Aggregates of the protein tau are proposed to drive pathogenesis in neurodegenerative diseases. Tau can be targeted by using passively transferred antibodies (Abs), but the mechanisms of Ab protection are incompletely understood. In this work, we used a variety of cell and animal model systems and showed that the cytosolic Ab receptor and E3 ligase TRIM21 (T21) could play a role in Ab protection against tau pathology. Tau-Ab complexes were internalized to the cytosol of neurons, which enabled T21 engagement and protection against seeded aggregation. Ab-mediated protection against tau pathology was lost in mice that lacked T21. Thus, the cytosolic compartment provides a site of immunotherapeutic protection, which may help in the design of Ab-based therapies in neurodegenerative disease.

Over the past decade, smartphones have become the point of convergence for many applications and services. There is a growing trend in which traditional smart-card based services like banking, transport and access control are being provisioned through smartphones. Smartphones with Near Field Communication (NFC) capability can emulate a contactless smart card; popular examples of such services include Google Pay and Apple Pay. Similar to contactless smart cards, NFC-based smartphone transactions are susceptible to relay attacks. For contactless smart cards, distance-bounding protocols are proposed to counter such attacks; for NFC-based smartphone transactions, ambient sensors have been proposed as potential countermeasures. In this study, we have empirically evaluated the suitability of ambient sensors as a proximity detection mechanism for contactless transactions. To provide a comprehensive analysis, we also collected relay attack data to ascertain whether ambient sensors are able to thwart such attacks effectively. We initially evaluated 17 sensors before selecting 7 sensors for in-depth analysis based on their effectiveness as potential proximity detection mechanisms within the constraints of a contactless transaction scenario. Each sensor was used to record 1000 legitimate and relay (illegitimate) contactless transactions at four different physical locations. Near Field Communication (NFC) has enabled mobile phones to emulate contactless smart cards. Similar to contactless smart cards, they are also susceptible to relay attacks. To counter these, a number of methods have been proposed that rely primarily on ambient sensors as a proximity detection mechanism (also known as an anti-relay mechanism). In this paper, we, for the first time in academic literature, empirically evaluate a comprehensive set of ambient sensors for their effectiveness as a proximity detection mechanism. We selected 15 out of a total of 17 sensors available via the Google Android platform for evaluation, with the other two sensors unavailable on widely-used handsets. In existing academic literature, only 5 sensors have been proposed with positive results as a potential proximity detection mechanism. Each sensor, where feasible, was used to record the measurements of 1000 contactless transactions at four different physical locations. A total of 252 random users, random sample of the university student population, were involved during the field trails. The analysis of these transactions provides an empirical foundation to categorically answer whether ambient sensors provide a strong proximity detection mechanism for security sensitive applications like banking, transport and high-security access control. After careful analysis, we conclude that no single evaluated mobile ambient sensor is suitable for such critical applications in realistic deployment scenarios. Antibodies targeting the middle capsid protein VP6 are particularly abundant, and as VP6 is only exposed inside cells, neutralisation must be post-entry. However, while a system of poly immune globulin receptor (pIgR) transcytosis has been proposed for anti-VP6 IgAs, the mechanism by which VP6-specific IgG mediates protection remains less clear. We have developed an intracellular neutralisation assay to examine how antibodies neutralise rotavirus inside cells, enabling comparison between IgG and IgA isotypes. Unexpectedly we found that neutralisation by VP6-specific IgG was much more efficient than by VP6-specific IgA. This observation was highly dependent on the activity of the cytosolic antibody receptor TRIM21 and was confirmed using an in vivo model of murine rotavirus infection. Furthermore, mice deficient in only IgG and not other antibody isotypes had a serious deficit in intracellular antibody-mediated protection. The finding that VP6-specific IgG protect mice against rotavirus infection has important implications for rotavirus vaccination. Current assays determine protection in humans predominantly by measuring rotavirus-specific IgA titres. Measurements of VP6-specific IgG may add to existing mechanistic correlates of protection.

It is well-known that the Dolev-Yao adversary is a powerful adversary. Besides acting as the network, intercepting, sending, and composing messages, he can remember as much information as he needs. That is, his memory is unbounded. We recently proposed a weaker Dolev-Yao like adversary, which also acts as the network, but whose memory is bounded. We showed that this Bounded Memory Dolev-Yao adversary, when given enough memory, can carry out many existing protocol anomalies. In particular, the known anomalies arise for bounded memory protocols, where there is only a bounded number of concurrent sessions and the honest participants of the protocol cannot remember an unbounded number of facts nor an unbounded number of nonces at a time. This led us to the question of whether it is possible to infer an upper-bound on the memory required by the Dolev-Yao adversary to carry out an anomaly from the memory restrictions of the bounded protocol. This paper answers this question negatively (Theorem 2). The second contribution of this paper is the formalization of Progressing Collaborative Systems that may create fresh values, such as nonces. In this setting there is no unbounded adversary, although bounded memory adversaries may be present. We prove the NP-completeness of the reachability problem for Progressing Collaborative Systems that may create fresh values.
