Core decompression (CD) is beneficial in the early stage of osteonecrosis of the femoral head (ONFH). Adjunctive bone marrow derived cell therapies (BMDCT) have been advocated which potentially aid the regenerative process.This study was conducted to determine potential benefit of CD + BMDCT in ONFH, in terms of disease progression, conversion to arthroplasty (primary outcomes), and functional outcomes and complication rates (secondary outcomes).A systematic review of literature was performed on 3 databases. Studies reporting CD + BMDCT (intralesional instillation) in ONFH, with a minimum follow up of 1 year and reporting the pre-defined outcome measures were included in the review. Meta-analysis consisted of two different arms: a comparative arm, to compare CD + BMDCT to CD alone, and a non-comparative meta-analysis arm, to determine pooled rates of disease progression, conversion to arthroplasty and complication rates.A total of 18 studies were included in the systematic review. CD + BMDCT had lower rates of disease progression (OR 0.19 [95% CI, 0.09, 0.40]) and conversion to arthroplasty (OR 0.20 [95% CI, 0.11, 0.40]) as compared to CD alone. Functional score (MD = -7.07 [95% CI, -12.28, -1.86]) and visual analog scale also showed better improvement with the use of CD + BMAC (MD = -10.39 [95% CI, -12.87, -7.90]). Increasing age and post-collapse stage at presentation were noted to have an adverse effect on the outcomes.CD + BMDCT was found to decrease disease progression and conversion to arthroplasty, and was noted to have better functional outcome scores as compared to CD alone.

Background: Alzheimer’s disease (AD) is the most common type of neurodegenerative dementia affecting people in their later years of life. The AD prevalence rate has significantly increased due to a lack of early detection technology and low therapeutic efficacy. Despite recent scientific advances, some aspects of AD pathological targets still require special attention. Certain traditionally consumed phytocompounds have been used for thousands of years to treat such pathologies. The standard extract of Gingko biloba (EGB761) is a combination of 13 macro phyto-compounds and various other micro phytocompounds that have shown greater therapeutic potential against the pathology of AD. Objective: Strong physiological evidence of cognitive health preservation has been observed in elderly people who keep an active lifestyle. According to some theories, consuming certain medicinal extracts helps build cognitive reserve. We outline the research employing EGB761 as a dual target for AD. Methods: This study investigates various inhibitory targets against AD using computational approaches such as molecular docking, network pharmacology, ADMET (full form), and bioactivity prediction of the selected compounds. Results: After interaction studies were done for all the phytoconstituents of EGB761, it was concluded that all four of the phytocompounds (kaempferol, isorhamnetin, quercetin, and ginkgotoxin) showed the maximum inhibitory activity against acetylcholinesterase (AChE) and GSK3β. Conclusion: The highly active phytocompounds of EGB761, especially quercetin, kaempferol, and isorhamnetin, have better activity against AChE and GSK3β than its reported synthetic drug, according to molecular docking and network pharmacology research. These compounds may act on multiple targets in the protein network of AD. The AChE theory was primarily responsible for EGB761’s therapeutic efficacy in treating AD.

The Proliferation of on-demand usage-based IT services, as well as the diverse range of cloud users, have led to the establishment of energy-hungry hefty cloud data centers. Therefore, cloud service providers are striving to reduce energy consumption for cost-saving and environmental sustainability issues of data centers. In this direction, Virtual Machine (VM) consolidated is a widely used approach to optimize hardware resources at cost of performance degradation due to unnecessary migrations. Hence, the motivation of the proposed approach is to minimize energy consumption while maintaining the performance of cloud data centers. This leads to a reduction in the overall cost and an increase in the reliability of cloud service providers. To achieve this goal Predictive Virtual Machine Consolidation (PVMC) algorithm is proposed using the exponential smoothing moving average (ESMA) method. In the proposed algorithm, the ratio of deviation to utilization is calculated for VM selection and placement. migrating the high CPU using VMs or we can restrict steady resource-consuming VMs from migration. The outcomes of the proposed algorithm are validated on computer-based simulation under a dynamic workload and a variable number of VMs (1–290). The experimental results show an improvement in the mean threshing index (40%, 45%) and instruction energy ratio (15%, 17%) over the existing policies. Hence, the proposed algorithm could be used in real-world data centers for reducing energy consumption while maintaining low service level agreement violations.

The lifestyle led by today’s generation and its negligence towards health is highly susceptible to various diseases. Developing countries are at a higher risk of mortality due to late-stage presentation, inaccessible diagnosis, and high-cost treatment. Thermography-based technology, aided with machine learning, for screening inflammation in the human body is non-invasive and cost-wise appropriate. It requires very little equipment, especially in rural areas with limited facilities. Recently, Thermography-based monitoring has been deployed worldwide at various organizations and public gathering points as a first measure of screening COVID-19 patients. In this article, we systematically compare the state-of-the-art feature extraction approaches for analyzing thermal patterns in the human body, individually and in combination, on a platform using three publicly available Datasets of medical thermal imaging, four Feature Selection methods, and four well-known Classifiers, and analyze the results. We developed and used a two-level sampling method for training and testing the classification model. Among all the combinations considered, the classification model with Unified Feature-Sets gave the best performance for all the datasets. Also, the experimental results show that the classification accuracy improves considerably with the use of feature selection methods. We obtained the best performance with a features subset of 45, 57, and 39 features (from Unified Feature Set) with a combination of mRMR and SVM for DB-DMR-IR and DB-FOOT-IR and a combination of ReF and RF for DB-THY-IR. Also, we found that for all the feature subsets, the features obtained are relevant, non-redundant, and distinguish normal and abnormal thermal patterns with the accuracy of 94.75% on the DB-DMR-IR dataset, 93.14% on the DB-FOOT-IR dataset, and 92.06% on the DB-THY-IR dataset.

Skin cancer is a life threatening disease and has caused significant loss to human health across the globe. Its prevalence has been increasing every year and is one of the common malignancies in case of organ transplant recipients of which 95% constitute the basal cell and squamous cell carcinomas. Around the 20th century, sunlight has proven to be the major cause of skin cancer. The prime factor that is responsible for causing skin cancer is the UV radiations. A novel hypothesis by US scientists stated that the cutaneous melanoma was mainly due to the recurrent exposure to the sun, whereas keratinocyte cancer occurred due to progressive accumulation of sun exposure. Management of skin cancer has been done via various approaches such as cryotherapy, radiotherapy, photodynamic therapy etc. Radiotherapy has proven to treat skin cancers to some extent after the discovery of X-rays, but the indication are uncertain since it depends upon the type of tumour and surgical treatment that is required for the patient. Due to various limitations of skin cancer treatment and increased severity, there is a requirement of cost effective, novel and efficient treatment. Various nanocarriers such as SLNs, magnetic nanoparticles, gold nanoparticles, carbon nanotubes etc have been the potential carriers in the management and prognosis of both non-melanoma and melanoma skin cancer. To extract the results, various research and review database along with patent reports have been studied and information has been compiled. A review also discuss about the role of various nanocarriers in the treatment and diagnosis of skin cancer.

Optimal blood pressure targets following successful mechanical thrombectomy remain uncertain. While some observational studies suggest that the relationship between blood pressure and outcomes follows a U -shaped curve, others suggest a linear relationship where lower is better. The recent BP-TARGET study (Blood Pressure Target in Acute Stroke to Reduce Hemorrhage After Endovascular Therapy) did not find a benefit to intensive blood pressure lowering regarding the risk of symptomatic intracranial hemorrhage, but it was not sufficiently powered to detect differences in functional outcomes. On its heels arrived the ENCHANTED2 (Enhanced Control of Hypertension and Thrombectomy Stroke Study)/mechanical thrombectomy trial, the first trial of intensive blood pressure lowering in patients with hypertension following successful mechanical thrombectomy powered to detect a difference in functional outcomes. The trial randomized patients to either a systolic blood pressure less than 120 or 140 to 180 mm Hg. The trial was terminated early due to safety concerns in the more intensive blood pressure–lowering group. In this emerging therapy critique, we explore concerns regarding the generalizability of ENCHANTED2/mechanical thrombectomy, including the high prevalence of intracranial atherosclerosis in the studied population. We survey mechanisms for poor outcomes in patients who receive overly aggressive blood pressure reduction following successful thrombectomy, such as poststroke autoregulatory compromise and persistent microcirculatory hypoperfusion. Finally, we advocate for a more moderate approach, pending further research.

Schiff base compounds are gaining importance daily in this scenario due to their various biological and catalytic activities. In this study, an efficient and eco-friendly synthesis of N-benzylideneaniline was carried out in polyethene glycol (PEG-400) as a greener medium at room temperature. PEG400 was inexpensive and non-toxic, providing a high yield and efficient medium for the synthesis of Schiff bases with excellent outcomes. Furthermore, FT-IR spectroscopy is used to characterize the newly created Schiff base. The recyclability of the catalyst was also studied up to six cycles.

Background: The aim of the present study was to determine the pattern of adverse drug reactions (ADRs) reported at ADR monitoring centre (AMC) in Punjab. Methods: This observational retrospective study was done in department of Pharmacology, GGS Medical College and Hospital, Faridkot from September 2020 to August 2021. A total of 148 ADRs were reported during the study period. Each ADR was analyzed for demographic data and characteristics of ADR. Assessment of causality, severity and preventability was done according to WHO UMC scale, modified Hartwig and Siegel scale and Modified Schumock and Thornton Preventability Scale respectively. Results: A total of 148 ADRs were reported from both outpatients and in patients of various departments. Most of the ADRs were found in males (55%) and patients of age group 31-45 years (33%). Majority of ADRs were reported from dermatology department (40%). Overall, 38% of ADRs were due to antimicrobial drugs. Most of the ADRs were reported as possible (57%), followed by probable (41%) as per WHO causality assessment. Most of the ADRs were moderate severity (83%). 97% of the ADRs were found to be definitely preventable type. Conclusions: We concluded that most of the ADRs were reported from antimicrobial drugs, so it is advisable to have close monitoring of the antimicrobial drug therapy to prevent ADRs in the patients. Although the majority of ADRs were moderate in nature but mostly were recovered. The study of ADRs in a particular institute using demographic patterns will contribute to patient safety by sensitizing the clinicians in that particular institute.

Periodontitis is an infection-driven inammatory condition that leads to destruction of the attachment apparatus of the tooth where, the major etiological factor leading to the destruction is plaque. Plaque act as a reservoir for the complex subgingival microbiota. Therefore, an inference here could be drawn that with the overall reduction of micro-organism, disease progression could be halted. Periodontal therapy aims at restoring balanced periodontal health along with the compatible oral microbiota. Treatment of periodontal disease aims at achieving a healthy periodontium and preventing the risk of recurrence or disease progression. Due to specic properties of biolm, the subgingival periodontal microbiota are more difcult to target which leads to dearth of clear protocols for the use of antibiotics and therefore, the development of specically designed strategies to treat the subgingival microbiota, as a biolm, is highly desirable. So, comes the use of antibiotics in periodontal therapy. Systemic antibiotics enter periodontal tissues via serum and can affect microorganisms outside the reach of adequate instrumentation and towards use of topical anti-infective chemotherapeutics. Antibiotic therapy delays subgingival recolonisation of the microbial pathogens by potentially suppressing the periodontal pathogens residing onto the oral surfaces. Antibiotic treatment strategies in conjugation with proper mechanical biolm disruption, eventually helps in maintaining the periodontal health by minimizing and eradicating subgingival periodontalpathogens and thereby, improves the status of clinical outcomes of periodontal therapy. This review henceforth, attempts to highlight the walk-on part of antibiotics in periodontal therapy.

Severe acute respiratory syndrome coronavirus 2 disease (COVID-19) has caused more than 6 million deaths globally. Understanding predictors of mortality will help in prioritizing patient care and preventive approaches. This was a multicentric, unmatched, hospital-based case-control study conducted in nine teaching hospitals in India. Cases were microbiologically confirmed COVID-19 patients who died in the hospital during the period of study and controls were microbiologically confirmed COVID-19 patients who were discharged from the same hospital after recovery. Cases were recruited sequentially from March 2020 until December–March 2021. All information regarding cases and controls was extracted retrospectively from the medical records of patients by trained physicians. Univariable and multivariable logistic regression was done to assess the association between various predictor variables and deaths due to COVID-19. A total of 2,431 patients (1,137 cases and 1,294 controls) were included in the study. The mean age of patients was 52.8 years (SD: 16.5 years), and 32.1% were females. Breathlessness was the most common symptom at the time of admission (53.2%). Increasing age (adjusted odds ratio [aOR]: 46–59 years, 3.4 [95% CI: 1.5–7.7]; 60–74 years, 4.1 [95% CI: 1.7–9.5]; and ≥ 75 years, 11.0 [95% CI: 4.0–30.6]); preexisting diabetes mellitus (aOR: 1.9 [95% CI: 1.2–2.9]); malignancy (aOR: 3.1 [95% CI: 1.3–7.8]); pulmonary tuberculosis (aOR: 3.3 [95% CI: 1.2–8.8]); breathlessness at the time of admission (aOR: 2.2 [95% CI: 1.4–3.5]); high quick Sequential Organ Failure Assessment score at the time of admission (aOR: 5.6 [95% CI: 2.7–11.4]); and oxygen saturation < 94% at the time of admission (aOR: 2.5 [95% CI: 1.6–3.9]) were associated with mortality due to COVID-19. These results can be used to prioritize patients who are at increased risk of death and to rationalize therapy to reduce mortality due to COVID-19.

Hereditary angioedema (HAE) is a rare genetic disease with significant mortbidity and mortality for which early diagnosis and effective therapy are critical. Many Asia-Pacific (AP) countries still lack access to diagnostic tests and evidence-based therapies. Epidemiological data from the AP is needed to formulate regional guidelines to improve standards of care for HAE.To investigate the estimated minimal prevalence, needs and potential interventions for the diagnosis and management of HAE in the AP.A structured questionnaire was distributed to representative experts from member societies of the Asia Pacific Association of Allergy, Asthma and Clinical Immunology (APAAACI). Patient profiles and presence of diagnostic facilities/ tests, regional / national HAE guidelines and patient support groups were reported and compared.Completed questionnaires were received from 14 representatives of 12 member countries/territories, representing 46% of the world population. Overall minimal prevalence of HAE in the AP region was 0.02 per 100,000 population, with significant heterogeneity across different centres. Only half and one-third had registered on-demand and prophylactic medications, respectively. Few had patient support groups (58%) or regional guidelines (33%), and their existence was associated with availability of HAE-specific medications. Availability of C1-inhibitor level testing was associated with a lower age of HAE diagnosis (p=0.017).HAE in the AP differs from Western countries. HAE-specific medications were only registered in a minority of countries/territories, but those with patient support groups or regional guidelines were more likely to have better access. AP specific consensus and guidelines are lacking and urgently needed.

To examine patient and disease characteristics, toxicity, and clinical outcomes for patients with advanced urothelial carcinoma (aUC) who are rechallenged with immune checkpoint inhibitor (ICI)-based therapy.In this retrospective cohort, we included patients treated with ICI for aUC after having prior ICI treatment. Endpoints included the evaluation of radiographic response and disease control rates with first and second ICI courses, outcomes based on whether there was a change in ICI class (anti-PD-1 vs. anti-PD-L1), and assessment of the reasons for ICI discontinuation.We identified 25 patients with aUC from 9 institutions who received 2 separate ICI courses. ORR with first ICI and second ICI were 39% and 13%, respectively. Most patients discontinued first ICI due to progression (n = 19) or treatment-related toxicity (n = 4). Thirteen patients received non-ICI treatment between the first and second ICI, and 12 patients changed ICI class (anti-PD-1 vs. anti-PD-L1) at rechallenge. Among 10 patients who changed ICI class, 8 (80%) had progressive disease as best response with second ICI, while among 12 patients re-treated with the same ICI class, only 3 (25%) had progressive disease as best response at the time of rechallenge. With second ICI, most patients discontinued treatment due to progression (n = 18) or patient preference (n = 2).A proportion of patients with aUC rechallenged with ICI-based regimens may achieve disease control, supporting clinical trials in that setting, especially with ICI-based combinations. Future studies are needed to validate our results and should also focus on identifying biomarkers predictive of benefit with ICI rechallenge.

The present study examines whether overweight and obese are inter related with low-grade systemic inflammation as measured by serum hs-CRP concentration. The present observational, cross-sectional study is from the department of medicine, CSS Hospital, Subharti Medical College, Meerut, to evaluate the profile of patients who came for routine health check-ups from December 2020 to January 2022. The study comprised 150 patients divided into three groups normotensives, normoglycemic adults of the age group 20-70 years based on BMI as Normal BMI (18.5-22.9 kg/m2), overweight (23-24.9 kg/m2) and obese (BMI>25 kg/m2), according to Asian standards by WHO. The mean age among the study subjects was 49.38±12.62 years. Out of 150 subjects, 83 (55.33%) were males and 67 (44.67%) were females. Normal, overweight, and obesity were found among 34%, 29.33%, and 36.67% of the subjects respectively. Normal and high hs-CRP was reported in 46% and 54% of the subjects respectively. The mean hs-CRP among the study subjects was 6.07±4.23. Mean FBS (mg/dL), WC (cm), TC (mg/dL), TGL (mg/dL), and HDL (mg/dL) among the study subjects was 102.94±13.03, 88.2±13.36, 161.84±52.56, 133.18±64.48 and 34.73±17.44 respectively. Mean hs-CRP was found to be highest in obese subjects (10.09±3.67) followed by overweight (6.74±2.48) and normal subjects (2.32±1.11). When hs-CRP was compared among normal vs overweight, normal vs obese, and overweight vs obese using a t-test, a significant difference was found. Pearson correlation analysis revealed a positive correlation between BMI, FBS (mg/dL), WC (cm), TC/HDL (mg/dL), TGL (mg/dL), and hs-CRP i.e. with an increase in these mentioned parameters, hs-CRP value also increases. Conclusion: Obesity is a state of chronic inflammation that has a strong link to hs-CRP levels.

Gurugram is emerging as one of India's most advanced cities. The combined impact of industrial and vehicular emissions makes the environment toxic. Recently, Gurugram has experienced severe air quality. In the present work, an assessment of seasonal, monthly, and diurnal variations of ambient air quality was carried out in Gurugram during the period of March 2021 to 2022 February. Seasonal and monthly concentrations of key air pollutants like particulate matter (PM2.5), nitrogen dioxide (NO2), carbon monoxide (CO) and ozone (O3) were examined at Vikas sadan, Gwal Pahari and Teri Gram in Gurugram city to study the most polluted seasons and months. Significantly higher mean concentrations of Particulate matter PM2.5 (406.94 μgm−3) and NO2 (353.96ppb) were seen during the colder months and seasons. O3 showed a consistent trend with variations during the year, with the highest concentration in winter (106.35µg/m3). PM2.5 and NO2 concentrations during the night were greater for all seasons when compared to diurnal values. O3 concentrations displayed diurnal tendencies that were the opposite of those of NO2 concentrations. The highest concentrations of ambient PM2.5, NO2, and CO were observed at the Vikas Sadan Monitoring Station. While the NISE Gwal Pahari station showed greater O3 values. The findings highlight the necessity of efficient air pollution control in Gurugram. To prevent public exposure to air pollutants, preventive measures like green spaces, using public transport, etc. must be adopted. The study contributes to a better understanding of air pollution by seasonal, monthly and diurnal assessment in the city of Gurugram.

Endometriosis refers to the extrauterine presence of hormonally active endometrial glands and stroma. This ectopic endometrial tissue exhibits cyclic bleeding, inflammation, fibrosis and leads to formation of adhesions. Endometriosis affects about 10% to 15% of women between 15 to 45 years of age. Clinical presentations vary from infertility, dysmenorrhoea, chronic pelvic pain, deep dyspareunia and even bleeding at external sites like the umbilicus. Besides involving fallopian tubes, bowel, liver, thorax, pericardium, pleura etc, the most commonly affected areas in the gastrointestinal tract are the descending colon, rectosigmoid, appendix, and ileo-caecum in descending order of frequency. This case series highlights some unusual presentations of endometriosis along with co-existence of other ovarian pathologies which are not frequently encountered in clinical practice. This series also highlights the role of preoperative radiology for an adequate clinical diagnosis and complete surgical excision.

Skin manifestations have always been alarming to the patient and often warrant complete clinical evaluation. A thorough understanding of relationship between gastrointestinal (GI) disorders and skin diseases can alert the clinician to an underlying occult GI disorder. The aim of the study was to review the cutaneous changes in patients with co-existent, often masked GI diseases and to observe the underlying pathophysiology of such lesions. This was a retrospective, record based observational study. The archived histopathology reports & medical records of patients over a period of 5 years were searched and all patients who underwent skin biopsy with co-existent GI diseases were included. Out of a total of 218 patients of GI diseases who had dermatological manifestations, only 44 were biopsied. Pruritis (n=175), koilonychia (n=52) and erythematous rash (n=35) were the most common clinical features. Psoriasis (n=4), vasculitis (n=4), Dermatitis Herpetiformis (n=3) and pyoderma gangrenosum (n=5) were often seen associated with benign GI diseases like Inflammatory Bowel Diseases, Chronic Liver Disease, Malabsorption syndrome etc. Sweets syndrome (n=2) and cutaneous metastatic nodules (n=3) were associated with underlying malignancy particularly of gall bladder and pancreas.

Coccidiosis caused by protozoan parasite of Eimeria species is one of the most important poultry diseases prevalent in different parts of globe including India. The parasite invades the intestinal cells and the disease is characterised by enteritis and diarrhoea leading to weight loss, altered feed conversion ratio, poor growth and eventually death of affected birds. A survey on commercial poultry farms was conducted in 10 districts of Haryana state, India, using a questionnaire to assess the status of coccidiosis, associated risk factors and management practices followed. On analysis of data, it was found that overall, 15 (28.8%) farms had history of coccidiosis. One-fourth (25%) of the farmers reported the use of anticoccidial drugs for chemoprophylaxis or therapeutic purposes at their farms. Background: Adverse drug reactions are important causes of mortality and morbidity in the patients. Early detection, evaluation and monitoring of ADRs is essential to improve public health. Methods: This was an observational, non-interventional and retrospective study conducted at the ADR monitoring centre of a tertiary care hospital of North India. Suspected ADR forms reported over a period of 4 years involving at least one chemotherapeutic drug with at least one dose were analyzed. Results: A total of 261 chemotherapeutic drugs associated ADRs were analyzed. Out of these, maximum numbers of ADRs were reported by males (54%). Maximum reporting was done by Skin and VD department (37.93%). Causality assessment was probable in maximum number of cases (54%). Most common ADRs were skin rashes (21.46%), followed by jaundice, urticaria and fixed drug eruptions. Maximum ADRs were suspected to be caused by Anti tubercular drugs (31.42%) followed by anticancer drugs (14.56%). Conclusions: ADRs due to antibiotics and anticancer drugs is a significant health problem.

Male hypogonadism (MH) is a clinical and biochemical syndrome caused by inadequate synthesis of testosterone. Untreated MH can result in long-term effects, including metabolic, musculoskeletal, mood-related, and reproductive dysfunction. Among Indian men above 40 years of age, the prevalence of MH is 20%–29%. Among men with type 2 diabetes mellitus, 20.7% are found to have hypogonadism. However, due to suboptimal patient-physician communication, MH remains heavily underdiagnosed. For patients with confirmed hypogonadism (either primary or secondary testicular failure), testosterone replacement therapy (TRT) is recommended. Although various formulations exist, optimal TRT remains a considerable challenge as patients often need individually tailored therapeutic strategies. Other challenges include the absence of standardized guidelines on MH for the Indian population, inadequate physician education on MH diagnosis and referral to endocrinologists, and a lack of patient awareness of the long-term effects of MH in relation to comorbidities. Five nationwide advisory board meetings were convened to garner expert opinions on diagnosis, investigations, and available treatment options for MH, as well as the need for a person-centered approach. Experts’ opinions have been formulated into a consensus document with the aim of improving the screening, diagnosis, and therapy of men living with hypogonadism.

Owing to widespread groundwater pollution, arsenic is one of the most extreme worldwide ecological toxicants and induces cytotoxic activity and other chronic diseases for millions of people and also animals. Because of geologic and anthropogenic activities, arsenic pollution is widely spread. Long-term toxicity caused by arsenic can lead to the development of arsenicosis related to skin lesions, hepatomegaly, splenomegaly, cognitive dysfunction, cardiovascular and peripheral vascular disease, hypertension, diabetes mellitus, hepatic fibrosis and hematological, gastrointestinal, immunological, endocrine and multicancer neurological disorders. Till date no effective treatment or medicine is available in global market to prevent arsenic induced toxicosis. Only use of safe water and well nourished foods can help to prevent further exposure. Recently, chelation therapy is recognized as a good remedial action to reduce and to eliminate the acute arsenic toxicity.

Cell identity is governed by the complex regulation of gene expression, represented as gene-regulatory networks1. Here we use gene-regulatory networks inferred from single-cell multi-omics data to perform in silico transcription factor perturbations, simulating the consequent changes in cell identity using only unperturbed wild-type data. We apply this machine-learning-based approach, CellOracle, to well-established paradigms—mouse and human haematopoiesis, and zebrafish embryogenesis—and we correctly model reported changes in phenotype that occur as a result of transcription factor perturbation. Through systematic in silico transcription factor perturbation in the developing zebrafish, we simulate and experimentally validate a previously unreported phenotype that results from the loss of noto, an established notochord regulator. Furthermore, we identify an axial mesoderm regulator, lhx1a. Together, these results show that CellOracle can be used to analyse the regulation of cell identity by transcription factors, and can provide mechanistic insights into development and differentiation.

The antibacterial activities of two previously reported copper (II) coordination complexes [Cu(HL1)Cl2] and [Cu(HL2)Cl2], and the two new complexes [Cu(HL3)Cl2] and [Cu(HL4)Cl2], where HL1 is 2-(((pyridin-2-ylmethyl)amino)methyl)phenol, HL2 is 2-((benzyl(pyridine-2-ylmethyl)amino)methyl)phenol, HL3 is 2-(((pyridin-2-ylethyl)amino)methyl)phenol and HL4 is 2-(((pyridin-2-ylmethyl)(quinolin-2-ylmethyl)-amino)methyl)phenol were investigated using agar well plate diffusion assay. The ligands were characterized using elemental analysis, IR, 1H-NMR, 13C-NMR spectroscopy and thermal analysis. The corresponding copper (II) complexes of each ligand were synthesized and characterized using elemental analysis, FT-IR and UV-Visible spectroscopy, electronic spectra and magnetic moment measurements, and thermal analysis. Additionally, a thorough investigation of the copper complexes as potent drug candidates was performed using web-based software SwissADME to predict their physicochemical and pharmacokinetics properties. All copper complexes were stable to air and moisture and showed excellent stability up to 200 °C. Electronic spectra of all copper complexes in methanol consisted of single band at 14,490–15,260 cm−1 depicting ligand field excitation (Cu (II) d–d band). Ligands were completely ineffective on gram positive bacteria; HL1 and HL3 showed moderate to high activity at concentration range of 0.2–1 mg/ml. Copper complexes were found to exhibit moderate to high activity against bacteria as compared to the ligands. SwissADME analysis of all copper (II) complexes justified their candidature as potential candidate for pharmaceutical applications.
