Abstract

Experimental cross sections for the He4(e,e′p)X reactions in the missing energy range from 0.017 to 0.022 GeV and up to a missing momentum of 0.632 GeV/c at xB=1.24 and Q2=2 (GeV/c)2 are reported. The data are compared to relativistic distorted-wave impulse approximation calculations for the He4(e,e′p)H3 channel. Significantly more events are observed for pm≥0.45 GeV/c than are predicted by the theoretical model, and striking fluctuations in the ratio of data to the theoretical model around pm=0.3GeV/c are possible signals of initial-state multinucleon correlations.

Abstract

Multiferroic heterostructure CoFeB/Pb(Mg1/3Nb2/3)O3–PbTiO3 is prepared by depositing CoFeB films on Pb(Mg1/3Nb2/3)O3–PbTiO3 single crystal through direct current magnetron sputtering. The electric field induced magnetism change of CoFeB film was studied by magneto-optical Kerr effect measurement. The magnetic hard/easy axis change was investigated under direct current electric field. Alternative current voltage magneto-optical Kerr effect test indicates the different magnetoelectric mechanism with the change of film thickness. The microwave frequency tunability was observed under the electric field and the tunable ferromagnetic resonance field reaches 190 Oe. Our study provides great opportunities for the application of electric-field tunable information storage devices.

Abstract

The inclusive cross-section of annihilation in flight $e^+e^-\rightarrow\gamma\gamma$ of 430 MeV positrons with atomic electrons of a thin diamond target has been measured with the PADME detector at the Laboratori Nazionali di Frascati. The two photons produced in the process were detected by an electromagnetic calorimeter made of BGO crystals. This measurement is the first one based on the direct detection of the photon pair and one of the most precise for positron energies below 1 GeV. This measurement represents a necessary step to search for dark sector particles and mediators weakly coupled to photons and/or electrons with masses ranging from 1 MeV to 20 MeV with PADME. The measurement agrees with the Next to Leading Order QED prediction within the overall 6% uncertainty., Comment: Submitted to PRD

Abstract

The recently approved Electron-Ion Collider (EIC) will provide a unique new opportunity for searches of charged lepton flavor violation (CLFV) and other new physics scenarios. In contrast to the $e \leftrightarrow μ$ CLFV transition for which very stringent limits exist, there is still a relatively large discovery space for the $e \to τ$ CLFV transition, potentially to be explored by the EIC. With the latest detector design of ECCE (EIC Comprehensive Chromodynamics Experiment) and projected integral luminosity of the EIC, we find the $τ$-leptons created in the DIS process $ep\to τX$ are expected to be identified with high efficiency. A first ECCE simulation study, restricted to the 3-prong $τ$-decay mode and with limited statistics for the Standard Model backgrounds, estimates that the EIC will be able to improve the current exclusion limit on $e\to τ$ CLFV by an order of magnitude., 11 pages, 8 figures, to be submitted to NIM

Abstract

Exclusive heavy quarkonium photoproduction is one of the most popular processes in EIC, which has a large cross section and a simple final state. Due to the gluonic nature of the exchange Pomeron, this process can be related to the gluon distributions in the nucleus. The momentum transfer dependence of this process is sensitive to the interaction sites, which provides a powerful tool to probe the spatial distribution of gluons in the nucleus. Recently the problem of the origin of hadron mass has received lots of attention in determining the anomaly contribution $M_{a}$. The trace anomaly is sensitive to the gluon condensate, and exclusive production of quarkonia such as J/$ψ$ and $Υ$ can serve as a sensitive probe to constrain it. In this paper, we present the performance of the ECCE detector for exclusive J/$ψ$ detection and the capability of this process to investigate the above physics opportunities with ECCE., 11 pages, 14 figures, 1 table

Abstract

The ECCE detector has been recommended as the selected reference detector for the future Electron-Ion Collider (EIC). A series of simulation studies have been carried out to validate the physics feasibility of the ECCE detector. In this paper, detailed studies of heavy flavor hadron and jet reconstruction and physics projections with the ECCE detector performance and different magnet options will be presented. The ECCE detector has enabled precise EIC heavy flavor hadron and jet measurements with a broad kinematic coverage. Abstract

The strong interaction is not well understood at low energy, or for interactions with low momentum transfer $Q^2$, but one of the clearest insights we have comes from Chiral Perturbation Theory ($\chi$PT). This effective treatment gives testable predictions for the nucleonic generalized polarizabilities -- fundamental quantities describing the nucleon's response to an external field. We have measured the proton's generalized spin polarizabilities in the region where $\chi$PT is expected to be valid. Our results include the first ever data for the transverse-longitudinal spin polarizability $\delta_{LT}$, and also extend the coverage of the polarizability $\bar{d_2}$ to very low $Q^2$ for the first time. These results were extracted from moments of the structure function $g_2$, a quantity which characterizes the internal spin structure of the proton. Our experiment ran at Jefferson Lab using a polarized electron beam and a polarized solid ammonia (NH$_3$) target. The $\delta_{LT}$ polarizability has remained a challenging quantity for $\chi$PT to reproduce, despite its reduced sensitivity to higher resonance contributions; recent competing calculations still disagree with each other and also diverge from the measured neutron data at very low $Q^2$. Our proton results provide discriminating power between existing calculations, and will help provide a better understanding of this strong QCD regime.

Abstract

Pressurised metered dose inhalers (pMDI) are one of the most common devices to deliver therapeutic treatment to patients with asthma or COPD. The atomisation mechanism responsible for droplet production is poorly understood because of the short length and timescales involved, making experimental investigations to characterise fluid flow structures and spray formation processes difficult. This article reports the findings of new high-speed and high-resolution imaging and temperature measurements seeking to improve the fundamental understanding of the atomisation mechanism of the pMDI. An annular flow regime of gaseous core in liquid annulus was observed in the orifice. Shock diamonds within the flow, viewed via Schlieren imaging, confirmed experimentally for the first time the choked nature of the orifice exit flow. Propellant formulation flow in the orifice was found to be superheated to a degree that significant flashing and evaporation at the orifice exit is likely to take place. This information points to a hybrid mechanism of predominantly aerodynamic atomisation with significant liquid flashing upon exit from the orifice producing a near instantaneous formation of respirable droplets. Large irrespirable droplets are produced by two mechanisms; breakup of liquid slugs ejected from the orifice, due to unsteady transient fluid structures in the actuator sump and stripping of ligaments from a liquid pool around the orifice exit, which subsequently break up into large droplets in the periphery of the plume. This new understanding will aid development of next generation, high efficiency pMDIs, particularly those employing low global warming potential propellants such as HFA152a or HFO1234ze(E).

12. [Risk factors for prostate cancer in male patients with MRI-negative and PSA-abnormal findings]

Abstract

To investigate the risk factors for clinically significant PCa diagnosed by transrectal ultrasound-guided systematic prostate biopsy in patients with MRI-negative and PSA-abnormal findings.From January 2014 to December 2017, 335 male patients with MRI-negative (PI-RADS 2.0 score ≤ 2) and PSA-abnormal (4－30 ng/ml ) findings underwent systematic prostate biopsy guided by transrectal ultrasound under local anesthesia in our department. We collected and analyzed the demographic data, clinical symptoms, complications, past history and PSA density (PSAD) of the patients.Clinically significant PCa was diagnosed in 21 (6.3%) of the 335 patients. Multivariate logistic regression analysis showed that the independent risk factors were higher age (AUC: 0.704, P0.01) and PSAD (AUC: 0.743, P0.01). The cutoff values of age and PSAD were 71 years and 0.18 ng/ml/ml, respectively.Higher age and PSAD are risk factors for clinically significant PCa. Prostate biopsy, even repeated or saturated puncture, is recommended for those aged71 years old or with PSAD0.18 ng/ml/ml so as to avoid missed diagnosis and unnecessary invasive biopsy as well. /.

Abstract

Short-range correlations (SRCs) have been identified as being responsible for the high-momentum tail of the nucleon momentum distribution, n(k). Hard, short-range interactions of nucleon pairs generate the high-momentum tail and imprint a universal character on n(k) for all nuclei at large momentum. Triple coincidence experiments have shown a strong dominance of np pairs, but these measurements involve large final-state interactions. This paper presents the results from Jefferson Lab experiment E08014 which measured inclusive electron scattering cross section from Ca isotopes. By comparing the inclusive cross section from Ca48 to Ca40 in a kinematic region dominated by SRCs we provide a new way to study the isospin structure of SRCs.

Abstract

PADME (Positron Annihilation into Dark Matter Experiment) is a fixed target experiment located at the Beam Test Facility (BTF) at the Laboratori Nazionali di Frascati (LNF) designed to search for a massive dark photon A ′ in the process e + e − → γ A ′ , using a positron beam of energy up to 550 MeV. The experiment exploits the missing mass technique which allows for a search of A ′ in a model independent way. A sensitivity on the mixing constant ϵ > 10−3 for a dark photon mass in the range 1 ≤ m A ′ ≤ 23.7 MeV /c2 can be achieved by collecting 4 × 1013 positrons on target. Run 2 data taking finished in December 2020 and allowed to reach an integrated luminosity of 5 × 1012 positrons on target.

Abstract

Natural killer (NK) cells have an important role in immune defense against viruses and cancer. Activation of human NK cell cytotoxicity toward infected or tumor cells is regulated by killer cell immunoglobulin-like receptors (KIRs) that bind to human leukocyte antigen class I (HLA-I). Combinations of KIR with HLA-I are genetically associated with susceptibility to disease. KIR2DS4, an activating member of the KIR family with poorly defined ligands, is a receptor of unknown function. Here, we show that KIR2DS4 has a strong preference for rare peptides carrying a Trp at position 8 (p8) of 9-mer peptides bound to HLA-C*05:01. The complex of a peptide bound to HLA-C*05:01 with a Trp at p8 was sufficient for activation of primary KIR2DS4+ NK cells, independent of activation by other receptors and of prior NK cell licensing. HLA-C*05:01+ cells that expressed the peptide epitope triggered KIR2DS4+ NK cell degranulation. We show an inverse correlation of the worldwide allele frequency of functional KIR2DS4 with that of HLA-C*05:01, indicative of functional interaction and balancing selection. We found a highly conserved peptide sequence motif for HLA-C*05:01–restricted activation of human KIR2DS4+ NK cells in bacterial recombinase A (RecA). KIR2DS4+ NK cells were stimulated by RecA epitopes from multiple human pathogens, including Helicobacter, Chlamydia, Brucella, and Campylobacter. We predict that over 1,000 bacterial species could activate NK cells through KIR2DS4, and propose that human NK cells also contribute to immune defense against bacteria through recognition of a conserved RecA epitope presented by HLA-C*05:01.

Abstract

Experimental cross sections for the $^4He(e,e'p)X$ reaction up to a missing momentum of 0.632 GeV/$c$ at $x_B=1.24$ and $Q^2$=2(GeV/$c$)$^2$ are reported. The data are compared to Relativistic Distorted Wave Impulse Approximation(RDWIA) calculations for $^4He(e,e'p)^3H$ channel. Significantly more events in the triton mass region are measured for $p_{m}$$>$0.45 GeV/$c$ than are predicted by the theoretical model, suggesting that the effects of initial-state multi-nucleon correlations are stronger than expected by the RDWIA model.

Abstract

Due to the lack of free neutron targets, studies of the structure of the neutron are typically made by scattering electrons from either $^2$H or $^3$He targets. In order to extract useful neutron information from a $^3$He target, one must understand how the neutron in a $^3$He system differs from a free neutron by taking into account nuclear effects such as final state interactions and meson exchange currents. The target single spin asymmetry $A_y^0$ is an ideal probe of such effects, as any deviation from zero indicates effects beyond plane wave impulse approximation. New measurements of the target single spin asymmetry $A_y^0$ at $Q^2$ of 0.46 and 0.96 (GeV/$c)^2$ were made at Jefferson Lab using the quasi-elastic $^3\mathrm{He}^{\uparrow}(e,e'n)$ reaction. Our measured asymmetry decreases rapidly, from $>20\%$ at $Q^2=0.46$ (GeV/$c)^2$ to nearly zero at $Q^2=0.96$ (GeV$/c)^2$, demonstrating the fall-off of the reaction mechanism effects as $Q^2$ increases. We also observed a small $��$-dependent increase in $A_y^0$ compared to previous measurements, particularly at moderate $Q^2$. This indicates that upcoming high $Q^2$ measurements from the Jefferson Lab 12 GeV program can cleanly probe neutron structure from polarized $^3$He using plane wave impulse approximation., 6 pages, 5 figures

Abstract

The PADME experiment, hosted at the Laboratori Nazionali di Frascati in Italy, is dedicated to the dark photon search. It looks for the reaction $e^{+}\,e^{-}\rightarrow A'\,\gamma$, where $A'$ indicates the dark photon. The $550\,\text{MeV}$ positrons beam that impinges on electrons, coming from an active diamond target, allows to scan $A'$ masses up to $23.7\,\text{MeV}$. In this context, the segmented electromagnetic calorimeter plays a fundamental role, since it measures the final photon four-momentum. It consists of $616$ BGO crystals displaced in a cylindrical shape of $\approx29\,\text{cm}$ radius with a central square hole of $5$ crystals side, to let Bremsstrahlung radiation pass, to limit the calorimeter trigger rate. Each crystal is read by a HZC XP1911 type B photomultipliers and their signal is digitised by means of a CAEN V1742 board, taking $1024$ samples at $1\,\text{GS/s}$. 19. Search for three-nucleon short-range correlations in light nuclei

Abstract

We present new data probing short-range correlations (SRCs) in nuclei through the measurement of electron scattering off high-momentum nucleons in nuclei. The inclusive He4/He3 cross section ratio is observed to be both x and Q2 independent for 1.52, our data support the hypothesis that a previous claim of three-nucleon correlation dominance was an artifact caused by the limited resolution of the measurement. While 3N-SRCs appear to have an important contribution, our data show that isolating 3N-SRCs is significantly more complicated than for 2N-SRCs.

Abstract

We report on a precise measurement of double-polarization asymmetries in electron-induced breakup of $^3\mathrm{He}$ proceeding to $\mathrm{pd}$ and $\mathrm{ppn}$ final states, performed in quasi-elastic kinematics at $Q^2 = 0.25\,(\mathrm{GeV}/c)^2$ for missing momenta up to $250\,\mathrm{MeV}/c$. These observables represent highly sensitive tools to investigate the electromagnetic and spin structure of $^3\mathrm{He}$ and the relative importance of two- and three-body effects involved in the breakup reaction dynamics. The measured asymmetries cannot be satisfactorily reproduced by state-of-the-art calculations of $^3\mathrm{He}$ unless their three-body segment is adjusted, indicating that the spin-dependent part of the nuclear interaction governing the three-body breakup process is much smaller than previously thought., Comment: 5 pages, 3 figures, submitted to Phys. Abstract

We report measurements of target- and double-spin asymmetries for the exclusive channel $\vec e\vec p\to e��^+ (n)$ in the nucleon resonance region at Jefferson Lab using the CEBAF Large Acceptance Spectrometer (CLAS). These asymmetries were extracted from data obtained using a longitudinally polarized NH$_3$ target and a longitudinally polarized electron beam with energies 1.1, 1.3, 2.0, 2.3 and 3.0 GeV. The new results are consistent with previous CLAS publications but are extended to a low $Q^2$ range from $0.0065$ to $0.35$ (GeV$/c$)$^2$. The $Q^2$ access was made possible by a custom-built Cherenkov detector that allowed the detection of electrons for scattering angles as low as $6^\circ$. These results are compared with the unitary isobar models JANR and MAID, the partial-wave analysis prediction from SAID and the dynamic model DMT. In many kinematic regions our results, in particular results on the target asymmetry, help to constrain the polarization-dependent components of these models., 20 pages, 15 figures, to appear in Phys. Abstract

We present deeply virtual $��^0$ electroproduction cross-section measurements at $x_B$=0.36 and three different $Q^2$--values ranging from 1.5 to 2 GeV$^2$, obtained from experiment E07-007 that ran in the Hall A at Jefferson Lab. The Rosenbluth technique was used to separate the longitudinal and transverse responses. Results demonstrate that the cross section is dominated by its transverse component, and thus is far from the asymptotic limit predicted by perturbative Quantum Chromodynamics. An indication of a non-zero longitudinal contribution is provided by the interference term $��_{LT}$ also measured. Results are compared with several models based on the leading twist approach of Generalized Parton Distributions (GPDs). In particular, a fair agreement is obtained with models where the scattering amplitude is described by a convolution of chiral-odd (transversity) GPDs of the nucleon with the twist-3 pion distribution amplitude. Therefore, neutral pion electroproduction may offer the exciting possibility of accessing transversity GPDs through experiment., Submitted to Physical Review Letters

Abstract

We report the first measurement of the target single-spin asymmetry, $A_y$, in quasi-elastic scattering from the inclusive reaction $^3$He$^{\uparrow}(e,e^\prime)$ on a $^3$He gas target polarized normal to the lepton scattering plane. Assuming time-reversal invariance, this asymmetry is strictly zero for one-photon exchange. A non-zero $A_y$ can arise from the interference between the one- and two-photon exchange processes which is sensitive to the details of the sub-structure of the nucleon. An experiment recently completed at Jefferson Lab yielded asymmetries with high statistical precision at $Q^{2}=$ 0.13, 0.46 and 0.97 GeV$^{2}$. These measurements demonstrate, for the first time, that the $^3$He asymmetry is clearly non-zero and negative with a statistical significance of (8-10)$\sigma$. Using measured proton-to-$^{3}$He cross-section ratios and the effective polarization approximation, neutron asymmetries of $-$(1-3)% were obtained. The neutron asymmetry at high $Q^2$ is related to moments of the Generalized Parton Distributions (GPDs). Our measured neutron asymmetry at $Q^2=0.97$ GeV$^2$ agrees well with a prediction based on two-photon exchange using a GPD model and thus provides a new, independent constraint on these distributions.

Abstract

We report on new p$(e,e^\prime p)��^\circ$ measurements at the $��^{+}(1232)$ resonance at the low momentum transfer region. The mesonic cloud dynamics is predicted to be dominant and rapidly changing in this kinematic region offering a test bed for chiral effective field theory calculations. The new data explore the low $Q^2$ dependence of the resonant quadrupole amplitudes while extending the measurements of the Coulomb quadrupole amplitude to the lowest momentum transfer ever reached. The results disagree with predictions of constituent quark models and are in reasonable agreement with dynamical calculations that include pion cloud effects, chiral effective field theory and lattice calculations. The reported measurements suggest that improvement is required to the theoretical calculations and provide valuable input that will allow their refinements.

Abstract

We report the measurement of beam-target double-spin asymmetries ($A_\text{LT}$) in the inclusive production of identified hadrons, $\vec{e}~$+$~^3\text{He}^{\uparrow}\rightarrow h+X$, using a longitudinally polarized 5.9 GeV electron beam and a transversely polarized $^3\rm{He}$ target. Hadrons ($\pi^{\pm}$, $K^{\pm}$ and proton) were detected at 16$^{\circ}$ with an average momentum $$=2.35 GeV/c and a transverse momentum ($p_{T}$) coverage from 0.60 to 0.68 GeV/c. Asymmetries from the $^3\text{He}$ target were observed to be non-zero for $\pi^{\pm}$ production when the target was polarized transversely in the horizontal plane. Abstract

Improving the thermal stability of diamond and other superhard materials has great significance in various applications. Here, we report the synthesis and characterization of bulk diamond–cBN–B4C–Si composites sintered at high pressure and high temperature (HPHT, 5.2 GPa, 1620–1680 K for 3–5 min). The results show that the diamond, cBN, B4C, B x SiC, SiO2 and amorphous carbon or a little surplus Si are present in the sintered samples. The onset oxidation temperature of 1673 K in the as-synthesized sample is much higher than that of diamond, cBN, and B4C. The high thermal stability is ascribed to the covalent bonds of B–C, C–N, and the solid-solution of B x SiC formed during the sintering process. The results obtained in this work may be useful in preparing superhard materials with high thermal stability.

Abstract

We present a precise measurement of double-polarization asymmetries in the $^3\vec{\mathrm{He}}(\vec{\mathrm{e}},\mathrm{e}'\mathrm{d})$ reaction. This particular process is a uniquely sensitive probe of hadron dynamics in $^3\mathrm{He}$ and the structure of the underlying electromagnetic currents. The measurements have been performed in and around quasi-elastic kinematics at $Q^2 = 0.25\,(\mathrm{GeV}/c)^2$ for missing momenta up to $270\,\mathrm{MeV}/c$. The asymmetries are in fair agreement with the state-of-the-art calculations in terms of their functional dependencies on $p_\mathrm{m}$ and $\omega$, but are systematically offset. Beyond the region of the quasi-elastic peak, the discrepancies become even more pronounced. Thus, our measurements have been able to reveal deficiencies in the most sophisticated calculations of the three-body nuclear system, and indicate that further refinement in the treatment of their two- and/or three-body dynamics is required., Comment: 6 pages, 4 figures, accepted for publication at Phys. Abstract

We studied simultaneously the 4He(e,e'p), 4He(e,e'pp), and 4He(e,e'pn) reactions at Q^2=2 [GeV/c]2 and x_B>1, for a (e,e'p) missing-momentum range of 400 to 830 MeV/c. The knocked-out proton was detected in coincidence with a proton or neutron recoiling almost back to back to the missing momentum, leaving the residual A=2 system at low excitation energy. These data were used to identify two-nucleon short-range correlated pairs and to deduce their isospin structure as a function of missing momentum in a region where the nucleon-nucleon force is expected to change from predominantly tensor to repulsive. Neutron-proton pairs dominate the high-momentum tail of the nucleon momentum distributions, but their abundance is reduced as the nucleon momentum increases beyond ~500 MeV/c. The extracted fraction of proton-proton pairs is small and almost independent of the missing momentum in the range we studied. Abstract

A metaproteomic survey of surface coastal waters near Palmer Station on the Antarctic Peninsula, West Antarctica, was performed, revealing marked differences in the functional capacity of summer and winter communities of bacterioplankton. Proteins from Flavobacteria were more abundant in the summer metaproteome, whereas winter was characterized by proteins from ammonia-oxidizing Marine Group I Crenarchaeota. Proteins prevalent in both seasons were from SAR11 and Rhodobacterales clades of Alphaproteobacteria, as well as many lineages of Gammaproteobacteria. The metaproteome data were used to elucidate the main metabolic and energy generation pathways and transport processes occurring at the microbial level in each season. In summer, autotrophic carbon assimilation appears to be driven by oxygenic photoautotrophy, consistent with high light availability and intensity. In contrast, during the dark polar winter, the metaproteome supported the occurrence of chemolithoautotrophy via the 3-hydroxypropionate/4-hydroxybutyrate cycle and the reverse tricarboxylic acid cycle of ammonia-oxidizing archaea and nitrite-oxidizing bacteria, respectively. Proteins involved in nitrification were also detected in the metaproteome. Taurine appears to be an important source of carbon and nitrogen for heterotrophs (especially SAR11), with transporters and enzymes for taurine uptake and degradation abundant in the metaproteome. Divergent heterotrophic strategies for Alphaproteobacteria and Flavobacteria were indicated by the metaproteome data, with Alphaproteobacteria capturing (by high-affinity transport) and processing labile solutes, and Flavobacteria expressing outer membrane receptors for particle adhesion to facilitate the exploitation of non-labile substrates. TonB-dependent receptors from Gammaproteobacteria and Flavobacteria (particularly in summer) were abundant, indicating that scavenging of substrates was likely an important strategy for these clades of Southern Ocean bacteria. This study provides the first insight into differences in functional processes occurring between summer and winter microbial communities in coastal Antarctic waters, and particularly highlights the important role that dark carbon fixation has in winter. © 2012 International Society for Microbial Ecology. Abstract

Heterotrophic marine bacteria play key roles in remineralizing organic matter generated from primary production. However, far more is known about which groups are dominant than about the cellular processes they perform in order to become dominant. In the Southern Ocean, eukaryotic phytoplankton are the dominant primary producers. In this study we used metagenomics and metaproteomics to determine how the dominant bacterial and archaeal plankton processed bloom material. We examined the microbial community composition in 14 metagenomes and found that the relative abundance of Flavobacteria (dominated by Polaribacter) was positively correlated with chlorophyll a fluorescence, and the relative abundance of SAR11 was inversely correlated with both fluorescence and Flavobacteria abundance. By performing metaproteomics on the sample with the highest relative abundance of Flavobacteria (Newcomb Bay, East Antarctica) we defined how Flavobacteria attach to and degrade diverse complex organic material, how they make labile compounds available to Alphaproteobacteria (especially SAR11) and Gammaproteobacteria, and how these heterotrophic Proteobacteria target and utilize these nutrients. The presence of methylotrophic proteins for archaea and bacteria also indicated the importance of metabolic specialists. Overall, the study provides functional data for the microbial mechanisms of nutrient cycling at the surface of the coastal Southern Ocean. © 2012 Society for Applied Microbiology and Blackwell Publishing Ltd.

33. Two processing pathways for the MHC class II-restricted presentation of exogenous influenza virus antigen

Abstract

The natural Ag influenza virus A was used to test the requirements for the HLA-DR1-restricted presentation of the epitopes 18-29 in the matrix protein and 307-318 in the hemagglutinin protein. CD4+ cytotoxic T cell clones of similar efficiency were used to detect presentation of these two epitopes. Presentation of the matrix epitope by APC pulsed with either inactivated virus particles or purified soluble protein followed the classical pathway in that 1) it required invariant chain expression, 2) it was blocked by inhibition of protein synthesis, and 3) it was dependent on a function(s) encoded in the MHC class II region. These characteristics suggest that peptides corresponding to the matrix epitope can only load onto newly synthesized class II molecules that were targeted to a processing compartment by the invariant chain. In contrast, presentation of the hemagglutinin epitope processed from virus particles followed a different pathway. First, presentation of hemagglutinin was independent of invariant chain expression. Second, a human B lymphoblastoid cell line in which protein synthesis was inhibited for 9 h was still able to present hemagglutinin even at very low doses of Ag. Third, a DR1-transfected mutant B cell line missing the MHC class II region was able to present hemagglutinin. Thus, mature class II alpha beta molecules can acquire immunogenic peptides derived from intact natural Ags for presentation to CD4+ T cells. This pathway may be useful for the binding of peptides derived from Ags that are rapidly degraded upon uptake into APC.

Abstract

In this paper, we tested a novel methodology of measuring critical dimension (CD) uniformity, or CDU, with electron beam (e-beam) hotspot inspection and measurement systems developed by Hermes Microvision, Inc. (HMI). The systems were used to take images of two-dimensional (2D) array patterns and measure CDU values in a custom designated fashion. Because this methodology combined imaging of scanning micro scope (SEM) and CD value averaging over a large array pattern of optical CD, or OCD, it can measure CDU of 2D arrays with high accuracy, high repeatability and high throughput.

Abstract

We present a search at Jefferson Laboratory for new forces mediated by sub-GeV vector bosons with weak coupling $\alpha'$ to electrons. Such a particle $A'$ can be produced in electron-nucleus fixed-target scattering and then decay to an $e^+e^-$ pair, producing a narrow resonance in the QED trident spectrum. Using APEX test run data, we searched in the mass range 175--250 MeV, found no evidence for an $A'\to e^+e^-$ reaction, and set an upper limit of $\alpha'/\alpha \simeq 10^{-6}$. Abstract

The MHC class I-specific inhibitory receptors on human and mouse NK cells have surprisingly different structures. The mouse receptors (Ly-49) are type II transmembrane glycoproteins of the C-type lectin family, whereas the human receptors (killer cell inhibitory receptors (KIR)) belong to the Ig superfamily. This difference prompted a search for Ig-like inhibitory receptors in mice. Here we show that gp49, a mouse mast cell protein of unknown function but with sequence similarity to KIR, is expressed in NK cells. The gp49 cytoplasmic tail, containing a sequence related to an inhibitory motif shared by KIR and Ly-49, delivered a strong inhibitory signal in both human and mouse NK cells when substituted for a KIR cytoplasmic tail. These data show that Ig-like receptors with inhibitory properties exist in both species and that mouse mast and NK cells may recognize common inhibitory ligands.

Abstract

The processing pathway for the MHC class II-restricted presentation of endogenous cytosolic Ag is distinct from the class I pathway since a cytosolic form of the influenza virus A hemagglutinin, expressed by a recombinant vaccinia virus, was presented by HLA-DR in a B cell mutant lacking the TAP1 subunit of the transporter for Ag presentation (TAP). In this report, two additional B cell mutants have been used to define the requirements of this TAP1-independent processing pathway. The first mutant, .61, lacks expression of both TAP1 and TAP2 genes, and of both LMP2 and LMP7 genes encoding proteasome subunits. As expected, class I-restricted presentation of the influenza virus matrix protein was totally deficient in mutant .61. In contrast, class II-restricted presentation of both the natural cytosolic matrix and the engineered cytosolic hemagglutinin proteins was functional in mutant .61. Thus, presentation of cytosolic Ag by class II molecules is independent of both TAP subunits and of the two MHC-encoded proteasome subunits. However, this endogenous processing pathway is dependent on at least one other function encoded in the class II region of the MHC as demonstrated with the second mutant, .174, in which a large deletion eliminates all expressed class II genes. Mutant .174 transfected with HLA-DR1 genes was previously shown to be defective in the presentation of exogenous Ag but normal in the presentation of short exogenous peptides. We show here that .174(DR1) is also defective in the presentation of cytosolic matrix and hemagglutinin proteins. This similar requirement for the class II-restricted presentation of either cytosolic Ag or internalized exogenous Ag suggests that both forms of Ag are ultimately targeted to the same cellular compartment for association with class II molecules.

Abstract

NK clones that were inhibited by target cell expression of HLA-B*2705 displayed peptide-specific recognition of HLA-B*2705. To evaluate the specificity of this recognition, synthetic versions of 14 endogenous ligands of HLA-B*2705 were tested for their ability to provide protection from NK-mediated lysis by binding to surface HLA-B*2705 molecules on RMA-S cells deficient in the transporter for Ag presentation. Several unrelated peptides inhibited lysis by the same NK clones. Despite similar capacities to stabilize HLA-B*2705 molecules on RMA-S cells, the 14 peptides differed widely in their abilities to provide protection. Single amino acid substitutions in both a protective and a nonprotective peptide revealed the importance of residues 7 and 8 in the peptide for recognition by NK clones, thus localizing the peptide influence to a polymorphic region of the alpha-helix of HLA class I molecules known to control discrimination among allelic variants of HLA-B and HLA-C by NK cells.

Abstract

The recently approved Electron-Ion Collider (EIC) will provide a unique new opportunity for searches of charged lepton flavor violation (CLFV) and other new physics scenarios. In contrast to the $e \leftrightarrow \mu$ CLFV transition for which very stringent limits exist, there is still a relatively large discovery space for the $e \to \tau$ CLFV transition, potentially to be explored by the EIC. With the latest detector design of ECCE (EIC Comprehensive Chromodynamics Experiment) and projected integral luminosity of the EIC, we find the $\tau$-leptons created in the DIS process $ep\to \tau X$ are expected to be identified with high efficiency. A first ECCE simulation study, restricted to the 3-prong $\tau$-decay mode and with limited statistics for the Standard Model backgrounds, estimates that the EIC will be able to improve the current exclusion limit on $e\to \tau$ CLFV by an order of magnitude., Comment: 11 pages, 8 figures, to be submitted to NIM
