Urban water system ecology has been severely damaged, urban waterlogging, water pollution, water resources shortage and other problems have become more serious, and it is urgent to repair and harness them. The concept of sponge city can effectively improve the state of urban water system. Therefore, this article aims to provide theoretical support for China's sponge city pollution water purification technology by analyzing the characteristics, hazards, and different sewage purification technologies of urban sewage.

Accumulating evidence indicates a correlation between circadian dysfunction and genomic instability. However, whether the circadian machinery directly regulates DNA damage repair, especially in double-strand breaks (DSBs), remains poorly understood. Here, we report that in response to DSBs, BMAL1 is activated by ATM-mediated phosphorylation at S183. Phosphorylated BMAL1 is then localized to DNA damage sites, where it facilitates acetylase CLOCK to load in the chromatin, regulating the acetylation of histone H4 (H4Ac) at DSB sites. In this way, the BMAL1-CLOCK-H4Ac axis promotes the DNA end-resection to generate single-stranded DNA (ssDNA) and the subsequent homologous recombination (HR). BMAL1 deficient cells display defective HR, accumulation of unrepaired DSBs and genome instability. Accordingly, depletion of BMAL1 significantly enhances the sensitivity of adrenocortical carcinoma (ACC) to DNA damage-based therapy in vitro and in vivo. These findings uncover non-canonical function of BMAL1 and CLOCK in HR-mediated DSB repair, which may have an implication in cancer therapeutics.

Swainsonine (SW) is the principal toxic ingredient of locoweeds, and is produced by fungi including Metarhizium anisopliae, Slafractonia leguminicola, and Alternaria oxytropis. A key enzyme in the SW synthesis pathway is a hybrid SWNK/NRPS. The downstream biosynthetic pathways of SW have been clearly analyzed by a knockout and complement of swnK in Metarhizium robertsii, but the upstream steps and the associated regulatory proteins have not been thoroughly investigated. To verify the role of swnK in the SW biosynthesis pathway of M. anisopliae, the concentration of SW and the swnK expression in M. anisopliae fermentation from 1st to 7th d were determined using LC-MS and RT-qPCR, respectively. M. anisopliae had the highest SW content and swnK expression on the 5th day of fermentation; We obtained a mutant strain (MT) by PEG-mediated homologous recombination (HR) to knock out swnK in the wild-type (WT) strain. And we obtained a complemented-type (CT) strain by transforming a modified PUC19 complementation vector containing the geneticin (G418) resistance gene and swnK. SW was not detected in the MT strain and reverted to its original level in the CT strain; We also used a Psilent-1 plasmid with Benomyl (ben)-resistant that interfered with swnK of wild-type strain (WT). The level of SW in the RNAi strain was markedly diminished. RNAi of swnK affected the formation of the cell wall in M. anisopliae. These results indicate that swnK plays a crucial role in the SW biosynthesis of M. anisopliae.

During the transformer winding deformation process, the leakage magnetic field around the winding will change accordingly. Therefore, it is an effective method to monitor and track the change of the leakage magnetic field and then analyze and judge the state of the transformer. This paper firstly uses Comsol Multiphysics software to establish a 110 kV transformer electromagnetic simulation calculation model. Based on the simulation results of magnetic leakage distribution, an installation plan for the internal magnetic leakage sensor of a 110 kV true transformer is determined. The measurement results of the true single short-circuit test under different working conditions verify the accuracy of the simulation model. Subsequently, a number of B-phase high-centered three-phase short circuit (H-M B) true type tests were carried out, and the relationship between the magnetic leakage distribution characteristics and the impedance change rate after each impact was analyzed. The results show that before the transformer is seriously deformed due to multiple short circuit shocks, the sensitivity of the impedance change rate to the winding deformation is low, and the first five shocks only increase from 0.11% to 0.39%. However, the difference ratio between the simulation value and the test value of magnetic flux leakage (MFL) has obvious changes in each small deformation. BX3 increases from 1.77% to 5.62%, and BX4 increases from 2.08% to 6.55%. The difference ratio of four shocks before winding deformation is more than 6%. Therefore, by monitoring the flux leakage magnetic induction intensity, when the difference ratio is greater than 6%, strengthen the vigilance, which can provide a certain basis for winding monitoring before serious deformation.

Infiltration of tumor-associated macrophages (TAMs) can promote tumorigenesis and development. C-C motif chemokine ligand 3 (CCL3) was reported to be derived from TAMs and tumor cells and facilitate the progression of several cancers. Nevertheless, whether CCL3 can be derived from TAMs and tumor cells of colon adenocarcinoma (COAD) is unclarified.Peripheral blood monocytes-derived macrophages were polarized by the conditioned medium from COAD cells to establish TAM-like macrophages (TAM1/2). RT-qPCR and western blotting were used for detection of expression levels of CCL3 and its receptors C-C motif chemokine receptor 1 (CCR1) and CCR5 in TAM1/2 and COAD cells. Immunofluorescence staining was utilized for evaluating CCL3, CD163 and CCR5 expression. The Akt signaling pathway-associated protein levels were measured by western blotting. Transwell assays were used for assessing cell migration and invasiveness.CCL3 displayed a high level in TAMs and cancer cells of COAD. CCL3 activated the Akt signaling pathway by binding to CCR5. CCL3-CCR5 axis facilitated COAD cell migration and invasiveness by activating the Akt signaling. CCL3 derived from both TAMs and cancer cells contributed to the malignant behaviors of COAD cells. High expression of CCL3/CCR5 was closely associated with poor prognoses of COAD patients.CCL3-CCR5 interaction promotes cell migration and invasiveness, and functions as a prognostic biomarker for COAD.

Many AI system designers grapple with how best to collect human input for different types of training data. Online crowds provide a cheap on-demand source of intelligence, but they often lack the expertise required in many domains. Experts offer tacit knowledge and more nuanced input, but they are harder to recruit. To explore this trade off, we compared novices and experts in terms of performance and perceptions on human intelligence tasks in the context of designing a text-based conversational agent. We developed a preliminary chatbot that simulates conversations with someone seeking mental health advice to help educate volunteer listeners at 7cups.com. We then recruited experienced listeners (domain experts) and MTurk novice workers (crowd workers) to conduct tasks to improve the chatbot with different levels of complexity. Novice crowds perform comparably to experts on tasks that only require natural language understanding, such as correcting how the system classifies a user statement. For more generative tasks, like creating new lines of chatbot dialogue, the experts demonstrated higher quality, novelty, and emotion. We also uncovered a motivational gap: crowd workers enjoyed the interactive tasks, while experts found the work to be tedious and repetitive. We offer design considerations for allocating crowd workers and experts on input tasks for AI systems, and for better motivating experts to participate in low-level data work for AI.

The traceability system has significantly contributed to ensure food safety and quality. However, the biggest difficulty in food traceability is the numerous links from field to table, and there is no stable strategic partnership between supply chain members and the lack of social responsibility of some practitioners. Thus, this study aims to seek the best traceability strategy for companies in centralized model and decentralized model, respectively. Therefore, we have constructed a differential game model based on the delay effect to determine the optimal traceability level and traceable goodwill and compare the profits of the food supply chain (FSC). The results show that the delay time is positively related to the level of traceability effort and has a high impact on the traceable goodwill. Companies in the FSC can formulate optimal traceability strategies based on delay time and foster improvement in food safety and quality.

Detecting 3D objects in a crowd remains a challenging problem since the cars and pedestrians often gather together and occlude each other in the real world. The Pointpillar is the leader in 3D object detection, its detection process is simple, and the detection speed is fast. Due to the use of maxpooling in the Voxel Feature Encode (VFE) stage to extract global features, the fine-grained features will disappear, resulting in insufficient feature expression ability in the feature pyramid network (FPN) stage, so the object detection of small targets is not accurate enough. This paper proposes to improve the detection effect of networks in complex environments by integrating attention mechanisms and the Pointpillar. In the VFE stage of the model, the mixed-attention module (HA) was added to retain the spatial structure information of the point cloud to the greatest extent from the three perspectives: local space, global space, and points. The Convolutional Block Attention Module (CBAM) was embedded in FPN to mine the deep information of pseudoimages. The experiments based on the KITTI dataset demonstrated our method had better performance than other state-of-the-art single-stage algorithms. Compared with another model, in crowd scenes, the mean average precision (mAP) under the bird’s-eye view (BEV) detection benchmark increased from 59.20% of Pointpillar and 66.19% of TANet to 69.91 of ours, the mAP under the 3D detection benchmark was increased from 62% of TANet to 65.11% of ours, and the detection speed only dropped from 13.1 fps of Pointpillar to 12.8 fps of ours.

Permanent magnet synchronous motors (PMSMs) with non-concentric magnetic poles (NCMP) structure are widely used as high-precision servo motors. The current subdomain method mainly equates the NCMP to a rectangular magnet to make each subdomain boundaries of magnet parallel when calculating the magnet eddy current loss (ECL), which results in a lower accuracy of the calculation. This paper divides the NCMP into non-concentric region and concentric region along the radial direction. The non-concentric region is further divided along the circumferential direction, and each divided region has a single thickness. Based on the subdomain method, a novel analytical method of magnet ECL in PMSMs with NCMPs is proposed. Moreover, the 3-D distribution coefficient of ECL in the NCMP is derived by the equivalent impedance method. The effects of eddy current reaction and stator slotting are taken into account to improve the calculation accuracy. Then, through theoretical derivation, the relationship between the minimum magnet loss and the pole parameters is obtained, which provides a reference for the design of NCMPs. Finally, the loss separation experiment is performed on four prototypes with NCMPs. The experimental results, finite element method results and analytical results are compared to verify the correctness of the model.

Dysregulation of pseudogenes, enhancement of fatty acid synthesis and formation of immunosuppressive microenvironment are important factors that promote the malignant progression of glioma. It is of great significance to search for the molecular mechanism of interaction between the three and then perform targeted interference for improving the treatment of glioma. In this study, we found that pseudogene transmembrane protein 198B (TMEM198B) was highly expressed in glioma tissues and cell lines, and it could promote malignant progression of glioma by regulating lipid metabolism reprogramming and remodeling immune microenvironment. Applying the experimental methods of gene interference, lipidomics and immunology, we further confirmed that TMEM198B promoted PLAG1 like zinc finger 2 (PLAGL2) expression by mediating tri-methylation of histone H3 on lysine 4 (H3K4me3) of PLAGL2 through binding to SET domain containing 1B (SETD1B). Increased PLAGL2 could transcriptional activate ATP citrate lyase (ACLY) and ELOVL fatty acid elongase 6 (ELOVL6) expression, and then influenced the biological behaviors of glioma cells via enhancing the de novo lipogenesis and fatty acid acyl chain elongation. At the same time, TMEM198B promoted macrophages lipid accumulation and intensification of fatty acid oxidation (FAO) through glioma-derived exosomes (GDEs), further induced macrophages to M2 polarization, which subsequently facilitated immune escape of glioma cells. In conclusion, our present study clarifies that the TMEM198B/PLAGL2/ACLY/ELOVL6 pathway conducts crucial regulatory effects on the malignant progression of glioma, which provides novel targets and new ideas for molecular targeted therapy and immunotherapy of glioma.

Awareness of radiation-related knowledge (RRK) and nuclear energy-related knowledge (NERK) among residents around a nuclear power plant (NPP), as well as their concerns about a NPP, were investigated. A face-to-face survey was conducted among 1,775 residents within 30 km around the NPP in Liaoning Province, China. A single-item Likert scale, Spearman's/Pearson's correlation coefficients, Student's t-test, ANOVA, and multiple-linear regression analysis were employed. Awareness of RRK and NERK among residents around the NPP was 27.7% and 36.6%, respectively. The anxiety level of respondents was negatively corelated with the distance from their residence to the NPP and age. Also, 55.6% of respondents thought that the publicity about nuclear energy/NPPs was insufficient, and 82.7% of respondents wanted to know relevant information about NPPs. Awareness of RRK and NERK among residents around the NPP was relatively low, which was related to education, occupation, and income. The anxiety level among residents was related to distance and age. The public was eager to know about RRK and NERK. These findings indicate that the publicity and education of RRK and NERK among residents around the NPP should be strengthened.

BackgroundPrevious studies have demonstrated that prophylactic cranial irradiation (PCI) could reduce the risk of brain metastases and prolong the overall survival (OS) of patients with small cell lung cancer (SCLC). However, it remains controversial whether the efficacy and safety of PCI would be subjected to the different characteristics of patients with extensive stage of SCLC. This meta-analysis aims to evaluate the efficacy and safety of PCI in patients with extensive stage SCLC.MethodsPubMed, Embase, and the Cochrane Library were searched for relevant studies from inception to May, 2021. Hazard ratios (HRs) were used to measure the OS and progression-free survival (PFS), and relative risks (RRs) were employed to calculate the incidence of brain metastases, survival rate, and adverse events. Summary results were pooled using random-effect models.ResultsThere were 1215 articles identified, and 15 trials were included, with a total of 1,623 participants. Patients who received PCI did not result in significantly improved OS [HR=0.87, 95%CI (0.70, 1.08) p=0.417] and PFS [HR=0.81, 95%CI (0.69, 0.95) p=0.001], compared with those who did not receive PCI, while patients who received PCI had a significantly decreased incidence of brain metastases [RR=0.57, 95%CI (0.45, 0.74), pp=0.154), 3-year (RR=0.97, p=0.072), 4-year (RR=0.71, p=0.101) and 5-year survival rates (RR=0.32, p=0.307), compared with non-PCI group, whereas the overall RR indicated that PCI was associated with a higher 1-year survival rate [RR=1.46, 95%CI (1.08, 1.97), p=0.013]. In addition, PCI treatment was shown to be associated with increased incidence of adverse events, including fatigue, dermatitis, anorexia, nausea, vomiting, malaise, and cognitive impairment.ConclusionThis meta-analysis suggests that PCI can reduce the incidence of brain metastases in extensive stage SCLC. Although PCI has no significant effect on the OS, it improves 1-year survival in patients with extensive stage SCLC. However, PCI does not significantly affect 2,3,4,5-year survival and may result in a significantly increased risk of adverse events.

Background The emergency of Omicron variants, spreading in China and worldwide, has sparked a new wave of the coronavirus disease 2019 (COVID-19) pandemic. The high infectivity and persistence of the pandemic may trigger some degrees of post-traumatic stress disorder (PTSD) for nursing students experiencing indirect trauma exposure to the epidemic, which hinders the role transition from students to qualified nurses and exacerbates the health workforce shortage. Thus, it’s well worth an exploration to understand PTSD and its underlying mechanism. Specifically, PTSD, social support, resilience, and fear of COVID-19 were selected after widely literature review. This study aimed to investigate the relationship between social support and PTSD among nursing students during COVID-19, to address the mediating role of resilience and fear of COVID-19 between social support and PTSD, and to provide practical guidance for nursing students’ psychological intervention. Methods From April 26 to April 30, 2022, 966 nursing students from Wannan Medical College were selected by the multistage sampling method to fill the Primary Care PTSD Screen for the fifth edition of the Diagnostic and Statistical Manual of Mental Disorders (DSM-5), Brief Resilience Scale, Fear of COVID-19 Scale, and Oslo 3 Items Social Support Scale. Data were analyzed by descriptive statistics, spearman’s correlation analysis, regression analysis, and path analysis. Results 15.42% of nursing students had PTSD. There were significant correlations between social support, resilience, fear of COVID-19, and PTSD (r =-0.291 ~ 0.353, P Conclusion The social support of nursing students not only directly affects PTSD, but also indirectly affects PTSD through the separate and chain mediating effect of resilience and fear of COVID-19. The compound strategies targeted at boosting perceived social support, fostering resilience, and controlling fear of COVID-19 are warranted for reducing PTSD.

Objective: Although great progress has been made in diagnosis and treatment of lung adenocarcinoma (LUAD), the clinical prognosis and overall survival of advanced LUAD are still poor. It has been proved that ferroptosis plays crucial role in LUAD. However, the prognosis of ferroptosis related genes (FRGs) in LUAD remains largely unclear. Thus, this study aims at exploring the FRGs signature in the prognosis of LUAD. Methods: FGRs,the gene expression data, and GSE72094 and GSE68465 datasets was downloaded from FerrDb database, The Cancer Genome Atlas (TCGA), and The Gene Expression Omnibus (GEO) database. The survival analysis was analyzed by R package “survminer” using Kaplan-Meier curve and log-rank test. The model’s predictive ability was confirmed using receiver operator characteristics (ROC) curve. Immune and stromal score of tumor sample was calculated with the ESTIMATE algorithm. The infiltrating score of 16 immune cells and the immune-related pathways (13) activity was detected by single-sample gene set enrichment analysis (ssGSEA). The Tumor Immune Dysfunction and Exclusion (TIDE) and R package “OncoPredict” (v0.2) were applied to predict the potential response to immune checkpoint blockade and the half-maximal inhibitory concentration (IC50) of each drug, respectively. Results: We constructed a novel 17 FRGs prognostic signature in LUAD, that show excellent predictive performance for the overall survival. Furthermore, we found that the high-risk group had a lower survival rate than that in the low-risk group. The ROC curve indicated the accuracy of the risk score model in judging the prognosis of LUAD. ESTIMATE and IC50 assay revealed that the risk scoring model was significantly associated with immune cell infiltration and response to targeted therapy. Conclusion: 17 FRGs signature showed outstanding advantages in the LUAD prognosis and dysregulated immune cell infiltration, suggesting that it might be a therapeutic alternative for LUAD by targeting ferroptosis.
