2. In vitro activity of sulopenem and comparator agents against Enterobacterales and anaerobic clinical isolates collected during the SENTRY Antimicrobial Surveillance Program

Objectives Physicians must leverage several factors when making antibiotic therapy decisions, including route of administration and duration of therapy. Oral administration provides several potential advantages including increased accessibility, prevention of hospitalizations and earlier discharges. Sulopenem—a broad-spectrum, synthetic penem β-lactam agent—uniquely possesses both oral and IV formulations along with noted stability among antimicrobial-resistant subsets. This study evaluated the in vitro activity of sulopenem and comparator agents against contemporary Enterobacterales and anaerobic clinical isolates predominantly from patients with bloodstream, intra-abdominal and urinary tract infections. Methods A contemporary collection of 1647 Enterobacterales and 559 anaerobic isolates was assembled from medical centres in Europe and the USA. Isolates were susceptibility tested using the CLSI reference methods: broth microdilution for Enterobacterales and agar dilution for anaerobes. Results Sulopenem demonstrated potent in vitro antimicrobial activity (MIC50/90, 0.03/0.25 mg/L) against Enterobacterales isolates regardless of infection type, inhibiting 99.2% of isolates at ≤1 mg/L. This activity was conserved against resistant phenotypes including ESBL-phenotype Escherichia coli (MIC50/90, 0.03/0.06 mg/L) and ESBL-phenotype Klebsiella pneumoniae (MIC50/90, 0.06/1 mg/L). Sulopenem maintained activity against ciprofloxacin-, nitrofurantoin- and trimethoprim/sulfamethoxazole-non-susceptible subsets (MIC50/90, 0.03–0.06/0.12–0.5 mg/L). Against anaerobic isolates, sulopenem (98.9% inhibited at ≤4 mg/L) and meropenem [98.4% susceptible (CLSI)] were the most active compounds tested. Conclusions The potent in vitro activity of sulopenem against this large collection of recent Enterobacterales and anaerobic clinical isolates from multiple infection types supports its further clinical evaluation in the treatment of intra-abdominal and urinary tract infections.

OBJECTIVE Timing of mobilization after chronic subdural hematoma (cSDH) surgery is highly heterogeneous among neurosurgical centers. Past studies have suggested that early mobilization may reduce medical complications without increasing recurrence, but evidence remains scarce. The purpose of this study was to compare an early mobilization protocol with a 48-hour bed rest practice, with a focus on the occurrence of medical complications. METHODS The GET-UP Trial is a prospective, randomized, unicentric, open-label study with an intention-to-treat primary analysis designed to evaluate the impact of an early mobilization protocol after burr hole craniostomy for cSDH on the occurrence of medical complications and functional outcomes. A total of 208 patients were recruited and randomly assigned to either an early mobilization group where they began head-of-bed elevation within the first 12 hours after surgery and proceeded to sedestation, orthostatism, and/or walking as rapidly as tolerated, or to a bed rest group where they remained recumbent with a head-of-bed angle inferior to 30° for 48 hours after surgery. The primary outcome was the occurrence of a medical complication (defined as either an infection, seizure, or thrombotic event) after surgery and until clinical discharge. Secondary outcomes included length of stay measured from randomization to clinical discharge, surgical hematoma recurrence at clinical discharge and 1 month after surgery, and Glasgow Outcome Scale–Extended (GOSE) assessment at clinical discharge and 1 month after surgery. RESULTS A total of 104 patients were randomly assigned to each group. No significant baseline clinical differences were observed before randomization. The primary outcome occurred in 36 (34.6%) patients included in the bed rest group and 20 (19.2%) in the early mobilization group (p = 0.012). At 1 month after surgery, a favorable functional outcome (defined as GOSE score ≥ 5) was observed in 75 (72.1%) patients in the bed rest group and 85 (81.7%) in the early mobilization group (p = 0.100). Surgical recurrence occurred in 5 (4.8%) patients in the bed rest group and 8 (7.7%) in the early mobilization group (p = 0.390). CONCLUSIONS The GET-UP Trial is the first randomized clinical trial to assess the impact of mobilization strategies on medical complications after burr hole craniostomy for cSDH. Early mobilization was associated with a reduction in medical complications without a significant effect on surgical recurrence, compared with a 48-hour bed rest protocol.

7. Antimicrobial activity of ceftibuten-avibactam against a global collection of Enterobacterales from patients with urinary tract infections (2021)

We evaluated the in vitro activity of ceftibuten-avibactam against Enterobacterales causing urinary tract infection (UTI). A total of 3216 isolates (1/patient) were consecutively collected from patients with UTI in 72 hospitals from 25 countries in 2021 then susceptibility tested by CLSI broth microdilution. Ceftibuten-susceptible breakpoints currently published by EUCAST (≤ 1 mg/L) and CLSI (≤ 8 mg/L) were applied to ceftibuten-avibactam for comparison. The most active agents were ceftibuten-avibactam (98.4%/99.6% inhibited at ≤ 1/ ≤ 8 mg/L), ceftazidime-avibactam (99.6% susceptible [S]), amikacin (99.1%S), and meropenem (98.2%S). Ceftibuten-avibactam (MIC50/90, 0.03/0.06 mg/L) was fourfold more potent than ceftazidime-avibactam (MIC50/90, 0.12/0.25 mg/L) based on MIC50/90 values. The most active oral agents were ceftibuten (89.3%S; 79.5% inhibited at ≤ 1 mg/L), levofloxacin (75.4%S), and trimethoprim-sulfamethoxazole (TMP-SMX; 73.4%S). Ceftibuten-avibactam inhibited 97.6% of isolates with an extended-spectrum β-lactamase phenotype, 92.1% of multidrug-resistant isolates, and 73.7% of carbapenem-resistant Enterobacterales (CRE) at ≤ 1 mg/L. The second most active oral agent against CRE was TMP-SMX (24.6%S). Ceftazidime-avibactam was active against 77.2% of CRE isolates. In conclusion, ceftibuten-avibactam was highly active against a large collection of contemporary Enterobacterales isolated from patients with UTI and exhibited a similar spectrum to ceftazidime-avibactam. Ceftibuten-avibactam may represent a valuable option for oral treatment of UTI caused by multidrug-resistant Enterobacterales.

A key to the six Australian genera of the wolf spider (Lycosidae Sundevall, 1833) subfamily Artoriinae Framenau, 2007 is provided, now including Artoria Thorell, 1877, Artoriopsis Framenau, 2007, Diahogna Roewer, 1960, Kangarosa Framenau, 2010, Kochosa gen. nov. and Tetralycosa Roewer, 1960. Kochosa gen. nov. is described to include 16 species: K. australia sp. nov. (type species; from New South Wales, Queensland, South Australia, Victoria, Western Australia), K. aero sp. nov. (Western Australia), K. asterix sp. nov. (New South Wales, Queensland, Tasmania and Victoria), K. confusa sp. nov. (Queensland), K. erratum sp. nov. (Queensland), K. fleurae sp. nov. (Victoria), K. mendum sp. nov. (Australian Capital Territory, New South Wales, Queensland), K. nigra sp. nov. (Queensland), K. obelix sp. nov. (Western Australia), K. queenslandica sp. nov. (Queensland), K. sharae sp. nov. (South Australia), K. tanakai sp. nov. (New South Wales, Queensland), K. tasmaniensis sp. nov. (Tasmania), K. timwintoni sp. nov. (Western Australia), K. tongiorgii sp. nov., (Queensland), and K. westralia sp. nov. (Western Australia). Kochosa gen. nov. differs distinctly from all other genera within the Artoriinae by somatic and genitalic morphology. Most conspicuous is a distinct off-white or yellowish-white cardiac mark on an otherwise generally uniformly dark abdomen. The cardiac mark is rendered by thick black setae, which are particularly dense posteriorly. The tegular apophysis of the male pedipalp is heavily reduced, generally forming a semi-transparent small lobe. In turn, the embolic division is often complex with a variety of apophyses. Kochosa gen. nov. generally inhabit mesic habitats such as temperate and tropical shrubs and forests along the eastern and south-eastern coast and in the south-western parts of Australia.

Introduction Acute kidney injury (AKI) has been identified as one of the most common and significant problems in hospitalized patients with COVID-19. However, studies examining the relationship between COVID-19 and AKI in low- and low-middle income countries (LLMIC) are lacking. Given that AKI is known to carry a higher mortality rate in these countries, it is important to understand differences in this population. Methods This prospective, observational study examines the AKI incidence and characteristics of 32,210 patients with COVID-19 from 49 countries across all income levels who were admitted to an intensive care unit during their hospital stay. Results Among patients with COVID-19 admitted to the intensive care unit (ICU), AKI incidence was highest in patients in LLMIC, followed by patients in upper-middle income countries (UMIC) and high-income countries (HIC) (53%, 38%, and 30%, respectively), whereas dialysis rates were lowest among patients with AKI from LLMIC and highest among those from HIC (27% vs. 45%). Patients with AKI in LLMIC had the largest proportion of community-acquired AKI (CA-AKI) and highest rate of in-hospital death (79% vs. 54% in HIC and 66% in UMIC). The association between AKI, being from LLMIC and in-hospital death persisted even after adjusting for disease severity. Conclusions AKI is a particularly devastating complication of COVID-19 among patients from poorer nations where the gaps in accessibility and quality of healthcare delivery have a major impact on patient outcomes.

A new monotypic genus in the orb-weaving spider family Araneidae Clerck, 1757 is described from Australia: Abbagen. nov., with Abba transversa (Rainbow, 1912) comb. nov. as the type species. It differs from all other genera in the family by somatic characters, specifically a patch of approximately five long spines on the prolateral surface of the first leg in males and an abdominal colouration with a pair of two central spots dorsally on a creamy-white surface. Specimens of A. transversacomb. nov. have been collected in Queensland and New South Wales, where the species is largely summer-mature. We also provide a genus level summary of all Australian Araneidae, currently consisting of 230 described species and eight subspecies in 46 genera.

O marketing e a comunicação online permitem às empresas comunicar e interagir com os seus clientes de forma eficaz e direcionada. Neste contexto, o presente estudo pretende determinar como e de que forma as Pequenas e Médias Empresas (PME’s) da Indústria Têxtil e Vestuário portuguesa, que operam no segmento B2B, usam as ferramentas de marketing digital. Pretende também “medir” a importância e eficácia atribuída pelas organizações a cada uma das ferramentas de marketing digital, os obstáculos que enfrentam na utilização destas ferramentas, os objetivos que pretendem atingir com a sua utilização, e que métricas utilizam para as avaliar. Este estudo utilizou uma metodologia quantitativa: foi desenvolvido e aplicado um questionário a uma amostra de empresas portuguesas que operam no setor ITV (Indústrias de Têxtil e Vestuário). Os principais resultados apontam para uma utilização relativamente frequente das redes sociais, do Search Marketing e do Website. Globalmente as empresas reconhecem a eficácia da maioria das ferramentas utilizadas. Os principais obstáculos à implementação destas ferramentas estão relacionados com a falta de recursos humanos especializados e com a falta de investimento nestas áreas.

Por tradição, em contexto de ensino e de aprendizagem da língua materna, tem-se verificado a hegemonia de uma perspectiva grafocêntrica da língua e da sua aprendizagem (SILVA et al. 2011, p. 7). A tomada de palavra, o falar, permite ao indivíduo dar opiniões, criticar, expor factos, discordar, defender-se, entre tantas outras possibilidades. É necessário que a abordagem do oral, em sala de aula, seja uma abordagem que projete o aluno para um perfil autônomo e com a capacidade de usar a língua em todas as situações. Assim, tendo consciência de que a oralidade constitui uma competência determinante para a integração e promoção social dos indivíduos, este artigo assume como objetivo orientador discutir o tratamento dado à oralidade no ensino de língua portuguesa como língua materna nos contextos brasileiro e português, a partir da retomada das documentações oficiais para o ensino obrigatório, de alguns que estejam ligados à elaboração de estratégias didáticas já desenvolvidos nos dois países, à análise de materiais didáticos ou às documentações oficiais para o ensino obrigatório. O foco coloca-se nas questões sobre o que ensinar e como ensinar. Para isso, utilizaremos como base a abordagem textual e discursiva por meio de autores que tratam da oralidade como objeto de estudo (MARCUSCHI, 2001; KOCH; ELIAS, 2006; 2009; AMOR, 2006; FIGUEIREDO, 2011) e metodologicamente utilizaremos uma perspectiva qualitativa bibliográfica, em que exploraremos as pesquisas sobre o tema a fim de efetuarmos um panorama do desenvolvimento deste domínio nos dois países.

18. Affinity of cefotiam for the alternative penicillin binding protein PBP3SAL used by Salmonella inside host eukaryotic cells

Background Following the invasion of eukaryotic cells, Salmonella enterica serovar Typhimurium replaces PBP2/PBP3, main targets of β-lactam antibiotics, with PBP2SAL/PBP3SAL, two homologue peptidoglycan synthases absent in Escherichia coli. PBP3SAL promotes pathogen cell division in acidic environments independently of PBP3 and shows low affinity for β-lactams that bind to PBP3 such as aztreonam, cefepime, cefotaxime, ceftazidime, ceftriaxone, cefuroxime and cefalotin. Objectives To find compounds with high affinity for PBP3SAL to control Salmonella intracellular infections. Methods An S. Typhimurium ΔPBP3 mutant that divides using PBP3SAL and its parental wild-type strain, were exposed to a library of 1520 approved drugs in acidified (pH 4.6) nutrient-rich LB medium. Changes in optical density associated with cell filamentation, a read-out of blockage in cell division, were monitored. Compounds causing filamentation in the ΔPBP3 mutant but not in wild-type strain—the latter strain expressing both PBP3 and PBP3SAL in LB pH 4.6—were selected for further study. The bactericidal effect due to PBP3SAL inhibition was evaluated in vitro using a bacterial infection model of cultured fibroblasts. Results The cephalosporin cefotiam exhibited higher affinity for PBP3SAL than for PBP3 in bacteria growing in acidified LB pH 4.6 medium. Cefotiam also proved to be effective against intracellular Salmonella in a PBP3SAL-dependent manner. Conversely, cefuroxime, which has higher affinity for PBP3, showed decreased effectiveness in killing intracellular Salmonella. Conclusions Antibiotics with affinity for PBP3SAL, like the cephalosporin cefotiam, have therapeutic value for treating Salmonella intracellular infections.

O presente trabalho trata-se de uma proposta de atividade que foi desenvolvida usando materiais de fácil aquisição, modelado com o software Geogebra com aplicação gráfica para resistores ôhmicos. O trabalho foi desenvolvido durante a disciplina Iniciação a Extensão do curso de Matemática da Universidade Federal do Acre (UFAC), durante o primeiro semestre do ano letivo de 2021. O trabalho poderá ser aplicado em aulas de Física e Matemática para alunos da educação básica. Além disso, o trabalho teve como objetivo mostrar a importância do uso das tecnologias digitais em sala de aula, da interação social e de inclusão dos alunos. A atividade proposta foi apresentada no término da disciplina, como proposta de interdisciplinaridade de ensinar Física e Matemática para alunos do ensino médio. A proposta de atividade foi ancorada na Teoria Significativa de Ausubel. Esse trabalho tem a intenção de divulgar e incentivar o ensino da lei de Ohm e de função linear numa perspectiva diferente das aulas usualmente lecionadas na educação básica.

Whole genome and transcriptome analysis of 213 Pseudomonas aeruginosa isolates resistant to antipseudomonal β-lactams collected in 30 countries was performed to evaluate resistance mechanisms against these agents.Isolates were susceptibility tested by reference broth microdilution. Whole genome and transcriptome sequencing were performed, and data were analysed using open-source tools. A statistical analysis of changes in the expression of5500 genes was compared to the expression of PAO1.The high-risk clones ST235 and ST111 were the most prevalent among90 sequence types (STs). Metallo-β-lactamase (MBLs) genes were detected in 40 isolates. AmpC and MexXY were the most common genes overexpressed in approximately 50% of the 173 isolates that did not carry MBLs. Isolates overexpressing pmrA and pmrB, the norspermidine production genes speD2 and speE2, and the operon arnBCADTEF-ugd were noted among strains resistant to ceftolozane-tazobactam and ceftazidime-avibactam, despite the lack of polymyxin resistance often associated to increased expression of these genes. Overexpression of MuxABC-OpmB, OprG, and OprE proteins were associated with resistance to ceftolozane-tazobactam in addition to the usual genes involved in cephalosporin, monobactam, and carbapenem resistance. Statistical analysis identified discrete mutations in ArmZ, OprD, and AmpC that correlated to antipseudomonal β-lactam resistance.P. aeruginosa resistance mechanisms are complex. This analysis suggests the role of multiple genes in resistance to antipseudomonal β-lactams, including some not commonly described.

A manufatura aditiva tem transformado os processos de fabricação e produção, sendo muito utilizado no ramo da indústria, medicina, odontologia, arquitetura e em desenvolvimento de produtos para quem deseja empreender. Esse processo tem como objetivo transformar um arquivo digital 3D em objeto real, sendo possível, em um mesmo equipamento imprimir uma peça mecânica de alta resistência, uma prótese medica ou até mesmo uma peça decorativa, depende da materia prima utilizada e seus parâmetros de impressão. A pesquisa aqui apresentada, tem como objetivo, explorar o uso de impressoras 3D no ramo de joalheria, com o objetivo de tornar-se uma aliada ao joalheiro, fazendo com que a produção seja mais rápida comparado com o método tradicional, mantendo a técnica da cera perdida mais eficiente, garantindo um produto padrão e possibilitando a produção em larga escala. Isso é devido que a impressora 3D tem a capacidade de imprimir várias jóias em poucos minutos, trabalho que uma pessoa manualmente demoraria horas. Desta maneira, foi realizado testes para identificar a impressão de um anel, o qual foi desenvolvido e modelado em 3D, posteriormente foi impresso em três diferentes tipos de tecnologias de impressoras 3D, como DLP, SLA e LCD. Em seguida as amostras de testes foram escaneadas e comparadas com o arquivo original onde, como resultados, apresentaram alta qualidade e ótima precisão, sendo possível seguir com o fluxo de trabalho para obter a joia final.

Coronavirus disease 2019 (COVID-19) is caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and has resulted in more than 6 million deaths worldwide. COVID-19 is a respiratory disease characterized by pulmonary dysfunction leading to acute respiratory distress syndrome (ARDs), as well as disseminated coagulation, and multi-organ dysfunction. Neutrophils and neutrophil extracellular traps (NETs) have been implicated in the pathogenesis of COVID-19. In this review, we highlight key gaps in knowledge, discuss the heterogeneity of neutrophils during the evolution of the disease, how they can contribute to COVID-19 pathogenesis, and potential therapeutic strategies that target neutrophil-mediated inflammatory responses.

25. Variáveis contemporâneas para o ensino integrado e formação integral

O objetivo deste trabalho consiste em identificar temas contemporâneos para o Ensino Integrado e Formação Integral no Curso Técnico em Agropecuária Integrado ao Ensino Médio do Instituto Federal Catarinense. Para tanto, realizamos uma análise documental, em que, partindo da ideia de Variáveis Contemporâneas e Equação Civilizatória, analisamos as diretrizes dos cursos técnicos do instituto e o projeto pedagógico do curso. Justificamos a escolha das Variáveis Contemporâneas ao utilizar como aporte teórico Bazzo (2016), Galeano (1999), Quijano (2000) e Grosfoguel (2008). Desta forma, constatou-se que educação ambiental, identidades de gênero e questões étnico-raciais, foram as principais variáveis identificadas e que é necessária a constante atualização dos temas contemporâneos nos documentos oficiais, além da reflexão do quanto, na prática, são abordados tais temas na formação dos estudantes.

Ceftolozane/tazobactam is an antipseudomonal cephalosporin combined with a well-established β-lactamase inhibitor.Methods: A total of 2038 Gram-negative organisms (376 P. aeruginosa and 1662 Enterobacterales) were collected consecutively using a prevalence-based approach from 11 medical centres. Organisms were susceptibility tested by broth microdilution according to CLSI guidelines. CLSI and EUCAST breakpoint criteria were used.Results: Ceftolozane/tazobactam was the most potent (MIC50/90, 0.5/4 mg/L) β-lactam agent tested against P. aeruginosa isolates, inhibiting 91.0% of the isolates at an MIC of ⤠4 mg/L. P. aeruginosa exhibited high rates of susceptibility to amikacin (92.0/92.0% [CLSI/EUCAST]) and colistin by EUCAST criteria only (99.2% intermediate [CLSI]/99.2% susceptible [EUCAST]). Ceftolozane/tazobactam (MIC50/90, 0.25/16 mg/L; 86.8/86.8% susceptible [CLSI/EUCAST]) and meropenem (MIC50/90, 0.03/0.12 mg/L; 93.0/93.3% susceptible [CLSI/EUCAST]) were the most active compounds tested against Enterobacterales. Isolates displayed susceptibility rates to other β-lactam agents, ranging from 81.5/77.7% for piperacillin/tazobactam, 66.0/64.5% for cefepime, and 65.3/60.9% for ceftazidime using CLSI/EUCAST breakpoints. Among the Enterobacterales isolates, 6.8% were carbapenem-resistant Enterobacterales (CRE) and 29.6% exhibited an extended-spectrum β-lactamase (ESBL) non-CRE phenotype. Ceftolozane/tazobactam showed good activity against ESBL non-CRE phenotype strains of Enterobacterales (MIC50/90, 0.5/8 mg/L; 84.8/84.8% susceptible), but not against isolates with a CRE phenotype (MIC50/90, > 32/> 32 mg/L).Conclusion: Ceftolozane/tazobactam was the most active β-lactam agent tested against P. aeruginosa and demonstrated higher in vitro activity than the available cephalosporins when tested against Enterobacterales from Asian countries.Keywords: Asia, ceftolozane/tazobactam, drug resistance, Enterobacterales, P. aeruginosa, surveillance

Adorno’s work, in particular the texts dealing with the relationship between music and social behaviors or structures, has been the target of criticism, especially in the second half of the 20th century, being considered by many to be generalist, dogmatic or even elitist. This work proposes the analysis of musical technologies not only as a set of compositional techniques, as Adorno does, but, in fact, as material conditions for the realization of a certain type of sound/music. The colonialist character of these technologies is also analyzed. Based on a review of some key concepts in Adornian theory, a dialogue is sought with more contemporary authors from the sociology of music.

28. Presenilin and APP Regulate Synaptic Kainate Receptors

Kainate receptors (KARs) form a family of ionotropic glutamate receptors that regulate the activity of neuronal networks by both presynaptic and postsynaptic mechanisms. Their implication in pathologies is well documented for epilepsy. The higher prevalence of epileptic symptoms in Alzheimer's disease (AD) patients questions the role of KARs in AD. Here we investigated whether the synaptic expression and function of KARs was impaired in mouse models of AD. We addressed this question by immunostaining and electrophysiology at synapses between mossy fibers and CA3 pyramidal cells, in which KARs are abundant and play a prominent physiological role. We observed a decrease of the immunostaining for GluK2 in the stratum lucidum in CA3, and of the amplitude and decay time of synaptic currents mediated by GluK2-containing KARs in an amyloid mouse model (APP/PS1) of AD. Interestingly, a similar phenotype was observed in CA3 pyramidal cells in male and female mice with a genetic deletion of either presenilin or APP/APLP2 as well as in organotypic cultures treated with γ-secretase inhibitors. Finally, the GluK2 protein interacts with full-length and C-terminal fragments of APP. Overall, our data suggest that APP stabilizes KARs at synapses, possibly through a transsynaptic mechanism, and this interaction is under the control the γ-secretase proteolytic activity of presenilin.SIGNIFICANCE STATEMENTSynaptic impairment correlates strongly with cognitive deficits in Alzheimer's disease (AD). In this context, many studies have addressed the dysregulation of AMPA and NMDA ionotropic glutamate receptors. Kainate receptors (KARs), which form the third family of iGluRs, represent an underestimated actor in the regulation of neuronal circuits and have not yet been examined in the context of AD. Here we provide evidence that synaptic KARs are markedly impaired in a mouse model of AD. Additional experiments indicate that the γ-secretase activity of presenilin acting on the amyloid precursor protein controls synaptic expression of KAR. This study clearly indicates that KARs should be taken into consideration whenever addressing synaptic dysfunction and related cognitive deficits in the context of AD.

The immunosuppressive effect of methotrexate has rarely been associated with reactivation of cutaneous leishmaniasis. Here we present a case of a cutaneous leishmaniasis patient with atypical clinical symptoms without splenomegaly but with cutaneous manifestations after treatment of rheumatoid arthritis with methotrexate and blood recovery of the parasite. Next-generation sequencing was used to identify Leishmania infantum chagasi in the patient’s blood sample.

Este estudo tem como objetivo discutir a possibilidade de interface entre a Linguística de Texto e o Funcionalismo norte-americano por meio da retomada dos seus pressupostos teóricos basilares, da elucidação dos seus pontos de contato e da ilustração das suas possibilidades de encaminhamento teórico-analítico. Para isso, são exploradas as bases das duas vertentes (cf. GIVÓN, 1995; BUTLER, 2003; BERNÁRDEZ, 2003; VAN DIJK, 1997; 2006; 2015; AUTOR, 2020b) e são apresentadas pesquisas de Santos (2003) e de Autor (2017; 2020), Autor (2020a) e Autor (2021). Diante de tais elucidações, há a defesa de uma agenda de pesquisas a partir do diálogo entre a Linguística de Texto e o Funcionalismo norte-americano.

31. Percepción de los padres acerca de la comunicación sobre sexualidad de sus hijos con trastorno del espectro autista

Objetivo: Explorar las percepciones de los padres en relación con la comunicación sobre sexualidad hacia sus hijos que se encuentran dentro del espectro autista. Método: Estudio cualitativo descriptivo y exploratorio, que fue desarrollado en tres centros de atención a niños y adolescentes autistas en los estados de Baja California y Sinaloa, México. Participaron cuatro madres y un padre de niños/jóvenes con Trastorno del Espectro Autista y para la recogida de datos fueron utilizadas entrevistas semiestructuradas. Los datos fueron analizados por medio de la técnica de análisis de contenido. Resultados: Fueron identificados dos temas: (1) Barreras para la comunicación sobre sexualidad, con cuatros subtemas (Inadecuada identificación de temas de sexualidad, baja autoeficacia para la comunicación sobre sexualidad, falta de profesionales capacitados y falta de herramientas o materiales adecuados para la comunicación sobre sexualidad); (2) Miedos y preocupaciones por la falta de comunicación sobre sexualidad. Conclusiones: Los padres reconocen los aspectos importantes que influyen en el tipo y la forma en que se abordan los temas sexuales, reduciéndose a la higiene personal y la masturbación, dejando de lado aquellos que implican mayor sensibilidad como el abuso sexual. Asimismo, los padres solo tocan los temas necesarios, conforme a la etapa de desarrollo del hijo.

Este trabalho discute as experiências acerca da organização e desenvolvimento de um congresso no formato remoto, promovido pela área de física da Universidade Federal do Acre (UFAC) durante o período da pandemia da doença por coronavírus 2019 (COVID-19). Discutimos como a partir do isolamento social imposto pela pandemia, a utilização de Tecnologias da Informação e Comunicação (TICs) permitiram a realização de um congresso inteiramente remoto, com palestras, minicursos, apresentações de trabalhos pelos alunos e discussões sobre os mais variados temas da física. Embora o congresso tenha como face a física, o presente trabalho é uma radiografia sobre aspectos sociais, inclusivos, metodológico-educacionais e de interação entre instituições escolares e universitárias. Analisamos a partir dos resultados obtidos, o impacto, a abrangência e a relevância que um congresso em modalidade remota pode promover aos estudantes, em particular no estado do Acre.

Este artigo apresenta um estudo acerca dos usos de anáforas encapsuladoras na construção do gênero entrevista por meio de seus aspectos estilísticos e composicionais. Os encapsulamentos anafóricos são definidos como um recurso coesivo expresso por Sintagmas Nominais que funcionam como uma estratégia resumitiva de informações de tamanhos variados. A partir de uma amostra formada por 24 entrevistas publicadas nos anos de 2018 e 2019 nas revistas Exame e Veja, foram coletados e analisados os dados. Partiu-se dos postulados de Bakhtin (1997[1979], que defende que os gêneros podem ser caracterizados pelos planos temático, estilístico e composicional. O artigo relaciona tais planos a partir da construção do estilo e do conteúdo composicional por meio dos usos dos encapsulamentos anafóricos e das diferentes partes das entrevistas. Foram muito importantes para a pesquisa os trabalhos sobre gêneros textuais (MARCUSCHI, 2005; 2008; BEZERRA, 2017), e os estudos sobre referenciação (FRANCIS, 1994; CONTE, 1996; BORREGUERO, 2006; PECORARI, 2014; BASTOS, 2018). Dentre as principais contribuições da pesquisa, estão a caracterização de como se constrói o gênero entrevista com ênfase no caráter fórico e na multifuncionalidade dos encapsuladores.

36. Biases of the Barotropic Atmospheric Circulation Variability in CMIP6 Models

Two ensembles of Atmospheric Model Intercomparison Project (AMIP) simulations, in the scope of the current Coupled Model Intercomparison Project (CMIP6), are compared with their fully coupled counterparts. The atmospheric models simulate less barotropic atmospheric circulation variability over the North Atlantic and more barotropic atmospheric circulation variability over the North Pacific when compared with reanalysis variability, at intraseasonal and interannual scales. The coupled climate simulations have smaller global barotropic variability than the corresponding AMIP simulations. The smaller variability of the coupled simulations results in no mean overestimation of the subtropical jet variability in the North Pacific, but further underestimation of the jet stream variability in the North Atlantic. The results suggest that the reduction of the biases, in the North Pacific barotropic atmospheric variability, of coupled climate simulations is achieved through compensating biases in the mean sea surface temperatures (SSTs). Moreover, the reduction of the positive biases in the North Pacific seems to be associated with a reduction of the excitation of the most unstable barotropic mode of the atmospheric circulation, which contributes also to a reduction of the barotropic atmospheric variability in the North Atlantic.

Bivalve molluscs (BM) are filter animals and bioaccumulators of substances from the environment. This characteristic allows a great absorption of nutrients that makes them a source of protein-rich foods. On the other hand, if there are toxic contaminants in the environment, their absorption by animals may occur. This happens with pesticides coming from agricultural and livestock production systems that can migrate to areas of BM crops. Considering the high nutritional value of bivalve molluscs and their positive impact on the human diet, these products must be carefully evaluated for the possible presence of toxic substances in order to guarantee their safety. Thus, the aim of this work was to implement and validate a multi-residue method using tandem mass spectrometry to evaluate pesticide residues commonly used in agricultural production systems present in these matrices. Extraction and cleaning steps were optimized and the method proved to be adequate to quantify 322 pesticides. The samples come from five different areas of culture of bivalve molluscs in the southeast, north and northeast regions of Brazil. The analysis of the mollusc samples showed the presence at the trace level of seven different pesticide residues in four of the five evaluated samples.

38. Bovine Lactoferrin Loaded Magnetic-Responsive Plasmonic Lipo-Somes for Therapeutic Applications

Bovine lactoferrin (bLf) is a milk-derived protein that exhibits multiple biological activities and has been explored towards different therapeutic applications. Since this molecule is susceptible to degradation and some of its properties depend on its tertiary structure, the encapsulation of bLf in stimuli-responsive therapeutic formulations provides an added value to potentiate its biological activities when administered. Plasmonic magnetoliposomes emerge as promising nanosystems for dual hyperthermia (magneto-photothermia) and local therapy, since the combination of magnetic and gold nanoparticles (NPs) in a single nanosystem (multifunctional liposomes) enables the targeting and controlled release of encapsulated drugs. In this work, plasmonic magnetoliposomes (PMLs) containing manganese ferrite nanoparticles (28 nm size) and gold nanoparticles (~ 5 nm size) functionalized with 11-mercaptoundecanoic acid or octadecanethiol, were synthesized and loaded with bLf. The structural, magnetic and optical properties of the nanoparticles were measured by TEM, SQUID and UV/vis/NIR absorption spectroscopy. Specific Absorption Rate was determined to assess the capabilities for magnetic and photothermal hyperthermia. Finally, Saccharomyces cerevisiae was used as an eukaryotic cellular model to assess the biological activity and the mechanism of entry of bLf-loaded PMLs, through counting of colony forming units and fluorescence microscopy, respectively. The results demonstrate that PMLs are mainly internalized through an energy- and temperature-dependent endocytic process, though the contribution of a diffusion component cannot be discarded. Most notably, only bLf-loaded PMLs exhibit cytotoxicity with an efficiency similar to free bLf, attesting their promising potential for bLf delivery in the context of therapeutic interventions.

40. Retinoblastoma protein activity revealed by CRISPRi study of divergent Rbf1 and Rbf2 paralogs

Retinoblastoma tumor suppressor proteins are highly conserved transcriptional corepressors, regulating the key transition from G1 to S phase of the cell cycle. The mammalian Rb family is composed of Rb, p107, and p130, which possess both overlapping and unique roles in gene regulation. Likewise, the Drosophila lineage experienced a gene duplication event, leading to the expression of the Rbf1 and Rbf2 paralogs. To uncover the significance of the multiplicity of the Rb family, and how gene regulatory roles have been apportioned between the family members, we made use of the CRISPRi system. We engineered dCas9 fusions to Rbf1 and Rbf2, and deployed them to gene promoters in developing Drosophila tissue to study their relative impact on target promoters. On some genes, both Rbf1 and Rbf2 are able to mediate potent repression, and this happens in a distance-dependent manner. In general, a greater repression effect is noted for promoter-proximal sites, suggesting that Rb proteins have short-range effects, and may directly impact function of the basal transcriptional machinery. Depending on context, Rbf1 or Rbf2 was more potent, suggesting that the paralogs possess non-identical functions, possibly due to primary sequence divergence. Notably, with this dCas9 tethering system, an Rbf1 mutant lacking the entire pocket domain, that has been generally assumed to be necessary for function, retained repression activity. Significantly, direct comparison of Rb activity on endogenous genes and transiently transfected reporters showed that only qualitative, but not key quantitative aspects of repression were conserved, indicating that the native chromatin environment is essential for understanding the context-specific effects of Rb activity. The data presented here points to the complexity of Rb-mediated transcriptional regulation in a living organism, undoubtedly impacted by the different promoter landscapes and the evolution of the Rb proteins themselves.

42. How can the most unstable barotropic mode of atmospheric models contribute for the explanation of atmospheric variability biases of climate models in the North Atlantic?

In this communication, we will present results from an analysis of the variability of the vertically averaged (i.e., barotropic) atmospheric circulation simulated by climate models, which integrate the current Coupled Model Intercomparison Project (CMIP6). The variabilities in two ensembles of Atmospheric Model Intercomparison Project (AMIP) simulations were compared with the variabilities in two ensembles of fully coupled simulation counterparts of the current CMIP6 (Castanheira and Marques, 2022).The atmospheric models simulate less variability of the barotropic atmospheric circulation over the Northern Atlantic and more variability over the North Pacific when compared with the corresponding variabilities in the ERA5 reanalysis (“observations”), at intraseasonal and interannual scales. When integrated over the whole globe, the variability in the coupled climate simulations is smaller than the variability in the corresponding AMIP simulations. The diagnosis of chronic illness in childhood implies frequent hospitalizations and, consequently, the interruption of school attendance. This study aimed to understand the process of school reintegration of children and adolescents with chronic illness from the mothers' perspective.A qualitative descriptive-exploratory study was conducted with mothers who experienced the process of school reintegration of their child or adolescent, aged between 8 and 17 years old, and diagnosed with chronic illness. The participants were recruited by convenience and interviewed at the paediatric unit of a children's hospital. Data collection was interrupted when the data set was sufficient to answer the research question. The interviews were analysed using inductive thematic analysis. The study was approved by the research ethics committee.Eleven interviews were conducted, 10 with mothers and one with a grandmother, who played the maternal role. Participants' age ranged between 33 and 58 years old. A theme was developed-"School reintegration under the maternal vigilance"-which encompasses four subthemes: (1) What matters? My child's health comes first; (2) How to keep in touch with the school? (3) Back to the school: Are we ready? (4) Sharing vigilance: reality and expectations. The themes highlighted a cyclical, dynamic, and subjective school reintegration process, constantly permeated by maternal vigilance.A new understanding about school reintegration was evidenced, from the perspective of mothers of children and adolescents with different chronic illnesses. Mothers and children experience a nonlinear and recurrent process of leaving and returning to school, surrounded by a lack of communication and continuity in school activities. The results of this study may assist health professionals in planning care focused on the needs of the school reintegration of this population.

O carcinoma de células escamosas é responsável por 95% das lesões malignas orais e possui como origem o epitélio de revestimento bucal, tendo como principais fatores de risco: tabagismo, alcoolismo, exposição excessiva ao sol sem proteção, desnutrição, entre outros. Para prevenir seu aparecimento, é necessário evitar hábitos nocivos, além disso, o autoexame é extremamente relevante para o diagnóstico precoce da lesão, possibilitando um tratamento imediato e consequentemente um prognóstico mais favorável. Esse trabalho tem como objetivo relatar um caso clínico vivenciado na Clínica- Escola do Unifeso, de uma lesão que de acordo com o laudo histopatológico foi diagnosticado uma forma rara e agressiva de câncer, o Carcinoma de Células Escamosas do tipo basalóide, que ocorre com maior frequência no trato aerodigestivo superior. Essa lesão é um subtipo do carcinoma de células escamosas e seu prognóstico depende do estadiamento da lesão e condições clínicas do paciente no momento do diagnóstico, tendo um tempo médio de 23 meses de sobrevida. Com isso, o tratamento de escolha foi o cirúrgico, seguido de radioterapia e radioquimioterapia.

Body image is a multidimensional concept that involves the mental image of the human body and the feeling of being oneself throughout existence. Treatment for breast cancer causes several bodily changes that affect women's body image.This meta-synthesis aims to synthesise and interpret primary qualitative studies on the experience of body image in women undergoing treatment for breast cancer.A qualitative meta-synthesis was conducted employing systematic searches in six databases (PubMed, CINAHL, SCOPUS, Web of Science, PsycINFO, and LILACS). Data analysis was performed according to thematic synthesis.Forty studies were included. Five descriptive themes were identified: (1) actively saying goodbye to body image; (2) the rupture of body image; (3) the sacrifice of body image in exchange for life; (4) body image as a vehicle for social expression; and (5) resignation of the alterated body image. These themes were understood through one analytical theme: Half-woman: body image of the woman with breast cancer.The experience of body image in the context of breast cancer is a dynamic phenomenon, which involves dismissal, rupture, and resignation and occurs mediated by interpersonal contact.

Copyright © 2022 The Author(s). An experimental and numerical study into the behaviour of concrete-filled double skin tubular (CFDST) stub columns is presented. A total of eight axial compression tests were carried out, four utilising conventional concrete and four with recycled aggregate concrete. The stub columns were circular in cross-section and each comprised an austenitic stainless steel outer tube and a carbon steel inner tube, of varying dimensions. Accordingly, hollow ratios of 0.67 and 0.55 were considered. The recycled coarse aggregate was made by crushing test specimens from a previous research project, and a replacement ratio of 50% was adopted. During the experiments, similar structural behaviour and failure modes were observed between the specimens with conventional and recycled aggregate concrete. To investigate the behaviour further, a finite element model was developed in ABAQUS; validation of the model against the experimental results from the current work as well as data available in the literature is described. The finite element model was employed to conduct a parametric study to examine the load-bearing contributions of the constituent components of CFDST sections and to assess the influence of the hollow ratio on the structural behaviour. The experimental and numerical ultimate loads are compared with the capacity predictions determined using available design procedures. Overall, the results show that CFDST stub columns with recycled aggregate concrete can achieve similar capacities to their conventional concrete counterparts, demonstrating the potential for the wider use of recycled aggregate concrete, towards more sustainable structural solutions. Lower water availability due to climate change has dramatically affected coffee yields and quality. Characteristics related to plants and soil can be explored to obtain more productive and sustainable coffee production under adverse crop conditions. To this end, the objective of this study was to evaluate the potential for using different agronomic practices to optimize water use in coffee plants and to mitigate the effects of lower water availability to the crop. In the plots, three soil management systems (polyethylene film, brachiaria and bare soil) were used. In the subplots, two types of fertilizers were used (conventional and controlled-release fertilizer). Five soil conditioners (coffee husk, gypsum, water-retaining polymer, organic compost, and the control) were distributed throughout the sub-subplots. The plant growth, soil moisture, leaf physiology and anatomy, and soil chemical properties of coffee plants were analysed. The management of the soil cover using polyethylene film or brachiaria stood out as agronomic practices that can be combined with coffee husk applications to enable the better use of water by coffee plants and to favour soil conditioning. Controlled-release fertilizer and gypsum allowed for improvements in the anatomical and physiological characteristics of the plants. The use of organic compost favoured greater water availability; however, it caused losses in terms of coffee growth and physiological parameters due to nutritional imbalance. Therefore, based on results, the use of polyethylene film, brachiaria, controlled-release fertilizer, coffee husks, and gypsum are potential agronomic practices for optimizing water use and mitigating the effects of water deficit in coffee plants

Este artigo apresenta resultados de uma pesquisa que analisou as práticas de escrita no uso do computador e da internet em um telecentro de uma comunidade quilombola situada ao norte de Minas Gerais/Brasil. O objetivo foi investigar como os jovens da comunidade quilombola integraram Tecnologias Digitais de Informação e Comunicação ao seu cotidiano, à medida que frequentavam o curso de inclusão digital no telecentro. Adotou-se uma perspectiva interpretativa de investigação, a partir de uma abordagem etnográfica. Os dados mostraram que, embora o curso de informática possa conter em sua programação um currículo prescrito para a inclusão do aluno no ambiente digital, essa inclusão pode estar mais relacionada à forma como os usuários se organizam e se envolvem em atividades usando computadores e a internet em suas práticas cotidianas ligadas às identidades culturais particulares.
