1. Single-stage post-compression of an Ytterbium fiber laser down to 20 fs

Abstract

We demonstrate single-stage post-compression of an Ytterbium fiber laser to about 20 fs based on spectral broadening in a gas-filled multipass cell. A compression factor of seven has been achieved with a throughput of 86%.

Abstract

Let n be either 2, or an odd integer greater than 1, and fix a prime p > 2(n + 1). Under standard "adequate image" assumptions, we show that the set of components of n-dimensional p-adic potentially semistable local Galois deformation rings that are seen by potentially automorphic compatible systems of polarizable Galois representations over some CM field is independent of the particular global situation. We also (under the same assumption on n) improve on the main potential automorphy result of [BLGGT14b], replacing "potentially diagonalizable" by "potentially globally realizable".

Abstract

Structural integrity and cellular homeostasis of the embryonic stem cell niche are critical for normal tissue development. In the telencephalic neuroepithelium, this is controlled in part by cell adhesion molecules and regulators of progenitor cell lineage, but the specific orchestration of these processes remains unknown. Here, we studied the role of microRNAs in the embryonic telencephalon as key regulators of gene expression. By using the early recombinerRx-Cremouse, we identify novel and critical roles of miRNAs in early brain development, demonstrating they are essential to preserve the cellular homeostasis and structural integrity of the telencephalic neuroepithelium. We show thatRx-Cre;Dicer(F/F)mouse embryos have a severe disruption of the telencephalic apical junction belt, followed by invagination of the ventricular surface and formation of hyperproliferative rosettes. Transcriptome analyses and functional experimentsin vivoshow that these defects result from upregulation ofIrs2upon loss oflet-7miRNAs in an apoptosis-independent manner. Our results reveal an unprecedented relevance of miRNAs in early forebrain development, with potential mechanistic implications in pediatric brain cancer.

Abstract

Summary: circSLC45A4 is the main RNA splice isoform produced from its genetic locus and one of the highest expressed circRNAs in the developing human frontal cortex. Knockdown of this highly conserved circRNA in a human neuroblastoma cell line is sufficient to induce spontaneous neuronal differentiation, measurable by increased expression of neuronal marker genes. Depletion of circSlc45a4 in the developing mouse cortex causes a significant reduction of the basal progenitor pool and increases the expression of neurogenic regulators. Furthermore, knockdown of circSlc45a4a induces a significant depletion of cells in the cortical plate. In addition, deconvolution of the bulk RNA-seq data with the help of single-cell RNA-seq data validates the depletion of basal progenitors and reveals an increase in Cajal-Retzius cells. In summary, we present a detailed study of a highly conserved circular RNA that is necessary to maintain the pool of neural progenitors in vitro and in vivo. : Suenkel et al. present a detailed study of the highly conserved circular RNA (circRNA) circSLC45A4 in human neuroblastoma cells and mouse embryonic cortex. She received her Graduate degree in chemistry in 2001 and her Ph.D. degree in 2006, both from the Universidad Autonomá de Madrid, Spain. She completed her education with several stays in the UniversitéBordeaux I, and a long-term Postdoctoral fellowship (2006?2009) at the Lawrence Berkeley National Lab, Berkeley, CA. She now has a Tenure Track position (Ramo?y Cajal) in Madrid. She received the Marie Curie Integration Grant (ATTOTREND) as a Fellow Researcher. She has published more than 45 peer reviewed articles in international journals working on atomic and molecular physics, in particular, developing new ab initio time-dependent treatments for the description of atoms and molecules subject to ultrashort intense laser pulses. Fernando Marti? was born in Madrid, Spain, in 1961. He graduated in Chemistry in 1984 and Physics in 1986 at the Universidad Autonomá de Madrid. He received his Ph.D. at the same university in 1986. He completed his postdoctoral studies at the University of Bordeaux (1988), the Université de Paris VI (1989?1990), and the Univesity of Chicago (1995?1996). He has been a Full Profesor at the Universidad Autonomá de Madrid since 2005. His research focuses on the theoretical modeling of photoexcitation and photoionization of atomic and molecular systems induced by synchrotron radiation and ultrashort laser pulses, as well as that of complex molecular systems, isolated or deposited on surfaces. He has published over 350 articles, and since 2011, he has been the recipient of an Advanced Grant from the European Research Council.

Abstract

We have expressed the neuroendocrine VGF protein in FRT rat thyroid cells to study the molecular mechanisms of its sorting to the regulated and polarized pathways of secretion. By immunoelectron microscopy, we have demonstrated that VGF localizes in dense-core granules. Rapid secretion of VGF is induced by PMA stimulation. Moreover, human chromogranin B, a protein of the regulated pathway, co-localizes in the same granules with VGF. In confluent, FRT monolayers on filters protein secretion occur from the apical cell domain. VGF deletion mutants have been generated. By confocal microscopy, we have found that in transient transfection, all mutant proteins are sorted into granules and co-localize with the full-length VGF. They all retain the apical polarity of secretion. We also found that intracellular VGF and its deletion mutants are largely in an aggregated form. We conclude that FRT thyroid cells correctly decode the sorting information of VGF. The signals present on the protein to enter the granules and to be secreted apically cannot be separated from each other and are not in just one discrete portion of the protein. We propose that selective aggregation might represent the signal for sorting VGF to the regulated, apical route.

Abstract

We demonstrate that dissociative ionization of O(2) can be controlled by the relative delay between an attosecond pulse train (APT) and a copropagating infrared (IR) field. Our experiments reveal a dependence of both the branching ratios between a range of electronic states and the fragment angular distributions on the extreme ultraviolet (XUV) to IR time delay. The observations go beyond adiabatic propagation of dissociative wave packets on IR-induced quasistatic potential energy curves and are understood in terms of an IR-induced coupling between electronic states in the molecular ion.
