Background Hexavalent chromium can promote centrosome amplification (CA) as well as tumorigenesis. Since CA can lead to tumorigenesis, it is plausible that the chromium promotes the development of cancer via CA. In the present study, we investigated the signaling pathways of the chromium-induced CA. Results Our results showed that sub-toxic concentration of chromium was able to cause CA in HCT116 cells, and decreased the expression of TMOD2 and NPM2. Furthermore, TMOD2 and NPM2 interacted to each other via their C-terminal and the N-terminal, respectively, which was inhibited by the chromium. Overexpression of TMOD2 and NPM2 increased their binding and significantly attenuated the CA. Moreover, TMOD2 and NPM2 were co-localized with the centrosomes. The chromium inhibited the centrosomeal localization of NPM2, which is reversed by the overexpression of TMOD2, C-termianl of TMOD2, but not the N-terminal of NPM2. Conclusion Our results suggest that the chromium induces CA via inhibiting the binding between TMOD2 and NPM2, and the dissociation of NPM2 from centrosomes.

Uncontrolled bleeding is one of the most important causes threatening human health, but quick hemostasis remains a challenge. We prepared porous cryogels with poly β-cyclodextrin (Pβ-CD) and quaternary ammoniated chitosan (QCs). Pβ-CD acts as a "water-grabbing agent" to assist QCs' ability to absorb and concentrate blood rapidly. The rat-tail amputation model and liver injury model exhibited that cryogels had excellent hemostatic performance. Moreover, cryogels showed good antibacterial activity and biocompatibility. Therefore, these cryogels can be used as potential hemostatic materials.

Heat shock is a feasible means to break rock mass in engineering duo to the large additional thermal stress caused by thermal expansion. This paper established a coupled thermal transfer and rock deformation model based on the energy conservation and the elastic deformation theory of rock. In the model, the failure and damage of rock are judged according to the Drucker-Prager criterion. A finite element method of COMSOL Multiphysics to study the characteristics of temperature, stress distribution and damage zone on a rock surface is proposed. Results show that the failure of rock mass occurs at the cusps of the heater first duo to stress concentration and then grows at both sides of the heater greatly.

OX2 (X=halogen) molecules was studied theoretically. Calculation results show that delocalized [Formula: see text] bonds exist in their electronic structures and O atoms adopt the sp2 type of hybridization, which violates the prediction of the valence shell electron pair repulsion theory of sp3 type. Delocalization stabilization energy is proposed to measure the contribution of delocalized [Formula: see text] bond to energy decrease and proves to bring extra-stability to the molecule. These phenomena can be summarized as a kind of coordinating effect.

The synthesis of the originally proposed structure of (±)-nicotlactone A, a potent antiviral lignan with three continuous chiral centers, is reported in 5 steps from methyl acrylate. The key steps of the synthesis included an In-catalyzed regioselective allylation and a Mn-catalyzed Mukaiyama hydration reaction. Our synthetic strategy also enabled us to get the other three epimers and investigate the structure–activity relationship. The NMR data of the synthesized compounds do not match that of the isolated sample, indicating that the structure of nicotlactone A remains to be reassigned. All the synthetic target compounds were evaluated for their anti-tobacco mosaic virus (anti-TMV) activity. Bioassay results indicated that (±)-8-demethylnicotlactone A displayed similar anti-TMV activity to the commercial agent ningnanmycin, thus being a promising candidate or lead compound for developing novel antiviral agents in crop protection.

Silicosis is a leading cause of occupational disease-related morbidity and mortality worldwide, but the molecular basis underlying its development remains unclear. An accumulating body of evidence supports gasdermin D (GSDMD)-mediated pyroptosis as a key component in the development of various pulmonary diseases. However, there is little experimental evidence connecting silicosis and GSDMD-driven pyroptosis. In this work, we investigated the role of GSDMD-mediated pyroptosis in silicosis. Single-cell RNA sequencing of healthy and silicosis human and murine lung tissues indicated that GSDMD-induced pyroptosis in macrophages was relevant to silicosis progression. Through microscopy we then observed morphological alterations of pyroptosis in macrophages treated with silica. Measurement of interleukin-1β release, lactic dehydrogenase activity, and real-time propidium iodide staining further revealed that silica induced pyroptosis of macrophages. Additionally, we verified that both canonical (caspase-1-mediated) and non-canonical (caspase-4/5/11-mediated) signaling pathways mediated silica-induced pyroptosis activation, in vivo and in vitro. Notably, Gsdmd knockout mice exhibited dramatically alleviated silicosis phenotypes, which highlighted the pivotal role of pyroptosis in this disease. Taken together, our results demonstrated that macrophages underwent GSDMD-dependent pyroptosis in silicosis and inhibition of this process could serve as a viable clinical strategy for mitigating silicosis.

The paper deals with a non-linear barotropic vorticity equation in a shear flow, the model is applied to get the non-linear Rossby solitary wave with the effects of topography and other external forcing. Basing on the perturbation method, the variable coefficient KdV equation is derived for Rossby waves. The periodic-like solution for the equation is obtained with the help of Jacobi elliptic functions, the solitary solutions can also be obtained in the limit case. The analysis indicates that the wave amplitude and velocity will be related to the topography effect. It is also shown that the external forcing plays an important role in evolution of the waves.

Visible-light-induced deaminative alkylation of Katritzky salts with silyl enol ethers has been developed. The reaction can proceed efficiently through electron donor-acceptor complex formation, avoiding the use of precious metal complexes or synthetically elaborate organic dyes. A series of functionalized γ-ketoesters was successfully obtained with good functional group tolerance and compatibility under mild and straightforward conditions.

Aim This study investigated the effects of sodium-glucose cotransporter 2 (SGLT2) inhibitors on renal outcomes in Asian patients with type 2 diabetes mellitus (T2DM). Materials and methods We searched Medline, EMBASE, and the Cochrane Library to identify randomized controlled trials published up to April 2020 that compared SGLT2 inhibitors with placebo or active comparator and reported any renal outcomes in Asian patients with T2DM. Random effects models and inverse variance weighting were used to calculate relative risks with 95% confidence intervals (CIs). Results We included 14 studies, totaling 3792 patients, in the analysis. In the short term, SGLT2 inhibitors significantly slowed estimated glomerular filtration rate (eGFR) decline (MD: 0.80; 95% CI: 0.66 to 0.94; p p p p = 0.11). However, dapagliflozin does appear to reduce albuminuria (p = 0.005). Lastly, SGLT2 inhibitors increased the incidence of adverse events (AEs) related to renal function (OR: 1.90; 95% CI: 1.24 to 2.91; p = 0.003), but did not increase the incidence of renal impairment (OR: 0.85; 95% CI: 0.40 to 1.81; p = 0.68). Conclusion The use of SGLT2 inhibitors in Asian patients with T2DM can help delay the decline of eGFR and reduce Scr and UA. Although SGLT2 inhibitors have no overall advantage in reducing albuminuria, dapagliflozin does appear to reduce albuminuria, and while they may increase the occurrence of AEs related to renal function, they do not increase the incidence of renal impairment.

Background: Burns are an underrated injury that can affect anyone, anytime and anywhere. Therefore, screening immune-related diagnostic markers of burn can be used to prevent serious complications and death.Methods: GEO database obtains GSE19743 and GSE37069, and uses the GPL570 platform. Cibersort algorithm was used to calculate the figure of immune cells in healthy control group and burn group. Immune cells were identified by Least Absolute Shrinkage and Selection Operator (LASSO) regression. “RobustRankAggreg” method was used to screen differentially expressed genes. DEGs' functional correlation was analyzed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Possible biomarkers were identified by Cytoscape's cytoHubba, and the diagnostic result was predicted Receiver Operator Characteristic Curve (ROC) analysis. Spearman's rank correlation analysis in R package was used to learn the correlation between the identified biomarkers and infiltrating immune cells. Results: After joint analysis of the two sets of data, 60 DEGs were obtained, of which 42 increased and 18 decreased. Generally speaking, the function of DEG is related to immune cells. In CytoHubba, 6 hub genes were obtained through the intersection of 10 algorithms. GZMB, OLFM4, ELANE, MMP9, LCN2 and CEACAM8 showed good specificity and sensitivity, respectively. The correlation between 6 effective biomarkers and 12 meaningfully different immune cells in burn group was analyzed. The outcomes demonstrated that LCN2 was correlated with M0 macrophages positively (R=0. 594, P < 0.001).Conclusion: CEACAM8 and LCN2 have great potential as new diagnostic biomarkers of immune dysfunction after burn.

Phthalates as a large group of environmental pollutants are used primarily as plasticizers and solvents, which have become a growing problem worldwide. Epidemiological results show that severity of heart disease is related to degree of environmental contamination. As the most usually used phthalate, di(2-ethylhexyl) phthalate (DEHP) has toxic effects on organism health and is also a major cause of heart damage. Ingestion of food, liquid, or dust contaminated with DEHP are major routes of exposure. The purpose of the present research was to determine the mechanism of cardiotoxicity in mice after exposure to DEHP. Here, male mice were treated by gavage with three different doses of (50, 200 and 500 mg/kg b.w.) DEHP for 28 days. Our research showed that DEHP brought about histopathological changes involving cardiomyocyte lysis and rupture, and ultrastructural damage such as dissolution and loss of mitochondrial cristae. Furthermore, DEHP induced oxidative stress and a significant decline in the antioxidant function, which activates nuclear factor E2-related factor 2 (Nrf2)/heme-oxygense-1 (HO-1) signaling pathways. Interestingly, DEHP resulted in lipid peroxidation and increased ferrous ion content, suggesting that ferroptosis occurred in mouse hearts. Therefore, our findings demonstrated that DEHP could induce cardiac ferroptosis via upregulation of HO-1. The present study provides novel evidence of HO-1 as a target for DEHP-induced cardiotoxicity.

Hydrogen-bonded organic frameworks (HOFs) have received tremendous attention in recent years due to the good designability. However, the pure organic nature of HOFs sometimes limits the application development and performance improvement. Functionalizing is an effective strategy to control and modulate material properties, which can achieve properties that cannot be achieved by a pristine material. Herein, a series of HOF-76⊃DSMI were synthesized through functionalizing the stable AIE-based HOF-76 by incorporating a red dye which complements the deficiency of the red component of HOF-76. Then, a single matrix white light-emitting diode (WLED) was fabricated by coating the HOF-76⊃DSMI material on a 460 nm blue LED with CIE chromaticity coordinates of (0.333, 0.329), a correlated colour temperature (CCT) of 5490 K and a colour rendering index (CRI) of 80.

The superior optical properties of lead-halide perovskites (LHPs) inspired significant research in cell imaging applications; many encapsulating processes have improved perovskite stabilities with comparable biosafety. Herein, facile solvent evaporation encapsulation based on an oil-in-fluorine emulsion for aqueous-stable and extremely nontoxic LHP microcapsules is described. Perfluorooctane dispersed the emulsifier fluorocarbon surfactant to form a continuous fluorine phase, while LHPs and polymethylmethacrylate (PMMA) were dispersed in 1,2-dichloroethane, then emulsified in the fluorine phase to form an oil-in-fluorine emulsion. CsPbBr3 microcapsules with a dense PMMA shell that protect fragile CsPbBr3 from the external environment and inhibit lead ion release were obtained after solvent evaporation. The CsPbBr3 microcapsules not only retained 91% of fluorescence intensity after exposure to water for 30 d but also possess extremely low cytotoxicity for MCF-7 cells. After exposure to 2 mg/mL of CsPbBr3 microcapsules for 48 h, the cell viability remained >90%. The intracellular uptake of CsPbBr3 microcapsules indicates its potential use in cell imaging.

Male infertility is a multifactorial pathological condition and may be a harbinger of future health. Phthalates are ubiquitous environmental contaminants that have been implicated in the global decline in male fertility. Among them, di-(2-ethylhexyl) phthalate (DEHP) is the most prevalently used. Lycopene (LYC) is a possible preventive and therapeutic agent for male infertility owing to its antioxidant properties. The blood-testis barrier (BTB) is formed between Sertoli cells where it creates a unique microenvironment for spermatogenesis.We hypothesize that phthalate caused male infertility and LYC plays an important role in phthalate-induced male fertility disorders.Hematoxylin-eosin (HE) staining, ultrastructure observation, and fluorescence microscopy were used to examine the morphological changes. RNA-Seq, and western blotting were conducted to detect gene and protein levels. Routine testing for sperm morphology and sperm-egg binding assay were conducted to examine the morphological structure and function of sperm. Cell scratch assay and transepithelial electrical resistance (TER) were used to detect cell migration capacity and barrier integrity.In vivo experiments, we showed that LYC prevented DEHP-induced impairment of BTB integrity, which provided a guarantee for the smooth progress of spermatogenesis. LYC improved DEHP-induced change in sperm parameters and fertilization ability. Subsequent in vitro experiments, LYC alleviated MEHP-induced disruption of intercellular junctions in mouse Spermatogonia cells (GC-1 cells) and mouse Sertoli cells (TM4 cells). In MEHP-induced BTB impairment models of Sertoli cells, treatment with LYC or overexpressing connexin-43 (Cx43) promoted cell migration capacity and normalized BTB integrity. Cx43 knockdown inhibited cell migration capacity and perturbed BTB reassembly in LYC preventing DEHP-induced BTB impairment.Our study provides evidence for the role of LYC in phthalates-induced spermatogenic disorders and points to Cx43 as a potential target for male fertility.

Background: To investigate the correlation between hypertriglyceridemic waist circumference (HTWC) phenotype and gestational diabetes mellitus (GDM).Methods: A total of 1083 patients with gestational age ≤8 weeks were divided into four groups: normal triglyceride and waist circumference group (group A, n=575), simple abdominal obesity group (group B, n=317), simple high triglyceride group (group C, n=125), and HTWC group (group D, n=66). General information and serum biochemical indicators were measured and recorded. Analysis of variance (ANOVA) and logistic regression analysis were used to evaluate the relationship between HTWC with GDM.Results: The prevalence of GDM in the HTWC group was significantly greater than in the other three groups. After adjustment by multivariate logistic regression analysis, the proportion of GDM in the HTWC group was 1.753 times higher than in group A.Conclusion: These findings suggest that there is a significant correlation between HTWC phenotype and GDM, indicating that the HTWC phenotype could be applied as a simple marker for identifying GDM risk factors.

Increasing acidity is an effective method for bacterial inactivation by inhibiting the synthesis of intracellular proteins at low pH. Photo-driven proton release probe can be used for the measurement of proton in hydrophobic condition. To develop fluorescent proton donor, two boron dipyrromethene derivatives (BDP-S and BDP-S2) were characterized by spectroscopic methods. Irradiation of BDP-S by white LED light resulted in efficient generation of acidic species with changes of fluorescence emission. The linear relationship between the pH value and the fluorescence intensity of BDP-S was obtained, indicating that BDP-S is a fluorescent visual proton donor. Light-induced antibacterial results indicate that BDP-S can significantly inhibit the growth of E. coli. The results prove that BDP-S is a very promising photoacid sterilant.

A visible-light-induced iodine-anion-catalyzed C–H stereoselective alkylation of enamides has been developed. Redox-active esters and Katritzky salts of amino acids were found to be amenable for decarboxylative/deaminative cross-coupling reactions, delivering various functionalized enamides with excellent functional group tolerance. The reaction proceeded via photoactivation of the transiently assembled chromophores, avoiding the use of exogenous precious photoredox catalysts.

The global rate of human male infertility is rising at an alarming rate owing to environmental and lifestyle changes. Phthalates are the most hazardous chemical additives in plastics and have an apparently negative impact on the function of male reproductive system. Ferroptosis is a recently described form of iron-dependent cell death and has been linked to several diseases. Transferrin receptor (TfRC), a specific ferroptosis marker, is a universal iron importer for all cells using extracellular transferrin. We aim to investigate the potential involvement of ferroptosis during male reproductive toxicity, and provide means for drawing conclusions on the effect of ferroptosis in phthalates-induced male reproductive disease. In this study, we found that di (2-ethylhexyl) phthalate (DEHP) triggered blood-testis barrier (BTB) dysfunction in the mouse testicular tissues. DEHP also induced mitochondrial morphological changes and lipid peroxidation, which are manifestations of ferroptosis. Four sewage sludge (SS) feedstocks with distinct properties were converted into biochars by pyrolysis at 300–700 °C, in order to clarify the effects of the composition difference of SS feedstocks. The yields of biochars present a positive correlation with the contents of ash in SS. Notedly, the contents of organic matter (OM) in SS largely determine the quality of biochars. SS feedstocks with high content of OM are more likely to form stable biochars with higher aromaticity/carbonization degree, and the formed biochars possess higher calorific values. The contents of residual OM in biochars derived from SS feedstocks with low content of OM likely fail to meet the needs of soil improvement (10 wt.%). Most of heavy metals (HMs) existing in raw SS are remained in biochars after pyrolysis. The biochar produced from SS feedstocks with high content of HMs usually contains higher contents of HMs. Surprisingly, the leachability of HMs in biochars is all weakened to some extent compared to raw SS. In addition, the biochars show higher thermal stability and pH values, and P/K nutrients are enriched in biochars. The biochars prepared from four SS feedstocks exhibit different adsorption ability of methylene blue, especially at low dosage of biochar.

CO2 flooding can effectively enhance the recovery of low-permeability reservoirs and realize CO2 geological storage. During the displacement process, the minimum miscible pressure (MMP) of CO2 and oil is an important parameter that affects the displacement effect and storage efficiency. However, the microscopic pore-throat structure of low-permeability reservoirs has significant influences on the fluids and phase behaviors. This paper presented a method to determine the miscible state of CO2 flooding based on the microscopic pore-throat structure. Firstly, a physic model was established to quantitatively characterize the microscopic pore-throat structure. Secondly, taking into consideration the P-R equation of state, the gas-liquid equilibrium in the narrow pore-throat was calculated. On this basis, a MMP prediction model was established correspongdingly by considering the multi-stage contact and mass transfer of CO2-oil. Finally, the results obtained by the proposed model were compared with the experimental results of CO2 flooding, and then the model was applied to the actual reservoir to predict plane distribution of MMP. The curves of MMP distribution and pressure drawdown between wells were combined to determine the position of miscible front and non-miscible area at different production stages. The results have shown that the MMP of core sample calculated by the model was 20.3 MPa, which was comparable to that of CO2 flooding experiment, e.g., 20 MPa, and thus indicatesd a high accuracy of the model. The MMP in the well control area of the Y29-101 well group was 19.8 MPa. During the unsteady flow stage, the miscible-phase front was 430 m from the injection well, while it was 310 m from the injection well during the stable flow stage. This method can accurately determine the specific phase distribution of CO2-oil in the formation, which is of great significance to promote the development of CO2 flooding and storage technology, improve the recovery of low permeability reservoirs, ensure energy supply and reduce carbon emission.
