Abstract

IntroductionLate-onset Alzheimer’s disease (LOAD, onset age > 60 years) is the most prevalent dementia in the elderly1, and risk is partially driven by genetics2. Many of the loci responsible for this genetic risk were identified by genome-wide association studies (GWAS)3–8. To identify additional LOAD risk loci, the we performed the largest GWAS to date (89,769 individuals), analyzing both common and rare variants. We confirm 20 previous LOAD risk loci and identify four new genome-wide loci (IQCK, ACE, ADAM10, and ADAMTS1). Pathway analysis of these data implicates the immune system and lipid metabolism, and for the first time tau binding proteins and APP metabolism. These findings show that genetic variants affecting APP and Aβ processing are not only associated with early-onset autosomal dominant AD but also with LOAD. Analysis of AD risk genes and pathways show enrichment for rare variants (P = 1.32 × 10−7) indicating that additional rare variants remain to be identified.

3. Phosphotyrosyl-Based Motifs in the Structure-Based Design of Protein­ Tyrosine Kinase-Dependent Signal Transduction Inhibitors

Abstract: Transmission of extracellular signals from the cell membrane to the nucJeus depends on modification of phosphorylation states of intracellular proteins. Tyrosine, serine and threonine residues are the principal amino acid targets of these phosphorylation events, with tyrosyl residues being particularly important for pathways mediated by growth factors and cytokines. Aberrations in phosphotyrosyl (pTyr)­ dependent signalling can contribute to a variety of diseases, including canc.ers, and for this reason selective modulation of pTyr-dependent signalling may afford new therapeutic approaches. The design of such therapeutics is facilitated by the functional compartmen­ talization of pTyr dependent signalling into three broad categories: ( l) the generation of pTyr residues by protein-tyrosine kinases (PTK); (2) pTyr-dependent protein-protein associations mediated by binding modules such as Src homology 2 (SH2) and phosphotyrosine binding (PTB) domains and (3) the dephosphorylation of pTyr residues by protein-tyrosine phosphatases (PTPs). The pTyr residue itself, which is a unifying component of this signalling triad, potentially affords a starting point for the design of antagonists. In the PTK, SH2/PTB and PTP domain signalling environments, the pTyr residue plays unique roles by participating in interactions characteristic to each. Therefore, depending on which aspects of the L-4'-phosphotyrosyl structure are emphasized, and the manner in which they are utilized, inhibitors can be potentially directed against distinct legs of the signalling triad. This review will provide examples of this, by examining several series of compounds that have been prepared as inhibitors of either PTKs, SH2/PTB domains or PTPs. Also described will be how the pTyr structure can serve as a thematic Rosetta stone for the development of signal transduction inhibitors.

4. 'Bubbling reefs' in the Kattegat: submarine landscapes of carbonate-cemented rocks support a diverse ecosystem at methane seeps

Abstract

Methane seeps in shallow waters in the northern Kattegat off the Danish coast form spectacular submarine landscapes - the 'bubbling reefs' - due to carbonate-cemented sandstone structures which are colonized by brightly coloured animals and plants. These structures may be 100 m2 in area and consist of pavements, complex formations of overlying slab-type layers, and pillars up to 4 m high. The carbonate cement (high-magnesium calcite, dolomite or aragonite) is 13C-depleted, indicating that it originated as a result of microbial methane oxidation. It is believed that the cementation occurred in the subsurface and that the rocks were exposed by subsequent erosion of the surrounding unconsolidated sediment. The formations are interspersed with gas vents that intermittently release gas, primarily methane, at up to 25 1 h-' The methane most likely originated from the microbial decomposition of plant material eposited during the Eemian and early Weichselian periods, i.e. l00 000 to 125 000 years B.P. Aerobic methane oxidation in the sediment was restricted Lo the upper 4 cm in muddy sand and to the upper 13 cm In coarse sand. Maximum aerobic methane oxidation rates ranged from 4.8 to 45.6 pm01 dm-3 d". The rock surfaces and epifauna around the seeps were also sites of methane-oxidizing activity. Integrated sulphate reduction rates for the upper 10 cm of muddy sand gave 4.2 to 26.6 mm01 m-2 d-' These rates are higher than those previously reported from similar water depths in the Kattegat but did not relate to the sediment methane content. Since gas venting occurs over several km2 of the sea floor in the Kattegat it is likely to make a significant local contribution to the cycling of elements in the sediment and the water column. The rocks support a diverse ecosystem ranging from bacteria to macroalgae and anthozoans. Many animals live within the rocks in holes bored by sponges, polychaetes and bivalves. Stable carbon isotope composition (6'") of tissues of invertebrates from the rocks were in the range -17 to -24 'A, indicating that methane-derived carbon makes little direct contribution to their nutrition. Within the sediments surrounding the seeps there is a poor metazoan fauna, in terms of abundance, diversity and biomass. This may be a result of toxicity due to hydrogen sulphide input from the gas.

Abstract

At least eight neurodegenerative diseases, including Huntington disease, are caused by expansions in (CAG)n repeats in the affected gene and by an increase in the size of the corresponding polyglutamine domain in the expressed protein. A hallmark of several of these diseases is the presence of aberrant, proteinaceous aggregates in the nuclei and cytosol of affected neurons. Recent studies have shown that expanded polyglutamine (Qn) repeats are excellent glutaminyl-donor substrates of tissue transglutaminase, and that the substrate activity increases with increasing size of the polyglutamine domain. Tissue transglutaminase is present in the cytosol and nuclear fractions of brain tissue. Thus, the nuclear and cytosolic inclusions in Huntington disease may contain tissue transglutaminase-catalyzed covalent aggregates. The (CAG)n/Qn-expansion diseases are classic examples of selective vulnerability in the nervous system, in which certain cells/structures are particularly susceptible to toxic insults. Quantitative differences in the distribution of the brain transglutaminase(s) and its substrates, and in the activation mechanism of the brain transglutaminase(s), may explain in part selective vulnerability in a subset of neurons in (CAG)n-expansion diseases, and possibly in other neurodegenerative disease. If tissue transglutaminase is found to be essential for development of pathogenesis, then inhibitors of this enzyme may be of therapeutic benefit.
