(Asteraceae), é uma espécie vegetal característica de regiões tropicais, muito utilizada na medicina popular como anti-inflamatória, no tratamento de problemas hepáticos e renais, bem como para disfunções estomacais e intestinais. Recentemente, estudos demonstraram potencial atividade esquistossomicida sobre vermes adultos de Schistosoma mansoni (linhagem BH) para o óleo essencial (OE). Assim, este projeto teve por objetivo avaliar as atividades antiproliferativa e mutagênica, além da composição química do OE de B. trimera cultivada no campo experimental do CPQBA/UNICAMP. Coletas mensais (Janeiro/2012 a Dezembro/2012) das partes aéreas de B. trimera foram realizadas para a preparação do OE. Além disso, na época de floração em Julho, foram coletadas amostras das partes aéreas de indivíduos machos e fêmeas de B. trimera, diferenciadas morfologicamente pelas inflorescências. Todos os OEs preparados por hidrodestilação foram analisados por cromatografia gasosa acoplada a detector de massas (CG/EM) e avaliados quanto à atividade antiproliferativa in vitro e quanto à mutagenicidade no modelo de indução de micronúcleos com bloqueio da citocinese, com e sem ativação metabólica. A análise por CG/EM indicou que os compostos majoritários dos OEs foram biciclogermacreno, E-cariofileno, germacreno D, ?-pineno, globulol, ?-mirceno e ?-cadineno. Também foi observada uma variação na proporção relativa dos compostos presentes nos OEs obtidos das plantas fêmeas em relação aos machos. No teste de atividade antiproliferativa, em linhagens tumorais e não-tumorais, observou-se um perfil de atividade citostática semelhante para todas as amostras, independente da época de coleta, com valores de GI50 maiores que 25 ?g/mL. No teste do micronúcleo (MN) sem ativação metabólica, os OEs coletados no inverno (Jul/2012) e primavera (Out/2012) induziram um aumento na frequência de MNs, em relação ao controle de células sem tratamento, nas concentrações de 25 e 12,5 ?g/mL, enquanto os OEs obtidos no verão (Jan/2012) e outono (Abr/2012) apresentaram indução de MNs, estatisticamente significativa, somente na concentração de 25 ?g/mL. Nos experimentos realizados com os OEs extraídos de partes aéreas dos indivíduos machos e fêmeas, verificou-se que as plantas fêmeas não apresentaram potencial mutagênico nas três concentrações testadas (6,25; 12,5 e 25 ?g/mL), enquanto o OE obtido a partir dos indivíduos machos induziu aumento na frequência de MNs nas mesmas concentrações. Esses resultados sugerem que os sesquiterpenos hidrocarbonetos que predominam no OE obtido das partes aéreas das plantas machos (54,07%) podem estar envolvidos na ação mutagênica. Por outro lado, no teste com ativação metabólica, verificou-se uma redução significativa na frequência de MNs induzida pelos OEs em relação ao teste sem ativação, sugerindo que a metabolização dos OEs resultou em compostos com menor potencial mutagênico. Finalmente, no teste preliminar de antimutagenicidade, evidenciou-se no pós-tratamento e no tratamento simultâneo do OE com o metilmetanosulfonato uma possível reversão ou redução da clastogenicidade causada por este indutor de MNs. Esses dados sugerem possíveis atividades antimutagênica e mutagênica do OE das partes aéreas de B. trimera, com diferença significativa no potencial indutor de MNs entre indivíduos machos e fêmeas e que essa atividade é reduzida na presença de enzimas metabólicas, sugerindo uma potencial segurança de uso in vivo Abstract: "Carqueja", Baccharis trimera (Less.) DC. (Asteraceae) is a native species from tropical regions widely used in folk medicine as anti-inflammatory, to treat liver and kidney problems as well as stomach and intestinal disorders. Recent studies have demonstrated the potential of B. trimera essential oil (EO) as schistosomicidal on adult worms of Schistosoma mansoni (BH strain). Therefore, this project aimed the evaluation of chemical composition, antiproliferative and mutagenic activities of EO obtained from B. trimera aerial parts cultivated in CPQBA/UNICAMP experimental field. B. trimera aerial parts were collected monthly (January/2012 to December/2012) and immediately submitted to EO extraction by hydrodistillation. Moreover, samples of males and females of B. trimera aerial parts morphologically differentiated by inflorescences were collected during the flowering season (July/2012). All EOs were analyzed by gas chromatography coupled to mass detector (GC/MS). They were also evaluated for antiproliferative activity in vitro and mutagenicity in cytokinesisblock micronucleus assay (CBMN), with and without metabolic activation. GC/MS analysis indicated that EOs major compounds were bicyclogermacrene, E-caryophyllene, germacrene D, ?-pinene, globulol, ?-myrcene and ?-cadinene, besides a variation on relative proportion of compounds between EOs of female and male plants. Antiproliferative assay in tumor and nontumor cell lines showed a similar cytostatic activity profile for all samples, independent of the collection time, with GI50 values above 25 ?g/mL. In CBMN assay without metabolic activation, EOs collected in winter (Jul/2012) and spring (Oct/2012) induced at 25 and 12.5 ?g/mL an increase in micronuclei frequency in comparison to untreated control cells, whereas EOs obtained in summer (Jan/2012) and autumn (Apr/2012) showed statistically significant micronuclei induction only at 25 ?g/mL. CBMN assay performed with male and female aerial parts EOs demonstrated that female plants showed no mutagenic potential in three concentrations (6.25, 12.5 and 25 ?g/mL) whereas male aerial parts EO induced an increase in micronuclei frequency at the same concentrations. These results suggested that sesquiterpenes hydrocarbons predominately present in male aerial parts EO (54.07%) should be involved in mutagenic action. Moreover, there was a reduction in micronuclei frequency induced by EOs when CBMN assay was carried out with metabolic activation, which suggests that enzymatic metabolism of EOs resulted in compounds with low mutagenic potential. Finally, preliminary evaluation of antimutagenicity has evidenced that simultaneous and post treatments of EO with methylmetanesulfonate (MMS) could reverse and/or reduce clastogenicity caused by this inducer of MNs. These data suggest possible antimutagenic and mutagenic activities of aerial parts of B. trimera EO, with a significant difference in potential inducer of MNs between males and females plants. DC., conhecida como carqueja, é característica de regiões tropicais e muito utilizada na medicina popular como anti-inflamatória, bem como para disfunções estomacais e intestinais. Considerando o potencial uso medicinal do óleo essencial de B. trimera cv. CPQBA 1 (OEBt), esta pesquisa teve por objetivo a continuação do estudo fitoquímico e da avaliação de segurança do uso de OEBt. Foram comparados dois lotes de OEs; o primeiro obtido por hidrodestilação das partes aéreas de B. trimera coletadas mensalmente (Janeiro a Dezembro de 2014) no campo experimental do CPQBA/UNICAMP (OEBt CPQBA) e o segundo adquirido comercialmente (OEBt comercial). A análise por cromatografia gasosa acoplada a detector de massas identificou, como majoritários, os compostos biciclogermacreno, E-cariofileno, germacreno D, beta-pineno, beta-mirceno e delta-cadineno no OEBt CPQBA, ao passo que acetato de carquejila e palustrol foram identificados em OEBt comercial. As amostras foram ainda comparadas quanto à capacidade antioxidante in vitro através dos métodos 2,2-diphenyl-1-picrylhydrazyl (DPPH¿), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS¿+) e Oxygen Radical Absorbance Capacity (ORAC). No teste ABTS¿+, OEBt comercial foi mais ativo do que o OEBt CPQBA, enquanto no teste DPPH¿, ambas as amostras apresentaram capacidade sequestrante similar; os resultados também sugeriram a possível participação do alfa-pineno, E-cariofileno, mirceno e beta-pineno na atividade de OEBt CPQBA. No modo ORAC lipofílico, o OEBt CPQBA apresentou atividade antioxidante superior ao OEBt comercial que pode ser, em parte, atribuída ao E-cariofileno. No teste de atividade antiproliferativa, ambas as amostras OEBts apresentaram perfil citostático similar frente às linhagens celulares tumorais e não tumorais. Além disso, a mutagenicidade das amostras foi estudada através dos modelos de indução de micronúcleos in vitro (células CHO-K1) e in vivo (medula óssea de camundongos) e o potencial genotóxico foi avaliado por meio do teste Cometa alcalino em células CHO-K1. O OEBt CPQBA induziu um aumento significativo na frequência de micronúcleos (MN) em células CHO-K1, independente da concentração avaliada, enquanto o OEBt comercial aumentou de maneira significativa a frequência de MN nas maiores concentrações; os compostos mirceno, E-cariofileno e beta-pineno podem ter, em parte, contribuído para a mutagenicidade do OEBt CPQBA. A presença do sistema de metabolização exógena (fração S9) reverteu os efeitos mutagênicos de ambos OEBts, sugerindo a formação de compostos menos mutagênicos após metabolização. No teste Cometa alcalino in vitro, o OEBt CPQBA foi menos genotóxico do que o OEBt comercial, sendo que o uso da fração S9 reduziu a fragmentação do DNA induzida por OEBt comercial. O experimento de toxicidade oral aguda determinou a dose de 800 mg/kg como dose máxima tolerável (DMT) para ambos OEBts e, para tratamentos repetidos, selecionou-se a dose de 300 mg/kg. O OEBt comercial foi mutagênico (aumento na frequência de MN em eritrócitos policromáticos, MNPCE) nas três doses avaliadas, enquanto OEBt CPQBA induziu um aumento significativo na frequência de MNPCE apenas nas duas maiores doses. Através da abordagem metabolômica de fingerprint (ESI-MS seguido de análise de PLS-DA com VIP score) e da técnica de microextração em fase sólida de voláteis dispersos em headspace, foi possível evidenciar diferenças na composição química dos OEBts CPQBA e comercial antes e após o tratamento com fração S9. Concluindo, o OEBt CPQBA apresenta um efeito antioxidante mais evidente, além de ser menos genotóxico/mutagênico do que OEBt comercial; essas diferenças biológicas estão relacionadas com as variações encontradas na composição química. Estes resultados indicam uma potencial segurança no uso terapêutico de OEBt CPQBA, o que poderá ser comprovado em estudos complementares Abstract: Baccharis trimera (Less.) DC., known as "carqueja", is a native species from tropical regions widely used in folk medicine as anti-inflammatory and to treat stomach and intestinal disorders. Considering the medicinal use of B. trimera cv. CPQBA 1 essential oil (EOBt), this research aimed to continue the phytochemical study and safety evaluation of EOBt. This study compared two batches of EOs; the first one was obtained by hydrodistillation of B. trimera aerial parts collected from January to December 2014 in the CPQBA/UNICAMP experimental field (EOBt CPQBA), while the second was a commercial sample (EOBt commercial). Gas chromatography coupled to the mass detector identified as major compounds the bicyclogermacrene, E-caryophyllene, germacrene D, beta-pinene, beta-myrcene and delta-cadinene in EOBt CPQBA, whereas carquejyl acetate and palustrol were identified in EOBt commercial. The samples were compared for in vitro antioxidant capacity using 2,2-diphenyl-1-picrylhydrazyl (DPPH¿), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS¿+) and Oxygen Radical Absorbance Capacity (ORAC). In the ABTS¿+ assay, EOBt commercial was more active than EOBt CPQBA, while in the DPPH¿ assay, both samples presented similar scavenger abilities; the results also suggested the possible participation of alpha-pinene, E-caryophyllene, myrcene and beta-pinene on EOBt CPQBA activity. In the ORAC lipophilic mode, EOBt CPQBA showed higher antioxidant activity than EOBt commercial and this antioxidant potential can be attributed, in part, to E-caryophyllene. In the antiproliferative activity assay, both EOBts samples presented a similar cytostatic profile against the tumor and non-tumor cell lines. In addition, the mutagenicity of the samples was studied using the in vitro (CHO-K1 cells) and in vivo (mouse bone marrow) micronuclei induction models while the genotoxic potential was evaluated by the in vitro (CHO-K1 cells) alkaline Comet assay. EOBt CPQBA increased the micronuclei (MN) frequency in CHO-K1 cells, concentration-independent, while EOBt commercial increased the MN frequency at the highest concentrations; myrcene, E-caryophyllene and beta-pinene may have contributed in part to the EOBt CPQBA mutagenicity. The presence of an exogenous metabolic system (S9 fraction) reversed both EOBts mutagenic effects, suggesting the formation of less mutagenic compounds after metabolization. In the in vitro alkaline Comet assay, EOBt CPQBA was less genotoxic than EOBt commercial and the use of S9 fraction reduced the EOBt commercial-induced DNA fragmentation. The acute oral toxicity experiment determined 800 mg/kg as the maximum tolerable dose (MTD) for both EOBts and from this, the dose of 300 mg/kg was selected for repeated treatments. EOBt commercial was mutagenic (dose-dependent increase in micronucleated polychromatic erythrocytes, MNPCE) in the three doses evaluated, while EOBt CPQBA increased MNPCE frequency only in the two higher doses. Through the fingerprint metabolomic approach (ESI-MS followed by PLS-DA analysis with VIP score) and the solid-phase microextraction technique of headspace volatiles, it was possible to show differences in the chemical composition of EOBts CPQBA and commercial before and after treatment with S9 fraction. Concluding, EOBt CPQBA has a more evident antioxidant effect in addition to being less genotoxic/mutagenic than EOBt commercial; these biological differences are related to chemical composition variations. Orientadores: Vitor Rafael Coluci, Gisela de Aragão Umbuzeiro Dissertação (mestrado) - Universidade Estadual de Campinas, Faculdade de Tecnologia Resumo: O 3-nitrobenzantrona (3-NBA) é um composto mutagênico e genotóxico gerado principalmente durante a queima de combustíveis fósseis que vem sendo encontrado em diferentes matrizes ambientais. Devido a essas características, é necessário se desenvolver técnicas de remediação para diminuir a concentração do 3-NBA no ambiente. Atualmente, nanomateriais tem sido utilizado para se remediar poluentes orgânicos presentes na água. Dentre eles, o óxido de grafeno (GO) tem sido utilizado em diversas aplicações devido a sua composição química versátil, grande área superficial e boa solubilidade que permitem adsorver física ou quimicamente poluentes orgânicos. O objetivo desta pesquisa foi estudar a capacidade de adsorção do 3-NBA pelo GO em água por meio de testes biológicos. Utilizamos o teste de Salmonella/microsoma (Teste de Ames) pelo fato do 3-NBA ser muito potente nesse tipo de teste. Resultados de testes Salmonella/microsoma demostraram que o GO é realmente de capaz de adsorver 3-NBA. Essa adsorção é dependente da quantidade de GO utilizada sendo maior a adsorção em maiores concentrações de GO. Observamos um aumento na adsorção de 3-NBA quando as amostras de GO foram lavadas por processos químicos envolvendo solução básica. Baseando-se nos modelos atuais para a estrutura do GO, sugerimos que esse aumento da capacidade pode ser atribuído tanto à remoção de resíduos superficiais da síntese do GO (conhecidos como debris) quanto à remoção parcial de grupos oxigenados presentes na superfície de GO Abstract: 3-Nitrobenzantrone (3-NBA) is a mutagenic and genotoxic compound, mainly generated during the burning of fossil fuels and that has been found in different environmental matrices. Due to these characteristics, the development of remediation techniques to reduce the 3-NBA environment concentration is extremely necessary. Currently, nanomaterials have been used to remediate organic pollutants present in the water. Among them, graphene oxide (GO) has been used in various applications due to its versatile chemical composition, large surface area and good solubility which allow physical or chemical adsorption of organic pollutants. The aim of this work was to study the capacity of GO to adsorb 3-NBA in water. We used biological tests to determine the GO adsorption capacity, especially the Salmonella/microsome test (Ames test) because the 3-NBA is very powerful in this type of test. Salmonella/microsome tests have shown that GO is really capable of adsorbing 3-NBA. This adsorption is dependent on the amount of GO used in the tests with higher GO concentrations leading to higher 3-NBA adsorption. Chemical processes involving basic washing GO with NaOH solution led to an increase in the adsorption capacity of the GO samples. We suggest that this increase can be attributed to the removal of surface residues from the GO synthesis (known as oxidation debris) and to the partial removal of oxygenated groups present in the GO surface, depending on models used to describe the GO chemical structure Mestrado Ambiente Mestra em Tecnologia FAPESP 2012/19982-5

Orientador: Gisela de Aragão Umbuzeiro Dissertação (mestrado) - Universidade Estadual de Campinas, Faculdade de Tecnologia Resumo: Cerca de 70% dos corantes utilizados em indústrias têxteis são corantes do tipo azo, que se caracterizam pelo grupo -N=N- ligado a sistemas aromáticos, sendo que a função azo inclui os principais tipos de corantes. A eliminação da cor no efluente é um grande desafio para o setor têxtil, já que, aproximadamente, 15% da produção mundial de corantes são descartados para o meio ambiente durante sua produção, processamento e aplicação. O corante comercial Disperse Red 1 utilizado neste trabalho é composto por seis corantes diferentes e surfactante, porém o corante principal em sua composição é o Disperse Red 1 (N-Ethyl-N-(2-hydroxyethyl)-4-(4- nitrophenylazo) aniline; CAS number 2872-52-8) com 60% em massa. O corante comercial foi submetido ao processo de cloração com gás cloro, simulando as condições utilizadas em Estações de Tratamento de Efluentes. A toxicidade do corante comercial CI Disperse Red 1 e do subproduto clorado foi avaliada em testes agudos com Ceriodapnhia dubia, Ceriodaphnia silvestrii, Daphnia similis, Daphnia magna, Hydra attenuata, e em testes crônicos com Pseudokirchneriella subcapitata, Ceriodapnhia dubia, e Hydra attenuata. A mutagenicidade foi avaliada com ensaio Salmonella/microssoma com as linhagens TA98, TA100 e YG1041. D. similis foi o organismo mais sensível ao corante comercial; e H. attenuata foi mais sensível ao subproduto clorado. A CE50 obtida para D. similis para o corante comercial é similar ao corante puro, mostrando que a toxicidade do corante comercial é devido ao Disperse Red 1. O subproduto clorado foi menos tóxico do que o corante para todos os organismos testados, com exceção de H. attenuata. Porém, o subproduto clorado foi mais mutagênico do que o corante para todas as linhagens testadas. Mais estudos são necessários para compreender os mecanismos envolvidos na toxicidade destes corantes, considerando sua alta toxicidade para organismos aquáticos, a fim de fornecer informações para a elaboração de corantes ambientalmente corretos. Os resultados obtidos neste trabalho podem fornecer informações úteis para a derivação de critérios para qualidade da água para esse corante Abstract: About 70% of dyes used in textiles industries are of type azo. These days 15% of the dye world production is discharged into the environment during its production, processing and application. Effluent that contains dyes must have their color removed in order to be in compliance with environmental regulations, but sometimes the treatment can lead to more toxic compounds. The commercial azo dye Disperse Red 1 used in this study is composed of six different dye constituents and surfactant; however the main dye of the commercial product is the Disperse Red 1 (N-Ethyl-N-(2-hydroxyethyl)-4-(4-nitrophenylazo) aniline; CAS number 2872-52-8). Commercial dye samples were treated with chlorine gas, in order to simulate the conditions used in effluent treatment plant. The ecotoxicity of the commercial dye and chlorinated byproduct was evaluated in acute tests with Ceriodaphnia dubia, Ceriodaphnia silvestrii, Daphnia similis, Daphnia magna, and Hydra attenuata as well as in chronic toxicity tests with Pseudokirchneriella subcapitata, Ceriodapnhia dubia and Hydra attenuata. Mutagenicity was evaluated using Salmonella/microsome assay with TA98, TA100 and YG1041 strains. The commercial dye Disperse Red 1 was similarly toxic to P. subcapitata, C. dubia and D. similis, even with different endpoints. The chlorinated byproduct was less toxic to all organisms tested than the dye, except for H. attenuata. The toxicity of the commercial dye is due to Disperse Red 1 itself and the surfactant does not seem to contribute to the toxicity, at least to D. similis. The chlorinated byproduct was more mutagenic than the commercial dye. More studies are necessary to address the mechanisms involved in the toxicity of this azo dye considering its high toxicity to aquatic organisms in order to provide information for the design of more environmental friendly dye products. The Salmonella reversion based Ames test is the most widely used method for mutagenicity testing. For rapid toxicity assessment of e.g. water samples and for effect-directed analysis, however, the Ames test suffers from lack of throughput and is regarded as a laborious, time consuming method. To achieve faster analysis, with increased throughput, a (downscaled) luminescent derivative of the Ames Salmonella/microsome fluctuation test has been developed through expression of the Photorhabdus luminescens luciferase in the Salmonella TA98 and TA100 strains. The applicability of this test is demonstrated by analysis of environmentally relevant compounds, a suspended particulate matter extract and an industrial effluent sample. Use of the luminescent reporter reduced the required detection time from 48 to 28 h with a specificity of 84% for responses reported in the literature to a set of 14 mutagens as compared to 72% in the unmodified fluctuation test. Testing of the same compounds in a downscaled luminescent format resulted in an 88% similarity with the response found in the regular luminescent format. The increase in throughput, faster analysis and potential for real-time bacterial quantification that luminescence provides, allows future application in the high-throughput screening of large numbers of samples or sample fractions, as required in effect-directed analysis in order to accelerate the identification of (novel) mutagens.

É provável que a presença de impurezas nas amostras possa ser uma das causas do possível potencial mutagênico. Dessa forma, esse estudo visa avaliar a atividade mutagênica do corante tartrazina com diferentes graus de pureza e possíveis interferentes presentes nas amostras, utilizando o ensaio Salmonella/microssoma a partir das linhagens recomendadas pela OECD 471. Os resultados obtidos demonstraram que o corante tartrazina ?99 % e o corante tartrazina comercial 90%, não apresentaram atividade mutagênica para as linhagens TA97a, TA98, TA100, TA1535 e TA102 demonstrando ausência de impurezas mutagênicas ou que as mesmas estejam em baixas concentrações nas amostras avaliadas Abstract: Although very useful and diverse applications in industry, for years the discussion on the genotoxic potential of the dye tartrazine has been addressed, since there are several controversial results in the literature. It is likely that the presence of impurities in the samples may be a cause of the possible mutagenic potential. Thus, this study aims to evaluate the mutagenic activity of the dye tartrazine with different degrees of purity and possible interferences present in the samples, using the Salmonella / microsome test from the lines recommended by the OECD 471. More than any other environmental chemicals, dioxins have been in the limelight of public interest for about 10 years. In addition to carcinogenicity, genetic risk is a cause for concern. Mutagenicity tests performed so far do not give a clear picture. The mutagenic potential of dioxins has to be considered weak or absent. Therefore, it seemed profitable to investigate comutagenicity and co-recombinogenicity of dioxins more thoroughly. The only useful method for investigating comutagenicity and co-recombinogenicity of dioxins in vivo is the spot test with mice. In this test system, a number of cocarcinogens and tumor promoters have shown comutagenic or co-recombinogenic effects. In the present study, tetrachlorodibenzo-p-dioxin (TCDD) and two environmental dioxin mixtures [pentachlorodibenzodioxin (PCDD) 1 and 2] were tested for genetic activity. Given alone, no mutagenic or recombinogenic effects could be observed. In combination with the carcinogenic mutagen ethyl nitrosourea (ENU) at concentrations of 128 micrograms/kg for PCCD 2, 314 micrograms/kg for PCDD 1, and 3 micrograms/kg for TCDD, a doubling of the genetic effectiveness of ENU was observed. The genetic risk can roughly be considered as 1:0.02 for TCDD:PCDD 2 and 1:0.01 for TCDD:PCDD 1. While PCDD 1 and 2 seem to enhance the mutagenic as well as the recombinogenic potential of ENU, TCDD showed mainly co-recombinogenic and antimutagenic activity. This characteristic indicates that TCDD is mainly a tumor promoter.

Flow cytometric measurements of the chromosomal DNA content have been used to develop a screening method for the detection of chemically- or physically-induced cytogenetic damage. The reproducibility of this flow cytogenetic assay was shown in a series of subcultures of a Chinese hamster cell clone. The accuracy and sensitivity was tested in cultures treated with chemical mutagens and x-rays. The clastogenic effectiveness was quantified and the dose-effect relationship was established by the increase of the coefficient of variation of the peak of the largest chromosome type in the flow histograms. Since structural chromosome aberrations cause an unequal division of the DNA at mitosis, it is expected that clastogenic effects can be detected also in whole cells of growing populations as an increased dispersion of the cellular DNA content. In order to test this feature, high resolution flow cytometric measurements were performed in x-irradiated hamster cells in vitro and mouse bone marrow cells in vivo.

A long-term test with yeast strain MP1, suitable for simultaneous detection of mutation, nonreciprocal recombination due to gene conversion, and reciprocal recombination has been developed. Principle of the method is the permanent and exactly calibrated supply of fresh media and removal of exhausted media allowing yeast cells to grow permanently for months. First experiments has shown that it is possible to detect the effect of carcinogens in concentrations being at least 1 order of magnitude lower than in short-term tests. Furthermore it was possible to get positive results with caprolactam, quercetin and chrysarobin, substances which are negative in short-term tests with yeast.
