Mining areas characterized by high underground water levels are one of the most important types of coal mining areas in China. In regions with high groundwater levels, the soil ecological environment is destroyed due to surface subsidence induced by coal mining and soil disturbances. There are a variety of soil factors each with different degrees of spatial variation, and the impact on soil microbial communities is particularly severe. In order to explore the change and driving mechanism of soil microbial community structure in coal mining subsidence areas with high underground water levels, we sought to elucidate these mechanisms by studying soil samples collected at different depths (SL: 0-20 cm, ML: 20-40 cm, DL: 40-60 cm) of a deep coal seam subsidence area (T1) and shallow coal seam subsidence area (T2) and their control non-subsidence areas (W1 and W2) within a typical coal mine area with high underground water levels in southwest Shandong Province. These soil samples were used for determination and analysis of their physicochemical properties and microbial diversity. The results show that coal mining subsidence has significant effects on the soil physicochemical properties and soil microbial community. With the increase in sampling depth, the soil water content (SWC), bulk density (BD), and soil pH increased, whereas the contents of soil alkali-hydrolyzable nitrogen (AN), available phosphorus (AP), available potassium (AK), and soil organic matter (SOM) decreased. Compared with the non-subsidence area, the soil alkalinity in the subsidence area was lower and the soil moisture content, affected by the underground water level, was higher; the richness and diversity of the microbial community was lower in the subsidence area despite its higher relative abundance of Actinobacteria, Chloroflexi, and Myxomycota species. In addition, species of Thelebolales and Pleosporales were dominant in T1 and T2, respectively. Soil pH was observed to be the most important physicochemical factor affecting microbial communities, followed by AN and AP. The results of our study provide a theoretical basis for soil ecological restoration and land reclamation in mining areas with high underground water levels.

Microvascular invasion (MVI) is an independent detrimental risk factor for tumor recurrence and poor survival in hepatocellular carcinoma (HCC). Competitive endogenous RNA (ceRNA) networks play a pivotal role in the modulation of carcinogenesis and progression among diverse tumor types. However, whether the ceRNA mechanisms are engaged in promoting the MVI process in patients with HCC remains unknown.A ceRNA regulatory network was constructed based on RNA-seq data of patients with HCC from The Cancer Genome Atlas (TCGA) database. In total, 10 hub genes of the ceRNA network were identified using four algorithms: "MCC," "Degree," "Betweenness," and "Stress." Transcriptional expressions were verified by in situ hybridization using clinical samples. Interactions between ceRNA modules were validated by luciferase reporting assay. Logistic regression analysis, correlation analysis, enrichment analysis, promoter region analysis, methylation analysis, and immune infiltration analysis were performed to further investigate the molecular mechanisms and clinical transformation value.The ceRNA regulatory network featuring a tumor invasion phenotype consisting of 3 long noncoding RNAs, 3 microRNAs, and 93 mRNAs was constructed using transcriptional data from the TCGA database. Systemic analysis and experimentally validation identified a ceRNA network (PVT1/miR-1258/DUSP13 axis) characterized by lipid regulatory potential, immune properties, and abnormal methylation states in patients with HCC and MVI. Meanwhile, 28 transcriptional factors were identified as potential promotors of PVT1 with 3 transcriptional factors MXD3, ZNF580, and KDM1A promising as therapeutic targets in patients with HCC and MVI. Furthermore, miR-1258 was an independent predictor for MVI in patients with HCC.The PVT1/DUSP13 axis is significantly associated with MVI progression in HCC patients. This study provides new insight into mechanisms related to lipids, immune phenotypes, and abnormal epigenetics in oncology research.

Sugar beet (Beta vulgaris L.) is an important sugar-producing and energy crop worldwide. The sugar beet pure line IMA1 independently bred by Chinese scientists is a standard diploid parent material that is widely used in hybrid-breeding programs. In this study, a high-quality, chromosome-level genome assembly for IMA1was conducted, and 99.1% of genome sequences were assigned to nine chromosomes. A total of 35,003 protein-coding genes were annotated, with 91.56% functionally annotated by public databases. Compared with previously released sugar beet assemblies, the new genome was larger with at least 1.6 times larger N50 size, thereby substantially improving the completeness and continuity of the sugar beet genome. A Genome-Wide Association Studies analysis identified 10 disease-resistance genes associated with three important beet diseases and five genes associated with sugar yield per hectare, which could be key targets to improve sugar productivity. Nine highly expressed genes associated with pollen fertility of sugar beet were also identified. The results of this study provide valuable information to identify and dissect functional genes affecting sugar beet agronomic traits, which can increase sugar beet production and help screen for excellent sugar beet breeding materials. In addition, information is provided that can precisely incorporate biotechnology tools into breeding efforts.

Tertiary lymphoid structures (TLSs) play key roles in tumour adaptive immunity. However, the prognostic value and molecular properties of TLSs in oesophageal squamous cell carcinoma (ESCC) patients have not been studied.The prognostic values of the presence and maturation status of tumour-associated TLSs were determined in 394 and 256 ESCC patients from Sun Yat-sen University Cancer Center (Centre A) and the Cancer Hospital of Shantou University Medical College (Centre B), respectively. A deep-learning (DL) TLS classifier was established with haematoxylin and eosin (HE)-stained slides using an inception-resnet-v2 neural network. Digital spatial profiling was performed to determine the cellular and molecular properties of TLSs in ESCC tissues.TLSs were observed in 73.1% of ESCCs from Centre A via pathological examination of HE-stained primary tumour slides, among which 42.9% were TLS-mature and 30.2% were TLS-immature tumours. The established DL TLS classifier yielded favourable sensitivities and specificities for patient TLS identification and maturation evaluation, with which 55.1%, 39.5% and 5.5% of ESCCs from Centre B were identified as TLS-mature, TLS-immature and TLS-negative tumours. Multivariate analyses proved that the presence of mature TLSs was an independent prognostic factor in both the Centre A and Centre B cohorts (p.05). Increased proportions of proliferative B, plasma and CD4+ T helper (Th) cells and increased B memory and Th17 signatures were observed in mature TLSs compared to immature ones. Intratumoural CD8+ T infiltration was increased in TLS-mature ESCC tissues compared to mature TLS-absent tissues. The combination of mature TLS presence and high CD8+ T infiltration was associated with the best survival in ESCC patients.Mature TLSs improve the prognosis of ESCC patients who underwent complete resection. The use of the DL TLS classifier would facilitate precise and efficient evaluation of TLS maturation status and offer a novel probability of ESCC treatment individualization.

Emerging evidence suggests different intratumoral microbiome exists in different tumors and plays a crucial function in carcinogenesis. However, whether intratumoral microbiome in esophageal squamous cell carcinoma (ESCC) impacts clinical outcomes and its mechanism remains unclear. 16S rDNA amplicon sequencing was performed on surgical resected samples from 106 ESCC patients including 25 paired tumorous and non-tumorous tissues (NTs) to analyze intratumoral microbiome abundance and its composition. Multiplex fluorescent immunohistochemistry (mfIHC) staining was used to profile the phenotypes of immune infiltrates in tumor microenvironment (TME). Both Shannon index and operational taxonomic units (OTUs), representing alpha-diversity of microbiome, were found significantly lower in tumors comparing to the corresponding NTs. Multivariate analysis revealed that the intratumoral Shannon index, the relative abundance of Lactobacillus and the pTNM stage were independently associated with patients’ overall survival. Furthermore, the relative abundance of Lactobacillus and Shannon index were both positively correlated with the proportions of PD-L1+ epithelial cells (ECs) and tumor-associated macrophages (TAMs, and Shannon index was negatively correlated with the proportions of natural killer cells (NKs) and activated cytotoxic T cells (aCTLs) in TME, indicating an immunosuppressive microenvironment. These findings underscored for the first time that the high abundance of intratumoral Lactobacillus and microbiome alpha-diversity were associated with poor long-term survival of ESCC patients, which might be achieved by increasing PD-L1 + ECs and PD-L1 + TAMs and decreasing aCTLs and NKs in TME.

With global urbanization and industrialization, environmental pollution and food safety problems caused by soil heavy metal pollution occur frequently. To realize the safe utilization of cultivated land resources in high-risk areas of heavy metal pollution, we present an approach to safe utilization classification and management in this study. A typical agricultural area around industrial and mining enterprises located in the economic belt of the Yangtze River Delta was chosen as the research area with cultivated soil as the research object. A total of 1,139 geochemical survey sampling sites and soil survey data were used for this research. Initially, the potential ecological risk index was used to assess the potential ecological risk of heavy metals in cultivated soil, and key soil physical and chemical indicators were chosen to assess soil resilience. Next, the safe utilization classification of cultivated land was carried out by combining the potential ecological risk of soil heavy metals with soil resilience. Then, the specific classification management strategy was developed according to ecological risk factor types and crop types. The results showed that the production and operation activities of industrial and mining enterprises in the study area contributed significantly to soil Cd and Hg pollution, and the potential ecological risk of heavy metals in the surrounding soil was high. The soil resilience of cultivated land in the study area was generally not high. The clay content and cation exchange capacity (CEC) were the main strong restrictive indicators, while CaO, soil organic carbon (SOC), and pH were the main medium restrictive indicators. Natural conditions such as topography, hydrogeology, and soil-forming parent material differed by region, resulting in different restrictive factors. There were 16 safe utilization types in the study area. Different management strategies were proposed based on various soil potential ecological risks, soil resilience, ecological risk factor types, and crop types. This study can offer fresh perspectives on the safe utilization classification and management of land resources in high-risk areas of soil pollution and serve as a reference for sustainable intensification.

One hundred sixty-two parallel bamboo strand lumber (PBSL) beams were tested for the bending properties under three-point bending with variable span. Two typical failure types were classified, type I was tension failure of the bamboo fiber located below the neutral axis at mid-span, while type II was shear failure which was mainly found in specimens with large depth-span ratio. The shear modulus $${\text{G}}_{{{\text{LR}}}}$$ of PBSL was also calculated according to Timoshenko’s bending theory. To evaluate the shear modulus, 45° off-axis compression tests had been done. The results showed that the mean values of shear modulus for $${\text{G}}_{{{\text{LT}}}}$$ , $${\text{G}}_{{{\text{TR}}}}$$ , and $${\text{G}}_{{{\text{LR}}}}$$ are 1254.5 MPa, 408.1 MPa, and 716.4 MPa, respectively. The bending test result was compared with the shear modulus measured by 45° off-axis compression test. It was found that the shear modulus measured by three-point bending test with variable span was significantly less than the shear modulus obtained by 45° off-axis compression test. The empirical equation of shear factor s corresponding to depth-span ratio h/l was proposed. The calculation results gained from the proposed formula are in good agreement with the 45° off-axis compression test results.

Increases in government debt are associated with a reduction in the yield spread between high-grade corporate bonds and long-term Treasuries and an increase in fiscal uncertainty. Consequently, increases in government debt significantly reduce the firm’s likelihood of acquisition. The effect is stronger among firms whose debt is a closer substitute for Treasuries and firms with greater exposure to fiscal uncertainty. A positive change in government debt motivates acquirers to avoid cash financing or more irreversible deals. The average deal quality is lower during periods of rising public debt, consistent with heightened fiscal uncertainty impeding monitoring and fostering “bad” deals. We examine how an M&A advisor’s position in the network of investment banks affects its ability to create value for acquirers in takeover transactions. We show that acquirers enlisting the services of more centrally positioned M&A advisors enjoy higher announcement abnormal returns and pay lower takeover premiums. Consistent with the idea that central network positions convey an information advantage, we find that the effects are stronger for acquirers facing greater target information asymmetry and for M&A advisors depending more on networks for target-specific information. The information advantage primarily comes from network contacts that had previously assisted the targets in equity issuance. Centrally positioned advisors charge premium fees; network banks appear to enjoy a significant advantage in the competition for future co-advisory appointments.

Objective Colon cancer (CC) is the third most common cancer with a high rate of incidence and mortality. Therefore, it is highly necessary to explore novel targets of CC. Methods The miRNA-seq and RNA-seq data of CC were accessed from the TCGA database. Differential analysis was performed using the "edgeR" package to identify differentially expressed miRNAs (DE_miRNAs). The downstream target genes of the target miRNA were then predicted by miRNA target prediction databases to identify the target mRNA. Normal colon cell line CCD-18Co and CC cell lines HCT-116, HT-29, SW620 and SW480 were chosen, and qRT-PCR was conducted to detect miR-141 expression in these cell lines. qRT-PCR and Western blot were carried out to determine PHLPP2 mRNA and protein expression, respectively. Dual-luciferase reporter gene assay was performed to verify the targeting relationship between miR-141 and PHLPP2 3'UTR. CCK-8 assay and colony formation assay were carried out to detect cell proliferation. Meanwhile, tumor xenograft model in nude mice was constructed to assess CC cell tumorigenic ability in vivo. Results miR-141 was markedly up-regulated in CC tissue. CC cell proliferation and in vivo tumorigenic ability were suppressed by miR-141 silencing but promoted by miR-141 over-expression. PHLPP2 was significantly down-regulated in cancer tissue. Dual-luciferase reporter gene assay indicated that miR-141 could bind to PHLPP2 3'UTR. PHLPP2 expression was noticeably elevated upon miR-141 deficiency but significantly inhibited upon miR-141 over-expression. CCK-8 and colony formation assay suggested that miR-141 facilitated CC cell proliferation by silencing PHLPP2. Conclusion miR-141 promotes CC cell proliferation by targeted silencing PHLPP2.

Education is one of the areas with a higher impact on digitalization, which takes various forms such as education through digital devices and technology to improve the learning process. Online and tangible computing platforms have become more interested in pursuing active teaching through educational technologies within the curriculum. Some common problems and considerations can be addressed, such as access, capacity, financing, periodic progress measurements, and evaluation results. The productive learning activity can be immediately and constantly monitored by proposing Digital Tangible Intelligent Monitoring systems (DTIMS). The decision tree method’s teaching methodology can efficiently perform this proposed system, which monitors the former challenges. At the same time, the next is resolved by a dynamic evaluation process based on the Internet of Things (IoT). The research is evaluated using the education systems currently adopted. The results highlighted the potential of the proposed model and helped to gain information in digital teaching. The simulation analysis is performed based on accuracy 97.69%, vulnerability 91.09%, and efficiency, proving the proposed framework’s reliability of 85.10%.

Hand, foot, and mouth disease (HFMD) caused by Enterovirus type 71 (EV71) is a serious threat to children's health. However, the pathogenic mechanism of EV71 is still unclear. Long non-coding RNAs (lncRNAs), some of which bind to miRNA as competitive endogenous RNAs (ceRNA) and weaken the silencing effect on the mRNA of downstream target genes, play a key role in regulating the viral infection process. In this study, through experimental verification, we found miR-4443 to be downregulated in cells infected with EV71. Next, by predicting lncRNAs that potentially regulate miR-4443, we found that EV71 infection induced upregulation of lncRNA ENST00000469812 and then further downregulated miR-4443 expression by direct interaction. We also demonstrated that nuclear protein 1 (NUPR1) is one of the target genes of miR-4443 and is involved in the ENST00000469812/miR-4443/NUPR1 regulatory axis. Finally, the ENST00000469812/miR-4443/NUPR1 regulatory axis exhibited a positive effect on EV71 replication. Here, we lay a foundation for exploring the pathogenic mechanism of EV71 and identify potential targets for HFMD treatment.

In recent years, there has been progress in the treatment of breast cancer; however, the prognosis is still poor due to recurrence and metastasis following conventional treatment. The anti-tumor peptide SA12 has been demonstrated to inhibit proliferation and arrest the cell cycle in MDA-MB-231 and MCF-7 breast cancer cells. In the present study, whether SA12 was able to inhibit the metastasis of breast cancer cells was investigated. Wound healing and Transwell assays were used to investigate the inhibition of SA12 on cell migration while, reverse transcription-quantitative PCR and western blot assays were used to identify the mechanism of action behind the effects of SA12 on cell migration. Results from the wound healing and Transwell assays revealed that SA12 significantly inhibited the migration of MDA-MB-231 and MCF-7 breast cancer cells following treatment with 100 µM SA12. Compared with that in the controls, the mRNA expression levels of cadherin 1 (CDH1), non-metastasis 23-H1 (nm23-H1) and breast cancer metastasis suppressor 1 (BRMS1) were increased in MDA-MB-231 and MCF-7 cells following treatment with 100 µM SA12. Furthermore, the protein expression levels of E-cadherin, NM23A and BRMS1 were also increased in MDA-MB-231 cells and MCF-7 cells following treatment with 100 µM SA12. In conclusion, SA12 inhibited the migration of MDA-MB-231 and MCF-7 breast cancer cells and enhanced the expression of the tumor metastasis suppressor genes, CDH1, nm23-H1 and BRMS1, which may be responsible for the SA12-induced inhibition of breast cancer cell metastasis.

In order to investigate the basic mechanical properties and stress strain relationship model for bamboo scrimber manufactured based on a new technique, a large quantities of experiments have been carried out. Based on the analysis of the test results, the following conclusions can be drawn. Two main typical failure modes were classified for bamboo scrimber specimens both under tension parallel to grain and tension perpendicular to grain. Brittle failure happened for all tensile tests. The slope values for the elastic stages have bigger discreteness compared with those for the specimens under tensile parallel to grain. The failure modes for bamboo scrimber specimens under compression parallel to grain could be divided into four. Only one main failure mode happened both for the bending specimens and the shear specimens. With the COV values of 28.64 and 25.72 respectively, the values for the strength and elastic modulus under tensile perpendicular to grain have the largest discreteness for bamboo scrimber. From the point of CHV values, the relationship among the mechanical parameters for bamboo scrimber were proposed based on the test results. Compared with other green building materials, bamboo scrimber manufactured based on a new technique has better mechanical performance and could be used in construction area. Three stress strain relationship models which are four-linear model, quadratic function model, and cubic function model were proposed for bamboo scrimber specimens manufactured based on a new technique. The latter two models gives better prediction for stress strain relationship in elastic plastic stage.

Objective The aim of this study was to evaluate the changes and diagnostic value of serum ADA activity in autoimmune diseases, including systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), ankylosing spondylitis (AS), and myasthenia gravis (MG). Methods Serum ADA activity, including total ADA (tADA) and its isoenzymes (ADA1 and ADA2), was determined in patients with different autoimmune diseases (144 RA, 114 SLE, 55 AS, 68 MG). The changes in serum ADA activity in patients were analysed. A receiver operating characteristic (ROC) curve analysis was applied to evaluate the diagnostic performance of serum ADA activity. Results Compared with healthy controls, the serum tADA activity in SLE patients was significantly increased (p0.05). The ROC analysis showed that the optimal cut-off value of serum tADA activity for SLE diagnosis was 10.5 U/L (79.8% specificity and 74.6% sensitivity; likelihood ratio (LR): 3.693; p Conclusion These findings suggest that serum tADA and ADA2 activity might play an important role in SLE progression.

Indoleamine 2,3 dioxygenase 1 (IDO1) is an attractive target for cancer immunotherapy. However, IDO1 inhibitors have shown disappointing therapeutic efficacy in clinical trials, mainly because of the activation of the aryl hydrocarbon receptor (AhR). Here, we show a post-transcriptional regulatory mechanism of IDO1 regulated by a proteasome-associated deubiquitinating enzyme, USP14, in colorectal cancer (CRC). Overexpression of USP14 promotes tryptophan metabolism and T-cell dysfunction by stabilizing the IDO1 protein. Knockdown of USP14 or pharmacological targeting of USP14 decreases IDO1 expression, reverses suppression of cytotoxic T cells, and increases responsiveness to anti-PD-1 in a MC38 syngeneic mouse model. Importantly, suppression of USP14 has no effects on AhR activation induced by the IDO1 inhibitor. These findings highlight a relevant role of USP14 in post-translational regulation of IDO1 and in the suppression of antitumor immunity, suggesting that inhibition of USP14 may represent a promising strategy for CRC immunotherapy.

The unsatisfactory sensory function reconstruction after flap transplantation to repair tissue defects and reconstruct organs results in decreased quality of life. Schwann cells (SCs) can promote sensory function reconstruction, but the underlying mechanism is not completely understood. The aim of the present study was to explore the in‑depth mechanism underlying SCs in sensory function reconstruction. Sciatic nerve transection and a repair animal model were performed to evaluate the effect of SC‑like cells (iSCs) and a neurotrophin 3 (NT‑3) chitosan conduit. SC RNA‑seq data indicated that the SOX2/fibronectin 1 (FN1) axis promoted proliferation and migration, which are the cytological bases of nerve regeneration. Subsequently, the effects of SOX2, FN1 and exosomes secreted by iSCs on SC proliferation and migration were assessed using scratch wound and EdU assays, respectively. The RNA‑seq of SCs indicated that SOX2 overexpression increased iSC viability and migration. Furthermore, SOX2 increased FN1 expression to promote nerve recovery by fibronectin fibrillogenesis. In addition, exosomes secreted by iSCs increased SC viability and migration. In conclusion, iSCs with an NT‑3 chitosan conduit promoted sciatic nerve recovery via the SOX2/FN1 axis and exosomes secreted by iSCs. Therefore, the present study identified potential effective therapeutic approaches for sciatic nerve repair.

Chemotherapy is one of the vital treatments for gastric cancer (GC) patients, especially those suffering advanced stages. Chemoresistance results in tumor relapse, leading to poor prognosis in GC patients; thus, identifying key regulators in this process might provide novel clues for GC therapy. Herein, we identify hyaluronan-mediated motility receptor (HMMR) as a key regulator of chemoresistance in GC. HMMR was found to be substantially up-regulated in 5-fluorouracil (5-Fu)-resistant GC biopsies and cell lines. High expression of HMMR significantly correlates with tumor relapse and predicts poorer prognosis in GC patients. Moreover, we observed that HMMR induced epithelial-mesenchymal transition and increased the cancer stem cell properties of GC, thus rendering resistance to chemotherapy. Importantly, silencing of HMMR effectively increased the susceptibility to 5-Fu therapy both in vitro and in vivo. Furthermore, we demonstrated that HMMR activates the TGF-β/Smad2 signaling pathway, which was required for the HMMR-mediated oncogenic effects and exhibited significant clinical relevance with HMMR expression. These findings reveal a critical role for HMMR in the chemoresistance of GC and suggest that HMMR might be a potential prognostic marker or therapeutic target against the disease.-Zhang, H., Ren, L., Ding, Y., Li, F., Chen, X., Ouyang, Y., Zhang, Y., Zhang, D. Aims Keloids are a dermal fibrotic disease whose etiology remains totally unknown and for which there is no successful treatment. Mechanical tension, in addition, is closely associated with the germination and development of keloids. In this study, we investigated the influence of human keloid-derived mesenchymal stem cells (KD-MSCs) on cell proliferation, collagen synthesis, and expressions of integrin αvβ3 under tension. Main methods KD-MSCs and human normal skin-derived mesenchymal stem cells (NS-MSCs) were isolated and cultured in stem cell medium with a gradual increase in the serum concentration. Cell proliferation and collagen synthesis were detected by Cell Counting Kit-8 (CCK-8) assay and hydroxyproline content analysis under tension respectively. We investigated the messenger RNA expressions of nine integrin subunits, including integrin units α2, α3, α5, αv, α8, α10, α11, β1, and β3, in KD-MSCs stimulated with tension. Identification of differentially expressed genes was performed by Western blot analysis and immunocytochemistry staining. Key findings We obtained high-purity KD-MSCs and NS-MSCs using the culture method of decreasing serum concentration gradient gradually. Furthermore, we found that tension enhances cell proliferation and collagen synthesis and promotes expressions of integrin αvβ3 in KD-MSCs. In addition, blocking experiments showed that increased integrin αvβ3 expression affects cell proliferation and collagen synthesis of KD-MSCs under tension. Significance Our results suggest that integrin αvβ3 receptor may be sensitive molecules of mechanical tension and could contribute to the occurrence and development of keloids. It could lead to novel targets for therapeutic intervention, treatment, and prevention of recurrence for keloid disorders.

The competing endogenous RNA (ceRNA) axis has been shown to play a critical role in the pathogenesis of various viral infections. Generally, the ceRNA network involves long non-coding RNAs (lncRNAs) that act as sponges for miRNA to regulate mRNA expression. However, no information is available regarding the involvement of ceRNA networks in Enterovirus type 71 (EV71) infections. In the present study, data obtained from Gene Expression Omnibus (GEO) database was analyzed using various bioinformatics tools. EV71 infection in rhabdomyosarcoma (RD) cells was associated with differential expression of six lncRNAs, 28 miRNAs, and 349 mRNAs. Gene function enrichment analysis suggested induction of cytoplasmic vesicle process upon EV71 infection. The ceRNA networks were constructed, in which 20 hub genes were predicted by protein-protein interaction. To confirm the MALAT1/miR-194-5p/DUSP1 ceRNA regulatory axis in EV71 infection, real-time quantitative polymerase chain reaction (qRT-PCR) and luciferase reporter assay were performed. The results of the study also revealed the involvement of the MALAT1/miR-194-5p axis in apoptosis induced by EV71 infection, while no association with autophagy was observed. Thus, the present study provided novel insights into the pathogenic mechanism of EV71 infection.

Objective The paper analyzes the influence of hot built-in objects on the performance of corrugated carton, and provides reference to select corrugated carton packing hot built-in objects. Methods Choose six sample corrugated carton packing hot built-in objects as follows: ordinary carton, waterproof coated carton, moisture proof starch adhesive carton, color printing varnish carton, color printing UV oil carton and color printing covering film carton. After loading hot built-in objects in the preset temperature, environment humidity and time conditions, the performance of carton was detected, and then the influence of hot built-in objects on the performance of carton was analyzed. Results Within the study conditions, as temperature, environment humidity and time change, the performance of six carton packing hot built-in objects showed different degree of change. After letting stand for single carton and stacking experiments for 24 h, the performance of carton also showed different degree of change. Conclusion According to the different storage temperature, environment humidity and user requirements, it is recommended to use a carton with good heat resistance.

TPS210 Background: Neoadjuvant chemoradiotherapy (NACRT) could bring tumor downstaging and pathological response (pCR), and also survival benefit for locally advanced rectal cancer (LARC) patients. Several single arm prospective clinical trials have investigated combination effect of immunotherapy (PD-1 or PD-L1 antibody) and NACRT in LARC patients, such as the VOLTAGE clinical trial. A randomized trial is needed to confirm the benefit of immunotherapy in this setting and explore predictive biomarkers. This is a clinical trial with two cohorts according the MMR/MSI status (clinicalTrials.gov, NCT04304209). Methods: In this study, LARC patients with pMMR/MSS/MSI-L tumor will enter cohort B and be randomized into two arms. Main inclusion criteria include: cT3-4N0M0 or cTxN+M0 rectal adenocarcinoma, pMMR/MSS/MSI-L confirmed by immunohistochemistry or gene test, aged 18-75y; ECOG performance 0-1; no previous anti-tumor treatment for rectal adenocarcinoma. Main exclusion criteria include: active autoimmune diseases or a history of autoimmune diseases, and inadequate main organ functions. Patients in the experimental arm will receive four cycles of neoadjuvant PD1 antibody Sintilimab, Capeox chemotherapy and concurrent radiotherapy, followed by curative surgery or watch and wait, then four cycles of adjuvant Capeox chemotherapy. Patients in the control arm will receive four cycles of neoadjuvant Capeox chemotherapy and concurrent radiotherapy, followed by curative surgery or watch and wait, then four cycles of adjuvant Capeox chemotherapy Primary outcome measure is pCR rate. Secondary outcome measures include acute toxicity, tumor regression grade, R0 resection rate, local recurrence, distant metastasis. Sample size for this cohort is 134. Whole exome sequencing, RNA sequencing and immunohistochemistry of the rectal primary tumor are planned for biomarker searching and synergy effect mechanism investigation.
