Background: Diabetic nephropathy (DN) is one of the most serious complications of diabetes mellitus and the main cause of the end-stage renal disease (ESRD). Activation of the NLRP3 inflammasome has been proven to play an important role in the development of DN. Thus, specific and direct targets of NLRP3 inflammasome assembly may have therapeutic potential. CY-09 is a new NLRP3 inflammasome specific inhibitor that has been shown to protect against non-alcoholic fatty liver disease (NAFLD) by inhibiting the activation of the NLRP3 inflammasome. However, its role in kidney disease, especially DN, has not been reported. Methods: In this study, we used HE staining to assess renal pathological damage in each group, and RT-PCR, immunofluorescence and WB were performed to detect the expression changes in inflammatory and fibrosis proteins. The apoptosis level was detected by TUNEL staining. Results: Here, we showed increased inflammation, oxidative stress, apoptosis and fibrosis in db/db mice, while CY-09 exerted renoprotection by inhibiting NLRP3 inflammasome activation. In vitro, CY-09 also inhibited NLRP3 and reduced caspase-1, IL-18, IL-1β and apoptosis in a dose-dependent manner. Conclusion: CY-09 effectively protects the kidney from hyperglycemia induced damage by inhibiting the NLRP3 inflammasome and may be a promising therapeutic strategy to prevent the progression of DKD.

Abstract: Autophagy is the process by which cells selectively remove damaged organelles or proteins. Chaperone-mediated autophagy (CMA) is a type of autophagy that degrades proteins containing the KFERQ pentapeptide in cells. CMA can degrade damaged or excess proteins and therefore plays an important role in maintaining protein balance in cells. CMA can also play a regulatory role by degrading key proteins in life activities, such as lipid and glucose metabolism. This review introduces the CMA process and describes the current commonly used CMA detection methods. In addition, we describe the role of CMA in glucose and lipid metabolism. Finally, we summarize the current role of CMA in metabolic diseases such as diabetic nephropathy (DN), alcoholic liver disease (ALD) and nonalcoholic fatty liver disease (NAFLD) and discuss the role of CMA as a potential therapeutic target for metabolic diseases.

Abstract: Long-term diabetes can lead to renal injury known as diabetic nephropathy (DN), which is a major cause of end-stage renal disease (ESRD). However, its pathogenesis has not been well explained. Adipose tissue is recognized as an important energy storage device for the body. Interestingly, many studies have shown that adipose tissue can also act as an endocrine organ by secreting a variety of adipokines to maintain homeostasis. Here, we summarize some of the adipokines that have been identified thus far to, more specifically, to emphasize their role in DN progression and propose that the “adipose-renal axis” may be a potential target for the treatment of DN.

Abstract: Diabetic nephropathy (DN) is a serious complication of diabetes mellitus and one of the main causes of end-stage renal disease (ESRD). There are many factors causing the progression of DN. Lipid metabolism disorder is a common clinical manifestation of DN, and ectopic renal lipid deposition was recently proposed as a key factor promoting the development of DN. Lipophagy is a newly discovered type of selective autophagy that can remove excessive lipids in cells to maintain lipid homeostasis. Recently, abnormalities in lipophagy have also been implicated in the progression of DN. Here, we discuss the formation of lipid droplets, describe lipophagy and its key regulatory signals, summarize the current research progress of lipophay in DN, and finally propose that lipophagy may be a potential target for the treatment of DN.

Background: Patients with Alzheimer’s disease (AD) have an increased risk of developing epileptiform discharges, which is associated with a more rapid rate of progression. This suggests that suppression of epileptiform activity could have clinical benefit in patients with AD. Objective: In the current study, we tested whether acute, intravenous administration of levetiracetam led to changes in brain perfusion as measured with arterial spin labeling MRI (ASL-MRI) in AD. Methods: We conducted a double-blind, within-subject crossover design study in which participants with mild AD (n = 9) received placebo, 2.5 mg/kg, and 7.5 mg/kg of LEV intravenously in a random order in three sessions. Afterwards, the participants underwent ASL-MRI. Results: Analysis of relative cerebral blood flow (rCBF) between 2.5 mg of levetiracetam and placebo showed significant decreases in a cluster that included the posterior cingulate cortex, the precuneus, the posterior part of the cingulate gyrus, while increased cerebral blood flow was found in both temporal lobes involving the hippocampus. Conclusion: Administration of 2.5 mg/kg of LEV in patients without any history of epilepsy leads to changes in rCBF in areas known to be affected in the early stages of AD. These areas may be the focus of the epileptiform activity. Larger studies are needed to confirm the current findings.

Background. Ectodysplasin A (EDA), a member of the TNF family, plays important roles in ectodermal development, while recent studies expanded its regulatory effects on insulin resistance and lipid metabolism. This study was the first time to investigate the correlation between circulating EDA and albuminuria in patients with T2DM. Methods. A total of 189 T2DM and 59 healthy subjects were enrolled in the study. We analyzed the concentrations of EDA by ELISA. Plasma glucose, insulin, HbA1c, lipids, creatinine, BUN, and UACR were also measured. Insulin resistance and pancreatic cell function were assessed by HOMA. Results. Circulating EDA concentration was significantly increased in T2DM patients and increased with the degree of albuminuria. EDA was positively correlated with age, FIns, HOMA-IR, HOMA-β, Scr, and UACR, and negatively correlated with eGFR. Linear stepwise regression showed that FIns, HOMA-β, and UACR were independent influencing factors of EDA. Logistic regression analysis showed that EDA was independently associated with the occurrence of albuminuria in T2DM. ROC curve showed that EDA had an area under the receiver operating curve of 0.701 [ 95 % CI = 0.625 − 0.777 , P < 0.001 ]. Conclusion. EDA is positively correlated with the degree of albuminuria in patients with T2DM and may be involved in the occurrence and progression of diabetic kidney disease (DKD).

We assessed inpatients’ psychotic symptoms as well as their aggressive and impulsive personality traits using the Positive and Negative Symptom Scale, the Barratt Impulsiveness Scale, and the Buss-Perry Aggression Questionnaire.Results: Compared with the scores of inpatients in the non-aggressive group, the total Buss-Perry Aggression Questionnaire, subscale, and Barratt Impulsiveness Scale behavioral factor scores in those in the aggressive group were higher (p < 0.05). The results of logistic regression analysis suggested that a high Positive and Negative Symptom Scale positive factor score (odds ratio = 1.07) and a high Buss-Perry Aggression Questionnaire physical aggression score (odds ratio = 1.02) were risk factors for aggressive behavior.Conclusion: Hospitalized patients with schizophrenia with more severe positive symptoms and aggressive traits may be more prone to aggressive behavior.Keywords: schizophrenia, aggressive behavior, impulsivity trait, aggressive trait, stable patients

Antibiotic resistance is one of the most serious health problems today and is expected to worsen in the coming decades. It has been suggested that antibiotic administration routes that bypass the human gut could potentially tackle this problem. In this work, an antibiotic hydrogel-forming microarray patch (HF-MAP) system, which can be used as an alternative antibiotic delivery technology, has been fabricated. Specifically, poly(vinyl alcohol)/poly(vinylpyrrolidone) (PVA/PVP) microarray showed excellent swelling properties with >600% swelling in PBS over 24 h. The tips on the HF-MAP were proven to be able to penetrate a skin model which is thicker than stratum corneum. The antibiotic (tetracycline hydrochloride) drug reservoir was mechanically robust and dissolved completely in an aqueous medium within a few minutes. In vivo animal studies using a Sprague Dawley rat model showed antibiotic administration using HF-MAP achieved a sustained release profile, in comparison with animals receiving oral gavage and intravenous (IV) injection, with a transdermal bioavailability of 19.1% and an oral bioavailability of 33.5%. The maximum drug plasma concentration for HF-MAP group reached 7.40 ± 4.74 μg/mL at 24 h, whereas the drug plasma concentration for both oral (5.86 ± 1.48 μg/mL) and IV (8.86 ± 4.19 μg/mL) groups peaked soon after drug administration and had decreased to below the limit of detection at 24 h. The results demonstrated that antibiotics can be delivered by HF-MAP in a sustained manner.

A 12-y-old Himalayan black bear suddenly developed depression, anorexia, cough, and dyspnea and died at the Ordos Zoo, China. At autopsy, the mesenteric and cranial mediastinal lymph nodes (LNs) were enlarged; the largest cranial mediastinal LN was ~13 cm in diameter. Scattered-to-diffuse, rounded-or-oval, gray, firm 1–6-mm nodules were observed on the surfaces of the spleen, liver, lungs, and small intestine. Histologically, the enlarged cranial mediastinal and mesenteric LNs, spleen, small intestine, lungs, and liver contained dense populations of neoplastic lymphoid cells (NLCs). The NLCs were round-or-oval with small amounts of mildly eosinophilic cytoplasm and round-or-oval hyperchromatic nuclei with indistinct nucleoli; the mitotic count was 55 in 2.37 mm2. Immunohistochemically, cell membranes and the cytoplasm of NLCs were CD3+, CD79a−, CD20−, CD15−, CD30−, and CD45RA−; hence, the NLCs were derived from T lymphocytes. To our knowledge, T-cell lymphoma has not been reported previously in a Himalayan black bear.

Background: Subjective cognitive decline (SCD) is considered as a preclinical hallmark of Alzheimer’s disease (AD). However, the characteristics of SCD associated with amyloid pathology remain unclear. Objective: We aimed to explore the associations between SCD characteristics with amyloid pathology. Methods: Using logistic regression analyses, we analyzed the associations between cerebrospinal fluid (CSF) amyloid pathology with AD risk factors, SCD-specific characteristics (onset of SCD within the last five years, age at onset ≥60 years, feelings of worse performance, informant confirmation of complaints, worries, other domains of cognition complaints), as well as subthreshold depressive and anxiety symptoms among individuals with SCD. Results: A total of 535 SCD individuals with available CSF Aβ42 information from the Chinese Alzheimer’s Biomarker and LifestylE (CABLE) study (mean age of 63.5 years, range 40 to 88 years; 47.10% female) were enrolled. The characteristics of informant confirmation of complaints (OR, 95% CI = 2.00, 1.19–3.36), subthreshold depressive symptoms (OR, 95% CI = 2.31, 1.05–5.09), and subthreshold anxiety symptoms (OR, 95% CI = 2.22, 1.09–4.51) were found to be significantly associated with pathological amyloid in multivariate analyses when adjusting for age, sex, education, and APOE ɛ4. Besides, age and females were observed risks for amyloid pathology in subscale analyses. Nonetheless, we did not find any associations of other SCD-specific characteristics with amyloid pathology in this study. Conclusion: Our study suggested that informant confirmed complaints and subthreshold psychiatric symptoms might be critical for discriminating AD-related SCD from non-AD related SCD.

ECG-derived respiration (EDR) is a low-cost and productive means for capturing respiratory activity. In particular, as the primary procedure in some cardiorespiratory-related studies, the quality of EDR is decisive for the performance of subsequent analyses.In this paper, we proposed a novel EDR method based on the feature derived from the first moment (mean frequency) of the power spectrum (FMS). After obtaining the EDR signal from the feature, we introduced the Interacting Multiple Model (IMM) smoother to enhance the similarity of the EDR signal to the reference respiration. The assessment of the approach consisted of two steps: (1) the performance of extracted feature was verified against R-peak misalignment and noise. (2) the enhancement of IMM smoother to EDR waveforms was evaluated based on waveform correlation and respiratory rate estimation. All the assessments were conducted under the Fantasia database and Drivers database.The FMS improved robustness against R peak offsets compared to most established feature-based EDR algorithms, but a slight 5% improvement of waveform correlation against RR interval-based feature under accurate R peaks. The IMM smoother performed similarly with the Kalman filter in the static database but realized the enhancement of some extent of the EDR waveform in the ambulatory database.The proposed method investigated frequency domain mapping of ECG morphological changes caused by respiratory modulation and explained the EDR signal as a non-stationary time series, which provided a direction of better fitting the natural respiration process and enhancing the EDR waveform.

Background and Objectives Moderate-intensity continuous training (MICT) is used to observe lipidomic effects in adults. However, the efects of MICT on lipid metabolism in adolescents remain unclear. Therefore, we aimed to longitudinally characterize the lipid profile in adolescents during different periods of 6-week MICT. Methods Fifteen adolescents undertook bicycle training at 65% of maximal oxygen consumption. Plasma samples were collected at four time points (T0, T1, T2, and T3). Targeted lipidomics was assessed by ultra-performance liquid chromatography–tandem mass spectrometry to characterize the plasma lipid profiles of the participants to identify the lipids present at differing concentrations and changes in lipid species with time. Results MICT afected the plasma lipid profiles of the adolescents. The concentrations of diglycerides, phosphatidylinositol, lysophosphatidic acid, lysophosphatidylcholine, and lysophosphatidylethanolamine were increased at T1, decreased at T2, and increased again at T3. Fatty acids (FAs) showed an opposite trend. Ether-linked alkylphosphatidylcholine and triglycerides were significantly increased and remained high. Sphingolipid concentrations initially decreased and then remained low. Therefore, a single bout of exercise had substantial efects on lipid metabolism, but by T3, fewer lipid species were present at significantly diferent concentrations and the magnitudes of the remaining diferences were smaller than those at earlier times. Among all the changed lipids, only DG(14:1/18:1), HexCer(d18:1/22:1) and FA(22:0) showed no significant correlations with any other 51 lipids (P < 0.05). Glycerides and phospholipids showed positive correlations with each other (P < 0.05), but FAs were significantly negatively correlated with glycerides and phospholipids while positively with other FAs (P < 0.05). Pathway enrichment analysis showed that 50% of the metabolic pathways represented were related to lipid metabolism and lipid biosynthesis. Conclusion MICT increases ether-linked alkylphosphatidylcholine and triglyceride concentrations. Diglyceride, phosphatidylinositol, and lysophosphatidylcholine concentrations initially rise and then decrease 6 weeks after MICT, but FA concentrations show an opposite trend. These changes might correlate with lipid metabolism or biosynthesis pathways.

This paper proposes a novel concept of using defect management technology to measure, predict, and optimize the management of the entire software project cycle. It first utilizes the software defect measurement GQIM model and prediction Gompertz model to track and quantify the defects of the entire project cycle. Then, the specific optimization strategy is proposed to manage the core processes of the project management, including project scope management, project time management, project quality management, project human resource management, project communication management, and project risk management. This study solves the quantification problems that are common in the current software project management process, such as complex testing procedures, high defect repair costs, and high communication costs. Furthermore, it contributes to promote the overall level of project management in the software industry. Taking the SDLM’s electronic banking software project management as an example, the various management quality indicators before and after the optimization of the project management process strongly indicate that using software defect management technology to optimize the software project management process is feasible and effective.

The Gametophyte factor1 (Ga1) locus in maize confers unilateral cross-incompatibility (UCI), and it is controlled by both pollen and silk-specific determinants. Although the Ga1 locus has been reported for more than a century and is widely utilized in maize breeding programs, only the pollen-specific ZmGa1P has been shown to function as a male determinant; thus, the genomic structure of the Ga1 locus and all the determinants that control UCI at this locus have not yet been fully characterized. Here, we used map-based cloning to confirm the determinants of UCI at the Ga1 locus and maize pan-genome sequence data to characterize the genomic structure of the Ga1 locus. The Ga1 locus comprises one silk-expressed PME (ZmGa1F) and eight pollen-expressed PMEs (ZmGa1P and ZmGa1PL1-7). Knockout of ZmGa1F in Ga1/Ga1 lines leads to the complete loss of the female barrier function. The expression of individual ZmGa1PL genes in a ga1/ga1 background endows ga1 pollen with the ability to overcome the female barrier of the Ga1 locus. These findings, combined with genomic data and genetic analyses, indicate that the Ga1 locus is modulated by a single female determinant and multiple male determinants, which are tightly linked. The results of this study provide valuable insights into the genomic structure of the Ga2 and Tcb1 loci and will aid applications of these loci in maize breeding programs.
