Este estudio examina la educación ambiental relacionada con el cambio climático que poseen los docentes de la Universidad Nacional de Asunción y propone medidas que puedan contribuir con el desarrollo sostenible. Se utiliza una investigación con enfoque cualitativo, con un diseño no experimental y del tipo descriptivo. Como instrumento de recolección de datos se emplea la técnica de entrevistas personales semiestructuradas, basadas en un cuestionario. Se identifica que la población de estudio cuenta con una percepción adecuada del cambio climático. Sin embargo, se evidencia que los miembros de dicha población poseen medianos niveles de conocimientos sobre las informaciones científicas que sustentan la temática. Por tanto, se observa la necesidad de fortalecer dichos conocimientos para evitar barreras en el momento de transmitir las ciencias a los estudiantes. Por otra parte, poseen una actitud positiva para contribuir con el cuidado del medioambiente. Asimismo, perciben la necesidad de que se introduzcan algunas propuestas pedagógicas que promuevan una mejor comprensión de la temática y favorezcan la formación de los futuros profesionales con valor de sostenibilidad.

5. Cultivation of previously uncultured microorganisms with a continuous-flow down-flow hanging sponge (DHS) bioreactor, using a syntrophic archaeon culture obtained from deep marine sediment as a case study

In microbiology, cultivation is a central approach for uncovering novel physiology, ecology, and evolution of microorganisms, but conventional methods have left many microorganisms found in nature uncultured. To overcome the limitations of traditional methods and culture indigenous microorganisms, we applied a two-stage approach: enrichment/activation of indigenous organisms by using a continuous-flow down-flow hanging sponge bioreactor and subsequent selective batch cultivation. Here, we provide a protocol for this bioreactor-mediated technique using activation of deep marine sediment microorganisms and downstream isolation of a syntrophic co-culture containing an archaeon closely related to the eukaryote ancestor (Candidatus Promethearchaeum syntrophicum strain MK-D1) as an example. Both stages can easily be tailored to target other environments and organisms by modifying the inoculum, feed solution/gases, attachment material and/or cultivation media. We anaerobically incubate polyurethane sponges inoculated with deep-sea methane seep sediment in a reactor at 10 °C and feed anaerobic artificial seawater medium and methane. Once phylogenetically diverse and metabolically active microorganisms are adapted to synthetic conditions in the reactor, we transition to growing community samples in glass tubes with the above medium, simple substrates and selective compounds (e.g., antibiotics). To accommodate for the slow growth anticipated for target organisms, primary cultures can be incubated for ≥6-12 months and analyzed for community composition even when no cell turbidity is observed. One casamino acid- and antibiotic-amended culture prepared in this way led to the enrichment of uncultured archaea. Through successive transfer in vitro combined with molecular growth monitoring, we successfully obtained the target archaeon with its partner methanogen as a pure syntrophic co-culture.

Cancer chemotherapy outcomes have improved markedly after the introduction of cytotoxic anticancer drugs, molecular target medicines, and immune checkpoint inhibitors (ICIs) into clinical practice. Chemotherapy regimens are specified treatment plans that include the dose of each anticancer drug, duration of treatment, and other supportive care drugs such as antiemetics. A chemotherapy regimen leads to the standardization of cancer pharmacotherapy, leading to medical safety and efficiency. Most molecular target medicines and ICIs have companion diagnostic agents (CDxs) that predict the treatment effect based on the gene variant (driver gene) and its expression level. In addition, oncogene panel tests have recently become covered by health insurance in Japan. However, there are some strict regulations and problems with their clinical use. Most molecular target medicines and ICIs have various side effects that are different from those of cytotoxic anticancer drugs. To provide more effective care and nursing to patients receiving cancer chemotherapy, clinical nurses need to understand the basic pharmacologic characteristics of each cancer medicine and provide patients with information about side effects and potential effects on daily life.

9. Marinagarivorans cellulosilyticus sp. nov., a cellulolytic bacterium isolated from the deep-sea off Noma-misaki, Japan

Cells from strain GE09T, isolated from an artificially immersed nanofibrous cellulose plate in the deep sea, were Gram-stain-negative, motile, aerobic cells that could grow with cellulose as their only nutrient. Strain GE09T was placed among members of Cellvibrionaceae , in the Gammaproteobacteria , with Marinagarivorans algicola Z1T, a marine degrader of agar, as the closest relative (97.4 % similarity). The average nucleotide identity and digital DNA–DNA hybridization values between GE09T and M. algicola Z1T were 72.5 and 21.2 %, respectively. Strain GE09T degraded cellulose, xylan and pectin, but not starch, chitin and agar. The different carbohydrate-active enzymes encoded in the genomes of strain GE09T and M. algicola Z1T highlights their differences in terms of target energy sources and reflects their isolation environments. The major cellular fatty acids of strain GE09T were C18 : 1 ω7c, C16 : 0 and C16 : 1 ω7c. The polar lipid profile showed phosphatidylglycerol and phosphatidylethanolamine. The major respiratory quinone was Q-8. Based on these distinct taxonomic characteristics, strain GE09T represents a new species in the genus Marinagarivorans , for which we propose the name Marinagarivorans cellulosilyticus sp. nov. (type strain GE09T=DSM 113420T=JCM 35003T).

10. Fundidesulfovibrio magnetotacticus sp. nov., a sulphate-reducing magnetotactic bacterium, isolated from sediments and freshwater of a pond

A sulphate-reducing magnetotactic bacterium, designated strain FSS-1T, was isolated from sediments and freshwater of Suwa Pond located in Hidaka, Saitama, Japan. Strain FSS-1T was a motile, Gram-negative and curved rod-shaped bacterium that synthesizes bullet-shaped magnetite (Fe3O4) nanoparticles in each cell. Strain FSS-1T was able to grow in the range of pH 6.5–8.0 (optimum, pH 7.0), 22–34 °C (optimum, 28 °C) and with 0–8.0 g l−1 NaCl (optimum, 0–2.0 g l−1 NaCl). Strain FSS-1T grew well in the presence of 50 µM ferric quinate as an iron source. The major fatty acids were anteiso-C15 : 0, iso-C15 : 0 and anteiso-C17 : 0. The major menaquinone was MK-7 (H2). Strain FSS-1T contained desulfoviridin, cytochrome c 3 and catalase, but did not contain oxidase. Strain FSS-1T used fumarate, lactate, pyruvate, malate, formate/acetate, succinate, tartrate, ethanol, 1-propanol, peptone, soytone and yeast extract as electron donors, while the strain used sulphate, thiosulphate and fumarate as electron acceptors. Fumarate was fermented in the absence of electron acceptors. Analysis of the 16S rRNA gene sequence showed that strain FSS-1T is a member of the genus Fundidesulfovibrio . The gene sequence showed 96.7, 95.0, 92.0, 91.2 and 91.4% similarities to the most closely related members of the genera Fundidesulfovibrio putealis B7-43T, Fundidesulfovibrio butyratiphilus BSYT, Desulfolutivibrio sulfoxidireducens DSM 107105T, Desulfolutivibrio sulfodismutans ThAc01T and Solidesulfovibrio magneticus RS-1T, respectively. The DNA G+C content of strain FSS-1T was 67.5 mol%. The average nucleotide identity value between strain FSS-1T and F. putealis B7-43T was 80.7 %. Therefore, strain FSS-1T represents a novel species within the genus Fundidesulfovibrio , for which the name Fundidesulfovibrio magnetotacticus sp. nov. is proposed (=JCM 32405T=DSM 110007T).

13. Rectinema subterraneum sp. nov, a chemotrophic spirochaete isolated from the deep terrestrial subsurface

A novel, obligately anaerobic bacterium (strain SURF-ANA1T) was isolated from deep continental subsurface fluids at a depth of 1500 m below surface in the former Homestake Gold Mine (now Sanford Underground Research Facility, in Lead, South Dakota, USA). Cells of strain SURF-ANA1Twere Gram-negative, helical, non-spore-forming and were 0.25–0.55×5.0–75.0 µm with a wavelength of 0.5–0.62 µm. Strain SURF-ANA1Tgrew at 15–50 °C (optimally at 40 °C), at pH 4.8–9.0 (pH 7.2) and in 1.0–40.0 g l−1NaCl (10 g l−1NaCl). The strain grew chemoheterotrophically with hydrogen or mono-, di- and polysaccharides as electron donors. The major cellular fatty acids in order of decreasing abundance (comprising >5% of total) were 10-methyl C16:0, iso-C15:0, C18:2and C18:0dimethyl acetal (DMA) and C20:0methylene-nonadecanoic acid. Phylogenetic analysis based on the 16S rRNA gene sequence of strain SURF-ANA1Tindicated a closest relationship with the recently characterizedRectinema cohabitans(99%). Despite high sequence identity, because of its distinct physiology, morphology and fatty acid profile, strain SURF-ANA1Tis considered to represent a novel species within the genusRectinema, for which the nameRectinema subterraneumsp. nov. is proposed. To our knowledge, this is the first report of an isolate within the phylumSpirochaetesfrom the deep (>100 m) terrestrial subsurface. The GenBank/EMBL/DDBJ accession numbers for the 16S rRNA gene and genomic sequences of strain SURF-ANA1Tare KU359248 and GCF 009768935.1, respectively. Intravenous administration of magnesium with a short hydration regimen is recommended for patients receiving high-dose cisplatin to protect against cisplatin-induced nephrotoxicity. However, the optimal dose of magnesium supplementation has not been clarified. The aim of this trial was to investigate the safety and efficacy of a short hydration regimen with 20 mEq of magnesium supplementation for lung cancer patients receiving cisplatin-based chemotherapy. The key eligibility criteria included cytologically or histologically diagnosed lung cancer, candidacy for cisplatin-based (≥ 60 mg/m2) chemotherapy or chemoradiotherapy, no prior chemotherapy, aged 20–75 years, and adequate renal function. Cisplatin was administered with pre-hydration with 20 mEq of magnesium sulfate. Mannitol was administered just before cisplatin infusion to enforce diuresis. The primary endpoint was the proportion of patients who underwent cisplatin-based chemotherapy with a short hydration regimen with 20 mEq of magnesium supplementation without a grade 2 or higher elevation in creatinine. Forty patients with a median age of 66 years (range 35–74) were prospectively enrolled. Median baseline creatinine was 0.71 mg/dL. Median dose of cisplatin in the first cycle was 80 mg/m2. In the first cycle, no patients developed grade 2 creatinine toxicity. During the treatment period, one patient developed grade 2 creatinine elevation; thus, the proportion of patients without a grade 2 or higher elevation in creatinine was 97.5% (95% confidence interval 86.8–99.9). A short hydration regimen with 20 mEq of magnesium supplementation is safe and feasible for patients with lung cancer receiving cisplatin-based chemotherapy.

In anticipation of the advent of UHD-1 (4K)/UHD-2 (8K) broadcasting in Japan, we developed ultrahigh-definition TV (UHDTV) studios to promote UHD content. Regarding UHD-1/4K productions, it was necessary to adapt to simultaneous production of 2K and 4K programs to achieve an efficient workflow. Thus, we used 12G-SDI (SMPTE ST 2082), which is highly compatible with the 2K equipment, and developed a realtime transform device and control system to simultaneously produce high-quality content in International Telecommunication Union-Rec.2020/HDR (high dynamic range) and Rec.709/SDR (standard dynamic range). We designed the transform algorithm based on our studies and experience. Also, to overcome the high bitrate issues of UHD-2/8K, we introduced a mezzanine compression approach in the 8K studio. To interface with various devices, a serial digital interface/internet protocol (SDI/IP) hybrid system was adopted in the studio, where we can produce programs of the highest quality in the world. We discuss the key points of the novel studio designs and highlight certain challenges to be addressed in the future.

16. UHD-2/8K Camera Recorder Using Organic CMOS Image Sensor

We developed an ultrahigh-definition (UHD)-2/8K camera recorder that uses a complementary metal–oxide semiconductor (CMOS) image sensor with an organic thin-film, instead of photodiodes for the opto-electrical conversion element. The advent of UHD-2/8K Super Hi-Vision broadcasts and an increase in the production of UHD-2/8K programs across a variety of genres necessitate the development of a sensor with a good signal-to-noise ratio (SNR) and high-dynamic range (HDR) to capture the full range of HDR video. To ensure successful use of this sensor, we studied the level diagrams (e.g., balancing of sensitivity, SNR, and dynamic range) that would best fit the sensor’s characteristics and use its strengths, and then proceeded accordingly. For the current UHD-2/8K location shooting, a workflow is needed to record raw data and post-process them into a practical video format. To implement a more efficient workflow, we chose to use Grass Valley HQX compression encoding while adding features that make it possible to store video data in a compressed form and simultaneously generate high-definition television (HDTV) proxy data. The workflow that we envisioned uses proxy data for offline editing and relinks them using online editing equipment.

17. An ultrasensitive nanofiber-based assay for enzymatic hydrolysis and deep-sea microbial degradation of cellulose

Substrates for enzymatic reactions, such as cellulose and chitin, are often insoluble in water. The enzymatic degradation of these abundant organic polymers plays a dominant role in the global carbon cycle and has tremendous technological importance in the production of bio-based chemicals. In addition, biodegradation of plastics is gaining wide attention. However, despite the significance, assaying these degradation reactions remains technically challenging owing to the low reaction rate, because only the surface of the substrate is accessible to the enzymes. We developed a nanofiber-based assay for the enzymatic hydrolysis of cellulose. This assay facilitated the quantification of the enzymatic hydrolysis of1 ng crystalline cellulose. Utilization of the assay for the functional screening of cellulolytic microorganisms revealed an unprecedented genetic diversity underlying the production of deep-sea cellulase. This study reiterates that interdisciplinary efforts, such as from nanotechnology to microbiology, are critical for solving sustainability challenges.

18. Sulfur-assisted urea synthesis from carbon monoxide and ammonia in water

- Abstract
Efficient conversion of carbon monoxide into urea in an aqueous ammonia solution was demonstrated through coupling with the elemental sulfur reduction to polysulfides. Polysulfides control the overall reaction rate while suppressing the accumulation of a by-product, hydrogen sulfide. These functions follow basic kinetic and thermodynamic theories, enabling prediction-based reaction control. This operational merit, together with the superiority of water as a green solvent, suggests that our demonstrated urea synthesis is a promising option for sulfur utilization beneficial for agricultural production.

SummaryMany age-dependent neurodegenerative diseases, like Alzheimer’s and Parkinson’s, are characterised by abundant inclusions of amyloid filaments. Filamentous inclusions of the proteins tau, amyloid-β (Aβ), α-synuclein and TDP-43 are the most common. Here, we used electron cryo-microscopy (cryo-EM) structure determination to show that residues 120-254 of the lysosomal type II transmembrane protein 106B (TMEM106B) also form amyloid filaments in the human brain. We solved cryo-EM structures of TMEM106B filaments from the brains of 22 individuals with neurodegenerative conditions, including sporadic and inherited tauopathies, Aβ-amyloidoses, synucleinopathies and TDP-43opathies, as well as from the brains of two neurologically normal individuals. We observed three different TMEM106B folds, with no clear relationship between folds and diseases. The presence of TMEM106B filaments correlated with that of a 29 kDa sarkosyl-insoluble fragment of the protein on Western blots. The presence of TMEM106B filaments in the brains of older, but not younger, neurologically normal individuals indicates that they form in an age-dependent manner.

The origin of eukaryotes remains unclear1–4. Current data suggest that eukaryotes may have emerged from an archaeal lineage known as ‘Asgard’ archaea5,6. Despite the eukaryote-like genomic features that are found in these archaea, the evolutionary transition from archaea to eukaryotes remains unclear, owing to the lack of cultured representatives and corresponding physiological insights. Here we report the decade-long isolation of an Asgard archaeon related to Lokiarchaeota from deep marine sediment. The archaeon—‘Candidatus Prometheoarchaeum syntrophicum’ strain MK-D1—is an anaerobic, extremely slow-growing, small coccus (around 550 nm in diameter) that degrades amino acids through syntrophy. Although eukaryote-like intracellular complexes have been proposed for Asgard archaea6, the isolate has no visible organelle-like structure. Instead, Ca. P. syntrophicum is morphologically complex and has unique protrusions that are long and often branching. On the basis of the available data obtained from cultivation and genomics, and reasoned interpretations of the existing literature, we propose a hypothetical model for eukaryogenesis, termed the entangle–engulf–endogenize (also known as E3) model., Isolation and characterization of an archaeon that is most closely related to eukaryotes reveals insights into how eukaryotes may have evolved from prokaryotes.

A novel slow-growing, facultatively anaerobic, filamentous bacterium, strain MO-CFX2T, was isolated from a methanogenic microbial community in a continuous-flow bioreactor that was established from subseafloor sediment collected off the Shimokita Peninsula of Japan. Cells were multicellular filamentous, non-motile and Gram-stain-negative. The filaments were generally more than 20 µm (up to approximately 200 µm) long and 0.5–0.6 µm wide. Cells possessed pili-like structures on the cell surface and a multilayer structure in the cytoplasm. Growth of the strain was observed at 20–37 °C (optimum, 30 °C), pH 5.5–8.0 (pH 6.5–7.0), and 0–30 g l−1 NaCl (5 g l−1 NaCl). Under optimum growth conditions, doubling time and maximum cell density were estimated to be approximately 19 days and ~105 cells ml−1, respectively. Strain MO-CFX2T grew chemoorganotrophically on a limited range of organic substrates in anaerobic conditions. The major cellular fatty acids were saturated C16 : 0 (47.9 %) and C18 : 0 (36.9 %), and unsaturated C18 : 1ω9c (6.0 %) and C16 : 1ω7 (5.1 %). The G+C content of genomic DNA was 63.2 mol%. 16S rRNA gene-based phylogenetic analysis showed that strain MO-CFX2T shares a notably low sequence identity with its closest relatives, which were Thermanaerothrix daxensis GNS-1T and Thermomarinilinea lacunifontana SW7T (both 85.8 % sequence identity). Based on these phenotypic and genomic properties, we propose the name Aggregatilinea lenta gen. nov., sp. nov. for strain MO-CFX2T (=KCTC 15625T, =JCM 32065T). In addition, we also propose the associated family and order as Aggregatilineaceae fam. nov. and Aggregatilineales ord. nov., respectively.

A novel hydrogenotrophic methanogen, strain HHBT, was isolated from a deep-sea hydrothermal vent chimney sample collected from Beebe Vent Field at the Mid-Cayman Spreading Center, Caribbean Sea. The cells were non-motile regular to irregular cocci possessing several flagella. The novel isolate grew at 60–80 °C, pH 5.0–7.4 and with 1–4 % of NaCl (w/v). The isolate utilized H2/CO2 as the only substrates for growth and methane production. The results of phylogenetic analyses of both 16S rRNA and mcrA gene sequences and comparative genome analysis indicated that HHBT represented a member of the order Methanococcales , and was closely related to the members of the genera Methanothermococcus and Methanotorris . The most closely related species were Methanothermococcus okinawensis IH1T and Methanotorris igneus Kol 5T in comparison of 16S rRNA gene sequences (each with 93 % identity), and Methanotorris formicicus Mc-S-70T in the case of deduced amino acid sequence similarity of mcrA genes (92 % similarity). The ANI and AAI values between HHBT and the members of the genera Methanothermococcus and Methanotorris were 69–72 % and 66–70 %, respectively. Although many of the morphological and physiological characteristics were quite similar between HHBT and the species of the genera Methanothermococcus and Methanotorris , they were distinguishable by the differences in susceptibility to antibiotics, formate utilization, growth temperature and NaCl ranges. On the basis of these phenotypic, phylogenetic and genomic properties, we propose that strain HHBT represents a novel species, of a novel genus, Methanofervidicoccus abyssi gen. nov., sp. nov. The type strain is HHBT (=JCM 32161T=DSM 105918T).

Chemotherapy-induced nausea and vomiting (CINV) can cause anorexia, weight loss and deterioration of patient quality of life. It is one of the most unpleasant adverse effects of chemotherapy treatment regimens. For the optimal treatment of gastrointestinal symptoms during urothelial carcinoma chemotherapy, the present study investigated the association between gastrointestinal symptoms and therapeutic effects of gemcitabine plus platinum [cisplatin (GC) or carboplatin (GCa)] therapies. The incidence and frequency of nausea/vomiting with GC split therapy (gemcitabine, 1,000 mg/m(2) on days 1 and 8; split-dose cisplatin, 35 mg/m(2) on days 1 and 8; 21-day schedule) and GCa therapy [gemcitabine, 750-1,000 mg/m(2) on days 1, 8 and 15; carboplatin, area under the blood concentration-time curve=5 mg min/ml (Calvert formula) on day 2; 28-day schedule] were lower compared with those of GC therapy (gemcitabine, 1,000 mg/m(2) on days 1, 8 and 15; single-dose cisplatin 70 mg/m(2) on day 2; 28-day schedule). However, no differences in therapeutic outcomes were observed among therapies. GCa therapy, regardless of renal function, and GC split therapy demonstrated significant increases compared with GC therapy in alleviating gastrointestinal symptoms associated with cancer chemotherapy in patients with urothelial carcinoma. Overall, these results suggested that split-dose cisplatin administration or the use of carboplatin instead of cisplatin may be useful in patients who experience CINV without compromising treatment effectiveness.

Although the Cockcroft-Gault equation is still used for the dose adjustment of many drugs that have been approved prior to creatinine standardization, the clinical impact of standardized creatinine in the dose adjustment of capecitabine is poorly understood. We focused on patients with borderline renal function and evaluated the tolerability and safety of capecitabine in patients who received capecitabine plus oxaliplatin (Cape-Ox). We retrospectively identified patients with resected colorectal cancer who had received adjuvant therapy with Cape-Ox regimen. Creatinine clearance (CrCL) was calculated by the Cockcroft-Gault equation with standardized creatinine measured using enzymatic methods, and adjusted CrCL was estimated by adding 0.2 (mg/dL) to the serum creatinine in the equation. We defined patients with "pseudo-normal" renal function as those who had an adjusted CrCL of ≤50 mL/min in patients with normal renal function (CrCL50 mL/min). We evaluated the tolerability and grade 2 or severer adverse events of capecitabine treatment. One hundred four patients had normal and 10 had impaired renal function (CrCL50 mL/min). Among the 104 patients with normal renal function, 23 (22.1%) had pseudo-normal renal function. Seventeen patients completed the eight cycles of Cape-Ox therapy without treatment delay or dose reduction, and all of them had truly normal renal function. The patients with pseudo-normal renal function were more likely to have grade 2 or severer thrombocytopenia than those with truly normal renal function. We should recognize correctly the clinical impact of standardized creatinine in the treatment of borderline renal function with Cape-Ox regimen in patients.

27. Polysulfide-assisted urea synthesis from carbon monoxide and ammonia in water

- Abstract
Efficient conversion of carbon monoxide into urea in an aqueous ammonia solution was demonstrated through coupling with the elemental sulfur reduction to polysulfides. This reaction starts with a simple mixture of carbon monoxide, ammonia, elemental sulfur, and a small amount of hydrogen sulfide for polysulfide formation, enabling an almost complete conversion of 1 atm of carbon monoxide to urea (95–100% yield) within 216, 64, and 32 h at 35 °C, 50 °C, and 65 °C, respectively. Polysulfides control the overall reaction rate while suppressing the accumulation of a by-product, hydrogen sulfide, to less than 1 Pa. These functions follow simple kinetic and thermodynamic theories, enabling prediction-based reaction control. This operational merit, together with the superiority of water as a green solvent, suggests that our demonstrated urea synthesis is a promising option for sulfur utilization beneficial for agricultural production.

This paper highlights our developments in hardware compression for production applications. The underlying compression scheme shows a minimal quality degradation through multiple encode/decode cycles, making it suitable for production systems. The Tiny Codec (TICO) codec takes an uncompressed UHDTV-2 bitstream of 48 Gbits/s (59.94 Hz, 10 bits, 4:2:2) and compresses it to fit into a single 12G-Serial Digital Interface (SDI) cable. The device can also be fitted with IP interfaces. These changes permit significant reductions in cable and switcher complexity.

AIM To investigate the real-world efficacy and safety of sofosbuvir/ribavirin (SOF/RBV) therapy for Japanese patients with genotype 2 hepatitis C virus (GT2-HCV). METHODS A total of 182 patients with GT2-HCV infection who received SOF/RBV therapy for 12 wk at our hospital were enrolled. The patients comprised 122 men and 60 women (age range: 17-84 years; mean age ± SD: 60.1 ± 12.1 years). Relationships between virological response and clinical data were examined by logistic regression analyses. RESULTS The proportions of patients with liver cirrhosis and history of hepatocellular carcinoma (HCC) were 29.0% and 17.3%, respectively. The proportion of patients with prior interferon (IFN)-based therapy was 25.6%. SOF/RBV therapy rapidly decreased HCV RNA levels. Several patients required RBV dose reduction because of anemia or fatigue. Four patients discontinued the therapy. The rates of sustained virological response at 12 wk after the end of treatment were 87.9% (intention to treat: 160/182) and 94.1% (per protocol: 159/169). Multivariate analyses showed that history of HCC or IFN-based therapy independently reduced the efficacy of SOF/RBV therapy. CONCLUSION SOF/RBV therapy for GT2-HCV is safe, highly tolerated, and effective. History of HCC or IFN-based therapy independently reduces the efficacy of this treatment.

Background Although opioid-induced nausea and vomiting (OINV) often result in analgesic undertreatment in patients with cancer, no randomized controlled trials have evaluated the efficacy of prophylactic antiemetics for preventing OINV. We conducted this randomized, placebo-controlled, double-blind trial to evaluate the efficacy and safety of prophylactic treatment with prochlorperazine for preventing OINV. Materials and Methods Cancer patients who started to receive oral oxycodone were randomly assigned in a 1:1 ratio to receive either prochlorperazine 5 mg or placebo prophylactically, given three times daily for 5 days. The primary endpoint was the proportion of patients who had a complete response (CR) during the 120 hours of oxycodone treatment. CR was defined as no emetic episode and no use of rescue medication for nausea and vomiting during 5 days. Key secondary endpoints were the proportion of patients with emetic episodes, proportion of patients with moderate or severe nausea, quality of life, and proportion of treatment withdrawal. Results From November 2013 through February 2016, a total of 120 patients were assigned to receive prochlorperazine (n = 60) or placebo (n = 60). There was no significant difference in CR rates (69.5% vs. 63.3%; p = .47) or any secondary endpoint between the groups. Patients who received prochlorperazine were more likely to experience severe somnolence (p = .048). Conclusion Routine use of prochlorperazine as a prophylactic antiemetic at the initiation of treatment with opioids is not recommended. Further research is needed to evaluate whether other antiemetics would be effective in preventing OINV in specific patient populations. Implications for Practice Prophylactic prochlorperazine seems to be ineffective in preventing opioid-induced nausea and vomiting (OINV) and may cause adverse events such as somnolence. Routine use of prophylactic prochlorperazine at the initiation of treatment with opioids is not recommended. Further research is needed to evaluate whether other antiemetics would be effective in preventing OINV in specific patient populations.

A 49-year-old man with chronic hepatitis B receiving treatment with entecavir visited a hospital with a complaint of abdominal pain. Computed tomography (CT) showed 2 liver tumors, each measuring 1cm in diameter, 1 in segment 7 and 1 in segment 4. Magnetic resonance imaging (MRI) showed a hypervascular tumor in segment 7 that appeared in a site different from that seen on CT. The liver tumor in segment 4 was not detected by MRI. Two months later, MRI showed a new liver tumor in segment 7/6 and that the liver tumor in segment 7 had increased to 2cm in diameter;blood tests showed eosinophilia. Enzyme-linked immunosorbent assay showed a high serum Toxocara antibody. The patient was diagnosed as having hepatic toxocariasis and was treated with albendazole for 8 weeks. After treatment, MRI showed that the liver tumors disappeared. Eosinophilia, multiple lesions, and the disappearance of the tumors were characteristic findings of visceral larva migrans.

AIM This study aimed to compare the incidence of infusion site reactions (ISRs) induced by intravenous administration of brand-name and generic vinorelbine (VNR) for treating non-small cell lung cancer. METHOD This single-centre retrospective cohort study was conducted by medical chart review of VNR infusions. ISRs were defined as symptoms around the infusion site, including pain, redness and swelling. ISRs requiring treatment were defined as ISRs requiring treatments including steroid ointments, vein repuncture and local steroid injections. RESULTS In all, 1973 VNR infusions were administered to 340 patients (brand-name 141 patients, generic 199 patients). ISRs and ISRs requiring treatment were observed in 161 and 100 patients, respectively. The ISR incidence per patient and per injection was significantly higher in generic VNR-treated patients than in brand-name VNR-treated patients (53.3% vs 39.0%, P = 0.0112 and 15.0% vs 9.9%, P = 0.0008, respectively). The frequency of ISRs requiring treatment was also significantly higher in the generic group (per patient 36.7% vs 19.2%, P = 0.0005; per injection 11.3% vs 5.5%, P < 0.0001). Multivariate analysis revealed that generic VNR was significantly associated with an increased risk of ISRs (per patient adjusted odds ratio [AOR] 1.775, P = 0.0155; per injection AOR 1.672, P = 0.004) and ISRs requiring treatment (per patient AOR 2.422, P = 0.0012; per injection AOR 2.286, P = 0.001). CONCLUSION Intravenous infusion of generic VNR was associated with an increased risk of ISRs. Further research is needed to elucidate the mechanism underlying the increased incidence of ISRs with generic VNR.

The dosage of cisplatin is adjusted according to creatinine clearance (Ccr) estimated by the Cockcroft-Gault formula, which is commonly used as a marker for renal function. NHK, Nippon Hoso Kyokai (Japan Broadcasting Corporation), is going to present highly realistic broadcasts of the 2020 Olympic Games in Tokyo via 8K Super Hi-Vision (also known as ultrahigh definition TV (UHDTV)), the world's most sophisticated broadcasting system. Here, we describe our preparations for the 8K broadcasting, particularly for the test broadcasting via satellite that began in 2016. In particular, we describe our activities related to standardization and development of program production equipment, play-out, and a distribution system for the 8K broadcasting. Regarding the production and distribution systems, we present the results of experimental satellite broadcasting performed in 2016. We also discuss the current status of the high dynamic range function for the 8K system, the multiformat production workflow, and remaining issues.

A method called “adaptive channel diversity” (ACD), which improves wireless performance in terms of reliability and latency, is proposed. With ACD, the optimum frequency channels are selected on the basis of the amount of dynamic radio interference. ACD was implemented in two prototype systems operating in the 2.4-GHz industry–science–medical band based on the ISA100.11a standard, namely, time-division multiple access and frequency channel hopping. According to an evaluation of the prototype systems, a 10−4 packet error rate was achieved within a 1.3-s latency for a 30-terminal star topology and within a 0.85-s latency for a 5-terminal multihop topology. These evaluation results are consistent with the results of simulations based on probabilistic analysis.

The occurrence of chalky rice (Oryza sativa L.) grains caused by high temperature is a serious problem in rice production. Of the several kinds of chalky grains, milky-white grains are not well analyzed. The milky-white rice grain phenomenon is caused by genetic factors as well as environmental and nutritional conditions. To analyze the genetic control system for rice grain quality, we raised recombinant inbred lines from progeny produced from 'Tsukushiroman' (high temperature sensitive) and 'Chikushi 52' (high temperature tolerant) cultivars. Quantitative trait locus (QTL) analysis revealed that the QTL on chromosome 4, linked to the simple sequence repeat marker RM16424, contributed substantially to the occurrence of milky-white grains, as it was detected over two experimental years. To validate the effect of the QTL, we developed near isogenic lines that have the 'Chikushi 52' segment on the short arm of chromosome 4 in the 'Tsukushiroman' genetic background, and that had a lower milky-white grain ratio than that of 'Tsukushiroman' when exposed to high temperatures during the ripening period. These results suggest that the 'Chikushi 52' allele on chromosome 4 suppresses the occurrence of milky-white grains and improves rice grain quality under heat stress during the grain ripening period.

Axitinib is a selective tyrosine kinase inhibitor of VEGF receptors, approved for advanced renal cell carcinoma (RCC). Associations between axitinib plasma exposure, genetic polymorphisms of ABC transporters and axitinib-induced toxicities have not been adequately explored. Twenty RCC patients treated with axitinib were enrolled in this study. Blood samples were collected 0, 0.5, 1, 2, 4, and 6 h after administration of axitinib on day 1 and at steady state. Plasma concentrations of axitinib were analyzed by UPLC–MS/MS. The ABCG2 (421C>A) and ABCB1 (1236C>T, 2677G>T/A, 3435C>T) genetic polymorphisms were determined by real-time PCR. ABCB1 haplotype was associated with increased dose-adjusted area under the plasma concentration–time curve (AUC) of axitinib at steady state. The incidence of fatigue during therapy was associated with high AUC0–6 of axitinib (P = 0.013). The treatment period without discontinuation or dose reduction due to adverse events in patients with high AUC0–6 of axitinib was significantly shorter than for those with low AUC0–6 (P = 0.024). No significant differences were found in the frequency of adverse events among the ABCG2 genotype and ABCB1 haplotype groups. Our results have demonstrated that adverse events leading to discontinuation or dose reduction in axitinib were associated with increased axitinib plasma exposure, but not directly with genetic polymorphisms of ABC transporters. Therefore, measurement of steady state axitinib plasma concentrations may be useful in avoiding adverse events in axitinib therapy.

Background: Serum lactate dehydrogenase (LDH) levels could be a prognostic factor for sorafenib-treated patients with several types of solid tumor because it reflects hypoxic circumstances in aggressive tumors. For hepatocellular carcinoma (HCC), however, the prognostic role of LDH has been controversial. Liver fibrosis can potentially cause hypoxia in the liver, which has not been previously studied in the patients with advanced HCC. Thus, we aimed to analyze the prognostic role of LDH based on the degree of fibrosis. Methods: Eighty-nine consecutive patients with HCC (Child-Pugh class A) who were treated using sorafenib were enrolled into this study. Pretreatment characteristics and changes in hepatic functional tests based on early response to sorafenib and serum LDH levels were analyzed. The degree of fibrosis was estimated using the aspartate aminotransferase (AST) to platelet ratio index (APRI), and the tumor response was evaluated after 3 months of sorafenib treatment. Results: Overall, five patients discontinued sorafenib within 4 weeks. For the other 84 patients, those with progressive disease (PD) had significantly high pretreatment LDH levels, which correlated with the APRI score but not with the tumor stage. Multivariate logistic analysis revealed that older age and lower pretreatment LDH levels were independent prognostic factors for a better response to sorafenib. In patients who discontinued sorafenib early, three experienced acute liver failure accompanied with an increase in serum LDH. Conclusions: We demonstrated that baseline serum LDH levels in HCC patients were affected by liver fibrosis but not by the tumor stage, and these LDH levels could be a marker for early response to sorafenib. A marked increase in serum LDH levels during sorafenib administration might also indicate subsequent acute liver failure. Close observation of serum LDH levels before and during sorafenib treatment could be useful in managing treatment of patients receiving this therapy.

We, herein, report a 61-year-old male patient with amyotrophic lateral sclerosis (ALS) complicated pneumomediastinum while using mechanical insufflation-exsufflation (MI-E) after recovery from riluzole (RZ)-induced interstitial lung disease (RZ-ILD). After the treatment of RZ-ILD, he required non-invasive mechanical ventilation (NIV) at minimal pressure settings and MI-E to manage ALS-related breathing and airway-clearance issues, respectively. After a while, he developed progressive worsening dyspnoea, and chest computed tomography revealed extensive pneumomediastinum that had spread to the area surrounding the oesophagus, the retrosternal space, and the pericardial space. He was treated with immediate discontinuation of MI-E; however, he had to keep using NIV to support his severe respiratory muscle involvement. Pneumomediastinum gradually reduced in size and no recurrence of pneumomediastinum occurred. The clinical course of our patient suggests that excessive coughing associated with MI-E combined with his previous RZ-ILD, which potentially renders his lungs vulnerable to airway pressure, may have been the aetiological factors for secondary pneumomediastinum, i.e. barotrauma. Clinicians should be aware of the risk of pneumomediastinum while using MI-E in patients with ALS, who have other pre-existing risk factors for pneumomediastinum, such as drug-induced ILD in our case., Highlights • Pneumomediastinum could occur while using MI-E in ALS patients who recovered from drug-induced interstitial lung disease. • MI-E could be a cause of secondary pneumomediastinum in patients with pre-existing risk factors for pneumomediastinum. • Pneumomediastinum should be considered when acute progressive dyspnoea develops during the use of MI-E in ALS patient.

High temperature stress during ripening increases the frequency of chalky grains, resulting in a lower market value for rice (Oryza sativa L.). Changes in starch properties and the accumulation pattern of storage proteins are proposed to be related to the occurrence of chalky grains. This study investigated changes in the accumulation of key storage compounds in the grains of Japanese highly palatable rice cultivars, subjected to high temperature stress when grown in a growth chamber and the field. The 13 kDa prolamin content was significantly reduced in a highly heat-sensitive cultivar, Tsukushiroman, whereas the 13 kDa prolamin content was not affected in a heat-tolerant cultivar, Genkitsukushi, even in a high temperature chamber condition (31/26 °C day/night), when compared with the control chamber condition (26/21 °C day/night) for both genotypes. In addition, grains grown in field conditions revealed that severely chalky grains had less 13 kDa prolamin than perfect grains in all five genotypes. Changes in amylose content and the distribution of amylopectin chain lengths did not explain the difference in grain appearance both for chamber and field experiments. These results strongly suggest that physiological processes linked with the synthesis of 13 kDa prolamin are associated with grain appearance in Japanese highly palatable under high temperature stress.

Satellites must endure the hostile environement during their launching to space via rocket; therefore, they should be exposed to the real launch conditions for ground testing, including all subsystems and components which should be carefully tested. Several solid-state-ceramic batteries have been selected to be evaluated under the launch environment after been evaluated under the space environment, which has shown so far good results. This paper focuses on the physical degradation and the electrical performances of the batteries based on the discharge capacity, the open-circuit voltage, and charge/discharge modes. Batteries have been exposed to shock, then tested under vibration within different frequencies’ levels with sinewave, sine burst, and random. Before and after the test, the physical properties of all batteries have been checked, several cycles of discharge and charge have been performed to check their performances and survivability after the evaluation test. With 95% of capacity, batteries could demonstrate their ability to withstand the launch conditions successfully, they could be able to operate during several cycles after the test, so far, showing no degradation on their performances within the limits. Also, the paper is providing the main requirements and criteria for batteries’ launch ground testing for the small satellite project.

The patient was an 81-year-old woman. At age 73, she developed difficulties in climbing stairs and swallowing, and became unable to open bottles at age 74. She had been walking with a cane since age 76. Accidental chest X-ray findings showed bilateral hilar lymphadenopathy at age 78. Angiotensin converting enzyme (ACE) was elevated. Lymphocyte proliferation was prominent in bronchoalveolar lavage fluid. Sarcoidosis was suspected, but she was followed without treatment due to lack of respiratory symptoms. She became unable to walk without assistance at age 80 and visited our hospital with a complaint of gait disturbance at age 81. Moderate diffuse muscle atrophy in extremities was evident. Muscle weakness of finger flexion and knee extension were remarkable. The muscle involvement pattern was similar to sporadic inclusion body myositis (sIBM). However, radiographically, rectus femoris and semitendinosus muscles are selectively preserved. This radiogaphic finding was consistent with chronic sarcoid myopathy (CSM). We reached a final diagnosis of CSM based on the presence of granulomas in the muscle biopsy specimen, BHL in fluorodeoxyglucose positron-emission tomography, the previous finding of elevated ACE, and bronchoscopy results. In conclusion, CSM is a treatable disease and should thus be differentiated from sIBM. This should not be done solely based on clinical findings, but instead, muscle biopsy should be performed. Moreover, muscle selectivity may be useful in distinguishing between CSM and sIBM.

Edge defined Film Fed Growth (EFG) method with molybdenum (Mo) crucible and die was studied for the growth of several oxide-based single crystals. At first, reactivity between the molybdenum (Mo) metal and oxide melts of Ce:(Lu,Y)2SiO5 (LYSO) Ce:Y2SiO5 (YSO), Ce:Gd3(Al,Ga)5O12 (GAGG), Ce:Lu3Al5O12 (LuAG), Ce:Y3Al5O12 (YAG) was investigated. The experimental and calculation results on the reactivity with Mo show that the aluminum garnets (LuAG, YAG) had the smallest reactivity with Mo metal. On the contrary, GAGG showed the strongest reaction with Mo. Then, the multiple-die grown shaped single crystal growth of Ce:LuAG was demonstrated by EFG method. Single crystal plates up to 320 mm in length were grown successfully. The homogeneity of dopant distribution, Mo contamination, luminescence and scintillation responses of the grown crystals were evaluated.

The succession of faunal composition of a whale-fall community is believed to depend on the progress of decomposition of the whale carcass, which itself is dependent on microbial activity in the sediment around the carcass. This means that the faunal succession could be reflected in the variation of the geochemical environment. In this study, we examined sperm whale carcasses placed in two areas of Japanese waters. In Sagami Bay at a depth of ca. 500 m, dive surveys were performed 2 weeks, 2 months, and 9 months after implantation, when mobile scavengers and Osedax worms were observed. Off Cape Nomamisaki, 12 carcasses were implanted at a depth of 200–300 m, and dive surveys were performed there 17, 29, and 41 months after implantation, and abundant chemosynthesis-based mussels were observed at these times. The recovered sediment cores from beneath and around the carcasses in Sagami Bay showed increases in 15N-enriched ammonium and 34S-depleted sulfide and decreases in 34S-enriched sulfate during the 9 months following implantation, and their stoichiometric relationship suggested that proteinaceous matter from the carcass was decomposed preferentially followed by active bacterial sulfate reduction. In comparison, relatively low ammonium but high sulfide concentrations after 41 months off Cape Nomamisaki suggest that primarily lipids from the almost skeletonized carcasses were decomposed in the sediment during bacterial sulfate reduction. Such variation of decomposed organic substrates and geochemical environments result from interaction with the animals inhabiting in and on the sediments adjacent the whale carcasses. It is possible that evolution of the geochemical environment plays an important role in faunal succession, from the enrichment opportunist to sulfophilic stages.

A novel, anaerobic bacterium, strain MO-SEDIT, was isolated from a methanogenic microbial community, which was originally obtained from marine subsurface sediments collected from off the Shimokita Peninsula of Japan. Cells were Gram-stain-negative, non-motile, non-spore-forming rods, 0.4–1.4 μm long by 0.4–0.6 μm wide. The cells also formed long filaments of up to about 11 μm. The strain grew on amino acids (i.e. valine, leucine, isoleucine, methionine, glycine, phenylalanine, tryptophan, lysine and arginine), pyruvate and melezitose in the presence of yeast extract. Growth was observed at 4–37 °C (optimally at 30 °C), at pH 6.0 and 8.5 (optimally at 7.0–7.5) and in 0–60 g l− 1 NaCl (optimally 20 g NaCl l− 1). The G+C content of the DNA was 32.0 mol%. The polar lipids of strain MO-SEDIT were phosphatidylglycerol, phosphatidyl lipids and unknown lipids. The major cellular fatty acids (>10 % of the total) were C14 : 0, C16 : 1ω9 and C16 : 0 dimethyl aldehyde. Comparative sequence analysis of the 16S rRNA gene showed that strain MO-SEDIT was affiliated with the genus Sedimentibacter within the phylum Firmicutes. It was related most closely to the type strain of Sedimentibacter saalensis (94 % sequence similarity). Based on the phenotypic and genetic characteristics, strain MO-SEDIT is considered to represent a novel species of the genus Sedimentibacter, for which the name Sedimentibacter acidaminivorans sp. nov. is proposed. The type strain is MO-SEDIT ( = JCM 17293T = DSM 24004T).

High-temperature stress during the grain-filling stage reduces grain quality of rice, and this is a serious problem in Japan, especially in the Kyushu region. To solve this problem, various heat-tolerant cultivars have been bred, such as ‘Nikomaru’, ‘Kumasannochikara’, ‘Genkitsukushi’, ‘Sagabiyori’, and ‘Otentosodachi’. When cultivated under high temperature after flowering, these heat-tolerant cultivars had lower percentages of chalky grains than in the heat-sensitive cultivar ‘Hinohikari’. All the heat-tolerant cultivars markedly decreased the nonstructural carbohydrate content in the stem under the high temperature compared to control condition during early grain-filling stage, which is considered to be a common trait of heat tolerance. Notably, ‘Sagabiyori’, ‘Genkitsukushi’, and ‘Nikomaru’ maintained a nucellar epidermis at 17 days after flowering (DAF) under high temperature, whereas the nucellar epidermis disappeared in ‘Hinohikari’. In addition, the expression of AGPS2b, thought to be a rate-limiting enzyme in starch synthesis, in ‘Kumasannochikara’, ‘Otentosodachi’, and ‘Nikomaru’ did not decrease under high temperature, whereas ‘Hinohikari’, ‘Sagabiyori’, and ‘Genkitsukushi’ could not maintain expression of the gene at 17 DAF. Moreover, the expression of Amy3E, a starch-degradation-related gene considered to induce grain chalkiness, in ‘Kumasannochikara’ at 17 DAF was not increased by high temperature. These results suggest that the heat-stress-tolerant cultivars have various mechanisms for dealing with high-temperature stress.

Silicon- and carbon-bridged polythiophenes, dithienosilole (DTS) and cyclopentadithiophene (DTC) homopolymers with 2-ethylhexyl or n-octyl substituents on the bridging atom, were synthesized and their optical and electrochemical properties were investigated. OFET devices containing the present polymer films as the active layers were fabricated to investigate their carrier transport properties. The device based on a DTS polymer with n-octyl substituents showed much higher carrier mobility than that of the DTC polymer with the same substituents.
