Acute respiratory infections (ARIs) are a common public safety threat with high morbidity and mortality in pediatric patients worldwide. Qinbaohong Zhike oral liquid (QBH), a marketed traditional Chinese medicine product, has been widely used to cure respiratory diseases. QBH is reported to have antitussive, expectorant, and antiasthmatic properties. However, its treatment effect against ARIs is not elucidated. This study aimed to explore the therapeutic efficacy of QBH in the treatment of ARIs-induced pneumonia. Network pharmacology was used to predict the possible targets of QBH against ARIs. Next, the tracheal lipopolysaccharide (LPS-)-induced acute lung injury (ALI) immature rat model was constructed to evaluate the therapeutic effect of QBH. Tandem mass tag (TMT-)-based quantitative proteomics was then used to screen the in-depth disease targets of QBH. QBH exerted a protective effect against LPS-induced ALI by inhibiting pulmonary pathological damage. QBH also reduced the levels of interleukin (IL)-6, tumor necrosis factor (TNF)-α, interferon (IFN)-γ, and granulocyte macrophage colony-stimulating factor (GM-CSF) in the serum and IL-1β, IL-6, IL-8, TNF-α, IFN-γ, and GM-CSF in the lung tissue. Based on proteomic data, olfactomedin 4 (OLFM4) related to immunity and inflammation was selected as a potential target. Western blot analysis further confirmed the moderating effect of QBH downregulation on OLFM4 in the lung tissue. Our findings demonstrated that QBH alleviated lung tissue damage and inflammatory reaction via inhibiting OLFM4 expression in LPS-challenged immature rats. Our research indicates that QBH may have therapeutic potential for treating ARIs-related ALI in pediatric patients, which also serves as a candidate target for drug therapy of ALI by intervening OLFM-related signaling pathways.

Magnetic Resonance Imaging (MRI) has been proven to be an efficient way to diagnose Alzheimer's disease (AD). Recent dramatic progress on deep learning greatly promotes the MRI analysis based on data-driven CNN methods using a large-scale longitudinal MRI dataset. However, most of the existing MRI datasets are fragmented due to unexpected quits of volunteers. To tackle this problem, we propose a novel Temporal Recurrent Generative Adversarial Network (TR-GAN) to complete missing sessions of MRI datasets. Unlike existing GAN-based methods, which either fail to generate future sessions or only generate fixed-length sessions, TR-GAN takes all past sessions to recurrently and smoothly generate future ones with variant length. Specifically, TR-GAN adopts recurrent connection to deal with variant input sequence length and flexibly generate future variant sessions. Besides, we also design a multiple scalelocation (MSL) module and a SWAP module to encourage the model to better focus on detailed information, which helps to generate high-quality MRI data. Compared with other popular GAN architectures, TR-GAN achieved the best performance in all evaluation metrics of two datasets. After expanding the Whole MRI dataset, the balanced accuracy of AD vs. cognitively normal (CN) vs. mild cognitive impairment (MCI) and stable MCI vs. progressive MCI classification can be increased by 3.61% and 4.00%, respectively.

Weak spectral signal detection technology is a new subject in the measurement engineering field. It obtains the characteristic information of the measured object based on the characteristics of light diffraction, interference, polarization and so on. Regarding light source entropy, the artificial light source of mercury argon lamp with a spectrum of 250 nm~918 nm is selected as the first research object. The grating monochromator with built-in grating scale lines (1200 g/mm, 600 g/mm) is selected as the light splitting device. Due to the weak spectral signal, the output signal is vulnerable to noise interference after a certain amount of photoelectric conversion. AD202 variable voltage coupling filtering technology is adopted. It means that in the signal isolation and amplification process, the AD202 variable voltage coupled filter amplifier chip is employed to design the current/voltage conversion circuit and the same direction proportional operation amplifier circuit. Meanwhile, a second-order low-pass filter circuit is designed based on the characteristics of the OP07 operational amplifier. For the data mining of multispectral image texture features, Contourlet transform is used, that is, the multispectral image texture features are fuzzy preprocessed to remove the adjacent range correlation knowledge. On this basis, the texture features of multispectral images are analyzed. In the experiment, Contourlet transform is adopted to transform multispectral images from spatial domain to frequency domain, which has the advantages of short extraction time, low cost, and high mining accuracy. A spectrum acquisition system based on photoelectric detection is designed. With chlorophyll as the research object, the sample solution to be tested has high photoelectric absorbance in the range of blue-violet light (400 nm~500 nm) and red-orange light (600 nm~700 nm).

[Image: see text] Andrographolide (andro) and its derivatives have been reported to have antitumor activity by arresting the cell cycle. However, the more precise mechanism has been controversial. Here, a proteome chip was used to screen drug targets in cells, and we discovered that andro can bind to PDCD2 (PD2), which has been shown to be associated with the cell cycle and mRNA nuclear export. Then, RNA-binding protein immunoprecipitation for PD2 was used to detect the quantity of cell cycle-related mRNAs, and the nuclear distribution difference analyses of these mRNAs in tumor cells after andro intervention, followed by systematic experiments, were performed to assess the downstream effects of this event in vivo and in vitro. Thus, the target spectrum of andro was revealed at the level of the human proteome chip for the first time, and this work demonstrated that andro, through targeting PD2, blocks the nuclear output of CDK mRNAs in the nucleus of tumor cells, further reduces the expression of cell CDK proteins, and finally causes tumor cell cycle arrest in phenotype and tumor tissue growth arrest in vivo.

As a conserved molecular chaperone, heat shock protein 90 (Hsp90) maintains the stability and homeostasis of oncoproteins and helps cancer cells survive. DNA-dependent protein kinase catalytic subunit (DNA-PKcs) plays a pivotal role in the non-homologous end joining pathway for DNA double-strand breaks (DSB) repair. Tumor cells contain higher levels of DNA-PKcs to survive by the hostile tumor microenvironment and various antitumor therapies. Here, we showed that increased levels of Hsp90α, Hsp90β, and DNA-PKcs correlated with a poor overall survival in hepatocellular carcinoma (HCC). We revealed that Hsp90 N-terminal domain and C-terminal domain have different effects on DNA-PKcs protein and mRNA levels. The stability of DNA-PKcs depended on Hsp90α N-terminal nucleotide binding domain. Transcription factor SP1 regulates the transcription of PRKDC (gene name of DNA-PKcs) and is a client protein of Hsp90. Inhibition of Hsp90 N-terminal by STA9090 decreased the location of Hsp90α in nucleus, Hsp90α-SP1 interaction, SP1 level, and the binding of Hsp90α/SP1 at the proximal promoter region of PRKDC. Because hyperthermia induces DSBs with increases level of DNA-PKcs, combined STA9090 treatment with hyperthermia effectively delayed the tumor growth and significantly decreased DNA-PKcs levels in xenografts model. Consistently, inhibition of Hsp90 increased the number of heat shock–induced γ-H2AX foci and delayed the repair of DSBs. Altogether, our results suggest that Hsp90 inhibitor STA9090 decreases DNA-PKcs protein stability and PRKDC mRNA level, which provide a theoretical basis for the promising combination therapy of hyperthermia and Hsp90 inhibitor in HCC.

Compartmentalized micelles (CMs) are promising tailor-made soft matters that mimic natural designed structures and functions. Despite the structure of complex CMs, manipulating CM structures accessibly and reversibly remains elusive. Here, we report the fabrication of CMs via a generally valid noncovalent postmodification process. Starting from precursor micelles (PMs) based on one diblock copolymer, aromatic modification leads to the compartmentalization of PMs into well-defined spherical CMs. Control over compartment number, size and distribution in CMs, and segment distribution in their linear hierarchical assemblies is attained by simply tuning the postmodification degree and solvent composition. We also demonstrate the reversible transformation between PM and CMs during several heating-cooling cycles, which endows the micelles with potential in reversible functional transitions

[Image: see text] Diosgenin, a component separated from Dioscorea plants, is an important starting material for steroid hormone drugs and semisynthetic steroids. In the work, two series of diosgenin derivatives were designed, synthesized, and evaluated for their cellular anticancer activities. Most of the target compounds exhibited good inhibitory activities against four cell lines, Aspc-1 (human colon adenocarcinoma cells), H358 (human nonsmall cell lung cancer cells), HCT116 (human colorectal adenocarcinoma cells), and SW620 (human metastatic pancreatic cancer cells). Among them, the representative compound 2.2f exhibited 7.9–341.7-fold antiproliferative activities against the above-mentioned four cell lines compared with the lead compound diosgenin.

Inspired by a lotus leaf, a 3D carbon fibre-cotton-based conical water evaporator with tuneable water supply is designed. By changing the number of branch water paths, a balance between the water supply flux and evaporation rate is achieved.

This paper proposes a saturated smooth adaptive controller for regulating a certain type of underactuated Euler–Lagrange systems (UELSs) with modeling uncertainties and control saturations based on a singular perturbation approach. Compared with relevent literature, the advantages of the proposed controller include: (1) it renders the UELS semiglobally asymptotically track the desired position without the violation of control input constraints; (2) high-order derivatives of positions are not required in its implementation. The Hoppensteadt’s Theorem is employed to show that the proposed saturated controller renders the UELS semiglobally asymptotically stable about the desired set point with the satisfaction of control input constraints. The control effectiveness is validated by simulations on a two-link compliant robot arm.

Natural active products (NAPs) are derived from chemical substances found in nature that have biological activity and medicinal potential. Screening and revealing the protein targets of NAPs is an indispensable link in the pharmacological and toxicological understanding of NAPs. Proteins are the main factors executing cell functions, and cells rely on the function of proteins to complete various activities in the life cycle. The important mechanism of action of drugs is to regulate cell biological activities by interacting with proteins and other macromolecules. At present, the classic way to screen protein targets is based on the molecular label tracing method, which has a long cycle and changes the molecular structure and pharmacological effects of NAPs. Due to the shortcomings of molecular labelling methods, in recent years, scientists have tried to develop a variety of label-free protein target identification methods for NAPs and have made a certain amount of progress. This article reviews the current protein target identification methods for NAPs with the aim of providing a reference for research on NAP protein targets.

In order to discover novel anti-pulmonary fibrosis agents, a series of new pirfennidone derivatives were designed and synthesized. All compounds have been investigated for their anti-pulmonary activity and confirmed by 13C and 1H nuclear magnetic resonance and high-resolution mass spectrometry. Preliminary studies on its biological activity showed that all target compounds showed different degrees of inhibition on pulmonary fibrosis, and most of the derivatives were significantly better than pirfenidone.

Chitosan hydrogel have presented great potential in biomedical applications due to its biocompatibility, biodegradability and characteristic similarity to native extracellular matrix. However, 3D printing of chitosan hydrogel often suffers from weak formability and poor mechanical property, limiting its further utilization. In this study, a novel chiotsan hydrogel is prepared from maleic chitosan (MCS) with high acrylate group substitution (i.e. 1.67) and thiol-terminated poly (ethylene glycol) (TPEG) via step-chain growth photopolymerization approach, which can overcome significantly the oxygen inhibition effect. Rheological property, microstructure, mechanical properties and in vitro degradation can be regulated by changing the thiol/acrylate molar ratio. There is strong intermolecular action between MCS and TPEG. Notably, photopolymerized MCS/TPEG hydrogel exhibited ~2-fold and ~ 10-fold increase in gelling rate and compressive strength, respectively, compared to pure chitosan hydrogel. Based on these results, 3D printing of chitosan hydrogel fabricated by simultaneous extrusion deposition and thiol-acrylate photopolymerization, demonstrates printing accuracy and improved scaffold stability. This 3D printing of chitosan hydrogel shows no cytotoxicity and can support adherence of L929 cells, suggesting its potential in biomedical applications such as tissue engineering and drug delivery.

Soil inorganic carbon (SIC), including mainly carbonate, is a key component of terrestrial soil C pool. Autotrophic microorganisms can assimilate carbonate as the main or unique C source, how microorganisms convert SIC to soil organic carbon (SOC) remains unclear. A systematic field survey (n = 94) was performed to evaluate the shift in soil C components (i.e., SIC, SOC, and microbial residues) along a natural salinity gradient (ranging from 0.5 ‰ to 19 ‰), and further to explore how microbial necromass as an indicator converting SIC into SOC in the Yellow River delta. We observed that SIC levels linearly decreased with increasing salinity, ranging from ∼12 g kg−1 (salinity 6 ‰). Additionally, the concentrations of SOC and microbial residues exponentially decreased from salinity 6 ‰, with the decline of 39% and 70%, respectively. Microbial residues and SOC was tightly related to the variations in SIC. The structural equation model showed the causality on explanation of SOC variations with SIC through microbial residues, which can contribute 89% of the variance in SOC storage combined with SIC. Taken together, these two statistical analyses can support that microbial residues can serve as an indicator of SIC transition to SOC. This study highlights the regulation of microbial residues in SIC cycling, enhancing the role of SIC playing in C biogeochemical cycles and enriching organic C reservoirs in coastal saline soils.

Solar driven interfacial vapor generation is considered to be an effective strategy to alleviate the impact of water crisis on human activities. However, great efforts of researchers have been devoted to improving the solar steam generation efficiency, while less attention has been paid to the long-term stability of evaporators. Herein, we proposed a robust core-shell structured evaporator prepared by a simple fast curing assisted spray-coating method. Enhancing the mechanical properties and cytocompatibility of decellularized heart valves is the key to promote the application of biological heart valves. In order to further improve the mechanical properties, the electrospinning and non-woven processing methods are combined to prepare the polylactic acid (PLA)/decellularized heart valve nanofiber-reinforced sandwich structure electrospun scaffold. The effect of electrospinning time on the performance of decellularized heart valve is investigated from the aspects of morphology, mechanical properties, softness, and biocompatibility of decellularized heart valve. Results of the mechanical tests show that compared with the pure decellularized heart valve, the mechanical properties of the composite heart valve were significantly improved with the tensile strength increasing by 108% and tensile strain increased by 571% when the electrospinning time exceeded 1 h. In addition, with this electrospinning time, the composite heart valve has a certain promoting effect on the human umbilical vein endothelial cells proliferation behavior. This work provides a promising foundation for tissue heart valve reendothelialization to lay the groundwork for organoid.

Root-associated fungal microbiota, which inhabit the rhizosphere, rhizoplane and endosphere, have a profound impact on plant growth and development. Sorghum bicolor (L.) Moench, also called broomcorn or sweet sorghum, is a multipurpose crop. The comparison between annual and perennial sweet sorghum cultivars in terms of plant growth, as well as their interactions with belowground fungal microbiota, is still poorly understood, although there has been growing interest in the mutualism between annual sweet sorghum and soil bacteria or bacterial endophytes. In this study, the perennial sweet sorghum cultivar N778 (N778 simply) and its control lines TP213 and TP60 were designed to grow under natural field conditions. Bulk soil, rhizosphere soil and sorghum roots were collected at the blooming and maturity stages, and then the fungal microbiota of those samples were characterized by high-throughput sequencing of the fungal ITS1 amplicon. Our results revealed that the alpha diversity of the fungal microbiota in rhizosphere soil and root samples was significantly different between N778 and the two control lines TP213 and TP60 at the blooming or maturity stage. Moreover, beta diversity in rhizosphere soil of N778 was distinct from those of TP213 and TP60, while beta diversity in root samples of N778 was distinct from those of TP213 but not TP60 by PCoA based on Bray–Curtis and WUF distance metrics. Furthermore, linear discriminant analysis (LDA) and multiple group comparisons revealed that OTU4372, a completely unclassified taxon but with symbiotroph mode, was enriched in sorghum roots, especially in N778 aerial roots at the blooming stage. Our results indicate that Cladosporium and Alternaria, two fungal genera in the rhizosphere soil, may also be dominant indicators of sorghum yield and protein content in addition to Fusarium at the maturity stage and imply that the perennial sweet sorghum N778 can primarily recruit dominant psychrotolerant bacterial taxa but not dominant cold-tolerant fungal taxa into its rhizosphere to support its survival below the freezing point.

Cellulose has been widely used in filtration owing to its abundance, low density, and high specific surface area. However, the use of cellulose-based scaffolds for filtration under high temperatures or with sparks is difficult to realize because cellulose is highly flammable. To develop an advanced cellulose-based filtration material with excellent flame-retardant and filtration performance, in this study, an ice template is employed to prepare a cellulose/expandable graphite (EG) aerogel with a direction porous structure for filtration. The effect of the EG concentration in the aerogel on the aerogel’s physical properties, as well as its flame-retardance and filtration performance is investigated. Experimental data indicates that a compound aerogel with a 30 wt% EG concentration is extinguished immediately after leaving the fire source and has a self-extinguishing time as short as 0.155 s. The formation of a direction porous structure in cellulose/EG aerogels provides a pathway for air movement during the filtration process. The use of EG could improve the filtration performance of cellulose aerogels by increasing the specific surface area. When the EG concentration in the aerogel is 30 wt%, the blocking efficiency of particle sizes less than 0.3 and 0.5 µm reaches 98 % and 99 %, respectively, and the blocking efficiency of particle sizes less than 1.0 µm is 96 %. As-prepared cellulose/EG aerogels have good mechanical properties and thermal stability, which implies that they can be used for filtration under high temperatures and sparks. Moreover, the filtration performance of cellulose/EG aerogels prepared using ice templates with direction porous structure and excellent flame-retardance can potentially be used for filtration-based applications under high temperature and electrical spark conditions. The use of EG and direction porous structures in cellulose aerogels provides a novel idea for the functionalization of cellulose scaffolds.
