We analyzed the chemical compositions of dust and sea‐salt particles in the EPICA Dome C (EDC) ice core during 26–7 kyr BP using an ice‐sublimation technique and compared the results with existing data of the Dome Fuji (DF) ice core. Combined with ion concentration data, our data suggested similar sea‐salt fluxes in both cores and significantly lower dust flux in the EDC core. The differences in modal size and aspect ratio of dust particles between the two cores support the dominance of Patagonian source suggested by earlier works. The compositions of calcic dust showed major change at ~17 kyr BP, possibly reflecting a relative increase in dust transported via the upper troposphere. The calcium sulfate fraction was higher in the DF core than in the EDC core after ~17 kyr BP, suggesting that higher Patagonian dust contribution to the DF region. Abundant NaCl particles were found in the DF core in comparison with the EDC core from the LGM to early Holocene, possibly because of the high concentration of terrestrial dust in the DF core that reduced acid availability for sea‐salt modification. During the Holocene, the lower NaCl fraction and Cl−/Na+ ratio in the EDC core suggested that most Cl− was lost to the atmosphere from snow at Dome C, while it was preserved at Dome Fuji as NaCl and solid solution.

Objectives To understand the perspectives of patients with rheumatoid arthritis (RA) about the risk of their relatives developing RA in the future, and about communicating with their relatives about risk and its modulation. Methods Twenty-one RA patients took part in semi-structured interviews. Results Participants reported willingness to communicate with relatives about their risk of developing RA, but described choosing which relatives to communicate with, on the basis of their perceived receptivity to such risk information. Participants described the potential for risk information to cause negative emotions. Some participants did not consider RA to be hereditable, and few reported smoking as a risk factor. Patients described a lack of public awareness about the causes of RA and the negative impact that RA has on quality of life. Awareness of this negative impact was identified as an important driver for predictive and preventive strategies. Participants held positive perceptions of predictive testing for RA, though the results of predictive tests were conceptualised as having a high degree of accuracy. Negative views of predictive testing were associated with an appreciation of the probabilistic nature of risk information. Participants felt that their relatives would prefer lifestyle modification over medication as a risk reduction strategy. Conclusions Information about risk factors for RA, and the potential impact of RA on quality of life, is needed to support family communication about RA risk. Management of expectations is needed in relation to the probabilistic nature of risk information, and appropriate support should be provided for negative psychological outcomes.

Events with high energy isolated electrons, muons or tau leptons and missing transverse momentum are studied using the full e^\pm p data sample collected by the H1 experiment at HERA, corresponding to an integrated luminosity of 474 pb^{-1}. Within the Standard Model, events with isolated leptons and missing transverse momentum mainly originate from the production of single W bosons. The total single W boson production cross section is measured as 1.14 \pm 0.25 (stat.) \pm 0.14 (sys.) pb, in agreement with the Standard Model expectation. The data are also used to establish limits on the WW\gamma gauge couplings and for a measurement of the W boson polarisation., Comment: 41 pages, 13 figures

Processes leading to a final state with at least two high transverse momentum leptons (electrons or muons) are studied using the full e ± p data sample collected by the H1 experiment at HERA. The data correspond to an integrated luminosity of 463 pb -1. Di-lepton and tri-lepton event classes are investigated. Cross sections of the production of e + e - and μ + μ - pairs are derived in a restricted phase space dominated by photon-photon collisions. In general, good agreement is found with Standard Model predictions. Events are observed with a total scalar sum of lepton transverse momenta above 100 GeV where the Standard Model expectation is low. In this region, combining di-lepton and tri-lepton classes, five events are observed in e + p collisions, compared to a Standard Model expectation of 0.96 ± 0.12, while no such event is observed in e - p data for 0.64 ± 0.09 expected. © 2008 Elsevier B.V. A search for a narrow baryonic resonance decaying to $K^0_s p$ or $K^0_s \bar p$ is carried out in deep inelastic ep scattering with the H1 detector at HERA. Such a resonance could be a strange pentaquark \thplns, evidence for which has been reported by several experiments. The $K^0_s p$ and $K^0_s \bar p$ invariant mass distributions presented here do not show any significant peak in the mass range from threshold up to 1.7 GeV. Mass dependent upper limits on $\sigma(ep \to e \thplf X)\times BR(\thplf \to K^0 p)$ are obtained at the 95% confidence level., Comment: 13 pages, 4 figures

Differential dijet cross sections are measured in photoproduction in the region of photon virtualities Q^2 < 1 GeV^2 with the H1 detector at the HERA ep collider using an integrated luminosity of 66.6 pb^{-1}. Jets are defined with the inclusive k_T algorithm and a minimum transverse momentum of the leading jet of 25 GeV is required. Dijet cross sections are measured in direct and resolved photon enhanced regions separately. Longitudinal proton momentum fractions up to 0.7 are reached. The data compare well with predictions from Monte Carlo event generators based on leading order QCD and parton showers and with next-to-leading order QCD calculations corrected for hadronisation effects., 18 pages, 5 figures, 5 tables

A search for the single production of doubly-charged Higgs bosons H ± ± in ep collisions is presented. The signal is searched for via the Higgs decays into a high mass pair of same charge leptons, one of them being an electron. The analysis uses up to 118 pb -1 of ep data collected by the H1 experiment at HERA. No evidence for doubly-charged Higgs production is observed and mass dependent upper limits are derived on the Yukawa couplings h e l of the Higgs boson to an electron-lepton pair. Assuming that the doubly-charged Higgs only decays into an electron and a muon via a coupling of electromagnetic strength h e μ = sqrt(4 π α em) ≃ 0.3, a lower limit of 141 GeV on the H ± ± mass is obtained at the 95% confidence level. For a doubly-charged Higgs decaying only into an electron and a tau and a coupling h e τ ≃ 0.3, masses below 112 GeV are ruled out. © 2006 Elsevier B.V. Deep-inelastic positron-proton interactions at low values of Bjorken-x down to x \approx 4.10^-5 which give rise to high transverse momentum pi^0 mesons are studied with the H1 experiment at HERA. The inclusive cross section for pi^0 mesons produced at small angles with respect to the proton remnant (the forward region) is presented as a function of the transverse momentum and energy of the pi^0 and of the four-momentum transfer Q^2 and Bjorken-x. Measurements are also presented of the transverse energy flow in events containing a forward pi^0 meson. In order to determine the aetiology of acute epiglottitis in adults, blood cultures, paired sera and a urine sample were obtained from 54 patients with fever and epiglottitis visualized by indirect laryngoscopy or by direct fibreoptic nasolaryngoscopy. Antibodies were determined against the capsular polysaccharide of Haemophilus influenzae type b (Hib), 3 pneumococcal antigens (a mixture of 23 capsular polysaccharides, C-polysaccharide and pneumolysin) and antistreptolysin O. Acute sera were examined by the polymerase chain reaction (PCR) for DNA of Hib and pneumococci. The urine samples were examined for Hib capsular antigen. Blood cultures were positive in 15 patients. In another 16, serology and/or PCR verified the aetiology. Hib was the cause in 14, pneumococci in 12 and group A streptococci in 5 patients. The aetiology remained unknown in 23/54 patients (43%). In conclusion, the addition of serology and PCR to blood cultures doubled the possibilities of verifying the aetiology of acute epiglottitis in adults.

Human NK cells (with CD3-/56+ phenotype) acquired features characteristic of apoptosis after incubation with autologous monocytes, as revealed by apoptotic nuclear morphology, degradation of DNA into oligonucleosomal fragments, and reduced nuclear interchalation of propidium iodide. In contrast, T cells (CD3+/56-) remained non-apoptotic. The monocyte-induced apoptosis in NK cells was prevented by catalase, a scavenger of hydrogen peroxide; whereas superoxide dismutase (a scavenger of superoxide anion), hydroxyl radical scavengers such as mannitol and deferoxamine, or the hypochlorus acid scavenger taurine did not prevent apoptosis. Sodium azide, a myeloperoxidase inhibitor, substantially reduced the monocyte-induced apoptosis in NK cells. Exogenous hydrogen peroxide, at concentrations exceeding 1 microns, induced apoptosis in both NK and T cells. Apoptosis induced by hydrogen peroxide occurred independently of synthesis of protein or mRNA and was blocked by the endonuclease inhibitor aurin tricarboxylic acid. Furthermore, oxidatively induced apoptosis in NK cells was inhibited by herbimycin A, indicating that apoptosis was dependent on protein kinases. Two to five times more hydrogen peroxide was required to induce apoptosis in T cells compared with NK cells. Similarly, NK cells were considerably more susceptible to apoptosis induced by the topoisomerase II inhibitor etoposide or by gamma-irradiation than were T cells. We conclude that monocyte-derived reactive oxygen metabolites kill NK cells by apoptosis and that NK cells are unusually sensitive to oxidatively as well as non-oxidatively induced apoptosis.

Differential dijet cross sections in diffractive deep-inelastic scattering are measured with the H1 detector at HERA using an integrated luminosity of 51.5 pb-1. The selected events are of the type ep --> eXY, where the system X contains at least two jets and is well separated in rapidity from the low mass proton dissociation system Y. The dijet data are compared with QCD predictions at next-to-leading order based on diffractive parton distribution functions previously extracted from measurements of inclusive diffractive deep-inelastic scattering. The prediction describes the dijet data well at low and intermediate zpom (the fraction of the momentum of the diffractive exchange carried by the parton entering the hard interaction) where the gluon density is well determined from the inclusive diffractive data, supporting QCD factorisation. A new set of diffractive parton distribution functions is obtained through a simultaneous fit to the diffractive inclusive and dijet cross sections. This allows for a precise determination of both the diffractive quark and gluon distributions in the range 0.05, Comment: 33 pages, 10 figures

Peripheral blood mononuclear cells (PBMC) from patients with aplastic anemia (AA) and healthy donors were compared with regard to their ability to produce soluble factors with inhibitory activity on in vitro granulopoiesis (GM-CFC). Although PBMC from AA patients produced enhanced levels of IFN-gamma as compared to controls, this lymphokine was found not to be the main inhibitor of in vitro granulopoiesis. Other, non-IFN related factors were potent inhibitors of both the mature and the immature precursors for GM-CFC, could act across the species barrier and were of low molecular weight. Also PBMC from healthy donors produced a non-IFN mediated GM-CFC inhibitory factor, but to a lesser degree and acting only on one type of myeloid precursors. The possible implications of these findings in relation to the etiology of AA will be discussed.

To examine the association between individual rheumatoid arthritis (RA) autoantibodies, sex and age at RA onset.Anti-CCP2, IgA-, IgG- and IgM-RF were analysed centrally in baseline sera from 1600 RA patients diagnosed within one year of RA symptom onset. Cut-offs for RF isotypes were determined at the 98th percentile based on RA-free controls, close to the 98.4% anti-CCP2 specificity.Anti-CCP2 was found in 1020 patients (64%), IgA RF in 692 (43%), IgG RF in 529 (33%) and IgM RF in 916 (57%) of the patients. When assessed one by one, anti-CCP2 and IgM RF were both associated with lower age at RA diagnosis. When assessed in one joint model, the association to IgM RF weakened and a strong association between IgA RF and higher age at RA diagnosis appeared. IgA RF and IgG RF associated with male sex, and IgM RF with female sex, with no difference for anti-CCP2. When the model was adjusted for sex, the association between IgM RF and age disappeared, whereas the strong associations between IgA RF and high age and between anti-CCP2 and low age at diagnosis remained. Further adjustments for smoking, shared epitope and inclusion year did not change the outcome. Univariate analyses stratified on anti-CCP2 and IgA RF status confirmed the findings.Anti-CCP associate with low, and IgA RF with high age at RA onset. RFs and anti-CCP2 display opposing association with sex. These results underscore that studies on RA phenotypes in relation to autoantibodies should accommodate age and sex.

A general method for solid-phase gene assembly on streptavidin-coated magnetic beads has been developed. The introduction of biotin in the 5'-end of the initiation oligonucleotide enables anchoring to the bead by means of the streptavidin-biotin interaction. The immobilization of one oligonucleotide enables controlled, stepwise annealing/ligation of successive 5'-phosphorylated oligonucleotides to rapidly build up predesigned gene constructs. In this report, we have assembled gene constructs of different lengths derived from the Plasmodium falciparum malaria blood-stage antigen Ag332. The encoded gene products were subsequently expressed in Escherichia coli using two parallel expression systems based on staphylococcal protein A and streptococcal protein G, respectively.

The Northern Hemisphere experienced dramatic changes during the last glacial, featuring vast ice sheets and abrupt climate events, while high northern latitudes during the last interglacial (Eemian) were warmer than today. Here we use high-resolution aerosol records from the Greenland NEEM ice core to reconstruct the environmental alterations in aerosol source regions accompanying these changes. Separating source and transport effects, we find strongly reduced terrestrial biogenic emissions during glacial times reflecting net loss of vegetated area in North America. Rapid climate changes during the glacial have little effect on terrestrial biogenic aerosol emissions. A strong increase in terrestrial dust emissions during the coldest intervals indicates higher aridity and dust storm activity in East Asian deserts. Glacial sea salt aerosol emissions in the North Atlantic region increase only moderately (50%), likely due to sea ice expansion. Lower aerosol concentrations in Eemian ice compared to the Holocene are mainly due to shortened atmospheric residence time, while emissions changed little.

A detailed history of Holocene volcanism was reconstructed using the sulfate record of the European Project for Ice Coring in Antarctica Dome C (EDC96) ice core. This first complete Holocene volcanic record from an Antarctic core provides a reliable database to compare with long records from Antarctic and Greenland ice cores. A threshold method based on statistical treatment of the lognormal sulfate flux distribution was used to differentiate volcanic sulfate spikes from sulfate background concentrations. Ninety-six eruptions were identified in the EDC96 ice core during the Holocene, with a mean of 7.9 events per millennium. The frequency distribution (events per millennium) showed that the last 2000 years were a period of enhanced volcanic activity. EDC96 volcanic signatures for the last millennium are in good agreement with those recorded in other Antarctic ice cores. For older periods, comparison is in some cases less reliable, mainly because of dating uncertainties. Sulfate depositional fluxes of individual volcanic events vary greatly among the different cores. A volcanic flux normalization (volcanic flux/Tambora flux ratio) was used to evaluate the relative intensity of the same event recorded at different sites in the last millennium. Normalized flux variability for the same event showed the highest value in the 1100–1500 AD period. This pattern could mirror changes in regional transport linked to climatic variations such as slight warming stages in the Southern Hemisphere (Southern Hemisphere Medieval Warming–like period?).

The strontium and neodymium isotopic signature of aeolian mineral particles archived in polar ice cores provides constraints on the geographic provenance of dust and paleo-atmospheric circulation patterns. Data from different ice cores drilled in the centre of the East Antarctic plateau such as EPICA-Dome C (EDC, 75°06′S; 123°21′E) and Vostok (78°28′S, 106°48′E) suggested a uniform geographic provenance for dust during Pleistocene glacial ages, likely from southern South America (SSA). In this work the existing dust isotopic data from EDC have been integrated with new data from Marine Isotopic Stage (MIS) 14 (about 536 ka before 1950AD) and in parallel some first results are shown for the new TALDICE ice core which was drilled on the edge of the East Antarctic Plateau (Talos Dome, 72°48′S, 159°06′E) on the opposite side with respect to SSA. Interestingly, the isotopic composition of TALDICE glacial dust is remarkably similar to that obtained from glacial dust from sites located in the East Antarctic interior. Overall, the glacial dust isotopic field obtained from six East Antarctic ice cores matches well South American data obtained from target areas. In this respect, it was recently suggested that dust exported long-range from South America originates from Patagonia and from the Puna-Altiplano plateau. To test this hypothesis, we analysed the isotopic composition of dust from an ice core drilled on the Illimani glacier (Bolivia, 16°37′S, 67°46′W; 6350 m a.s.l.) in order to obtain information on the isotopic composition of regional mineral aerosol uplifted from the Altiplano area and likely transported over a long distance. Altogether, ice core and source data strongly suggest that the westerly circulation pattern allowed efficient transfer of dust from South America to the East Antarctic plateau under cold Quaternary climates. Isotopic data support the hypothesis of a possible mixing of dust from Patagonia and from the Puna-Altiplano plateau. Interestingly, high glacial dust inputs to Antarctica are characterized by less radiogenic Nd values, an issue suggesting that enhanced dust production in Patagonia was associated with the activation of a secondary source. © The Author(s) 2020., Smoldering multiple myeloma (SMM) is an asymptomatic precursor state of multiple myeloma (MM). Recently, MM was redefined to include biomarkers predicting a high risk of progression from SMM, thus necessitating a redefinition of SMM and its risk stratification. We assembled a large cohort of SMM patients meeting the revised IMWG criteria to develop a new risk stratification system. We included 1996 patients, and using stepwise selection and multivariable analysis, we identified three independent factors predicting progression risk at 2 years: serum M-protein >2 g/dL (HR: 2.1), involved to uninvolved free light-chain ratio >20 (HR: 2.7), and marrow plasma cell infiltration >20% (HR: 2.4). This translates into 3 categories with increasing 2-year progression risk: 6% for low risk (38%; no risk factors, HR: 1); 18% for intermediate risk (33%; 1 factor; HR: 3.0), and 44% for high risk (29%; 2–3 factors). Addition of cytogenetic abnormalities (t(4;14), t(14;16), +1q, and/or del13q) allowed separation into 4 groups (low risk with 0, low intermediate risk with 1, intermediate risk with 2, and high risk with ≥3 risk factors) with 6, 23, 46, and 63% risk of progression in 2 years, respectively. The 2/20/20 risk stratification model can be easily implemented to identify high-risk SMM for clinical research and routine practice and will be widely applicable., The data collection study was conducted by the International Myeloma Foundation as an International Myeloma Working Group project supported in part by a grant funded by Janssen.

Given sex‐related differences in brain disorders, it is of interest to study if there is a sex difference in the permeability of the blood‐cerebrospinal fluid barrier (BCSFB) and the blood‐brain barrier (BBB). The CSF/serum albumin ratio (QA lb) is a standardized biomarker that evaluates the function of these barriers. In previous studies, contradictory results have been reported with respect to sex difference using this quotient, possibly because of small population sizes and heterogeneity with respect to ages. QA lb measurements in more than 20 000 patients between 1 and 90 years visiting our hospitals revealed a significant sex difference in all age groups also when excluding patients with pathologically high CSF albumin > 400 mg/L. Similar pattern was found in 335 healthy volunteers in similar age intervals. Although also other factors are likely important, our observation is consistent with lower integrity of the brain barriers in males. If the difference in QA lb is caused mainly by a difference in barrier function, this may require different drug doses and strategies for efficient central nervous system (CNS) delivery in males and females, as well as it may indicate differences in brain metabolism. Moreover, our study emphasizes that different reference values should be used both for different ages and sexes.
