In the current study, bismuth ferrite nano-sorbent was synthesized and utilized as a sorbent for the dispersive solid-phase extraction of methylprednisolone from exhaled breath samples.

Diethylene glycol monoethyl ether (DEGME) is a hydroalcoholic solvent that gained tremendous attention in the cosmetics, food, nanoformulations, and pharmaceutical industries. Due to its physicochemical features, it has been widely used as a penetration enhancer, surfactant, and solubilizer. Among numerous tradenames defined for DEGME -- Carbitol® (by Dow Chemical Co., USA), and Transcutol® HP, CG, and P. (by Gattefossé Co., France) -- are known to be employed in pharmaceutical industries. Transcutol® CG is utilized only in cosmetics; however, Transcutol® P and Carbitol® are both used in various pharmaceutical topical dosage forms such as creams, gels, etc. Additionally, Transcutol® HP is used in all administration routes. In view of this, the application of DEGME is highlighted in the areas of industry and pharmaceutical sciences. Moreover, in this review the prominent characteristics, pharmacokinetics, and toxicity of DEGME are examined and it is suggested that DEGME is a promising solvent/solubilizer with comparable assignments to other conventional excipients.

Mesalazine is a low-permeable and low-soluble drug, which makes it a class IV drug in the Biopharmaceutics Classification System. Hence, its solubilization can be helpful for various stages of formulation development. The purpose of this study was to investigate the solubilization manner and thermodynamics of mesalazine in ternary solvent combinations of {ethanol (1) + propylene glycol (2) + water (3)} using the shake-flask technique at (298.2-313.2) K. In the following, the mathematical representation of the acquired solubility data using some popular models was evaluated. The accuracies of the applied models were described by percentages of mean relative deviation (MRD%). Based on obtained results (MRD%10.0), it can be concluded that the trained models can adequately predict the solubility of mesalazine in the investigated ternary solvent combinations. The findings also revealed that the solution composition and temperatures greatly influence the solubility of mesalazine. In addition, the thermodynamic characteristics of the mesalazine dissolution process indicate that the mesalazine dissolution process is endothermic and entropy-driven. The generating data in the current work also expands the available solubility database for mesalazine in the solvent mixtures.

Serum levels of inflammatory cytokines and uremic toxins, and their inter-correlations with the diversity of Bacteroidaceae, Bifidobacteriaceae, Prevotellaceae and Lactobacillaceae families in intestinal microbiota were investigated in patients with end stage renal disease (ESRD).Stool and blood samples from 20 ESRD patients on maintenance hemodialysis were collected. DNA genome of the bacterial composition of the stool samples was extracted and evaluated by the sequencing analysis of 16S rRNA genes. Serum levels of inflammatory cytokines and uremic toxins were then analyzed.The mean serum concentrations of TNF-α, IL-6, indoxyl sulfate (IS) and p-cresol (PC) were 305.99 ​± ​12.03 ​ng/L, 159.95 ​± ​64.22 ​ng/L, 36.76 ​± ​5.09 ​μg/mL and 0.39 ​± ​0.15 ​μg/mL, respectively. The most significant positive correlation was observed between Prevotellaceae family and total antioxidant capacity (TAC), Lactobacilli species and CRP and PC, as well as Scardovia wiggsiae and IS (p ​ ​0.001). A negative correlation was also found between Bacteroides clarus and PC. Patients with ESRD on maintenance hemodialysis had elevated levels of PC and IS and increased levels of the inflammatory markers. The most positive correlation was found between microbiota and CRP and PC, while the most negative one was between microbiota and IL-1 and TAC.The abundance and diversity of Bacteroidaceae, Bifidobacteriaceae, Prevotellaceae and Lactobacillaceae families and their correlations with clinical parameters could provide benefits in the ESRD patients but they could not promote the symptoms.

In the present study, the solubility values of amlodipine besylate (AMB) in the binary mixtures of polyethylene glycol 400 and ethanol were determined using the shake-flask method at 293.2 to 313.2 K under atmospheric pressure (≈ 85 kPa) and correlated to some cosolvency and activity coefficient models. The accuracy of each model was marked by the mean relative deviations (MRD%) of the back-calculated solubility data. Also, the apparent thermodynamic properties of AMB dissolution were computed by the van't Hoff and Gibbs equations.

This work aims to obtain the solubility, density and thermodynamic parameters of deferiprone in propylene glycol and ethanol. For this purpose, a shake-flask technique was applied for solid–liquid equilibration and the spectrophotometry method was employed for solubility measurement. Solubility and density of deferiprone in non-aqueous mixtures of propylene glycol and ethanol were measured in the temperatures 293.2–313.2 K. Some equations including van’t Hoff, the Jouyban-Acree, the Jouyban-Acree-van’t Hoff, the mixture response surface and modified Wilson equations were used for the mathematical data modeling. The apparent thermodynamic parameters of the deferiprone dissolution process were computed and reported.

Aim: To report a correlative model to calculate the solubility of ketoprofen in mono-solvents at various temperatures. Methodology: Previously reported solubility data of ketoprofen in a number of mono-solvents at various temperatures are re-analyzed using a recently developed model employing Abraham, Hansen and Catalan parameters as input values. The accuracy of the model is evaluated by computing the standard deviation of residuals (SDRs) and compared with those of previous models. Results: The new model provided very accurate correlation for the solubility of ketoprofen in the investigated mono-solvents at various temperatures. The obtained SDR is 2.20 where the SDRs of previously reported models are 1.54 to 2.56. The new model correlates whole solubility data using a single model, whereas other models correlate the solubility data in each mono-solvent using a separate set of model constants. Conclusion: The trained model provided reasonably accurate results for ketoprofen by using Abraham, Hansen and Catalan parameters of the mono-solvents as input values.

Aim: To develop a one-step vortex-assisted liquid-liquid extraction (VALLE) method, without the need for evaporation and reconstitution steps, to establish a rapid and straightforward treatment procedure based on capillary electrophoresis-diode array detection (CE-DAD) for the determination of vanillylmandelic acid (VMA) in human urine. Methodology: Optimization of VALLE and CE-DAD procedures were studied in detail. The effects of various experimental parameters, such as the type of the extraction solvent, sample pH, salt addition, and extraction time were investigated. Also, CE separation conditions including background electrolyte type, concentration, and pH, injection time and separation voltage were optimized as well. Results: A successful separation of VMA was achieved in less than 6 min using a basic background electrolyte composed of 60 mmol·L-1 acetate/ACN (acetonitrile) 50% (v/v) (final apparent pH is 5.73). The linear response was obtained over the concentration range from 1.0 to 14 μg·mL-1. The limit of detection and quantification were 0.30 and 1.0 μg·mL-1, respectively. The intra- and inter-day precisions were found to be less than 4.3%. The extraction recoveries of VMA were between 95%-97%. Conclusion: The developed method is found to be a simple, rapid, and reliable method for quantitative analysis of urinary VMA.

Background: There is only limited data for solubility of codeine phosphate in binary systems available, which comes with uncertainties about the prediction accuracy of common thermodynamic models. Methods: This study investigated the codeine phosphate dissolution in N-methyl-2-pyrrolidone (NMP) and ethanol system using shake-flask method and mathematically described generated data by different thermodynamic models. The density as another property were also determined and fitted to results of the Jouyban-Acree equation. The mean relative deviations were obtained for confirming the model’s accuracy. Moreover, ,, and of the dissolution of codeine phosphate in the NMP and ethanol system were calculated using the desired equations at Thm. Results: The dissolution process of codeine phosphate was identified as endotherm, the solubility in the binary mixtures was best at higher mass fractions of NMP and finally, the model predictions were deemed as excellent based on a mean relative deviation that was generally below eight percent. Conclusion: The results of this study could expand the available solubility database for codeine phosphate.

The exhaled breath condensate (EBC) presents a simple and non-invasive alternative approach for bioequivalence assessments and therapeutic drug monitoring of inhaled drugs. EBC better represents the drug at the site of action and eliminates the possibility of the contribution of a swallowed portion of the dose when systemic bioavailability is used for assessment. This review summarizes the recently reported analytical methods for the quantification of drugs in EBC. It also discusses the difficulties in the bioequivalence evaluation criteria of generic orally inhaled drug products suggested by various regulatory agencies that may be eliminated using the EBC analysis approach.

This study presents solubility determination and the thermodynamic analysis of celecoxib (CXB) in two kinds of solvent mixtures of N-methyl-2-pyrrolidone (NMP) + ethanol and NMP + 1-propanol at various temperatures (293.15–313.15 K). The density of CXB-free mixtures of NMP + 1-propanol at various temperatures was also measured since the data was not available from the literature. In both systems, the solubility raised with temperature and increased with increasing the amount of NMP in the blends. Measured solubility data were fitted using four mathematical cosolvency models. The resulting MRD% values for all examined models were less than 6.0. The measured densities of the CXB saturated solutions were calculated using an adopted version of Jouyban-Acree model. Further, predictive versions of the model which were trained using the density of drug-free mixed solvents were also used to predict the density of CXB saturated solutions with an overall MRD% of 1.3.

Background: To discover the optimal solvent amounts for using in a particular application, it is vital to achieve some useful information in regard with suitable neat or mixed solvent and drugs equilibrium solubility in them. It is known that the low solubility of drugs such as bosentan (BST) in water negatively effects in vitro and in vivo kinetics of dissolution, affecting in turn its bioavailability along with making several difficulties around designing its liquid formulations. Methods: Solubility of BST in some mixtures of polyethylene glycol 400 (PEG 400) and water was experimentally determined at T = (293.15 to 313.15) K by using a common shake-flask technique followed by UV-visible spectroscopic method. The experimental solubility data in PEG 400 mass fraction (w1) of 0.0 to 1.0 at 298.15 K and in w1=0.0, 0.5 and 1.0 at other temperatures were then correlated by cosolvency models including the Jouyban-Acree, the Jouyban-Acree-van’t Hoff, and the double log-log models and some un-measured solubility data were predicted based on the obtained trained models. Results: The results presented that the aqueous solubility of BST is increased by increasing mass fraction of PEG 400 as well as increasing temperature and reached the maximum value in neat PEG 400 at 313.15 K. Conclusion: The BST solubility in water improved by addition of PEG 400 into it. According to the average relative deviations obtained from the back-computed data with trained models which were < 8.0%, it concluded that the selected models were able to predict the un-measured data with high reliability.

The solubility of 6-methylcoumarin in water, ethanol and nine aqueous ethanol mixtures was determined from 293.15 to 323.15 K. 6-Methylcoumarin solubility expressed in mole fraction was correlated with some well-known correlation/prediction models. Gibbs energy, enthalpy and entropy, for dissolution and mixing processes, were computed using van't Hoff and Gibbs equations. The plot of enthalpy vs. Gibbs energy of dissolution exhibited negative slopes from water to w(1) = 0.20 and from w(1) = 0.90 to ethanol but positive slopes in the interval 0.20 w(1) w(1) = 0.20 is entropy-driven that could be attributed to water molecules releasing originally bounded as 'icebergs' around the drug-non-polar groups. In mixtures of 0.20 w(1) < 0.90, enthalpy-driving mechanism for drug transfer from more-polar to less-polar systems is observed, probably owing to better solvation by ethanol. Inverse Kirkwood-Buff integrals show that 6-methylcoumarin is preferentially solvated by water in water-rich mixtures but preferentially solvated by ethanol in 0.24 x(1) < 1.00.

In this study, gold nanoparticles (AuNPs) modified with β-cyclodextrin (β-CD) were used to assist with electromembrane extraction (EME) and were coupled with capillary electrophoresis (CE) and ultraviolet (UV) detection (CE-UV) for the extraction and measurement of methadone from plasma samples. A β-CD-modified AuNP-reinforced hollow fiber (HF) was utilized in this work. The β-CD-modified AuNPs act as an absorbent and provide an extra pathway for the analyte extraction. For obtaining the effect of the presence of β-CD-modified AuNPs in the HF pores, the extraction efficiency of the EME and β-CD-modified AuNPs/EME techniques were compared. Different parameters influencing the extraction efficacy of the EME and β-CD-modified AuNPs/EME methods were optimized. Optimal extractions were performed with 1-octanol as the organic solvent in the supported liquid membrane (SLM), with an applied voltage of 10 V as the driving force across the SLM, and with pH 7.0 in the donor solutions with a stirring speed of 1000 rpm after 20 min and 25 min for the β-CD-modified AuNPs/EME and EME methods, respectively. Under optimal conditions, compared with the EME method, the β-CD-modified AuNPs/EME method exhibited increased extraction efficacy in a short time.
