Herein, we report anecdotes of manatee (family Trichechidae) behavior based upon accounts made by experienced observers. We have categorized the anecdotes into the broad categories of Conservation and Management (interactions with boats, people, and capture nets; rehabilitation and release), Behavioral Ecology (navigation, mother–calf interactions, interactions with conspecifics, mating behavior and birthing, anti-predator behavior, foraging and eating), and Cognition (sensory perception; learning, memory, and knowledge; interactions with objects, other species, and people; and mimetic behavior). These are all subjects that pertain to the goals of the Florida Manatee Recovery Plan of 2001. We found that many accounts report similar behaviors, suggesting that these may be fruitful lines of further inquiry. One subcategory, Navigation, appears to offer many research areas amenable to systematic study. The apparent behavioral flexibility and knowledge shown in manatee actions in several domains suggest that investigations of manatee intelligence are likely to be revealing.

We report the results from follow-up observations of two Roche-lobe filling hot subdwarf binaries with white dwarf companions predicted to have accretion disks. ZTF J213056.71+442046.5 (ZTF J2130) with a 39-minute period and ZTF J205515.98+465106.5 (ZTF J2055) with a 56-minute period were both discovered as subdwarf binaries with light curves that could only be explained well by including an accretion disk in their models. We performed a detailed high-resolution spectral analysis using Keck/ESI to search for possible accretion features for both objects. We also employed polarimetric analysis using the Nordic Optical Telescope (NOT) for ZTF J2130. We did not find any signatures of an accretion disk in either object, and placed upper limits on the flux contribution and variation in degree of polarisation due to the disk. Owing to the short 39-minute period and availability of photometric data over six years for ZTF J2130, we conducted an extensive $O - C$ timing analysis in an attempt to look for orbital decay due to gravitational wave radiation. No such decay was detected conclusively, and a few more years of data paired with precise and consistent timing measurements were deemed necessary to constrain $\dot P$ observationally., 10 pages, 8 figures. A promising progenitor scenario for Type Ia supernovae (SNeIa) is the thermonuclear detonation of a white dwarf in a close binary system with another white dwarf. After the primary star explodes, the surviving donor can be spontaneously released as a hypervelocity runaway. One such runaway donor candidate is LP 398-9, whose orbital trajectory traces back $\approx 10^5$ years to a known supernova remnant. Here we report the discovery of carbon-rich circumstellar material around LP 398-9, revealed by a strong infrared excess and analyzed with follow-up spectroscopy. The circumstellar material is most plausibly composed of inflated layers from the star itself, mechanically and radioactively heated by the past companion's supernova. We also detect a 15.4 hr periodic signal in the UV and optical light curves of LP 398-9, which we interpret as surface rotation. The rotation rate is consistent with theoretical predictions from this supernova mechanism, and the brightness variations could originate from surface inhomogeneity deposited by the supernova itself. Our observations strengthen the case for this double-degenerate SNIa progenitor channel, and motivate the search for more runaway SNIa donors., Accepted to MNRAS

Ulnar claw hand usually occurs when the ulnar nerve is damaged distally. Claw hand deformity is characterized by metacarpophalangeal hyperextension and interphalangeal flexion, making it impossible to oppose the fingers and thumb. Bouvier’s test is used to guide the procedure. Palliative surgery requires prior preparation of paralytic hands. In case of a positive Bouvier's test, Zancolli’s lasso technique is preferred because of its effectiveness. Capsuloplasty with anteroposterior transosseous fixation is used if the bone is strong enough and when flexor digitorum profondus muscle is inactive and does not allow flexor digitorum superficialis tendon transfer. In case of a negative Bouvier’s test with interphalangeal extension deficit of 45 degrees of less, direct interossei muscle restoration techniques by active transfers are performed. If the interphalangeal extension deficit is more than 45 degrees, proximal interphalangeal arthrodesis is indicated. Wartenberg first described actively irreducible abduction of the little finger. Wartenberg’s sign is seen when ulnar paralysis occurs, and during ulnar nerve regeneration. Treatment of isolated Wartenberg's sign consists of re-routing the extensor digiti minimi. Among the other techniques, Belmahi's "tie lasso" is preferred when flexible claw hand is associated with Wartenberg’s sign.

Palliative tendon transfer is an integral part of radial nerve palsy treatment. It can be considered in the first weeks when the possibility of nerve repair by direct suture or nerve grafting is not feasible or reasonable. Mostly, it is discussed secondarily when it is too late for nerve surgery and motor recovery cannot be expected, or after failure or incomplete recovery after nerve repair. The goal of tendon transfers is to restore wrist, finger and thumb extension. For wrist extension, the use of pronator teres is well accepted. The best tendon transfer for finger extension is debated. This can be restored doing a flexor carpi ulnaris (FCU), flexor carpi radialis or flexor digitorum superficialis (FDS) to extensor digitorum communis transfer. Regarding thumb extension and abduction, a palmaris longus (PL) or one FDS tendon to the rerouted extensor pollicis longus (EPL) transfer can be performed. If a transfer is done on the EPL without rerouting it, abduction can be restored by doing a tendon transfer to the abductor pollicis longus (APL) or an APL tenodesis. The different tendon transfer options are selected based on the surgeon’s preference, and most importantly, discussed with the patients to define the objectives together. The transfer is chosen based on the clinical examination (high or low radial nerve palsy, tendon available for transfer like PL, wrist mobility) and based on the patient’s needs and expectations (activities requiring the FCU, finger independence, independence of thumb extension or abduction). If the surgical rules and the postoperative instructions for rehabilitation are followed, tendon transfers for radial nerve palsy regularly produce very satisfactory results.

Misophonia – an unusually strong aversion to certain sounds – can cause significant distress and disruption to those who have it but is an enigma in terms of our scientific understanding. A key challenge for explaining misophonia is that, as with other disorders, it is likely to emerge from an interaction of traits that also occur in the general population (e.g., sensory sensitivity, anxiety) and that are transdiagnostic in nature (i.e., shared with other disorders). With a large sample of participants (N=1430), we performed a cluster analysis (based on responses to questions relating to misophonia) and identified two misophonia subgroups differing in severity, as well as a third group without misophonia. A subset of this sample (N=403) then completed a battery of measures designed to assess sensory sensitivity and clinical co-morbidities. Clinical symptoms were limited to the most severe group of misophonics (including autistic traits, migraine with visual aura, anxiety sensitivity, obsessive-compulsive traits). Both the moderate and severe groups showed elevated attention-to-detail and hypersensitivity (across multiple senses). A novel symptom network model of the data shows the presence of a central hub linking misophonia to sensory sensitivity which, in turn, connects to other symptoms in the network (relating to autism, anxiety, etc.). In this way, the severity of misophonia can be understood as a propagation from core features, which are sensory-attentional in nature, to peripheral clinical symptoms.

Background: Patients with mitral valve prolapse (MVP) may develop adverse outcomes even in the absence of mitral regurgitation or left ventricular (LV) dysfunction. Purpose: To investigate the prognostic value of mitral annulus disjunction (MAD) and myocardial fibrosis at late gadolinium enhancement (LGE) cardiac MRI in patients with MVP without moderate-to-severe mitral regurgitation or LV dysfunction. Materials and Methods: In this longitudinal retrospective study, 118 144 cardiac MRI studies were evaluated between October 2007 and June 2020 at 15 European tertiary medical centers. Follow-up was from the date of cardiac MRI examination to June 2020; the minimum and maximum follow-up intervals were 6 months and 156 months, respectively. Patients were excluded if at least one of the following conditions was present: cardiomyopathy, LV ejection fraction less than 40%, ischemic heart disease, congenital heart disease, inflammatory heart disease, moderate or worse mitral regurgitation, participation in competitive sport, or electrocardiogram suggestive of channelopathies. In the remainder, cardiac MRI studies were reanalyzed, and patients were included if they were aged 18 years or older, MVP was diagnosed at cardiac MRI, and clinical information and electrocardiogram monitoring were available within 3 months from cardiac MRI examination. The end point was a composite of adverse outcomes: sustained ventricular tachycardia (VT), sudden cardiac death (SCD), or unexplained syncope. Multivariable Cox regression analysis was performed. Results: A total of 474 patients (mean age, 47 years ± 16 [SD]; 244 women) were included. Over a median follow-up of 3.3 years, 18 patients (4%) reached the study end point. LGE presence (hazard ratio, 4.2 [95% CI: 1.5, 11.9]; P = .006) and extent (hazard ratio, 1.2 per 1% increase [95% CI: 1.1, 1.4]; P = .006), but not MAD presence (P = .89), were associated with clinical outcome. LGE presence had incremental prognostic value over MVP severity and sustained VT and aborted SCD at baseline (area under the receiver operating characteristic curve, 0.70 vs 0.62; P = .03). Conclusion: In contrast to mitral annulus disjunction, myocardial fibrosis determined according to late gadolinium enhancement at cardiac MRI was associated with adverse outcome in patients with mitral valve prolapse without moderate-to-severe mitral regurgitation or left ventricular dysfunction.

Since its inception50 years ago, clean intermittent catheterization (CIC) has become ubiquitous in managing lower urinary tract dysfunction in children. Emphasis has been on its impact on daily life, but little on its implementation and adjustment in families. The aim of the current study was to discover how families learned to implement and manage their child's CIC needs by interviewing caregivers, adolescents, and young adults about their experiences. Interviews were designed to uncover facilitators and barriers to beginning CIC to initiate potential improvements in a multidisciplinary approach.A semi-structured interview guide was developed and piloted initially to 12 families for validation. Between August 2018 and October 2019, 40 families (52 interviews of caregivers and patients12 years) were then interviewed with open-ended questions interspersed with more specific ones to generate discussion. Transcripts were coded using Dedoose software to create a base list with emergent codes. Inductive and deductive methods were employed to generate themes. Consensus was reached during successive team meetings.Five major and several subthemes emerged regarding implementation of CIC by caregivers and patients learning CIC for the first time. THEME 1: Parental reaction to CIC: Caregivers described benefits of an adjustment period on learning their child's need for CIC. Prenatal information to caregivers of spina bifida children gave them time to mentally process the need for CIC. THEME 2: Ease of learning CIC: impact of age and gender: caregivers identified advantages of initiating CIC in infancy. Caregivers speculated CIC was physically easier in boys than girls due to meatus location. Developmentally ready children expressed a desire for independence and privacy as they learned to initiate CIC. THEME 3: The impact of additional caregiver support in learning and performing CIC: presence of multiple caregivers optimized learning and implementation of CIC. Having secondary caregivers available provided peace of mind and more flexibility in maintaining reliable CIC care. Patients learning CIC found it helpful to have a parent present at the teaching session. Occasionally, female caregivers reported feelings of anger and frustration when male caregivers were reluctant to be involved in catheterization, irrespective of their child's gender. THEME 4: Satisfaction with healthcare team's approach: The healthcare team's responsiveness to their learning needs affected how they mastered CIC. The healthcare team's teaching and reassurance helped build caregiver confidence. Developmentally appropriate children were able to learn self-catheterization when supported by the healthcare team. Patients learning self-CIC articulated having a supportive healthcare team was helpful with implementation. THEME 5: Effect of CIC on employment status relative to job changes, insurance, and daycare: implementing and performing CIC presented a spectrum of issues related to employment. Educating employers regarding CIC facilitated a caregiver's ability to both remain at work and administer to their child. Caregivers underscored the importance of adequate insurance when considering employment choices. Concerns about daycare availability affected caregivers' work schedules.It is anticipated this information will aid healthcare personnel to more effectively teach and initiate CIC in families, and in individuals learning for the first time. The findings should serve as the basis for conducting future patient satisfaction studies, which would determine the effectiveness and reproducibility of these approaches.

Congenital anomalies of the kidneys and urinary tract (CAKUT) are the most common cause of chronic kidney disease among children and adults younger than 30 yr. In our previous study, whole-exome sequencing (WES) identified a known monogenic cause of isolated or syndromic CAKUT in 13% of families with CAKUT. However, WES has limitations and detection of copy number variations (CNV) is technically challenging, and CNVs causative of CAKUT have previously been detected in up to 16% of cases.To detect CNVs causing CAKUT in this WES cohort and increase the diagnostic yield.We performed a genome-wide single nucleotide polymorphism (SNP)-based CNV analysis on the same CAKUT cohort for whom WES was previously conducted.We evaluated and classified the CNVs using previously published predefined criteria.In a cohort of 170 CAKUT families, we detected a pathogenic CNV known to cause CAKUT in nine families (5.29%, 9/170). There were no competing variants on genome-wide CNV analysis or WES analysis. In addition, we identified novel likely pathogenic CNVs that may cause a CAKUT phenotype in three of the 170 families (1.76%).CNV analysis in this cohort of 170 CAKUT families previously examined via WES increased the rate of diagnosis of genetic causes of CAKUT from 13% on WES to 18% on WES + CNV analysis combined. We also identified three candidate loci that may potentially cause CAKUT.We conducted a genetics study on families with congenital anomalies of the kidney and urinary tract (CAKUT). We identified gene mutations that can explain CAKUT symptoms in 5.29% of the families, which increased the percentage of genetic causes of CAKUT to 18% from a previous study, so roughly one in five of our patients with CAKUT had a genetic cause. These analyses can help patients with CAKUT and their families in identifying a possible genetic cause.

A new strategy for the activation of glycosyl halide donors to be utilized in glycosylation reactions is presented, utilizing the ferrocenium (Fc) complexes [FcB(OH)2]SbF6 and FcBF4 as promoters. The scope of the new system has been investigated using glycosyl chloride and glycosyl fluoride donors in combination with common glycosyl acceptors, such as protected glucose. The corresponding glycosylation products were formed in 95 to 10% isolated yields with α/β ratios ranging from 1/1 to β only (2 to 14 h reaction time at room temperature, 40 to 100% ferrocenium promoter load).

Aims To investigate the effect of losartan on preventing bladder fibrosis and protecting renal function in rats with neurogenic paralysis bladder (NPB). Materials and methods Rats were assigned to the transecting spinal nerves group (TSNG), transecting spinal nerves + losartan group (LSTG), and control group (CG). On Day 32 postsurgery, bladder capacity (BC), bladder compliance (ΔC), bladder leakage pressure (Pves.leak) of TSNG and LSTG while BC, ΔC, and bladder threshold pressure (Pves.thre) of CG were measured by cystometry in each cohort. Renal function and the expression quantity of Angiotensin Ⅱ (Ang II) in blood were detected, in addition Ang II, Ang II Type 1 receptor (AT1), transformation growth factor β1 (TGFβ1), Collagen Ⅲ, and collagen fibrin in the bladder tissue were detected too. Results ΔC in TSNG and LSTG decreased significantly compared to the CG. Pves.leak in TSNG and LSTG were significantly higher than Pves.thre in CG. Renal function of both TSNG and LSTG decreased significantly compared with the CG, but renal function in LSTG was better than in TSNG. Ang Ⅱ in blood and bladder tissue in TSNG and LSTG increased significantly compared with CG. AT1 was expressed in the bladder tissue of all rats. The TGFβ1, Collagen Ⅲ, and collagen fibrin expression level increased significantly in TSNG compared with LSTG and CG, while these levels were not significantly different between CG and LSTG. Conclusion Losartan might prevent NPB fibrosis by stopping the upregulated signaling of Ang II/AT1/TGFβ1 and consequently may reduce kidney damage from occurring.

Ferrocenium catalysis is a vibrant research area, and an increasing number of ferrocenium-catalyzed processes have been reported in the recent years. However, the ferrocenium cation is not very stable in solution, which may potentially hamper catalytic applications. In an effort to stabilize ferrocenium-type architectures by inserting a bridge between the cyclopentadienyl rings, we investigated two ferrocenophanium (or ansa-ferrocenium) cations with respect to their stability and catalytic activity in propargylic substitution reactions. One of the ferrocenophanium complexes was characterized by single crystal X-ray diffraction. Cyclic voltammetry experiments of the ferrocenophane parent compounds were performed in the absence and presence of alcohol nucleophiles, and the stability of the cations in solution was judged based on the reversibility of the electron transfer. The experiments revealed a moderate stabilizing effect of the bridge, albeit the effect is not very pronounced or straightforward. Catalytic propargylic substitution test reactions revealed decreased activity of the ferrocenophanium cations compared to the ferrocenium cation. It appears that the somewhat stabilized ferrocenophanium cations show decreased catalytic activity.

Catalyst decomposition has a negative effect on catalytic activity, and knowledge of decomposition pathways can assist with catalyst development. Ferrocenium cations have been employed as catalysts in a number of organic transformations, and we investigated the stability of a number of ferrocenium salts in solution. The observed rate decomposition constants for [Fc]Cl, [Fc]PF6, [Fc]BF4, [Fc]CSA [Fc = ferrocenium, CSA = camphor-10-sulfonate (β)], [AcFc]SbF6, (AcFc = acetylated ferrocene), and [FcB(OH)2]SbF6 [FcB(OH)2 = ferrocenylboronic acid] were determined in CH2Cl2 solution by time-resolved UV-vis spectroscopy. The rate decomposition constants depended on the nature of the counterion, with [Fc]Cl being the most stable complex in solution. The decomposition rate constants dropped by roughly an order of magnitude in most cases when the experiments were performed in nitrogenated solvent, demonstrating that the decomposition is mainly an oxidative process. The cosolvent HFIP (1,1,1,3,3,3-hexafluoropropan-2-ol) slowed the decomposition of the ferrocenium cations as well. Many catalytic or stoichiometric reactions of ferrocenium cations are performed with alcohols; we determined that hexan-1-ol is decomposed over the course of 16 hours, but not oxidized in the presence of a ferrocenium cation. Finally, the different ferrocenium cations were employed in a test reaction to determine catalytic activity. The nucleophilic substitution of hydroxyl groups in a tertiary propargylic alcohol by an alcohol is catalyzed by all complexes, and, again, a counterion dependency of the catalytic activity was observed. Also, HFIP increases the catalytic activity of the ferrocenium cations. The research has importance in the development of ferrocenium-based catalyst systems, because changes in the counterion as well as the architecture of the ferrocenium cation have an influence on stability and catalytic activity.

Intensity mapping (IM) of spectral lines has the potential to revolutionize cosmology by increasing the total number of observed modes by several orders of magnitude compared to the cosmic microwave background (CMB) anisotropies. In this paper, we consider IM of neutral hydrogen (HI) in the redshift range $0 \lesssim z \lesssim 3$ employing a halo model approach where HI is assumed to follow the distribution of dark matter (DM) halos. If a portion of the DM is composed of ultralight axions then the abundance of halos is changed compared to cold dark matter below the axion Jeans mass. With fixed total HI density, $\Omega_{\rm HI}$, assumed to reside entirely in halos, this effect introduces a scale-independent increase in the HI power spectrum on scales above the axion Jeans scale, which our model predicts consistent with N-body simulations. Lighter axions introduce a scale-dependent feature even on linear scales due to its suppression of the matter power spectrum near the Jeans scale. We use the Fisher matrix formalism to forecast the ability of future HI surveys to constrain the axion fraction of DM and marginalize over astrophysical and model uncertainties. We find that a HIRAX-like survey is a very reliable IM survey configuration, being affected minimally by uncertainties due to non-linear scales, while the SKA1MID configuration is the most constraining as it is sensitive to non-linear scales. Including non-linear scales and combining a SKA1MID-like IM survey with the Simons Observatory CMB, the benchmark "fuzzy DM" model with $m_a = 10^{-22}\text{ eV}$ can be constrained at the 10% level. IntroductionThe objective was to report on the experience of the French Army Health Service in the management of blast injury of the hands related to warfare explosive devices.MethodsA retrospective study was conducted in the Percy Military Hospital (role 4 medical treatment facility) among French soldiers who presented with a combat-related blast injury of the hand between 2002 and 2018. The functional result was assessed by the disabilities of the arm, shoulder and hand (DASH) and the Orthotics and Prosthetics User Survey (OPUS, upper extremity functional status) scores. Proximal amputations (PAs) and distal amputations (DAs) were distinguished for the analysis.ResultsFifteen patients with a mean age of 31±8 years were included. They totalised 20 blasted hands. There were 16 traumatic amputations: 8 in each of the PA and DA groups. Twelve patients had additional injuries, four of which were polytraumatic. Skin closure time and flap use were higher in the DA group. Only one thumb reconstruction was performed. At a mean follow-up of 6.5±4 years, the number of amputees wearing a prosthesis was higher in the PA group. The mean DASH and OPUS scores were 35.5%±24.0% and 64.0%±19.0%, respectively, with no difference between the two groups.ConclusionThe severity of hand blasts related to warfare explosive devices requires the systematic application of damage control surgery. PAs are frequent and secondary reconstruction options are limited. The functional result is poor and similar between proximal and distal amputees.

Two sets of macrocyclic, bio-inspired, non-heme ligands are utilized for the synthesis of NiII, PdII and PtII complexes. The ligands consist of a 16-atom macrocycle, formed by four methylene bridged NHC moieties, with imidazole or benzimidazole as building blocks. The complexes exhibit a square planar coordination geometry and are characterized by NMR, ESI-MS, elemental analysis, SC-XRD and UV/Vis. For complexes incorporating benzimidazole, an evaluation of luminescence properties is performed, and is found that phosphorescence is present for the PdII derivative and there is fluorescence for the PtII derivative. Stability studies in cell culture medium are performed for subsequent MTT assays. Here, the NiII complexes show low to no activity, and PdII and PtII complexes exhibit remarkable low IC50 values in cisplatin resistant A2780cisR cells.

Carbohydrates are a large class of natural products that play key roles in a number of biological processes such as in cellular communication or disease progression. Carbohydrates are also used as vaccines and pharmaceuticals. Their synthesis through glycosylation reactions is challenging, and often stoichiometric amounts of promoters are required. Transition metal catalyzed glycosylation reactions are far less common, but can have advantages with respect to reaction conditions and selectivity. The review intends to approach the topic from the catalysis and carbohydrate perspective to encourage researchers from both the fields to perform research in the area. The article covers the basics in glycosylation and catalysis chemistry. The catalysts for the reaction can be roughly divided into two groups. In one group, the catalysts serve as Lewis acids. In the other group, the catalysts play a higher sophisticated role, are involved in all elementary steps of the mechanism and remain coordinated to the substrate throughout the whole catalytic cycle. Based on selected examples, the main trends in transition metal catalyzed glycosylation reactions are explained. Lewis acid catalysts tend to require a somewhat higher catalyst load compared to other organometallic catalysts. The reaction conditions such as the temperature and time depend in many cases on the leaving group employed. An outlook is also presented. The article is not meant to be comprehensive; it outlines the most common transition metal catalyzed processes with the intention to bring the catalysis and carbohydrate communities together and to inspire research activities in both areas.

Congenital anomalies of the kidney and urinary tract (CAKUT) are the most common cause of chronic kidney disease in the first three decades of life, and in utero obstruction to urine flow is a frequent cause of secondary upper urinary tract malformations. Here, using whole-exome sequencing, we identified three different biallelic mutations in CHRNA3, which encodes the α3 subunit of the nicotinic acetylcholine receptor, in five affected individuals from three unrelated families with functional lower urinary tract obstruction and secondary CAKUT. Four individuals from two families have additional dysautonomic features, including impaired pupillary light reflexes. Functional studies in vitro demonstrated that the mutant nicotinic acetylcholine receptors were unable to generate current following stimulation with acetylcholine. Moreover, the truncating mutations p.Thr337Asnfs(∗)81 and p.Ser340(∗) led to impaired plasma membrane localization of CHRNA3. Although the importance of acetylcholine signaling in normal bladder function has been recognized, we demonstrate for the first time that mutations in CHRNA3 can cause bladder dysfunction, urinary tract malformations, and dysautonomia. These data point to a pathophysiologic sequence by which monogenic mutations in genes that regulate bladder innervation may secondarily cause CAKUT.

A manatee’s primary modality to detect a vessel on a possible collision course is hearing as underwater visibility is limited in many manatee habitats and their visual acuity is poor. We estimate a Florida manatee’s ability to detect the sound of an approaching boat and vocalizations in four different soundscapes in Sarasota Bay, FL. Background noise samples were collected every 5 minutes for a two-week period during winter and summer at each location (2019 or 2020). Sound levels in third octave bands (0.5, 1, 2, 4, and 8 kHz) were measured and compared to manatee auditory hearing thresholds and to sound levels of an approaching boat traveling at a slow, medium, or fast speed. Background sound levels in a wider band (1–20 kHz) were calculated to model vocal communication space at each location. We found that a manatee’s estimated ability to detect an approaching boat differs greatly among locations, with time of day, and by season, and that fast boats are predicted to be detected later than slow boats. Latency of boat noise detection is estimated to sharply increase when considering unusually loud background noise levels. We suggest that such uncommonly loud conditions (e.g. 95th percentile sound level), not just typical conditions (median sound level), are important to consider for understanding the problem of manatee-boat collisions. Additionally, background noise impacts estimated vocal communication space and may limit the ability of vocal-mediated mother-calf cohesion. Altogether, a manatee’s ability to detect acoustic signals of interest is expected to vary greatly spatially and temporally.

Laborpraktika stellen in naturwissenschaftlichen Fächern einen elementaren Bestandteil des Curriculums dar. Die Studierenden erlernen in diesem Lehr-Lernformat das universitäre Experimentieren. In diesem Beitrag werden zwei Beispiele vorgestellt, wie konventionelle Laborpraktika mit eLearning-Angeboten ergänzt werden können. Auf Basis der Evaluation werden die Potenziale und Grenzen der unterschiedlichen eLearning-Bestandteile vor dem Hintergrund aktueller Entwicklungen digitaler Experimentierumgebungen sowie der Lernwirksamkeit analysiert. Basierend auf den Erkenntnissen werden Empfehlungen zur Weiterentwicklung von Laborpraktika mittels Augmented bzw. Virtual Reality abgeleitet.

Purpose Congenital anomalies of the kidneys and urinary tract (CAKUT) constitute the leading cause of chronic kidney disease in children. In total, 174 monogenic causes of isolated or syndromic CAKUT are known. However, syndromic features may be overlooked when the initial clinical diagnosis of CAKUT is made. We hypothesized that the yield of a molecular genetic diagnosis by exome sequencing (ES) can be increased by applying reverse phenotyping, by re-examining the case for signs/symptoms of the suspected clinical syndrome that results from the genetic variant detected by ES. Methods We conducted ES in an international cohort of 731 unrelated families with CAKUT. We evaluated ES data for variants in 174 genes, in which variants are known to cause isolated or syndromic CAKUT. In cases in which ES suggested a previously unreported syndromic phenotype, we conducted reverse phenotyping. Results In 83 of 731 (11.4%) families, we detected a likely CAKUT-causing genetic variant consistent with an isolated or syndromic CAKUT phenotype. In 19 of these 83 families (22.9%), reverse phenotyping yielded syndromic clinical findings, thereby strengthening the genotype–phenotype correlation. Conclusion We conclude that employing reverse phenotyping in the evaluation of syndromic CAKUT genes by ES provides an important tool to facilitate molecular genetic diagnostics in CAKUT.

Although rhenium (Re) is a rare element, it is an economically important one due to its use as a catalyst in the chemical preparation of gasoline and in technologically vital alloys. While the cytotoxicity of specific rhenium compounds has been tested against various cell lines, there is a dearth of information about the toxicity of Re-compounds, since Re is considered a nontoxic element. However, recent studies with Re-compounds as anticancer agents have demonstrated that some these compounds are effective in combating cancer, and therefore it is important to examine the toxicity of Re-compounds in the environment. In addition, information regarding speciation of Re-compounds, the assays involved in testing the toxicity of the compounds, and the available biological studies are provided. Re has been used in biological studies as a radiotracer, due in part to the ease in producing 188Re and recently 186Re. Re has some properties that are similar to technetium and is being used with increasing frequency to replace technetium (Tc) or in conjunction with Tc. This review describes the toxicity of Re materials reported in the literature. In addition, these reported toxicity studies are placed in the context of ecosystems showing ecotoxicity, as well as describing speciation and subsequently toxicity of various species in a variety of cytotoxicity assays.

This review focuses on cytotoxic rhenium compounds in terms of IC50 values, their mode of action in biological systems and their status in clinical trials. Biological studies of different rhenium compounds ordered by the oxidation state of rhenium are presented. Numerous rhenium complexes are reported, with the greatest number of compounds containing a Re(I)(CO)3+ core. A wide range of complexes has been designed using a combination of organometallic ligands, N- or S-based ligands, peptides, multidentate ligands and oxo groups. Design concepts based on membrane permeability and lipophilicity, membrane receptor targets and specific enzyme targets are presented. The cytotoxicity parameter IC50 is shown for organometallic compounds, coordination complexes, clusters and Re(oxo) complexes. In addition, a brief summary of in vivo studies is given. A further summary of rhenium compounds subjected to clinical trials is presented to provide information about the classes of rhenium compounds that have been tested in human beings and the approaches used in these studies. Moreover, the comparability of the IC50 values among the cytotoxicity studies is critically assessed to provide the basis for a summary of the most potent rhenium compounds according to their reported IC50 values for each type of cancer. The summary of the structures for the most cytotoxic complexes allows the identification of structural similarities and basic features that could lead to their cytotoxicity and might be useful for future investigations. Finally, the information from the analysis of the rhenium compounds subjected to cellular studies is compared to data on the rhenium compounds that have been involved in in vivo evaluation and clinical trials.

The synthesis and characterization of a novel macrocyclic Au(III) N-heterocyclic carbene (NHC) imidazolyl complex, a novel macrocyclic tetra-NHC benzimidazole ligand, and the corresponding Ag(I) and Au(I) complexes are presented. Single-crystal X-ray diffraction analysis of the Au(I) benzimidazolyl complex 3 reveals an unusual structure, differing from the respective Au(I) imidazolyl complex 4. Both complexes have a Au4L2 composition; however, 3 has two C–Au(I)–C units acting as a connection between the two ligands with two Au(I) atoms being linearly coordinated inside the cavity of the macrocyclic ligand. In the case of complex 4, the structure shows a box-type coordination with all four Au(I) atoms being located between the two ligands. Stability studies in cell culture medium are performed for subsequent MTT assays and they show an unprecedented proton-to-deuterium exchange of the methylene bridge of the Au(III) imidazolyl complex. In MTT assays, the tetranuclear acyclic Au(I) complex 5 displays the lowest IC50 values in MCF-7, PC3, and A2780cisR cells with a selective cytotoxicity for MCF-7 and A2780cisR cells.

Human studies have shown loss of diversity of the gut microbiome following hematopoietic stem cell transplantation (HSCT) in association with significant gut injury caused by the preparative regimen. Prolonged antibiotic use worsens loss of microbiome diversity and increases risk of complications such as graft-versus-host disease (GVHD). Our data support the hypothesis that loss of intestinal commensals that produce short-chain fatty acids (SCFAs) may increase dysbiosis. Here, we report an extensive longitudinal examination of changes in the luminal SCFAs in children undergoing allogeneic HSCT, and the relationship of those changes to the microbiota and antibiotic exposure. We found significant and progressive alterations in butyrate, and in additional SCFAs in stool in the first 14 days after transplant, a finding not observed in published mouse studies. SCFA levels were lower in children receiving antibiotics with activity against anaerobic organisms. Moreover, day 14 post-HSCT butyrate and propionate levels are lower in children who went on to develop GVHD, although our disease population was small. These data provide insight into the mechanism of prior observations that loss of diversity and increased antibiotic use are associated with GVHD following HSCT. Our findings offer potential modifiable targets to reduce risk of GVHD and improve survival after HSCT.

Learning about the nature of science is an important aspect of becoming part of the culture of practicing physicists. While these topics have a long-standing tradition in U.S. curricula they are currently not part of German educational standards or curricula. And while there is some research concerning primary and secondary school students’ beliefs in this area, there are hardly any data about German university students. In this study we query 100 physics students in their first to fifth semester at a German university for their views of nature of science. We then compare them to a sample of physics professors at German universities from an earlier study. The students exhibit quite adequate views in some areas but could improve on their knowledge of the variability of scientific methods and the tentativeness and limited certainty of scientific knowledge—two issues they have in common with the professors. If we interpret the sample longitudinally, we see positive development in the first but worsening in the latter area.
