Abstract: The SARS-CoV-2 Omicron BA.1 variant emerged in 20211 and has multiple mutations in its spike protein2. Here we show that the spike protein of Omicron has a higher affinity for ACE2 compared with Delta, and a marked change in its antigenicity increases Omicron’s evasion of therapeutic monoclonal and vaccine-elicited polyclonal neutralizing antibodies after two doses. mRNA vaccination as a third vaccine dose rescues and broadens neutralization. Importantly, the antiviral drugs remdesivir and molnupiravir retain efficacy against Omicron BA.1. Replication was similar for Omicron and Delta virus isolates in human nasal epithelial cultures. However, in lung cells and gut cells, Omicron demonstrated lower replication. Omicron spike protein was less efficiently cleaved compared with Delta. The differences in replication were mapped to the entry efficiency of the virus on the basis of spike-pseudotyped virus assays. The defect in entry of Omicron pseudotyped virus to specific cell types effectively correlated with higher cellular RNA expression of TMPRSS2, and deletion of TMPRSS2 affected Delta entry to a greater extent than Omicron. Furthermore, drug inhibitors targeting specific entry pathways3 demonstrated that the Omicron spike inefficiently uses the cellular protease TMPRSS2, which promotes cell entry through plasma membrane fusion, with greater dependency on cell entry through the endocytic pathway. Consistent with suboptimal S1/S2 cleavage and inability to use TMPRSS2, syncytium formation by the Omicron spike was substantially impaired compared with the Delta spike. The less efficient spike cleavage of Omicron at S1/S2 is associated with a shift in cellular tropism away from TMPRSS2-expressing cells, with implications for altered pathogenesis.

Abstract: Although two-dose mRNA vaccination provides excellent protection against SARS-CoV-2, there is little information about vaccine efficacy against variants of concern (VOC) in individuals above eighty years of age1. Here we analysed immune responses following vaccination with the BNT162b2 mRNA vaccine2 in elderly participants and younger healthcare workers. Serum neutralization and levels of binding IgG or IgA after the first vaccine dose were lower in older individuals, with a marked drop in participants over eighty years old. Sera from participants above eighty showed lower neutralization potency against the B.1.1.7 (Alpha), B.1.351 (Beta) and P.1. (Gamma) VOC than against the wild-type virus and were more likely to lack any neutralization against VOC following the first dose. However, following the second dose, neutralization against VOC was detectable regardless of age. The frequency of SARS-CoV-2 spike-specific memory B cells was higher in elderly responders (whose serum showed neutralization activity) than in non-responders after the first dose. Elderly participants showed a clear reduction in somatic hypermutation of class-switched cells. The production of interferon-γ and interleukin-2 by SARS-CoV-2 spike-specific T cells was lower in older participants, and both cytokines were secreted primarily by CD4 T cells. We conclude that the elderly are a high-risk population and that specific measures to boost vaccine responses in this population are warranted, particularly where variants of concern are circulating., Nature, 596 (7872), ISSN:0028-0836, ISSN:1476-4687

Abstract: The kinetics of the immune changes in COVID-19 across severity groups have not been rigorously assessed. Using immunophenotyping, RNA sequencing, and serum cytokine analysis, we analyzed serial samples from 207 SARS-CoV2-infected individuals with a range of disease severities over 12 weeks from symptom onset. An early robust bystander CD8+ T cell immune response, without systemic inflammation, characterized asymptomatic or mild disease. Hospitalized individuals had delayed bystander responses and systemic inflammation that was already evident near symptom onset, indicating that immunopathology may be inevitable in some individuals. Viral load did not correlate with this early pathological response but did correlate with subsequent disease severity. Immune recovery is complex, with profound persistent cellular abnormalities in severe disease correlating with altered inflammatory responses, with signatures associated with increased oxidative phosphorylation replacing those driven by cytokines tumor necrosis factor (TNF) and interleukin (IL)-6. Abstract: Transmission of SARS-CoV-2 is uncontrolled in many parts of the world; control is compounded in some areas by the higher transmission potential of the B.1.1.7 variant1, which has now been reported in 94 countries. It is unclear whether the response of the virus to vaccines against SARS-CoV-2 on the basis of the prototypic strain will be affected by the mutations found in B.1.1.7. Here we assess the immune responses of individuals after vaccination with the mRNA-based vaccine BNT162b22. We measured neutralizing antibody responses after the first and second immunizations using pseudoviruses that expressed the wild-type spike protein or a mutated spike protein that contained the eight amino acid changes found in the B.1.1.7 variant. The sera from individuals who received the vaccine exhibited a broad range of neutralizing titres against the wild-type pseudoviruses that were modestly reduced against the B.1.1.7 variant. This reduction was also evident in sera from some patients who had recovered from COVID-19. Decreased neutralization of the B.1.1.7 variant was also observed for monoclonal antibodies that target the N-terminal domain (9 out of 10) and the receptor-binding motif (5 out of 31), but not for monoclonal antibodies that recognize the receptor-binding domain that bind outside the receptor-binding motif. Introduction of the mutation that encodes the E484K substitution in the B.1.1.7 background to reflect a newly emerged variant of concern (VOC 202102/02) led to a more-substantial loss of neutralizing activity by vaccine-elicited antibodies and monoclonal antibodies (19 out of 31) compared with the loss of neutralizing activity conferred by the mutations in B.1.1.7 alone. The emergence of the E484K substitution in a B.1.1.7 background represents a threat to the efficacy of the BNT162b2 vaccine.

Abstract: The flat spectrum radio quasar (FSRQ) PKS 1510-089 is known for its complex multiwavelength behavior, and is one of only a few FSRQs detected at very high energy (VHE, $E>100\,$GeV) $\gamma$-rays. VHE $\gamma$-ray observations with H.E.S.S. and MAGIC during late May and early June 2016 resulted in the detection of an unprecedented flare, which reveals for the first time VHE $\gamma$-ray intranight variability in this source. While a common variability timescale of $1.5\,$hr is found, there is a significant deviation near the end of the flare with a timescale of $\sim 20\,$min marking the cessation of the event. The peak flux is nearly two orders of magnitude above the low-level emission. For the first time, curvature is detected in the VHE $\gamma$-ray spectrum of PKS 1510-089, which is fully explained through absorption by the extragalactic background light. Optical R-band observations with ATOM reveal a counterpart of the $\gamma$-ray flare, even though the detailed flux evolution differs from the VHE ightcurve. Interestingly, a steep flux decrease is observed at the same time as the cessation of the VHE flare. In the high energy (HE, $E>100\,$MeV) $\gamma$-ray band only a moderate flux increase is observed with Fermi-LAT, while the HE $\gamma$-ray spectrum significantly hardens up to a photon index of 1.6. A search for broad-line region (BLR) absorption features in the $\gamma$-ray spectrum indicates that the emission region is located outside of the BLR. Radio VLBI observations reveal a fast moving knot interacting with a standing jet feature around the time of the flare. As the standing feature is located $\sim 50\,$pc from the black hole, the emission region of the flare may have been located at a significant distance from the black hole. If this correlation is indeed true, VHE $\gamma$ rays have been produced far down the jet where turbulent plasma crosses a standing shock., Comment: 25 pages, 17 figures. Abstract: The constantly improving sensitivity of ground-based and space-borne observatories has made possible the detection of high-energy emission (X-rays and gamma-rays) from several thousands of extragalactic sources. Enormous progress has been made in measuring the continuum flux enabling us to perform imaging, spectral and timing studies. An important remaining challenge for high-energy astronomy is measuring polarization. The capability to measure polarization is being realized currently at X-ray energies (e.g. with IXPE), and sensitive gamma-ray telescopes capable of measuring polarization, such as AMEGO, AdEPT, e-ASTROGAM, etc., are being developed. These future gamma-ray telescopes will probe the radiation mechanisms and magnetic fields of relativistic jets from active galactic nuclei at spatial scales much smaller than the angular resolution achieved with continuum observations of the instrument. In this white paper, we discuss the scientific potentials of high-energy polarimetry, especially gamma-ray polarimetry, including the theoretical implications, and observational technology advances being made. Weed and GIS survey of 60 cereal and pulse fields in Morocco are combined with crop sampling for stable isotope analysis to frame assessment of agricultural labour intensity in terms of manuring, irrigation, tillage and hand-weeding. Under low management intensity weed variation primarily reflects geographical differences, whereas under high management intensity fields in disparate regions have similar weed flora. Manured and irrigated oasis barley fields are clearly discriminated from less intensively manured rain-fed barley terraces in southern Morocco; when fields in northern and southern Morocco are considered together, climatic differences are superimposed on the agronomic intensity gradient. Barley δ13C and δ15N values clearly distinguish among the Moroccan regimes. An integrated approach combines crop isotope values with weed ecological discrimination of low- and high-intensity regimes across multiple studies (in southern Morocco and southern Europe). Analysis of archaeobotanical samples from EBA Tell Brak, Syria suggests that this early city was sustained through extensive (low-intensity, large-scale) cereal farming.

Abstract: The high radio frequency polarization imaging of non-thermal emission from active galactic nuclei (AGN) is a direct way to probe the magnetic field strength and structure in the immediate vicinity of supermassive black holes (SMBHs) and is crucial in testing the jet-launching scenario. To explore the the magnetic field configuration at the base of jets in blazars, we took advantage of the full polarization capabilities of the Global Millimeter VLBI Array (GMVA). With an angular resolution of $\sim$50 micro-arcseconds ($\mu$as) at 86 GHz, one could resolve scales up to $\sim$450 gravitational radii (for a 10$^9$ solar mass black hole at a redshift of 0.1). We present here the preliminary results of our study on the blazar BL~Lac. Our results suggest that on sub-mas scales the core and the central jet of BL Lac are significantly polarized with two distinct regions of polarized intensity. Abstract: Blazars are among the most variable objects in the universe. They feature energetic jets of plasma that launch from the cores of these active galactic nuclei (AGN), triggering activity from radio up to gamma-ray energies. Spatial localization of the region of their MeV/GeV emission is a key question in understanding the blazar phenomenon. The flat spectrum radio quasar (FSRQ) PKS 1502+106 has exhibited extreme and correlated, radio and high-energy activity that triggered intense monitoring by the Fermi-GST AGN Multi-frequency Monitoring Alliance (F-GAMMA) program and the Global Millimeter VLBI Array (GMVA) down to $��$3 mm (or 86 GHz), enabling the sharpest view to date towards this extreme object. Here, we report on preliminary results of our study of the gamma-ray loud blazar PKS 1502+106, combining VLBI and single dish data. We deduce the critical aspect angle towards the source to be $��_{\rm c} = 2.6^{\circ}$, calculate the apparent and intrinsic opening angles and constrain the distance of the 86 GHz core from the base of the conical jet, directly from mm-VLBI but also through a single dish relative timing analysis. Finally, we conclude that gamma rays from PKS 1502+106 originate from a region between ~1-16 pc away from the base of the hypothesized conical jet, well beyond the bulk of broad-line region (BLR) material of the source., 4 pages, 1 figure, proceedings of the 12th European VLBI Network Symposium and Users Meeting - EVN 2014, 7-10 October 2014, Cagliari, Italy. Abstract: Very long baseline interferometry at millimetre/submillimetre wavelengths (mmVLBI) offers the highest achievable spatial resolution at any wavelength in astronomy. The anticipated inclusion of ALMA as a phased array into a global VLBI network will bring unprecedented sensitivity and a transformational leap in capabilities for mmVLBI. Building on years of pioneering efforts in the US and Europe the ongoing ALMA Phasing Project (APP), a US-led international collaboration with MPIfR-led European contributions, is expected to deliver a beamformer and VLBI capability to ALMA by the end of 2014 (APP: Fish et al. 2013, arXiv:1309.3519). This report focuses on the future use of mmVLBI by the international users community from a European viewpoint. Firstly, it highlights the intense science interest in Europe in future mmVLBI observations as compiled from the responses to a general call to the European community for future research projects. A wide range of research is presented that includes, amongst others: - Imaging the event horizon of the black hole at the centre of the Galaxy - Testing the theory of General Relativity an/or searching for alternative theories - Studying the origin of AGN jets and jet formation - Cosmological evolution of galaxies and BHs, AGN feedback - Masers in the Milky Way (in stars and star-forming regions) - Extragalactic emission lines and astro-chemistry - Redshifted absorption lines in distant galaxies and study of the ISM and circumnuclear gas - Pulsars, neutron stars, X-ray binaries - Testing cosmology - Testing fundamental physical constants, Replaced figures 2 and 3: corrected position SRT. Abstract: When manufacturing free-form concrete parts, casting approaches are most commonly adopted. While casting allows good surface finish, geometric repeatability and easy replication of parts, it is inflexible and costly to setup. Expensive mould tools, with limited life, must be created before manufacturing can begin. These mould tools must then be safely stored to ensure future identical parts can be manufactured. If damaged, recreating exact replica mould tooling is an expensive and time-consuming process. The cost and difficulty of manufacturing new tooling therefore prohibits the economic manufacture of many bespoke parts, or results in significant lead time increases and project delays. An alternative approach, that is gaining increasing interest, is the use of 3D printing for concrete. A robot is used to guide a concrete deposition nozzle in a layer by layer deposition path, similar to polymer fused deposition modelling. However, in Concrete 3D Printing the combination of complex material rheological properties, and the need for high volumetric deposition rates, means achieving net shape precision parts from the deposition step is extremely challenging. To address this issue, a hybrid robotic 3D printing and milling system has been developed, with integrated in-process structured-light metrology, to enable the production of precision concrete parts with well controlled geometric features. The integrated robot manufacturing cell is described. Examples structures that demonstrate the ability to create and replicate complex concrete parts with improved surface properties are illustrated. Components with surface features that enable concrete assemblies are studied to illustrate the capability of the hybrid process to manufacture challenging parts with demanding geometric requirements. These structures are used to quantify the performance of the current system and illustrate the future potential for the approach.

Abstract: We present the current expectations for the design and physics program of an e+e- linear collider of center of mass energy 500 GeV -- 1 TeV. We review the experiments that would be carried out at this facility and demonstrate its key role in exploring physics beyond the Standard Model over the full range of theoretical possibilities. We then show the feasibility of constructing this machine, by reviewing the current status of linear collider technology and by presenting a precis of our `zeroth-order' design.
