Background: Congenital disorder of glycosylation caused by mutation of the DOLK(DOLK-CDG) is a group of rare autosomal recessive diseases with an early-onset age and poor prognosis. DOLK-CDG can cause the dysfunction of multiple systems and organs such as the heart, skin, nerves, and bones. Case Presentation: We report a child with DOLK-CDG diagnosed and treated in the Affiliated Hospital of Qingdao University. The child was born with neonatal asphyxia, Ichthyoid rash, and congenital heart disease. His fingers of both the hands looked like lotus roots, and the palm and foot were covered by a white membrane. He was hospitalized with a severe infection at 4 months after birth. Physical examination showed that he was complicated with development delay and hypotonia. He experienced convulsions 1 hour after admission and died of multiple organ failure 2 hours after admission. Blood samples were taken for genetic testing before the child died. The results showed that there was a novel compound heterozygous mutation in DOLK, c.1268C＞G (P.P423R）and c.1581_1583del (P.527_528del). Conclusion: This mutation is new and not included in the human gene mutation library. The discovery of the novel mutation broadened the mutation spectrum of DOLK. At the same time, we sorted out the DOLK-CDG gene mutation sites and related clinical manifestations reported by August 2021 through a literature review.

Methane's global warming potential (GWP) is much larger than carbon dioxide and contributes significantly to global warming. Solar chimney power plant (SCPP) integrated with photocatalytic reactors can capture and remove atmospheric methane, and generate electrical power without fossil energy consumption simultaneously. In this paper, the performance of the flow characteristics, the CH4 removal, the CO2 emission reduction, and the power generation were analyzed for the SCPP integrated with different types of photocatalytic reactors under ambient crosswind (ACW). The results revealed that the SCPP integrated with a honeycomb reactor was more stable for the degradation of CH4 than that with a plate reactor. With an increase in ACW, the removal rate of atmospheric CH4 was reduced to a constant value of 0.41 g/s for the honeycomb reactor and 0.11 g/s for the plate reactor. The SCPP integrated with a honeycomb reactor achieved a maximum power generation of 88.31 kW, which was 1.63 times than that of the conventional SCPP when G = 857 W/m2 and ACW = 0 m/s. In addition, the improved SCPP could reduce CO2 emissions by 85.04 kg/h when G = 857 W/m2, ACW = 0 m/s, and △P = 320 Pa.

Wool fiber is widely used for clothing in our daily life due to its excellent physical and mechanical properties. However, in the daily laundry process, wool products can shrink severely by strong mechanical action in a warm and moist environment due to the fiber surface scales. Aiming to overcome the difficulty of washing delicate clothes such as wool and silk, Panasonic Intellectual Property Management Co. Ltd. invented a new washing machine for delicate clothes, based on tapping actions. The washing performance of wool sweaters in tapping washing is compared with that of hand washing and gentle and normal machine washing. The results show that tapping washing always gives wool sweaters the best appearance, dimensional stability and mechanical property up to 10 washing cycles. The changes in appearance and mechanical properties of wool sweaters are mainly due to the rate of felting shrinkage. By comparing the magnitude, duration, number of mechanical actions and direction of the forces acting on the fabric in tapping washing, hand washing and gentle machine washing, it can be concluded that the direction of forces acting on the fabric has a significant effect on wool fabric felting shrinkage, and unidirectional force has less effect on felting shrinkage than multidirectional forces. Tapping washing can give the lowest felting rate to wool samples because the forces in tapping washing on all parts of the fabric have a similar direction.

To achieve efficient delivery and precise release of chemotherapy drugs at tumor sites, an active targeting multi-responsive drug delivery platform was developed. Here, doxorubicin hydrochloride (DOX) was loaded onto polydopamine (PDA), which were coated by the cystamine-modified hyaluronic acid (HA-Cys), designated as DOX@PDA-HA (PDH). The combination of PDA and HA-Cys endowed the nanoplatform photothermal conversion, tumor-targeting, and pH/redox/NIR sensitive drug release capacity. Moreover, HA could be degraded by the excess hyaluronidase (HAase) in the tumor microenvironment (TME), promoting DOX release, and further enhancing the effect of chemotherapy. Experimental results demonstrated PDH good biocompatibility, high loading rate, targeted drug delivery, and efficient tumor cell killing ability. This ingenious strategy based on PDH showed huge potential in photothermal/chemotherapy combination treatment of cancer.

Botryosphaeria canker and fruit rot caused by the fungus Botryosphaeria dothidea is one of the most destructive diseases of apple worldwide. Our data indicated that the phosphatase cascade Nem1/Spo7-Pah1 plays important roles in the regulation of fungal growth, development, lipid homeostasis, environmental stress responses, and virulence in B. dothidea .

Pokkah Boeng disease (PBD) is one of the main diseases affecting sugarcane in China, seriously damaging sugarcane production and economic development. Therefore, it is important to clarify the pathogenic mechanisms of this disease and to provide a theoretical basis for the breeding of PBD-resistant sugarcane strains.

High expression of immune checkpoints (ICs) and senescence molecules (SMs) contributes to T cell dysfunction, tumor escape, and progression, but systematic evaluation of them in co-expression patterns and prognosis in acute myeloid leukemia (AML) was lacking. Three publicly available datasets (TCGA, Beat-AML, and GSE71014) were first used to explore the effect of IC and SM combinations on prognosis and the immune microenvironment in AML, and bone marrow samples from 68 AML patients from our clinical center (GZFPH) was further used to validate the findings. High expression of CD276, Bcl2-associated athanogene 3 (BAG3), and SRC was associated with poor overall survival (OS) of AML patients. CD276/BAG3/SRC combination, standard European Leukemia Net (ELN) risk stratification, age, and French-American-British (FAB) subtype were used to construct a nomogram model. Interestingly, the new risk stratification derived from the nomogram was better than the standard ELN risk stratification in predicting the prognosis for AML. A weighted combination of CD276 and BAG3/SRC positively corrected with TP53 mutation, p53 pathway, CD8+ T cells, activated memory CD4+ T cells, T-cell senescence score, and Tumor Immune Dysfunction and Exclusion (TIDE) score estimated by T-cell dysfunction. High expression of ICs and SMs was associated with poor OS of AML patients. Cell senescence deters the activation of various oncogenes. Induction of senescence is therefore a potentially effective strategy to interfere with vital processes in tumor cells. Sphingosine-1-phosphate receptor 1 (S1PR1) has been implicated in various cancer types, including ovarian cancer. The mechanism by which S1PR1 regulates ovarian cancer cell senescence is currently elusive. In this study we demonstrate that S1PR1 was highly expressed in human ovarian cancer tissues and cell lines. S1PR1 deletion inhibited the proliferation and migration of ovarian cancer cells, which concurred with cell cycle arrest in the G1/S phase. S1PR1 deletion enhanced cisplatin-induced ovarian cancer cell senescence. Treatment of ovarian cancer cells with sphingosine-1-phosphate (S1P) led to increased expression of 3-phosphatidylinositol-dependent protein kinase 1 (PDK1), decreased expression of large tumor suppressor 1/2 (LATS1/2), and increased activity of the yes-associated protein (YAP). Opposite results were obtained in S1PR1-knockout or pharmacological inhibition in ovarian cancer cells. LATS1/2 inactivation caused by S1PR1 deletion increased YAP expression, inhibited senescence, and lead to increased S1PR1 expression. Transcriptional regulation of S1PR1 by YAP was confirmed by chromatin immunoprecipitation. Accordingly, the S1PR1-PDK1-LATS1/2-YAP pathway regulates ovarian cancer cell senescence and does so through a YAP-mediated feedback loop. S1PR1 constitutes a druggable target for the induction of senescence in ovarian cancer cells. Pharmacological intervention in the S1PR1-PDK1-LATS1/2-YAP signaling axis may augment the efficacy of standard chemotherapy.

Aims Most studies focus on the effects of biofertilizer on the bacterial and fungal communities, and we still lack an understanding of biofertilizer on the protistan community. Here, the effects of biofertilizer containing Paenibacillus triticisoli BJ-18 on composition and interaction of the protistan community in the wheat rhizosphere were investigated. Methods and Results Biofertilizer application altered soil physicochemical properties and the protistan community composition, and significantly induced an alpha diversity decline. Random forecast and redundancy analysis demonstrated that nitrogenase activity and available phosphorus were the main drivers. Trichomonas classified to the phylum Metamonada was enriched by biofertilizer, and was significantly positive connected with soil nitrogenase activity and some function genes involved in nitrogen-fixation and nitrogen-dissimilation. Biofertilization loosely connected biotic interactions, while it did not affect the stability of the protistan community. Besides, biofertilizer promoted the connections of protists with fungi, bacteria, and archaea. Combined with biotic networks (protists, fungi, bacteria, and archaea) and interactions between protists and soil physicochemical properties/function genes, protists may act as keystone taxa potentially driving soil microbiome composition and function. Significance and Impact of the Study Overall, these results suggest that the biofertilizer is a driver of the soil protistan community, contributing to ecosystem functioning.

Heat shock protein 70 (HSP70) is a critical stress protein for repressing inflammation, however, its role in AKI is not fully understood. The current study aimed to determine the protective effect of HSP70 on septic AKI and its underlying mechanisms.Methods: Hsp70.1 knockout and wildtype mice were used for creating sepsis model by cecal ligation and puncture (CLP). Renal function, histological changes, pro-inflammatory cytokines, and apoptosis were analyzed with H&E, PAS, ELISA, western-blot, and immunofluorescence. Moreover, the effects of HSP70 on renal proximal tubular epithelial (HK-2) cells with LPS were assessed by measuring the levels of nuclear factor kappa B (NF-κB) signaling and downstream cytokines, viability, and apoptosis using western-blot, qRT-PCR, flow-cytometry, and immunofluorescence. Immunoprecipitate and immunoblotting were used for determining the interaction of HSP70 with tumor necrosis factor receptor-associated factor 6 (TRAF6). Exogenous HSP70 was applied to further identify its biological significance at the cellular and animal level.Results: Hsp70.1 deficiency significantly aggravated renal dysfunction with increasing serum levels of BUN, SCr, kidney injury molecule-1 (KIM-1), and neutrophil gelatinase-associated lipocalin (NGAL), and shortened survival in CLP mice. Furthermore, hsp70.1 knockout caused renal-tissue structural damage, especially proximal tubular, and inflammatory cascade and increased apoptotic cells, along with elevated Bax, caspase 3 and cleaved caspase 3, as well as decreased Bcl2 in vivo and vitro. Significantly, HSP70 directly interacted with TRAF6 in HK-2 cells, leading to suppression of inflammatory response and apoptosis. Moreover, exogenous HSP70 alleviated renal damage, decreased apoptosis and elevated survival rate in septic AKI in vivo and vitro.Conclusion: Our findings demonstrated that HSP70 played a critical role in sepsis-induced AKI via interaction with TRAF6 and inhibiting inflammation and apoptosis.Keywords: heat shock protein 70, sepsis, acute kidney injury, tumor necrosis factor receptor-associated factor 6, nuclear factor kappa B, apoptosis

This letter presents our study on a millimeter wave (mm-wave) orthomode transducer (OMT) based on novel double-ridged waveguide junction. By reducing the length of ridges in the square waveguide, the ridge width of proposed junction is about three times wider than that of conventional junction. This benefits the fabrication and mechanical performance of double-ridged waveguide OMT in the higher mm-waveband or terahertz band. The simulated reflection coefficients for both orthogonal modes are less than -20 dB over 37% bandwidth. As practical application, an E-band OMT based on the novel junction is fabricated. A smooth-horn is integrated with the OMT as a dual-polarized antenna. The measured results show that reflection coefficients less than -15 dB, isolation better than 39 dB, and cross-polarization discrimination (XPD) higher than 23 dB are achieved from 71 to 86 GHz.

Exploiting two-dimensional room temperature ferromagnetic materials is always a significant and valuable work. However, the actual number of satisfied materials with intrinsic ferromagnetism is very limited. Here, the van der Waals Cr4Te5 epitaxial films prepared by pulsed laser deposition technique were confirmed to hold ferromagnetic ordering state until to 300 K. We find that the Cr4Te5 films can easily achieve the epitaxial growth along a single orientation for the hexagonal Al2O3(0001) substrate. However, as for cubic SrTiO3(001) and multiphase structure Mica substrate, the Cr4Te5 films only show an uniaxial growth instead of epitaxial growth. Based on the investigation of electronic transport in metallic regime, it is revealed that the interaction of electronmagnon scattering only exists in the lower temperature region. Our work render two-dimensional vdW Cr4Te5 crystal a very promising material for developing practical spintronic nanodevices.

The transcriptional initiation of genes is inextricably bound with the functions of cis-regulatory sequences. The pig is one of the most important livestock species and an ideal animal model for biomedical studies. At the same time, the liver is a critical organ with diverse and complex metabolic functions. Here, we performed Cleavage Under Targets and Tagmentation (CUT&Tag) coupled with high-throughput sequencing to profile the chromatin landscape of histone H3 lysine 27 acetylation (H3K27ac), histone H3 lysine 4 monomethylation (H3K4me1), and CCAAT enhancer-binding protein β (C-EBPβ) in the 70-d-old porcine liver, compared the different profiles among the three markers and their associated stitched-enhancers by stitching and sorting the peaks within 12.5 kb (Pott and Lieb, 2015) and generated the porcine liver-specific super-enhancers (SEs) by the combination of three markers. Compared to typical enhancers (TEs) and other stitched-enhancers, liver-specific SEs showed a higher density of cis-motifs and SNPs, which may recruit more tissue-specific vital TFs. The expression profiles in fetal and 70-d-old pigs proved that a large proportion of SE-associated genes were up-regulated and were more related to hepatic metabolisms and detoxification pathways. Our results illustrated the difference and connection among promoter and enhancer markers, identified the features of liver SEs and their associated genes, and provided novel insight into cis-element identification, function, and liver transcriptional regulation.

BackgroundWith the ongoing development of the information society, the Internet and smartphones have become an essential way of life, but also fostered the problem of mobile phone dependence. Physical activity and subjective well-being have both been shown to correlate with mobile phone dependence, but the impact of subjective well-being on the relationship between physical activity and mobile phone dependence is not fully understood. This study investigates subjective well-being as a potential mediating variable in the relationship. It also investigates whether psychological capital moderates the association between subjective well-being and mobile phone dependence.MethodsA total of 9,569 students from 38 universities in Jiangsu Province were selected. Participants were surveyed via the online questionnaire distribution platform Questionnaire Star. Common method bias test and Pearson correlation tests were used to analyze the study indicators, and the theoretical model for this study was validated using Process plug-in developed by Hayes and set at p ResultsThe levels of physical activity, subjective well-being, and psychological capital were all significantly higher for male students than female students. However, female students had a significantly higher level of mobile phone dependence. As predicted, there was a direct negative correlation between physical activity and mobile phone dependence, and subjective well-being mediated that relationship. Psychological capital moderated the relationship between subjective well-being and mobile phone dependence. It also positively moderated the indirect effect between physical activity and mobile phone dependence via subjective well-being.

Skin and subcutaneous tissue tightening is usually treated by noninvasive photothermal treatment for medical esthetics purpose, while the underlying mechanism remains to be elucidated. Here, we hypothesized that adipocyte injury, as a stimulator, may regulate extracellular matrix (ECM) production by increasing ALOX15 in macrophages, which could lead to fibroblast activation. In this study, we show that lipolysis was induced by laser heating (45°C for 15 min) in patients and rats, and adipocyte thermal injury stimulates the ECM production in fibroblasts by ALOX15 that was increased in cocultured macrophages. These phenomena were evidenced by the ALOX15 knockdown. In addition, ALOX15 metabolite 12(S)-HETE activated p38 MAPK signaling pathway that mediated the production of ECM in fibroblast. In summary, the results of this study demonstrate that the mechanisms of adipose photothermal injury-induced skin and/or subcutaneous tissue tightening may have clinical relevance for noninvasive or minimally invasive photothermal therapeutics.

This letter presents our study on a compact and low-loss E-band bandpass filter (BPF) based on multilayer groove gap waveguide (GGW). Different from other reported planar GGW filters and vertically stacked substrate integrated waveguide (SIW) filters, the proposed filter realizes a cross-coupled quadruplet topology by introducing a pair of unique circular capacitive holes across layers, innovative ridges (capacitive iris), and conventional inductive irises on the same layer, which reduces the insertion loss (IL) while having a smaller size. By tuning the dimensions of the coupling structures and cavities, the filter is optimized to exhibit a superior performance in simulation. The designed filter is fabricated using CNC machining and verified in experiment. The measured results agree with the simulated ones quite well, showing an average IL of 0.45 dB and an 8% relative 3-dB bandwidth at a center frequency of 73.65 GHz. Therefore, the proposed multilayer GGW structure opens a new avenue in designing high-performance filters in millimeter-wave bands.

Variable Renewable Energy (VRE), i.e., wind and solar photovoltaics (PVs), is being installed in rapidly increasing amounts around the world. Growth in VRE is being spurred by ambitious zero-carbon targets set by countries and individual states across the globe. The European Union approved a carbon neutrality target for 2050 in 2019. Japan’s newly appointed prime minister announced the same target in 2020, and the Chinese government set goals to peak carbon emissions before 2030 and become carbon neutral by 2060.
