Abstract

Progress in RNA metabolism and function studies relies largely on molecular imaging systems, including those comprising a fluorogenic dye and an aptamer-based fluorescence-activating tag. G4 aptamers of the Mango family, typically combined with a duplex/hairpin scaffold, activate the fluorescence of a green light-emitting dye TO1-biotin and hold great promise for intracellular RNA tracking. Here, we report a new Mango-based imaging platform. Its key advantages are the tunability of spectral properties and applicability for visualization of small RNA molecules that require minimal tag size. The former advantage is due to an expanded (green-to-red-emitting) palette of TO1-inspired fluorogenic dyes, and the truncated duplex scaffold ensures the latter. To illustrate the applicability of the improved platform, we tagged Mycobacterium tuberculosis sncRNA with the shortened aptamer-scaffold tag. Then, we visualized it in bacteria and bacteria-infected macrophages using the new red light-emitting Mango-activated dye.

2. Assessment of naphazoline effect on erythrocyte superoxide dismutase activity in rats under experimental modeling of acute radiation sickness

Abstract

Aim to assess the activity of superoxide dismutase in erythrocytes of rats after a single intramuscular injection of naphazoline in radiofrequency dose and in experimental simulation of acute radiation sickness. Material and methods. The activity of erythrocytes superoxide dismutase (SOD) after single intramuscular injection of nafazoline in experimental modeling of acute radiation sickness was investigated. The pharmacological properties and mechanisms of radioprotective action of nafazoline were clarified based on the dynamics of activity of erythrocytes SOD in intact and irradiated animals with the dose of 7,4 Gy. Results. The superoxide dismutase activity was found to increase significantly 60 min after single intramuscular administration of nafazolin at a dose of 5 mg/kg. Under experimental conditions of acute radiation disease modeling, a decrease in superoxide dismutase activity was observed within the first hour after exposure, evidencing the direct involvement of the antioxidant system components in the inactivation of free-radical reaction products. Conclusion. The radioprotective properties of naphazoline may be due not only to reduction of oxygen delivery to the cells of radiosensitive tissues and inhibition of their metabolism by effect on 2-adreno- and imidazoline receptors, but also by activation of antioxidant system links.

Abstract

The article deals with the problem of the formation of adolescent tolerance and examines the influence of subjective control on the tolerance of older adolescents. The education system, having unique opportunities to expand knowledge about other people who are different from us, their views, culture, as one of the most important tasks, puts forward the problem of forming tolerance among the younger generation. The relevance of this socio-pedagogical problem is due to the need to find effective means of forming tolerance among students. The study was conducted on the basis of the municipal budgetary educational institution "General education institution Gymnasium No. 11 named after A.N. Kulakov" in Krasnoyarsk. The sample of the study was 133 subjects. The female sample is 71 people, the male sample is 62 people, students of grades 9-11. The obtained data and statistical analysis confirmed the dependence of the level of tolerance of older adolescents on sex differences. Girls are tolerant, they are characterized by tolerant forms of behavior, while boys, on the contrary, are more often intolerant, both to the outside world and to people. With the help of Kendall's rank correlation coefficient, data were processed and compared using the methods "Level of subjective control" and "Tolerance Index". The data confirmed the relationship between the level of subjective control and the level of tolerance of older adolescents. Externals have a low tolerance for others, internals have a higher level of tolerance. In addition, the data showed that girls have a higher level of tolerance than boys.

Abstract

INTRODUCTION: TDespite the achievements of the modern medicine, use of some medical drugs (MDs) is associated with both the absence of a significant therapeutic effect due to peculiarities of the physico-chemical interaction in the internal environment of an organism, and with the adverse effect on organs and tissues. The advanced technologies of creation of micro- and nanoparticles will permit to improve pharmacokinetics and pharmacodynamics of the drug, its bioavailability and solubility, the ability of crossing blood-brain barriers, and to reduce undesirable systemic effects. MDs using micro- and nanoparticles, have a targeted effect on the focus of pathological lesion. An important additional advantage is a possibility of using micro- and nanoparticles in development of long-acting MDs. The main active substances immobilized to micro- and nanoparticles, open up new prospects for effective treatment of different pathological conditions (neoplasms, diseases of cardiovascular and central nervous system, inflammatory processes, wounds), and for realization of new imaging capabilities in foci of a pathological process, which is especially important in diagnostic procedures. CONCLUSION: The article presents a summary of the ideas of methods of micro- and nanoencapsulation, and assessments of the prospects for the development of drugs for the correction of pathological conditions using innovative technologies.

Abstract

The subject of the work is the development and implementation of a program for the regulation of negative psychological states of karate athletes from 8 to 14 years old. The purpose of this study is to develop an effective program aimed at correcting the negative psychological states of karate athletes from 8 to 14 years old during preparation for competitive activities. In our work, we analyze the methods of psychological regulation of karate athletes from 8 to 14 years old during the preparation for competitive activity. In the teaching of the karate section, there are a number of problems associated with a decrease in the cognitive activity of students, with a decrease in the level of health culture, which is due to a number of factors: improper nutrition of athletes, bad habits (alcohol, smoking), a small amount of sports in life (the development of physical inactivity), workload in training, etc. These factors force teachers to look for new ways in teaching and educating students. In this work, the psychological states of karate athletes from 8 to 14 years old for competitions were studied. We have developed a program for regulating the psychological states of karate athletes from 8 to 14 years of age in the pre-competition period of training by organizing work with model situations in which athletes can detect psychological difficulties associated with motivation to achieve success, anxiety and stress, as well as together with the coach to find ways to resolve them, during which there will be an increase in motivation to achieve success and stress resistance and reducing the level of anxiety. In the course of the study, after testing the program, changes in the level of anxiety (its decrease) and motivation (its increase) occurred in 83.3% of respondents.

10. Distribution of amanitine-containing macromycetes in the territory of Russia

Abstract

Introduction. The toxic effect of amanitin-containing macromycetes, or phalloidin syndrome, is accompanied by low patient survival and it remains as one of the leading causes of urgent liver transplantation. As a result, the problems of biodiversity and the distribution of amanitin-containing fungi leading to this condition appear in a relevant light for toxicology. Materials and methods. The study was carried out in two stages. At the first stage, a preliminary list of amanitin-containing macromycetes was compiled by means of a system search in the PubMed database. At the second stage of the study, the presence and occurrence of amanitin-containing taxa in Russia were analyzed. Results. Based on the literature sources and data of the Laboratory of Geography and Systematics of Fungi of the Komarov Botanical Institute of the Russian Academy of Sciences, maps of the administrative-territorial occurrences in Russia of twenty-one species of amanitin-containing macromycetes were compiled: five of Amanita, one of Galerina and fifteen of Lepiota. The species concepts of some poisonous taxa and their key identification features were reviewed. The debatable ideas about the toxicity of two Conocybe species are highlighted. Limitations. The study takes into account only the specific occurrence of poisonous fungi in various territories of Russia, without considering the density and frequency of their growth, which may make it difficult to assess the relationship between the data presented and the likelihood of poisoning by the species in question in individual administrative units. Conclusion. As a result of the study, the boundaries of the distribution of poisonous fly agaric and Galerina species were identified, as well as the territories with the highest registered diversity of Lepiota: the Moscow Region and the Primorsky Territory.

13. Preliminary Stages of Investigation of Smuggling of Weapons of Mass Destruction

Abstract

The article presents the criminalistic characteristics of weapons of mass destruction, as well as their components, objects and substances from which they can be made, focuses on the high level of public danger that weapons of mass destruction carry, especially in connection with the activation of extremist and terrorist groups. The paper considers and summarizes the complex of investigative and operational measures that the investigative body is obliged to carry out at the preliminary stage of the investigation of the smuggling of weapons of mass destruction, as well as materials and equipment that can be used in its creation, provides a description of the main (typical) investigative situations in the commission of this crime. The author comes to the conclusion that only with the close cooperation of all law enforcement agencies, the crimes under investigation on the territory of Russia can be solved as soon as possible, and the conclusion is also made about the possible involvement of unfriendly states, criminal communities and terrorist organizations in the smuggling of weapons of mass destruction.

Abstract

Single-domain antibodies (sdAbs, VHHs, or nanobodies) are a promising tool for the treatment of both infectious and somatic diseases. Their small size greatly simplifies any genetic engineering manipulations. Such antibodies have the ability to bind hard-to-reach antigenic epitopes through long parts of the variable chains, the third complementarity-determining regions (CDR3s). VHH fusion with the canonical immunoglobulin Fc fragment allows the Fc-fusion single-domain antibodies (VHH-Fc) to significantly increase their neutralizing activity and serum half-life. Previously we have developed and characterized VHH-Fc specific to botulinum neurotoxin A (BoNT/A), that showed a 1000-fold higher protective activity than monomeric form when challenged with five times the lethal dose (5 LD50) of BoNT/A. During the COVID-19 pandemic, mRNA vaccines based on lipid nanoparticles (LNP) as a delivery system have become an important translational technology that has significantly accelerated the clinical introduction of mRNA platforms. We have developed an mRNA platform that provides long-term expression after both intramuscular and intravenous application. The platform has been extensively characterized using firefly luciferase (Fluc) as a reporter. An intramuscular administration of LNP-mRNA encoding VHH-Fc antibody made it possible to achieve its rapid expression in mice and resulted in 100% protection when challenged with up to 100 LD50 of BoNT/A. The presented approach for the delivery of sdAbs using mRNA technology greatly simplifies drug development for antibody therapy and can be used for emergency prophylaxis.

Abstract

CP4 3HDM is a three-Higgs-doublet model based on the $CP$ symmetry of order 4 (CP4). Imposing CP4 leads to remarkable connections between the scalar and Yukawa sectors and unavoidably generates tree-level flavor-changing neutral couplings (FCNC). It remains unclear whether FCNC can be sufficiently suppressed in the CP4 3HDM. In this paper, we systematically explore this issue. We first develop an efficient scanning procedure which takes the quark masses and mixing as input and expresses the FCNC matrices in terms of physical quark observables and quark rotation parameters. This procedure allows us to explore the FCNC effects for all the Yukawa sectors possible within the CP4 3HDM. We find that, out of the eight possible CP4 Yukawa sectors, only two scenarios are compatible with the $K$, $B$, $B_s$ and, in particular, $D$-meson oscillation constraints. 17. Application aspects of medications for inhalation at the prehospital stage of medical evacuation

Abstract

This study aimed to determine the aspects of inhalation application of drugs as medical means of protection against lesions by factors of radiation, chemical, and biological nature at the prehospital stage of medical evacuation. Advantages of inhalation administration of drugs and use of individual inhalers of portable type over parenteral and oral administration methods are described. The existing drugs that are most suitable for inhalation at the prehospital stage of medical evacuation include emergency prevention and treatment of toxic pulmonary edema, analgesics, antiradiation, drugs for arresting radiation primary reaction, agents for accelerated radionuclides elimination, and antibacterial and antiviral agents. This list is conditioned by the rapid achievement of the protective effect of the drug during inhalation, both due to the local action in the area of the entrance gate of the intake of damaging agents and the accelerated absorption and systemic action on target organs. This study presents data on existing and promising inhalation drugs (antidotes for warfare agents, recombinant forms of acetyl- and butyrylcholinesterase) and technical means of their delivery (a metered aerosol inhaler, a metered dry powder inhaler) in the Armed Forces of the European North Atlantic Treaty Organization bloc and the United States for the prevention and treatment of damage e to chemical agents, as well as the prospects of radiomitigator usage (granulocyte-macrophage colony-stimulating factor) by inhalations for acute radiation syndrome treatment and inhalation of complexones of radioactive isotopes. The prospects of inhalation in combination with bronchodilators in the development of acute respiratory failure and toxic pulmonary edema on the background of poisoning with the use of a metered-dose powder inhaler are described. The introduction and use of a metered-dose dry powder inhaler at the prehospital stage of medical evacuation determines the need for its development, taking into account the specifics of the drug usage, as well as the need to unify the dosage form of drugs in combination with one type of inhaler (single-dose or multi-dose).

Abstract

Xanthine derivatives have been a great area of interest for the development of potent bioactive agents. Thirty-eight methylxanthine derivatives as acetylcholinesterase inhibitors (AChE) were designed and synthesized. Suzuki–Miyaura cross-coupling reactions of 8-chlorocaffeine with aryl(hetaryl)boronic acids, the CuAAC reaction of 8-ethynylcaffeine with several azides, and the copper(I) catalyzed one-pot three-component reaction (A3-coupling) of 8-ethynylcaffeine, 1-(prop-2-ynyl)-, or 7-(prop-2-ynyl)-dimethylxanthines with formaldehyde and secondary amines were the main approaches for the synthesis of substituted methylxanthine derivatives (yield 53–96%). The bioactivity of all new compounds was evaluated by Ellman’s method, and the results showed that most of the synthesized compounds displayed good and moderate acetylcholinesterase (AChE) inhibitory activities in vitro. The structure-activity relationships were also discussed. The data revealed that compounds 53, 59, 65, 66, and 69 exhibited the most potent inhibitory activity against AChE with IC50 of 0.25, 0.552, 0.089, 0.746, and 0.121 μM, respectively. The binding conformation and simultaneous interaction modes were further clarified by molecular docking studies.

Abstract

The Advisory Board chaired by the chief specialist in infectious diseases of the Ministry of Health of Russian Federation, Professor V.P. Chulanov was held on June 18, 2022 in Saint Petersburg. Aim. The main purpose of the Board was following discussion: the analysis of the real-world data of levilimab as an anticipatory therapy for COVID-19 in hospitalized patients; the review of the experience and perspectives of levilimab as an anticipatory anti-inflammatory option for outpatient patients who meet defined clinical and laboratory criteria. Results. The analyzed data on clinical efficacy and safety formed the basis of recommendations proposed by experts for the use of levilimab in the inpatient and outpatient medical care for COVID-19.18 июня 2022 г. в Санкт-Петербурге состоялся Экспертный совет под председательством главного внештатного специалиста по инфекционным болезням Минздрава России профессора В.П. Чуланова. Цель. Рассмотрение следующих вопросов: анализ опыта применения препарата левилимаб в качестве патогенетической противовоспалительной терапии COVID-19 у госпитализированных пациентов; обсуждение результатов применения и перспектив упреждающей терапии левилимабом на догоспитальном этапе медицинской помощи с определением оптимальных критериев назначения препарата. Результаты. С учетом проанализированных данных по клинической эффективности и безопасности экспертами предложены рекомендации по применению препарата левилимаб на стационарном и догоспитальном этапах медицинской помощи при COVID-19.

Abstract

To analyze the results of direct and transgenerational effects of radio frequency electromagnetic fields (RF-EMF) on the model organism of crustaceansThe results of our study show that exposure of RF-EMF at juvenile period can significantly affect the fertility and size of irradiated daphnids and their offspring of the first generation. The decrease in fertility may be associated with a cytotoxic effect on the cells of irradiated animals. The reduction in the size of the terminal spine and the body of individuals is an indicator of the negative impact of radiation on the protective strategy of the crustacean population. The reproductive process is restored by the second generation. The results of our study provide further insights into the possible mechanisms underlying the in vivo effects of RF-EMF.

Abstract

Modern biomedical technologies development affords to provide the doctor with colossal amount of information about patients organism condition. However, the opportunity of using this data for medical diagnosis fully now is a distantive perspective only. The reason is a humans limited ability in assessment and interpretation this data arrays. The solution seems in artificial intelligence and expert systems wide introduction to medicine. Currently, almost all authors consider various options for constructing artificial neural networks as a way to implement artificial intelligence. This approach, which goes back to the fundamental theorem of A.N. Kolmogorov, the works of V.I. Arnold and Hecht-Nielsen [3], demonstrates excellent capabilities in a number of pattern recognition problems, which are reduced to revealing hidden details against the background of input noises. Much less often is mentioned such a method of modeling formal thinking as expert systems, which arose in the 1960s and then went into the shadows. Since the inception of cybernetics, computer programmers have tried to reproduce the mechanism of human thinking, that is, the task was to teach the computer to "think". The first known results in the field of creating and using intelligent systems were laid by the work of Norbert Wiener and G.S. Altshuller. At the same time, the creation of intelligent systems was reduced to the development of programs that solve problems using a variety of heuristic methods based on the property of human thinking to generalize.

The ratio of the radiation energy of the plasma relativistic microwave amplifier at a frequency of the input signal to the noise energy is measured. In the absence of the input signal, the noise radiation is detected. It is shown that, under certain conditions, the amplifier noise energy sharply decreases, as compared to the noise energy measured in the absence of the input signal.

Abstract

Many authors note the difficulties in qualifying weapons of mass destruction as an object of a criminal offense. The article is devoted to the problems of understanding the essence of the concept of "weapons of mass destruction" in modern criminal law and directly addresses the issues related to the disclosure of the concept of the subject in the investigation of this type of crime. The article analyzes the concept of "weapons of mass destruction" as one of the elements of the criminalistic characteristics of its smuggling, which serves as the basis for putting forward versions and seeing tactical prospects for investigating this type of crime. The article considers the role of the customs authorities of Russia as a paramilitary and law enforcement structure by its status, which provides national security of the state.

Abstract

A supersonic flow around a cylinder with a flat end is studied for the case in which a single particle is launched from the end surface towards the incident flow. Numerical modeling is carried out with allowance for the gas-dynamic interaction of the particle with the shock layer. High-resolution, adaptive, rectangular grids are used to obtain detailed spatial and temporal flow patterns. The calculation results are compared with experimental data. It is shown that a single, large particle moving along the axis of symmetry against the incident flow and crossing the bow shock wave causes a significant restructuring of the flow and the formation of complex shock-wave and vortex structures. An important role in the formation of the flow structure is played by the formation of a toroidal vortex, which leads to an “inviscid” separation of the incident flow from the axis of symmetry. A distinctive feature of the flow around a cylinder with a flat end is the oscillatory flow regime caused by alternating stages of growth and decay of a toroidal vortex. The patterns of the oscillating flow and the oscillation frequency obtained via numerical simulation are in good agreement with the experimental data.

Abstract

The paper is devoted to the numerical study of the shock layer–particle interaction in a supersonic flow past bodies. The shock wave associated with a moving particle and the flow in the particle wake is taken into consideration. Distinctive features of the numerical technique are the adaptive Cartesian sliding grids of high resolution, a ghost cells immersed boundary method for the realization of conditions on curvilinear boundaries, parallelization of computations on graphics processors. The results of computational experiment obtained allows to study the characteristic shock and vortex structures formed during the passage of a particle rebounding from the surface through the bow shock wave. The oscillations observed for a flow over the fla-tended cylinder are discussed.

Abstract

The article is devoted to understudied issues in the intellectual heritage of professor archpriest Dumitru Staniloae, an outstanding Romanian theologian of the twentieth century. More precisely, the article examines the journalistic articles that reflected his philosophical, cultural, social, political, and religious views, which as a whole can be organized into a model of “political ideology”. At the same time, the author considers the attitude to the issues of combining the Orthodox worldview, patriotism, and statehood in the context of developing the conceptual and ideological field of the idea of “Romanism”, which was established in the situation of territorial and social changes in Romania during the interwar period. In this regard, the religious “Romanian traditionalism” becomes the key triad of “Orthodoxy — collegiality — tradition”, which is equally dissociated from the materialistic-atheistic ideologies of both the democratic West with its individualism and capitalism, and the Bolshevik East with its authoritarianism and collectivism. The views of the Romanian theologian are presented on the material of some of his publications in the journal Gandirea and the newspaper Telegraful roman. Understanding the specifics of the ideological field of the Orthodox Romanian national and state self-identification, in the context of the formation of a new map of Europe in the 30s, can be taken into account when analyzing elements of the conceptual paradigm of Orthodox “political theology”.

Abstract

During the long-term operation of buildings, damage is accumulated in structural elements leading to violations of strength bonds, especially during seismic impacts. Therefore, for buildings and structures located in areas having high seismic activity, periodic monitoring of the technical condition of buildings is recommended. Given the dependence of dynamic characteristics on the integral strength properties of structures, monitoring should be based on a study of the dynamic characteristics of building structures. The present studies were carried out using a microdynamic method based on a determination of the transfer functions between vibrations at the nodal points of the structure and microseismic noise measured at the building base. The actual mechanical properties of the supporting structures take into account the spatial work of the building as a whole under the action of loads. Affecting any design, earth-quakes are acknowledged to result in vibration effects leading to structural instability including base soils. The direction of mechanical impact development during an earthquake must also be taken into account. The paper presents the results of dynamic response studies into microseismic effects affecting an 8-storey frame building. The initial natural frequencies and modes of vibration are determined along with the damping constants to form a basis for the monitoring of the changes in the technical condition of the building following a given period of operation.

Abstract

Anaphylatoxin C3a is a small signaling polypeptide that is generated during complement activation. C3a is involved in the regulation of various innate and adaptive immune system processes; however, the role of C3a in macrophage differentiation and polarization is poorly elucidated. Here we showed that C3a impairs alternative M2 polarization of human macrophages and suppressed CD206, IL1Ra and CCL22 expression. C3a leads to a decrease of nuclear receptor PPARγ expression via the ERK1/2 signaling pathway, resulting in repressed PPARγ-dependent activation of CD36, FABP4 and LXRα genes and blunted response to an LXR ligand TO901317. Using small interfering RNA and agonist/antagonist approaches we showed that C3a decreases CD206, IL1Ra and CCL22 transcription at least partly in a PPARγ-dependent manner in M2 macrophages. Moreover, C3a impairs efferocytosis by M2 macrophages and inhibits their migratory activity. By contrast, macrophages treated with C3a during differentiation show blunted response to lipopolysaccharide stimulation owing to downregulation of TLR4 and lipid raft content. At the same time, differentiation of macrophages with C3a does not change M1 polarization in interferon gamma (IFNγ) and IFNγ + lipopolysaccharide-treated macrophages. These data provide a novel role of complement system and C3a in the regulation of M2 macrophage polarizations and suggest crosstalk between C3a, TLR4, PPARγ and LXR signaling pathways.

Abstract

The purpose of this study is to assess the effect of doping ZrO2 ceramics with MgO on radiation swelling and polymorphic transformations, as a result of irradiation with heavy ions. Interest in these types of materials is due to the great prospects for their use as structural materials for new-generation reactors. The study established the dependences of the phase composition formation and changes in the structural parameters following a change in the concentration of MgO. It has been established that the main mechanism for changing the structural properties of ceramics is the displacement of the cubic c-ZrO2 phase by the Zr0.9Mg0.1O2 substitution phase, which leads to an increase in the stability of ceramic properties to irradiation. It has been determined that an increase in MgO concentration leads to the formation of an impurity phase Zr0.9Mg0.1O2 due to the type of substitution, resulting in changes to the structural parameters of ceramics. During studies of changes in the strength properties of irradiated ceramics, it was found that the formation of a phase in the Zr0.9Mg0.1O2 structure leads to an increase in the resistance to cracking and embrittlement of the surface layers of ceramics.

Abstract

Despite the widespread use of the COVID-19 vaccines, the search for effective antiviral drugs for the treatment of patients infected with SARS-CoV-2 is still relevant. Genetic variability leads to the continued circulation of new variants of concern (VOC). There is a significant decrease in the effectiveness of antibody-based therapy, which raises concerns about the development of new antiviral drugs with a high spectrum of activity against VOCs. We synthesized new analogs of uracil derivatives where uracil was substituted at the N1 and N3 positions. Antiviral activity was studied in Vero E6 cells against VOC, including currently widely circulating SARS-CoV-2 Omicron. All synthesized compounds of the panel showed a wide antiviral effect. In addition, we determined that these compounds inhibit the activity of recombinant SARS-CoV-2 RdRp. Our study suggests that these non-nucleoside uracil-based analogs may be of future use as a treatment for patients infected with circulating SARS-CoV-2 variants.
