A variety of techniques exist for tracking and detection of pedestrian traffic.The “proof of concept” or the utility of these methods is often illustrated by analysis of a video or photographs produced by the researcher as part of the development process of the algorithms.Although these images are often based on actual human subjects, they lack portability and ground truth or at best require tedious hand mapping to record ground truth.Hence, each algorithm is developed and tested for a unique situation.Consequently, as an alternative process we propose using gaming techniques to generate pedestrian and crowd like movements that readily produce ground truth referenced via data logs.For this initial study, we have used modifications of the Reynolds flocking model to generate crowd like behavior.Using these algorithms and open-source software platforms, we generated reference crowds and then added individual pedestrian behavior within the simulated crowd.Various detection methods were applied to differentcrowd scenarios to explore and assess the utility of detection methods, illustrate the possibilities of this technique, and demonstrate an initial screening for a detection algorithm.Although not a final proof of a detection process, this method allows facile, rapid, and comparative initial evaluation of the methods under consideration.

IL-6 is a multifunctional cytokine involved in hemopoiesis, immune regulation, inflammation, neural development, and infection. IL-6 belongs to a family of related cytokines that includes leukemia inhibitory factor, oncostatin M, IL-11, ciliary neurotropic factor, and cardiotropin-1, all of which initiate signaling through a receptor-associated gp130. IL-6 induces homodimerization of gp130 and activates the Jak/STAT pathway of signal transduction. In addition, IL-6 stimulates the mitogen-activated protein kinases designated ERK (extracellular signal-regulated kinase)-1 and -2. Activation of ERK-1 and -2 may involve the Src homology-2 containing proteins Shc and Grb2. Here we provide evidence that Shc could function as signaling molecules for IL-6 in DeFew-IL-6R/gp130 cells, a human B lymphoma cell line engineered to express high levels of both the IL-6R (p80) and the gp130 subunit. IL-6 was shown to promote the rapid tyrosine phosphorylation of gp130, Jak2, and Shc proteins. Moreover, Shc associated both in vivo and in vitro with phosphorylated gp130 through the Shc-Src homology-2 domain. We also report that Shc bound to activated Jak2 by using the Shc amino terminal phosphotyrosine interaction domain. Following IL-6 stimulation, Shc physically associated with Grb2. Thus, the data point to Shc proteins as a functional link between the Jak2 and Ras pathways of IL-6 signal transduction.

Background: Although diabetes mellitus is of high concern in Iran, and the level of control is unacceptable, few qualitative studies have been carried out to reflect the experiences of patients on the barriers and motivators to self-care. This study aimed to explore a culturally based experience of Iranian diabetic patients regarding the personal and environmental barriers to and facilitat­ing factors for diabetes self-care. Methods: Six focus groups were conducted among type 2 diabetic patients in the Charity Foundation for Special Diseases' diabetes clinic. Purposeful sampling was used. Newly diagnosed patients (less than six months) and all type 1 diabetic pa­tients were excluded. Three focus groups were held on for each sex. A total of 43 patients participated in the study. Frame­work analysis was used to extract the themes from the data. Results:Data analysis showed five main barriers: physical barriers (such as physical effects of diabetes); psychological bar­riers (such as health beliefs); educational barriers (such as lack of knowledge about diabetes); social barriers (such as group pressure); and care system barriers (such as service availability). Along with the barriers, there were some motivators that the participants mentioned as a stimuli to control their diabetes. They include beliefs about diabetes, perceived responsibil­ity for family, religious beliefs, and the views of significant others. Conclusion:Culturally based interventions are needed to improve diabetes care management in Iran. In addition to personal factors, diabetes health educators should pay attention to the environmental factors when they develop programs.

Galaxies in rich clusters with z $\gtrsim$ 0.3 are observed to have a higher fraction of photometrically blue galaxies than their nearby counterparts. This raises the important question of what environmental effects can cause the termination of star formation between z $\approx$ 0.3 and the present. The star formation may be truncated due to ram-pressure stripping, or the gas in the disk may be depleted by an episode of star formation caused by some external perturbation. To help resolve this issue, surface photometry was carried out for a total of 70 early-type galaxies in the cluster Cl1358+62, at z $\sim$ 0.33, using two-color images from the Hubble Archive. The galaxies were divided into two categories based on spectroscopic criteria: 24 are type K+A (e.g., strong Balmer lines, with no visible emission lines), while the remaining 46 are in the control sample with normal spectra. Radial color profiles were produced to see if the K+A galaxies show bluer nuclei in relation to their surrounding disks. Specifically, a linear gradient was fit to the radial color profile of each galaxy. We find that the K+A galaxies on average tend to have slightly bluer gradients towards the center than the normals. A Kolmogorov-Smirnov two-sample test has been applied to the two sets of color gradients. The result of the test indicates that there is only a $\sim$2% probability that the K+A and normal samples are drawn from the same parent distribution. There is a possible complication from a trend in the apparent magnitude vs. color gradient relation, but overall our results favor the centralized star formation scenario as an important process in the evolution of galaxies in dense clusters., 16 pages, 12 figures, accepted for publication in AJ

For many years our knowledge on hepatitis C virus (HCV) replication has been based on in vitro experiments or transfection studies. Recently, the first reliable system for studying viral replication in tissue culture cells was developed. Taking advantage of this system, we examined in detail the localization of viral nonstructural (NS) proteins in cells containing functional replication complexes. By fractionation experiments and immunomicroscopy, we observed that all NS proteins were associated with the endoplasmic reticulum (ER) membranes, confirming the hypothesis that the ER is the site of membrane-associated HCV RNA replication. Interestingly, NS3 and NS4A were preferentially localized in endoplasmic reticulum cisternae surrounding mitochondria, suggesting additional subcellular compartment-related functions for these viral proteins. Furthermore, the immunoelectron microscopy revealed the loss of the organization and other morphological alterations of the ER (convoluted cisternae and paracrystalline structures), resembling alterations observed in liver biopsies of HCV-infected individuals and in flavivirus-infected cells.
