Open Access article, published by EDP Sciences, under the terms of the Creative Commons Attribution License (http://creativecommons.org/licenses/by/4.0), which permits unrestricted use, distribution, and reproduction in any medium, provided the original work is properly cited., We directly measured twenty overhanging cliffs on the surface of comet 67P/Churyumov-Gerasimenko extracted from the latest shape model and estimated the minimum tensile strengths needed to support them against collapse under the comet's gravity. We find extremely low strengths of around 1 Pa or less (1 to 5 Pa, when scaled to a metre length). The presence of eroded material at the base of most overhangs, as well as the observed collapse of two features and the implied previous collapse of another, suggests that they are prone to failure and that the true material strengths are close to these lower limits (although we only consider static stresses and not dynamic stress from, for example, cometary activity). Thus, a tensile strength of a few pascals is a good approximation for the tensile strength of the 67P nucleus material, which is in agreement with previous work. We find no particular trends in overhang properties either with size over the ~10-100 m range studied here or location on the nucleus. There are no obvious differences, in terms of strength, height or evidence of collapse, between the populations of overhangs on the two cometary lobes, suggesting that 67P is relatively homogenous in terms of tensile strength. This paper presents an analysis of the physical properties of craters on asteroid (21) Lutetia, derived from images acquired by OSIRIS, the high-resolution cameras onboard ESA's spacecraft Rosetta. Crater morphology on (21) Lutetia fits very well with the general picture of what was known for previously visited small bodies, with a typical depth to diameter ratio of 0.12. We discuss here the distribution of this parameter all across the surface, but also region by region, and see how it can vary from one location to another and help to distinguish between different geological units. In a later section of the paper we study in more details Beatica region where a deep ejecta blanket filled most of the craters, and estimate the thickness profile of this ejecta based on our analysis of the d / D . We find a good agreement with existing scaling laws, and use this to constrain the scale of the original event that reshaped the surface around the North pole of the asteroid. Finally, we report on the observations of avalanches in several crater flanks, and the presence of many asymmetrical craters with flow-like features, and discuss the evidence for widespread fine material all over the surface.

Dust jets, i.e. fuzzy collimated streams of cometary material arising from the nucleus, have been observed in-situ on all comets since the Giotto mission flew by comet 1P/Halley in 1986. Yet their formation mechanism remains unknown. Several solutions have been proposed, from localized physical mechanisms on the surface/sub-surface (see review in Belton (2010)) to purely dynamical processes involving the focusing of gas flows by the local topography (Crifo et al. 2002). While the latter seems to be responsible for the larger features, high resolution imagery has shown that broad streams are composed of many smaller features (a few meters wide) that connect directly to the nucleus surface. We monitored these jets at high resolution and over several months to understand what are the physical processes driving their formation, and how this affects the surface. Using many images of the same areas with different viewing angles, we performed a 3-dimensional reconstruction of collimated jets, and linked them precisely to their sources on the nucleus. Results.We show here observational evidence that the Northern hemisphere jets of comet 67P arise from areas with sharp topographic changes and describe the physical processes involved. We propose a model in which active cliffs are the main source of jet-like features, and therefore the regions eroding the fastest on comets. NASA's Deep Impact spacecraft met its intended fate on 4 July this year when it collided with Comet Tempel 1 at a relative velocity of over 36,000 km per hour. Telescopes all around the world and beyond it were trained on the event in order to glean information about the internal structure of a comet. Results from OSIRIS cameras on ESA's Rosetta spacecraft provided continuous coverage from 3 days before to 10 days after the impact. The impact created a crater 60 metres in diameter and the nature of the debris points to the comet as an ‘icy dustball’ rather than the more familiar concept of a ‘dirty snowball’. Comets spend most of their life in a low-temperature environment far from the Sun. They are therefore relatively unprocessed and maintain information about the formation conditions of the planetary system, but the structure and composition of their nuclei are poorly understood. Although in situ1 and remote2 measurements have derived the global properties of some cometary nuclei, little is known about their interiors. The Deep Impact mission3 shot a projectile into comet 9P/Tempel 1 in order to investigate its interior. Here we report the water vapour content (1.5 × 1032 water molecules or 4.5 × 106 kg) and the cross-section of the dust (330 km2 assuming an albedo of 0.1) created by the impact. The corresponding dust/ice mass ratio is probably larger than one, suggesting that comets are ‘icy dirtballs’ rather than ‘dirty snowballs’ as commonly believed4. High dust velocities (between 110 m s-1 and 300 m s-1) imply acceleration in the comet's coma, probably by water molecules sublimated by solar radiation. We did not find evidence of enhanced activity of 9P/Tempel 1 in the days after the impact, suggesting that in general impacts of meteoroids are not the cause of cometary outbursts.

In the field of cardiovascular research, the pig is considered to be an excellent animal model of human diseases. It is well-known that primary cultures of endothelial cells (ECs) are a powerful tool for the study of vascular physiology and pathology, and, according to the principles of the Three Rs, their use results in a substantial reduction in the numbers of experimental animals required. However, a limitation of EC culture is that the cells are not in their physiological context. Here, we describe and characterise a method for the culture of porcine vessels that overcomes the limitation of EC cultures, with the advantage of reducing the number of animals used for research purposes. The organ cultures were set-up by using an aortic cylinder obtained from the arteries of control pigs sacrificed for other experimental purposes. In order to characterise the method, vascular endothelial growth factor (VEGF) secretion, matrix metalloproteinase (MMP) activation and the vessel's structural features were evaluated during organ culture. These analyses confirm that the culture of aortic cylinder lumen, in a medium specific for ECs, results in a stable system in terms of VEGF and MMP secretion. The ECs do not undergo cell division during the organ culture, which is also the case in vivo, if no stimulation occurs. Overall, we show that this novel system closely resembles the in vivo context. Importantly, porcine aortas can be collected from either veterinary surgeries or slaughterhouses, without having to sacrifice animals specifically for the purposes of this type of research.

A simple and efficient method for producing multitransgenic animals is required for medical and veterinary applications. Sperm-mediated gene transfer (SMGT) is an effective method for introducing multiple genes into pigs (Sus, Sus scrofa). The major benefits of this technique are the high efficiency, low cost, and ease of use compared with that of other methods: Sperm-mediated gene transfer does not require embryo handling or expensive equipment. The aim of this study was to investigate the influence of SMGT treatment and exogenous DNA uptake on sperm quality. Even after a coincubation with a 20-fold larger amount (100 microg/mL) of DNA than usual (5 microg/mL), sperm quality parameters were not significantly affected, confirming the hypothesis that the SMGT protocol itself or the amount of bound DNA do not compromise the possibility of an extended employment of SMGT. More importantly, we found that semen used for in vitro fertilization 24h after DNA uptake gave good cleavage (60% vs. 58%, treated vs. control) and developmental rates definitely positive (41% vs. 48%, treated vs. control). These good results are connected to a competitive efficiency of transformation (62%) due to the numerous improvements in SMGT technique. We demonstrate that SMGT-treated spermatozoa retain good quality and fertilization potential for at least 24h, expanding the possibility to apply transgenesis in field conditions in swine, where the greatest hurdles are fertilization timing and plain procedure.

Mental stress is a physiological condition that has a strong negative impact on the quality of life, affecting both the physical and the mental health. For such a reason, accurate measurements of stress level can be helpful to provide mechanisms for prevention and treatment. This paper proposes a procedure for the classification of different mental stress levels by using physiological signals provided by low invasive wearable devices. 17 healthy volunteers participated in this study. Three different mental states were elicited in them: a resting condition, a stressful cognitive state, and a sustained attention task. The acquired physiological signals were: a one lead electrocardiogram (ECG), a respiratory signal, a blood volume pulse (BVP), and 14 channels of a 10–20 electroencephalogram (EEG). For all subjects, 59 time series of 300 samples each were structured by including the RR series, the respiratory series, the pulse arrival time (PAT) series, and the delta, theta, alpha, beta power series of the 14 EEG channels. Different classifiers were implemented to assess the mental stress level starting from a pool of 3481 features computed from the aforementioned physiological quantities, using the Network Physiology paradigm. The highest achieved accuracy was 84.6%, from logistic regression and random forest classifiers, cross validated by mean of leave-one-person-out analysis. A further analysis was carried out to evaluate the classification accuracy using only cardio-respiratory signals, since the latter are more suitable to be used in real-life scenarios. In this case, the highest achieved accuracy was 76.5% obtained by the random forest classifier.

Flow cytometric separation of X and Y chromosome-bearing spermatozoa has been demonstrated to be effective in pigs, allowing the use of boar sexed semen in in vitro trials. Sperm Mediated Gene Transfer (SMGT) is a widely used and efficient technique for the creation of transgenic animals. The present research intended to prove that it is possible to associate sperm sexing with the SMGT technique in order to speed up the assessment of homozygous lines of transgenic pigs. In the first experiment, the sorting protocol was modified in order to obtain the highest DNA uptake by sorted spermatozoa. In the second experiment, spermatozoa that had undergone only sperm sorting, only SMGT, or both procedures (Sorted-SMGT) were used for in in vitro fertilization of in vitro matured oocytes. In the third experiment, transformed blastocysts of the desired gender (male) were obtained with Sorted-SMGT in an in vitro fertilization trial. The method we developed here allowed us to produce transgenic swine blastocysts of pre-determined gender, giving a positive answer at the aim to couple SMGT and sperm sorting in swine, obtaining fertile spermatozoa able to produce transgenic embryos of pre-determined gender.
