Vascular endothelial function and atherosclerosis are known to be important risk factors for cardiovascular disease. However, it remains unknown whether remnant cholesterol (RC) correlates with vascular endothelial function and atherosclerosis as represented by flow-mediated vasodilation (FMD) and brachial-ankle pulse wave velocity (baPWV). Therefore, in this study, we aimed to investigate this in the general population.In this study, we examined 13,237 subjects who have undergone blood lipid, FMD, and baPWV measurements. Participants were divided into four groups based on RC quartiles. Multivariable linear regression models were used to calculate odds ratios for FMD and baPWV according to the RC levels.A significant negative relationship was found between RC and FMD (β=-0.14, p=0.014), whereas RC was positively associated with baPWV (β=21.42, p＜0.001), especially in the male and without chronic disease medication populations. The population was divided into three groups according to their lipids: dyslipidemia group, nondyslipidemia but RC increased group (RC ＞0.78 mmol/L), and nondyslipidemia and RC normal group (RC ≤ 0.78 mmol/L). The FMD of the three groups was 7.09%±3.36%, 7.39%±3.38%, and 7.57%±3.54%, respectively. The baPWV of the three groups was 1445.26±261.56 cm/s, 1425.04±265.24 cm/s, and 1382.73±267.75 cm/s. Significant differences were noted between the groups.The findings indicated that a higher RC was an independent predictive factor for participants with endothelial function and atherosclerosis. It is important to use RC as a risk management indicator of vascular function, especially for those with normal conventional lipid parameters but increased RC.

Background: A potential role of the antimicrobial peptide LL-37, which is upregulated after infection, in the pathogenesis of Alzheimer’s disease (AD) was identified. However, the clinical relevance of LL-37 in AD is not clear yet. Objective: This study aims to investigate the association of circulating LL-37 with longitudinal cognitive decline and neurodegeneration among older adults with memory complaints. Methods: This cohort study recruited 357 older adults with memory complaints. Participants were followed-up for two years and the cognitive functions were assessed using the Mini-Mental State Examination (MMSE). Serum LL-37, pTau181, and tTau levels were determined at baseline. Associations of baseline LL-37 with longitudinal cognitive decline and change of neurodegenerative biomarkers were analyzed. Results: No difference was found in the slope of longitudinal cognitive decline during follow-up between the low and high LL-37 group, adjusting for age, sex, education, body mass index, APOE ɛ4 carrier status, comorbidities, and baseline MMSE scores (difference in slope: 0.226, 95% CI: –0.169 to 0.621). Higher LL-37 levels were associated with longitudinal cognitive decline, as indicated by a decrease of MMSE scores of 3 points or above during follow-up (OR = 2.11, 95% CI: 1.32 to 3.38). The high LL-37 group had larger slopes of the increase in neurofilament light (difference in slope: 3.759, 95% CI: 2.367 to 5.152) and pTau181 (difference in slope: 0.325, 95% CI: 0.151 to 0.499) than the low LL-37 group. Conclusion: These findings support an association of the antimicrobial peptide LL-37 with AD from a clinical perspective.

We measure the black hole mass and investigate the accretion flow around the local ($z=0.0502$) quasar PG 1119+120. Spectroscopic monitoring with Calar Alto provides H$\beta$ lags and linewidths from which we estimate a black hole mass of $\log \left(M_{\bullet}/\mathrm{M}_{\odot} \right) = 7.0$, uncertain by $\sim0.4$ dex. High cadence photometric monitoring over two years with the Las Cumbres Observatory provides lightcurves in 7 optical bands suitable for intensive continuum reverberation mapping. We identify variability on two timescales. Slower variations on a 100-day timescale exhibit excess flux and increased lag in the $u'$ band and are thus attributable to diffuse bound-free continuum emission from the broad line region. Faster variations that we attribute to accretion disc reprocessing lack a $u'$-band excess and have flux and delay spectra consistent with either $\tau \propto \lambda^{4/3}$, as expected for a temperature structure of $T(R) \propto R^{-3/4}$ for a thin accretion disc, or $\tau \propto \lambda^{2}$ expected for a slim disc. Decomposing the flux into variable (disc) and constant (host galaxy) components, we find the disc SED to be flatter than expected with $f_{\nu} \sim \rm{const}$. Modelling the SED predicts an Eddington ratio of $\lambda_{\rm Edd} > 1$, where the flat spectrum can be reproduced by a slim disc with little dust extinction or a thin disc which requires more dust extinction. While this accretion is super-Eddington, the geometry is still unclear, however a slim disc is expected due to the high radiation pressure at these accretion rates, and is entirely consistent with our observations., Comment: 23 pages, 16 figures, 10 tables. Background: Olfactory dysfunction appears prior to cognitive decline, and thus it has been suggested to be an early predictor of Alzheimer’s disease. However, it is currently not known whether and how olfactory threshold test could serve as a quick screening tool for cognitive impairment. Objective: To define olfactory threshold test for screening cognitive impairment in two independent cohorts. Methods: The participants are comprised of two cohorts in China, 1,139 inpatients with type 2 diabetes mellitus (T2DM, Discovery cohort) and 1,236 community-dwelling elderly (Validation cohort). Olfactory and cognitive functions were evaluated by Connecticut Chemosensory Clinical Research Center test and Mini-Mental State Examination (MMSE), respectively. Regression analyses and receiver operating characteristic (ROC) analyses were carried out to determine the relation and discriminative performance of the olfactory threshold score (OTS) regarding identification of cognition impairment. Results: Regression analysis showed that olfactory deficit (reducing OTS) was correlated with cognitive impairment (reducing MMSE score) in two cohorts. ROC analysis revealed that the OTS could distinguish cognitive impairment from cognitively normal individuals, with mean area under the curve values of 0.71 (0.67, 0.74) and 0.63 (0.60, 0.66), respectively, but it failed to discriminate dementia from mild cognitive impairment. The cut-off point of 3 showed the highest validity for the screening, with the diagnostic accuracy of 73.3% and 69.5%. Conclusion: Reducing OTS is associated with cognitive impairment in T2DM patients and the community-dwelling elderly. Therefore, olfactory threshold test may be used as a readily accessible screening tool for cognitive impairment.

This research investigated the impacts of model prediction on the optimization of hybrid energy systems using a system consisting of solar panels, batteries, a proton exchange membrane fuel cell (PEMFC), and a chemical hydrogen generation system. A PEMFC has several advantages, such as low operating temperatures, fast response times, high power density, and environmental friendliness, and it can convert hydrogen into electricity. However, because hydrogen costs are an important consideration, the PEMFC is usually integrated with hybrid energy systems to guarantee system sustainability. Therefore, in this study, a whole-year household load and solar radiation data were applied to optimize the system components and power management, thereby reducing the system cost by 42.43% and improving system sustainability by 7.05%. The system responses showed that some hydrogen consumption might be saved if the solar and load profiles could be foreseen. Two prediction models were developed that could accurately forecast the radiation and load profiles. Next, a second-year dataset was employed to verify the effectiveness of the model prediction. The results showed that the system cost was reduced by 40.20% without model prediction and by 44.06% with model prediction compared to the original system settings. Finally, experiments to illustrate the feasibility of the hybrid energy system were conducted using prediction models. Based on the results, the model prediction was deemed effective in improving the performance of hybrid energy systems.

The purpose of this study was to investigate the anti-inflammatory effects of EU-Idd both in vivo and in vitro. In vivo, we used the collagen-induced arthritis (CIA) rat model to investigate the efficacy of EU-Idd on rheumatoid arthritis. Hematoxylin-eosin staining and Safranin O-fast green staining were used to evaluate the pathological status of the ankle joints in CIA rats. Micro-CT scanning was used to investigate bone erosion of the ankle joints. In vitro, the effect of EU-Idd on Th17 cell differentiation was identified by flow cytometry. TRAP staining was used to detect osteoclast cells. HFLS-RA model cells, induced by tumor necrosis factor-α(TNF-α), were used to evaluate the anti-inflammatory effects of EU-Idd while the levels of related inflammatory cytokines and JAK2/STAT3 proteins were detected by RT-qPCR and western blotting. EU-Idd alleviated joint inflammation in CIA rats and exerted protective effects on the ankle joints. EU-Idd also prevented the differentiation of CD4+ T cells into Th17 cells, reduced the number of osteoclasts, and improved the expression levels of bone metabolism-related proteins including OPG and RANKL. Moreover, EU-Idd inhibited the invasion and migration of HFLS-RA cells and downregulated the expression of related inflammatory cytokine genes and the protein expression levels of p-JAK2 and p-STAT3, both in vivo and in vitro. EU-Idd exerts anti-inflammatory and osteoprotective effects by regulating the JAK2/STAT3 pathway in rheumatoid arthritis. These results are beneficial to excavate new pharmaceutical ingredients for rheumatoid arthritis from iridoid.

We compile optical and mid-infrared light curves from the time-domain surveys (i.e. CRTS, PTF, ZTF, and ASAS-SN) and Wide-field Infrared Survey Explorer (WISE) archive for a selected sample of active galactic nuclei (AGNs) with Hβ reverberation mapping (RM) measurements. We measure the time lags (and thus torus sizes) of W1 (∼3.4 $\mu$ m) and W2 (∼4.6 $\mu$ m) band light curves relative to the optical one using the MICA method. Through Hβ RM, the sample has well-measured AGN properties, therefore allowing us to reliably constrain the relations between torus sizes and AGN properties. We perform linear regressions for the relations between torus sizes and 5100 Å luminosities ($R\propto L_{5100}^{\beta }$ ) in two cases: β = 0.5 and β set free. The latter case yields β ≈ 0.37 ± 0.028 for both W1 and W2 bands, shallower than the expected value of 0.5, possibly due to the dependence of torus size on accretion rate. For β = 0.5, by combining with the previous K band RM measurements, we obtain the characteristic broad-line region (BLR) and tours sizes following RBLR:RK :RW1:RW2 = 1.0:6.2:9.2:11.2. We investigate the deviations of the W1 and W2 band observed torus sizes from the corresponding best-fitting relations (with β = 0.5) and find that they both are correlated with accretion rate. As the accretion rate increases, the torus sizes tend to be shortened compared to the anticipated sizes from the best-fitting relations, similar to the behaviour found in BLRs. Such behaviours can be explained by the self-shadowing effect of slim discs. This is further supported by ratios of the W1 and W2 band torus sizes to BLR sizes, which do not show significant correlations with AGN properties.

The vertebrate brain and spinal cord arise from a common precursor, the neural tube, which forms very early during embryonic development. To shape the forming neural tube, changes in cellular architecture must be tightly co-ordinated in space and time. Live imaging of different animal models has provided valuable insights into the cellular dynamics driving neural tube formation. The most well-characterised morphogenetic processes underlying this transformation are convergent extension and apical constriction, which elongate and bend the neural plate. Recent work has focused on understanding how these two processes are spatiotemporally integrated from the tissue- to the subcellular scale. Various mechanisms of neural tube closure have also been visualised, yielding a growing understanding of how cellular movements, junctional remodelling and interactions with the extracellular matrix promote fusion and zippering of the neural tube. Additionally, live imaging has also now revealed a mechanical role for apoptosis in neural plate bending, and how cell intercalation forms the lumen of the secondary neural tube. Here, we highlight the latest research on the cellular dynamics underlying neural tube formation and provide some perspectives for the future.

Background: Previous cross-sectional studies have identified a possible link between Helicobacter pylori (H. pylori) infection and dementia. However, the association of H. pylori infection with longitudinal cognitive decline has rarely been investigated. Objective: This cohort study aims to demonstrate the effects of H. pylori infection on longitudinal cognitive decline. Methods: This cohort study recruited 268 subjects with memory complaints. Among these subjects, 72 had a history of H. pylori infection, and the rest 196 subjects had no H. pylori infection. These subjects were followed up for 24 months and received cognitive assessment in fixed intervals of 12 months. Results: At baseline, H. pylori infected, and uninfected participants had no difference in MMSE scores. At 2 years of follow-up, H. pylori infected participants had lower MMSE scores than uninfected participants. H. pylori infection was associated with an increased risk of longitudinal cognitive decline, as defined by a decrease of MMSE of 3 points or more during follow-up, adjusting for age, sex, education, APOE ɛ4 genotype, hypertension, diabetes, hyperlipidemia, and smoking history (HR: 2.701; 95% CI: 1.392 to 5.242).
