1. Iteratively enhanced semidefinite relaxations for efficient neural network Verification

Abstract: We propose an enhanced semidefinite program (SDP) relaxation to enable the tight and efficient verification of neural networks (NNs). The tightness improvement is achieved by introducing a nonlinear constraint to existing SDP relaxations previously proposed for NN verification. The efficiency of the proposal stems from the iterative nature of the proposed algorithm in that it solves the resulting non-convex SDP by recursively solving auxiliary convex layer-based SDP problems. We show formally that the the solution generated by our algorithm is tighter than state-of-the-art SDP-based solutions for the problem. We also show that the solution sequence converges to the optimal solution of the non-convex enhanced SDP relaxation. The experimental results on standard benchmarks in the area show that our algorithm achieves the state-of-the-art performance whilst maintaining an acceptable computational cost.

2. A semidefinite relaxation based branch-and-bound method for tight neural network verification

Abstract: We introduce a novel method based on semidefinite program (SDP) for the tight and efficient verification of neural networks. The proposed SDP relaxation advances the present SoA in SDP-based neural network verification by adding a set of linear constraints based on eigenvectors. We extend this novel SDP relaxation by combining it with a branch-and-bound method that can provably close the relaxation gap up to zero. We show formally that the proposed approach leads to a provably tighter solution than the present SoA. We report experimental results showing that the proposed method outperforms baselines in terms of verified accuracy while retaining an acceptable computational overhead.

Abstract: To date, results have been inconsistent in whether mothers show higher parental sensitivity to their infant than fathers do. The context in which sensitivity is measured may play a role in these inconsistent findings, but this has not been examined yet. The aim of the current study was to test context as a source of variability in parental sensitivity, comparing maternal and paternal sensitivity to infants in four different observational settings. Participants included 109 families with their 4-month-old infants. Parental sensitivity was observed during a routine caregiving session, free play episode, and the baseline and reunion of the Still Face Procedure. Results demonstrated that parental sensitivity showed weak to strong stability (correlations) across the four contexts. Furthermore, overall levels of parental sensitivity were higher in more naturalistic contexts (routine caregiving > free play > Still Face). Lastly, mothers and fathers were overall equally sensitive across contexts. Our findings highlight the importance of taking context into account when observing parental sensitivity in research as well as practice. 6. Narrative coherence across the transition to parenthood: Primiparous parents' representations of their child in relation to parental sensitivity

Abstract: Narrative coherence reflects parents' ability to provide a believable, clear, relevant, and internally consistent story about their child. Parents demonstrating more narrative coherence have been theorized to show higher parental sensitivity, but this has not been examined in a normative sample, nor across the transition to parenthood, and only once in fathers. The aim of this study was to examine stability and change in narrative coherence across the transition to parenthood in mothers and fathers, as well as the relation between pre- and postnatal narrative coherence and postnatal parental sensitivity. The sample consisted of 105 primiparous expecting parents. Narrative coherence was measured at 36-weeks pregnancy and when the child was 4 months old, using the Five Minute Speech Sample procedure. Parental sensitivity was observed in three episodes. Results demonstrated that narrative coherence was moderately stable (correlations) across the transition to parenthood in fathers only. Both mothers' and fathers' narrative coherence improved over time. Furthermore, mothers and fathers were overall equally coherent, and maternal and paternal narrative coherence were positively interrelated during pregnancy only. Lastly, our findings showed weak evidence for the theorized link between narrative coherence and parental sensitivity: only postnatal narrative coherence predicted paternal sensitivity, only during free play. Our findings give new insight in the development of narrative coherence across the transition to parenthood, and how it relates to actual parenting. More research is needed to confirm our findings and further explore this topic.

7. Natural killer cells inhibit sepsis caused by feces-induced acute peritonitis via regulation of angiogenesis

Abstract: Sepsis is associated with acute peritonitis, which can be induced by lipopolysaccharide (LPS) exposure and feces. Generally, LPS induces mono-microbial peritonitis, whereas feces cause poly-microbial peritonitis; the latter is a more complicated and closer to the clinical diseases. However, the immune responses to sepsis caused by feces-induced acute peritonitis have not been reported. In particular, the roles of natural killer (NK) cells, which generally have pro-angiogenic effects, on feces-induced sepsis are still unclear. Accordingly, in this study, we assessed the roles of NK cells in an acute sepsis model in mice. NK cells (108/kg) were injected via the tail vein into mice with acute sepsis, and nitric oxide (NO), inflammatory factors, and angiogenic factors were tested to explore the effects of NK cells on sepsis. In mice with sepsis, we observed a significant increase in survival in mice after the transfusion of NK cells. Interestingly, the levels of NO, interleukin-10, and vascular endothelial growth factor (VEGF) decreased. Consistent with our hypothesis, the transfusion of NK cells into mice with sepsis blocked inflammation and promoted angiogenesis. Overall, these findings suggest that NK cells may block sepsis by modulating the VEGF pathway.

8. The role of infant attention and parental sensitivity in infant cognitive development in the Netherlands and China

Abstract: Infant attention and parental sensitivity are important predictors of later child executive function (EF). However, most studies have investigated infant and parent factors in relation to child EF separately and included only mothers from Western samples. The current study examined whether both infant attention at 4 months and parental sensitivity at 4 and 14 months were related to infant EF (i.e., inhibition, working memory, and cognitive flexibility) at 14 months among 124 Dutch and 63 Chinese first-time mothers and fathers and their infants. Findings revealed that parental sensitivity at 4 months was not correlated with infant EF abilities at 14 months. However, infant attention at 4 months was significantly related to 14-month working memory, but not to inhibition and cognitive flexibility. Maternal sensitivity at 14 months was significantly related to 14-month inhibition, but not to working memory and cognitive flexibility. No country differences were found in the relation among 4-month infant attention, parental sensitivity, and EF outcomes. Results show that both infant and parent factors are associated with early EF development and that these correlates of early EF skills may be similar in Western and non-Western samples.

Abstract: Background and objectives: Long-term peritoneal dialysis (PD) is accompanied by low-grade intraperitoneal inflammation, may eventually lead to peritoneal membrane injury with high solute transport rate and ultrafiltration failure. Osteopontin(OPN) is highly expressed with the pro-inflammatory cytokines stimulation in many cell types, and evolves in the process of tissue fibrosis. This study aimed to investigate the potential of OPN as a new indicator of peritoneal injury. Methods: We analyzed a total of 125 PD patients with end-stage renal disease, including 16 patients with continuous ambulatory PD(CAPD)-related peritonitis and 109 patients without peritonitis in a single renal center. The OPN levels in the overnight peritoneal effluents or in serum were analyzed using ELISA. In HMrSV5 cells, The OPN and fibronectin(FN) protein expression were identified using western blot analysis. Results: The OPN levels in overnight drained dialysate were significantly correlated with D/P Cr (P < 0.0001, R =0.54) and D/D0 glucose (P < 0.0001 R=-0.39). Logistical regression analysis showed that the OPN levels in peritoneal effluents was an independent predictive factor for the increased peritoneal solute transport rate (PSTR) (p < 0.001). The area under the receiver operating characteristic (ROC) curve of the OPN-PSTR model for identifying PSTR was 0.88, with 95% confidence interval (CI):0.81-0.95. Abstract: We have conducted three searches for correlations between ultra-high energy cosmic rays detected by the Telescope Array and the Pierre Auger Observatory, and high-energy neutrino candidate events from IceCube. Two cross-correlation analyses with UHECRs are done: one with 39 cascades from the IceCube ‘high-energy starting events’ sample and the other with 16 high-energy ‘track events’. The angular separation between the arrival directions of neutrinos and UHECRs is scanned over. The same events are also used in a separate search using a maximum likelihood approach, after the neutrino arrival directions are stacked. To estimate the significance we assume UHECR magnetic deflections to be inversely proportional to their energy, with values 3◦ , 6◦ and 9 ◦ at 100 EeV to allow for the uncertainties on the magnetic field strength and UHECR charge. A similar analysis is performed on stacked UHECR arrival directions and the IceCube sample of through-going muon track events which were optimized for neutrino point-source searches.

Abstract: Most still‐face paradigm (SFP) studies have been done in Western families with infant–mother dyads. The present study investigated the SFP pattern in 123 Dutch and 63 Chinese 4‐month‐old infants with mothers and fathers. The classic SFP effect was found for positive affect and gaze in both countries. For negative affect, Chinese infants showed a different SFP pattern than Dutch infants. With fathers, in-fants displayed a less pronounced SFP pattern for positive affect and an increase from the still face to the reunion for negative affect. Only a minority of infants showed the ex-pected SFP pattern across episodes. Our findings support that infant emotion expression is influenced by parent gen-der and cultural context. An interesting avenue for further study is the exploration of the origins of within‐ and be-tween‐gender and culture differences in affective communi-cation between parents and infants.

Abstract: Background: definitions of child maltreatment vary widely between studies, and even more so between different cultural contexts. Objective: in this pilot study, we examine between-country variations in maternal notions about what constitutes child maltreatment. Participants and setting: the sample consisted of 466 mothers recruited in Chile, China, Greece, Iran, the Netherlands, Portugal, South Africa, Turkey, and Uruguay. Methods: all mothers completed a new Q-sort measure, ranking 90 parenting behaviors linked to subtypes of maltreatment (emotional neglect, emotional abuse, physical neglect, and physical abuse) from least to most detrimental to child development. Results: between-country agreement regarding the harmfulness of the parenting behaviors was high (r=.45), but there were different patterns of reported harmfulness of subtypes of maltreatment (although driven mostly by deviating patterns in the South African sample). Further, there were significant country effects on the number and type of behaviors labeled as maltreatment (pre=.15), and the number of items labeled as requiring intervention (pre=.19). Abstract: Background: Chronic kidney disease (CKD) is a major global public health challenge. We investigated determinants of CKD and their clinical utility in an ethnically diverse Southeast Asian population. Methods: Electronic health records (EHR) of adults ≥40 years who visited any one of 4 government polyclinics in Singapore from January 1, 2012 to December 31, 2015 were analyzed. CKD was defined as an estimated glomerular filtration rate 2 or 1+ dipstick proteinuria excretion, based on 2 measurements ≥3 months apart. CKD-associated factors and their clinical utility for predicting odds of CKD were investigated using multiple logistic regression analysis. Results: Based on the study criteria, 25.9% (95% CI 25.6–26.2) of the 88,765 eligible study individuals had CKD. The factors (OR and 95% CI) independently associated with CKD were older age ≥65 years (2.54 [2.44–2.64] vs. ≤65 years), respectively; men (1.13 [1.09–1.18]); Malay (1.27 [1.20–1.33]) and Indian (0.77 [0.71–0.83]) vs. Chinese ethnicity; overweight (body mass index [BMI] ≥27.5 kg/m2; 1.10 [1.04–1.16]) vs. normal weight (BMI 18 to 2); government (1.22 [1.15–1.31]) vs. private housing; and with hypertension (3.32 [3.09–3.56]), diabetes (6.93 [6.67–7.20]) or stroke (1.46 [1.36–1.56]) vs. without each co-morbidity, respectively. The area under the receiver operating characteristic curve (95% CI) for the model to predict the probability of CKD using hypertension, diabetes, and age was 0.808 (0.805–0.811). Only 28.5% (27.9–29.1%) of individuals with CKD had physician documentation of their CKD status. However, documentation of CKD status was associated with age ≥65 years (1.11 [1.04–1.20] vs. Conclusions: CKD prevalence in our Southeast Asian population is high and under-documented even in high-risk patients. Our findings highlight factors associated with CKD, and the predictive value of hypertension, diabetes, and advancing age as EHR-based screening targets for CKD. Our results also suggest that complementary educational efforts will be needed to increase physician detection and optimize the management of CKD, especially in high risk and marginalized groups across all clinics in Singapore, and possibly in the region.

Abstract: The Telescope Array (TA) experiment is located in the western desert of Utah, USA, and observes ultra high energy cosmic rays (UHECRs) in the Northern hemisphere. At the highest energies, $E>10$~EeV, the shape of cosmic ray energy spectrum may carry an imprint of the source density distribution along the line of sight different in different directions of the sky. In this study, we search for such directional variations in the shape of the energy spectrum using events observed with the Telescope Array's surface detector. We divide the TA field of view into two nearly equal-exposure regions: the "on-source" region which we define as $\pm 30^\circ$ of the supergalactic plane containing mostly nearby structures, and the complementary "off-source" region where the sources are further away on average. We compare the UHECR spectra in these regions by fitting them to the broken power law and comparing the resulting parameters. We find that the off-source spectrum has an earlier break at highest energies. The chance probability to obtain such or larger difference in statistically equivalent distributions is estimated as $6.2\pm1.1\times10^{-4}$ ($3.2\sigma$) by a Monte-Carlo simulation. The observed difference in spectra is in a reasonable quantitative agreement with a simplified model that assumes that the UHECR sources trace the galaxy distribution from the 2MRS catalogue, primary particles are protons and the magnetic deflections can be neglected., Comment: 17 pages 6 figures, submitted to Physical Review Letters, (31.May.2017)

Abstract: In this work we are reporting on the measurement of the proton-air inelastic cross section $\sigma^{\rm inel}_{\rm p-air}$ using the Telescope Array (TA) detector. Based on the measurement of the $\sigma^{\rm inel}_{\rm p-air}$ the proton-proton cross section $\sigma_{\rm p-p}$ value is also determined at $\sqrt{s} = 95_{-8}^{+5}$ TeV. Detecting cosmic ray events at ultra high energies with Telescope Array enables us to study this fundamental parameter that we are otherwise unable to access with particle accelerators. The data used in this report is the hybrid events observed by the Middle Drum fluorescence detector together with the surface array detector collected over five years. The value of the $\sigma^{\rm inel}_{\rm p-air}$ is found to be equal to $567.0 \pm 70.5 [{\rm Stat.}] ^{+29}_{-25} [{\rm Sys.}]$ mb. The total proton-proton cross section is subsequently inferred from Glauber Formalism and Block, Halzen and Stanev QCD inspired fit and is found to be equal to $170_{-44}^{+48} [{\rm Stat.}] _{-17}^{+19} [{\rm Sys.}] $mb.

Abstract: Adenosine A2A receptors (A2AR) are located postsynaptically in striatopallidal GABAergic neurons, antagonizing dopamine D2 receptor functions, and are also located presynaptically at corticostriatal terminals, facilitating glutamate release. To address the hypothesis that these two A2AR populations differently control the action of psychostimulants, we characterized A2AR modulation of cocaine-induced effects at the level of DARPP-32 phosphorylation at Thr-34 and Thr-75, c-Fos expression, and psychomotor activity using two lines of cell-type selective A2AR knockout (KO) mice with selective A2AR deletion in GABAergic neurons (striatum-A2AR-KO mice), or with A 2AR deletion in both striatal GABAergic neurons and projecting cortical glutamatergic neurons (forebrain-A2AR-KO mice). We demonstrated that striatum-A2AR KO mice lacked A2ARs exclusively in striatal GABAergic terminals whereas forebrain-A2AR KO mice lacked A2ARs in both striatal GABAergic and glutamatergic terminals leading to a blunted A2AR-mediated facilitation of synaptosomal glutamate release. The inactivation of A2ARs in GABAergic neurons reduced striatal DARPP-32 phosphorylation at Thr-34 and increased its phosphorylation at Thr-75. Conversely, the additional deletion of corticostriatal glutamatergic A2ARs produced opposite effects on DARPP-32 phosphorylation at Thr-34 and Thr-75. This distinct modulation of DARPP-32 phosphorylation was associated with opposite responses to cocaine-induced striatal c-Fos expression and psychomotor activity in striatum-A2AR KO (enhanced) and forebrain-A2AR KO mice (reduced). Thus, A2ARs in glutamatergic corticostriatal terminals and in GABAergic striatal neurons modulate the action of psychostimulants and DARPP-32 phosphorylation in opposite ways. We conclude that A2ARs in glutamatergic terminals prominently control the action of psychostimulants and define a novel mechanism by which A2ARs fine-tune striatal activity by integrating GABAergic, dopaminergic and glutamatergic signaling. © 2013 Shen et al.

Abstract: In a phase II trial, the activity of EO9, a new bioreductive alkylating agent, was assessed. EO9 was used as second-line chemotherapy in breast cancer patients and as first-line chemotherapy for patients with gastric, pancreatic and colorectal cancer. EO9 was given as a 5 min i.v. infusion at a weekly dose of 12 mg/m(2). 92 patients were entered; 22 with breast cancer, 26 with colon cancer, 24 with pancreatic cancer and 20 with gastric cancer. In general, the drug was well tolerated with nausea and vomiting occurring in 26.42 and 13.3% of courses, respectively. Reversible proteinuria was the main toxicity occurring in 45% of courses. Antitumour activity was not observed. At this dose and schedule, EO9 is not an active drug in the type of tumour studied. Copyright (C) 1996 Elsevier Science Ltd

Abstract: A rumen simulation technique (Rusitec) apparatus with eight 800 ml fermentation vessels was used to investigate the effects of rumen degradable protein (RDP) level and non-fibre carbohydrate (NFC) type on ruminal fermentation, microbial growth, and populations of ruminal cellulolytic bacteria. Treatments consisted of two NFC types (starch and pectin) supplemented with 0 g/d (low RDP) or 1.56 g/d (high RDP) sodium caseinate. Abstract: Antibodies are a powerful tool for structure/function studies of platelet proteins. However, classic immunisation frequently elicits antibody responses against domains of minor functional interest. Robust strategies to generate antibodies against defined domains would be of significant interest in post-genome research. In this study, we report a new strategy using a combination of DNA vaccination and V gene phage display that allows the rapid generation of domain specific single-chain Fv antibodies (scFvs). This system was validated using the I-domain of alpha2 integrin as a model. The alpha2beta1 integrin, which is expressed on many cell types, is the dominant collagen attachment receptor on platelets, functioning in close interplay with the collagen signalling receptor glycoprotein VI. A novel set of I-domain specific antibodies was obtained by a DNA vaccination/V gene repertoire cloning approach. Mice were first immunised with a DNA vaccine in which the alpha2 I-domain is expressed as a fusion protein with fragment C of tetanus toxoid (FrC-TT). Then the heavy and kappa light chain variable gene repertoires were rescued from immune splenocytes using antibody phage display. A total of four alpha2 I-domain specific scFvs were isolated by selection on recombinant I-domain or native platelet alpha2beta1 integrin. Characterisation of the scFvs indicated that they recognised distinct epitopes that had profound differences in accessibility between native and recombinant I-domain. Our data suggest DNA immunisation and phage display represent versatile alternatives to protein immunisation and hybridoma-fusion techniques for the isolation of recombinant antibody reagents. This approach will be particularly useful for the generation of domain or splice-variant specific antibodies that recognise native protein.

Abstract: In a multicentre randomized trial of the EORTC-ECSG, we have treated 38 chemotherapy naive patients with advanced non-small-cell lung cancer (NSCLC) with EO9, a novel bioreductive alkylating indoloquinone. The drug was given at two different dose schedules by a single bolus i.v. injection: arm A 12 mg/m2 weekly and arm B 22 mg/m2 every three weeks. All together 185 courses were administered (145 in arm A and 40 in arm B). The major toxicity was reversible proteinuria more frequently occurring in the three-weekly schedule (arm A 34.5% vs. arm B 62.5%). Creatinine elevation, fluid retention and pericardial or pleural effusion were also recorded in a limited number of patients. Other common toxicities more frequent in the three-weekly administration were asthenia (21% vs. 35% of cycles), nausea (15% vs. 27.5% of cycles) and Vomiting (5% vs. 17.5% of cycles). Toxicities were mainly of grade I and II. No responses have been observed. Five patients (26%) on arm A and eight (53%) on arm B experienced stable disease. Abstract: Total elbow arthroplasty has become a relatively common procedure in the last decade, and the number of primary total elbow replacements performed is likely to continue to increase as the population ages. The incidence of technically demanding prosthesis revisions involving complex problems such as major bone loss is therefore expected to increase. We report 3 cases of total elbow revision arthroplasty, all of which represented patients with severe bone loss. They were all treated using the impaction allografting technique and uncemented custom-made semiconstrained prostheses partially coated with hydroxyapatite manufactured at the bioengineering department of our institution.

Abstract: The present study was designed to investigate the perceptions of caring among student nurses and how these develop throughout the course of a programme of pre-registration nurse education. A 35-item version of the caring dimensions inventory was administered to a cohort of nursing students in a department of nursing in Scotland at entry to the programme, after 12 months and after 24 months on the programme. Caring was largely perceived through a technical dimension, demonstrated by factor analysis, but other dimensions such as intimacy, support and unnecessary and inappropriate aspects of nursing also became apparent as students progressed through the programme.

Abstract: Mild hypothermia, applied either during or soon after cerebral ischemia, has been shown to confer robust neuroprotection against brain injury in experimental stroke and in patients recovering from cardiac arrest. However, the mechanism underlying hypothermic neuroprotection is not completely understood. In this study, the effect of mild hypothermia on the induction of oxidative DNA damage, an early harmful event during post-ischemic reperfusion that triggers both necrotic and apoptotic cell death in the brain, was studied using the rat model of middle cerebral artery occlusion (MCAO) and reperfusion. Rats were subjected to 2-hr MCAO and reperfusion of various durations up to 3 days. Selective brain hypothermia (33 degrees C) was induced at the onset of ischemia and terminated at the beginning of reperfusion, and this significantly decreased infarct volume 72 hr later. Correlated with this protective effect, intraischemic mild hypothermia markedly attenuated the nuclear accumulations of several oxidative DNA lesions, including 8-oxodG, AP sites, and DNA single-strand breaks, after 2-hr MCAO. Consequently, harmful DNA damage-dependent signaling events, including NAD depletion, p53 activation, and mitochondrial translocation of PUMA and NOXA, were reduced during post-ischemic reperfusion in hypothermia-treated brains. These results suggest that the attenuation of oxidative DNA damage and DNA damage-triggered pro-death signaling events may be an important mechanism underlying the neuroprotective effect of mild hypothermia against ischemic brain injury.

Abstract: Computerized tomographic dimensions of the foramen magnum of 63 achondroplastic individuals were compared to standards established for nonachondroplastic individuals. The size of the foramen magnum in patients with achondroplasia was small at all ages, particularly in those with serious neurologic problems. The data suggest that measurement of the foramen magnum may identify achondroplastic individuals at high risk of developing neurologic complications.

Abstract: We have conducted three searches for correlations between ultra-high energy cosmic rays detected by the Telescope Array and the Pierre Auger Observatory, and high-energy neutrino candidate events from IceCube. Two cross-correlation analyses with UHECRs are done: one with 39 cascades from the IceCube `high-energy starting events' sample and the other with 16 high-energy `track events'. The angular separation between the arrival directions of neutrinos and UHECRs is scanned over. The same events are also used in a separate search using a maximum likelihood approach, after the neutrino arrival directions are stacked. To estimate the significance we assume UHECR magnetic deflections to be inversely proportional to their energy, with values $3^\circ$, $6^\circ$ and $9^\circ$ at 100 EeV to allow for the uncertainties on the magnetic field strength and UHECR charge. Abstract: We use the multiscale simulation approach, which combines the first-principles calculations and grand canonical Monte Carlo simulations, to comprehensively study the doping of a series of alkali (Li, Na, and K), alkaline-earth (Be, Mg, and Ca), and transition (Sc and Ti) metals in nanoporous covalent organic frameworks (COFs), and the effects of the doped metals on CO2 capture. The results indicate that, among all the metals studied, Li, Sc, and Ti can bind with COFs stably, while Be, Mg, and Ca cannot, because the binding of Be, Mg, and Ca with COFs is very weak. Furthermore, Li, Sc, and Ti can improve the uptakes of CO2 in COFs significantly. However, the binding energy of a CO2 molecule with Sc and Ti exceeds the lower limit of chemisorptions and, thus, suffers from the difficulty of desorption. By the comparative studies above, it is found that Li is the best surface modifier of COFs for CO2 capture among all the metals studied. Therefore, we further investigate the uptakes of CO2 in the Li-doped COFs. Our simulation results show that at 298 K and 1 bar, the excess CO2 uptakes of the Li-doped COF-102 and COF-105 reach 409 and 344 mg/g, which are about eight and four times those in the nondoped ones, respectively. As the pressure increases to 40 bar, the CO2 uptakes of the Li-doped COF-102 and COF-105 reach 1349 and 2266 mg/g at 298 K, respectively, which are among the reported highest scores to date. In summary, doping of metals in porous COFs provides an efficient approach for enhancing CO2 capture. © 2010 American Chemical Society.

44. Identification, replication and characterization of epigenetic remodelling in the aging genome

Abstract: Background: Aging is a complex biological process that involves numerous changes at various levels through remodelling of multiple biological processes and regulatory mechanisms including epigenetics. Recent analysis of the DNA methylome has reported large numbers of epigenetic markers associated with human aging. Validation and characterization of the significant findings are necessary both for obtaining reliable results and for better describing the age-associated epigenetic modifications. Methods: Based on genome-wide DNA methylation data measured in a large collection of older individuals in the Lothian Birth Cohorts of 1921 and 1936, we performed an association study to identify age-associated CpGs and replicated the findings in two independent cohorts of Danish twins. The double-replicated CpGs were characterized by distribution over gene regions and by their location in relation to CpG islands conditional on their age-dependent patterns of methylation status. The replicated CpGs were further characterized by their involvement in gene-sets or canonical pathways to study their functional implications. Abstract: Dendritic cells (DCs) are the most potent antigen-presenting cells able to trigger the adaptive immune response to specific antigens. When non-self-antigens are captured, DCs switch from an "immature" to a "mature" state to fulfill their function. Among the several surface proteins involved in DCs maturation, the role of aquaporins (AQPs) is still poorly understood. Here we investigated the expression profile of Aqps in murine bone marrow derived dendritic cells (BMDCs). Among the Aqps analyzed, Aqp9 was the most expressed by DCs. Its expression level was significantly upregulated 6 h following LPS exposure. Chemical inhibition of Aqp9 led to a decreased inflammatory cytokines secretion. BMDCs from AQP9-KO mice release lower amount of inflammatory cytokines and chemokines and increased release of IL-10. Despite the reduced release of inflammatory cytokines, Aqp9-KO mice were not protected from DSS induced colitis. All together, our data indicate that AQP9 blockade can be an efficient strategy to reduce DCs inflammatory response but it is not sufficient to protect from acute inflammatory insults such as DSS induced colitis.

Abstract: Using a focal cerebral ischemia model in rats, brain ischemia-induced changes in expression levels of mRNA and protein, and activities of proprotein convertase 2 (PC2) in the cortex were examined. In situ hybridization analyses revealed a transient upregulation of the mRNA level for PC2 at an early reperfusion hour, at which the level of PC2 protein was also high as determined by immunocytochemistry and western blotting. When enzymatic activities of PC2 were analyzed using a synthetic substrate, a significant decrease was observed at early reperfusion hours at which levels of PC2 protein were still high. Also decreased at these reperfusion hours were tissue levels of dynorphin-A(1-8) (DYN-A(1-8)), a PC2 substrate, as determined by radioimmunoassay. Further examination of PC2 protein biosynthesis by metabolic labeling in cultured neuronal cells showed that in ischemic cells, the proteolytic processing of PC2 was greatly attenuated. Finally, in mice, an intracerebroventricular administration of synthetic DYN-A(1-8) significantly reduced the extent of ischemic brain injury. In mice those lack an active PC2, exacerbated brain injury was observed after an otherwise non-lethal focal ischemia. We conclude that brain ischemia attenuates PC2 and PC2-mediated neuropeptide processing. This attenuation may play a role in the pathology of ischemic brain injury.
