Climate change is strongly influencing regions of Antarctica but the consequences on microevolutionary processes have been little studied. Patterns of population genetic diversity were analysed in the Antarctic bivalve Nuculana inaequisculpta (Protobranchia: Nuculanidae) from a fjord with 70 years of documented climate-forced glacier retreat. Thirty-nine individuals from five sites at different distances from the glacier terminus were collected, and the COI gene was sequenced from each individual. No statistically significant genetic differentiation was found between sites nor a significant correlation between the proximity of glaciers and genetic diversity, suggesting a high dispersal capability and therefore, a planktonic larval stage for this species. Nevertheless, we encourage increasing the sample size and number of loci in future studies to confirm our findings.

Diet of the Tristan spiny lobster Jasus paulensis was examined using gut content and Stable Isotope Analyses (SIA) to assess whether diets differed among three islands in the Tristan da Cunha archipelago (Inaccessible, Nightingale and Tristan islands), and between small and large lobsters at two depth intervals. Gut fullness was significantly less at Nightingale Island, suggesting overall less food there. Diet differed significantly among all islands, depths and sizes, although multidimensional scaling (MDS) and constrained canonical analysis of principle coordinates (CAP) plots showed considerable overlap and variability in diet. Lobster diet from Tristan Island showed a greater within-island similarity and was significantly less diverse, suggesting a more impoverished reef and narrower feeding niche, while dietary differences between size and depth reflected prey availability. Carbon and nitrogen stable isotope signatures differed among the islands, and carbon signatures between depths. Jasus paulensis appeared to be an opportunistic omnivore, with diet reflecting impoverished reef habitats. In addition, our data showed no evidence of soya remnants in the gut contents or tissue signatures of lobsters at Nightingale Island, following a soya spill there four years previously. However, gut fullness was less at Nightingale. This may have increased the attractiveness of baited traps, possibly explaining an observed rise in CPUE after the spill.

One of the paradigms underlying sandy beach ecology is the overriding control by physical processes; thus, biological interactions (i.e. food availability, competition and predation) are believed to play a role structuring macrofaunal communities only in benign habitats such as dissipative beaches. Moreover, sandy beaches are characterized by low in-situ productivity, so their food webs rely heavily on marine inputs. Studies have shown that estuarine organic matter plays a key role in influencing the dynamics of marine ecosystems. However, very few studies have tested the role of estuarine input on sandy beaches. Here, we aim to determine the impact of estuarine input on the food web of a sandy beach macrobenthic community. To this end, particulate organic matter (POM) samples from the marine environment and the estuary, as well as macrobenthic samples from the beach, were analysed for their stable isotope (SI) signature. Our results indicated that the POM SI signatures were not different along the beach, but differences were recorded between marine and estuarine sources. Bayesian mixing models indicated that the organisms did not make use of the estuarine POM at the beginning of the wet season, but relied more heavily on this resource towards the end of the wet season. This leads to the conclusion that changes in estuarine flow throughout the wet season can impact the trophic structure of macrobenthos communities, confirming a link between lotic and marine communities. Moreover, SI signatures suggest that the species collected here exhibit overlapping trophic niches, indicating high level of inter-specific competition. This highlights that species in low-productivity areas, such as the one studied here, can experience high levels of competition even in physically controlled environments such as sandy beaches.

The seismic stratigraphy, architecture and evolution of a large subtropical embayment, Maputo Bay, Southern Mozambique is described. Nine seismic units are documented; and show repeated phases of incised valley formation and transgressive infilling. Trangressive infilling alternates between the classic wave-dominated transgressive succession of fluvial and central estuarine bay deposits and the filling by intertidal flat and tidal bedforms developed during stages of inlet restriction and shallowing associated with barrier growth at stillstand intervals. Imprinted on the succession are several key facets of subtropical stratigraphy, hurricane-generated erosional surfaces and beachrock horizons. The upper stratigraphy comprises the highstand bayhead delta, a prograding feature constructed by several flood events. In comparison with literature from other similar systems such as lagoons, coastal lakes and estuaries, a generic three stage stratigraphic model of large marine embayments is proposed that includes incision during glaciations; transgressive infilling during deglaciations; and shallowing during the stillstand episodes of the recent deglaciation.

Rationale Chemical lipid extractions, as means of standardizing sample preparations, have been identified as important for comparability of studies. Unfortunately, these methods are expensive, because of the costly chemicals and the need to analyse two sets of samples, one for δ(13) C values (treated) and another for δ(15) N values (untreated). To avoid this, studies have suggested mathematical solutions to the problem. Our study intends to (i) determine the applicability of the five most common mathematical correction models and (ii) which of the widely applied chemical extraction methods is the most suitable for a variety of marine organisms. Methods Muscle, heart and liver samples were collected from eight different species. The tissues were treated with Bligh and Dyer, Folch and Soxhlet extraction methods and analysed in a Europa 20-20 mass spectrometer. Predicted lipid-extracted δ(13) C values were calculated from untreated tissue values using the five most common mathematical models. Results The results indicated that the mathematical methods could not be accurately applied to any of the eight species used in this study, highlighting current issues with accepted isotope methodologies. The Folch chemical extraction removed the highest amount of lipid, suggesting it is the most suitable delipidation method. Conclusions Analysing two samples, one treated one not, remains the best method to obtain accurate δ(13) C isotope values of muscle tissue. By using this approach every study will obtain two datasets, eventually providing a suitable collective dataset for determining how isotopic signatures are affected by delipidation and potentially producing better mathematical correction models in future.

Eddies are important not just because they have momentum and can transfer water properties, but also because they play an important role in increasing the trophic energy available to organisms, thereby enhancing primary productivity. This study was conducted to test the ‘suitcase hypothesis’ – the inclusion of organic matter (OM), plankton and larvae within the single water mass of an eddy, which are then highly conserved and transported to another location. Here we hypothesize that particulate OM (POM) and zooplankton from the continental shelf of Madagascar become trapped as the eddy forms and are transported westwards. We analysed stable isotope signatures of POM and zooplankton from samples collected from the continental shelf (CS) as well as within an eddy that had recently formed off the south-west coast of Madagascar. There was no statistical difference in the POM isotopic signature or C/N ratios between the continental shelf or any of the eddy regions. However, some C/N ratio values could suggest that some OM from close to the coastal region could have been transported by the eddy. This was not the case for zooplankton, however, with isotopic signatures on the CS greatly differing from those in the eddy. Several factors could account for these differences including feeding behaviour or tissue turnover rate. We conclude that our study did not gather enough evidence to support (or reject) the ‘suitcase’ hypothesis, and further studies are needed in order to fully understand the transport of material, including zooplankton and larvae, by eddies.

The South African coast contains abundant estuaries and lagoons, most of which originated as river valleys incised during Quaternary sea-level fall and subsequently drowned and/or infilled during rising interglacial sea levels. This chapter discusses these changes and highlights the geomorphological and sedimentological evolution of several southern African estuaries during the Pleistocene to present. The development, infilling and positioning of incised valley systems is mainly controlled by sea-level variation as well as fluvial and marine sediment supply. Most contemporary estuaries in southern Africa show dramatic responses to shorter term sea-level fluctuations and sediment supply during the Holocene. Barring anthropogenic interference, the dynamic behaviour of estuaries, dictated by their transient position and geomorphic character within an incised valley, is considered to fluctuate on centennial to millennial timescales.

Stable isotope measurements are an important tool for ecosystem trophic linkage studies. Ideally, fresh samples should be used for isotopic analysis, but in many cases organisms must be preserved and analysed later. In some cases dyes must be used to help distinguish organisms from detritus. Since preservatives and dyes are carbon-based, their addition could influence isotopic readings. This study aims to improve understanding of the effects of sample storage method, dye addition and acidification on the δ(15)N and δ(13)C values of zooplankton (Euphasia frigida and Undinula vulgaris). Zooplankton was collected and preserved by freezing, or by the addition of 5% formalin, 70% ethanol, or 5% formalin with added Phloxine B or Rose Bengal, and stored for 1 month before processing. Samples in 5% formalin and 70% ethanol were also kept and processed after 3 and 9 months to study changes over time. Formalin caused the largest enrichment for δ(13)C and a slight enrichment for δ(15)N, while ethanol produced a slight depletion for δ(13)C, and different effects on δ(15)N depending on the species. In formalin, dyes depleted the δ(13)C values, but had variable effects on δ(15)N, relative to formalin alone. Acidification had no significant effect on δ(15)N or δ(13)C for either species. Long-term storage showed that the effects of the preservatives were species-dependent. Although the effects on δ(15)N varied, a relative enrichment in (13)C of samples occurred with time. This can have important consequences for the understanding of the organic flow within a food web and for trophic studies. .

In an optical trap, micron-sized dielectric particles are held by a tightly focused laser beam. The optical force on the particle is composed of an attractive gradient force and a destabilizing scattering force. We hypothesized that using anti-reflection-coated microspheres would reduce scattering and lead to stronger trapping. We found that homogeneous silica and polystyrene microspheres had a sharp maximum trap stiffness at a diameter of around 800 nm—the trapping laser wavelength in water—and that a silica coating on a polystyrene microsphere was a substantial improvement for larger diameters. In addition, we noticed that homogeneous spheres of a correct size demonstrated anti-reflective properties. Our results quantitatively agreed with Mie scattering calculations and serve as a proof of principle. We used a DNA stretching experiment to confirm the large linear range in detection and force of the coated microspheres and performed a high-force motor protein assay. These measurements show that the surfaces of the coated microspheres are compatible with biophysical assays.

The international literature highlights the importance of understanding the role and fate of terrestrial organic matter (OM) in the marine system, yet little is known about this on the oligotrophic east coast of South Africa. Within the Bight, three important processes make OM biologically available: (1) a topographically induced upwelling cell (north), (2) a Cyclonic Eddy (south), and (3) riverine input. We hypothesise that: (i) these OM sources differ substantially in their isotope ratios, and (ii) zooplankton at the inshore region has an isotope signature linked to localised fluvial fluxes. Particulate OM samples were collected at five locations along the Bight and in three main estuaries during austral summer and winter seasons in 2010 and subjected to isotopic and mixing model analysis. Riverine input played an important role in this ecosystem inshore during the wet season. Only marine OM played a role as a biological driver offshore and at all sites during the dry season. Although there were indications that the upwelling cell did occur, it was not confirmed as active in either season. The current study provides valuable insights into the Bight's biological functioning and principle insights into the fate of terrestrial organic carbon on the east coast of southern Africa.
