Pulmonary rehabilitation (PR) is a cost-effective intervention with well-known benefits on exercise capacity, symptoms and quality of life in patients with chronic respiratory diseases. Despite the compelling evidence of its benefits, PR implementation is still suboptimal, and maintenance of PR benefits is challenging. To overcome these pitfalls, there has been a growing interest to develop novel models for PR delivery. Digital health is a promising solution, as it has the potential to address some of the most reported barriers to PR uptake and adherence (such as accessibility issues), help maintain the positive results following a PR programme, and/or promote patients’ adherence to a more active lifestyle through physical activity (tele-)coaching. Despite the accelerated use of digital health to deliver PR during the COVID-19 pandemic, there are still several factors that contribute to the resistance in the adoption of digital health, such as the lack of evidence on its effectiveness, low acceptability by patients and healthcare professionals, concerns about implementation and maintenance costs, inequalities in access to the internet and technological devices, and data protection issues. Nevertheless, the trend towards reducing technology costs and the higher availability of digital devices, as well as the greater ease and simplicity of use of devices, enhance the opportunities for future development of digitally enabled PR interventions. We consider real 2-step metric nilpotent Lie algebras associated to graphs with possibly repeated edge labels as constructed by Ray in 2016. We determine how the structure of the egde labeling within the graph contributes to the abelian factor in these Lie algebras. Furthermore, we explicitly compute the abelian factor of the 2-step nilpotent Lie algebras associated with some families of graphs such as star graphs, cycles, Schreier graphs, and properly edge-colored graphs. Aims: Research objectively evaluating physical activity (PA) and sleep in adults with hypermobile Ehlers–Danlos syndrome (hEDS) and generalized hypermobility spectrum disorder (G-HSD) is lacking. Furthermore, it is not clear to what extent frequently occurring symptoms in these patients are related to their PA and sleep. Therefore, a cross-sectional study was performed to objectively evaluate, and identify factors contributing to, PA and sleep in adults with hEDS and G-HSD. Methods: Twenty female adults with hEDS, 23 with G-HSD, and 32 healthy controls participated. Physical activity and sleep were measured using two tri-axial ActiGraphs worn over seven consecutive days. Furthermore, questionnaires evaluating frequently occurring symptoms were completed. Regression analysis was performed to determine major contributors to PA and sleep. Results: Daily step counts were significantly lower in both patient groups compared to the control (CTR) group (p lessthan 0.04) and to the recommended 7500 steps (p≤0.001). Other PA and sleep variables did not differ between the groups. In the hEDS group, body mass index and kinesiophobia were related to PA, explaining 53% of the variance in step counts. In the G-HSD group, 18.5% of the variance in step counts could be attributed to the variance in pain impact. Conclusion: Adults with hEDS and G-HSD had lower step counts than healthy peers, which may be partially due to kinesiophobia and the impact of pain respectively. No differences in objectively measured sleep parameters were identified. Treatment focusing on fear-avoidance beliefs and pain relief could potentially increase daily step counts and benefit overall health in these patients.

The current study shows the research on phytochemical concerning the phenolic components of Algerian pomegranate fruit extract (Punica granatum L.) using Ultra Performance Liquid Chromatography Diode-Array Detection Electrospray Ionization Mass Spectrometry (UPLC-DAD-ESI-MS). In this study; about more than 50 phenolic compounds, including 40 phenolic acids, 5 flavonoids, 2 coumarins and ellagitannins were identified. The profile is characterized by the presence of major peak at retention time 0.59, related to Bergaptan diglucoside (40%). This latter compound and many compounds such as digalloyl lactone citric acid, dicafeoyl galloyl dihydroxymalonic acid, ferruloyl diglucosyl citrate, dicafeoyl galloyl malonyl citrate, protocatechic cafeoyl coumaroyl rhamnoside, isorhamnetin glucosyl rhamnoside, dihydrokaemferol ferruloyl cafeoyl, and bergapten triglucoside methylsulphide were first time reported and identified in the hydroalcoholic fruit extract of Algerian Punica granatum. This acquired information may be useful to validate the fruit extract of Punica granatum, as source of bioactive compounds.

Purpose:KPT-8602 (Eltanexor) is a second-generation exportin-1 (XPO1) inhibitor with potent activity against acute lymphoblastic leukemia (ALL) in preclinical models and with minimal effects on normal cells. In this study, we evaluated whether KPT-8602 would synergize with dexamethasone, vincristine, or doxorubicin, three drugs currently used for the treatment of ALL.Experimental Design:First, we searched for the most synergistic combination of KPT-8602 with dexamethasone, vincristine, or doxorubicin in vitro in both B-ALL and T-ALL cell lines using proliferation and apoptosis as a readout. Next, we validated this synergistic effect by treatment of clinically relevant B- and T-ALL patient-derived xenograft models in vivo. Finally, we performed RNA-sequencing (RNA-seq) and chromatin immunoprecipitation sequencing (ChIP-seq) to determine the mechanism of synergy.Results:KPT-8602 showed strong synergism with dexamethasone on human B-ALL and T-ALL cell lines as well as in vivo in three patient-derived ALL xenografts. Compared with single-drug treatment, the drug combination caused increased apoptosis and led to histone depletion. Mechanistically, integration of ChIP-seq and RNA-seq data revealed that addition of KPT-8602 to dexamethasone enhanced the activity of the glucocorticoid receptor (NR3C1) and led to increased inhibition of E2F-mediated transcription. We observed strong inhibition of E2F target genes related to cell cycle, DNA replication, and transcriptional regulation.Conclusions:Our preclinical study demonstrates that KPT-8602 enhances the effects of dexamethasone to inhibit B-ALL and T-ALL cells via NR3C1- and E2F-mediated transcriptional complexes, allowing to achieve increased dexamethasone effects for patients.

T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive type of leukemia caused by accumulation of genetic alterations in T-cell progenitors. However, for many genes it remains unknown how loss-of-function mutations contribute to disease development. Single-cell CRISPR screening in ex vivo cultured primary murine pro-T cells allowed us to study the transcriptomic impact of such alterations and to link this with effects on cell proliferation and survival. We first targeted 17 well-studied T-cell regulators and defined key transcriptional signatures, such as NOTCH, MYC, STAT and E2F. A second screen targeted 42 poorly characterized genes and identified gene clusters with E2F/MYC and STAT/NOTCH signatures having opposing roles. These analyses identified a cluster of genes, including multiple chromatin modifiers, regulating Myc expression. Additionally, Spi1 was identified as an essential gene for cell survival, associated with the MYC regulon. Bcl11b inactivation conferred the strongest growth advantage and was associated with JAK/STAT upregulation, corresponding with publicly available patient data. Bcl11b inactivation together with mutant JAK3 transformed pro-T cells to cytokine-independent growth. With this data, we characterized tumor suppressors and oncogenes in T-ALL, providing insight in the mechanisms of leukemia development.

INSIDER (Improved Nuclear SIte characterization for waste minimization in D&D operations under constrained EnviRonment) was a European project funded under the H2020-EURATOM programme and launched in June 2017. The project was coordinated by the French Commissariat a l'energie atomique et aux energies alternatives (CEA), it had a total duration of 4 years and covered a budget of 4 Meuro. INSIDER's work was performed by 5 technical working groups (WG) which brought together 18 institutions from 10 countries, leading to a total of 68 participating researchers. The objective of the project was to optimise the radiological characterisation of nuclear installations in constrained environments in order to obtain an accurate estimate of the content of contaminated materials as well as to optimise the quantity of contaminated materials to be treated as waste. The focus of this paper is on the statistical analysis of an interteam comparison of measurement results (dose rate, total gamma measurement, and gamma spectrometry) made in situ at the BR3 reactor, Belgium. BackgroundLong-term outcome data of coronavirus disease 2019 (COVID-19) survivors are needed to understand their recovery trajectory and additional care needs.MethodsA prospective observational multicentre cohort study was carried out of adults hospitalised with COVID-19 from March through May 2020. Workup at 3 and 12 months following admission consisted of clinical review, pulmonary function testing, 6-min walk distance (6MWD), muscle strength, chest computed tomography (CT) and quality of life questionnaires. We evaluated factors correlating with recovery by linear mixed effects modelling.ResultsOf 695 patients admitted, 299 and 226 returned at 3 and 12 months, respectively (median age 59 years, 69% male, 31% severe disease). About half and a third of the patients reported fatigue, dyspnoea and/or cognitive impairment at 3 and 12 months, respectively. Reduced 6MWD and quadriceps strength were present in 20% and 60% at 3 months versus 7% and 30% at 12 months. A high anxiety score and body mass index correlated with poor functional recovery. At 3 months, diffusing capacity for carbon monoxide (DLCO) and total lung capacity were below the lower limit of normal in 35% and 18%, decreasing to 21% and 16% at 12 months; predictors of poor DLCO recovery were female sex, pre-existing lung disease, smoking and disease severity. Chest CT improved over time; 10% presented non-progressive fibrotic changes at 1 year.ConclusionMany COVID-19 survivors, especially those with severe disease, experienced limitations at 3 months. At 1 year, the majority showed improvement to almost complete recovery. To identify additional care or rehabilitation needs, we recommend a timely multidisciplinary follow-up visit following COVID-19 admission.

TAL1 is ectopically expressed in about 30% of T-cell acute lymphoblastic leukemia (T-ALL) due to chromosomal rearrangements leading to the STIL-TAL1 fusion genes or due to non-coding mutations leading to a de novo enhancer driving TAL1 expression. Analysis of sequence data from T-ALL cases demonstrates a significant association between TAL1 expression and activating mutations of the PI3K-AKT pathway. We investigated the oncogenic function of TAL1 and the possible cooperation with PI3K-AKT pathway activation using isogenic pro-T-cell cultures ex vivo and in vivo leukemia models. We found that TAL1 on its own suppressed T-cell growth, in part by affecting apoptosis genes, while the combination with AKT pathway activation reduced apoptosis and was strongly driving cell proliferation ex vivo and leukemia development in vivo. As a consequence, we found that TAL1+AKTE17K transformed cells are more sensitive to PI3K-AKT pathway inhibition compared to AKTE17K transformed cells, related to the negative effect of TAL1 in the absence of activated PI3K-AKT signaling. We also found that both TAL1 and PI3K-AKT signaling increased the DNA-repair signature in T cells resulting in synergy between PARP and PI3K-AKT pathway inhibition. Occasionally, a vexing problem is solved by a sudden epiphany with the Aha! as its felt component. Although insight occurs sometimes, the mundane way to solve problems is analytically (i.e., non-insight). At first glance, non-insight appears to depend on the availability and taxation of cognitive resources to execute the step-by-step approach, whereas insight does not or to a lesser extent. However, this remains debated. To address whether both solution types depend (in)differentially on cognitive resources, we assessed the involvement of the prefrontal cortex, which is closely linked to it. Following the neurovisceral integration model, we used vagally mediated heart rate variability (i.e., vmHRV) to index prefrontal resource availability (i.e., trait) and prefrontal resource taxation (i.e., state). Participants (N = 68) solved 70 compound remote associates word puzzles solvable with insight and non-insight. Before, during, and after solving the word puzzles, we measured vmHRV. Our results showed that resting-state vmHRV (i.e., trait) showed a negative association with behavioural performance for both solution types. This might reflect inter-individual differences in inhibitory control. As the solution search requires one to think of remote associations, inhibitory control might hamper rather than aid this process. Furthermore, we observed, for both solution types, a vmHRV increase from resting-state to solution search (i.e., state), lingering on in the post-task recovery period. This could mark the increase of prefrontal resources to promote an open-minded stance, essential for divergent thinking, which arguably is crucial for this task. Our study revealed that both solution types might have more in common than meets the eye.

The uniqueness of MALT1 protease activity in controlling several aspects of immunity in humans has made it a very attractive therapeutic target for multiple autoimmune diseases and lymphoid malignancies. Despite several encouraging preclinical studies with MALT1 inhibitors, severe reduction in regulatory T cells and immune-mediated pathology seen in MALT1 protease-dead (MALT1-PD) mice and some, but not all, studies analysing the effect of prolonged pharmacological MALT1 protease inhibition, indicates the need to further unravel the mechanism of MALT1 protease function. Notably, the contribution of individual MALT1 substrates to the immune defects seen in MALT1-PD mice is still unclear. Previous in vitro studies indicated a role for MALT1-mediated cleavage of the E3 ubiquitin ligase HOIL-1 in the modulation of nuclear factor-kappa B (NF-kappa B) signalling and inflammatory gene expression in lymphocytes. Here, we addressed the immunological consequences of inhibition of HOIL-1 cleavage by generating and immunophenotyping MALT1 cleavage-resistant HOIL-1 knock-in (KI) mice. HOIL-1 KI mice appear healthy and have no overt phenotype. NF-kappa B activation in T or B cells, as well as IL-2 production and in vitro T-cell proliferation, is comparable between control and HOIL-1 KI cells. Inhibition of HOIL-1 cleavage in mice has no effect on thymic T-cell development and conventional T-cell homeostasis. Likewise, B-cell development and humoral immune responses are not affected. Together, these data exclude an important role of MALT1-mediated HOIL-1 cleavage in T- and B-cell development and function in mice.

Global warming and plastic pollution are two human-induced environmental stressors of rising concern due to their potential impact on ocean health. To tackle sustainability concerns on the production of conventional petroleum-based polymers, bio-based polymers, i.e. polymers originated from natural feedstocks, are seen as a potential alternative. However, just as conventional polymers, plastic items from bio-based polymers can leach additives and other associated substances into the (marine) environment. To date, the ecotoxicological effects of leachates from bio-based polymers are still unclear, but previous reports have demonstrated that leachates from petroleum-based polymers can induce adverse effects in marine invertebrates. Also, in the environment, organisms are subjected to a myriad of environmental stressors, among which global warming, and the combination of stressors is often not considered in environmental risk assessments. To increase our understanding of the combined effects of plastic leachates from either bio-based or petroleum-based polymers and elevated temperature (global warming), in this work we used the case study of polylactic acid (PLA) and polypropylene (PP), at control (22°C) and elevated water temperatures (25°C). To do so, we exposed newly hatched larvae of a harpacticoid copepod Nitokra spinipes to plastic leachates (80 g/L) as well as selected additives of both polymers as positive controls, at each temperature. Cancer cells outgrowing in distant organs of metastasis rewire their metabolism to fuel on the available nutrients. While this is often considered an adaptive pressure limiting metastasis formation, some nutrients available at the metastatic site naturally or through changes in organ physiology may inherently promote metastatic growth. We find that the lung, a frequent site of metastasis, is a lipid-rich environment. Moreover, we observe that pathological conditions such as pre-metastatic niche formation and obesity further increase the availability of the fatty acid palmitate in the lung. We find that targeting palmitate processing inhibits spheroid growthin vitroand metastasis formation in lean and obese mice. Mechanistically, we discover that breast cancer cells use palmitate to synthesize acetyl-CoA in a carnitine palmitoyltransferase 1a (CPT1a)-dependent manner. Lysine acetyltransferase 2a (KAT2a), whose expression is promoted by palmitate availability, relies on the available acetyl-CoA to acetylate the NF-κB subunit p65. This favors nuclear location of p65 and activates a pro-metastatic transcriptional program. Accordingly, deletion of KAT2a phenocopies CPT1a silencingin vitroas well asin vivoand patients with breast cancer show co-expression of both proteins in metastases across palmitate-rich metastatic sites. In conclusion, we find that palmitate-rich environments foster metastasis growth by increasing p65 acetylation resulting in elevated NF-κB signaling.

Previous research has demonstrated that the test coverage of libraries can be expanded by using existing test inputs from their dependent projects. In this paper, we propose an algorithm for test transplantation based on test slicing. The algorithm extracts test inputs, isolates them by creating mocks, and then transplants the test code onto the test suite of the libraries. To achieve test slicing, we dynamically execute the tests in the dependent project and create its graph of histories. Then, we traverse back from the interesting object state and collect the corresponding edges. Finally, we reverse the collected edges and create a sequence of method calls to reconstruct the same object state. We have implemented a proof-of-concept in Pharo-Smalltalk, in this paper we discuss the lessons learned so far.

Saponins are amphiphilic molecules of pharmaceutical interest and most of their biological activities (i.e., cytotoxic, hemolytic, fungicide, etc.) are associated to their membranolytic properties. These molecules are secondary metabolites present in numerous plants and in some marine animals, such as sea cucumbers and starfishes. Structurally, all saponins correspond to the combination of a hydrophilic glycan, consisting of sugar chain(s), linked to a hydrophobic triterpenoidic or steroidic aglycone, named the sapogenin. Saponins present a high structural diversity and their structural characterization remains extremely challenging. Ideally, saponin structures are best established using nuclear magnetic resonance experiments conducted on isolated molecules. However, the extreme structural diversity of saponins makes them challenging from a structural analysis point of view since, most of the time, saponin extracts consist in a huge number of congeners presenting only subtle structural differences. In the present review, we wish to offer an overview of the literature related to the development of mass spectrometry for the study of saponins. This review will demonstrate that most of the past and current mass spectrometry methods, including electron, electrospray and matrix-assisted laser desorption/ionization ionizations, gas/liquid chromatography coupled to (tandem) mass spectrometry, collision-induced dissociation including MS3 experiments, multiple reaction monitoring based quantification, ion mobility experiments, and so forth, have been used for saponin investigations with great success on enriched extracts but also directly on tissues using imaging methods.

Ipomoea aquatica is a common green leafy vegetable that has numerous uses in traditional medicine. This study focused on the determination of the cytotoxic, antiradical, and antidiabetic properties of various fractions of the I. aquatica methanolic extract, as well as on the tentative identification of some bioactive compounds in the same fractions. The cytotoxicity was determined by the brine shrimp lethal test. The antioxidant activities of the I. aquatica fractions were investigated through 3 assays. The antidiabetic activity (in vitro) was measured by α-glucosidase and α-amylase inhibition assays. Phytochemical qualitative analyses demonstrated the presence of alkaloids, terpenoids, phenols, and flavonoids in the ethyl acetate-methanol and methanol fractions. The total phenolic and total flavonoid contents were found to be highest in the ethyl acetate-MeOH fractions. The evaluation of the cytotoxicity showed that the hexane-dichloromethane fraction is the most toxic, while the others are moderately toxic. The antioxidant activity assays showed that the ethyl acetate-MeOH fractions are the most potent, while the α-glucosidase and α-amylase assays revealed that the hexane-dichloromethane fraction might contain a potent antidiabetic agent. Some bioactive substances in the MeOH fractions, such as salicylic acid glucoside, 1-O-sinapoyl-β-D-glucose derivative, and dihydroferulic acid derivative, were tentatively identified. To the best of our knowledge, this is the first report to detect and identify these compounds in this species. Based on the results of this study, it may be concluded that I. aquatica is a potent antioxidant agent and could be a good candidate as a natural antioxidant in food and therapeutics.

Peripheral T-cell lymphoma (PTCL) is a heterogeneous group of hematological cancers arising from the malignant transformation of mature T cells. In a cohort of 28 PTCL cases, we identified recurrent overexpression of MYCN, a member of the MYC family of oncogenic transcription factors. Approximately half of all PTCL cases was characterized by a MYC expression signature. Inducible expression of MYCN in lymphoid cells in a mouse model caused T-cell lymphoma that recapitulated human PTCL with an MYC expression signature. Integration of mouse and human expression data identified EZH2 as a key downstream target of MYCN. Remarkably, EZH2 was found to be an essential cofactor for the transcriptional activation of the MYCN-driven gene expression program, which was independent of methyltransferase activity but dependent on phosphorylation by CDK1. Software start-ups have gained attention from the software engineering research community in the recent past. However, the systematic adoption of existing engineering practices for use in start-ups has not attracted that much attention. The smooth transitioning towards software product lines in particular can be a major stumbling block for start-ups that must broaden their product portfolio to deal with divergent demands imposed by the market and the costs of maintaining their products. We conducted a preliminary study within two software start-ups, which revealed the motivating factors and benefits that would lead to the migration from a single product into a product line. Despite the benefits, tackling the challenges foreseen and the identification of features and their relations in the current product is the crucial first step towards implementing an appropriate highly-configurable product portfolio.

Agricultural operations are important sources of organic dust containing particulate matter (PM) and endotoxins, which have possible negative health con-sequences for both humans and animals. Dust concen-trations and composition in calf barns, as well as the potential health effects for these animals, are scarcely documented. The objective of this study was to mea-sure PM fractions and endotoxin concentrations in calf barns and study their associations with lung consoli-dation, respiratory tract inflammation, and infection in group-housed calves. In this cross-sectional study, samples from 24 dairy farms and 23 beef farms were collected in Belgium from January to April 2017. PM1.0, PM2.5 and PM10 (defined as particulate matter passing through a size-selective inlet with a 50% efficiency cut-off at a 1.0-μm, 2.5-μm, and 10-μm aerodynamic diam-eter, respectively) were sampled during a 24-h period using a Grimm aerosol spectrometer (Grimm Aerosol Technik Ainring GmbH & Co. KG). Endotoxin concen-tration was measured in the PM10 fraction. Thoracic ultrasonography was performed and broncho-alveolar lavage fluid was collected for cytology and bacteriology. Average PM concentrations were 16.3 μg/m3 (standard deviation, SD: 17.1; range: 0.20–771), 25.0 μg/m3 (SD: 25.3; range: 0.50–144.9), and 70.3 μg/m3 (SD: 54.5; range: 1.6–251.2) for PM1.0, PM2.5, and PM10, respec-tively. Mean endotoxin in the PM10 fraction was 4.2 endotoxin units (EU)/μg (SD: 5.50; range: 0.03–30.3). Concentrations in air were 205.7 EU/m3 (SD: 197.5; range: 2.32–901.0). Lung consolidations with a depth of ≥1, ≥3, and ≥6 cm were present in 43.1% (146/339), 27.4% (93/339), and 15.3% (52/339) of the calves, re-spectively. Exposure to fine (PM1.0) PM fractions was associated with increased odds of lung consolidations of ≥1 cm (odds ratio, OR: 3.3; confidence interval (CI): 1.5–7.1), ≥3 cm (OR: 2.8; CI: 1.2–7.1), and ≥6 cm (OR: 12.3; CI: 1.2–125.0). The odds of having lung consolidations of ≥1 cm (OR: 13.9; CI: 3.4–58.8) and ≥3 cm (OR: 6.7; 1.7–27.0) were higher when endotoxin concentrations in the dust mass exceeded 8.5 EU/μg. Broncho-alveolar lavage fluid neutrophil percentage was positively associated with PM10 concentration, and epithelial cell percentage was negatively associated with this fraction. Concentration of PM2.5 was positively as-sociated with epithelial cell percentage and isolation of Pasteurella multocida. Although concentrations of fine dust are lower in calf barns than in poultry and pig housings, in this study they were associated with pneumonia in calves. There are over 500 species of the genus Artemisia in the Asteraceae family distributed over the globe, with varying potentials to treat different ailments. Following the isolation of artemisinin (a potent anti-malarial compound with a sesquiterpene backbone) from Artemisia annua, the phytochemical composition of this species has been of interest over recent decades. Additionally, the number of phytochemical investigations of other species, including those of Artemisia afra in a search for new molecules with pharmacological potentials, has increased in recent years. This has led to the isolation of several compounds from both species, including a majority of monoterpenes, sesquiterpenes, and polyphenols with varying pharmacological activities. This review aims to discuss the most important compounds present in both plant species with anti-malarial properties, anti-inflammatory potentials, and immunomodulating properties, with an emphasis on their pharmacokinetics and pharmacodynamics properties. Additionally, the toxicity of both plants and their anti-malaria properties, including those of other species in the genus Artemisia, is discussed. As such, data were collected via a thorough literature search in web databases, such as ResearchGate, ScienceDirect, Google scholar, PubMed, Phytochemical and Ethnobotanical databases, up to 2022. A distinction was made between compounds involved in a direct anti-plasmodial activity and those expressing anti-inflammatory and immunomodulating activities or anti-fever properties. For pharmacokinetics activities, a distinction was made between compounds influencing bioavailability (CYP effect or P-Glycoprotein effect) and those affecting the stability of pharmacodynamic active components.

The Shadow Program Committee (PC) is an initiative/program that provides an opportunity to Early-Career Researchers (ECRs), i.e., PhD students, postdocs, new faculty members, and industry practitioners, who have not been in a PC, to learn rst-hand about the peer-review process of the technical track at Software Engi- neering (SE) conferences. This program aims to train the next generation of PC members as well as to allow ECRs to be recog- nized and embedded in the research community. By participating in this program, ECRs will have a great chance i) to gain expe- rience about the reviewing process including the restrictions and ethical standards of the academic peer-review process; ii) to be mentored by senior researchers on how to write a good review; and iii) to create a network with other ECRs and senior researchers (i.e., Shadow PC advisors). The Shadow PC program was rst introduced to the SE research community at the Mining Software Repositories (MSR) confer- ence in 2021. The program was led by Patanamon Thongta- nunam and Ayushi Rastogi (Shadow PC Co-chairs) with support from Shadow PC Advisor Co-Chairs (Foutse Khomh and Serge Demeyer), PC Co-Chairs of the technical track (Meiyappan Na- gappan and Kelly Blincoe), and the General Chair of the con- ference, Gregorio Robles. To promote and facilitate the Shadow PC program at SE conferences in the future, this report provides details about the process and a re ection on the Shadow PC pro- gram during MSR2021.
