Penelitian ini berjudul “Upaya Guru Dalam Meningkatan Hasil Belajar Fiqih Wudhu Di Mi Ma’arif Sutawinangun”. Rumusan masalah dalam penelitian ini adalah “Bagaimanakah Kemampuan Guru Dalam Mempraktik Wudhu Pada Peserta Didik di MI Ma’Arif Sutawinangun?”. Tujuan dilakukannya penelitian ini Untuk Mengetahui Bagaimanakah Kemampuan Guru Dalam Mempraktik Wudhu Pada Peserta Didik di MI Ma’Arif Sutawinangun. Penelitian yang penulis lakukan ini adalah termasuk dalam penelitian tindakan kelas. Dalam perjalanan mengumpulkan data, penulis menggunakan metode wawancara, observasi dan dokumentasi. Sedangkan untuk analisisnya, penulis menggunakan teknik analisis kualitatif deskriptif. Hasil penelitian ini adalah Penggunaan metode dapat memberikan peningkatan aktivitas guru dan anak di dalam kelas pada saat proses belajar mengajar berlangsung. Karena kegiatan praktik wudhu dengan metode demonstrasi di pada Mi Ma’arif Sutawinangun, meningkatkan konsentrasi anak-anak terhadap suatu objek yang sedang diperhatikannya dan dapat memperagakan kegiatan wudhu. Adapun hasil penelitian guru fiqh berupaya atau berusaha dengan akal dan ikhtiar agar peserta didik memahami materi wudhu dengan baik dan benar melalui Rencana Pelaksanaan Pemebelajaran (RPP) dan terkadang melakukan praktik, memberikan hadiah (reward) dan menggunakan infocus. Dan kesulitan atau mempunyai beberapa factor penghambat ketika ditugaskan untuk praktik wudhu kurang focus dan factor pendukung memalui media youtobe atau menggunakan power point.

3. Pembagian Harta Warisan berdasarkan Metode Hukum Waris Islam dan Kompilasi Hukum Islam

- Abstract
Penelitian ini bertujuan untuk mempelajari bagaimana metode pembagian harta warisan yang berdasarkan Hukum Waris Islam dan Kompilasi Hukum Islam mampu menyelesaikan permohonan atau sengketa pembagian harta warisan di antara orang-orang beragama Islam melalui lembaga Peradilan Agama Kendari. Penelitian ini dilaksanakan pada Kantor Pengadilan Agama Kendari. Penelitian ini menggunakan pendekatan yuridis normatif, dimana data dikumpulkan melalui studi bahan-bahan kepustakaan serta wawancara lalu kemudian dianalisa dengan metode deskriptif kualitatif merupakan argumetasi hukum yang logis dan sistematis sesuai dengan rumusan masalah yang telah dirumuskan. Hasil Penelitian menunjukan bahwa : 1. Dalam pembagian harta warisan, orang-orang yang beragama islam diberi kebebasan untuk memilih hukum mana yang digunakan apakah menggunakan hukum waris islam (fiqh/faraidh), menggunakan hukum waris perdata barat (BW) atau menundukan diri pada Kompilasi Hukum Islam. 2. Dalam pembagian harta warisan, mengikuti langkah-langkah sebagai berikut : a. Penentuan Ahli Waris berdasarkan Golongan atau kelompok. b. Menentukan bagian masing-masing Ahli Waris. c. Menghitung kadar atau takaran perolehan harta warisan dari ahli waris. d. Pelaksanaan pembagian/penyerahan Harta Warisan kepad Ahli Waris. 3. Dalam pembagian harta warisan berdasarkan metode Hukum Waris Islam dan Kompilasi Hukum Islam di Pengadilan Agama Kendari diperoleh fakta-fakta bahwa penyelesaian sengketa Pembagian Harta Warisan ada yang menempuh jalan damai, dan ada yang tidak dikabulkan disebabkan oleh karena gugatan/permohonan cacat formil.

- Abstract
This study aims to determine the process of implementing psychosocial assistance to child victims of sexual crimes in Kendari City and to find out what obstacles are faced by implementing psychosocial assistance to child victims of sexual crimes. This research was conducted in Kendari City, the Integrated Service Center for Women and Children Empowerment, hereinafter abbreviated as P2TP2A, and the Kendari City Social Service. This writing is done by conducting field studies by conducting interviews as well as obtaining documents related to the problems in the author's writing by using Qualitative methode. This Research aims to reduce the case problem of sexual crimes. Interviews were conducted by asking questions that were relevant to the topic posed. The second approach is to describe descriptively various interview results and then analyze the data. The process of handling victims in this case the implementation of psychosocial assistance is carried out only as an examination process only to complete the examination file at the police. If the police receive a complaint report, the police will contact P2TP2A and the Social Service to seek psychosocial assistance for children who are victims of sexual crimes. On the other hand, if P2TP2A or the Social Service have received reports of sexual crimes against children, P2TP2A and the Social Service will coordinate with the Police so that perpetrators of these sexual crimes can be prosecuted.

Latar belakang: Fase awal kehidupan, terutama masa sejak dalam kandungan hingga tahun kedua kehidupan anak, merupakan masa yang sangat penting bagi pertumbuhan dan perkembangan anak. Deteksi dini defisit perkembangan sangat penting untuk identifikasi dini perbedaan perkembangan pada anak prasekolah, termasuk memantau keluhan orang tua tentang kemungkinan masalah perkembangan pada anak. Tujuan: Tujuan dari kegiatan ini diharapkan dapat mendeteksi dini tumbuh kembang anak usia pra sekolah. Metode: Kegiatan ini dilaksanakan di KBTK Aisyiyah Sumberejo Klaten. Instrumen yang digunakan berupa timbangan digital dan pengukur tinggi badan serta form Kuesioner Pra Skrinig Perkembangan (KPSP) dari Kemenkes RI. Hasil: Status gizi sebagian besar siswa tergolong status gizi normal. Gambaran perkembangan anak di KBTK Aisyiyah Sumberejo Klaten sebagian besar sesuai (88,64%). Kesimpulan: Pertumbuhan siswa di KBTK Aisyiyah Sumberejo Klaten sebagian besar normal dan perkembangan siswa di KBTK Aisyiyah Sumberejo Klaten sebagian besar sesuai dengan umur. Kata Kunci: anak usia pra sekolah, deteksi dini, tumbuh kembang ________________________________________________________________________________________ Abstract Background: The early years of life, especially the period from the womb to the first two years of life, are extremely important for the growth and development of a child. Early detection of developmental gaps is very important for early detection of developmental gaps in preschool children. This includes monitoring parent complaints about child development issues. Objective: to help recognize the early growth and development of preschoolers. Method: This activity took place on KBTK Aisyiyah Sumberejo Klaten. The instruments used were a digital scale and height scale and the Indonesian Ministry of Health's Developmental Pre-Screening Questionnaire (KPSP) form. Result: Nutritional status measurements indicated that most of the students belonged to normal nutritional status. The description of child development in KBTK Aisyiyah Sumberejo Klaten is mostly appropriate (88.64%). Conclusion: student development at KBTK Aisyiyah Sumberejo Klaten is mostly normal and student development at KBTK Aisyiyah Sumberejo Klaten is mostly appropriate. - Abstract
In Indonesia, the Infant Mortality Rate in 2021 is 20.1/1000 live births, although this rate is much better and decreased compared to 2020 of 28.1/1000 live births.. In Semarang Regency the incidence of low birth weight is 5.2/1000 live births which is still a concern of the government in efforts to reduce low birth weight. One of the factors that affect birth weight is the nutritional status of pregnant women. Purpose: This study aims to determine the relationship between nutritional status and birth weight. This research is a correlation study with a cross-sectional design. This research was conducted in September 2022 - January 2023 at BPM Bidan Sri Harti. The population of this study were all newborns born at BPM Bidan Sri Harti in 2022 totaling 162 respondents. The sample in this study using total sampling. Data collection techniques use secondary data compiled using a master table. This study was analyzed bivariately using the Chi Square test. There is a significant relationship between the nutritional status of pregnant women and birth weight, with a p-value of 0.000 (

9. Effect of mineral nanoparticles obtained from thermal spring water on COVID-19

Background: Coronavirus disease 2019 (COVID-19) is presently a life-threatening condition, and despite of multiple attempts, no functional method has been introduced against this pandemic problem yet. In this study, for the first time, the possible antiviral aspects of mineral nanoparticles (MNPs) obtained from a natural source, thermal spring water, were evaluated. Methods: At first, MNPs obtained from the Gishki thermal spring, Kerman province, Iran, were characterized by transmission electron microscope (TEM). Then, the presence of mineral elements in MNPs was identified by X-ray fluorescence (XRF), Energy-dispersive X-ray spectroscopy (EDX), inductively coupled plasma mass spectrometry (ICP-MS), and MTT assay. Finally, 17 cases suspected of COVID-19 were randomly selected, and their nasal swab samples were exposed to two concentrations of MNPs (50 and 100 u/mL). Results: The results of real-time polymerase chain reaction (RT-PCR) test manifested that MNPs had a destructive effect on 4 (33%) COVID-19 cases. Conclusion: Therefore, MNPs of thermal spring water may act as an obstacle against COVID-19.

Introduction: Chemotherapy is one of the management applied to cervical cancer patients. Chemotherapy is beneficial to kill cancer cells, but it affects normal body cells, such as hair roots, skin, and changes in physical appearance. These physical changes result in poor self-assessment and stress. This study was conducted to describe the correlation between body image and stress level of cervical cancer patients undergoing chemotherapy at the Gynecology Polyclinic, Dr. Kariadi General Hospital Medical Center Semarang. Methods: This type of research is quantitative using a correlational research design. The research process was carried out in 2020 at Dr. Kariadi General Hospital Medical Center Semarang with the number of samples obtained was 87 respondents. Results: It showed that there was a relationship between body image and stress levels in cervical cancer patients receiving chemotherapy (r = -0.313 and p-value = 0.003). With a change in physical appearance, patients would feel something different which affects psychological situations and further causes stress. Conclusions: The level of stress has a significant correlation with the body image score of cervical cancer.

Gerbosari Village has a history of landslides with intensity and risk of 56 occurrences over 5 years. Gerbosari Village, Samigaluh District, Kulonprogo Regency is located at geographic coordinates 7◦ 38 '45.33 "- 7◦ 41' 35.24" LS and 100◦ 9 '20.80 "- 110◦ 11' 16.52" BT with topographic conditions at an altitude around 500 - 1000 mdpl. This study aims to determine the subsurface structure of landslide-prone areas in the form of weathered layer thickness and the effect of slope in landslide-prone areas so that it can be used in making micro zonation maps of landslide-prone areas. This study uses 43 microtremor data with a distance between points of 650 m. The microtremor signal was analysed using the horizontal to vertical spectrum ratio ( HVSR) method. From the measurement results, it is obtained that the value of the dominant frequency ranges from 1 - 22 Hz, the value of the amplification factor is obtained in the range of 1 - 10.5, the value of the peak ground acceleration ranges from 60 - 300 cm/s2, the thickness of the weathered layer is obtained in the range of 12 - 22 meters. Based on the results of the slope analysis, the study area is on a slope classified as a bit steep - very steep.

Exclusive breastfeeding is very important for the growth and intelligence of baby’s brain. Data on Exclusive breastfeeding in Central Sulawesi Province is still below the nasional Exclusive breastfeeding coverage, which is 61.9%. Based on information obtained at the village midwife Bahari Makmur, Lamala District, Banggai Regency, in 2021 from January to September there were 90% of babies who did not get exclusive breastfeeding, and it was known that babies were given formula milk to replace breast milk and were given additional food in the form of solid food for their babies. before the predetermined time, namely the age of the baby is less than 6 months. The purpose of the study was to describe the knowledge of exclusive breastfeeding in pregnant women in the Bahari Makmur Village. Methods this type of research uses descriptive quantitative with a cross sectional approach. The population in this study were all pregnant women for the September 2021 period with a total of 33 people in Bahari Makmur Village, Lamala District, Banggai Regency, Central Sulawesi Province, using a total sampling technique. The research instrument used a questionnaire and the data analysis used was univariate analysis. Results mothers who have good knowledge are 2 respondents (6.1%) and those who have less knowledge are 21 respondents (63.6%). Conclusion most pregnant women have less knowledge as many as 21 respondents (63.6%). Suggestions for health workers are that health services, especially midwives, are more active in improving health promotion by using media counseling or health education about exclusive breastfeeding such as leaflets or videos so that mothers are more interested and more active in exclusive breastfeeding ABSTRAKASI Eksklusif sangat penting bagi pertumbuhan dan kecerdasan otak bayi. Data pemberian ASI eksklusif di Provinsi Sulawesi tengah masih berada dibawah cakupan ASI Eksklusif nasional yaitu 61,9%. Berdasarkan informasi yang diperoleh di bidan desa Bahari Makmur, Kecamatan Lamala, Kabupaten Banggai tahun 2021 bulan Januari sampai September terdapat 90% bayi yang tidak mendapatkan ASI eksklusif, dan diketahui bayi diberikan susu formula untuk menggantikan ASI serta diberikan makanan tambahan berupa makanan makanan padat pada bayinya sebelum waktu yang telah ditentukan yaitu usia bayi kurang dari 6 bulan. Tujuan penelitian untuk mengetahui gambaran pengetahuan ASI eksklusif pada ibu hamil di Desa Bahari Makmur. Jenis penelitian ini menggunakan deskriptif kuantitatif dengan pendekatan cross sectional. Populasi dalam penelitian ini adalah seluruh ibu hamil periode September tahun 2021 sejumlah 33 orang di Desa Bahari Makmur Kecamatan Lamala Kabupaten Banggai Provinsi Sulawesi Tengah dengan menggunakan teknik total sampling. Intrumen penelitian menggunakan kuisioner dan analisis data yang digunakan adalah analisis univariat. Hasil penelitian bahwa ibu yang memiliki pengetahuan baik sebanyak 2 responden (6,1%) dan yang memiliki pengetahuan kurang sebanyak 21 responden (63,6%). Simpulan penelitian ini sebagian besar ibu hamil memiliki pengetahuan kurang sebanyak 21 responden (63,6%). Saran bagi tenaga kesehatan diharapkan pelayanan kesehatan khususnya bidan lebih giat lagi dalam meningkatkan promosi kesehatan dengan menggunakan media penyuluhan atau pendidikan kesehatan tentang ASI eksklusif seperti lefleat atau video agar ibu lebih tertarik dan semakin giat lagi dalam memberikan ASI eksklusif

20. Knowledge transformation on the economic recovery in post-pandemic

- Abstract
Economic recovery is the restoration of the business cycle due to the crisis marked by increased activity in various sectors. Meta-analysis is applied to find novelty through qualitative research development. Knowledge mapping during the pandemic was carried out through an analysis of 995 scientific journals obtained from searches on Google Scholar regarding the concept of economic recovery. This study finds that the scientific evolutionary research found several topics, including privatization, forecast, zakat, adoption, free zone, financing, and pandemic crisis. The novelty of the issues obtained is indicated to have a new value that has the potential to be further developed for the dissemination and diversity of knowledge. Knowledge development on the concept of economic development is expected to improve economic conditions in a business environment experiencing turbulence due to health crises.

The purpose of this research is that learning in Indonesia continues. The Covid-19 pandemic in Indonesia has made learning carried out online. Therefore, Learning Tool Development is also carried out online by utilizing E-Learning. The e-Learning used is zoom, quizizz, and google classroom. With online learning, it is hoped that students will remain enthusiastic in carrying out learning, even though there are many obstacles experienced by students during this online learning. Learning tools provided online are expected to train students' critical thinking in ways or strategies to solve mathematical problems or problems. By designing learning tools that are attractive, creative, and efficient so that students do not get bored during online learning, and can use smartphones positively and wisely. Learning kits consist of lesson plans, quizizz-based worksheets, and YouTube audio-visual materials. The purpose of this research is for learning to continue even in the new normal era due to the Covid-19 pandemic, by utilizing technology and information media such as E-learning.

Penelitian ini bertujuan untuk mengetahui bagaimana kebijakan teoritis dalam industrilisasi dan perubahan fungsi sosial hak milik atas tanah Penelitian ini bersifat yuridis normative yaitu penelitian yang difokuskan untuk mengkaji kaidaih-kaidah atau norma-norma dalam hukum positif. yaitu norma-nonna hukum yang terdapat dalam peraturan perundang-undangan di bidang pertanahan, yang terdiri dari Undang-undang, Peraturan Pemerintah, Peraturan Presiden, Keputusan Presiden, Perattran Menteri Negara/ Kepala Bsdan Pertsnahan Nasional, Keputusan Menteri dan Keputusan Pengadilan bahwa fungsi sosial hak milik atas tanah dianggap menjadi dua kepentingan yakni kepentingan individu pemilik tanah dan kepentingan di luar individu pemilik tanah. Munculnya pembangunan industri melahirkan perubahan-perubahan yang ada gilirannya menentukan eksistensi fungsi sosial hak milik atas tanah. Fungsi sosial yang diidealkan dalam normatif aktualisasinya sangat ditentukan oleh perubahan sosial yang terjadi. Sosial-kultural yang semakin majemuk dan heterogen menentukan aktualisasi fungsi sosial tersebut. Fungsi sosial sebagai nilai, berkembang dan berubah berbeda dari satu tempat ke tempat lain, dari waktu ke waktu dan dari kelompok manusia satu kepada kelompok manusia yang lain. Fungsi sosial dipahami berdasar keajengan (regularities, nomos) atau keseregaman (uniformities), bukan sebagai norma positif dan bukan sebagai keharusan-keharusan semata. - Abstract
The world economic recession caused by the Covid-19 Pandemic distracted companies from maintaining business continuity, the consumer goods sector index declined in the first quarter of 2020 to reach its lowest level since 2013. Business continuity is defined as the company's capability to align its resources to increase opportunities growth and prospects in the future, business continuity has a strategic role in supporting the long-term value creation of the company. This study aims to provide empirical evidence regarding aspects of business continuity in the value-added perspective of human capital. The sample used in this study were 49 companies that are members of the non-cyclicals consumer sector for the 2016-2020 period. The analytical tool used in this research is simple linear regression. The results of this study indicate that the added value of human capital can improve aspects of the company's business continuity.

29. Dissemination of Economic Resilience in Scientific Evolution to Strengthen Community Economy in Facing Covid-19 Pandemic Through Bibliometric Analysis

- Abstract
Economic resilience is the financial ability of a nation that relies on the utilization of existing resources in dealing with various problems whose sources are internal or external to create community welfare. Economic resilience is currently experiencing a severe test for all countries due to covid-19 pandemic, which has an impact on the economic crisis. A qualitative approach is applied by actualizing bibliometric analysis of the concept of economic resilience, which is the main focus. The research data is sourced from scientific journals on the concept of economic resilience, which was explored during the time span of the pandemic. The results of the study state that several theories found have a novel value that is rarely published and can help accelerate economic resilience, including disaster risk reduction, independent village, global value chain, ecology resilience, global recession, banking crisis, regional resilience, and village government. The main clusters derived from economic resilience include economic recovery, economic crisis, adaptation strategy, community empowerment, economic vulnerability, financial inclusion, circular economy, and good governance. Knowledge development needs to be carried out to disseminate current theories and issues so that researchers or academics can study and explore more so that the derived knowledge can help provide solutions and improve economic conditions.

[EN] Searches for dark matter (DM) have not provided any solid evidence for the existence of weakly interacting massive particles in the GeV-TeV mass range. Coincidentally, the scale of new physics is being pushed by collider searches well beyond the TeV domain. This situation strongly motivates the exploration of DM masses much larger than a TeV. Secluded scenarios contain a natural way around the unitarity bound on the DM mass, via the early matter domination induced by the mediator of its interactions with the Standard Model. High-energy neutrinos constitute one of the very few direct accesses to energy scales above a few TeV. An indirect search for secluded DM signals has been performed with the ANTARES neutrino telescope using data from 2007 to 2015. 35. Epidemiology of Bacterial Infections in the Maternal Intensive Care Unit of Ibn Tofail Hospital, Marrakech

Background: The purpose of this study is to evaluate the epidemiology and analyse the level of antibiotic resistance of Ab strains in the intensive care unit of the ibn Tofail Hospital of the Mohammed VI University Hospital of Marrakech. Results: Microbiologically, 200 samples were analyzed, of which urine was the most frequent sample with a rate of 36% of all samples, followed by blood cultures with a rate of 36%, catheters in 12%, vaginal samples in 7% and protected distal samples in 3%. The analysis of the resistance profile of germs to antibiotics showed that for Beta-lactams: 81% are extended-spectrum beta-lactamases (ESBL) 16% low level penicillinase (LLP) and high level penicillinase (HLP) at 3%.

- Abstract
The coverage of newborns receiving IMD in Indonesia is 77.6%, in Lampung Province at 84.2%, and IMD at Lampung Timur is 79.4%. Family support is one of the factors that influence the success of IMD. The purpose of this study was to determine the relationship between family support and the success of IMD at the PMB IPO Krisna Itik Rendai, East Lampung in 2021. The type of research used in this research is quantitative. The design used in this study was analytic observation with a cross sectional design. The population in this study was taken from all pregnant women who were interpreted to give birth in December 2021 to January 2022 with a total of 31 people. The sample in this study amounted to 31 people. The sampling technique uses total sampling. The research instrument used a questionnaire and an observation sheet. Data analysis using chi square. Pregnant women experienced anemia in the case group 30 Mothers with good family support found 11 (78.6%) respondents succeeded in doing IMD and 3 (21.4%) respondents did not succeed in IMD. Background: Multidrug resistant Acinetobacter baumannii, (MRAB) is an important cause of hospital acquired infection. The purpose of this study is to evaluate the epidemiology and analyze the level of antibiotic resistance of Ab strains in the intensive care unit of the ibn Tofail Hospital of the Mohammed VI University Hospital of Marrakech. Material and method: This study is a retrospective review of a city hospital epidemiology data base and includes 42 isolates of Acinetobacter baumannii (AB) from 120 patients. The antibiogram was performed according to European Committee on Antimicrobial Susceptibility Testing (EUCAST) recommendations. Results: In our study, all bacteriological samples were taken from the intensive care unit. The mean age was 48.2 years with extremes ranging from 19 to 72 years.88 patients were male (73.4% of all cases) and 32 female (26.6%).The most frequent reason for hospitalization in the intensive care unit was severe head trauma in 35 cases (29%).During the study period, out of 120 samples received, 42 Ab strains were isolated (35% of samples). These strains were mainly isolated from blood cultures (40%). Conclusions: The diffusion of multi-resistant Ab strains in intensive care units seems to be alarming, hence the need to implement an effective strategy to fight nosocomial infections.

41. A Cross-sectional Study to Investigate the Association of Depression among COPD Patients at a Secondary Care Hospital in Nilgiris

Aim: Depression is highly affected comorbid condition with Chronic Obstructive Pulmonary Disease (COPD) patients remaining unrecognized and under-diagnosed. Co-morbid depression affects the prognosis of the disease leading to an increase in the mortality rate. Objective: The main objective of the study was to assess and evaluate the association of depression among COPD patients by using the Hamilton Depression Scale (HAM-D). Study Design, Place and Duration: A cross-sectional study conducted at Government headquarters hospital, Ooty, consisting of 82 COPD patients, who were assessed for depression. Results: Among 82 COPD patients, 17% (n=14) of the patients found without depression, 17% (n=14) of the patients suffered from mild depression, followed by 35.3% (n=29) of the patients from moderate depression, 25.6% (n=21) of the patients suffered from severe depression and 4.8% (n=4) patients diagnosed with very severe depression. Thus, the prevalence of depression in our study population was 82.9%. Depression was found to be significantly associated with age (p =0.000), alcoholism (p =0.012) and duration of disease (p= 0.000). A positive correlation was obtained between depression and age, duration of disease. Conclusion: The study revealed that the severity of depression is strongly associated with age, consumption of alcohol, and duration of COPD.

- Abstract
Autophagy is a conserved catabolic process that maintains cellular homeostasis. Autophagy supports lung tumorigenesis and is a potential therapeutic target in lung cancer. A better understanding of the importance of tumor cell-autonomous versus systemic autophagy in lung cancer could facilitate clinical translation of autophagy inhibition. Here, we exploited inducible expression of Atg5 shRNA to temporally control Atg5 levels and to generate reversible tumor-specific and systemic autophagy loss mouse models of KrasG12D/+;p53−/− (KP) non–small cell lung cancer (NSCLC). Transient suppression of systemic but not tumor Atg5 expression significantly reduced established KP lung tumor growth without damaging normal tissues. In vivo13C isotope tracing and metabolic flux analyses demonstrated that systemic Atg5 knockdown specifically led to reduced glucose and lactate uptake. As a result, carbon flux from glucose and lactate to major metabolic pathways, including the tricarboxylic acid cycle, glycolysis, and serine biosynthesis, was significantly reduced in KP NSCLC following systemic autophagy loss. Furthermore, systemic Atg5 knockdown increased tumor T-cell infiltration, leading to T-cell-mediated tumor killing. Importantly, intermittent transient systemic Atg5 knockdown, which resembles what would occur during autophagy inhibition for cancer therapy, significantly prolonged lifespan of KP lung tumor-bearing mice, resulting in recovery of normal tissues but not tumors. Thus, systemic autophagy supports the growth of established lung tumors by promoting immune evasion and sustaining cancer cell metabolism for energy production and biosynthesis, and the inability of tumors to recover from loss of autophagy provides further proof of concept that inhibition of autophagy is a valid approach to cancer therapy.Significance:Transient loss of systemic autophagy causes irreversible damage to tumors by suppressing cancer cell metabolism and promoting antitumor immunity, supporting autophagy inhibition as a rational strategy for treating lung cancer.See related commentary by Gan, p. 4322

LKB1 and KRAS are the third most frequent co-mutations detected in non-small cell lung cancer (NSCLC) and cause aggressive tumor growth. Unfortunately, treatment with RAS-RAF-MEK-ERK pathway inhibitors has minimal therapeutic efficacy in LKB1-mutant KRAS-driven NSCLC. Autophagy, an intracellular nutrient scavenging pathway, compensates for Lkb1 loss to support Kras-driven lung tumor growth. Here we preclinically evaluate the possibility of autophagy inhibition together with MEK inhibition as a treatment for Kras-driven lung tumors. We found that the combination of the autophagy inhibitor hydroxychloroquine (HCQ) and the MEK inhibitor Trametinib displays synergistic anti-proliferative activity in KrasG12D/+;Lkb1-/- (KL) lung cancer cells, but not in KrasG12D/+;p53-/- (KP) lung cancer cells. In vivo studies using tumor allografts, genetically engineered mouse models (GEMMs) and patient-derived xenografts (PDXs) showed anti-tumor activity of the combination of HCQ and Trametinib on KL but not KP tumors. We further found that the combination treatment significantly reduced mitochondrial membrane potential, basal respiration, and ATP production, while also increasing lipid peroxidation, indicative of ferroptosis, in KL tumor-derived cell lines (TDCLs) and KL tumors compared to treatment with single agents. Moreover, the reduced tumor growth by the combination treatment was rescued by ferroptosis inhibitor. Taken together, we demonstrate that autophagy upregulation in KL tumors causes resistance to Trametinib by inhibiting ferroptosis. Therefore, a combination of autophagy and MEK inhibition could be a novel therapeutic strategy to specifically treat NSCLC bearing co-mutations of LKB1 and KRAS.

Since 2015 the LIGO and Virgo interferometers have detected gravitational waves from almost one hundred coalescences of compact objects (black holes and neutron stars). This article presents the results of a search performed with data from the ANTARES telescope to identify neutrino counterparts to the gravitational wave sources detected during the third LIGO/Virgo observing run and reported in the catalogues GWTC-2, GWTC-2.1, and GWTC-3. This search is sensitive to all-sky neutrinos of all flavours and of energies $>100$ GeV, thanks to the inclusion of both track-like events (mainly induced by $\nu_\mu$ charged-current interactions) and shower-like events (induced by other interaction types). Neutrinos are selected if they are detected within $\pm 500$ s from the GW merger and with a reconstructed direction compatible with its sky localisation. No significant excess is found for any of the 80 analysed GW events, and upper limits on the neutrino emission are derived. Using the information from the GW catalogues and assuming isotropic emission, upper limits on the total energy $E_{\rm tot, \nu}$ emitted as neutrinos of all flavours and on the ratio $f_\nu = E_{\rm tot, \nu}/E_{\rm GW}$ between neutrino and GW emissions are also computed. Finally, a stacked analysis of all the 72 binary black hole mergers (respectively the 7 neutron star - black hole merger candidates) has been performed to constrain the typical neutrino emission within this population, leading to the limits: $E_{\rm tot, \nu} < 4.0 \times 10^{53}$ erg and $f_\nu < 0.15$ (respectively, $E_{\rm tot, \nu} < 3.2 \times 10^{53}$ erg and $f_\nu < 0.88$) for $E^{-2}$ spectrum and isotropic emission. In the quest for high-energy neutrino sources, the Astrophysical Multimessenger Observatory Network has implemented a new search by combining data from the High Altitude Water Cherenkov (HAWC) Observatory and the Astronomy with a Neutrino Telescope and Abyss environmental RESearch (ANTARES) neutrino telescope. Using the same analysis strategy as in a previous detector combination of HAWC and IceCube data, we perform a search for coincidences in HAWC and ANTARES events that are below the threshold for sending public alerts in each individual detector. Data were collected between 2015 July and 2020 February with a live time of 4.39 yr.
