, and increased the migratory and invasive capabilities of ccRCC cells. These effects were reversed by MITF suppression. These results suggest that MITF is a potential therapeutic target for the treatment of ccRCC.

Autophagy plays an important role in the survival of cancer cells under stressful conditions, such as nutrient or oxygen deficiency. Therefore, autophagy inhibition is being considered as a novel therapeutic strategy for cancer. Decursin is a natural compound derived from Angelica gigas; it has been used in the treatment of various diseases, including cancer. However, the mechanism by which decursin regulates autophagy in gastric cancer and other carcinomas remains unclear. Here, we demonstrated that decursin reduced the growth and induced cell cycle arrest in gastric cancer cells in vitro. Decursin blocked autophagic flux by reducing the expression of lysosomal protein cathepsin C (CTSC) and attenuating its activity, thereby causing autophagic dysregulation (i.e., accumulation of LC3 and SQSTM1). Decursin also inhibited cell proliferation and cell cycle progression by inhibiting CTSC and E2F3, both of which were linked to gastric cancer aggressiveness. The antitumor effects of decursin were confirmed in vivo. We established spheroid and patient-derived organoid models and found that decursin decreased the growth of spheroids and patient-derived gastric organoids, as well as modulated the expression of CTSC and autophagy-related proteins. Hence, our findings uncovered a previously unknown mechanism by which decursin regulates cell growth and autophagy and suggests that decursin may act as a potential therapeutic agent that simultaneously inhibits cell growth and autophagy.

Personal identification methods using conventional fog computing services are not secure and efficient enough. This paper suggests a method for controlling fog server access rights by using ECG signals. When a user asks personal identification to control the fog server, wearable devices are used to measure ECG signals, and personal identification is confirmed through matching by using a computing device. The public ECG data is used for the experiment by means of the suggested network in order to examine the potential method for controlling the suggested fog server access rights. The experiment results revealing recognition performance of 99.81% of 52 participants in the experiment supports the fact that the method for controlling fog server access rights by using ECG signals in this paper is demonstrated as an effective method.

In this study, an intelligent predictor is designed for predicting the direction of dinghy booms and coaching dinghy sailing using the designed predictor with information obtained from multiple sensors attached to the dinghy and the sailor. For this purpose, we designed a Takagi-Sugeno-Kang-based linguistic model with interval prediction based on fuzzy granulation, which can be realized by using a context-based fuzzy c-means clustering. The sensors used in this study are cameras, GPS, and an anemometer. The GPS and cameras were attached to the dinghy, and the anemometer was installed on a separate boat near the dinghy. Features are extracted from obtained data to interpret them discretely without sequential data. The boom direction was predicted by collecting information from the dinghy driven by an expert wearing a marked suit to predict the optimal direction. The constructed database was randomly divided into a training set (60%) and a validation set (40%) for a 10-fold cross-validation. The experimental results revealed that in the prediction of the dinghy boom direction, the proposed predictor showed performance improvements of 25.2% and 17.9% on the training and validation sets, respectively, when compared to the previous predictors.

Background: CX chemokine receptor 7 (CXCR7) is frequently overexpressed in a variety of tumors and plays a significant role in tumor growth and metastasis. Thus, inhibition of CXCR7 is an important potential anticancer treatment strategy. Decursin, a derivative from Angelica gigas Nakai has been reported to have anti-tumor activity in various cancers. However, little is known about its anti-tumor activity and underlying mechanisms of action in head and neck squamous cell cancer (HNSCC). Methods: CXCR7-overexpressing HNSCC cell lines were established in vitro. The effects of decursin on cell growth, motility, migration, and invasion were investigated using CCK-8, wound healing, and transwell assays. In addition, the mechanism by which decusin shows antitumor activity was investigated. Results: Overexpression of CXCR7 significantly increased cell proliferation, migration, invasion, and cell motility compared with control cells. Treatment with decursin decreased membrane expression of CXCR7 in a concentration dependent manner and significantly inhibited tumor progression by suppressing cell growth, migration, invasion, and motility. Decursin was found to induce G0/G1 cell cycle arrest and regulate cell cycle-related proteins such as Cyclin A and CDK2 in HNSCC cells. In addition, decursin inhibited CXCR7-induced STAT3/c-MYC activation. Conclusion: These results demonstrate that CXCR7 promotes cancer progression through the STAT3/c-Myc pathway in HNSCC and that decursin is a natural product that may target CXCR7 for HNSCC treatment. Primary adrenal insufficiency due to tumor involvement is very rare. We herein report a case of adrenal insufficiency caused by cancer metastasis to a remaining single adrenal gland in a renal cell carcinoma patient who had undergone radical nephrectomy including removal of the right adrenal gland. The patient presented with abdominal discomfort, nausea, and vomiting which had begun about 3 months earlier. A rapid adrenocorticotropic hormone (ACTH) test showed no cortisol response with upper normal level of baseline ACTH. The patient was treated with prednisolone after diagnosis of primary adrenal insufficiency due to adrenal metastasis. The symptoms improved after prednisolone replacement. The case reminds clinicians the diagnosis of primary adrenal insufficiency due to tumor metastasis should be considered in patients with nonspecific gastrointestinal symptoms.

This paper addresses the performance index (PI) of an incremental granular model (IGM) with information granules of linguistic intervals. For this purpose, IGM is designed by combining a linear regression (LR) and an interval-based granular model (GM). The fundamental scheme of IGM construction comprises two essential phases: (1) development of LR as a basic model and (2) design of a local granular model, which attempts to reduce errors obtained by the LR model. Here, the local interval-based GM is based on an interval-based fuzzy clustering algorithm, which is materialized by information granulations. The PI of IGM is calculated by multiplying the coverage with specificity property, because the output of IGM is not a numerical value but a linguistic interval value. From the concept of coverage and specificity, we can construct information granules, thus, it is justified by the available experimental proof and presented as clearly defined semantics. To validate the PI method, an experiment is conducted on concrete compressive strength for civil engineering applications. The experimental results confirm that the PI of IGM is an effective performance evaluation method.

Purpose: Autophagy is a series of processes in which biomolecules are digested and recycled, and it also provides energy for the development and progression of cancer. Thus, autophagy inhibitors can be used to suppress the progression of cancer. In this study, we investigated whether decursin, which is derived from natural products, inhibits the growth and autophagy of cancer cells. Methods: The gastric cancer cell lines SNU216 and NCI-N87 were treated with or without decursin. Cell growth was evaluated by Cell-Counting Kit-8, colony formation, and BrdU assays, as well as by propidium iodide staining, and autophagic flux was evaluated by western blotting, LC3 puncta, GFP-RFP-LC3, and transmission electron microscopy. Finally, tumor spheroids and patient-derived gastric cancer organoids were used to confirm the effect of decursin in three-dimensional culture. Results: Decursin reduced the growth and anchorage-dependent viability of gastric cancer cells. LC3 cleavage and LC3-GFP puncta were increased by decursin in a time- and concentration-dependent manner. However, the increase in autophagosome formation did not lead to an increase in autophagic flux. Decursin downregulated cathepsin C (CTSC), a lysosomal protease, irrespective of cellular pH. In addition, knockdown of CTSC reduced the mRNA and protein levels of E2F3, resulting in cell-cycle arrest. Decursin also reduced cell growth in both tumor spheroids and patient-derived gastric cancer organoids. Conclusion: The results demonstrated that decursin inhibited tumor growth and progression by suppressing CTSC-mediated autophagic flux and cell-cycle progression. Supplemental Digital Content is available in the text, The aim of this study was to evaluate the outcomes of patients with advanced or recurrent ovarian cancer treated with cisplatin combined with topotecan as second- or higher-line palliative chemotherapy. We retrospectively reviewed the medical records of patients with advanced or recurrent ovarian cancer, who were treated with cisplatin (50 mg/m2 on day 1) and topotecan (0.75 mg/m2 on days 1–3). Treatment response, progression-free survival (PFS) and overall survival (OS) were analyzed, and laboratory data were reviewed to evaluate toxicities. Thirty one patients were treated with cisplatin and topotecan. The objective response rate (ORR) was 22.6%, and the disease control rate (DCR) was 61.3%. The median PFS was 3.7 months (95% confidence interval [CI], 2.3–5.2 months) and the median OS was 44.5 months (95% CI, 35.5–53.5 months). The ORR (33.3% vs. 0%; P = .012) was significantly better in the platinum-sensitive group compared to the platinum-resistant group. In this paper, we perform the performance comparison of image fusion methods from infrared and visible images. Here, we first use intensity-based registration method that image is aligned based on the relative intensity patterns for image registration. We also compare the performances of Sparse Representation (SR)-based and Multiscale Transform (MST)-based image fusion algorithms from OSU color-thermal database. Finally, we evaluate performance index by mutual information and entropy from final fusion images. It was found from the results that the presented image registration and fusion methods showed good performances.

Molecular dynamics (MD) simulations together with accurate physics-based force fields are employed to determine the mobility of atomic oxygen in amorphous ice at low temperatures, characteristic for conditions in interstellar ices. From the simulations it is found that the mobility of atomic oxygen ranges from 60 to 480 Å2 ns−1 in amorphous ice at temperatures between 50 and 200 K. Hence, the simulations establish that atomic oxygen is mobile to a certain degree and a chemical mechanism for water formation involving oxygen mobility is a realistic scenario. This is also confirmed by the computed migration barriers for oxygen diffusion by multiple umbrella sampling simulations, which yield barriers for diffusion in the range of 0.7–1.9 kcal mol−1. The physics-based force field – based on a multipolar expansion of the electrostatic interactions – yields more pronounced energetics for oxygen migration pathways compared to the conventional point-charge models employed in typical simulations. Once formed, the computed solvation free energy suggests that atomic oxygen thermodynamically prefers to be localized inside amorphous ice and is available for chemical reaction, which may be relevant to water formation in and on grains.

Background: The atypical chemokine receptor, ACKR3, has been shown to play as a tumor promoter, which is upregulated in various types of cancer. However, the biological role of ACKR3 and its underlying mechanism in head and neck squamous cell carcinoma (HNSCC) is still unknown. In this study, we investigated the functional role of ACKR3 and the underlying molecular mechanism of tumor progression in HNSCC. Methods: We examined the ACKR3 expression and its association with clinicopathological characteristics in 103 cases of HNSCC by immunohistochemical staining. The biological roles of ACKR3- and ACKR3-induced downstream signaling were investigated in HNSCC cell through ACKR3 overexpression and knockdown of ACKR3 in vitro and in vivo. Results: We detected differential expression of ACKR3 in HNSCC cells and tissues. High ACKR3 expression was significantly associated with invasion depth of tumor (T status; P = 0.007), lymph node involvement (N status; P = 0.004), and higher stage III/IV (P = 0.02). Overexpression of ACKR3 dramatically enhanced cell migration and invasion in HNSCC cells in vitro, and promoted lymph node metastasis in vivo. ACKR3 overexpression also induced the epithelial-mesenchymal transition. Vimentin, Slug, and Twist were increased but E-cadherin and Ep-CAM were decreased by ACKR3 expression. Upregulation of TGF-β1 was observed in ACKR3 overexpression, and TGF-β1/Smad signaling activation and Akt phosphorylation were induced in HNSCC cells with ACKR3 overexpression. Treatment with a PI3K inhibitor reduced Slug and Twist levels while suppression of Smad2/3 signaling by siRNA reduced Akt phosphorylation, as well as Slug and Twist. Furthermore, inhibition of Smad2/3 decreased tumor cell migration and invasion in HNSCC. ACKR3 knockdown using siRNA in HNSCC cells reduced the secretion of TGF-β1 and also recovered the cell migratory and invasive behavior of HNSCC cells. Conclusions: ACKR3 expression was associated with an aggressive tumor behavior in HNSCC. ACKR3 contributed to migration and invasion of HNSCC cell through the TGF-β1/Smad signaling axis and epithelial-mesenchymal transition in vitro, and was involved in lymph node metastasis in vivo, suggesting that ACKR3 plays an important role during HNSCC progression and metastasis. In the results of investigating the role of LED light quality in enhancing the ornamental value of indoor foliage plants, amber and red light increased plant height, leaf width, and leaf stalk, and the consequent tree shape decreased the ornamental value. The chlorophyll content increased significantly under white light and compound light. With regard to the effect of plant leaf color on ornamental value, the value of lightness was markedly enhanced by red light. As to the functionality of plants according to photosynthetic activity, plants such as Dieffenbachia, Clusia, and Dracaena were found favorable to those staying indoors for a longtime from morning to evening. Spathiphyllum, and Ficus were found to be recommendable for indoor spaces used actively during afternoon because their photosynthesis was activated in the afternoon. With regard to power consumption according to light quality, white light consumed 119 W/hour, around 45% lower than that of fluorescent lamps, so it is considered the optimal artificial light quality that can enhance energy efficiency. Red light consumed 72 W/hour, only 33% of that of fluorescent lamps, but it was not considered the optimal light quality because plant growth was poor under the light quality. White light and compound light were found to be the ideal light sources for improving the functionality and ornamental value of indoor plants and reducing the cost of maintenance, but because compound light hinders people from recognizing the original color of plants and makes their eyes easily tired, white light was considered the optimal light satisfying all of the ornamental value, economic efficiency and functionality resulting from plant growth.

To evaluate changes in macular thickness, ganglion cell layer/inner plexiform layer (GCL/IPL) thickness, and retinal nerve fiber layer (RNFL) thickness in normal eyes and glaucomatous eyes using spectral domain optical coherence tomography (SD-OCT). We enrolled 89 eyes (all left eyes), including 45 (of 45 patients) eyes with glaucoma and 44 (of 44 patients) normal eyes. The data from macular measurements using spectral domain optical coherence tomography were analyzed according to groups divided by age and glaucoma status. The macular thickness analysis, GCL/IPL thickness, and RNFL thickness values determined by SD-OCT scans were compared among the groups. Mean macular thickness decreased significantly with age or glaucoma. Mean GCL/IPL thickness decreased significantly in glaucomatous eyes in all sectors but did not decrease with age. Mean RNFL thickness, which was divided into four quadrants (superior, nasal, inferior, and temporal), decreased significantly in glaucomatous eyes at all quadrants and decreased in the temporal quadrant with age in non-glaucomatous eyes. No significant differences were detected between eyes with normal tension glaucoma (NTG) and primary open angle glaucoma (POAG) in all sectors of mean GCL/IPL thickness, RNFL thickness, and macular thickness. No significant difference in mean thickness was detected between eyes with NTG and POAG. Some of the sectors of RNFL thickness decreased with age or glaucoma. GCL/IPL thickness, however, decreased in glaucomatous eyes but not with age. Therefore, GCL/IPL thickness is less influenced by age when monitoring patients with glaucoma or suspect glaucoma.

This paper is concerned with speech enhancement using Phase-Error based Filters (PEF) and Excitation Source (ES) information in car environments. For this purpose, we firstly use ES information to determine the time-delay from speech signals obtained by two microphones for sound source localization. Then, the phase-error based filters are performed by prior knowledge regarding the time delay obtained by ES information and the phases of the signals recorded by the microphones. The experimental results showed the effectiveness of the presented method for speech enhancement.

In this paper, the fundamental idea of Incremental Granular Models (IGM) introduced by Pedrycz and Kwak (2007) is followed and their comprehensive design framework is developed. In contrast to typical rule-based systems encountered in fuzzy modeling, the underlying principle of IGM is to consider a two-phase development. First, we build a Linear Regression (LR) model which could be treated as a global model. Next, all modeling errors are compensated by a collection of fuzzy rules that capture more localized nonlinearities of the system as a local model. Here we expand local granular models into Radial Basis Function Networks (RBFN) or Adaptive Neuro-Fuzzy Networks (ANFN) with the use of information granules through Context-based Fuzzy C-Means (CFCM) clustering in the design of incremental model. Numerical studies concern two datasets coming from the machine learning repository. The experimental results revealed that both Incremental RBFN (IRBFN) and Incremental ANFN (IANFN) showed good performance in comparison to the previous works.

Although navigation systems and automatic identification technologies are in use for nautical safety of ocean vessels, ship collisions are increasing due to the growing number of overseas transportations. Since these accidents are mostly caused by the watch officers and crews` human mistakes, it is essential to develop a new set of systems to prevent such misfortunes. Therefore, we suggest a control and management system which monitors crews in order to conduct secure on-board navigation and nautical safety. We also materialized a mobile terminal system and portable device for the crews to check the status of the ship, and we designed a platform for an integrated surveillance system to assure a harmless voyage. Lastly, we attested the mutual connections of integrated safety-related data.
