Human lactoferrin (hLF) is an iron-binding protein with antimicrobial and immunomodulatory activities. hLF cDNA was transferred into grass carp via electroporated sperm. The production of transgenic fish was as high as 55% tinder the best parameters. 2(11) pulses and 20-min incubation. 103. The Classification of Pavement Crack Image Based on Beamlet Algorithm

Pavement distress, the various defects such as holes and cracks, represent a significant engineering and economic concern. This paper based on Beamlet algorithm using MATLAB software to process the pavement crack images and classify the different cracks into four types: horizontal, vertical, alligator, and block types. Experiment results show that the proposed method can effectively detect and classify of the pavement cracks with a high success rate, in which transverse crack and longitudinal crack detection rate reach to 100%, and alligator crack and block crack reach more than 85%.

Recombinant “all-fish” growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant “fast-growing” effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the F1 generation were genetically analyzed, which gave solid evidence for the hypothesis that 2–3 chromosomes are integrated with transgene. In addition, the distinct biological effects for multisite-integrated transgenes were further discussed. The present study opens a door for the breeding of “fast-growing” transgenic fish.

A DR300 Broad Band ADCP mounted on a vessel moving at a speed of 2–3 m/s was used to measure the profile of suspended sediment concentrations (SSCs) at the entrance to Jiaozhou Bay, Shandong Peninsula, where the water is characterized by low SSCs. The echo intensity data produced by the ADCP were regressed against the SSCs derived using the filtration method. The results show that the calibrated relationship can be used to calculate the SSC, with a relative error of 30%. Therefore, it is feasible to measure the SSC (even if the concentration is low) using the ADCP mounted on a moving vessel. Compared with OBS, ABS and other instruments for SSC measurements, the ADCP represents a potentially powerful tool to retrieve SSC data in continental shelf waters.

Angular correlations between charged trigger and associated particles are measured by the ALICE detector in p-Pb collisions at a nucleon-nucleon centre-of-mass energy of 5.02 TeV for transverse momentum ranges within 0.5 < P-T,P-assoc < P-T,P-trig < 4 GeV/c. The correlations are measured over two units of pseudorapidity and full azimuthal angle in different intervals of event multiplicity, and expressed as associated yield per trigger particle. Two long-range ridge-like structures, one on the near side and one on the away side, are observed when the per-trigger yield obtained in low-multiplicity events is subtracted from the one in high-multiplicity events. The excess on the near-side is qualitatively similar to that recently reported by the CMS Collaboration, while the excess on the away-side is reported for the first time. The two-ridge structure projected onto azimuthal angle is quantified with the second and third Fourier coefficients as well as by near-side and away-side yields and widths. The yields on the near side and on the away side are equal within the uncertainties for all studied event multiplicity and p(T) bins, and the widths show no significant evolution with event multiplicity or p(T). In order to study whether marrow stromal cells (MSCs) can be induced into nerve-like cells in vitro, and the mechanism, the MSCs in Wistar rats were isolated and cultured, and then induced with DMSO and BHA in vitro. The expression of specific marking proteins in neurons, glia and neural stem cells were detected before preinduction, at 24 h of preinduction, at 6 h, 24 h, and 48 h of neuronal induction by using immunohistochemistry and Western blotting. The ultrastructural changes after the inducement were observed. The results showed that after the inducement, many MSCs turned into bipolar, multipolar and taper, and then intersected as network structure. At the same time, some MSCs had the typical neuron-like ultrastructure. Immunohistochemistry revealed that NeuN and Nestin expression was detectable after inducement, but there was no GFAP and CNP expression. Western blotting showed the expression of Nestin was strong at 6 h of neuronal induction, and decreased at 24 h, 48 h of the induction. NeuN was detectable at 6 h of neuronal induction, and increased at 24 h, 48 h of the induction. It was concluded MSCs were induced into neural stem cells, and then differentiated into neuron-like cells in vitro.

The ALICE experiment at the LHC has studied J/psi production at mid-rapidity in pp collisions at sqrt{s}=7 TeV through its electron pair decay on a data sample corresponding to an integrated luminosity L_int = 5.6nb-1. The fraction of J/psi from the decay of long-lived beauty hadrons was determined for J/psi candidates with transverse momentum p_t>1.3 GeV/c and rapidity |y| 1.3 GeV/c, |y| < 0.9) = 8.3 +- 0.8(stat.) +- 1.1(syst.) + 1.5 - 1.4(syst. pol.) micro barn. The cross section for the production of b-hadrons decaying to J/psi with p_t>1.3 GeV/c and |y, Comment: The most important modification with respect to v1 is a correction factor that is applied to all cross section values, which comes from the Erratum to "K. 109. Multi-objective imperfect maintenance for dependent competing risk systems with multiple degradation processes

- Abstract
Multiple competing risks are one of the important topics in reliability field, especially degradation processes and random shocks. This research aims to relax the independent assumption by considering that there exist dependent relationships not only among multiple degradation measures but also between degradation measure and random shocks. In reality, many systems have multiple components with more than one degradation measure which is dependent with each other due to their interplaying functions or common usage history. Independent assumption may underestimate system reliability estimation under many cases. Random shocks will also contribute to the system failure through two ways: (1) one is working directly on the degradation processes; (2) the other is causing immediate failure to the system. We develop a new methodology to formulate the reliability prediction model for the gradually degradating systems subject to multiple dependent competing risks of degradation processes and random shocks. Two kinds of random shocks are considered: (1) fatal shocks, which fail the system immediately; (2) non-fatal shocks, which exhibit two effects on the system degradation process, including sudden degradation increment and degradation rate acceleration. The dependency between degradation processes and random shocks are modulated by a time-scaled covariate factors while the dependency among degradation processes are fitted by copula method. Also the reliability and state probability estimation for the systems are derived under the research scope of multi-state system using both analytical and Monte Carlo simulation for the dependent competing-risk systems. Different maintenance policy models involving imperefect preventive maintenance for this dependent model are introducted and compared with each other. Multi-objective optimization is applied to consider two important targets simultaneously in maintenance issues, including long-run expected cost rate and system availability.

Sliding is a common behavior used by driver on level road during vehicle operation. So it has practical significance to analyze the variation discipline of sliding vehicle's longitudinal dynamic parameters, such as sliding speed, deceleration rate, coasting resistance (including rolling resistance and air drag) in order to operate vehicle properly and reasonably. Taking some diesel passenger vehicle as experimental object and combining theoretical analysis with experimental verification, thermodynamic models about vehicle's longitudinal dynamic parameters were formulated and developed by use of least square principle and linear regression in this work. The models show that sliding speed and deceleration rate decrease with the sliding time, while coasting resistance and deceleration rate decrease with sliding speed. But the reduction rate of sliding speed and coasting resistance is not consistent. Finally, the accuracy of these models is scrutinized against actual repeated test results from the vehicle. In the final analysis, it is seen that there is an excellent agreement in all of these evaluations.

The using of self-developed shock force sensor to detect the steel ball size whether meet the requirements, stamping equipment running status whether is normal; the using of innovative design eddy current flaw detection sensor which is used on-line to detect defects of the steel ball materials, to effectively control the quality of the steel ball; the using of special signal processing modules and embedded computer systems to achieve real-time sensor signal acquisition and processing, to quickly and accurately determine the status of equipment operation and product quality; the using of a pneumatic schematic design of the products qualified for high-speed sorting institutions to realize the pass rate of steel ball sorting. Experiment is proved that the system can be effectively monitor the status of high-speed stamping equipment and to be achieved the separation of the steel ball which is above 98% pass rate, the control system is stable and reliable.

To improve performance of traffic signal control system in urban area, a novel method is proposed in this paper. The roads, vehicles and the traffic control systems are all modeled as intelligent agents. Wireless communication network provides the possibility of the cooperation of vehicles and roads. Based on all the information from vehicles and roads, a traffic control policy can be planned online according to the updated situation on the roads. The optimum intersection signals can be learned automatically on line based on reinforcement learning. An intersection signal control system is studied as an example of the method with a Q-learning based algorithm. The simulation results show that the proposed intersection signal control can improve traffic efficiency by about 30% over the traditional method.

The yeast vectors described, pYEV and pYEVB, were designed for parallel polymerase chain reaction (PCR) cloning of open reading frames (ORFs) and immediate protein expression in Pichia pastoris. The pYEV vector was used to clone PCR fragments obtained by using Taq or similar polymerase mixes, which leave an A-base overhang. The other vector pYEVB, with the same features for blunt-end ligation of PCR products, was developed to be complementary to pYEV. These two plasmids were linearized using the restriction enzymes BfuI and SchI, respectively. The purified PCR products, without any other treatments, were cloned into the linearized vectors, followed by selection on plates supplemented with X-gal. Only desired recombinants carrying the target gene in the correct orientation can give typical blue colonies. This screening technique is based on a lacO reconstruction strategy that can produce a full-length lacO to exhaust endogenous LacI and switch on the transcription of lacZ in the host. The recombinant plasmids extracted from the blue colonies can be linearized by SalI and transformed into P. pastoris for immediate expression. By using these two vectors, researchers could be saved from the tedious and time-consuming conventional cloning procedures.

115. Is There a Difference in the Association between Percent Mammographic Density and Subtypes of Breast Cancer? Luminal A and Triple-Negative Breast Cancer

BACKGROUND: Mammographic density is a potentially modifiable risk factor for breast cancer. To what extent mammographic density is a predictor for both hormone receptor-positive and hormone receptor-negative tumors is unclear. Even less is known about whether mammographic density predicts subtypes of breast cancer defined by expression status of the three receptors: estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor-2 (HER-2). METHODS: We estimated the association of percent mammographic density with subtypes of invasive breast cancer among 479 population-based female breast cancer patients and 376 control subjects ages 35 to 64 years. The expression status of ER, PR, and HER-2 was assessed using immunohistochemistry methods in a single laboratory. We considered ER+ or PR+ plus HER-2- tumors as luminal A breast cancer and ER-/PR-/HER-2- tumors as triple-negative breast cancer. We used unconditional logistic regression methods to estimate odd ratios (95% confidence intervals) for both case-control and case-case comparisons. RESULTS: Mammographic density was associated with increased risk of both invasive breast cancer subtypes, luminal A and triple-negative, in the case-control analysis. Results from case-case comparisons yielded no differences between the two subtypes among all women combined or in analyses done separately by race (White versus African American women) or menopausal status (premenopausal versus postmenopausal women; all P values > 0.05). CONCLUSIONS: Our results suggest that percent mammographic density is positively associated with both luminal A and triple-negative breast cancer.

VI is an instrument based on computer, the application of which is one of the hottest topics in computer applications development and research fields. Remote inspection is the most emergent problem in the product security inspection laboratory. The application of networked VI becomes necessary and feasible. In this paper, we developed a Distributed Real-time Online Inspection System to control VI remotely using LabVIEW. Firstly, the architecture of the system is introduced. Secondly, the design of system and key techniques are discussed. Finally, the test of controlling VI remotely is successfully performed. It provides us a high reference value for the VI networked inspection. Since the System use the bottom layer programming technique which makes the bottom layer interfaces independent and multithreading technique ensures the system’s good transplantability, stability, reliability, and efficiency. Our experiments show that the system works well in both local and remote inspection.

The enthalpies of the crystal transformation from I to II and from II to III and the melting enthalpy of 2,2,2-trinitroethyl-4,4,4-trinitrobutyrate (TNETB) are determined by means of Calvet microcalorimeter. On cooling, the supercooing from liquid to solid does not appear, and form II will transform to form I when 71.8° C is reached. The phase diagrams of TNETB-2,4,6-trinitrotoluene (TNT) and TNETB-polyester systems have been constructed by differential scanning calorimetry (DSC). The eutectic temperatures are 56° C and 34° C respectively. The compositions corresponding to the eutectic points are 52 and 46 weight percent TNETB respectively.

Gonadotropin-releasing hormone (GnRH) is a conservative neurodecapeptide family, which plays a crucial role in regulating the gonad development and in controlling the final sexual maturation in vertebrate. Two differing cGnRH-II cDNAs of common carp, namely cGnRH-II cDNA1 and cDNA2, were firstly cloned from the brain by rapid amplification of cDNA end (RACE) and reverse transcription- polymerase chain reaction (RT-PCR). The length of cGnRH-II cDNA1 and cDNA2 was 622 and 578 base pairs (bp), respectively. The cGnRH-II precursors encoded by two cDNAs consisted of 86 amino acids, including a signal peptide, cGnRH-II decapeptide and a GnRH-associated peptide (GAP) linked by a Gly-Lys-Arg proteolytic site. The results of intron trapping and Southern blot showed that two differing cGnRH-II genes in common carp genome were further identified, and that two genes might exist as a single copy. The multi-gene coding of common carp cGnRH-II gene offered novel evidence for gene duplication hypothesis. Using semi-quantitative RT-PCR, expression and relative expression levels of cGnRH-II genes were detected in five dissected brain regions, pituitary and gonad of common carp. With the exception of no mRNA2 in ovary, two cGnRH-II genes could be expressed in all the detected tissues. However, expression levels showed an apparent difference in different brain regions, pituitary and gonad. According to the expression characterization of cGnRH-II genes in brain areas, it was presumed that cGnRH-II might mainly work as the neurotransmitter and neuromodulator and also operate in the regulation for the GnRH releasing. Then, the expression of cGnRH-II genes in pituitary and gonad suggested that cGnRH-II might act as the autocrine or paracrine regulator.

Objective Heat shock protein 90 (HSP90) is a chaperone for several client proteins involved in transcriptional regulation, signal transduction, and cell cycle control. HSP90 is abundantly expressed by a variety of tumour types and has been recently targeted for cancer therapy. The objective of this study is to determine the role of Hsp90 in regulating the migration of Mesenchymal stem cells and to determine the mechanism. We hypothesised that inhibition of Hsp90 impairs the MSCs migration via PI3K/Akt and ERK1/2 signalling pathways. Methods The MSCs were cultured from femoral and tibia. The ability for MSCs cells to migrate is to be determined by the wound healing assay and transwell assay. The activity of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) were estimated by gelatin –zymography. The mRNA levels of MMP-2, MMP-9, CXCR4 and VCAM-1 were detected by real-time PCR. The protein expression of MMP-2, MMP-9 and ERK1/2, phospho-ERK1/2, Akt and phospho-Akt were determined by Western-blot. Results Treatment with RhHsp90α significantly enhances MSCs migration from 9.83±2.48 to 48.65±2.81 cells. Treatment with sirhsp90α significantly decreased MSCs migration compared with treatment of hsp90α from 63.33±9.61 to 13.00±4.38 cells. Pretreat with 17-AAG, wortmannin, U0126, decreased MSCs migration to 13.33±1.29, 15.33±2.1, 16.5±3.3 cells, respectively. Treatment with RhHsp90α enhanced the MSCs secretion of MMP-2 and MMP-9, as well as significantly increasing the activity of MMP-9, and increasing the expression of CXCR4 and VCAM-1. PI3K/Akt and ERK signalling pathways mediate the promotion of MSCs migration by RhHsp90α. Conclusions Our data showed that Hsp90 promotes MSCs migration, elevate the expression of MMP-2, MMP-9, CXCR4 and VCAM-1. PI3K/Akt and ERK signalling pathways mediates these effects. Hsp90 is a candidate drug for enhancement of MSCs migration.

Objective Coronary artery disease (CAD) has emerged as the dominant etiologic factor in patients with heart failure. The Objective of this work was to identify new biomarkers and establish serum protein fingerprint models for early diagnosis and preoperative evaluation of luminal stenosis severity of CAD. Methods 48 eligible case subjects, having >50% luminal stenosis in at least one major coronary artery (defined as coronary artery disease (CAD) by angiography), 39 patients with luminal stenosis between 25% and 50% (defined as coronary atherosclerosis), and 52 eligible health individuals, were recruited randomly in this study. The above 139 samples were analysed by Surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS). Results The clinical and biochemical characteristics in three groups matched well. To detect CAD, thirty samples from patients with CAD (stenosis ≥50%) and thirty-two samples from the control group were analysed and designated as pattern 1. This pattern 1 model comprised ten potential biomarkers with m/z of 4276, 5326, 4481, 4320, 4205, 5814, 5551, 3689, 5344 and 6134 Da, respectively. The peaks at 5326, 5814, 5551, 5344, 6134 Da were expressed greatly in samples from patients with CAD, but weakly expressed in healthy individuals; and the other five peaks were expressed greatly in healthy individuals but weakly expressed in patients with CAD. Pattern 1 had a specificity of 78.1% and a sensitivity of 80.0%, as evaluated by leave-one-out crossvalidation. The remaining 38 serum samples, as a blind test set, were analysed on the second day to test pattern 1. The specificity and sensitivity of the blind test were 83.3 and 72.2%, respectively. To distinguish different severities of coronary artery stenosis, twenty-seven samples from patients with coronary atherosclerosis (between 25% and 50%) were analysed and compared with thirty CAD (stenosis ≥50%) patient samples. Pattern 2 was established similarly to distinguish coronary atherosclerosis (stenosis between 25% and 50%) from CAD. Pattern 2 comprised seven potential biomarkers with m/z of 3280, 3689, 5326, 4289, 6135, 3164 and 6439 Da, respectively. While the peaks with m/z 5326 and 6135 Da, were expressed more significantly in CAD samples than in coronary atherosclerosis (stenosis between 25% and 50%), the other five biomarkers appeared to be expressed in the opposite manner. This model had a specificity of 92.6% and a sensitivity of 93.3%, as evaluated by leave-one-out crossvalidation. Pattern 2 was tested blindly in another 30 serum samples on the second day. The specificity and sensitivity of the blind test were 75.0% and 77.8%, respectively. Conclusion Taken together, the SELDI-TOF-MS technique combined with bioinformatics approaches can not only facilitate the discovery of better biomarkers for CAD and its severity, but also provide a useful tool for molecular diagnosis.

Objective ischaemic heart disease is the leading cause of morbidity and mortality all over the world. Cardiomyocytes from bone marrow-derived mesenchymal stem cells (MSCs) offer great potential for repairment of the infracted heart. However, this approach has been limited by inefficient differentiation of MSCs into cardiomyocytes. To overcome such a problem, the underlying regulation mechanisms for cardiac differentiation should be elucidated. MicroRNAs (miRNAs) are small noncoding RNAs of ∼23 nucleotides that control post-transcriptional gene expression. Recently, miRNA has been widely shown to regulate key cellular events such as cell proliferation, cell differentiation. The purpose of this study is to determine the role of miRNAs during cardiac differentiation from MSCs. Method Firstly, we established a model of cardiac differentiation from rat bone marrow-derived MSCs using 10 μM 5-Aza, and performed a global miRNA analysis using EXIQON miRNA array to identify characteristic miRNA at different stage of differentiation. After being validated by real-time qRT-PCR and target gene prediction, several miRNAs such as miRNA-145 were further chosen to reveal its function during cardiac differentiation. Results miRNA profiling revealed that miR-145 expression increased during cardiac differentiation, especially at 12 days of treatment (2.25-fold change vs untreated MSCs). Compared to other tissues such as liver, brain, kidney, miRNA-145 expression is highest in rat heart tissue. Gain-of-function methods using pre-miR-145 showed the enchancement of cardiac differentiation, confirmed by immunocytochemical staining with cardiac-specific myosin heavy chain antibody. Conclusion miR-145 may be a critical regulator for cardiomyocyte lineage in our 5-Aza-induced differentiation system.

Single later blastula nuclei from AB strain of zebrafish (Danio rerio) were transplanted into enucleated unfertilized eggs of Long fin strain. Of 1119 cloning embryos, 14 reconstructed embryos developed into fry. DNA fingerprinting systems of the cloned fish were similar to those of the nuclear donor fish, but were distinctly different from those of the unclear recipient fish. It confirmed that the genetic material originated from nuclear donor cell other than from nuclear recipient egg. The research suggested that the basic technique for nuclear transplantation performed with different strains of zebrafish has made a breakthrough. It should be helpful for the study of some important developmental problems such as gene function, the regulation of gene expression during animal development, the developmental potential of a nucleus and the interactions between the donor nucleus and the recipient cytoplasm, etc.

We present the first measurements of the longitudinal double-spin asymmetry $A_{LL}$ for dijets with at least one jet reconstructed within the pseudorapidity range $0.8 < \eta < 1.8$. The dijets were measured in polarized $pp$ collisions at a center-of-mass energy $\sqrt{s}$ = 200 GeV. Values for $A_{LL}$ are determined for several distinct event topologies, defined by the jet pseudorapidities, and span a range of parton momentum fraction $x$ down to $x \sim$ 0.01. The measured asymmetries are found to be consistent with the predictions of global analyses that incorporate the results of previous RHIC measurements. They will provide new constraints on $\Delta g(x)$ in this poorly constrained region when included in future global analyses.
