Variety assortment enhancement is a normal task that involves enhancing the assortment's quality and upgrading varieties. The findings of a research of imported grape varieties in the southeast of Kazakhstan, namely in the Almaty region's bottom-mountain zone, are presented in this article. The onset and conclusion of the main phenological phases of a grape plant throughout the vegetative period were directly influenced by the climatic and meteorological conditions of this district. In comparison to the recognized variety Almaty early-maturing, it has been proven that types Priusadebny, Iyulsky, and Kuibyshevsk early-maturing have a high degree of eyes wintering buds and may provide a high-quality crop in this location. Resumo O aprimoramento da variedade é uma tarefa normal que envolve o aprimoramento da qualidade da variedade e o aprimoramento das variedades. As descobertas de uma pesquisa de variedades de uvas importadas no sudeste do Cazaquistão, ou seja, na zona de base da montanha da região de Almaty, são apresentadas neste artigo. O início e a conclusão das principais fases fenológicas de uma videira ao longo do período vegetativo foram diretamente influenciados pelas condições climáticas e meteorológicas deste distrito. Em comparação com a variedade reconhecida Almaty de maturação precoce, foi comprovado que os tipos Priusadebny, Iyulsky e Kuibyshevsk de maturação precoce têm um alto grau de gomos de inverno de olhos e podem fornecer uma safra de alta qualidade neste local.

We describe a gall midge Parampelomyia yukawai Elsayed and Tokuda gen. nov. sp. nov. belonging to the subtribe Schizomyiina (Diptera: Cecidomyiidae: Asphondyliini) based on an integrative taxonomic study. This species forms barely-swollen flower bud galls on the porcelain berry Ampelopsis brevipedunculata var. heterophylla (Vitaceae) in Japan. The new genus is distinguishable from all known schizomyiine genera by tarsomere I lacking a ventroapical extension, the bulbous base of the protrusible portion of the ovipositor, the fused and sclerotized female cerci, the bidentate gonostylus, and the larval terminal abdominal segment that bears two corniform, two asetose and six setose papillae. The new genus is compared with and separated from the similar genera Schizomyia and Ampelomyia morphologically and phylogenetically.

Resveratrol is a polyphenol with antioxidant properties and possible beneficial effects on human health. Grapes, peanuts, berries, cacao beans and red wine contain resveratrol. Resveratrol attracts attention due to its bioactive properties, however, the concentration of this compound is not high in grape and wine. Therefore, different studies have been carried out to increase resveratrol level in these products. Several factors such as the grapevine variety, the climatic conditions and the viticultural practices used to create stress on the vine affect the level of resveratrol. Winemaking technologies applied during pre-fermentation, fermentation and post–fermentation stages could also have an effect on the concentration of this stilbene. In addition, recent studies have evaluated biotechnological approaches through the use of different bacteria and yeast strains to produce wine with increased resveratrol content. In this review, the most important factors contributing to increase the resveratrol concentration in grapes and wines are examined. Besides, analytical methods to determine resveratrol content in wine are addressed.

Monitoring the quality attributes of grapes is a practice that allows the state of ripeness to be checked and the optimal harvest time to be identified. A non-destructive method based on hyperspectral imaging (HSI) technology was developed. Analyses were carried out directly in the field on a ‘Sangiovese’ (Vitis vinifera L.) vineyard destined for wine production, by using a Vis/NIR (400–1000 nm) hyperspectral camera. One vineyard row was analysed on 13 different days during the pre-harvest and harvest time. The soluble solids content (SSC) expressed in terms of °Brix was measured by a portable digital refractometer. Afterwards, the grape samples were split in two classes: the first one composed by the samples characterised by a °Brix lower than 20 (not-ripe), while the second one by the samples with a °Brix higher than 20 (ripe). Grape mean spectra were extracted from each hyperspectral image and used to predict the SSC by partial least squares regression (PLS), and to classify the samples into the two classes by PLS discriminant analysis (PLS-DA). SSC was predicted with a R2 = 0.77 (RMSECV = 0.79 °Brix), and the samples were correctly classified with a percentage from 86 to 91%. Even if the number of wavelengths was limited, the percentages of correctly classified samples were again within the above-mentioned range. The present study shows the potential of the use of HSI technology directly in the field by proximal measurements under natural light conditions for the prediction of the harvest time of the ‘Sangiovese’ red grape.

The aim of this study was to characterize the changes in berry anatomy during the development of grape (Vitis vinifera L., and Vitis vinifera × Vitis labrusca) with different shapes. Paraffin sectioning was used to examine the structural parameters of the cells. The results revealed that, with the development of berries, the transverse and longitudinal diameters of the flesh cells gradually increased, revealing certain regularity. However, the transverse and longitudinal diameters of the epidermal and sub-epidermal cells were different between varieties, reflecting the specificity of the varieties. The growth of the transverse and longitudinal diameters of the berries was found to be completed in the early stage of development. A combination of correlation analysis and size analysis for each cell layer revealed that, due to the small cross-sectional area of the epidermal and sub-epidermal cells, the influence of these cells on the transverse and the longitudinal diameters of the berries would be small. In conclusion, the longitudinal and transverse diameters of the grape berries were mainly determined by the longitudinal and transverse diameters of the flesh cells. The different shapes of the grape berries could mainly be attributed to the different growth rates of the flesh cells in the longitudinal and transverse directions. These different rates of growth led to different lengths and widths of the berry.

The grape is a very well-liked fruit that is valued for its distinct flavor and several health benefits, including antioxidants, anthocyanins, soluble sugars, minerals, phenolics, flavonoids, organic acids, and vitamins, which significantly improve the product’s overall quality. Today’s supply chain as a whole needs quick and easy methods for evaluating fruit quality. Thus, the objective of this study was to estimate the quality attributes of Flame Seedless grape berries cultivated under various agronomical management and other practices using color space coordinates (berry L*, berry a*, and berry b*) as inputs in an artificial neural network (ANN) model with the best topology of (3-20-11). Satisfactory predictions based on the R2 range, which was 0.9817 to 0.9983, were obtained for physical properties (i.e., berry weight, berry length, and berry diameter as well as berry adherence strength) and chemical properties (i.e., anthocyanin, total soluble solids (TSS), TSS/titratable acidity, total sugars, titratable acidity, reducing sugars, and non-reducing sugars). Meanwhile, we also performed a contribution analysis to analyze the relative importance of CIELab colorimeter parameters of berries L*, a*, and b* to determine the main fruit quality. In terms of relative contribution, berry b* contributed relatively largely to berry weight, berry adherence strength, TSS, TSS/titratable acidity, titratable acidity, total sugars, reducing sugars, and non-reducing sugars and a* contributed relatively largely to anthocyanin, berry length, and berry diameter. The developed ANN prediction model can aid growers in enhancing the quality of Flame Seedless grape berries by selecting suitable agronomical management and other practices to avoid potential quality issues that could affect consumers of them. This research demonstrated how color space coordinates and ANN model may well be utilized to evaluate the Flame seedless grape berries’ quality.

Background and Aims Although plastid genes are widely used in phylogenetic studies, signals of positive selection have been scarcely investigated in the grape family. The plastomes from 91 accessions of Vitaceae were examined to understand the extent to which positive selection is present and to identify which genes are involved. Moreover, the changes through time of genes under episodic positive selection were investigated and the hypothesis of an adaptive process following the Cretaceous–Palaeogene (K/Pg) transition about 66 million years ago was tested. Methods Different codon-substitution models were used to assess pervasive and episodic positive selection events on 70 candidate plastid genes. Divergence times between lineages were estimated and stochastic character mapping analysis was used to simulate variation over time of the genes found to be under episodic positive selection. Key Results A total of 20 plastid genes (29 %) showed positive selection. Among them, 14 genes showed pervasive signatures of positive selection and nine genes showed episodic signatures of positive selection. In particular, four of the nine genes (psbK, rpl20, rpoB, rps11) exhibited a similar pattern showing an increase in the rate of variation close to the K/Pg transition. Conclusion Multiple analyses have shown that the grape family has experienced ancient and recent positive selection events and that the targeted genes are involved in essential functions such as photosynthesis, self-replication and metabolism. Our results are consistent with the idea that the K/Pg transition has favoured an increased rate of change in some genes. Intense environmental perturbations have influenced the rapid diversification of certain lineages, and new mutations arising on some plastid genes may have been fixed by natural selection over the course of many generations.

(1) Background: Italia is a seeded grape variety widely cultivated in the Aegean Region in Turkey, whereas Bronx Seedless is a seedless grape variety, preferred by consumers due to its pink berries and interesting flavor. The goal was to study the volatile compounds of these table grapes throughout berry ripeness. (2) Methods: The volatile compounds were analyzed by GC-MS in six different phenological stages (3) Results: Bronx Seedless grapes presented a higher content of seven terpenes, three aldehydes, one fatty acid, three alcohols, one C6 compound, total aldehydes and total alcohols, and a lower content of eleven terpenes, one fatty acid, four esters, one alcohol, four C6 compounds and its total content than Italia table grapes. The concentration of most of the volatile compounds analyzed increased from “begin of berry touch” to “berries ripe for harvest” stages. Terpenes content in both varieties at harvest was lower than 1.0 mg L−1. β-ionone presented the highest odor activity value (OAV) in both varieties. Bronx Seedless grapes presented higher OAV for (Z)-3-hexenal and cedrol, and lower hexanal to (E)-2-hexenal ratio than Italia grapes. (4) Conclusions: Both varieties could be classified as neutral aromatical varieties and it is probable that to achieve a better aromatic quality, Bronx Seedless should be harvested later than Italia.

Anthocyanin plays an important role in wine color and quality. The anthocyanin concentration in grapes depends on the variety and is influenced by environmental factors, such as temperature, sunlight, soil, and grapevine practice. In this study, the anthocyanin profiles of two varieties, namely Cabernet Sauvignon and Merlot, from five regions (11 plots) in Xinjiang were examined. The climatic parameters performed noticeable difference between northern and southern of Xinjiang region. Result showed that northern of Xinjiang possessed the relatively lower humidity and higher average temperature in comparison with southern area. In addition, the total anthocyanin profiles of Cabernet sauvignon, which in Changji, Wushitala, Heshuo respectively, were higher than of other plots. This result might be caused by which those have relatively higher altitude than in other plots. For merlot, the higher anthocyanin concentrations performed in southern of Xinjiang, which could be contributed to the lower average temperature and higher relative humidity. Comparing Cabernet sauvignon and Merlot, the latter was easily affected by environment factors, and the anthocyanin composition and concentration of Merlot showed significant differences in different regions.

This study aimed to evaluate the wine composition from vine clones of Merlot (181, 346, 347, 348) and Cabernet Sauvignon (R5, 18A, 163, 338, 341). The grapes were harvested between 2010 and 2012, then on the same day, taken to the winery facility for winemaking. After crushing and destemming the grapes, the musts were transferred to glass containers, where the alcoholic and later malolactic fermentations occurred. Next, the wines were bottled and analyzed, where the classical variables were determined by physicochemical methods, volatile compounds by gas chromatography, and minerals by atomic absorption spectrophotometry. Data were submitted to ANOVA, Tukey test, correlation analysis, and Principal Component Analysis. The clone effect was significant for some variables; however, year and varietal group exerted greater effects. The Merlot group generally had higher values of ethanol, absorbance at 520 nm, color intensity, total polyphenols index, and methanol. On the other hand, the Cabernet Sauvignon group presented higher pH, ashes, alkalinity of ashes, hue, and potassium. Therefore, the three-year average data demonstrate that the evaluated Merlot and Cabernet Sauvignon clones are suitable to produce quality wines in the region, especially Merlot 181, 346, and 348 and Cabernet Sauvignon 18A and 163.

Canopy shading is a widely used viticultural strategy for mitigating early grape berry ripening caused by global warming. In this study, we covered half of the canopy with a black shade cloth from the pea-size stage to harvest. In the fruit zone, canopy shade treatment (ST) reduced daily average solar radiation by about 74.6 % compared to the untreated control (UC), and significantly reduced daily average temperature. ST leaves were found to have lower net assimilation rates and higher internal CO2 concentration than in UC, which resulted in reduced yield, bunch weight, cane starch and berry total soluble solids. A delayed development stage was found in ST berries which had lower pH and higher titratable acid than UC. ST increased both berry and wine anthocyanin concentration while significantly decreasing flavonol concentration. ST wines had higher concentrations of a number of ester compounds and β-damascenone than UC wines, thus significantly enhancing the floral and fruity aroma of ST wines. Higher C6/C9 and fatty acids concentrations in ST berries may have caused higher ester concentrations in ST wines. The results showed significant effects on metabolites in berries and wines caused by canopy shade.

Exogenous gibberellin (GA) was widely used to improve berry quality through inducing parthenocarpic seedless berries in grapes. We revealed that auxin response factors (ARFs), the key transcription factors in response to auxin, might respond to GA involving modulation of grape parthenocarpy. However, the underlying molecular mechanism in this process remains yet unclear. Here, a total of 19 VvARF members were identified in the ovaries during GA-induced grapes’ parthenocarpy. Interestingly, almost all members were GA-responsive factors, of which 9 could be classified in plant hormone signal transduction (KO04075) and involved in the tryptophan metabolic pathway (K14486). Moreover, VvARFs were predicted to have 310 interacted proteins involved in 19 KEGG pathways. Of them, 32 interacted proteins participated in the KO04075 pathway, including auxin (IAA), salicylic acid (SA), abscisic acid (ABA), cytokinin (CTK), and ethylene signaling pathways by responding to GA-mediated multi-hormone crosstalk. Further analysis demonstrated that VvARF4-2 might be the major factor in the modulation of GA-induced parthenocarpy via the crosstalk of IAA, CTK, SA, and ethylene signaling, followed by VvARF6-1 and VvARF9 involved in SA and ABA signaling pathways, respectively. Finally, we developed a VvARFs-mediated regulatory network by responding to GA-mediated multi-hormone crosstalk during grape parthenocarpy. Collectively, our findings provided novel insights into the regulatory network of VvARFs in GA-guided multi-hormone signaling to modulate grape parthenocarpy, which has great implications for the molecular breeding of high quality seedless grape berries.

Two lactic acid bacteria (LAB) strains of Lactobacillus plantarum 21802 and Lactobacillus brevis 6239 were selected and combined for fermentation of grape juice. The changes of sugars, organic acids, volatiles, phenolic compounds as well as antioxidant capacity of the grape juice were characterized. Results showed that fermentation with LAB led to a slight decrease in sugar content and increased in organic acid content. The sugars were mainly consumed at the initial stage of fermentation, while the malic acid was consumed completely during the fermentation. The contents of total phenolics and flavonoids in grape juice decreased gradually during the fermentation process. As compared with the initial contents in unfermented grape juice, the contents of proanthocyanin B2, rutin, isoquercitrin, rhamnose-3-O-glucoside in the final fermented grape juice were increased by 26.83%, 249.24%, 138.48%, 139.60%, respectively, while the contents of gallic acid, proanthocyanin B1, catechin and epicatechin were decreased by 24.49%, 14.85%, 71.68%, 42.52%, respectively. The mice fed with the fermented grape juice showed higher activity of superoxide dismutase and lower level of malondialdehyde both in serum and liver tissue than those fed with the unfermented grape juice. Fermentation with LAB also enhanced the flavor profile by increasing the total number and content of volatile compounds, especially those of monoterpene alcohols, which are the main and characteristic aroma volatile compounds in Muscat aroma grapes. This research confirmed that fermentation with LAB might be a desirable option for improving the flavor and nutritional attributes of grape juice by altering the ratio of sugar to organic acid, improving the phenolic compounds, enhancing the antioxidant capacity, intensifying of the characteristic aroma and pleasant flavor of the grape juice.

In harvesting operations, simulation verification of hand–eye coordination in a virtual canopy is critical for harvesting robot research. More realistic scenarios, vision-based driving motion, and cross-platform interaction information are needed to achieve such simulations, which are very challenging. Current simulations are more focused on path planning operations for consistency scenarios, which are far from satisfying the requirements. To this end, a new approach of visual servo multi-interaction simulation in real scenarios is proposed. In this study, a dual-arm grape harvesting robot in the laboratory is used as an example. To overcome these challenges, a multi-software federation is first proposed to establish their communication and cross-software sending of image information, coordinate information, and control commands. Then, the fruit recognition and positioning algorithm, forward and inverse kinematic model and simulation model are embedded in OpenCV and MATLAB, respectively, to drive the simulation run of the robot in V-REP, thus realizing the multi-interaction simulation of hand–eye coordination in virtual trellis vineyard. Finally, the simulation is verified, and the results show that the average running time of a string-picking simulation system is 6.5 s, and the success rate of accurate picking point grasping reached 83.3%. A complex closed loop of “scene-image recognition-grasping” is formed by data processing and transmission of various information. It can effectively realize the continuous hand–eye coordination multi-interaction simulation of the harvesting robot under the virtual environment.

In order to realize the goals of the EU Farm to Fork strategy, grape growers are introducing new grape-growing technologies. Among the new trends, “0-pesticide residue” protection is quite a promising one. Field trials were carried out in vineyards located in the Mediterranean part of Slovenia in 2021 and 2022 to test the “0-pesticide residue” (ZPR) grape protection system with the goal of producing wine without pesticide residues above the limit of 0.001 mg kg−1. The standard integrated grape protection program (IP) was compared to the ZPR program. The level of infection of leaves and grapes by fungal pathogens did not significantly increase due to the implementation of the ZPR spray program. The amount of yield and quality of yield were not decreased significantly, but a small financial loss of EUR 70–400 ha−1 appeared at ZPR grape production when compared to the IP production system. The ZPR system enabled a significant decrease in pesticide residue concentration in wine at a rate of 27 applied pesticide active substances in a rage from 20% to 99%. The goal of producing wine without pesticide residues above the limit concentration of 0.001 mg kg−1 was not completely achieved in these experiments, but we came very close to it with the tested spraying programs. Further finetuning of pesticide positioning and alternative plant protection products in 0-pesticide residue systems is needed.

The establishment and application of a two-crop-a-year cultivation system depends on the particularity of climatic conditions in subtropical regions. The different temperature, light, and water conditions throughout the growing season of summer and winter grapes are the fundamental reasons for differences in primary and secondary metabolites. We performed ultra-high-performance liquid chromatography–triple quadrupole mass spectrometry (UPLC-QQQ-MS)-based metabolomics on ‘Kyoho’ grapes under a two-crop-a-year cultivation system. In total, 1062 metabolites were identified and classified into 10 different categories, while flavonoids were the largest group, with 285 metabolites. Moreover, 876 metabolites were different among the four developmental stages, and 551 were different between the summer and winter grapes during the same growth period. Hierarchical clustering analysis (HCA) and principal component analysis (PCA) clearly distinguished developmental and growth-season differences based on the detected metabolites. Of note, flavonoids were the most important compounds responsible for the differences in berry composition during the growth and developmental seasons. The content of most flavonoids was higher in the winter grapes, but some were also found at higher levels in summer grapes, such as kaempferol-4′-O-glucoside, leucocyanidin, and cyanidin-3-O-glucosylglucoside. Additionally, myricetin-3-O-arabinoside was consistently higher in winter grapes than in summer grapes during all four developmental stages. The extreme high temperature and higher relative humidity were important reasons for the lower flavonoid content in the summer grapes than in the winter grapes. Moreover, the stronger light intensity in the early development of the winter grapes had a positive effect on the accumulation of flavonoids, especially flavonols and flavan-3-ols. This study provides new insights into the metabolism of flavonoids in grapes under a two-crop-a-year cultivation system and explores the climatic causes of the differences in the metabolites in the two crops of grapes.

A fermentation of Fiano di Avellino grape must was carried out at 9°C with the aim of selecting cryotolerant yeast strains and testing their fermentative performances and volatile production following molecular characterization. A total of 20 yeast cultures were isolated at different fermentation stages. Based on molecular identification and characterization, Metschnikowia (M.) pulcherrima, Hanseniaspora (H.) uvarum, Staremerella (St.) bacillaris, Saccharomyces (S.) cerevisiae, S. kudriavzevii, and S. paradoxus were found to be the yeast species dominating the fermentation. S. paradoxus has been rarely isolated in vineyards and never in the cellar environment. Moreover, in this study, S. kudriavzevii is detected for the first time in vine-wine environments. Both S. kudriavzevii and S. paradoxus co-occurred with S. cerevisiae when grapes were micro-fermented at low temperatures. The growth kinetics of the three species were greatly affected by the fermentation temperature. As a consequence, Fiano wines obtained with S. kudriavzevii and S. paradoxus significantly differed from those made by S. cerevisiae in terms of chemical and volatile composition.

Fruit zone leaf removal is a vineyard management practice used to manage bunch rots, fruit composition, and crop yield. We were interested in evaluating fruit zone leaf removal effects on bunch rot, fruit composition, and crop yield in ‘Chardonnay’ grown in the U.S. state of Georgia. The experiment consisted of seven treatments: no leaf removal (NO); prebloom removal of four or six leaves (PB-4, PB-6), post–fruit set removal of four or six leaves (PFS-4, PFS-6), and prebloom removal of two or three leaves followed by post–fruit set removal of two or three leaves (PB-2/PFS-2, PB-3/PFS-3). Although leaf removal reduced botrytis bunch rot and sour rot compared with NO, effects were inconsistent across the two seasons. Fruit zone leaf removal treatments reduced titratable acidity (TA) and increased soluble solids compared with NO. PB-6 consistently reduced berry number per cluster, cluster weight, and thus crop yield relative to PFS-4. Our results show that post–fruit set fruit zone leaf removal to zero leaf layers aids in rot management, reduces TA, increases soluble solids, and maintains crop yield compared with no leaf removal. We therefore recommend post–fruit set leaf removal to zero leaf layers over no leaf removal if crops characterized by relatively greater soluble solids-to-TA ratio and reduced bunch rot are desirable for winemaking goals.

In this work, both direct and indirect cell-based antioxidant profiles were established for 27 plant extracts and 1 algal extract. To evaluate the direct antioxidant effects, fluorescent AOP1 cell assay was utilized, which measures the ability of different samples to neutralize intracellular free radicals produced by a cell-based photo-induction process. As the intestinal barrier is the first cell line crossed by the product, dose response curves obtained from Caco-2 cells were used to establish EC50 values for 26 out of the 28 natural extracts. Among them, 11 extracts from Vitis, Hamamelis, Syzygium, Helichrysum, Ilex and Ribes genera showed remarkable EC50s in the range of 10 µg/mL. In addition to this, a synergistic effect was found when combinations of the most potent extracts (S. aromaticum, H. italicum, H. virginiana, V. vinifera) were utilized compared to extracts alone. Indirect antioxidant activities (i.e., the ability of cells to trigger antioxidant defenses) were studied using the ARE/Nrf2 luminescence reporter-gene assay in HepG2 cells, as liver is the first organ crossed by an edible ingredient once it enters in the bloodstream. Twelve extracts were subjected to an intestinal epithelial barrier passage in order to partially mimic intestinal absorption and show whether basolateral compartments could maintain direct or indirect antioxidant properties. Using postepithelial barrier samples and HepG2 cells as a target model, we demonstrate that indirect antioxidant activities are maintained for three extracts, S. aromaticum, H. virginiana and H. italicum. Our experimental work also confirms the synergistic effects of combinations of post-intestinal barrier compartments issued from apical treatment with these three extracts. By combining cell-based assays together with an intestinal absorption process, this study demonstrates the power of cell systems to address the issue of antioxidant effects in humans.

BACKGROUND: The grape volatile fraction determines to a great extent its aroma, which is one of the most important characteristics influencing wine quality and consumer preferences. Grapevine downy mildew (GDM) is one of the most important and devastating diseases of grapevines worldwide. In this study, the impact on the volatile composition of cv. Verdicchio grapes of classical copper formulations, was compared to that of alternative products. Thus, 11 treatments were foliar applied throughout one grapevine cycle. RESULTS: Most of the volatile compounds present in the grapes were not affected by the treatments used in order to prevent GDM. In the case of the total C norisoprenoids, some differences were found between grapes untreated control and those applied with the grapefruit seed extract. Moreover, the content of alcohols was smaller in grapes from Bordeaux mixture treatment and higher in the samples from chitosan application. CONCLUSION: Therefore, from the qualitative point of view of the berry, treatment with tested products alternative to copper is recommended, which minimize the environmental and health problems that this heavy metal brings to the soil and in the winery, since it means that their applications did not affect the grape aroma.

Multiple grape powdery mildew (PM) genetic resistance (R) loci have been found in wild grape species. Little is known about the defense responses associated with each R locus. In this study, we compare the defense mechanisms associated with PM resistance in interspecific crosses segregating for a single R locus from Muscadinia rotundifolia (Run1, Run1.2b, Run2.1, Run2.2), Vitis cinerea (Ren2), V. romanetii (Ren4D and Ren4U), and the interspecific hybrid Villard blanc (Ren3). By combining optical microscopy, visual scoring, and biomass estimation, we show that the eight R loci confer resistance by limiting infection at different stages. We assessed the defense mechanisms triggered in response to PM at 1 and 5 days post-inoculation (dpi) via RNA sequencing. To account for the genetic differences between species, we developed for each accession a diploid synthetic reference transcriptome by incorporating into the PN40024 reference homozygous and heterozygous sequence variants and de novo assembled transcripts. Most of the R loci exhibited a higher number of differentially expressed genes (DEGs) associated with PM resistance at 1 dpi compared to 5 dpi, suggesting that PM resistance is mostly associated with an early transcriptional reprogramming. Comparison of the PM resistance-associated DEGs showed a limited overlap between pairs of R loci, and nearly half of the DEGs were specific to a single R locus. The largest overlap of PM resistance-associated DEGs was found between Ren3+, Ren4D+, and Ren4U+ genotypes at 1 dpi, and between Ren4U+ and Run1+ accessions at 5 dpi. The Ren3+, Ren4D+, and Ren4U+ were also found to have the highest number of R locus-specific DEGs in response to PM. Both shared and R locus-specific DEGs included genes from different defense-related categories, indicating that the presence of E. necator triggered distinct transcriptional responses in the eight R loci.

A total of 13 samples of grapes (bunches) without apparent fungal contamination were analyzed. The samples were collected during the 2019 harvest from Vrbové village in the Small Carpathian region of Slovakia. For the isolation of fungi were used the direct plating technique on DRBC plates. The plates were incubated aerobically at 25 ±1 °C for one week in the dark. The data obtained from the cultivation of the grape berry samples revealed a high diversity of fungal species (a total of 1044 isolates were obtained). Alternaria and Rhizopus were the main components of the wine grape mycobiota of the Vrbovský subregion at harvest time (92%, each), followed by Cladosporium (85%), Penicillium (77%), Botrytis and Epicoccum (54%, each). The most abundant genera found by descending order were Penicillium (25%), Alternaria (24%), Cladosporium (20%), and Rhizopus (12%) and only in minor percentage by Aspergillus (3%) among others. The main fungal species isolated from genera Penicillium and Aspergillus were Penicillium expansum (57% RD) and A. section Nigri (97% RD). Of 17 analyzed Penicillium strains, 65% were able to produce at least one of the six mycotoxins analyzed in in vitro conditions by means of thin-layer chromatography method: citrinin, griseofulvin, patulin, cyclopiazonic acid, penitrem A, and roquefortin C.

This study was carried out to evaluate the socio-economic structure and organic farming activities of grape producers in Besni. A face-to-face survey was conducted with producers producing organic grapes. Data were obtained by interviewing a total of 50 producers. The number of organic products grown between 2005 and 2018 increased from 205 to 213 in Turkey. The number of producers engaged in cultivation in this area has increased more than 5 times compared to 2005. It reached nearly 80.000 in 2018. The area of organic vegetable production increased from 203.811 to 626.884 hectares in Turkey. While the production area increased 2.5 times, the production amount increased almost 4 times. Today, a variety of products in organic vegetable production, planting area, the number of farmers has increased in Turkey. The findings were obtained education level of the producers, the number of individuals in the family, record keeping, annual total income, newspaper reading, grape yield, grape cultivation area, property and rental land, credit usage, weed control, organic farming knowledge level, organic farming by using variables such as thoughts and grape variety. There are serious differences between conventional agriculture and organic farming. The family structure is effective in production activities. It has been determined that the thoughts of organic farming have changed depending on the age and experience of the producers. Organic grape producers have turned to organic farming due to their desire to sell their products at higher prices with state supports. Also, it has been suggested that organic farming is important in terms of the sustainability of living life all over the world. Increasing producer income and environmental awareness are among the main reasons for the transition to organic farming.

BACKGROUND: The pathway of anthocyanin biosynthesis, and its alterations leading to berry colour modification, are well known in grape skin. This variability could affect both quantity and quality of pigment accumulation. OBJECTIVE: The present work is focused on 15 grapevine cultivars selected to represent a high variability in the phenotypical colour traits in order to highlight new polymorphisms related to the flavonoid pathway. METHODS: Twenty-one genes involved in the biosynthetic pathway of anthocyanins were studied via targeted resequencing and were correlated with phenotypic data (anthocyanin profiles and spectroscopy indices). RESULTS: Single nucleotide polymorphism (SNP) and InDel (insertion/deletion) polymorphisms were detected. Out of 1751 polymorphic loci, 68% were SNPs and 32% were InDels (568). Cluster analysis and SPLS-DA were used to investigate the genetic relationships among the cultivars, confirming the large range of phenotypical variability. Statistically significant correlations were detected between accumulation of cyanidin-based anthocyanins and genetic polymorphisms in two structural genes and one transcription factor putatively involved in the anthocyanin biosynthetic pathway. CONCLUSIONS: The understanding of the polymorphisms related to the anthocyanin accumulation could support future selection of new pink table grape varieties with increased appeal on the consumers.

The seedlessness of grape derived from stenospermocarpy is one of the most prized traits of table or raisin grapes. It is controlled by a complex genetic system containing one major dominant gene and multiple minor recessive genes. Here, we collected dense variation data from high-depth resequencing data of seeded, seedless, and wild relative grape genomes sequenced to > 37x mean depth. Variant calls were made using a modified variant calling pipeline that was suitable for highly diverse interspecific grape accessions. The modified pipeline enabled us to call several million more variants than the commonly recommended pipeline. The quality was validated by Sanger sequencing data and subsequently supported by the genetic population structure and the phylogenetic tree constructed using the obtained variation data, results of which were generally consistent with known pedigree and taxonomic classifications. Variation data enabled us to confirm a major dominant gene and identify minor recessive loci for seedlessness. Incidentally, we found that grape cultivar Rizamat contains an ancestral chromosomal region of the major gene in Sultanina, a predominant seedlessness donor cultivar. Furthermore, we predicted new candidate causal genes including Vitvi01g00455, Vitvi08g01528, and Vitvi18g01237 associated with the minor seedless-regulating loci, which showed high homology with genes that regulate seed development in Arabidopsis. This study provides fundamental insights relevant to variant calling from genome resequencing data of diverse interspecific hybrid germplasms such as grape and will accelerate future efforts aimed at crop improvement.

The object of this study was to investigate the influence of bagging or the combination of umbrella and bags on berry composition, especially anthocyanin accumulation, in ‘Kyoho’ (V. labruscana) berry skins. Five different colors and bag materials commonly used in production were chosen for this study: white, green, yellow, nonwoven and transparent, and the umbrellas were made of plastic material. The composition and content of anthocyanins in the berry skin of ‘Kyoho’ were analyzed by HPLC-MS. The results showed that bagging or a combination of umbrella and bags on anthocyanin concentration in ‘Kyoho’ grapes showed inconsistent effects that depended on the colors and materials of the bags and the use of an umbrella or not. Compared with the control, the inhibition effects of white, green and yellow bags on the soluble solids content and the total and individual anthocyanin concentrations in ‘Kyoho’ grapes reached significant levels; effects of green and yellow bags were the most significant among them. The plastic umbrella used in this study also presented a lower relative light intensity and produced more even-colored clusters. White, green, and yellow bags and combinations of umbrella and bags significantly increased some stable anthocyanin proportions to varying degrees, such as coumaroylated and methylated anthocyanins, compared to the control.

Western Carpathians are historically the northern range traditional grapevine growing areas since the Middle Ages. The mean anual temperature has increased by about 1.1oC during the last century in Slovakia. Elevated temperature impacted the growing conditions of grapevine. Together with the increased temperatures, higher frequency of drought periods and parameters are evident. Traditional grape producing areas are facing new challenges. Except for the accelerated grapevine phenology, pathogene infection pressure and occurence of pests including new ones, as well as the quality of grapes influencing wine quality, are changing. In order to evaluate the drought impact on the quality parameters of grapes the locality of the Cultivar Testing Station in Dolné Plachtince which belongs to the Central Slovakian grape producing region was chosen. Interanual variability of the drought impact on the grape quality was evaluated according to Palmer drought severity index (PDSI). The 1990–2014 period was used as a basis for evaluation. The period with the phenological phase crucial for the grape quality formation was determined. Two groups of cultivars with different ripening periods were selected. Pinot Gris, Muscat Ottonel Weiss, Müller Thurgau represented the early ripening cultivars (OIV earliness code 4 and 5), whereas Grüner Veltliner, Riesling, Welschriesling represented the late ripening cultivars (OIV code 8 and 9) were used. The cumulative values of PDSI for the crucial periods were calculated. The PDSI values as well as the sugar and acid contents were correlated to find the strength of relation between them. Short drought periods did not influence the grape quality significantly, while long drought periods caused a decrease of the acid content and an increase of the sugar content. Though the tendency was clear, the correlation level was mostly low. The most sensitive period in this sense was July–September; however, it was influenced by the ripening term of individual cultivars. The results suggest the necessity of a thorough approach to cultivar selection, taking into account its vitality and ability to preserve a satisfactory acid content in grapes by the harvest date.

Aspergillus spp. are a common contaminant of grapes and a major source of mycotoxins. China is the largest producer of grape in the world now, however, the toxigenic Aspergillus population on grape in this country is still largely unknown. In this study, a total of 345 strains were isolated from grapes of 13 main grapevine producing regions in China. Based on calmodulin gene sequences, eight species within Aspergillus section Nigri were identified. Among them, Aspergillus tubingensis (48.7%) was predominant, followed by Aspergillus welwitschiae (20.6%) and Aspergillus aculeatinus (11%). Average of contamination level was up to 64.19%, and we found the occurrence of section Nigri species on the surface of fresh grapes was significantly influenced by the climate (P2 (FB2) producers. Of those, Aspergillus carbonarius was the main OTA producer and A. welwitschiae and Aspergillus niger were the main FB2 producers with different toxigenic ability. Our results highlight the potential risk of OTA and FB2 contamination by A. carbonarius and A. welwitschiae on grape in China, respectively; management strategies should be considered for preventing and reducing the toxigenic Aspergillus and its mycotoxins.

Cheap-change is a very practical and economical method for changing cultivar in vineyards using chip-budding grafting method. This method has many advantages over other classical grafting applications (cleft, notch and bark). A unique hand machine has been developed and training system is largely preserved in this method. It is seen that it is applied in a very practical way and approximate 2700 grapevines can be grafted daily with a team of 6 people in studies has been observed. In wine grapes (Vitis vinifera L. ‘Chardonnay’, ‘Sauvignon Blanc’, ‘Öküzgözü’, ‘Boğazkere’ and ‘Narince’) and table grapes (Vitis vinifera L. ‘Sultana’ and ‘Superior Seedless’), cheap-change “chip-budding grafting method” has been made in İzmir/Torbali and Halilbeyli regions in 2013 and 2014. Data like grafting take (%), yield (kg vine-1), average shoot length (cm) and grafting number (per day) are recorded. The grafting can be repeated several times in the same season. The grafting takes has ranged from 85.83% to 95.33% in first applications. In this method, there is no grapevine loss and the grape yield continues in the following year. This study was conducted to determine the effects of three different bud load levels (15 buds, 25 buds and 30 buds m-2) and two different leaf removal (25% and 50% on each vine shoot) treatments on number of bursted bud, midday leaf water potential (?mid), stomatal conductance (gs), total soluble solids (TSS) and titratable acidity (TA) of own-rooted ‘Sultana Seedless’ (Vitis vinifera L.) in 2016 and 2017 vegetation season in Alaşehir, Turkey. As a by-product of the winemaking process, grape seeds have been receiving increased attention worldwide due to a number of beneficial nutritional properties. Moreover, grape seed oil has also been gaining prominence for its high content of unsaturated fatty acids, primarily linoleic acid (C18:2) (72- 76%). The objective of this paper was to determine the influence of three different grape seed drying temperatures (20, 40 and 60 °C) on the quality of cold- pressed oils from five different grape varieties (Pinot noir, Pinot blanc, Muscat, Cabernet sauvignon and Graševina). The oils extracted were tested for the acid number (as an indicator of hydrolysis), the percentage of free fatty acids and the peroxide value (as an indicator of autoxidation). The peroxide values of the oil samples considered increased with the increasing drying temperature of grape seeds. The highest peroxide value was observed in the oil samples produced from the Graševina seeds (regardless of the grape seed drying temperature), whereas the Muscat oil samples exhibited the lowest peroxide values. The acid number and the percentage of free fatty acids of the oil samples considered were found to be variety specific and independent of the grape seed drying temperature

During the year 2018, the symptoms of bunch rot on Shine Muscat (Vitis vinifera L.) were observed in Kimcheon-si, Gyeongbuk province in Korea. The disease appears on the Shine Muscat as a black rot due to prolific fungal sporulation after it has invaded into the Shine Muscat which look completely empty and dryness. Colonies of these fungi are present on the Shine Muscat skin from fruit setting and increase in amount from early season to harvest, while become peak at ripening stage. To isolate the causal agent, small fragments (2 to 3 mm) of decayed tissue from the lesion margin were placed onto potato dextrose agar (PDA) plates. Fungal colonies on PDA produced dense white aerial mycelium and then covered with dark black conidial heads. These heads were large and radiate, and vesicles were globose (2.12–32.0×2.0–3.1 μm). Based on morphological and cultural characteristics, this fungus was identified as Aspergillus tubingensis. To confirm its identity, the internal transcribed spacer, β-tubulin, and RNA polymerase II was sequenced for molecular identification. BLAST search indicated 99% identity with A. tubingensis. The pathogenicity test on healthy grape of Shine Muscat produced bunch rot, as the original symptoms. To select effective fungicides for the control of brunch rot, an in vitro antifungal activity of seven fungicides were evaluated against the growth of A. tubingensis. Five fungicides (dipenoconazole, tebuconazole, metconazole, iminoctadine, and captan) exhibited significantly strong suppression of the mycelial growth of A. tubingensis.

No. of bitstreams: 0 Previous issue date: 2020-04-01 The effect of pectinolytic enzyme preparation (PEP) produced by the fungus Thermomucor indicae-seudaticae-N31 (PEP-N31) on total phenolic content, concentrations of methanol and color of grape juice was studied. Positive results were found when PEP-N31 was used to extract phenolic compounds after the grapes had been blanched for 3 min and macerated for 1 h. The resulting juice had better yield, color characteristics and higher phenolic content (1637.21 mg.L-1, as gallic acid equivalent, or GAE) than the conventionally prepared juice (1422.59 mg GAE.L-1), and it was very similar to the juice obtained through the treatment with a commercial enzyme (1682.10 mg GAE.L-1). The concentration of methanol in the juice produced with the PEP-N31 was less than 200 mg.L-1. These results encourage the use of PEP produced by Thermomucor indicae-seudaticae-N31 by the grape-processing industry. RNA editing is a post-transcriptional process of modifying genetic information on RNA molecules, which provides cells an additional level of gene expression regulation. Unlike mammals, in land plants, RNA editing converts C-to-U residues in organelles. However, its potential roles in response to different stressors (heat, salt, and so on) remains unclear. Grape is one of the most popular and economically important fruits in the world, and its production, like other crops, must deal with abiotic and biotic stresses, which cause reductions in yield and fruit quality. In our study, we tested the influence of the environmental factor temperature on RNA editing process in the whole mRNA from grape organelle. In total, we identified 122 and 627 RNA editing sites in chloroplast and mitochondria respectively with the average editing efficiency nearly ~ 60%. The analyses revealed that number of non-synonymous editing sites were higher than that of synonymous editing sites, and the amino acid substitution type tends to be hydrophobic. Additionally, the overall editing level decreased with the temperature rises, especially for several gene transcripts in chloroplast and mitochondria (matK, ndhB, etc.). We also found that the expression level of most PPR genes decreased with the temperature rises, which may contribute to the decline of RNA editing efficiency at high temperature. Our findings suggested that the RNA editing events were very sensitive to heat stress; the changes of amino acid in RNA editing genes may contribute to the stress adaption for grape. In this study, the effects of various digestive models, influencing factors and dietary supplements on the bioaccessibility of tebuconazole in table grapes were compared. The Caco-2 cell model was employed to reveal the transfer behavior of tebuconazole. The results indicated that digestion time is the main factor affecting bioaccessibility. With an increase in time, the tebuconazole in grapes was almost completely dissolved, with bioaccessibility reaching 98.5%, whereas dietary fiber reduced bioaccessibility. Tebuconazole undergoes carrier-free passive transport in permeable cells in the Caco-2 cell model. These findings have practical application value for correctly evaluating the harmful level of pollutants in the matrix to human body.

Grapes are rich in phenolic compounds, being important for human health with anti-inflammatory, antiatherosclerotic, antimutagenic, anticarcinogenic, antibacterial, antiviral, and antimicrobial activity. The winemaking of the grapes generates significant amounts of waste. These wastes contain bioactive compounds in their biomass that can be used as a source of food improvement or as a source of nutrition supplementation. This study looks at the content of bioactive compounds, the polyphenolic profile, and the antioxidant activity in different white and red grape pomaces. The investigation of bioactive characteristics (total polyphenols, total flavonoids, catechins, tannins, and antioxidant activity) was carried out by UV-Vis spectrophotometric methods, while the individual polyphenolic composition was investigated by target and screening UHPLC-HRMS/MS analysis. Principal components (PCA) and the heat maps analysis allows the discrimination between the grape pomace resulted from white grape cultivars (Muscat Ottonel and Tamaioasa Romaneasca) and red grape pomaces (Cabernet Sauvignon, Merlot, Feteasca Neagra, Burgund Mare, Pinot Nore), with the identification of the specific phenolic compounds for each grape pomace type.

(1) Background: Stenospermocarpic (Sultani Çekirdeksiz and Black Kishmish), parthenocarpic (Black Corinth), and seeded varieties (Ekşi Kara and Gök Üzüm) are used for raisin production. To our knowledge, there is little available information about the biochemical characteristics of raisins produced from these varieties. (2) Methods: Some metabolites, such as hormones, sugars, vitamins, minerals, and amino acids, including enzymatic activity, were determined in different raisin varieties. (3) Results: Seedless raisin varieties presented higher content of several hormones, vitamins, and minerals, as antioxidant capacity than the raisins produced from seeded varieties. Contrary to this, seeded raisin varieties presented higher contents of most measured sugars and amino acids than the raisins produced from seedless varieties. (4) Conclusions: Biological mechanisms of pollination and fertilization induced modifications in the primary and secondary metabolism of grapes, considerably affecting biochemical compounds and the antioxidant capacity of raisins.

With the frequent occurrence of extreme natural disasters, unfavorable growth environment is a common phenomenon in the life cycle of plants. In recent years, universal stress proteins (USPs) have attracted extensive attention in the field of plant science for their expression patterns and functional analysis. However, the characterization of the USP family remains unclear in grape. In this study, we identified 30 VvUSPs in the grape genome, which could be divided into three classes according to their encoded protein sequences, and this classification was reflected by the distribution of conserved motifs. Gene duplication analysis demonstrated that segmental duplication was an important pathway in the expansion of the VvUSP family. The expression patterns of 12 VvUSPs were significantly different between tissues, implying they had different functions in various tissues. Cis-acting element and expression analysis showed that most of the promoter regions of VvUSPs contained sequences responsive to hormones and stress elements, especially the promoter region of VIT_16s0013g01920. In conclusion, our findings provide comprehensive information for the further investigation of the genetics and protein functions of the USP gene family in grape.

Bronx Seedless and Italia (Vitis vinifera L.) are a variety preferred by consumers owing to their exciting flavour and widely cultivated in Aegean Region in Turkey. The aim was to identify the biogenic amines of these table grapes during berry ripeness. The biogenic amines were analyzed by HPLC in six different berry phenological stages. Italia grapes presented lower biogenic amine content than Bronx Seedless table grapes. The concentration of most of the biogenic amines analyzed linearly raised from the beginning of berry touch to when berries ripen for harvest stages. The most common biogenic amines in grape varieties were putrescine, followed by histamine, agmatine, and tyramine. There was also a positive correlation between all biogenic amines of the two grape varieties. The weakest correlation was found between spermine and cadaverine, whereas the strongest correlation was found among dopamine, trimethylamine, norepinephrine, tyramine, and histamine amines. The present study is the first report of a synthesis study regarding the effect of B.A.s on quality characteristics throughout berry ripeness in grape varieties containing foxy and muscat tastes. The concentration and composition of biogenic amines identified for both varieties might provide helpful information regarding human health and the vintage.

Conventional irrigation methods decrease greenhouse soil aeration, which leads to restricted root growth and reduced soil fungal abundance in greenhouse grapes. In this study, aerated irrigation equipment was used to investigate the effects of aerated irrigation on the biomass accumulation, root growth, and soil fungal community structure of grape seedlings. The results show that aerated irrigation significantly increased the root length, root surface area, root volume, and number of root tips by 38.5%, 32.1%, 62.1%, and 23.4%, respectively, at a soil depth of 20–40 cm (p ≤ 0.05). The chao1 index and ACE index of fungi at different soil depths under aerated irrigation were higher than those without aerated treatment; aerated irrigation changed the relative abundance of dominant fungi in rhizosphere soil. At a soil depth of 20–40 cm, aerated irrigation increased the abundance of Fusarium by 42.2%. Aerated irrigation also contributed to the abundance of the beneficial fungal genera Mortierella, Cladosporium, and Glomus. At a soil depth of 0–20 cm, the abundance of Mortierella in the soil that received aerated treatment was 180.6% higher than in the control treatment. These findings suggest that aerated irrigation is a promising strategy for the promotion of grape root growth and biomass accumulation, and it can also increase the abundance of some beneficial fungi.

Sucos de uvas são fontes importantes de flavonóides, principalmente flavan-3-óis em suas formas monoméricas de catequinas [(+)-catequina e (-)-epicatequina]. Os objetivos deste trabalho foram caracterizar e quantificar fenólicos totais (TP), monômeros de catequinas (CAT+EPI), atividade sequestrante de radicais livres (RSA) e específicos atributos sensoriais, durante o processo e armazenamento de sucos de uva comerciais. Sucos concentrados (68° Brix) do s cultivares Concord (CCJ) e Isabel (CIJ) foram amostrados durante o processo e após a concentração e armazenados a 5°C. Sucos de uva pasteurizados integ rais (14 a 19° Brix) de mesmos cultivares (PCJ e PIJ) foram mantidos à temperatura ambiente e luz indireta por 10 meses. Ambas situações simularam as condições de armazenamento industriais/comerciais. TP foi determinado pelo método de Folin-Ciocalteu; CAT+EPI por CLAE com detecção por fluorescência e RSA em ensaio com DPPH. Para o teste sensorial, a análise descritiva quantitativa (ADQ) foi utilizada com a seguinte lista pré-determinada de atributos e descritores: cor, adstringência, gosto amargo, gosto doce e sabor característico de suco de uva. As principais substâncias presentes nos sucos foram investigadas por espectrometria de massa com ionização por ¿electrospray¿ e infusão direta (ESI-MS), ambos no modo íon negativo. Durante o processamento, CCJ demonstrou maior RSA e TP em relação a CIJ com certa variação em cada etapa. CCJ apresentou 50% maior teor de TP, enquanto que a RSA foi em média 25% superior. No armazenamento, a preservação de TP variou entre 93% (CCJ) e 84% (PCJ) e a de RSA entre 87% (PIJ) e 85% (CCJ e PCJ). A retenção de (+)-catequina no período esteve entre 63% (PCJ) e 52% (PIJ) e a de (-)-epicatequina ente 32% (CCJ) e 15% (CIJ). Sucos concentrados mostraram maiores concentrações de catequinas e CCJ apresentou o maior teor de TP entre as amostras. Os conteúdos de TP e catequinas foram distintos entre os sucos, porém a RSA foi similar. PIJ obteve maior valor de RSA ¿por unidade de fenólico¿. Nos sucos concentrados, peonidina e peonidina-3-O-glucosídeo deram lugar a malvidina e dimetoxi-flavilium como substâncias significativas durante o processo; malvidina e piceatanol-O-glucosídeo decresceram após oito meses de armazenamento e dimetoxi-flavilium manteve-se estável. No aspecto sensorial, sucos concentrados sob refrigeração demonstraram maior intensidade em atributos desejáveis como cor, gosto doce e sabor característico após oito meses. CCJ apresentou maior estabilidade durante o armazenamento e seus atributos característicos foram adstringência, gosto amargo e intensidade de cor. CIJ foi caracterizado por gosto doce e sabor de suco de uva. As análises e respectivos resultados demonstraram que o processamento e as condições de armazenamento aplicados comercialmente mantiveram o a capacidade antioxidativa e a qualidade sensorial de sucos de uva concentrados Abstract: Epidemiological studies have demonstrated that polyphenol-rich foods and beverages exert beneficial physiological effects. Grape juices are relevant sources of flavonoids, particularly flavan-3-ols, mostly in the monomeric forms of catechins [(+)-catechin and (-)-epicatechin]. The objectives of this work were to characterize and quantify phenols (TP), catechin monomers (CAT+EPI), radical scavenging activity (RSA) and specific sensory attributes during processing and storage of commercial grape juices. Concentrated Concord (CCJ) and Isabel (CIJ) grapes juices (68° Brix) were sampled during processing an d after concentration and stored at 5°C. Pasteurized ready-to-drink juices (1 4 to 19° Brix) of the same grape cultivars (PCJ and PIJ) were kept at room temperature under indirect lighting for 10 months, both simulating industrial/commercial storage conditions. TP were determined using the Folin-Ciocalteu method; CAT+EPI by HPLC with fluorescence detection and RSA by the DPPH assay. For the sensory analysis, the quantitative descriptive analysis (QDA) was used with a pre-determined list of attributes: colour, astringency, bitterness, sweetness and characteristic grape juice flavour. The major components of juices were investigated by direct infusion and electrospray ionization mass spectrometry (ESI-MS) both in the negative ion mode. During processing, CCJ showed higher RSA and TP contents than CIJ with slight variations at each step. Although CCJ showed 50% higher TP contents, RSA was on average 25% higher. During storage, TP retention ranged from 93% (CCJ) to 84% (PCJ) and RSA from 87% (PIJ) to 85% (CCJ and PCJ). (+)-Catechin retention during storage ranged between 63% (PCJ) and 52% (PIJ) and (-)-epicatechin between 32% (CCJ) and 15% (CIJ). Concentrated juices of both cultivars showed notably higher catechin amounts and CCJ presented the greatest phenolic contents. Despite the marked differences in TP and CAT+EPI contents, similar RSA values were observed for all the juices. PIJ yielded the highest RSA ¿per phenolic unit¿, with the lowest TP content. In the concentrated juices, peonidin and peonidin-3-O-glucoside gave way to malvidin and dimethoxy-flavylium as significant components during processing; malvidin and piceatanol-O-glucoside decreased after 8-month storage whereas dimethoxy-flavylium was maintained. Regarding the sensory aspects, concentrated juices stored under refrigeration received higher ratings for desirable attributes such as colour, sweetness and flavour than juices aged in bottles at room temperature. CCJ was more stable during storage with sensory characteristics of astringency, bitterness and higher colour intensity while CIJ was characterized by sweetness and juice flavour. A novel clonal variety of Vitis vinifera was identified from “Chardonnay” using inter-simple sequence repeat (ISSR) markers and called “bud mutation. ” The metabolomic profiles in Chardonnay and bud mutation berries indicated essential differences in the expression of key genes in the pathways of 2-C-methyl-D-erythritol-4-phosphate (MEP) and lipoxygenase-hydroperoxide lyase (LOX-HPL). Bud mutation fruits also matured 10 days earlier than Chardonnay and have higher carotenoid, sugar, and acidic compound contents. Furthermore, the gene expression was examined in the biosynthetic pathways of two ripening-associated hormones, abscisic acid (ABA) and jasmonic acid (JA), which significantly increased in bud mutation compared with the Chardonnay fruit. The synthesis and metabolism of amino acids, terpenes, fatty acids, volatile components, and specialized metabolites significantly increased in bud mutation. Therefore, in comparison with Chardonnay, bud mutation is considered a highly aroma-producing grape variety for an improvement in the beverage industry.

In the Valpolicella area (Verona, Italy) Vitis vinifera cv. Corvina is the main grape variety used to produce Amarone wine. Before starting the winemaking process, the Corvina grapes are stored in a withering (i.e., dehydrating) warehouse until about 30% of the berry weight is lost (WL). This practice is performed to concentrate the metabolites in the berry and enrich the Amarone wine in aroma and antioxidant compounds. In compliance with the guidelines and strict Amarone protocol set by the Consorzio of Amarone Valpolicella, withering must be carried out by setting the grapes in a suitable environment, either under controlled relative air humidity (RH) conditions and wind speed (WS)—no temperature modification is to be applied—or, following the traditional methods, in non-controlled environmental conditions. In general, the two processes have different dehydration kinetics due to the different conditions in terms of temperature, RH, and WS, which affect the accumulation of sugars and organic acids and the biosynthesis of secondary metabolites such as stilbenes and glycoside aroma precursors. For this study, the two grape-withering processes were carried out under controlled (C) and non-controlled (NC) conditions, and the final compositions of the Corvina dried grapes were compared also to evaluate the effects on the organoleptic characteristics of Amarone wine. The findings highlighted differences between the two processes mainly in terms of the secondary metabolites of the dried grapes, which affect the organoleptic characteristics of Amarone wine. Indeed, by the sensory evaluation, wines produced by adopting the NC process were found more harmonious, elegant, and balanced. Finally, we can state how using a traditional system, grapes were characterised by higher levels of VOCs (volatile compounds), whilst wines had a higher and appreciable complexity and finesse.

Replant disease significantly hinders the development of the grape industry, and the imbalance of the rhizosphere microecological environment is one of the fundamental reasons hindering grape replants. Peanut is a common intercropping crop, and whether the root exudates of peanut can alleviate grape replant obstacles is still unknown. In this study, the effects of exogenous peanut root exudates on replanting grapevine growth, and the microbial community structure of grapevine replant soils were studied. The results showed that peanut root exudates could promote the growth of replanting grapevine seedlings; enhance root vigor and SOD activity, increasing 55.18% and 95.71%, respectively; and reduce the MDA content of root, decreasing 31.10%. After peanut exudate treatment, the growth of Fusarium solanum, an important harmful fungus that is an obstacle to grape replant, was inhibited. The relative abundances of Gaiella in bacteria and Cystobasidium and Mortierella in fungi increased, and the potential pathogen fungi Fusicolla decreased. Peanut root exudates also modified the soil bacterial and fungal community in a certain range and increased the interaction among the bacteria of grapevine rhizosphere soil. However, they loosened the interaction among fungi. There are extensive mutualistic interactions among bacteria or fungi in grape rhizosphere assemblages after peanut exudates treatment. Therefore, peanut root exudates might be helpful in changing the soil microbial environment and alleviating the grape replanting obstacle.

To guide grape picking robots to recognize and classify the grapes with different maturity quickly and accurately in the complex environment of the orchard, and to obtain the spatial position information of the grape clusters, an algorithm of grape maturity detection and visual pre-positioning based on improved YOLOv4 is proposed in this study. The detection algorithm uses Mobilenetv3 as the backbone feature extraction network, uses deep separable convolution instead of ordinary convolution, and uses the h-swish function instead of the swish function to reduce the number of model parameters and improve the detection speed of the model. At the same time, the SENet attention mechanism is added to the model to improve the detection accuracy, and finally the SM-YOLOv4 algorithm based on improved YOLOv4 is constructed. The experimental results of maturity detection showed that the overall average accuracy of the trained SM-YOLOv4 target detection algorithm under the verification set reached 93.52%, and the average detection time was 10.82 ms. Obtaining the spatial position of grape clusters is a grape cluster pre-positioning method based on binocular stereo vision. In the pre-positioning experiment, the maximum error was 32 mm, the mean error was 27 mm, and the mean error ratio was 3.89%. Compared with YOLOv5, YOLOv4-Tiny, Faster_R-CNN, and other target detection algorithms, which have greater advantages in accuracy and speed, have good robustness and real-time performance in the actual orchard complex environment, and can simultaneously meet the requirements of grape fruit maturity recognition accuracy and detection speed, as well as the visual pre-positioning requirements of grape picking robots in the orchard complex environment. It can reliably indicate the growth stage of grapes, so as to complete the picking of grapes at the best time, and it can guide the robot to move to the picking position, which is a prerequisite for the precise picking of grapes in the complex environment of the orchard.

In this study, phenolic extractability of Cabernet Sauvignon grapes from two California regions (Sonoma County and Central Coast) and its relation with skin cell wall composition was investigated. Phenolic grape composition, wine phenolic content as well as berry and pomace cell wall composition of three sites per region were determined. Grape cell wall material (CWM) composition, and thus pomace CWM composition, was impacted by the growing region. The process of fermentation modified CWM composition, solubilizing some of the compounds such as pectin and polysaccharides making pomace CWM composition from different sites more similar in the case of Sonoma County and more different for the samples grown in the Central Coast. Growing region had a significant impact on grape phenolics, particularly on flavan−3-ols and polymeric phenols, whereas polymeric pigments and anthocyanin contents were more similar among samples. Wines made from Sonoma County grapes showed higher anthocyanin and polymeric phenol content when compared to wines made from Central Coast grapes. Comparing wine to grape phenolic composition suggests a large difference in extractability based on region. Of all the CWM components analyzed, only lignin and the amount of cell wall isolated were found to have a significant impact on phenolic extractability.

Phenolic compounds are responsible for color and, with it, the quality of red wines. Its content will depend, among other factors, on the oenological techniques applied in the winery and, among them, the maceration time. Long maceration times can generate logistical problems at the moment of maximum grape entry into the winery. Applying techniques that accelerate the extraction of phenolic compounds from the grape solid parts to the must-wine would allow reducing the maceration time. Among the techniques of interest, microwaves use electromagnetic waves to produce dipole rotation and ion conduction, which can increase the elasticity of cell walls and cause the destruction of the cell membrane, facilitating the phenolic compound extraction. To study this effect, crushed grapes of the Cabernet Sauvignon variety were treated with this technology, macerated for 72 h and 7 days, and compared with respect to a control wine without any treatment. The results showed that the application of microwaves to the crushed grape favored a rapid extraction of the phenolic compounds, increasing the color of the wine and with only 72 h of maceration, the wines from treated grapes showed a similar phenolic content and chromatic characteristics to a control wine with 7 maceration days. The extraction of the less astringent skin tannins and the formation of stable pigments was also favored indicating that this technology may allow reducing the maceration time during the winemaking process without affecting the quality and stability of the wine.

(1) Background: To solve the problems of incomplete coloration and quality decline caused by unreasonable use of regulators in grapes, this study clarified the differences in the effects of a hormone-type growth regulator (AUT) and two commercial regulators on grape coloration and quality through field experiments. (2) Methods: The color indexes (brightness (L*), red/green color difference (a*), yellow/blue color difference (b*), and color index for red grapes (CIRG)) of grape fruit were measured using a CR-400 handheld color difference meter. The titratable acid content, total phenol content, and total sugar content were measured using anthrone colorimetry, folinol colorimetry, and NaOH titration, respectively, and the chalcone isomerase activity, phenylalanine ammoniase activity, dihydroflavol reductase activity, and anthocyanin content were measured using a UV spectrophotometer. (3) Results: The a*, total sugar and total phenol contents, and chalcone isomerase (CHI) and phenylalanine ammoniase (PAL) activities of grape fruit in the AUT treatment significantly increased, while the titratable acid content significantly decreased, compared to those in the CK treatment. The expressions of the differentially expressed genes (DEGs) trpB and argJ in AUT treatment were significantly up-regulated. The expressions of the differentially expressed metabolites (DEMs) phenylalanine and 4-oxoproline were significantly up-regulated, while those of 3,4-dihydroxybenzaldehyde and N-acetyl glutamate were significantly down-regulated. The CIRG significantly increased by 36.4% compared to that in the CK, indicating improved fruit coloration. (4) Conclusion: The AUT could shorten the color conversion period of grape fruit and improve the coloration, taste, and tolerance to saline and alkaline stresses.
