In recent years, several nanocarrier-based drug delivery systems, such as polymeric nanoparticles, solid lipid nanoparticles, metallic nanoparticles, liposomes, and others, have been explored to target and treat a wide variety of diseases. Their employment has brought many benefits, not only to human medicine but also to veterinary medicine, albeit at a slower rate. Soon, the use of nanocarriers could revolutionize the animal health sector, and many veterinary therapies will be more effective as a result. The purpose of this review is to offer an overview of the main applications of nanocarriers in the veterinary field, from supplements for animal health and reproduction to nanovaccines and nanotherapies. Among the major pathologies that can affect animals, special attention is given to canine osteosarcoma (OSA): a comparison with human OSA is provided and the main treatment options are reviewed emphasizing the benefits that nanocarriers could bring in the treatment of this widespread disease.

In this study, an overview of the regulations in force in some extra-EU states belonging to different geoeconomic areas is provided, starting from the current EU legislation on cosmetic products. We focused on their legislative frameworks and the location of the relevant regulatory documentation. Furthermore, for each state considered, our analysis examined the notification/authorization processes, approaches to animal testing, and allowed/prohibited ingredient lists, as these aspects are considered to be among the primary restrictions hindering the cosmetic market. It can be observed that many states are working towards standardising their regulations to promote greater international trade. However, it is essential to recognize that different countries belonging to distinct geoeconomic areas may have unique requirements, and harmonization may not necessarily be the best solution. It is crucial to consider different needs and preferences when approaching the global regulation of the cosmetic industry.

In this work, poly(N-isopropylacrylamide) (PNIPAM) based chemically cross-linked hydrogels are used as novel precipitation and stabilization media for solid lipid nanoparticles (SLNs) for the first time. The hydrogels and the hybrid thermoresponsive composite hydrogels with SLNs were characterized by SEM, DSC, DLS and rheometric analysis. The results showed that the SLNs obtained directly in the gel matrix by the newly devised method were well-dispersed and remained stable for one month. A remarkable advantage of this approach is that it yields the thermoresponsive nanocomposite hydrogels in a single step. This approach is a significant advancement in the preparation of hybrid thermoresponsive nanocomposite systems based on smart gels and SLNs for their use in biomedical applications.

Several semisynthetic, low-cardiotoxicity doxorubicin (DOXO) conjugated have been extensively described, considering the risk of cytotoxicity loss against resistant tumor cells, which mainly present drug efflux capacity. Doxorubicin 14-[4-(4-phenyl-5-thioxo-5H-[1,2]dithiol-3-yl)]-benzoate (H2S-DOXO) was synthetized and tested for its ability to overcome drug resistance with good intracellular accumulation. In this paper, we present a formulation study aimed to develop naked and decorated H2S-DOXO-loaded lipid nanoparticles (NPs). NPs prepared by the “cold dilution of microemulsion” method were decorated with hyaluronic acid (HA) to obtain active targeting and characterized for their physicochemical properties, drug entrapment efficiency, long-term stability, and in vitro drug release. Best formulations were tested in vitro on human-sensitive (MCF7) and human/mouse DOXO-resistant (MDA-MDB -231 and JC) breast cancer cells, on human (U-2OS) osteosarcoma cells and DOXO-resistant human/mouse osteosarcoma cells (U-2OS/DX580/K7M2). HA-decoration by HA-cetyltrimethyl ammonium bromide electrostatic interaction on NPs surface was confirmed by Zeta potential and elemental analysis at TEM. NPs had mean diameters lower than 300 nm, 70% H2S-DOXO entrapment efficiency, and were stable for almost 28 days. HA-decorated NPs accumulated H2S-DOXO in Pgp-expressing cells reducing cell viability. HA-decorated NPs result in the best formulation to increase the inter-cellular H2S-DOXO delivery and kill resistant cells, and therefore, as a future perspective, they will be taken into account for further in vivo experiments on tumor animal model.

Osteosarcoma (OSA) is the most frequently diagnosed primary malignant bone tumor in humans and dogs. In both species, standard chemotherapy can be limited by multidrug resistance of neoplastic cells, which prevents intracellular accumulation of cytotoxic drugs, resulting in chemotherapy failure. In this study, a lipophilic ester of doxorubicin (C12DOXO) was loaded into nanoparticles (NPs) using the “cold microemulsion dilution” method. The resulting NPs were then coated with calcium phosphate (CaP) in two different ways to have calcium or phosphate ions externally exposed on the surface. These systems were characterized by determining mean diameter, zeta potential, and drug entrapment efficiency; afterward, they were tested on human and canine OSA cells to study the role that the coating might play in increasing both drug uptake into tumor cells and cytotoxicity. Mean diameter of the developed NPs was in the 200–300 nm range, zeta potential depended on the coating type, and C12DOXO entrapment efficiency was in the 60–75% range. Results of studies on human and canine OSA cells were very similar and showed an increase in drug uptake and cytotoxicity for CaP-coated NPs, especially when calcium ions were externally exposed. Therefore, applications in both human and veterinary medicine can be planned in the near future.

Osteomyelitis is a bone marrow infection which generally involves cortical plates and which may occur after bone trauma, orthopedic/maxillofacial surgery or after vascular insufficiency episodes. It mostly affects people from the Third World Countries, the elderly and patients affected by systemic diseases e.g. autoimmune disorders, AIDS, osteoporosis and microvascular disease. The highest percentage of osteomyelitis cases (almost 75%) is caused by Staphylococcus spp., and in particular by Staphylococcus aureus (more than 50%). The ideal classification and the diagnosis of osteomyelitis are two important tools which help the physicians to choose the best therapeutic strategies. Currently, common therapies provide an extensive debridement in association with intravenous administration of antibiotics (penicillin or clindamycin, vancomycin and fluoroquinolones among all for resistant microorganisms), to avoid the formation of sequestra. However, conventional therapeutic approach involves several drawbacks like low concentration of antibiotics in the infected site, leading to resistance and adverse effects due to the intravenous administration. For these reasons, in the last years several studies have been focused on the development of drug delivery systems such as cements, beads, scaffolds and ceramics made of hydroxyapatite (HA), calcium phosphate (CaP) and β-tricalcium phosphate (β-TCP) which demonstrated to be biocompatible, poorly toxic and capable to allow osteointegration and a prolonged drug release. The aim of this review is to provide a focus on current therapies and latest developed drug delivery systems with particular attention on those based on CaP and its derivatives, hoping that this work could allow further direction in the field of osteomyelitis.

Some approaches to enhance the entrapment efficiency within nanoparticulate systems, chiefly of hydrophilic molecules, developed mainly by the Turin University Pharmaceutical Technology group, are discussed. Several approaches have been developed: the first entails associating anticancer hydrophilic molecules to liposomes, developing lipophilic prodrugs of these molecules, and encapsulating them in liposomes. The transformation of hydrophilic drugs into lipophilic prodrugs can also overcome problems of poor entrapment efficiency and rapid release from polymer nanocarriers. Examples are nanospheres and nanocapsules produced from various PEGylated poly(alkylcyanoacrylate) copolymers. Strategies have also been developed to enhance hydrophilic drug entrapment in solid lipid nanoparticles (SLN). Hydrophobic ion pairing was designed to enable various antitumor drugs to be entrapped in SLN, produced by the coacervation method. Another technique comprises the covalent linkage of antitumoral or antiviral drugs to a squalenoyl-derived chain, affording bioconjugates that self-assemble as stable nanoparticles. A further development comprises mesoporous silica nanoparticles with immobilized hydrophilic antioxidants, for topical applications: they were complexed with hydrophilic antioxidants (Trolox ® or rutin). Polymer-shelled and perfluoropentane-cored nanobubbles have also been designed, as versatile multifunctional carriers for the delivery of gases, drugs, and genes; the size range is below 500 nm, with shell thickness in the 30–50 nm range.

Vaginal infections such as bacterial vaginosis (BV), chlamydia, gonorrhea, genital herpes, candidiasis, and trichomoniasis affect millions of women each year. They are caused by an overgrowth of microorganisms, generally sexually transmitted, which in turn can be favored by alterations in the vaginal flora. Conventional treatments of these infections consist in systemic or local antimicrobial therapies. However, in the attempt to reduce adverse effects and to contrast microbial resistance and infection recurrences, many efforts have been devoted to the development of vaginal systems for the local delivery of antimicrobials. Several topical dosage forms such as aerosols, lotions, suppositories, tablets, gels, and creams have been proposed, although they are sometimes ineffective due to their poor penetration and rapid removal from the vaginal canal. For these reasons, the development of innovative drug delivery systems, able to remain in situ and release active agents for a prolonged period, is becoming more and more important. Among all, nanosystems such as liposomes, nanoparticles (NPs), and micelles with tunable surface properties, but also thermogelling nanocomposites, could be exploited to improve local drug delivery, biodistribution, retention, and uptake in vulvovaginal tissues. The aim of this review is to provide a survey of the variety of nanoplatforms developed for the vaginal delivery of antimicrobial agents. A concise summary of the most common vaginal infections and of the conventional therapies is also provided.

Floxuridine is a very effective drug with high potency in the treatment of various tumors but its utility is limited by its low efficiency of cellular uptake. In order to improve the floxuridine efficiency of cellular uptake, lipophilic prodrug of floxuridine (3',5'-distearoyl-5-fluoro-2'-deoxyuridine) was synthetized and loaded into behenic acid nanoparticles produced by fatty acid coacervation technique. Generally, spherical shaped SLN with mean diameters below 300 nm were obtained. Distearoyl-floxuridine was loaded in SLN with high entrapment efficiency (from 70.8 to 82.8%). In Vitro cytotoxicity studies on different human cancer cell lines (M14, HT-29 and MDA-MB231) were performed in order to test the ability of distearoyl-floxuridine-SLN to inhibit the cancer cell growth. In MTT test distearoyl floxuridine SLN showed a greater efficacy than floxuridine on all cancer cell lines revealing an efficiency about 100 times higher. Also clonogenic assay showed a higher cytotoxicity of distearoyl-floxuridine-SLN compared to floxuridine but the difference between the formulations was only about 10 times. In conclusion, SLN proved to be a promising vehicle to increase the floxuridine efficacy in cancer therapy.

Despite eye drops being widespread s for drug therapy of the anterior segment of the eye, they are also characterized by low drug bioavailability, due to rapid washout by tearing and nasolachrymal drainage. Moreover, effective drug delivery to the posterior segment of the eye is very difficult and alternative administration routes (periocular and intravitreal) are usually employed, because the blood retinal barrier is the major obstacle to systemic drug delivery. Therapeutic efficiency, as well as compliance and safety of ocular drugs, administered either to the anterior, or posterior segment of the eye, can benefit from a nanotechnological approach. Nanocarriers based on lipids are among the most biocompatible; their versatility of production and ease of functionalization allow the entrapment of many active molecules. In particular, mucoadhesion, with consequent increase of retention at the cornea, as well as permeation enhancement, are the most important challenges for topical lipid nanoparticle delivery to the anterior segment of the eye.

Multidrug resistance (MDR) is a critical hindrance to the success of cancer chemotherapy. The main thing responsible for MDR phenotypes are plasma-membranes associated with adenosine triphosphate (ATP) Binding Cassette (ABC) drug efflux transporters, such as the P-glycoprotein (Pgp) transporter that has the broadest spectrum of substrates. Curcumin (CURC) is a Pgp inhibitor, but it is poorly soluble and bioavailable. To overcome these limitations, we validated the efficacy and safety of CURC, loaded in biocompatible solid lipid nanoparticles (SLNs), with or without chitosan coating, with the goal of increasing the stability, homogeneous water dispersibility, and cellular uptake. Both CURC-loaded SLNs were 5&ndash, 10-fold more effective than free CURC in increasing the intracellular retention and toxicity of doxorubicin in Pgp-expressing triple negative breast cancer (TNBC). The effect was due to the decrease of intracellular reactive oxygen species, consequent inhibition of the Akt/IKK&alpha, &beta, /NF-kB axis, and reduced transcriptional activation of the Pgp promoter by p65/p50 NF-kB. CURC-loaded SLNs also effectively rescued the sensitivity to doxorubicin against drug-resistant TNBC tumors, without signs of systemic toxicity. These results suggest that the combination therapy, based on CURC-loaded SLNs and doxorubicin, is an effective and safe approach to overcome the Pgp-mediated chemoresistance in TNBC.

Central nervous system disorders are already prevalent and steadily increasing among populations worldwide. However, most of the pharmaceuticals present on world markets are ineffective in treating cerebral diseases, because they cannot effectively cross the blood brain barrier (BBB). Solid lipid nanoparticles (SLN) are nanospheres made from biocompatible solid lipids, with unique advantages among drug carriers: they can be used as vehicles to cross the BBB. This review examines the main aspects surrounding brain delivery with SLN, and illustrates the principal mechanisms used to enhance brain uptake of the delivered drug.

Nanoparticles are rapidly developing as drug carriers because of their size-dependent properties. Lipid nanoparticles (LNPs) are widely employed in drug delivery because of the biocompatibility of the lipid matrix.Many different types of LNPs have been engineered in the last 20 years, the most important being solid lipid nanoparticles (SLNs), nanostrucured lipid carriers (NLCs), lipid-drug conjugates (LDCs) and lipid nanocapsules (LNCs). This review gives an overview of LNPs, including their physico-chemical properties and pharmacological uses. Moreover, it highlights the most important innovations in the preparation techniques of LNPs, aimed to encapsulate different molecules within the lipid matrix. Finally, it gives a short perspective on the challenges of drug delivery, which are a potential field of application for LNPs: cancer therapy, overcoming the blood-brain barrier and gene and protein delivery.LNPs are a safe and versatile vehicles for drug and active delivery, suitable for different administration routes. New technologies have been developed for LNP preparation and studies are currently underway in order to obtain the encapsulation of different drugs and to deliver the active molecule to the site of action.

Cyclosporine-loaded solid lipid nanoparticles were prepared according to the coacervation method: various stabilizers were used in order to obtain nanoparticles with different surface charge. These formulations were tested on rabbit corneas. Corneal irritation was measured according to opacity and permeability test; the interaction of labeled solid lipid nanoparticles with the cornea was studied by fluorescence microscopy; drug accumulation and permeation through the cornea were studied after labeling cyclosporine with a fluorescent probe. Nanoparticles were not irritant to the cornea, according to the opacity and permeability test. Cationic (chitosan coated) solid lipid nanoparticles enhanced drug accumulation and permeation through rabbit cornea, compared to anionic and non-ionic solid lipid nanoparticles and other reference formulations, probably due to the interaction of chitosan coated nanoparticles with corneal epithelial cells.

Aim: Recently, ‘coacervation’ has been proposed as a new method to prepare fatty acid solid lipid nanoparticles (SLNs). The aim of this work was to encapsulate methotrexate, a hydrophilic anticancer drug, within SLNs obtained by coacervation, through hydrophobic ion pairing and to evaluate the potential efficacy in in vitro and in vivo breast tumor models of drug-loaded nanoparticles. Materials & Methods: Methotrexate-loaded SLN efficacy was evaluated in vitro towards MCF-7 and Mat B-III cell lines (human and murine breast tumor cell lines). Pharmacokinetics of drug-loaded nanoparticles was evaluated in male Wistar rats and biodistribution in a breast tumor model (Mat B-III) in female Fisher rats. Results: Drug- loaded SLNs showed an increased cytotoxicity towards MCF-7 and Mat B-III cell lines compared with free drug. After intravenous administration, drug plasmatic concentration was increased and a major drug accumulation within neoplastic tissue was shown when the drug was loaded in SLNs, compared with drug solution alone. Encapsulation of the drug within nanoparticles also increased its oral uptake after duodenal administration. Conclusion: SLNs are promising vehicles for the delivery of methotrexate, since an increase of efficacy in vitro and a preferential accumulation in breast cancer in vivo were shown. Curcumin (CU) loaded solid lipid nanoparticles (SLNs) of fatty acids (FA) were prepared with a coacervation technique based on FA precipitation from their sodium salt micelles in the presence of polymeric non-ionic surfactants. Myristic, palmitic, stearic, and behenic acids, and different polymers with various molecular weights and hydrolysis grades were employed as lipid matrixes and stabilisers, respectively. Generally, spherical-shaped nanoparticles with mean diameters below 500 nm were obtained, and using only middle-high hydrolysis, grade-polymer SLNs with diameters lower than 300 nm were produced. CU encapsulation efficiency was in the range 28-81% and highly influenced by both FA and polymer type. Chitosan hydrochloride was added to FA SLN formulations to produce bioadhesive, positively charged nanoparticles. A CU-chitosan complex formation could be hypothesised by DSC analysis, UV-vis spectra and chitosan surface tension determination. A preliminary study on HCT-116 colon cancer cells was developed to evaluate the influence of CU-loaded FA SLNs on cell viability.

Insulin-loaded solid lipid nanoparticles (SLN) were prepared according to a solvent dilution method from O/W emulsions using isovaleric acid as organic phase. Insulin was derivatized with fluorescein isothyocianate (FITC) obtaining a fluorescent marker to be used in in vivo experiments. FITC-insulin and native insulin–loaded SLN were quite similar with regard to their mean sizes and encapsulation efficiency. SLN intestinal uptake was then investigated administering FITC-insulin loaded SLN on healthy male Wistar rats. Significant drug accumulation within intestinal lymphatic system was recovered, but the immune system seems to play an important role in SLN degradation: further studies are necessary to improve the results on blood glucose level.

Chitosan is an excipient which has been studied thoroughly in research works thanks to its positive characteristics such as muco-adhesiveness and ability to open epithelial-tight-junctions. In this article, lipophilic stearoyl chitosan (ST-CS) was synthetized in order to anchor this polymer to lipid nanoparticles and prepare ST-CS-coated nanoparticles (ST-CS-NP) using the microemulsion cold dilution technique. Curcumin (CURC) was used as model drug. CURC-ST-CS-NP were characterized by dimensional analysis, zeta potential, drug entrapment, drug release, tested in vitro on Human Umbilical Vein Endothelial Cell (HUVEC) cells to study its cytotoxicity and on human pancreatic cancer cells (PANC-1) to determine inhibition ability, tested in rats to determine CURC blood profiles and biodistribution. CURC-ST-CS-NP had mean diameters in the range 200&ndash, 400 nm and CURC entrapment up to 73%. These systems did not show cytotoxicity on HUVEC cells at all tested dilutions and revealed to be more effective than free CURC solution on PANC-1 cells at 5 and 10 &micro, M CURC. Blood profile studies evidenced as CURC entrapment in NP prolonged the permanence of drug in the systemic circulation compared to CURC solution due to a certain stealth property of NP, probably attributable to hydrophilic chitosan coating. Biodistribution studies showed a smaller CURC concentration in RES organs when CURC-ST-CS-NP were administered.

Stearic acid solid lipid nanoparticles (SLN) were prepared according to the coacervation technique, consisting in fatty acid precipitation of alkaline salt from a solution. SLN can encapsulate Amphotericin B with high efficiency in its monomeric form. SLN markedly increase in vitro drug toxicity towards Saccharomyces cerevisiae (taken as model yeast), expressed as potassium leakage. Yeast susceptibility towards Amphotericin B is increased when the drug is loaded in nanoparticles compared to the free drug, even if inhibition is limited by drug encapsulation within lipid matrix.

Coacervation technique, a solvent-free, feasible and versatile method, based on a phase transformation from soap micellar solution into fatty acid solid particles by acid addition, was used to prepare cisplatin loaded solid lipid nanoparticles (SLN) of stearic acid. Different polymers were tested to stabilise SLN suspensions. Solubilisation of cisplatin, a hydrophilic antitumor agent, within sodium stearate micelles was possible by the formation of a hydrophobic ion-pair with sodium dioctylsulfosuccinate. Several SLN were produced, whose particle size were in the 275–525 nm range. Cisplatin encapsulation efficiency up to 90 % was obtained, depending on both stearic acid concentration and stabilisers type and concentration. The in vitro cisplatin release showed a burst effect of about 10–20 %, corresponding to the non-encapsulated drug, and then a complete drug release was reached after 24 h.

The purpose of this study was to improve the stability of dithranol, an effective drug for topical treatment of psoriasis. The influence of several formulations (microemulsions, O/W emulsion, gel emulsion, and gel) on the photodegradation kinetics of dithranol was investigated. The photodegradation rate was found to be related with the initial drug concentration and the nature of the vehicle. Solid lipid nanoparticles (SLN) were prepared by solvent injection technique to investigate whether the inclusion in the lipid matrix could increase the stability of the drug. Physicochemical characterization of the particles by optical microscopy, photon correlation spectroscopy (PCS) and differential scanning calorimetry (DSC) revealed that solvent injection is a suitable approach for dithranol-loaded SLN preparation. The obtained particle sizes were between 230 and 270 nm; up to 92% of drug was entrapped in the SLN. Solid lipid nanoparticles (SLN) made of different triglycerides (TG) in the presence and in the absence of various modified α- and γ-cyclodextrins (CD) were prepared by the solvent injection technique. A new synthesis of lipophilic derivatives of γ- CD was developed in this work. Curcumin (CU), a natural polyphenol with antitumor, antioxidant and anti-inflammatory properties, was used as model drug. SLNs mean sizes were in the 250–800 nm range and afforded CU entrapment efficiency in the 12–85% range. The presence of CD derivatives with almost the same chain length of TG induced an improvement of nanoparticle characteristics decreasing mean size values and increasing CU entrapment efficiency. A significant reduction in CU photodegradation was noted only when the drug was vehicled in tristearin-SLN, which became less pronounced in the presence of CD-derivatives, determining a loss in photoprotection. The hydrolytic stability of curcumin was highly improved by drug loading in tristearin-SLN, and only slightly by loading it in tricaprin-SLN, and this seemed not to be influenced by the presence of CD derivatives. Skin uptake studies revealed an increase in CU skin accumulation when CU was loaded in SLN obtained with all CD derivatives, particularly with most lipophilic one.

Dry emulsions were investigated for their possible use in cosmetic and pharmaceutical fields and an extract of Vitis vinifera leaves was obtained and characterized in terms of quality and quantitative levels, which can be introduced in a selected dry emulsion formulation. Several pre-dry emulsions were prepared, six of which were chosen as a consequence of their satisfying physico-chemical and over-time stability characteristics. They were successively freeze-dried and reconstituted after optimising the reconstitution modalities and characterized by optical microscopy, granulometric and thermal analysis, FTIR analysis, rheology, and texture studies. The best formulation was chosen to introduce Vitis vinifera dry extract prepared by percolation: a fair increase in anthocians stability was noted over time in dry emulsions as compared with classic emulsions.

The photochemical degradation of the sunscreen p‐octyl‐methoxy‐cinnamate (OMC) was studied in different media: Finsolv TN (a fluid ester), O/W emulsion and gel‐emulsion. OMC photolysis was performed using an UVB lamp and the degradation kinetic was studied in presence of various initial concentration of the active. The study was performed comparing the naked to the SLNs form of the active and the experiments were monitored with HPLC/UV‐VIS apparatus. Results suggest that lower is the concentration of the UVB sunscreen in the formulations more pronounced is its photodegradation rate. In all the cases the inglobation of OMC into SLNs leads to an over additive UV‐blocking effect.

Elastic positively-charged liposomes of soya lecithin were prepared using sucrose monopalmitate as deforming agent, and l-alanine benzyl ester or stearylamine as cationic charge-inducing agent. The positive charge of the liposomes was confirmed by zeta potential measurements and their elasticity by filtration through microporous filter. Positively-charged liposomes had a higher acyclovir entrapment efficiency than negatively-charged liposomes. Elastic positively-charged liposomes containing acyclovir, applied non-occlusively to pig-ear skin, determined a skin flux similar to that of non-elastic positively-charged liposomes, prepared using soya lecithin, cholesterol and stearylamine, but higher than that of non-elastic negatively-charged liposomes or control suspension. Skin deposition of acyclovir from elastic positively-charged liposomes was higher (up to 600 nmol/cm 2 ) than that from non-elastic positively- or negatively-charged liposomes (respectively 344 nmol/cm 2 , 237 nmol/cm 2 ) and than that from elastic negatively-charged liposomes (435 nmol/cm 2 ). The enhanced ACV accumulation within the skin obtained with elastic positively-charged liposomes might help to optimize drug targeting, creating new opportunities for well-controlled, modern, topical application of ACV in the treatment of Herpes simplex.

Glioblastoma, the most common primary brain tumor in adults, has an inauspicious prognosis, given that overcoming the blood-brain barrier is the major obstacle to the pharmacological treatment of brain tumors. As neoangiogenesis plays a key role in glioblastoma growth, the US Food and Drug Administration approved bevacizumab (BVZ), an antivascular endothelial growth factor antibody for the treatment of recurrent glioblastoma in patients whose the initial therapy has failed. In this experimental work, BVZ was entrapped in solid lipid nanoparticles (SLNs) prepared by the fatty-acid coacervation technique, thanks to the formation of a hydrophobic ion pair. BVZ activity, which was evaluated by means of four different in vitro tests on HUVEC cells, increased by 100- to 200-fold when delivered in SLNs. Moreover, SLNs can enhance the permeation of fluorescently labelled BVZ through an hCMEC/D3 cell monolayer-an in vitro model of the blood brain barrier. These results are promising, even if further in vivo studies are required to evaluate the effective potential of BVZ-loaded SLNs in glioblastoma treatment.

Several deformable liposomes were formulated using hydrogenated soya lecithin and sodium cholate, polisorbate 80, dipotassium glycyrrhizinate, or saccharose monopalmitate. The lipid:surfactant w/w ratio necessary to obtain elastic vesicles depended on the O/W surfactant and ranged from 4∶1 to 20∶1. The liposomes obtained were able to entrap α‐tocopherol and tocopheryl acetate up to 0.17% w/w. Elastic liposomes, whose deformability was confirmed by filtration through microporous filters and differential scanning calorimetry measurements, when non‐occlusively applied on pig ear skin, determined negligible skin fluxes of α‐tocopherol, while skin deposition significantly increased compared with reference solutions or normal liposomes. Moreover, the entrapment of the vitamin either in elastic or in normal liposomes increased its photo‐stability under UVB irradiation.

W/O/W multiple emulsions with ethylene oxide free emulsifiers, compatible with the skin, were prepared. They were fresh and easy to spread on the skin. The emulsion showed a shear thinning flux. Their multiplicity was confirmed with optical microscope analysis. FeSO4 and K3Fe(CN)6, when introduced separately in the inner and outer aqueous phases of the emulsions, did not give coloration, but when introduced together in the same aqueous phase, give a blue color due to their reaction. This experiment confirmed the multiplicity of these systems. The external aqueous phase of the emulsions was confirmed by Brilliant Blue up take from one of the multiple emulsions prepared higher than that from its W/O primary emulsion. The stability of the systems was confirmed by tests of breaking to centrifuge, of storage at 40°C and by freeze‐thaw cycles.

Several liposome dispersions, gel-emulsions and o/w microemulsions were developed using soy lecithin and non-ionic, non-ethoxylated, glucose-and sucrose-derived, well-tolerated and skin compatible emulsifiers. The systems prepared were characterised by determining their mean sizes, which in the case of liposomes markedly decreased in the presence of sucrose esters (from 450 to 350 nm) and, in the case of microemulsion were about 30 nm. α-tocopherol (vitamin E) was then added to the formulations and its stability against photodegradation under UVB light was studied. The best protective effect was noted when α-tocopherol was dissolved in the inner phase of o/w microemulsion in the presence of ascorbic acid in the outer phase, as no decrease in vitamin E content occurred after 9 h of irradiation. Liposome dispersions, particularly when formulated with sucrose monoesters as auxiliary surfactants, and gel emulsions also significantly increased α-tocopherol photostability compared to ethanol or surfactant solutions. Skin deposition of α-tocopherol within intact pig ear skin from liposomes, gel emulsions and microemulsions were assessed and compared with values for ethanol solutions. Only gel emulsions and microemulsions gave rise to significant skin depositions of α-tocopherol.

o/w emulsions with zinc oxide and salicylic acid were prepared at a pH between 4.0 and 5.2 in order to determine the best pH at which both molecules are simultaneously present in the system. The emulsion's structure and the presence of zinc oxide crystals were investigated by optical microscopy. Zinc oxide caused a slight increase in pH of the emulsions over time. The emulsion that had the most stable pH was that at pH 4.6. Salicylic acid diffusion from these systems, studied as a function of pH and the amount of zinc oxide crystals, was maximal at pH 4.6 and was found to be unrelated to the apparent partition coefficient or to the un-dissociated molar fraction of salicylic acid. Zinc oxide decreased the diffusion of salicylic acid from all emulsions.

Hydroxypropyl methylcellulose (HPMC) was used to increase the viscosity of aqueous solutions of sodium lauroyl glutamate (SLG) and potassium lauroyl hydrolyzed collagen (KLHC), as viscosity modifiers commonly used in toiletry products fail to thicken in the presence of these surfactants. A surfactant–polymer link was indicated by a change of surface tension as a function of surfactant and polymer concentration and by viscosity trends of surfactant–polymer mixtures around critical micellar concentration (CMC). A positive surfactant–polymer interaction was suggested by an experimental design of Doehlert. Such an interaction affects the viscosity of the system and the solubility of two lipophilic dyes in the micellar core of the surfactant–polymer complexes. Hydroxypropyl methylcellulose prevents SLG precipitation by lowering its Kraft point.

The aim of this study was to evaluate the possibility of using liposomes for skin delivery of dipotassium glycyrrhizinate (KG), an anti-inflammatory agent employed in treating acute and chronic dermatitis, and of formulating such liposomes in an oil-in-water emulsion (O/W). KG had emulsifying properties and the possibility of producing elastic liposomes was verified. Liposomes containing soya lecithin (PC) or hydrogenated soya lecithin (HPC) mixed with KG in w/w ratios of 2:1, 4:1 or 8:1 were prepared by the solvent evaporation method and then passed through a high pressure homogeniser. Liposome size and entrapment efficiency were determined and the interaction between KG and HPC was investigated using differential scanning calorimetry (DSC). Transepidermal permeation through intact pig skin and skin deposition of KG from liposomes and O/W emulsion containing liposomes were assessed and compared with values for aqueous control solutions. No marked differences were observed between PC and HPC liposomes. Liposome sizes ranged from 90 to 120 nm. Entrapment efficiency depended on the lipid:KG ratio; the maximum efficiency was obtained at 4:1 w/w. KG interacted with liposomes disrupting and fluidising the lipid bilayer, forming elastic liposomes able to penetrate through membrane pores of diameter much smaller than their own diameter. The liposome structure was maintained when dispersed in an O/W emulsion. The skin fluxes were less than the HPLC detection limit for all systems, while skin deposition increased 4.5-fold compared with aqueous solutions when KG was formulated in liposomes.

Vitamin A and vitamin A palmitate photostability were tested in different media. Ethanol and octyl octanoate solutions of these two vitamins, as such and with the addition of sunscreens (3,4 methylbenzilidencanfora, butyl methoxy dibenzoylmethane and octyl methoxycinnamate) or beta-carotene and butylated hydroxy toluene, were analysed spectrophotometrically after UVB or UVA irradiation. An O/W fluid emulsion with 0.5% w/w of retinyl palmitate, with and without butylated hydroxy toluene, was prepared. The oil containing the vitamin was extracted with HCl and aluminium sulfate and analysed spectrophotometrically after UVB or UVA irradiation. The fluid emulsion containing retinyl palmitate with and without butylated hydroxy toluene was stored at different temperatures and analysed every week spectrophotometrically for a month. Of the sunscreens tested butyl methoxy dibenzoylmethane showed the strongest protective action towards vitamin A and vitamin A palmitate, whereas beta-carotene did not protect either vitamin. Butylated hydroxy toluene inhibited the photodegradation of both vitamins dissolved in octyl octanoate, suggesting that oxygen may be involved in their degradation. O/W emulsion promoted slightly the degradation of vitamin A ester. Butylated hydroxy toluene protected retinyl palmitate from degradation induced by light and heat.

General ingredients include solid lipid(s), surfactant(s) and water. The term lipid is used here in a broad sense and includes triglycerides (e.g. tristearin), partial glycerides, fatty acids (e.g. stearic acid), steroids (e.g. cholesterol) and waxes (e.g. cetyl palmitate). All classes of surfac‐ tants (with respect to charge and molecular weight) have been used to stabilize the lipid dispersion [2].

Paclitaxel loaded solid lipid nanoparticles (SLN) of behenic acid were prepared with the coacervation technique. Generally, spherical shaped SLN with mean diameters in the range 300–600 nm were obtained. The introduction of charged molecules, such as stearylamine and glycol chitosan into the formulation allowed to obtain positive SLN with Zeta potential in the 8-20 mV range and encapsulation efficiency in the 25–90% range.Blood–brain barrier (BBB) permeability, tested in vitro through hCMEC/D3 cells monolayer, showed a significantly increase in the permeation of Coumarin-6, used as model drug, when vehicled in SLN. Positive-charged SLN do not seem to enhance permeation although stearylamine-positive SLN resulted the best permeable formulation after 24 h.Cytotoxicity studies on NO3 glioblastoma cell line demonstrated the maintenance of cytotoxic activity of all paclitaxel-loaded SLN that was always unmodified or greater compared with free drug. No difference in cytotoxicity was noted between neutral and charged SLN.Co-culture experiments with hCMEC/D3 and different glioblastoma cells evidenced that, when delivered in SLN, paclitaxel increased its cytotoxicity towards glioblastoma cells.

Several water-in-oil in water multiple emulsions carrying calcium thioglycolate in the inner aqueous phase were prepared as models for depilatory products. Rheology steady-state flow experiments revealed the multiple systems to be adequately stable after 3-month storage. The effective multiplicity of the emulsions was assessed by optical microscopy and confirmed by thioglycolic acid release experiments. The stability of thioglycolic acid against oxidative degradation at 25°C was far higher in the W/O/W emulsion than in an O/W reference emulsion, while at 45°C the degradation rate was quite similar in both systems.

Some inorganic pigments are used as ingredients for sunscreens as they can absorb, reflect or scatter UV radiation. Titanium dioxide and zinc oxide, which are used for this purpose, are semiconductor oxides and show photocatalytic activity. As a consequence, they can promote transformation of organic molecules upon absorption of radiation. This work studies the activity of some of these pigments, provided by cosmetic producers, towards phenol, chosen as a model aromatic molecule. An attempt is made to explain the different activity of the various pigments, coated or uncoated, towards phenol.

The ability of three matrices to model the barrier properties of the lipid domain of stratum corneum (SC) against permeation of seven steroids was studied. Model matrices were water and oleic acid/oleate; a mixture of unsaturated and saturated fatty acids/soap; or a more complex matrix also containing phospholipids, sphingolipids, cholesterol and ceramides. Permeability coefficients (K.) were similar in the three models, supporting the hypothesis that the barrier to steroid permeation is determined by the structural organization of the lipids, not by the chemical structure of individual substances. Parabolic relationships were found between K values and octanol/water partition coefficients (Poct) of the steroids, with an optimum permeability at log Poct, of 3·0. All three models showed good resistance to permeability by steroids. Several topical products contain nanometric TiO2 (nano-TiO2), which is a useful and safe component that absorbs UV light and does not cross skin barrier. However, nano-TiO2 may impregnate the first layer of the skin (stratum corneum, SC) and generate free radicals, even under low UV irradiation. These properties, largely dependent on TiO2 surface chemistry, may modulate the transdermal drug permeation. To investigate how TiO2 surface properties affect drug permeation, amphotericin in two different media, in the presence of three differently coated samples, was applied on skin and the flux measured. The naked, but not the coated, nano-TiO2 showed enhancer property, with a fourfold increase of the drug flux. Only the positively-charged, naked TiO2 strongly adhered to and altered the SC structure. The oxidative potential towards formate anion and linoleic acid was assessed and a molecular mechanism to elucidate increased skin permeability proposed. To enhance the drug permeation, both a surface charge-driven adhesion and an oxidative disorganization of the SC lipids are required. By modulating TiO2 surface charge (coating) and its oxidative potential (crystalline phase), the enhancer effect of nano-TiO2 may be tuned and turned up or down when transdermal penetration of drug has to be favored or impaired.

Solid lipid nanoparticles (SLN) of fatty acids (FAs) were prepared with a new, solvent-free technique based on FAs precipitation from their sodium salt micelles in the presence of polymeric non-ionic surfactants: this technique was called 'coacervation'. Myristic, palmitic, stearic, arachidic and behenic acid were employed as lipid matrixes. Spherical shaped nanoparticles with mean diameters ranging from 250 to approximately 500 nm were obtained. Different aqueous acidifying solutions were used to precipitate various FAs from their sodium salt micellar solution. Good encapsulation efficiency of Nile Red, a lipophilic model dye, in stearic acid nanoparticles was obtained. The coacervation method seems to be a potentially suitable technique to prepare close to monodisperse nanoparticles for drug delivery purposes.

A method to produce solid lipid nanoparticles (SLN) from W/O/W multiple emulsions was developed applying the solvent-in-water emulsion-diffusion technique. Insulin was chosen as hydrophilic peptide drug to be dissolved in the acidic inner aqueous phase of multiple emulsions and to be consequently carried in SLN. Several partially water-miscible solvents with low toxicity were screened in order to optimize emulsions and SLN composition, after assessing that insulin did not undergo any chemical modification in the presence of the different solvents and under the production process conditions. SLN of spherical shape and with mean diameters in the 600-1200 nm range were obtained by simple water dilution of the W/O/W emulsion. Best results, in terms of SLN mean diameter and encapsulation efficiencies, were obtained using glyceryl monostearate as lipid matrix, butyl lactate as a solvent, and soy lecithin and Pluronic F68 as surfactants. Encapsulation efficiencies up to 40% of the loaded amount were obtained, owing to the actual multiplicity of the system; the use of multiple emulsion-derived SLN can be considered a useful strategy to encapsulate a hydrophilic drug in a lipid matrix.

Insulin-loaded solid lipid nanoparticles (SLN), obtained by the solvent-in-water emulsion-diffusion technique, were produced using isovaleric acid (IVA) as organic phase, glyceryl mono-stearate (GMS) as lipid, soy lecithin and sodium taurodeoxycholate (TDC) as emulsifiers. IVA, a partially water-miscible solvent with low toxicity, was used to dissolve both insulin and lipids. SLN of spherical shape were obtained by simple water dilution of the O/W emulsion. Analysis of SLN content after processing showed interesting encapsulation efficiency with respect to therapeutic doses; moreover, insulin did not undergo any chemical modification within the nanoparticles and most of it remained stable after incubation of the SLN with trypsin solution. The biological activity of insulin, i.e. the ability to decrease glycemia in rats, was not negatively influenced by the SLN production process, as after subcutaneous administration of insulin extracted from SLN to animals, the blood glucose levels were quite similar to those obtained after administration of a conventional insulin suspension. Consequently, SLN seem to have interesting possibilities as delivery systems for oral administration of insulin.

Hemp-seed oil has several positive effects on the skin: thanks to its unsaturated fatty acid (PUFA) content it alleviates skin problems such as dryness and those related to the aging process. We present a comparative study of hemp-seed and olive oils, determining some physicochemical indices and evaluating their stability against oxidation. The peroxide value of hemp-seed oil was below 20, the threshold limit for edible oils. Hemp-seed oil was less stable against peroxidation than olive oil, but MDA and MONO assays showed its stability to be above expectations. The chlorophyll contained in extra virgin olive oil had a higher photostability than that contained in hemp-seed oil, possibly due to the larger amount of antioxidant in the olive oil. A certain amount of Vitamin E was found in hemp-seed oil. Since quality analyses indicated that hemp-seed oil is relatively stable, emulsions were prepared with the two oils, and their stability and rheological characteristics were tested. Some of the resulting gel-emulsions were suitable for spraying on the skin.

Vitamin A palmitate photostability in relation to UVA and UVB was tested in hydroxy ethyl cellulose hydrogels at pH 4.0, 5.6, 7.0, and 8.0, alone and with the addition of sunscreens (3,4-methylbenzilidencamphor or butyl methoxy dibenzoylmethane) or an antioxidant (butylated hydroxy toluene). The photostability of vitamin A palmitate was also tested in encapsulated systems (Tagravit A1 microcapsules, Lipotec liposomes, phosphatidylcholine liposomes, and Lipotec nanocapsules) dispersed in gels at pH 5.6 and 7.0. The stability of retinyl palmitate over time in hydroxy ethyl cellulose hydrogels at pH 5.6 and 7.0 (stored one month at 25 degrees C or 40 degrees C), alone or with butylated hydroxy toluene, was also tested. The stability of retinyl palmitate over time in encapsulated systems, dispersed in gels at pH 5.6 and 7.0, was also studied. O/W emulsions were also prepared to compare the stability of vitamin A palmitate introduced in a lipophilic/hydrophilic medium (O/W emulsions) and a hydrophilic medium (hydrogels). HPLC analysis showed that encapsulated systems such as Lipotec nanocapsules, Tagravit A1 microcapsules, phosphatidylcholine liposomes, and Lipotec liposomes protect the vitamin A ester over time from hydrolysis and from oxidation to retinaldeide and retinoic acid, and that Lipotec nanocapsules and phosphatidylcholine liposomes also improve the vitamin's photostability. A change in pH (from 5.6 to 7.0) of the gels did not influence the vitamin ester's stability. pH levels of 4.0 and 8.0 determined a decrease in the stability of retinyl palmitate in the gels. A high concentration of sunscreens improved the photostability of retinyl palmitate in the gels at pH 5.6 and 7.0. Butylated hydroxy toluene protected retinyl palmitate from degradation induced by light at all the pH levels studied and by heat at pH 5.6 and 7.0, as can be seen from the study of the photostability of vitamin A palmitate under UVB and UVA and of stability over time. Rheological studies showed a slight decrease in the viscosity of the gels after UVB-UVA irradiation and a higher decrease in the viscosity of the gels and the emulsions after storage at 25 degrees C and 40 degrees C. This decrease can be attributed to a partial degradation of hydroxy ethyl cellulose and of emulsifier, as can be seen from the decrease in shear stress versus shear rate values under these conditions of storage, denoting a depolymerization of the rheological modifier.

Deformable liposomes were prepared to investigate the effectiveness of dermal administration of methotrexate (MTX). The phospholipids used to prepare the liposomes were soybean lecithin (PC) or hydrogenated lecithin (HPC) and dipotassium glycyrrhizinate (KG) as surfactant. The lipid/KG ratio (w/w) was 2:1 and 4:1. Liposomes size, entrapment efficiency and MTX release through dialysis membrane were determined and the interaction between MTX and liposomes was investigated using differential scanning calorimetry. The MTX amount permeated through pig skin were three- to four-fold higher using liposomes containing KG compared to those from water solution or normal liposomes. No significant differences were observed between PC-KG liposomes and HPC-KG liposomes. At the end of the skin permeation assay using deformable liposomes, up to 50% of the administered dose was found in the skin. This capability depends on the self-regulating carrier deformability. These results suggest that liposomes containing KG may be of value for the topical administration of MTX in the treatment of psoriasis.

In recent years, transparent dispersions or diluted milks have been used as sunscreens. These products contain water-soluble sunscreen agents, and quite frequently are washed away from the skin. However, O/W microemulsions are now being prepared as transparent vehicles for sunscreens. They are waterproof, nonsticky, and easily spreadable. The microemulsions are prepared by using pseudoternary diagrams, by combining lipids with surfactant blends and a polar phase. Soya lecithin and decylpolyglucose produce transparent systems with the lowest percentage of surfactants. These microemulsions contain 4-methylbenzilidene camphor or octylmethoxycinnamate as sunscreen agents. Cyclomethicone, menthol, and allantoin give products a good skin feel, and stearyl methicone gives the waterproof effect. These systems show a Newtonian flux. Little permeation of the sunscreens' trough lipophilic and hydrophilic membrane is evidenced.

Microcalorimetric studies on alkylphosphate-butanol and alkylphosphate-butanol-lecithin systems were carried out as a first step to study the role of butanol (used as cosurfactant) in the formation of microemulsions. The enthalpy of solubilization: ΔH, of the same amount of butanol (1·806 mol kg−1) in aqueous sodium monoalkyl-phosphates, with a hydrocarbon chain varying from 6 to 10 carbon atoms, was investigated as a function of alkylphosphate concentration. Measurements were performed in the presence and in the absence of lecithin. The enthalpy values measured upon addition of butanol to the mixtures were negative in all cases. Below the critical micelle concentration (CMC) of the alkylphosphates, the heats of butanol dissolution were found to decrease upon increasing the concentration of the alkylphosphate. This behaviour was related to the dissolution of butanol in water and to the formation of mixed butanol-alkylphosphate and butanol-alkylphosphate-lecithin micelles. Above the CMC essentially constant ΔH values were found. The ΔH measured may be the sum of simultaneous effects—heat of alcohol dissolution and the simple alkylphosphate micelle dissociation or alkylphosphate-lecithin mixed aggregate dissociation and the heat of mixed micelle formation. The ΔH at the plateaus were linearly related to the corresponding CMC values of the alkylphosphates.
