White sponge nevus is a rare benign autosomal dominant disorder with variable penetrance. It is characterized by asymptomatic white plaques affecting mainly the oral mucosa. Careful clinical and histopathological examination is indicated to exclude other more serious conditions presenting as oral white lesions. Herein, we present a new case of oral white sponge nevus in a 17-year-old Iranian male with no familial background.

Visceral leishmaniasis (VL) is systemic zoonotic parasitic infection that is a health problem in some tropical and subtropical countries. The purpose of our study is to determine the seroprevalence of canine visceral leishmaniasis (CVL) in owned dogs of the Sarab area and to identify the species of Leishmania isolated from these dogs. We also compared the sensitivities and specificities of two polymerase chain reaction (PCR) assays (kDNA and ITS1) used for Leishmania infantum identification with culture, microscopic detection and enzyme-linked immunosorbent assay (ELISA) methods as well as validate the PCR techniques for the molecular diagnosis of CVL. Sera from 384 dogs of 30 villages around Sarab, were tested by ELISA and buffy coat blood fractions after sampling tested with PCR by specific primers (kDNA, ITS-18sRNA). Thirty-five dogs were seropositive by ELISA. The seroprevalence rate (SPR) of CVL was 9.1% (CI, 95% 6.6 -12.4). The most important serological result was a high proportion of seropositivity for leishmaniasis. Out of 361 (94%) asymptomatic dogs, 31 (8.6%) were seropositive, and out of 23 (6%) symptomatic dogs, 4 (17.4%) were seropositive. Agreement betweenthe ELISA test and clinical signs was 86.7%. Each assay was performed on 60 blood samples. PCR of kDNA (7/60 positives, 11.8%) was the most sensitive of the assays examined, followed by ELISA (3/60, 5%) and ITS1-PCR (2/60, 3.4 %). All diagnostic assays were highly specific (100 %) and had positive predictive values (PPV) >90% and negative predictive values (NPV) >88% for CVL. As expected, kDNAPCR proved to be the most sensitive (87.5 %) assay for leishmanial DNA in peripheral blood. This study shows that kDNA-PCR is significantly more sensitive than the other parasitological and serological methods, allowing the identification of infected dogs even before the appearance of serum L. infantum antibodies. Because kDNA-PCR is the most reliable, sensitive, and also a rapid diagnostic assay for CVL, it should be employed as the new standard for routine diagnosis.Key words: Leishmania infantum, polymerase chain reaction, kinetoplast DNA, enzyme-linked immunosorbent assay, Visceral leishmaniasis, dogs, prevalence.

Background: Cancer/Testis Antigens (CTAs) are a sub-group of tumor-associated antigens which are expressed normally in germ line cells and trophoblast, and aberrantly in a variety of malignancies. One of the most important CTAs is Developmental Pluripotency Associated-2(DPPA2) with unknown biological function. Considering the importance of DPPA2 in developmental events and cancer, preparing a suitable platform to analyze DPPA2 roles in the cells seems to be necessary. Methods: In this study, the coding sequence of DPPA2 gene was amplified and cloned into the retroviral expression vector to produce recombinant retrovirus. The viral particles were transducted to Esophageal Squamous Cell Carcinoma (ESCC) cell line (KYSE-30 cells) and the stable transducted cells were confirmed for ectopic expression of DPPA2 gene by real-time PCR. Results: According to the critical characteristics of retroviral expression system such as stable and long time expression of interested gene and also being safe due to deletion of retroviral pathogenic genes, this system was used to induce expression of DPPA2 gene and a valuable platform to analyze its biological function was prepared. Transduction results clearly showed efficient overexpression of the gene in target cells in protein level due to high level of GFP expression. Conclusion: Such strategies can be used to produce high levels of desired protein in target cells as a therapeutic target. The produced recombinant cells may present a valuable platform to analyze the effect of DPPA2 ectopic expression in target cells. Moreover, the introduction of its potential capacity into the mouse model to evaluate the tumorigenesis of these cancer cells in vivo leads to an understanding of the biological importance of DPPA2 in tumorigenesis. In addition, our purified protein can be used in a mouse model to produce specific antibody developing a reliable detection of DPPA2 existence in any biological fluid through ELISA system.

Objective(s): The limited homing potential of bone-marrow-derived mesenchymal stem cells (BM-MSC) is the key obstacle in MSC-based therapy. It is believed that chemokines and chemokine receptor interactions play key roles in cellular processes associated with migration. Meanwhile, MSCs express a low level of distinct chemokine receptors and they even lose these receptors on their surface after a few passages which influence their therapeutic applications negatively. This study investigated whether treatment of BM-MSCs with hypoxia-mimicking agents would increase expression of some chemokine receptors and cell migration. Materials and Methods: BM-MSCs were treated at passage 2 for our gene expression profiling. All qPCR experiments were performed by SYBR Green method in CFX-96 Bio-Rad Real-Time PCR. The Boyden chamber assay was utilized to investigate BM-MSC homing. Results: Possible approaches to increasing the expression level of chemokine receptors by different hypoxia-mimicking agents such as valproic acid (VPA), CoCl2, and desferrioxamine (DFX) are described. Results show DFX efficiently up-regulate the CXCR7 and CXCR4 gene expression while VPA increase only the CXCR7 gene expression and no significant change in expression level of CXCR4 and the CXCR7 gene was detectable by CoCl2 treatment. Chemotaxis assay results show that pre-treatment with DFX, VPA, and Cocl2 enhances significantly the migration ability of BM-MSCs compared with the untreated control group and DFX treatment accelerates MSCs homing significantly with a higher rate than VPA and Cocl2 treatments. Conclusion: Our data supports the notion that pretreatment of MSC with VPA and DFX improves the efficiency of MSC therapy by triggering homing regulatory signaling pathways.
