In their book Media Ethics at Work: True Stories from Young Professionals, editors Lee Anne Peck and Guy S. Reel do not deviate from this approach. Instead, they refine it.Along with their twenty-two contributors, the two have created a lengthy body of case studies not from seasoned professionals working for major metropolitan newspapers or big-city television stations. Instead, they have created cases using information from students working for their student media outlets and professionals within a few years of graduation. As such, the authors contend, their cases are more like to resonate with students in college-level ethics classes.As the cases selected cover all the major topics in a typical ethics course, from plagiarism to knowing when a source is off-the-record to digital manipulation of photographs, the book is an excellent text for an ethics course or supplement to a reporting course or for training in a student media environment.The book gets off to a seemingly slow start, reviewing the philosophical theories of Socrates, Plato, Aristotle, Kant, Ross, and Rawl among others. Anticipating a quick jump into the cases, this may seem like an unnecessary delay. However, it proves to be a foundation for the book. Just as the plot of a successful novel recurs throughout the book, the references to the individuals who developed ethical theories recur through this book and becomes part of what makes this book useful and unique.Through twenty-three cases, the authors acknowledge such fundamental ideas as the following: "Few ethical dilemmas are clear cut." "Standards might include transparency, accuracy and fairness." "Making ethical choices is a skill that gets better with experience." "Even if you can legally publish such information, that doesn't make it ethical to do so." Acknowledging the tenets of various professional codes of ethics, they add such concepts as "[I]t's clearly important to demonstrate care in your professional work, thus avoiding unnecessary harm."The authors also give suggestions on where young professionals can turn in times of crisis, including professional organizations, mentors in the workplace, co-workers and former professors. "Both scholarly approaches and professional standards can help a practitioner reach a decision," say the authors in their discussion about a young public relations professional asked to impersonate a twelve-year-old online. Consistent references to the codes of ethics of major and minor media associations will prove a consistent resource to students. …

Prostate cancer is the most frequently diagnosed cancer in males in developed countries. To identify common prostate cancer susceptibility alleles, we genotyped 211,155 SNPs on a custom Illumina array (iCOGS) in blood DNA from 25,074 prostate cancer cases and 24,272 controls from the international PRACTICAL Consortium. Twenty-three new prostate cancer susceptibility loci were identified at genome-wide significance (P < 5 × 10−8). More than 70 prostate cancer susceptibility loci, explaining ~30% of the familial risk for this disease, have now been identified. On the basis of combined risks conferred by the new and previously known risk loci, the top 1% of the risk distribution has a 4.7-fold higher risk than the average of the population being profiled. These results will facilitate population risk stratification for clinical studies.

Abstract: The purpose of this study was to (1) examine the psychometric properties of the Behavioral Health Screen Depression Subscale (BHS-DS), including internal consistency, factor structure, convergent and discriminant validity, and (2) identify cut-off scores on the BHS-DS to distinguish patients with depression risk and their level of severity among an acute clinical sample of adolescents in residential psychiatric care. Participants included 861 youths ages 10–24 at residential treatment centers who completed a battery of measures, including the BHS-DS and Patient Health Questionnaire (PHQ-9). Factor analysis, bivariate correlation, and Cronbach’s α suggested the BHS-DS is a unidimensional scale with evidence of internal consistency reliability and convergent and discriminant validity. Receiver operating characteristic curve analysis identified four clinical cut-off scores (no, mild, moderate, and severe depression risk). Analysis of variance, Cohen’s kappa (κ), and Wilcoxon signed-rank test results suggested the four groups differ from each other and that the BHS-DS exhibits moderate agreement with the PHQ-9. The BHS-DS demonstrates good psychometric properties among a sample of residential youth, offering clinicians a good alternative to the PHQ-9 in screening for depression.

To determine the clinical features and outcomes of choroidal melanoma initially masquerading as central serous chorioretinopathy (CSCR).Retrospective case series.All patients with choroidal melanoma, initially misdiagnosed as CSCR elsewhere and evaluated by the Ocular Oncology Service at Wills Eye Hospital from 2004 to 2022, were included.A retrospective detailed review of patient charts and imaging was performed for all patients included in the study. Paired t tests and chi-squared tests were performed for data analysis.The primary outcome measures included clinical characteristics, ultrasonography, OCT, fundus autofluorescence, fluorescein angiography, and indocyanine green angiography. The secondary outcome measures included treatment results, such as the final visual acuity, tumor control, radiation-related complications, and melanoma-related metastasis and death.There were 22 patients (mean age, 48 years; 16 men) in this cohort. The mean interval between initial CSCR diagnosis and suspicion of choroidal melanoma was 50 months (median, 50 months; range, 0-242 months). At tumor diagnosis, the melanoma was submacular in 16 (73%) patients and extramacular in 6 (27%) patients. The mean tumor thickness was 3.4 mm (median, 2.5 mm; range, 1.4-10.7 mm), and the mean basal diameter was 9.2 mm (median, 8.0 mm, range, 4.5-22.0 mm). Features enabling differentiation of choroidal melanoma from CSCR (affected versus unaffected eye) included choroidal thickness asymmetry (100%300 μm versus 21%300 μm; P = 0.005), ipsilateral choroidal surface irregularity (100% versus 0%; P0.001), loss of choroidal vascular detail on OCT (100% versus 0%; P0.001), presence of multiple pinpoint leaks on angiography (100% versus 0%; P0.001), and contralateral lack of autofluoresence abnormalities (75% versus 6%; P = 0.001). Management of the choroidal melanoma included plaque radiotherapy (19, 86%), enucleation (2, 9%), or treatment elsewhere (1, 5%). On follow-up (mean, 6 years), vision loss of ≥ 3 Snellen lines (9 patients, 47%), metastasis (3 patients, 14%), and death (1 patient, 5%) were noted.Patients with presumed CSCR, especially if chronic, should be evaluated for a possible thin underlying choroidal melanoma with a dilated fundus examination and multimodal imaging.

Spatial data from high-resolution images abound in many scientific disciplines. For example, single-molecule localization microscopy, such as stochastic optical reconstruction microscopy, provides super-resolution images to help scientists investigate co-localization of proteins and hence their interactions inside cells, which are key events in living cells. However, there are few accurate methods for analyzing co-localization in super-resolution images. The current methods and software are prone to produce false-positive errors and are restricted to only 2-dimensional images. In this paper, we develop a novel statistical method to effectively address the problems of unbiased and robust quantification and comparison of protein co-localization for multiple 2- and 3-dimensional image datasets. This method significantly improves the analysis of protein co-localization using super-resolution image data, as shown by its excellent performance in simulation studies and an analysis of co-localization of protein light chain 3 and lysosomal-associated membrane protein 1 in cell autophagy. Moreover, this method is directly applicable to co-localization analyses in other disciplines, such as diagnostic imaging, epidemiology, environmental science, and ecology.

Skipped spawning, or variation in spawning periodicity, occurs in many annual spawning fish species and is an important consideration for population management. We assessed plasma sex steroid concentrations and measured gonad size and ovarian follicle diameter as metrics to nonlethally identify mass ovarian follicular atresia, which may contribute to skipped spawning in Burbot Lota lota. We maintained wild fish in captivity and exposed them to increasing water temperatures during a 3-wk period before the spawning season to induce mass ovarian follicular atresia. We collected ovarian follicles, blood plasma, and gonadal sonograms from fish weekly between January 28, 2018, and March 25, 2018. We histologically analyzed ovarian follicles to confirm stage of maturity. We measured concentrations of plasma sex steroids testosterone (T) and estradiol-17β (E2) by radioimmunoassay. We measured gonad diameter and circumference by ultrasonography and ovarian follicle diameter by image analysis. Mean plasma T concentration decreased from 8.94 ng/mL during late vitellogenesis to 1.83 ng/mL during atresia, suggesting that plasma T concentrations may be used to identify mass ovarian follicular atresia. We do not recommend using plasma E2 concentrations to identify mass ovarian follicular atresia because E2 concentrations rapidly decreased during the completion of vitellogenesis and the initiation of atresia in Burbot; therefore, plasma E2 may not accurately identify mass ovarian follicular atresia. Mean gonad diameter measured by ultrasonography decreased from 4.05 cm during late vitellogenesis to 3.65 cm during atresia. Mean diameter of ovarian follicles decreased during the final week of the study, suggesting that ovarian follicle diameter may be used to identify advanced mass ovarian follicular atresia. The nonlethal tools assessed—plasma sex steroid concentrations, ultrasonography, and ovarian follicle diameter—enable fisheries biologists to determine the occurrence and frequency of mass ovarian follicular atresia among Burbot in Lake Roosevelt and may be applied to other Burbot populations.

Abstract. Background: Suicide is a serious public health problem for LGBTQ+ adolescents and young adults. Attachment-based family therapy (ABFT) is one of the few suicide treatment models that has been tested with this population. The challenge remains in how to implement ABFT into the ecology of LGBTQ+ service settings. Aims: In this study, we tested the feasibility, acceptability, and preliminary effectiveness of ABFT in LGBTQ+ community settings. Method: Ten participants were enrolled in 16 weeks of ABFT across three LGBTQ+ organizations. Feasibility (treatment completion) and acceptability (Working Alliance Inventory [WAI]; opinions about treatment [OAT]) were measured for youth and caregivers. Depression and suicide outcomes were assessed for nine participants. Results: ABFT was demonstrated to be both feasible and acceptable. All participants completed treatment, and there were no dropouts. Adolescents/young adults and their caregivers reported high WAI and adequate OAT scores throughout treatment. There was a significant decrease in suicidal ideation (β = −12.16, t(10) = −3.14, p < .01). Decreases in depression, however, were not significant (β = −1.83, t(9.11) = −0.88, p = .40). Limitations: The small sample limits our ability to estimate treatment effectiveness. A larger effectiveness trial is warranted. Conclusion: ABFT is a promising treatment for LGBTQ+ service settings.

Two major treatment modalities for retinoblastoma, intraarterial chemotherapy (IAC) and intravitreal chemotherapy (IVitC), have superseded external beam radiotherapy for eye salvage. In this new setting our objectives were to evaluate the indications for plaque radiotherapy, complications, and recurrence rates.Retrospective detailed review of patient's charts was performed for all subjects treated with plaque radiotherapy for retinoblastoma between January 2015 and December 2020.A total of 12 eyes of 12 patients were included. Mean age at plaque insertion was 45 months (median 29, range 17-150). The treatment dose was 40 Gy to the tumor apex. The indication for plaque radiotherapy was salvage therapy in 11 eyes (92%) and primary treatment in one eye (8%). At last follow-up from plaque insertion (mean 36 months, range 3-67), four (33%) patients had visual acuity better than 0.5 LogMAR and four (33%) had visual acuity worse than 1.0 LogMAR. Radiation-related complications were: one (8%) vitreous haemorrhage, two (16%) non-proliferative radiation retinopathy and one (8%) cataract. Recurrence was detected in four (33%) patients at a mean of 7.8 months (median 5, range 1-20) post-plaque. Globe salvage rate was 75%, as three eyes required enucleation, one to treat recurrence of the tumor treated with plaque and two to treat recurrence of other tumors.In the current era of retinoblastoma management, a role for plaque radiotherapy remains for salvage or primary treatment in eyes with localised active tumor, providing tumor control in 66%. Close observation is recommended to both detect recurrence and radiation-related complications.
