A man in his 20s presented with a 2-week history of fever, fatigue and diarrhoea. On arrival to the emergency department, he had clinical findings of sepsis. The care team initially suspected sepsis secondary to bacterial colitis and administered antibiotics. Further workup including a stool PCR assay for gastrointestinal pathogens failed to establish a diagnosis, and he had no evidence of immune compromise. Colonoscopy revealed mucosal ulceration presumed to be ulcerative colitis. Histopathology obtained after discharge revealed severe colitis with cytomegalovirus (CMV) inclusions. Serological studies indicated a primary CMV infection. To our knowledge, this is the first report of a primary CMV infection presenting as severe colitis and systemic disease in a young immunocompetent patient without underlying disease.

Cre-lox system is a powerful and widely used research tool developed to understand the specific role of genes. It allows to control the spatial and temporal expression of genes in experimental models. Several limitations including toxicity of Cre recombinase or incomplete excision of floxed loci have been reported in the past. To date, this is the first research study reporting that gut microbes also influence the expected phenotype of Paneth cell depletion in the genetically modified Math1lox/loxVilCreERT2 mouse model.

Early insights and opportunities of AI-powered pair-programming tools.

The Solid Oxide Fuel Cell (SOFC) offers major benefits, namely electrical efficiency and fuel versatility, for hybrid-electric aircraft propulsion. The combined development of novel SOFC concepts and corresponding manufacturing processes are regarded as key objectives for the SOFC activities at Airbus Central Research and Technology. Airbus is designing, manufacturing and testing SOFC concepts with the aim to achieve highest gravimetric power densities. Novel manufacturing technologies for metallic and ceramic materials have the potential to enable lighter functional layers for SOFCs with increased intrinsic mechanical stability and surface area. Different cell concepts with an optimized current collection are currently under development at Airbus. The micro-tubular and monolithic SOFC are seen as the most promising concepts, whereas around 2 kW/kg on a cell level has recently been achieved in a performance test at Airbus.

Record linkage is an important tool to enhance database integration. This is even more valuable in a scenario with more hefty budget cuts and a growing drop in response rate in traditional surveys. This strategy makes it possible to expand the crossing alternatives with variables not present in the original base. However, there are many different data pairing methods exposed in the literature. In this sense, the objective of this paper is to compare well-known methods of record linkage. The comparison was made in synthetic dataset. To compare the methods, it was adopted a quantitative approach based on the Precision, Recall, and F-Statistics metrics, using two comparison functions: Levenshtein and Jaro-Winkler. Among the six types of classifiers analyzed, the supervised methods had the best results.

However, the substrate-binding mode and the molecular mechanism of enzymatic PET hydrolysis are still under intense investigation, and controversial hypotheses have been presented. To help unravel the inherent mechanism of biocatalytic PET degradation at the atomic level, we performed solid-state NMR measurements of a cutinase from Thermobifida fusca (TfCut2) embedded in trehalose glasses together with chemically synthesized, amorphous 13C(═O)-labeled oligomeric PET. The resulting ternary enzyme-PET-trehalose glassy system enabled advanced solid-state NMR methods for real-time tracking of the enzymatic PET degradation and the investigation of PET chain dynamics. Combined with enhanced-sampling molecular dynamics simulations, specific enzyme–substrate interactions during the degradation process could also be monitored. Our results demonstrate that the PET chain is first cleaved by TfCut2 in blocks of at least one repeat unit and further to terephthalic acid and ethylene glycol. Moreover, the second step (formation of final hydrolysis products) appears to be rate-limiting in such reactions. The observed dynamic changes and interfacial protein contacts of 13C-labeled PET carbonyl groups suggest that only one PET repeat unit is bound to the enzyme during the degradation process while the rest of the PET chain is only loosely confined to the active site. These results, not accessible by using conventional solution enzyme samples and small nonhydrolyzable substrates, provide a better understanding of the biocatalytic PET degradation mechanism of polyester hydrolases, P.F. was supported by a scholarship of the Deutsche Bundesstiftung Umwelt (DBU grant 20018/565). Por muitos anos, criou-se um grande estigma na sociedade em relação à temática do HIV (Vírus da Imunodeficiência Humana) e da aids (Síndrome da Imunodeficiência Adquirida), seja pelos equívocos em relação à condição e ao modo de transmissão do vírus, seja pelo preconceito em relação às pessoas vivendo com HIV. É imprescindível que, como profissionais da área das ciências, possamos iniciar um processo de desmistificação de tais preconceitos ainda tão enraizados e, a partir de uma iniciativa didática, possamos promover a divulgação científica para estudantes nos últimos anos do ensino fundamental. O uso de Tecnologias da Informação e Comunicação (TIC) se mostra de grande utilidade pela familiaridade que se há com ela. O objetivo deste trabalho é abordar a temática HIV/aids por meio da construção de uma cartilha e produção de podcast como ferramentas didáticas para o ensino de ciências. Metodologicamente, o estudo foi desenvolvido pela pesquisa bibliográfica (para o desenvolvimento da cartilha) e entrevista (podcast).

Higher education plays a crucial role in supporting a society based on sustainable development through the facilitation of students’ acquisition of competencies for sustainable development. A suitable arena in which to integrate these competencies can be courses built on project-based learning, though knowing how to support the students’ learning can be difficult and a continued challenge. In this work, we study first and second year bachelor level PBL courses and examine the effect of choosing a project theme related to sustainability. Specifically, we look at the students’ own assessment of integration of sustainability aspects in their projects, and their development of the normative competency, critical thinking competency, and self-awareness competency fromUNESCO’s key competencies for sustainability, as a function of project theme. Through a survey, where students were asked to assess themselves and their own projects on a seven-point Likert scale, we found that while having sustainability-related project themes does have some effect, the effect is limited compared to the development of other competencies in the course. Along with how the project theme affects the development of the investigated competencies, the need for targeted support in order to facilitate the students’ development of sustainability related competencies is discussed.

Focus: To get to know some of the creatures in the deep-sea that use color and the lack of light to camouflage themselves. Learning objectives: With these TWO activities, we use vision to explore life forms in the deep sea which have adapted to extreme conditions. Through the activities, the pupils will also become familiar with some organisms well-hidden at the bottom of the ocean. Focus: To learn about foraminifera through practicing and presenting the song “The Foraminifera Boogie”. Pupils and teachers can also decide to play instruments along with the recorded soundtrack. Learning objectives: In this activity, pupils will practice listening to one another and the music. They will learn lyrics and use their voices to sing the song. This can be combined with playing actual musical instruments or using instruments the pupils have made themselves (for example see the activity “Cold Seeps Symphony”). This activity brings together music with the scientific story of the Foraminifera. Las experiencias formativas basadas en Aprendizaje-Servicio (ApS) que se vienen implementando en los últimos años en diferentes contextos universitarios tanto en España como en otros países permiten demostrar la validez de esta metodología como herramienta didáctica efectiva y de calidad en la enseñanza superior. El proyecto TRADAPS, en desarrollo en la Universidad de Salamanca dentro del grado en Traducción e Interpretación, pretende contribuir a corroborar la idoneidad de este enfoque en el ámbito concreto de los estudios de traducción. Este proyecto inició su andadura en el curso 2021-2022 con el propósito de diseñar una línea de trabajo en ApS que permitiera al alumnado realizar Trabajos de Fin de Grado en colaboración con ONG que comparten la misión de mejorar la calidad de vida y la inclusión social de colectivos de migrantes en situación de vulnerabilidad motivada por barreras lingüísticas y culturales. Actualmente, se realiza el seguimiento de dos Trabajos de Fin de Grado desarrollados en colaboración con las organizaciones españolas Cepaim y Salamanca Acoge., Recent training experiences based on Service-Learning (SL) conducted at university level in Spain and other countries demonstrate the validity of this methodology as an effective, high-quality training tool in higher education contexts. The TRADAPS project currently underway within the BA degree in Translation and Interpreting taught at the University of Salamanca was proposed with the aim of substantiating the suitability of this approach in the specific field of translation studies. This project, launched in 2021-2022, was aimed at designing a line of work in ApS which could enable students to complete final degree projects or dissertations in collaboration with particular NGOs which share the mission of improving the quality of life and enhancing social inclusion of migrant communities in situation of vulnerability due to linguistic and cultural barriers. Two Bachelor’s Degree Final Projects are currently being developed in collaboration with two Spanish NGOs: Cepaim and Salamanca Acoge.

Die roboterassistierte abdominothorakale Ösophagusresektion ist ein etabliertes Verfahren und wird am Universitätsklinikum Heidelberg circa 60 Mal im Jahr durchgeführt. Das Operationsprinzip besteht in der transthorakalen und abdominellen Entfernung des Ösophagus sowie einer onkologischen Lymphadenektomie und stellt somit einen Zwei-Höhlen-Eingriff dar. Die Rekonstruktion erfolgt mittels Magenschlauchbildung und anschließendem Magenhochzug.

A protocol for the mechanochemical synthesis of copper(I)/N-heterocyclic carbene complexes using cheap and readily available K3PO4 as base has been developed. This method employing a ball mill is amenable to typical simple copper(I)/NHC complexes but also to a sophisticated copper(I)/N-heterocyclic carbene complex bearing a guanidine moiety. In this way, the present approach circumvents commonly employed silver(I) complexes which are associated with significant and undesired waste formation and the excessive use of solvents. The resulting bifunctional catalyst has been shown to be active in a variety of reduction/hydrogenation transformations employing dihydrogen as terminal reducing agent.

Devido ao Estatuto da Pessoa com Deficiência (PcD), desde 2015, diversas universidades têm promovido políticas de inclusão nos cursos superiores e, apesar de todo esforço, os resultados advindos dessas ações não são suficientes para garantir a permanência deles nas universidades, principalmente aquelas relacionadas ao quesito arquitetônico ou espacial. Devido a esta problemática, este tabalho teve por objetivo identificar demandas de acessibilidade e indicar, através de ferramentas de Cadastro Territorial e Multifinalitário e tomada de decisão, intervenções arquitetônicas de forma a garantir o acesso e a permanência de PcD na Universidade Federal de Santa Catarina (UFSC). Para tanto, foram realizados diversos levantamentos in loco, remotamente e via questionários, para identificar problemas de acessibilidade arquitetônica no Campus Professor David Ferreira Lima da UFSC, a fim de validar a metodologia. Através dos resultados obtidos foi possível verificar a existência de diversos problemas de acessibilidade arquitetônica no Campus, concluindo que o método aplicado se mostrou válido para indicar as intervenções que se fazem necessárias. Como resultados também foram sugeridas alterações para atender a acessibilidade através de projetos. Foi concluído, também, que a integração de PcD à equipe fortaleceu e ampliou a formação humana e cidadã dos membros envolvidos, principalmente os estudantes de Engenharia Civil, futuros responsáveis, também, por realizar construções mais acessíveis.

Understanding the nucleation and growth mechanisms of nanocrystals under hydro- and solvothermal conditions is key to tailoring functional nanomaterials. High-energy and high-flux synchrotron radiation is ideal for characterization by powder X-ray diffraction and X-ray total scattering in real time. Different versions of batch-type cell reactors have been employed in this work, exploiting the robustness of polyimide-coated fused quartz tubes with an inner diameter of 0.7 mm, as they can withstand pressures up to 250 bar and temperatures up to 723 K for several hours. Reported here are recent developments of the in situ setups available for general users on the P21.1 beamline at PETRA III and the DanMAX beamline at MAX IV to study nucleation and growth phenomena in solvothermal synthesis. It is shown that data suitable for both reciprocal-space Rietveld refinement and direct-space pair distribution function refinement can be obtained on a timescale of 4 ms.

The microbial metabolic quotient (MMQ; mg CO2-C mg MBC-1 h-1), defined as the amount of microbial CO2 respired (MR; mg CO2-C kg soil-1 h-1) per unit of microbial biomass C (MBC; mg C kg soil-1), is a key parameter for understanding the microbial regulation of the carbon (C) cycle, including soil C sequestration. Here, we experimentally tested hypotheses about the individual and interactive effects of multiple nutrient addition (NPK+micronutrients) and herbivore exclusion on MR, MBC, and MMQ across 23 sites (5 continents). Our sites encompassed a wide range of edaphoclimatic conditions, thus we assessed which edaphoclimatic variables affected MMQ the most and how they interacted with our treatments. Soils were collected in plots with established experimental treatments. MR was assessed in a five-week laboratory incubation without glucose addition, MBC via substrate-induced respiration. MMQ was calculated as MR/MBC and corrected for soil temperatures (MMQsoil). Using LMMs and SEMs, we analysed how edaphoclimatic characteristics and treatments interactively affected MMQsoil. MMQsoil was higher in locations with higher mean annual temperature, lower water holding capacity, and soil organic C concentration, but did not respond to our treatments across sites as neither MR nor MBC changed. We attributed this relative homeostasis to our treatments to the modulating influence of edaphoclimatic variables. For example, herbivore exclusion, regardless of fertilization, led to greater MMQsoil only at sites with lower soil organic C (-1.7%). Our results pinpoint the main variables related to MMQsoil across grasslands and emphasize the importance of the local edaphoclimatic conditions in controlling the response of the C cycle to anthropogenic stressors. By testing hypotheses about MMQsoil across global edaphoclimatic gradients, this work also helps to align the conflicting results of prior studies.

A partir da centralização da problemática da violência sexual perpetrada contra mulheres em âmbito de relações afetivas/conjugais na ordem hegemônica e androcêntrica do gênero, o presente artigo tem como escopo demonstrar a maneira com a qual o Direito alicerçou e continua a alicerçar o discurso sexista. Nesse sentido, busca-se detectar de onde tais formações discursivas emergem, a fim de desconstruí-las e dar espaço a falas e práticas outras. Para tanto, utilizar-se-á da teoria queer e das perspectivas feministas decoloniais. A metodologia utilizada nesse artigo foi a de natureza exploratória, com abordagem qualitativa, partindo-se de uma análise legislativa e doutrinária, bem como do exame de acórdãos proferidos no Tribunal de Justiça do Mato Grosso do Sul (TJMS) entre os anos de 2016 e 2020, por meio do procedimento bibliográfico e documental.

Profound knowledge of nerve variations is essential for clinical practice. It is crucial for interpreting the large variability of a patient's clinical presentation and the different mechanisms of nerve injury. Awareness of nerve variations facilitates surgical safety and efficacy. Clinically significant anatomical variations can be classified into two main groups: variability in the course of the nerve and variability of structures surrounding the nerve. In this review article we focus on the most common nerve variants of the upper extremity and their clinical relevance.

To enhance their properties, such as stability or binding affinity, they are usually chemically modified. This includes, among other techniques, cyclization of the peptide chain by bridging, modifications to the backbone, and incorporation of unnatural amino acids. One approach previously established, is the use of halogenated aromatic amino acids. In principle, they are thereby enabled to form halogen bonds (XB). In this study, we focus on the -R-CF2X moiety (R = O, NHCO; X = Cl, Br) as an uncommon halogen bond donor. These groups enable more spatial variability in protein–protein interactions. The chosen approach via Fmoc-protected building blocks allows for the incorporation of these modified amino acids in peptides using solid-phase peptide synthesis.Results and Discussion: Using a competitive fluorescence polarization assay to monitor binding to Mdm4, we demonstrate that a p53-derived peptide with Lys24Nle(&epsiv;NHCOCF2X) exhibits an improved inhibition constant Ki compared to the unmodified peptide. Decreasing Ki values observed with the increasing XB capacity of the halogen atoms (F ≪ Cl < Br) indicates the formation of a halogen bond. By reducing the side chain length of Nle(&epsiv;NHCOCF2X) to Abu(γNHCOCF2X) as control experiments and through quantum mechanical calculations, we suggest that the observed affinity enhancement is related to halogen bond-induced intramolecular stabilization of the α-helical binding mode of the peptide or a direct interaction with His54 in human Mdm4.Graphical Abstract: Keywords: halogen bonding, solid-phase peptide synthesis, unnatural amino acids, fluorescence polarization assay, oncological target, MdmX

Zusammenfassung Einleitung Das Erscheinungsbild des Bauchnabels nach DIEP-Lappenplastik oder Abdominoplastik spielt eine wesentliche Rolle bei der ästhetischen Wahrnehmung (1). Obgleich der Nabel keine Funktion hat, besteht kein Zweifel an der Bedeutung, die die Nabelform für das Selbstwertgefühl der Patientinnen gerade nach einer Brustkrebserkrankung hat (2). In der vorliegenden Studie haben wir zwei der bevorzugten in der Literatur beschriebenen Techniken an 72 Patientinnen hinsichtlich des ästhetischen Ergebnisses, der Komplikationen und der Sensibilität verglichen: das kaudale Läppchen (kuppelförmige Form) und die querovale Form des Nabels. Patienten und Methode Zweiundsiebzig Patientinnen, die zwischen Januar 2016 und Juli 2018 eine DIEP-Lappenplastik zur Brustrekonstruktion erhielten, wurden in dieser Studie retrospektiv erfasst. Es wurden zwei Techniken zur Nabelrekonstruktion verglichen: die querovale Nabelform und die Umbilikoplastik unter Verwendung eines kaudalen Läppchens, welche in einer kuppelförmigen Form des Bauchnabels resultiert. Um die ästhetischen Ergebnisse zu vergleichen, wurde eine Evaluation durch die Patientinnen und eine Bewertung durch drei unabhängige Plastische Chirurg*innen mindestens 6 Monate postoperativ durchgeführt. Patientinnen und Chirurg*innen wurden gebeten, das allgemeine Erscheinungsbild des Nabels, einschließlich der Narbenbildung und der Form, mit einer Punktzahl zwischen 1 und 6 zu bewerten (1=Sehr gut, 2=gut, 3=befriedigend, 4=ausreichend, 5=mangelhaft, 6=ungenügend). Des Weiteren wurde die Sensibilität des Nabels durch Befragung der Patientinnen und das Auftreten von Wundheilungsstörungen untersucht. Ergebnisse Beide Techniken zeigten ähnliche Werte der ästhetischen Zufriedenheit (p=0,49) im Rahmen der Selbsteinschätzung durch die Patientinnen. Die Plastischen Chirurg*innen bewerteten die Technik mit kaudalem Läppchen als signifikant besser gegenüber der Nabelplastik mit querovaler Form (p=0,042). Beim kaudalen Läppchen traten mehr Wundheilungsstörungen auf (11,1%) im Vergleich zum querovalen Nabel. Dies war jedoch nicht signifikant (p=0,16). Eine operative Revision war nicht erforderlich. Beim Nabel mit kaudalem Läppchen zeigte sich eine Tendenz zur Verbesserung der Sensibilität (60% gegenüber 45%), die jedoch nicht signifikant war (p=0,19). Schlussfolgerung Die Patientenzufriedenheit zeigte ähnliche Ergebnisse für die beiden Methoden der Umbilikoplastik. Beide Techniken wurden im Durchschnitt mit einem guten Ergebnis bewertet. Andererseits bewerteten die Chirurg*innen die Umbilikoplastik mit kaudalem Läppchen als ästhetisch ansprechender.

Background: Preterm infants are at risk for neurodevelopmental deficits. An association between retinopathy of prematurity (ROP) and impaired cognitive outcome has already been described. However, less is known about the impact of ROP on visual motor integration (VMI), which is a prerequisite not only for fine motor abilities but also for further school skills. Therefore, the aim of this study was to retrospectively investigate the impact of ROP on VMI at preschool age. Methods: The study was conducted at the Medical University of Vienna, including patients born between January 2009 and December 2014 with a gestational age of less than 30 weeks and/or a birth weight of less than 1,500 g. VMI was determined by Beery-Buktenica Developmental Test of Visual Motor Integration (Beery VMI) at the age of 5 years. Results: Out of 1,365 patients, 353 met inclusion criteria for this study. Two hundred sixteen of them had no ROP, while 137 had ROP (stage 1: n = 23, stage 2: n = 74, stage 3: n = 40). Mean value of the Beery VMI score was significantly lower in the ROP group compared to the No-ROP group (90 ± 16 vs. 99 ± 14; p < 0.01). By correcting for other important medical conditions, ROP still had a significant impact on Beery VMI score (p < 0.01). Particularly, lower scores were found for stage 2 (p < 0.01) and stage 3 (p < 0.01). Conclusion: Beery VMI scores were significantly lower in preterm infants with ROP stage 2 and 3 than in infants without ROP. This study shows the negative impact of ROP on VMI skills at preschool age, even after adjustment for key demographic and medical characteristics.

International audience; The rapid identification of early hits by fragment-based approaches and subsequent hit-to-lead optimization represents a challenge for drug discovery. To address this challenge, we created a strategy called 'DOTS' that combines molecular dynamic simulations, computer-based library design (chemoDOTS) with encoded medicinal chemistry reactions, constrained docking, and automated compound evaluation. To validate its utility, we applied our DOTS strategy to the challenging target syntenin, a PDZ domain containing protein and oncology target. Herein, we describe the creation of a 'best-in-class' sub-micromolar small molecule inhibitor for the second PDZ domain of syntenin validated in cancer cell assays. Key to the success of our DOTS approach was the integration of protein conformational sampling during hit identification stage and the synthetic feasibility ranking of the designed compounds throughout the optimization process. This approach can be broadly applied to other protein targets with known 3D structures to rapidly identify and optimize compounds as chemical probes and therapeutic candidates.

The structure of power underlying the hegemonic control Chichen Itza held over the Northern Maya Lowlands has been debated for decades. In this article, we present the idea of a dominant discourse on masculinities, which played a fundamental role in both practice and on a symbolic level among the strategies designed to support this emblematic pre-Columbian capital. Our discussion of archaeological evidence will focus on spaces where men are represented, where they would meet and carry out rituals. We contend that gallery-patios such as Structure 2D6 served as instruction and socialization locales for groups of warriors. The architectural configuration of this building is very similar to a series of venues at Chichen Itza and other Mesoamerican cities. In these spaces, associated iconography depicts male individuals in processions and ritual practice, including sacrifice and self-sacrifice. We argue that the gallery of Structure 2D6 was a semi-public, performative space, whose theatricality combined the central alignment of a sacrificial stone and a throne or altar with the presence of several patolli boards carved into the building's plaster floor. Chemical analyses of plastered surfaces testify to intense activities taking place around all three of these features.
