The clarity of nights is the major factor that should be carefully considered for optical/infrared astronomical observatories in site-testing campaigns. Cloud coverage is directly related to the amount of time available for scientific observations at observatories. In this article, we report on the results of detailed night-time cloud statistics and continuous observing me derived from ground-based all-sky cameras at the Muztagh-ata site from 2017 to 2021. Results obtained from acquisition data show that the proportion of the annual observing me at the Muztagh-ata site is 65%, and the best period with the least cloud coverage and longer continuous observing time is from September to February. We made a comparison of the monthly mean observing nights obtained from our all-sky cameras and CLARA dataset, results show that the discrepancy between them may depend on the cloud top heights. On average, this site can provide 175 clear nights and 169 nights with at least 4 hours of continuous observing time per year., Comment: 11 pages, 9 figures

Background Hepatocellular carcinoma (HCC) is the seventh most commonly occurring cancer and the second most common cause of cancer-related death worldwide. Despite improvements in early detection and treatment, the morbidity and mortality remain high because of complex molecular mechanisms and cellular heterogeneity in HCC. However, novel model is still needed to predict the survival and clinical immunotherapy response in HCC.Methods 13 iron metabolism-related gene sets were identified from the GSEA. DEGs associated with iron metabolism were calculated between patients who survived Results Here we established a gene signature using iron metabolism-related genes form the Cancer Genome Atlas (TCGA), and the survival outcomes were validated from International Cancer Genome Consortium(ICGC). Distinct subtypes (high- and low-risk subtypes) were characterized by different clinical outcomes. The high-risk subtype was featured by better survival outcomes, upregulated cell cycle relevant pathways and better response for immunotherapy.Conclusions Our finding suggested that the novel model may be useful as a biomarker for prognostic predication, immunotherapy and cell cycle inhibitors may be efficacious for high-risk subtype of HCC patients.

CA extern The secondary structures of hepatitis C virus (HCV) RNA and the cellular proteins that bind to them are important for modulating both translation and RNA replication. However, the sets of RNA-binding proteins involved in the regulation of HCV translation, replication and encapsidation remain unknown. Here, we identified RNA binding motif protein 24 (RBM24) as a host factor participated in HCV translation and replication. Knockdown of RBM24 reduced HCV propagation in Huh7.5.1 cells. An enhanced translation and delayed RNA synthesis during the early phase of infection was observed in RBM24 silencing cells. However, both overexpression of RBM24 and recombinant human RBM24 protein suppressed HCV IRES-mediated translation. Further analysis revealed that the assembly of the 80S ribosome on the HCV IRES was interrupted by RBM24 protein through binding to the 5′-UTR. RBM24 could also interact with HCV Core and enhance the interaction of Core and 5′-UTR, which suppresses the expression of HCV. Moreover, RBM24 enhanced the interaction between the 5′- and 3′-UTRs in the HCV genome, which probably explained its requirement in HCV genome replication. Therefore, RBM24 is a novel host factor involved in HCV replication and may function at the switch from translation to replication.

Blood pressure (BP) regulation is a complex homeostatic process involving multiple organ systems, including the cardiovascular, renal, and central and peripheral nervous systems.1 Inability to properly regulate BP can result in the development of hypertension, which is associated with an increased risk of stroke, myocardial infarction, and cardiac and renal failure,2–4 and is the leading determinant of risk of death worldwide.5 In addition to genetic and environmental factors known to play a role in the development of hypertension,6 body weight and obesity are highly correlated with hypertension in humans.7,8 This is particularly alarming given the increased prevalence of obesity, which now affects 1 in 3 people in the United States.9 The close association between obesity, hypertension, and other cardiovascular disorders suggests an important role for adipose tissue (AT) in the regulation of vascular tone and BP. Until the mid 1900s, AT was considered to be a form of connective tissue with the primary function of storing energy in the form of lipids.10 However, AT is now recognized to be an important endocrine organ that can secrete polypeptides and metabolites, known as adipokines, which are involved in the normal regulation of various physiological processes, including BP regulation and vascular function.10,11 In the setting of obesity, AT undergoes complex remodeling. This is marked by a significant increase in tissue mass, through adipocyte hyperplasia and hypertrophy. Changes in structural and cellular composition also occur, including changes in AT extracellular matrix, composition of lipid droplets, and infiltration of immune cells such as macrophages and lymphocytes, which results in low-grade inflammation.10,11 Most importantly, obesity shifts the secretory profile of AT, which in turn contributes to the dysregulation of BP and eventual development of hypertension.12 This is thought …

The Daocheng site is one of the three candidate sites for the Large Optical/infrared Telescope (LOT) of China. It was discovered by Yunnan Observatories during the survey of potential sites for the next-generation large-aperture solar telescopes of China. This paper describes the overview of the site, the observation platform and the monitor instrument. In addition, simple statistical results are presented (from November, 2016 up to December, 2017). Detailed data results can refer to the overview of LOT site testing and data analysis articles, which were published during the same period.

On the basis of field soil sampling, this paper investigated the distribution characteristics of soil different nitrogen (N) forms and its influence factors in the different typical zonal soils. Aiming at the paroxysmal fault of aircraft power system, this paper proposes a new method based on inverse problem of fuzzy optimization and deduces the corresponding fault diagnosis model. Firstly, establish the hybrid model of aircraft power system by Petri net. Secondly, analyze the relationship between fault symptoms and fault causes. Thirdly, combine the inverse problem with fuzzy diagnosis theory and get the inverse problem model of fuzzy optimization, the problem of diagnosis amounts to solving a fuzzy relational equation. Finally, solve the fuzzy relational equation by the numerical analysis method and do fault diagnosis by several fuzzy diagnosis rules. A case study demonstrates that the proposed model is feasible.

The majority of hepatitis C virus (HCV) infection develops chronic infection, which causes steatosis, cirrhosis and hepatocellular carcinoma. However, understanding HCV chronicity and pathogenesis is hampered by its narrow host range, mostly restricted to human and chimpanzee. Recent endeavour to infect a variety of humanized mice has not been able to achieve persistent HCV infection unless the essential innate immune responsive genes are knocked out. Nevertheless, such immune-compromised humanized mice still lacked HCV infection-induced hepatopathogenesis. Here we report that transgenic mice in ICR background harboring both human CD81 and occludin genes (C/OTg) are permissive to HCV infection at a chronicity rate comparable to humans. In this mouse model, HCV accomplishes its replication cycle, leading to sustained viremia and infectivity for more than 12 months post infection with expected fibrotic and cirrhotic progression. Host factors favorable for HCV replication, and inadequate innate immune-response may contribute to the persistence. Lastly, NS3/4 protease inhibitor telaprevir can effectively inhibit de novo RNA synthesis and acute HCV infection of C/OTg mice. Thus, chronic HCV infection with complete replication cycle and hepatopathologic manifestations is recapitulated, for the first time, in immune-competent mice. This model will open a new venue to study the mechanisms of chronic hepatitis C and develop better treatments.

Subgenomic replicons of hepatitis C virus (HCV) have been widely used for studying HCV replication. Here, we report a new subgenomic replicon based on a strain isolated from a chronically infected patient. The coding sequence of HCV was recovered from a Chinese chronic hepatitis C patient displaying high serum HCV copy numbers. A consensus sequence designated as CCH strain was constructed based on the sequences of five clones and this was classified by sequence alignment as belonging to genotype 2a. The subgenomic replicon of CCH was replication-deficient in cell culture, due to dysfunctions in NS3 and NS5B. Various JFH1/CCH chimeric replicons were constructed, and specific mutations were introduced. The introduction of mutations could partially restore the replication of chimeric replicons. A replication-competent chimeric construct was finally obtained by the introduction of NS3 from JFH1 into the backbone of the CCH strain.

How does an organism's internal state direct its actions? At one moment an animal forages for food with acrobatic feats such as tree climbing and jumping between branches. At another time, it travels along the ground to find water or a mate, exposing itself to predators along the way. These behaviors are costly in terms of energy or physical risk, and the likelihood of performing one set of actions relative to another is strongly modulated by internal state. For example, an animal in energy deficit searches for food and a dehydrated animal looks for water. The crosstalk between physiological state and motivational processes influences behavioral intensity and intent, but the underlying neural circuits are poorly understood. Molecular genetics along with optogenetic and pharmacogenetic tools for perturbing neuron function have enabled cell type-selective dissection of circuits that mediate behavioral responses to physiological state changes. Here, we review recent progress into neural circuit analysis of hunger in the mouse by focusing on a starvation-sensitive neuron population in the hypothalamus that is sufficient to promote voracious eating. We also consider research into the motivational processes that are thought to underlie hunger in order to outline considerations for bridging the gap between homeostatic and motivational neural circuits.

Analytical optimal hedging rules are derived for two-objective reservoir operation based on the characteristics of economic and multilevel environmental water demands. This approach is employed to balance current and future economic and environmental water demands using a two-period model. According to the marginal values of utilities, the derived three-step hedging rule curves (THRCs) consist of three steps and six stages. The Huaihe River basin of China is taken as an example to regress the THRCs from a two-objective optimization model. Results show that the derived THRCs can be regressed from the optimal release decision, thereby confirming the derivations and theoretical analysis. The shapes of the THRCs are sensitive to the weight parameters of the objectives and capture the core concern of multiobjective reservoir operation. The THRCs tend to widen and flatten the stages with increasing storage capacity, which implies more water storage and more consideration for future demands.

Recent findings suggest the reward system encodes metabolic value independent of taste, provoking speculation that the hedonic value of taste could be derived from nutritional value as a secondary appetitive property. We therefore dissociated and compared the impact of nutrition and taste on appetitive behavior in several paradigms. Though taste alone induces preference and increased consumption, in the absence of nutritional value its reinforcing properties are greatly diminished and it does not, like sucrose, induce increased responding over time. In agreement with behavioral data, saccharin-evoked (but not sucrose-evoked) dopamine release is greatly attenuated following pre-exposure, suggesting that nutritional value is critical for dopamine-mediated reward and reinforcement. Further supporting the primacy of nutrition over taste, genetically increased dopaminergic tone enhances incentive associated with nutritional value with minimal impact on taste-based, hedonic incentive. Overall, we suggest that the sensory-hedonic incentive value associated with taste functions as a conditioned stimulus that requires nutritional value to sustainably organize appetitive behavior.

Research on the biology of addiction has advanced significantly over the last 50 years expanding our understanding of the brain mechanisms underlying reward, reinforcement and craving. Novel experimental approaches and techniques have provided an ever increasing armory of tools to dissect behavioral processes, neural networks and molecular mechanisms. The ultimate goal is to reintegrate this knowledge into a coherent, mechanistic framework of addiction to help identify new treatment. This can be greatly facilitated by using tools that allow, with great spatial and temporal specificity, to link molecular changes with altered activation of neural circuits and behavior. Such specificity can now be achieved by using optogenetic tools. Our review describes the general principles of optogenetics and its use to understand the links between neural activity and behavior. We also provide an overview of recent studies using optogenetic tools in addiction and consider some outstanding questions of addiction research that are particularly amenable for optogenetic approaches.

We investigated whether changes in abscisic acid (ABA) content in leaves of Mikania micrantha infected by the holoparasite Cuscuta campestris at five growth stages, influenced the host stomatal conductance (gs), transpiration rate (E) and net photosynthetic rate (PN). C. campestris infection caused a negative effect on gs, E and PN of the host plants. ABA content in host leaves infected by C. campestris was significantly lower at 6 d after parasitization (DAP) and significantly higher at 13 and 33 DAP, relative to uninfected controls. In the parasite, ABA content was lowest at 13 DAP and then sharply increased to the maximum at 26 DAP. Moreover, the ABA content in the parasite was always lower than in the infected host leaves. The results suggest that an increase in host ABA concentration contributes to reduced host gs, E and PN in the holoparasitic C. campestris — M. micrantha association.

Both the dorsal and ventral striatum have been demonstrated to have a critical role in reinforcement learning and addiction. Dissecting the specific function of these striatal compartments and their associated nigrostriatal and mesoaccumbens dopamine pathways, however, has proved difficult. Previous studies using lesions to isolate the contribution of nigrostriatal and mesoaccumbens dopamine in mediating the locomotor and reinforcing effects of psychostimulant drugs have yielded inconsistent and inconclusive results. Using a naturally occurring mutant mouse line, aphakia, that lacks a nigrostriatal dopamine pathway but retains an intact mesoaccumbens pathway, we show that the locomotor activating effects of cocaine, including locomotor sensitization, are dependent on an intact nigrostriatal dopamine projection. In contrast, cocaine reinforcement, as measured by conditioned place preference and cocaine sensitization of sucrose preference, is intact in these mice. In light of the well-established role of the nucleus accumbens in mediating the effects of psychostimulants, these data suggest that the nigrostriatal pathway can act as a critical effector mechanism for the nucleus accumbens highlighting the importance of intrastriatal connectivity and providing insight into the functional architecture of the striatum.

The Large sky Area Multi-Object Spectroscopic Telescope (LAMOST) General Survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects both in the pilot survey and the first year general survey are included in the LAMOST First Data Release (DR1). The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The general survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2,955,336 spectra, of which 1,790,879 spectra have observed signal-to-noise S/N >10. All data with S/N>2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2,204,696 spectra, of which 1,944,329 are stellar spectra, 12,082 are galaxy spectra and 5,017 are quasars. The DR1 includes not only spectra, but also three stellar catalogues with measured parameters: AFGK-type stars with high quality spectra (1,061,918 entries), A-type stars (100,073 entries), and M stars (121,522 entries). This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. Aiming at the lack of technology for generator incipient condition monitoring and fault prediction, proposed SVM(Support Vector Machine) is introduced to generator incipient fault prediction. This paper takes the parametric faults of synchronous generator as an example, selects the output voltage as a monitoring signal, and combines with SVM regression prediction algorithms to achieve synchronous generators incipient fault prediction.

Fault tree modeling and analysis methods are being developed in academic and industrial. This paper designed a real-time diagnose method based on the .NET platform, combining the fault tree analysis (FTA) and real-time systems. Use SQL Server as the knowledge-base to store the information. Take the fault tree information and monitoring data as inputs, diagnose the real-time alarm of the system using minimal cut set algorithm (MCS).Finally test and verify the method by simulation.

Objective: Dopamine (DA) is critical for motor performance, motor learning, and corticostriatal plasticity. The relationship between motor performance and learning, and the role of DA in the mediation of them, however, remain unclear. Methods: To examine this question, we took advantage of PITx3-deficient mice (aphakia mice), in which DA in the dorsal striatum is reduced by 90%. PITx3-deficient mice do not display obvious motor deficits in their home cage, but are impaired in motor tasks that require new motor skills. We used the accelerating rotarod as a motor learning task. Results: We show that the deficiency in motor skill learning in PITx3(/) is dramatic and can be rescued with levodopa treatment. In addition, cessation of levodopa treatment after acquisition of the motor skill does not result in an immediate drop in performance. Instead, there is a gradual decline of performance that lasts for a few days, which is not related to levodopa pharmacokinetics. We show that this gradual decline is dependent on the retesting experience. Interpretation: This observation resembles the long-duration response to levodopa therapy in its slow buildup of improvement after the initiation of therapy and gradual degradation. We hypothesize that motor learning may play a significant, underappreciated role in the symptomatology of Parkinson disease as well as in the therapeutic effects of levodopa. We suggest that the important, yet enigmatic long-duration response to chronic levodopa treatment is a manifestation of rescued motor learning. ANN NEUROL 2010;67:639 – 647

This study was aimed to investigate the specific anti-leukemia cell effect of cytotoxic T lymphocytes (CTLs) induced by HL-60 or K562 cell-sensitized dendritic cells (DCs) from umbilical cord blood. 12 units of human umbilical cord blood (UCB) were collected and the mononuclear cells (MNCs) were isolated from UCB, then cultured with granulocyte monocyte colony- stimulating factor (GM-CSF), interleukin 3 (IL-3), recombinant human stem cell factor (SCF) and EPO for 3 - 4 weeks. Flow cytometry was used to determine the number of DCs and cell surface antigens before and after culture with monoclonal antibodies including CD83, CD1a, CD11c and CDw123. HL-60 and K562 were frozen-thawed, and released their tumor antigen peptides (TAP). The CTLs were produced by sensitizing T lymphocytes with DC-loaded HL-60 and K562 cell antigens. The test of (3)H-TdR incorporation was used to detect the immunostimulation activity of DCs. MTT assay was applied to evaluate specific cytotoxicity of CTL on leukaemia cells. The results indicated that the MNCs of UCBs cultured with GM-CSF, IL-3, EPO and SCF were shown to differentiate into CD1a(+) CD11c(+) CD83(+) CDw123(+) DCs. Numbers of DCs from UCBs remarkably increased in 2 - 4 weeks and then decreased. After culture with cytokines DCs increased (10.6 - 28.2) x 10(5)/ml in actual numbers. The CTL induced by DC pulsed with HL-60, K562 frozen-thawed lysates were effective to kill HL-60 and K562. Cytotoxicity of CTL to HL60 and K562 were (42.04 +/- 8.46)% and (31.25 +/- 11.07)% respectively. It is concluded that the MNCs of UCBs cultured with cytokines of GM-CSF, SCF, EPO and IL-3 can differentiate into CD1a(+), CD83(+), CD11c(+) and CDw123(+) DCs. The CTL induced by DCs pulsed with HL-60, K562 frozen-thawed lysates can effectively kill HL-60 and K562. These DCs as antigen presenting cells play an important role in cancer immunotherapy.
