Abstract

4 pages, Coral gardens are increasingly being reported at 40–200 m depth in the Mediterranean Sea. These coral assemblages are composed of gorgonians from shallow coastal rocky bottoms extending their distribution to deeper areas, as well as by gorgonians and black corals with distribution restricted to the continental shelf and upper slope. Gorgonians and black corals are among the main structural species in deep benthic ecosystems, providing shelter and reproductive grounds for a highly diverse associated fauna. Due to their branching morphology and erect position, these deep coral gardens are extremely vulnerable to several fishing activities (bottom trawling, long-lines and trammel net fishing) and should consequently be protected through an ecosystem-based fishery management

7. Molecular Cloning of Silicatein Gene from Marine Sponge Petrosia ficiformis (Porifera, Demospongiae) and Development of Primmorphs as a Model for Biosilicification Studies

Abstract

In some sponges peculiar proteins called silicateins catalyze silica polymerization in ordered structures, and their study is of high interest for possible biotechnological applications in the nanostructure industry. In this work we describe the isolation and the molecular characterization of silicatein from spicules of Petrosia ficiformis, a common Mediterranean sponge, and the development of a cellular model (primmorphs) suitable for in vitro studies of silicatein gene regulation. The spicule of P. ficiformis contains an axial filament composed of 2 insoluble proteins, of 30 and 23 kDa. The 23-kDa protein was characterized, and the full-length cDNA was cloned. The putative amino acid sequence has high homology with previously described silicateins from other sponge species and also is very similar to cathepsins, a cystein protease family. Finally, P. ficiformis primmorphs express the silicatein gene, suggesting that they should be a good model for biosilicification studies.

8. ABA- and cADPR-mediated effects on respiration and filtration downstream of the temperature-signaling cascade in sponges

Abstract

Recently, the thermosensing pathway in sponges (Porifera) was elucidated. The thermosensor triggering this cascade is a heat-activated cation channel, with the phytohormone abscisic acid (ABA), cyclic ADP-ribose (cADPR) and calcium acting as intracellular messengers, similarly to the drought-stress signaling cascade in higher plants. Here, we investigated the functional effects downstream of the temperature-signaling pathway in Axinella polypoides (Porifera, Demonspongiae). Short-term stimulation followed by long-term depression of amino acid incorporation, oxygen consumption and water filtration were observed after exposure of the sponge to a brief heat stress or to micromolar ABA. These effects could be prevented by the targeted interruption of the signaling pathway either at the level of the cation channel thermosensor or at the level of the cADPR-induced intracellular calcium increase. Moreover, release of cyclase activity into the sea water and generation of extracellular cADPR were observed following brief heat stress. Intact sponge cells were sensitive to extracellular cADPR and addition of purified cyclase increased sponge respiration similarly to heat stress. This is the first observation of functional effects exerted on Metazoa by the phytohormone ABA: conservation of the ABA/cADPR stress-signaling cascade points to its early evolution in a common precursor of modern Metazoa and Metaphyta. The functional effects induced by extracellular cyclase/cADPR suggest an evolutionary origin of cADPR as an ancient stress hormone in Porifera.

Abstract

The by-the-wind sailor Velella velella is a cosmopolitan, pleustonic colonial hydrozoan, common in the Mediterranean Sea. It forms offshore aggregations that occasionally strand on the coasts, following strong wind conditions. Colonies are frequently discovered around Janthina pallida Janthina pallida, a pleustonic gastropod that feeds on them and floats on the secretion of a bubble raft. Because of the massive occurrence of these species along the Italian coasts, and their often neglected ecological roles, it is pivotal to increase the knowledge of their genetic structure. Analyses of the genetic variability of V. velella and J. pallida, stranded on the beach in springs 2016 and 2017 at ten locations along the Ligurian and Tyrrhenian seas, show the existence of panmictic Mediterranean populations. Nevertheless, V. velella showed high genetic variability with a high number of singleton haplotypes, while J. pallida showed a recent population expansion. Considering the ecological and socio-economic importance of gelatinous zooplankton in today's changing marine ecosystems, these results lay the basis for a better understanding of the ecological role and population dynamics of Mediterranean pleustonic species.

Abstract

The Mediterranean red coral, Corallium rubrum (L.), has been a valuable economic resource for more than 2000 years. The Sicily Channel and surrounding areas are one of the most famous red coral fishing grounds of the whole region, hosting the deepest ever found living colonies and large sub-fossil red coral deposits; the so-called Sciacca banks are a unique location in the whole Mediterranean Sea. In this paper, a morphometric description of this sub-fossil population is presented for the first time from studies of colonies in the collection of several coral factories from Torre del Greco (Naples), with radiocarbon age estimations and growth rate evaluations. From the results of this study, after several thousand years Sciacca red coral colonies maintained the organic matrix structure with evident annual discontinuities, allowing estimations of the annual growth rate (about 0.3 mm/year) and the average population age (about 33.5 years). These resulting data are similar to the values determined for deep-dwelling living red coral populations. The radiocarbon dating evidenced a range of ages, from 8300 to 40 years before 1950 CE, mostly falling between 2700 and 3900 YBP, suggesting that colonies accumulated over a wide span of time. In view of the tectonically active nature of the area, several catastrophic events affected these ancient populations, maintaining them in a persistent state of early-stage, structurally similar to the those in current over-exploited areas.

Abstract

The colonization of epilithic diatoms on artificial hard substrates (marble, quartzite and slate) was investigated on a seasonal basis in a subtidal site of the northern Adriatic Sea to determine if substrate-dependent differences in colonization occurs and to define the seasonal variations of microepilithic communities in terms of abundance, biomass and community structure. Artificial substrates (smoothed small discs mounted on a Plexiglas sheet) were placed at a depth of 8 m in April 2003, July 2003, January 2004 and February 2004. The discs were collected after 6–7 weeks for counting and taxonomic identification of the diatoms. The community structure of epilithic diatoms showed a dominance of motile species over the entire study period, followed by erect, adnate and tube-dwelling diatoms. Diatom density showed a marked seasonal variability, ranging from 365±407 (winter 2004) to 557156±82602 cells cm-2 (spring 2004). Biomass ranged from 0.02±0.01 to 17.53±3.20 μg C cm-2. Abundance and biomass values did not present any significant differences for the three substrates examined.

Abstract

A wealth of data are available on the seasonal variation of plankton communities, but few studies take into consideration the circannual cycles of benthic organisms. In fact, the macrobenthos is generally considered as composed by slow-growing organisms showing variations mainly in relation to substrate competition. On the contrary, hydroids are an important group of macrobenthos that, in temperate but also in tropical waters, undergo sharp seasonal cycles. For example, on the Portofino Promontory (Ligurian Sea), about half of the recorded species thrive in winter and 30% in the summer, the rest being constant in presence. Here, we revise the available data about the environmental factors switching the passage from quiescent to active phases. The available data suggest that a mosaic of physical factors, biotic interactions, and internal clues triggers the hydroid cyclic behaviour.

Abstract

Chemical analysis of the Indonesian soft coral Sinularia sp. (order Alcyonacea, family Alcyoniidae) afforded two new alkaloids, named sinulasulfoxide (1) and sinulasulfone (2), characterized by an amide linkage between a phytanic acid moiety and an uncommon sulfur-containing unit. Their complete stereostructures were elucidated by interpretation of MS and NMR data along with CD analysis and chemical modifications. Sinulasulfoxide (1) proved to moderately inhibit LPS-induced NO release.

Abstract

Two new steroidal derivatives, named clionastatins A and B, have been isolated from the burrowing sponge Cliona nigricans. These molecules are tri-and tetra-chlorinated androstane derivatives, respectively, and they represent the first polyhalogenated steroids found in a natural organism, either marine or terrestrial, and the first examples of halogenated androstanes in nature. Both clionastatins proved to be potently cytotoxic.
