The aim of the work was to obtain a producer strain with increased lysine accumulation using the chemical mutagenesis method. Methods. To achieve the goal, we used the method of treating the lysine-producing strain with the chemical mutagen N-methyl-N-nitro-N-nitrosoguanidine, cultivating the resulting clone and determining the accumulation of lysine in flasks and a bioreactor. Results. The optimal concentrations and duration of mutagen action for the production of mutant microorganisms were found. Сlones with the maximum lysine accumulation were selected. Mutagenesis was carried out consecutively three times. As a result, lysine-producing strain Brevibacterium sp. IMV B-7796 auxotrophic regarding leucine and threonine with maximum accumulation of the target amino acid was obtained. Conclusions. The lysine producer Brevibacterium sp. IMV B-7796 was obtained, which produced 65.0 g/dm3 of lysine in a bioreactor under conditions of periodic cultivation with feeding. The Brevibacterium sp. IMV B-7796 strain was proposed as a basis for the creation of a genetically modified strain with increased accumulation of lysine for further use in industrial lysine technology.

The aim of the study was to establish the optimal cultivation conditions for increasing accumulation of riboflavin by the producer strain Bacillus subtilis. As an object of the study there were used strains of B. subtilis from “Collection of strains of microorganisms and plant lines for food and agricultural biotechnology” of the State Enterprise “Institute of Food Biotechnology and Genomics of the National Academy of Sciences of Ukraine”. The percentage (10%) and the period of cultivation (16 hours) of the seed material necessary for accumulation of riboflavin were determined. The effect of the carbon source on the accumulation of riboflavin was studied and it was shown that the greatest accumulation (5.2 g/dm3) was with the use of glucose. The dynamics parameters of riboflavin accumulation over time were investigated and the optimal cultivation period was determined (68 hours). The optimal cultivation conditions were selected, which increased the accumulation of riboflavin in the culture fluid (glucose concentration — 120 g/dm3, temperature — 38 oC, and pH of the medium — 7.0) more than twice. It was concluded that the accumulation of riboflavin could be increased by changing the cultivation conditions.

The purpose of this study was to investigate the effect of curcumin on cytokine response and angiotensin-converting activity in terms of intrahippocampus administration of ß-amyloid peptide in rats. Animals with experimental model of Alzheimer’s disease received nasal therapy aqueous solution of curcumin. Recorded concentration of cytokines (interleukin-1ß, interleukin-6, interleukin-10, tumor necrosis factor-α) and angiotensin-converting activity in the parts of the brain (cerebral cortex and hippocampus) and blood serum, as well as indicators of conditional reflex escapers action. Install a reduction in cytokine response intrahippocampus administering Aß42_Human and angiotensin-converting activity inhibition in the brain but not in the blood serum of animals under the action of curcumin. Recorded improvement in cognitive performance in rats with a model of Alzheimer’s disease as a result of curcumin therapy. Nasal treatment with an aqueous solution of curcumin has anti-inflammatory effect in the targeted parts of the brain (сerebral cortex and hippocampus), but inhibits angiotensin-converting activity.

Creation of liposomal forms of curcumin (diferuloilmetane) was investegated in the work. The methods of preparation and determination of the obtained liposomes sizes and percentage of curcumin in liposomes are given. Soybean phospholipids, egg yolk phospholipids and sunflower phospholipids were selected for preparation of “empty” liposomes and liposomes loaded curcumin: C-SPC-L (curcumin encapsulated in SPC liposomes), C-EPC-L (curcumin encapsulated in EPC liposomes) and C-SFPC-L (curcumin encapsulated in liposomes SFPC). Physical properties of different liposomes such as average size (Dl) and encapsulation efficiency (EE) were studed. Using photon correlation spectroscopy, the average size of three types of curcumin-loaded liposomes were found as Dl = 111 ± 3 nm for C-SPC-L, Dl = 98 ± 5 nm for C-EPC-L and Dl = 109 ± 5 nm for C-SFPC-L. Encapsulation efficiency showed the following values: EEC-SPC-L = 83 ± 4%, EEC-EPC-L = 82 ± 3%, and EEC-SFPC-L = 85 ± 4%. We determined that by using the methods of freezing and thawing or sonication it was possible to isolate liposomes sized in the range 100– 120 nm. Upon that the obtained liposomal form of curcumin was stable and freeze-thaw method was optimal for liposomal form and provides a relatively high level (more than 80%) of curcumin inclusion.

The purpose of our work was to study generic and specific affinity of domestic strainsproducers, comparative productivity butanol strains-producers screening, fermentation of sugars being a part of renewable lignocelluloses raw materials and to determine the conditions for the butanol yield increasing. The objects of research were strains Clostridium acetobutylicum ІМВ В-7407 (IFBG C6H), IFBG C4B and IFBG C7P from «Collection microorganism’s stains and plants line for food and agriculture biotechnology» of Institute of Food Biotechnology and Genomics of the National Academy of Sciences of Ukraine. It was determined that domestic butanol-producing strains were relatively more productive and might be promising for improvement technology of butanol production.

The results of studies of hepatoprotective activity of biopreparations of defatted soy and sunflower lecithins under carbon tetrachloride intoxication of the experimental animals are given. Analyze of biochemical and histological data under soy and sunflower lecithin exposure changed in terms of poisoning enables to conclude that both biological products of defatted lecithin slowed the growth of mass of animal liver and transaminase activity and stimulated liver regeneration as a result of polyploidy and hypertrophy of hepatocytes, resuscitated beam and lobulation organ and created contacts between hepatocytes and venous vessels as well. It is evidenced that repair processes are activated in condition of their injection under carbon tetrachloride intoxication.

Dry lecithin, which is a mixture of polar phospholipids, neutral lipids, free fatty acids, glycolipids, carbohydrates, and small amounts of moisture, is a promising object for biologic-hepatoprotectors creation. One of its pharmacological activity displays is its influence on serum lipids, in particular transport forms of these lipids. The influence of dry soy lecithin and sunflower on hyperlipoproteinemia ratio and other lipid disorders is studied. It is shown that low-fat dry soybean lecithin showed hypocholesterolemic activity against all studied forms of serum cholesterol. Nonfat dry sunflower lecithin had similar but somewhat less prominent effect. However reduced concentration of high density lipoprotein cholesterol under sunflower lecithin developed right before soya lecithin. Both lecithin prevented the development of dyslipidemia induced carbon tetrachloride. [ABSTRACT FROM AUTHOR]

Liposomes are hollow particles which content is limited to a lipid membrane. They belong to a large family of vesicular (bubble) structures formed by amphiphilic molecules. Liposomes consisting of one or more phospholipid bilayers were first described in the mid 60s. In a relatively short period liposomes transformed from a simple model that mimics the cell membrane into the object of active research and variety of practical applications. Currently a large number of laboratory techniques create liposomes and relatively small number of technologies used in the industry are available. However, most of ones have limitations in conditions of the biologically active substances inclusion in liposomes due to their sensitivity to mechanical and/or chemical damage.
