Abstract

MicroRNAs (miRNAs) are small non-coding RNAs that regulate critical cell processes such as cell proliferation, apoptosis and differentiation by modulating gene expression. MiRNAs deregulation has been observed extensively in cancer. Elegant studies have demonstrated that miRNAs are involved in the initiation and progression of several malignancies. In this review we will address the role of miRNAs in the diagnosis and prognosis of cancer. The development of new drugs mimicking or blocking miRNAs will be discussed.

Abstract

The ZEUS inclusive differential cross-section data from HERA, for charged and neutral current processes taken with e+ and e- beams, together with differential cross-section data on inclusive jet production in e+ p scattering and dijet production in \gamma p scattering, have been used in a new NLO QCD analysis to extract the parton distribution functions of the proton. The input of jet data constrains the gluon and allows an accurate extraction of \alpha_s(M_Z) at NLO; \alpha_s(M_Z) = 0.1183 \pm 0.0028(exp.) \pm 0.0008(model) An additional uncertainty from the choice of scales is estimated as \pm 0.005. Abstract

The production of D^*+, D^0, D^+, D_s^+ and Lambda_c^+ charm hadrons and their antiparticles in ep scattering at HERA was measured with the ZEUS detector using an integrated luminosity of 79 pb^-1. The measurement has been performed in the photoproduction regime with the exchanged-photon virtuality Q^2 < 1 GeV^2 and for photon-proton centre-of-mass energies in the range 130 < W < 300 GeV. The charm hadrons were reconstructed in the range of transverse momentum p_T(D, Lambda_c) > 3.8 GeV and pseudorapidity |eta(D, Lambda_c)| < 1.6. The production cross sections were used to determine the ratio of neutral and charged D-meson production rates, R_u/d, the strangeness-suppression factor, gamma_s, and the fraction of charged D mesons produced in a vector state, P_v^d. The measured R_u/d and gamma_s values agree with those obtained in deep inelastic scattering and in e^+e^- annihilations. The measured P_v^d value is smaller than, but consistent with, the previous measurements. The fractions of c quarks hadronising as a particular charm hadron, f(c -> D, Lambda_c), were derived in the given kinematic range. The measured open-charm fragmentation fractions are consistent with previous results, although the measured f(c -> D^*+) is smaller and f(c -> Lambda_c^+) is larger than those obtained in e^+e^- annihilations. Cross sections for e(+)p neutral current deep inelastic scattering have been measured at a center-of-mass energy of roots=318 GeV with the ZEUS detector at HERA using an integrated luminosity of 63.2 pb(-1). The double-differential cross section, d(2)sigma/dxdQ(2), is presented for 200 GeV2 lt Q(2) lt 30 000 GeV2 and for 0.005 lt x lt 0.65. The single- differential cross sections dsigma/dQ(2), dsigma/dx and dsigma/dy are presented for Q(2) gt 200 GeV2. The effect of Z- boson-exchange is seen in dsigma/dx measured for Q(2) gt 10 000 GeV2. The data presented here were combined with ZEUS e(+)p neutral current data taken at roots=300 GeV and the structure function F-2(em) was extracted. All results agree well with the predictions of the Standard Model.

Abstract

The photoproduction of bottom quarks in events with two jets and a muon has been measured with the ZEUS detector at HERA using an integrated luminosity of 110 ${\mathrm{pb}}^{\mathrm{\ensuremath{-}}1}$. The fraction of jets containing b quarks was extracted from the transverse momentum distribution of the muon relative to the closest jet. Differential cross sections for bottom production as a function of the transverse momentum and pseudorapidity of the muon, of the associated jet and of ${x}_{\ensuremath{\gamma}}^{\mathrm{jets}},$ the fraction of the photon's momentum participating in the hard process, are compared with MC models and QCD predictions made at next-to-leading order. The latter give a good description of the data.

Abstract

A study of the $\overline{\ensuremath{\nu}}$-jet mass spectrum in ${e}^{+}\stackrel{\ensuremath{\rightarrow}}{p}\overline{\ensuremath{\nu}}X$ events at a center-of-mass energy 300 GeV has been performed with the ZEUS detector at the HERA collider at DESY using an integrated luminosity of 47.7 ${\mathrm{pb}}^{\ensuremath{-}1}.$ The mass spectrum is in good agreement with that expected from standard model processes over the $\overline{\ensuremath{\nu}}$-jet mass range studied. No significant excess attributable to the decay of a narrow resonance is observed. By using both ${e}^{+}\stackrel{\ensuremath{\rightarrow}}{p}{e}^{+}X$ and ${e}^{+}\stackrel{\ensuremath{\rightarrow}}{p}\overline{\ensuremath{\nu}}X$ data, mass-dependent limits are set on the s-channel production of scalar and vector resonant states. Couplings to first-generation quarks are considered and limits are presented as a function of the ${e}^{+}q$ and $\overline{\ensuremath{\nu}}q$ branching ratios. These limits are used to constrain the production of leptoquarks and R-parity violating squarks.

Abstract

UCRs are a subset of conserved sequences (100% between orthologous regions of human, rat and mouse genomes) located in both intra- and intergenic regions. There are 481 UCRs described and more than 50% of all the UCRs (256/481) has been classified as non exonic (with no evidence of coding protein), while the other 47% have been designated either exonic (overlap mRNAs of known genes), or possible exonic (with no clear evidence of overlap with protein coding genes) (Bejerano et al, Science 2004). Recently, our group reported that a subset of transcribed UCRs is aberrantly expressed in leukemia (Calin et al, Cancer Cell 2007). Because of the high degree of conservation, the UCRs are likely to be functional and may be involved in the phylogeny of mammals. Since hematopoiesis is regulated by signaling pathways and transcription factors that are highly conserved throughout phylogeny, we hypothesize that UCRs may be differentially expressed in hematopoietic tissues and may play a role in the regulation of this process. In this study we analyzed the UCRs expression in normal hematopoiesis by performing microarray analysis of hematopoietic precursors (HPCs) obtained by culturing human bone marrow (non-mobilized) CD34+ selected cells with different cytokine combinations for 2 weeks to stimulate differentiation to the erythrocyte (E), megakaryocyte (MK), monocyte (M) and granulocyte lineages (G). The following cytokine combinations were used: E (TPO/SCF/IL-3); G (G-CSF, GM-CSF, SCF, IL-3, IL-3); MK (TPO, SCF, IL-3) and M (M-CSF, GM-CSF, IL-6, IL-3 and SCF). Differentiation to the selected lineages was monitored every 3 days using morphology, special staining (benzidine) and flow cytometry analysis using appropriate lineage specific antibodies. Human bone marrow CD34+ and peripheral blood (PB) CD3+ (pan-T cells) and CD19+ (B-lymphocytes) selected cells were obtained from 3 different donors. Total RNA was obtained from the E and M cultures at days 7, 9, 11 and 14; G (days 10 and 14) and MK at day 14 and was hybridized in duplicate to the UCRs microarray chip (OSU version 4). After normalization of the array data with quantiles we analyzed the data using the BRB tools. Unsupervised analyses of the data revealed that samples segregated mainly in 6 main clusters corresponding to the M, E, G and Mk lineages, PB lymphocytes and CD34+ cells. Next we performed a series of two class analysis where we compared the UCRs expression of CD34+ cells vs. differentiated cells (E, G, MK, M, T and B-lymphocytes) one at a time (i.e. CD34+ cells vs. E). In table 1 we showed the top UCRs differentially expressed between in vitro differentiated HPCs/PB lymphocytes and CD34+ cells (+ or − sign reflects if the UCRs is in the sense (+) or antisense orientation (−). We validated the array results for uc.283+, uc.285+ and uc.161+ in a panel of differentiated CD34+ HPCs and CD34+ cells using qRT-PCR. In summary we have characterized the UCRs expression during hematopoietic differentiation of HSC and identified distinctive signatures associated with particular lineages. This research has the potential to identify novel regulators of hematopoiesis and may give insights into basic biology of gene expression and cell fate determination.

Abstract

The photoproduction reaction γp → μ+ μ-p has been studied in ep interactions using the ZEUS detector at HERA. The data sample corresponds to an integrated luminosity of 43.2pb-1. The Υ meson has been observed in photoproduction for the first time. The sum of the products of the elastic Υ(1S), Υ(2S),Υ(3S) photoproduction cross sections with their respective branching ratios is determined to be 13.3 ± 6.0(stat.)-2.3+2.7(syst.) pb at a mean photon-proton centre of mass energy of 120 GeV. The cross section is above the prediction of a perturbative QCD model. © 1998 Elsevier Science B.V. All rights reserved.
