- Abstract
Purpose: To evaluate the effect of therapeutic resources on the biomechanical characteristics of normal and grafted skin after a burn. Methods: Forty-four participants, both male and female with an average age of 35.89 (±11.53) years, with second- and third-degree burns of both normal skin and scars (skin sheet graft) were evaluated. Participants were randomly chosen, with a crossover design, and a washout of 7 days after each intervention. Biomechanical parameters of the skin, such as distensibility and viscoelasticity, were assessed by Cutometer® before and after 0, 10, 20, and 30 minutes of intervention. This included therapeutic ultrasound and paraffin, with or without vacuum massage. Results: All groups evaluated in this study demonstrated a profile of increased distensibility of their skin in the period immediately after the application of the resources. However, there was a progressive reduction of the effects in the consecutive times tested, most likely due to the probable "therapeutic window”. Participants with scars, showed a decrease in viscoelasticity in all groups except those treated with paraffin. Conclusion: The findings of this study indicate that the post-burn scars after a skin graft present different biomechanical characteristics compared to normal skin, before and after stimulation with various therapeutic interventions.

- Abstract

People with SCI have compromised their muscle recruitment and are more predisposed to secondary problems. In order to recover or enhance the lower limbs functions, rehabilitation programs are the most used treatment and, more recently, in combination with robots. The use of robots in SCI rehabilitation brings benefits for both the patient and the therapist since it can provide more control, accuracy, and variety of training modes, providing new outcomes to rehabilitation. Electromyography (EMG) activity is often used to trigger orthosis, prosthesis and exoskeletons in robotic rehabilitation. EMG can be recorded using either surface or intramuscular (invasive) detection systems. The use of intramuscular electrodes allows the detection of electric potentials closer to the muscle fibres and the register of EMG activity from deeper muscles. The high selectivity contrariwise, impose also some limitations as the activity reflection of only a small number of active motor units whose fibres are close to the position of the detection site. The European project EXTEND, where this master thesis is inset has, as of purpose, to develop neural interfaced assistive wearable robots for spinal cord injury. In this context, it was performed the analysis of the viability on the use of intramuscular recordings and the characterisation of signal quality and robustness against mechanical interference. Intramuscular electrodes showed a flaw compared to the superficial due to their maintenance in its initial local position, leading to loss of the muscle signal during the task. However, in compensation, they showed less interference from external means (like the exoskeleton) showing cleaner signals of mechanical noise., La lesión de la médula espinal (LME) afecta cada vez a más personas en todo el mundo.

This study aimed to evaluate the antiparasitic and growth potentials, weight gain and hematologic response of goats feed with the supplement of Cucurbita moschata seeds, Solanum tuberosum barks and Musa spp. leaves and citrus pulp. The trials were carried out at the States of Sergipe (10°57'52" S, 37°03'42" W) and São Paulo (22° 57'26" S, 47°13'06" W). Goats used in this research were males (SRD) with natural infection, 4?6 months old, average weight of 19.79 ± 5.53Kg and feed with 1kg/animal of Guabi¿ animal food. Distribution of the groups in Sergipe 30 goats: GV/SE ¿ goats treated with Ivermectina¿ - 1ml/25kg; GS/SE ¿ goats fed with 0.8Kg/animal of the commercial animal food + 100g of C. moschata seed flour (FSA) + 100g of S. tuberosum seed flour (FCB); GC/SE ¿ Control, the experiment lasted 60 days. In São Paulo: 24 goats: GV/SP ¿ goats treated with Ripercol¿ - 2mL/20Kg and Neguvon¿ 1mL/20Kg; GM/SP ¿ feed with 0.740Kg of the commercial animal food + 130g of Musa sp. leaves and 130g citrus pulp; GC/SP ¿ Control, the experiment lasted 90 days. Parameters analyzed fortnightly were counting of eggs/oocysts per gram of feces (OPG/OoPG), clinical individual evaluation (Famacha¿), morphometric analysis, weighing, and 3 blood collections (at days 0, 37 and 90). At the end of the experiment, the goats were euthanized, the cleaned carcasses were weighed, the gastrointestinal tract were taken to the lab (LDIP/Unit ¿ SE and L3/Unicamp ¿ SP) for recovering the parasites and part of the muscle was removed to evaluation of the centesimal composition. All co-products used were examined regarding phytochemical composition and antioxidant activity. The results were analyzed using an analysis of variance (ANOVA) and Pearson¿s correlation coefficient (? ? 0.05). In Sergipe it was found species of Eimeria sp. (603.4 OoPG) and OPG general reduction of 70.73% in the GS and of 83.82% in GV at the end of the experiment without statistical difference (p = 0.074). Groups of nematodes of Trichostrongyloidea superfamily, Strongyloides papillosus and Trichuris sp. presented reduction of 56.42, 86.22 and 97.60% in GV group, and of 70.47, 61.82 and 98.22% in GS group. GS group had the highest weight gain (34.86%), body length measurements (57.10 ± 7.27cm) and height of the withers (58.10 ± 3.81cm), with mean positive correlation of 0.65 for live final weight and r = 0.82 for warm carcass. GV and GS groups presented similar centesimal composition. In extracts from S. tuberosum bark, the antioxidant activity was of IC50 = 0.116mg/mL and vitamin E and linoleic acid were identified from the C. moschata seeds. In São Paulo it was found Eimeria sp. (807.6 ± 78.96 OoPG) and effectiveness in GM and GV groups was of 69.05% and 45.24%, respectively, at the end of the experiment with statistical difference (p = 0.021). Hematologic evaluations revealed discreet normochromic macrocytic anemia in GV group at time 0. Weight gain in GM group was 47.59% higher than the other groups (p = 0.001). Helminthes recovered belonged to the families Strongyloidae, Oxiuridae, Trichostrongylidae, Trichuridae and Anaplocephalidae. By the phytochemical analysis of hydroethanolic extracts from Musa sp. it was identified rutin, coumarin and E vitamin, and in the citric pulp, caffeic acid, coumarin and hesperidin. These identified compounds may have contribute to OPG reduction, to enhance productiveness (growth), meat quality (GS/SE), and induction and continuity of the leukocytarian response against the helminthic infections (GM/SP). In this way, the aforementioned co-products are an alternative for the decrease of helminthic infections, boosting economic benefits to goat culture Doutorado Relações Antrópicas, Meio Ambiente e Parasitologia Doutora em Biologia Animal

fungi - Abstract
Endophytic fungi represent ubiquitous microbial organisms able to live in the tissues of different plants around the world and represent a prolific source of bioactive metabolites. In the present study, the endophytic fungus Aspergillus calidoustus was isolated from the medicinal plant Acanthospermum australe (Asteraceae), and identified using molecular, physiological and morphological methods. A methylene chloride crude extract of A. calidoustus has been produced and subjected to antifungal bioassay-directed fractionation which resulted in the isolation of the two bioactive compounds: ophiobolin K and 6-epi-ophiobolin K. These pure compounds displayed antifungal activity against fungal plant pathogens, protozoal activity against Trypanosoma cruzi, and cytotoxic activity against human tumoral cell lines. The results show that A. calidoustus was able to produce the antifungal and cytotoxic metabolites ophiobolin K and 6-epi-ophiobolin K, which may help the fungus to colonise and occupy the substratum as well as survive in natural environments.

Depression in children and adolescents, as well as in adults, is one of the most common mental disorders. Since its appearance in these age groups leads to serious consequences in adulthood, it is critical to identify depressive symptoms in early stages. Therefore, self-report instruments play a key role in this contexto since they allow to easily, reliably and validly have access to ways the subject thinks, feels and acts. The aim of this study was to translate, adapt and study the Center for Epidemiological Studies Depression Scale for Children (CES-DC) (Weissman, Orvash & Padian, 1980) for portuguese adolescents. The sample consists of 417 adolescents, aged between 12 and 18 years old, attending the 3rd cycle of basic education and secondary education schools. To study the convergent and divergent validity of the CES-DC, the Depression Anxiety Stress Scales (DASS 21) (Lovibond & Lovibond, 1995) was used, along with the Children's depression Inventory (CDI) (Sailor, 1994) and the Students' Life Satisfaction Scale (SLSS) (Marques, Pais-Ribeiro & Lopez, 2007) that evaluate, respectively, the negative emotional states (depression, anxiety and stress), depressive symptoms and overall satisfaction with life. The results show that the evaluated scale presents a good internal consistency, an adequate temporal stability as well as good validity. They also suggest that this is a three-dimensional scale: humor factor, interpersonal factor and happiness factor. These data allow for the progress of psychology, regarding the assessment and clinical practice among adolescents. The usefulness of the questionnaire should be verified in other age groups, especially in children with less than 12 years old and in a clinical sample. Despite these limitations, the results suggest that CES-DC is useful in the assessment questionnaire of depressive symptoms in adolescents.

The bromeliad Nidularium minutum Mez is a cold tolerant species from the Atlantic Rainforest, which grows in the subtropical latitude, where temperatures range from 2 to 30 degrees C. We hypothesized that N. minutum plants would implement rapid metabolic adjustments to ensure survival during sudden cold events that occur in seasons other than winter. N. minutum plants were cultivated at 10 degrees C. Under this condition, there was an observed increase in soluble sugar content and nitric oxide (NO) emission, as well as a high nitrate reductase (NR) activity observed within the 72 h of cold exposure, when compared with plants maintained at 25 degrees C. These responses coincided with a decrease in antioxidant activity and an increase in lipid peroxidation (LPO). After 72 h of cold exposure, a subset of plants were de-acclimated at 25 degrees C. These plants displayed reduced soluble sugar concentrations, NO emissions, NR activity and LPO. Additionally, there was an increase in anti-oxidant activity, which indicated that the plants were recovering from cold stress. Furthermore, it was found that plants maintained at 10 degrees C for 168 h acclimated to the cold by reducing the NO content and maintaining the increased sugar concentrations, thus resulting in a less intense stress response. We conclude that rapid changes in NO content, sugar concentrations and antioxidant activity are metabolic adjustments that occur during the acclimation and de-acclimation processes of this bromeliad to low temperatures. During abrupt thermal changes in subtropical regions, such adjustments could enhance cold tolerance and increase the chances of survival for this plant.
