In this study, spermatheca and eggs of Pyrrhocoris apterus (Heteroptera: Pyrrhocoridae were examined by light microscope and SEM. The eggs have ~1.30 mm length and 0.94 mm width. Newly laid eggs turn from white to yellow as soon as embryonic differentiation. P. apterus egg surface shows polygonal (usually hexagonal and pentagonal) shapes. The eggs have pipe shaped 6-7 micropylar projections. The egg-burster which is a dark T-shaped or triangular pattern, explosive becomes visible when the embryo is well developed. The nimph, which has completed its embryonic development, comes out of the egg with peristaltic movements. The spermatheca of P. apterus consists of a bulb, a pump, a flange of pump, and duct. The morphology of the egg and spermatheca is useful for classification, because they show a great diversity among species and genera.

The hunting behavior of antlion larvae is unique in the insect world and soil characteristics may strongly affect their microhabitat selection. Therefore, the substrate type of sand pit construction is expected to have an important role in their traps. Different types of soil substrates can be inhabited by Myrmeleon formicarius Linnaeus, 1767 larvae. This study was conducted during 2018-2019 June-August in the Amanos Mountains of Turkey to examine assess the relationship between the microhabitat preference of larvae of M. formicarius. In this study, five substrate types were determined as particularly according to their habitat preference. In this study, the preference of fine-grained soils is found more adaptive because of the pits are constructed in these substrates are more functional for longer periods and they are much more successful in trapping their preys than the coarse-grained soil pits. Furthermore, it was found that there is a direct relationship between the pit size and substrate particle structure. Therefore, the detection and selection of microhabitats under more convenient ecological conditions, which increase capture success and elucidate the hunting strategy, may be carried out during strong soil substrate selection for Myrmeleon Linnaeus species. It was concluded that there is no strong relationship between the soil substrate type, pH, and trap diameter.

Amaç: Bağışıklığı baskılanmış insanlarda ve hayvanlarda yaşamı tehdit eden enfeksiyonlara neden olabilen Cryptococcus neoformans ve Cryptococcus gattii, çevresel odaklardan hassas olan konağa bulaşır. Patojen biyoklimatik değişkenlere uygun şekilde çevresel kolonizasyon gösterir. Daha önceki MaxEnt makine öğrenmesi ile yapılan analizlerde, Alanya bölgesi muhtemel kolonizasyon bölgesi olarak saptanmıştır. Bu çalışmanın amacı, ülkemiz için önemli bir turizm bölgesi olan Alanya ve yakın çevresindeki kovuklu ağaç florasında C. neoformans ve C. gattii kolonizasyonunun araştırılmasıdır.Gereç ve yöntem: Alanya şehir merkezi ve yakınındaki Dim çayı çevresinden, üzerinde geniş kovuk olduğu belirlenen 49 zeytin (Olea europea), 17 keçiboynuzu (Ceratonia siliqua) ve 13 okaliptüs (Eucalyptus camaldulensis) olmak üzere toplam 79 ağaçtan standart eküvyon yöntemi ile örnekleme yapıldı. Bifenil ve kloramfenikol içeren Staib agar kültür için kullanıldı. Üreyen maya benzeri, kahverengi pigmentli koloniler konvansiyonel yöntemler ve kanavanin-glisin-bromtimol agar reaksiyonu ile tanımlandı. Bulgular: Alanya şehir merkezinde sadece bir (%2,04) zeytin (O. europea) ağacında C. neoformans kolonizasyonu saptandı. Sonuç: Bu çalışmada, C. neoformans'ın Türkiye'nin önemli turizm kenti olan Alanya şehir merkezinden izole ettiği ilk çevresel kolonizasyonu rapor edilmiştir. Çevresel insan patojenik mantarlarının kolonizasyonu için ülkemizin insan yerleşimlerinin ve turizm potansiyeli yüksek bölgelerinin sistematik olarak taranması riskli alanların bulunmasına yardımcı olacaktır.

Introduction: The worldwide growth of aquaculture has led to persistent infections and the emergence of antibiotic-resistant bacteria (ARB). The present study identified the similarity and correlativity of antibiotic susceptibility in the autochthonous bacterial flora of carps cultured in the East Kolkata Wetland and peri-urban Kolkata, India using clustering algorithms based on minimal inhibitory concentration (MIC) data. Materials and Methods: Motile Aeromonas spp. and Escherichia coli (50 each) isolated in selective media from carps and their environment were tested for susceptibility to oxytetracycline (OTC) and chloramphenicol (CH) using the agar-disc diffusion assay. The MICs of these two antibiotics were determined using the agar dilution method and clustered using the BioNumerics 7.6 software package. Results: The MICs of OTC and CH varied from 0.39 to 50 µg/ml and 1.56 to >100 µg/ml, respectively. Dendrogram-based cluster analysis of motile aeromonads showed relatively high internal homogeneity, as >5 subgroups were obtained under the main clusters. Escherichia coli also showed high internal homogeneity. Dendrogram-based advanced nodal cluster analysis of motile aeromonads as a group yielded a greater number of clusters. Conclusion: The varied susceptibility among motile aeromonads and E. coli isolated from an aquaculture environment with no history of antibiotic use implied the possible contamination of carps with ARB from domestic and hospital effluents. Nevertheless, E. coli strains isolated from this environment exhibited high heterogeneity in antibiotic susceptibility, which is a serious cause for concern.

Introduction: Pathogens have persisted as major agents in the progress of infections that constitute a threat to human health the world over. Extracting remedies from plants is a longstanding human tradition. This study assesses the antimicrobial action of extracts of turmeric (Curcuma longa) and garlic (Allium sativum) against several bacterial clinical isolates. Materials and Methods: The phytochemical constituents of ethanolic and aqueous extracts of turmeric and garlic and their susceptibility in single and combined absorptions were determined using a Clinical and Laboratory Standards Institute standard. Results: Steroids, flavonoids, terpenoids, and alkaloids were detected in both extracts, whereas anthraquinone was only present in turmeric. On the other hand, tannins and saponins were present in extracts of garlic. Ethanolic extract of garlic showed a zone of inhibition measuring 12 mm at the absorption of 100 mg/ml against Staphylococcus aureus, a zone of inhibition 10 mm at a concentration of 100 mg/ml against Escherichia coli and a zone of inhibition of 7 mm at a concentration of 100 mg/ml against Klebsiella pneumoniae. The aqueous extract of the same garlic was effective against the tested bacteria. Ethanolic extract of turmeric showed the largest zone of inhibition of 13 mm at a concentration of 100 mg/ml against Staphylococcus aureus and 7 mm at a concentration of 100 mg/ml against Klebsiella pneumoniae. Equally, the aqueous extract of turmeric showed the highest zone of inhibition of 11 mm at a concentration of 100 mg/ml on Staphylococcus aureus and 7 mm against Klebsiella pneumoniae. The aqueous extract of garlic was active against the isolate, and the combination of the two extracts was active against the tested organism. Conclusion: Spices can be used as effective antimicrobial agents against both Gram-positive and Gram-negative bacteria in both individual and combined forms.

Salinity is one of the major limiting factors for sustainable crop production and the growth of beneficial microorganisms. Screening of biocontrol agent Trichoderma harzianum isolates under saline conditions could be necessary to select more efficient strains to alleviate salt stress through biopriming in plants. The present study aimed to investigate the antagonistic capacity of 14 T. harzianum isolates against Macrophomina phaseolina as well as the growth, protease activity, and proline content under different concentrations of salt. All isolates except Th15 and Th18 exhibited an over 85% inhibition against M. phaseolina at 0 mM NaCl in c and showed a noticeable decline in their antagonistic capacity as salt concentration increased. Colony growth decreased with increasing salt concentrations in all isolates tested. The growth of all isolates at 0 mM was significantly higher than the other NaCl treatments except at 70 mM NaCl. Protease activity also declined with the increased level of salt. Isolates displayed a wide range of protease expression patterns even at the same salinity as in the case of proline content. Unlike protease enzyme, proline content significantly increased at 240 mM NaCl. Salinity played a significant role in T. harzianum isolates with regards to the growth, antagonistic capacity, protease activity, and proline content.

The aim of this study is to determine the nitrogen activities of canola cultivars (Elvis, Orkan, Licrown, Licord, Contact and Oase) in the presence and absence of nitrogen. Özet Uluslararası Virüs Sınıflandırma Komitesi (International Committee on Taxonomy of Viruses; ICTV) tarafından yayımlanan raporlara göre tür düzeyinde tanımlanan virüslerin sayısı son birkaç yılda 5.561’den (ICTV 2018b) 9.110’a (ICTV 2020) ulaşmıştır. Yeni geliştirilen moleküler biyoloji teknikleri ve yaygınlaşan saha çalışmaları yanında, virüslerin sadece enfeksiyöz patojenler olarak görüldüğü ve tanımlandığı geleneksel bakışın aksine virüslerin aşı çalışmaları veya genetik temelli tedaviler için biyolojik araçlar olarak da incelendiği günümüzün yeni anlayışı tanımlanan tür sayısındaki hızlı artışın önemli bir nedeni olmuştur. Son yıllarda viral enfeksiyonların epidemiyolojisindeki hızlı değişimler ve pandemik virüslerin küresel düzeydeki yıkıcı etkileri de bilim dünyasının salgın potansiyeli olan virüsler başta olmak üzere viroloji alanına karşı ilgisinin artmasına yol açmıştır. In addition to newly developed molecular biology techniques and widespread field studies, today's new understanding in which viruses are also examined as biological tools for vaccine studies or genetic-based treatments, contrary to the traditional view that viruses are seen and defined only as infectious pathogens, has been an important reason for the rapid increase in the number of defined species. In recent years, the rapid changes in the epidemiology of viral infections and the devastating effects of pandemic viruses at the global level have led to an increase in the scientific world's interest in the field of virology, especially viruses with epidemic potential. Alphaviruses, which have spread all over the world, cause epidemics in humans and animals, and some species are associated with high-mortality infections, are mainly transmitted by mosquitoes. Alphaviruses are primarily associated with arthritogenic and encephalitic infections in humans, but some species are not infectious or pathogenic to humans. Among the members of the genus Alphavirus, there are species that infect sea creatures such as fish and seals through aquatic insect parasites, as well as species that stand out as safe tools in experimental studies because they can only infect mosquitoes. Moreover, recombinant alphaviruses and alphaviral replicases are used as biological tools in the design of replicative mRNAs and in the development of mRNA-based vaccines. While there are available attenuated and inactivated vaccines developed mainly for use in horses for common encephalitic alphavirus species, development studies are still ongoing for vaccines and antivirals that can be used in humans for different alphavirus types. The purpose of this study was to examine the relationship between Kick-Box athletes’ freedom perceived in time and thrill seeking levels. Appropriate sampling method was used for sample selection. Sample of the study was active kick-box athletes who live in Turkey. There were 355 male and 238 female, totally 593 participants voluntarily accepted to attend this study. Data were collected from demographic information form, the “Leisure Perceived Freedom Scale in Leisure-Time” developed by Witt and Ellis (1985) and “Thrill Seeking Scale” developed by Hoyle et al. (2002). In statistical analysis, one way ANOVA, t-test and follow-up TUKEY test were used. Before the statistical analysis, a normality test was checked and it was found that data were distributed normally. Results showed that males’ freedom perceived in time levels were significantly higher than those in females. Females’ thrill seeking level was significantly higher than males’ thrill seeking level. In the marital status variable, married participants’ thrill seeking level was higher than those in single participants., Bu araştırmanın amacı: Kick boks sporu ile ilgilenen bireylerin serbest zamanda algıladıkları özgürlük düzeyleri ve heyecan arayışı arasındaki ilişkiyi ortaya çıkarmaktır. Araştırmaya Türkiye sınırları içerisinde yaşayan ve kick boks branşında aktif olan sporculardan uygun örnekleme yöntemi ile belirlenmiştir. Bu araştırmada 355 erkek 238 kadın olmak üzere toplamda 593 katılımcı yer almıştır. Katılımcılardan kişisel bilgi formu, serbest zamanda algılanan özgürlük seviyelerini ölçmek için Witt ve Ellis tarafından (1985) geliştirilen “Serbest Zamanda Algılanan Özgürlük Ölçeği-SZAÖ” ve Hoyle ve arkadaşlarının (2002) geliştirdiği heyecan arayışı ölçeği kullanılmıştır. Veri analizinden önce normallik testi uygulanmış ve verilerin normal dağılım sağladığı belirlenmiştir. Veri analizinde tek yönlü ANOVA ve t-testi kullanılmıştır. Anlamlı farkın kontrolü için Tukey testi uygulanmıştır. Bulgulara göre serbest zaman algısının alt boyutlarından bilgi ve beceri alt boyutunda erkeklerini bilgi ve beceri değerlerinin kadın katılımcılara göre istatistiksel açıdan anlamlı olduğu tespit edilmiştir. Heyecan arayışı değerleri incelendiğinde ise kadın katılımcıların ortalama değerlerinin erkeklere oranla daha fazla olduğu tespit edilmiştir. Medeni durum değişkenine göre heyecan arayışı ölçeğinde bekâr katılımcıların evli katılımcılara göre daha yüksek bir ortalamaya sahip oldukları tespit edilmiştir.

Turkey is among the endemic countries in terms of tularemia. Tularemia, a zoonotic disease, is an important public health problem that occasionally causes epidemics and sporadic cases in our country. Transmission can often be via infected/contaminated animal contact, consumption of contaminated water, or vectors. The disease most commonly presents with lymphadenopathy in the neck and is often misdiagnosed. Since serological methods are necessary in the diagnosis of the disease, if it is not considered, it creates a problem in the diagnosis of the disease. The biggest known outbreak of the disease in our country was in Antalya. Many variables, such as seasonal changes and increased migration, have caused the epidemiology of the disease to change. In this review study, we aimed to evaluate the province of Antalya in terms of tularemia., Türkiye tularemi açısından endemik ülkeler arasında bulunmaktadır. Zoonotik bir hastalık olan tularemi, ülkemizde zaman zaman salgınlara ve sporadik vakalara neden olan önemli bir halk sağlığı sorunudur. Sıklıkla bulaş yolu, enfekte/kontamine hayvan teması, kontamine suların tüketilmesi veya vektörler aracılıklı olabilir. Hastalık en sık boyunda lenfadenopati ile kendini göstermekte ve çoğu zaman yanlış tanı almaktadır. Hastalığın tanısında serolojik yöntemlerin gerekli olması nedeniyle akla gelmemesi durumunda hastalık tanısının konulmasında sorun oluşturmaktadır. Hastalığın ülkemizdeki bilinen en büyük salgını Antalya ilinde olmuştur. Mevsimsel değişiklikler ve artan göçler gibi birçok değişken hastalık epidemiyolojisinin değişmesine neden olmuştur. Biz de bu derleme çalışmasında, Antalya ilinin tulareminin açısından değerlendirmesini yapmayı amaçladık.

Molineria capitulata is an ornamental plant that has traditionally been used to treat several chronic diseases. The present study was designed to examine the antioxidant, cytotoxic, thrombolytic, anti-inflammatory, and analgesic activities of a methanolic extract of M. capitulata leaves (MEMC) using both experimental and computational models. Previously established protocols were used to perform qualitative and quantitative phytochemical screening in MEMC. A computational study, including molecular docking and ADME/T analyses, was performed. The quantitative phytochemical analysis revealed the total phenolic and flavonoid contents as 148.67 and 24 mg/g, respectively. Antioxidant activity was assessed by examining the reducing power of MEMC, resulting in absorbance of 1.87 at 400 µg/mL, demonstrating a strong reduction capacity. The extract exhibited significant protection against blood clotting and showed the highest protein denaturation inhibition at 500 µg/mL. In both the acetic acid-induced writhing and formalin-induced paw-licking models, MEMC resulted in significant potential pain inhibition in mice. In the computational analysis, 4-hydroxybenzaldehyde, orcinol glucoside, curcapital, crassifogenin C, and 2,6-dimethoxy-benzoic acid displayed a strong predictive binding affinity against the respective receptors. These findings indicated that M. capitulata possesses significant pharmacological activities to an extent supported by computational studies.

In the study, the relationship between fish size and otolith measurements in the Fries's goby ( Lesueurigobius friesii ) were investigated. Fish specimens were collected by trawl operations from Izmir Bay between September 2005 and August 2006. A total of 100 Lesueurigobius friesii specimens were used for the study. Their total lengths (TL) and weights were recorded by a fish ruler and a digital scale with an accuracy of 0.1 cm and 0.01 g respectively. After that the fish were dissected and their sagittal otoliths were removed. Otolith lengths and heights were measured with an Olympus SZ61 stereo microscope with a micrometric ocular. Otoliths were weighted by a digital scale with an accuracy of 0.0001 g. The specimens used in the analysis were ranged between 5.5 and 8.7 cm total length and from 14.8 to 56.4 g in total weight. The length–weight relationship equations were estimated as W=0.0927*L 2.9431 (r=0.942). Otolith length (OL), otolith height (OH) and otolith weight (OW) of the species were ranged between 2.6–4.1 mm, 2.2–3.4 mm and 0.0049–0.0204 g, respectively. The total length–otolith length, total length–otolith height and otolith length–otolith height measurements showed significant linear relationships. The relations between the otolith weight–otolith length total length–otolith weight were found to be exponential.

Özet Bu çalışmada 10 yıl önce izole edilenC. parapsilosis, C. albicansveC. tropicalissuşları ile günümüz izolatlarındaki antifungal duyarlılık değişimini karşılaştırmayı amaçladık. Çalışmaya 10 yıldan daha uzun süre önce izole edilmiş 17 stokCandidaizolatı ve 47 yeni izolat olmak üzere toplam 64 klinikCandidaizolatı dahil edildi. Klinik izolatların dağılımı;C. parapsilosis25,C. albicans20,C. tropicalis9,C. glabrata6,C. krusei3 veC. lusitaniae1 izolat şeklinde idi. Çalışma izolatları flukonazol ve vorikonazol için ve ayrıca dirençli suşların tedavisinde önemli bir seçenek olan kaspofungin için disk difüzyon yöntemi ile test edildi. Sonuçlar CLSI M60 ve ayrıca CLSI M44-A2 kriterlerine göre değerlendirildi. Çalışma grubu 39 erkek, 21 kadın ve 4 çocuktan oluşurken, yaş ortalaması 55.4 (±21.2) idi.C. parapsilosisiçin uzun dönem stoklanmış izolatlarda flukonazol direnci saptanmazken, yeni izolatların%14.3’ünün (2/14) doza bağımlı duyarlı olduğu ve %35.7’sinin (5/14) flukonazole dirençli olduğu saptandı.C. parapsilosisiçin direnç oranındaki artış (0/11’den 5/14’e; p = 0.03768) ve antifungal duyarlılık zon çaplarının ortalamasındaki düşüş (30.1 mm’den 16.9 mm’ye; pC. parapsilosisiçin vorikonazol duyarlılık zon çaplarının ortalamasındaki düşüş (33.5 mm’den 21.4 mm’ye; p C. albicansveC. tropicalisiçin de zon çapı ortalamaları düşüş gösterirken bu düşüşler istatistiksel olarak anlamlı düzeylerde değildi. Abstract In this study, we aimed to compare the antifungal susceptibility ofC. parapsilosis,C. albicansandC. tropicalisstrains isolated 10 years ago with present isolates. A total of 64 clinicalCandidaisolates were included in the study, including 17 stockCandidastrains isolated more than 10 years ago and 47 present isolates.Distribution of clinical isolates was as follow: 25C. parapsilosis, 20C. albicans, 9C. tropicalis, 6C. glabrata, 3C. krusei, and 1 isolateC. lusitaniae. All isolates were tested for fluconazole and voriconazole by disk diffusion method, and were also tested for caspofungin, an important option in the treatment of resistant strains. The results were evaluated according to the CLSI M60 and CLSI M44-A2 criteria. The study group consisted of 4 children, 21 women, and 39 men, while the mean age was 55.4 (± 21.2). While no fluconazole resistance was found for the long-term stockedC. parapsilosisisolates, it was found that 14.3% (2/14) of the new isolates were dose-dependent susceptible and 35.7% (5/14) were fluconazole resistant. The increase in the resistance rate (from 0/11 to 5/14; p = 0.03768) forC. parapsilosisand the decrease in the mean of the antifungal sensitivity zone diameters (30.1 to 16.9; p C. parapsilosis(33.5 to 21.4; p C. albicansandC. tropicalis, but these decreases were not statistically significant. No decrease was observed in caspofungin zone diameters for all three species.In conclusion, differing fromC. albicansandC. tropicalis, it was found that there was a significant decrease in fluconazole susceptibility and fluconazole and voriconazole mean susceptibility zone diameters forC. parapsilosisisolates.

In this study, It is aimed to determine the effect on yield and yield components of basil (Ocimum basilicum L.) of different nitrogen doses (0, 3, 6, 9, 12 and 15 kg da-1). The study was carried out in a randomized complete block design with three replications under Eskişehir ecological conditions in 2019-2020. The plant height, fresh herb yield, fresh and dry leaf yields, essential oil content and essential oil yield of Ocimum basilicum L. were investigated in the study and the effect of different nitrogen doses on examined all parameters were found to be significant. The results (2019 and 2020 mean) obtained from the experiment showed that fresh herba yield, fresh leaf yield, dry leaf yield and essential oil yield ranged between 748.00-1617.80 kg da-1, 449.55-1062.40 kg da-1, 89.76-166.63 kg da-1 and 0.79-1.24 L da-1, respectively. In addition, essential oil contents were determined between 0.72-0.88%. Considering the results obtained from the study, the most suitable nitrogen dose that can be recommended for dry leaf and essential oil production in basil (Ocimum basilicum L.) cultivation under Eskişehir and similar ecological conditions is 12 kg da-1 and 9 kg da-1, respectively., Bu araştırmada, farklı azot dozlarının (0, 3, 6, 9, 12 ve 15 kg da-1) reyhan (Ocimum basilicum L.) ‘ın verim ve verim öğeleri üzerine etkisinin belirlenmesi amaçlanmıştır. Deneme sonucunda (2019 ve 2020 yılları ortalaması) taze herba verimi, taze yaprak verimi, kuru yaprak verimi ve uçucu yağ verimi sırasıyla 748.00-1617.80 kg da-1, 449.55-1062.40 kg da-1, 89.76-166.63 kg da-1 ve 0.79-1.24 L da-1 arasında değişmiştir. Uçucu yağ oranları ise %0.72-0.88 arasında belirlenmiştir. Çalışmadan elde edilen sonuçlar dikkate alındığında, Eskişehir ve benzer ekolojik koşullarda reyhan (Ocimum basilicum L.) yetiştiriciliğinde kuru yaprak üretimi için 12 kg da-1, uçucu yağ üretimi için ise 9 kg da-1 azot dozu tavsiye edilebilir.

Elektronik burun (e-burun), kahve, şarap ve bira gibi içeceklerin değerlendirmesinde kullanılan düşük maliyetli, koku üzerine yoğunlaşan bir cihazdır. Kahve, farklı kavurma derecelerinde instant veya içmeye hazır formda bulunan insanlık tarihinin en popüler içeceklerinden birisidir. Bu çalışmada geniş tüketim ağı bulunan Arabika ve Robusta türü kahvelerin tasarlanan elektronik burun cihazı ile ayrımlarının yapılması amaçlanmıştır. Bu amaç doğrultusunda MQ (MQ135, MQ2, MQ3, MQ5, MQ6, MQ7 ve MQ8) sensörleri kullanılarak bir elektronik burun düzeneği tasarlanmıştır ve kahve analizleri gerçekleştirilmiştir. Numunelerden alınan sinyaller arduino kartla işlenip MATLAB programına aktarılmıştır. Tasarlanan e-burun cihazının kahveleri ayırıp ayıramadığı ise sınıflandırma algoritmaları kullanılarak belirlenmiştir. Cihazdan alınan sinyal değerleri doğrusal ayırma analizi (LDA), Bayes, en yakın komşu (kNN-3, kNN-5, kNN-7 ve kNN-9), karar ağacı, oneR ve destek vektör analizi (SVM) algoritmaları kullanılarak sınıflandırılmıştır. Arabika ve Robusta ikili ayrımında en iyi sınıflandırma cevabı MQ7 sensöründen, karar ağacı algoritma analizi ile elde edilmiştir. MQ135 % 79.63 oranında ayrım yaparken, MQ2, % 78.46 ve MQ 7 % 80.92 oranında ayrım yapabildiği sonucuna varılmıştır. Elde edilen sinyallerin yorumlanmasında yürütülen sınıflandırma çalışmaları sonucunda en iyi sınıflandırmalar karar ağaçlarından (decision trees) elde edilmiştir ve diğer sınıflandırma yöntemlerine göre daha yüksek başarı sağladığı görülmüştür. Yürütülen bu çalışma ile kahve sektörü için önemli bir sorun olan daha ucuz kahvelerin kaliteli olanlar ile karıştırılması durumu hızlı ve ekonomik bir şekilde dizayn edilen e-burun düzeneği sayesinde tespit edilmiştir.

In this study, thirty-one Lactobacillus delbrueckii subsp. bulgaricus and thirty-four Streptococcus thermophilus were screened for β-galactosidase enzyme activities and specific activities. The β-galactosidase enzyme activities were determined by using o-nitrophenyl-beta-D-galactopyranoside (o-NPG) as a substrate. L. delbrueckii bulgaricus strains showed specific activity between 0.186-6.500 U/mg, while S. thermophilus strains exhibited specific activity between 0.172-5.064 U/mg. The highest specific enzyme activities among bacteria cultures were determined at L. delbrueckii subsp. bulgaricus ZN541 (6.500 U/mg) and S. thermophilus Z1052 (5.064 U/mg) strains. The ZN541 and Z1052 strains that showed high specific β-galactosidase activity were selected and the effects of different conditions (pH, temperature, lactose concentration, and fermentation time) on the activities of β-galactosidase enzymes of these strains were determined. L. delbrueckii subsp. bulgaricus suşları 0.186-6.500 U/mg arasında spesifik aktivite gösterirken, S. thermophilus suşları 0.172-5.064 U/mg arasında spesifik aktivite göstermiştir. L. delbrueckii subsp. bulgaricus ZN541 (6.500 U/mg) ve S. thermophilus Z1052 (5.064 U/mg) suşlarının yüksek spesifik aktivite yeteneğine sahip oldukları tespit edilmiştir. Yüksek spesifik β-galaktozidaz aktivitesi gösteren ZN541 ve Z1052 suşları seçilerek, farklı koşulların (pH, sıcaklık, laktoz konsantrasyonu ve fermantasyon süresi) bu suşlara ait β-galaktozidaz enzimlerin aktivitelerine etkileri belirlenmiştir. ZN541 suşunda optimum enzim aktivitesi için gereken pH’ın 6.2, sıcaklığın 42°C, laktoz konsantrasyonunun %2 ve fermantasyon süresinin 24 saat olduğu tespit edilmiştir. Z1052 suşunda ise optimum enzim aktivitesi için gereken pH’ın 6.8, sıcaklığın 42°C, laktoz konsantrasyonun %4 ve fermantasyon süresinin 24 saat olduğu belirlenmiştir.

Sinirbilim, milyonlarca nöron ve milyarlarca sinapstan oluşan insan beyninin yapı ve fonksiyonlarını anlamaya çalışır. Laboratuvar ortamında böylesine gelişmiş bir sistem için model oluşturmak ve incelemek mümkün değildir. Yirmi yılı aşkın süredir, insan sinir sistemine benzer özellikleri ve kolay incelenebilir yapısı nedeniyle Caernohabditis elegans (C. elegans), nöral ağların davranışını anlamak için faydalı bir model olarak öne çıkmaktadır. For more than two decades, Caernohabditis elegans (C. elegans) has come to the fore as a useful model for understanding the behavior of neural networks, due to its characteristics similar to the human nervous system and its easily observable structure. The created models facilitate quantitative analysis of behavior and neural activities and understanding the functioning of neural networks. Thus, research can be done at both the cell and the organism level. Since the purpose of neuroscience studies is to understand how the information transmitted from sensory neurons under the influence turns into an output response by motor neurons and to understand the role of the effect in making these responses a repetitive, consistent behavior; C. elegans, which has the advantage of being the first animal whose genome has been completely sequenced, is easy to observe with different techniques, and sheds light on the mammalian neural-behavioral pattern, has been used quite efficiently in these studies. In these researches, it is thought that there has been an increase after the 2000s, depending on the developments related to the medium and characterization. Research in this area has increased exponentially from the early 2000s until 2021. When neuroscience studies with C. elegans were examined by country, it was seen that countries such as America and Germany were the leading ones. While 67 of the 245 publications made in SCI-Expanded journals on this subject were neuroscience researches, 40 of them were made by researchers working in multidisciplinary fields. In this study, the general characteristics of C. elegans and its place in neuroscience research will be mentioned and the distribution of these studies by years and countries will be evaluated.

Bu araştırma yakıt olarak kömür kullanan iki termik santralin yer aldığı Çatalağzı bölgesinde yapılmıştır. Çalışmada, 39 noktadan biyomonitör karayosunu (Hypnum cupressiforme Hedw.) örnekleri toplanmış ve bu örneklerde bazı polisiklik aromatik hidrokarbonların (PAH’lar) ve iz elementlerin atmosferik birikimleri araştırılmıştır. Karayosunu örneklerindeki PAH’ların ekstraksiyonu ultrasonik banyo kullanılarak diklorometan ile yapılmış ve ardından bir temizleme işlemi sonrasında elde edilen numuneler ultraviyole dedektörlü yüksek performans sıvı kromatografisi (HPLC-UV) ile analiz edilmiştir. Karayosunu örneklerinde ölçülen toplam PAH konsantrasyonları 53,7 ile 1707,9 ng/g arasında değişiklik göstermiştir. Fenantren, floranten ve piren baskın PAH’lar olarak tespit edilmiştir. İz elementler ile ilgili olarak, karayosunu örnekleri mikrodalgada çözünürleştirme işlemi sonrasında indüktif eşleşmiş plazma-kütle spektrometresi (ICP-MS) ile analiz edilmiştir. Yosun örneklerinde iz elementler için bulunan konsantrasyon sıralaması Zn>Pb>Cu>Cr>Ni>As>Cd şeklindedir. PAH’ların ve metallerin hem kendi içlerinde hem de birbirleriyle olan muhtemel ilişkileri Pearson korelasyon analizi ile incelenmiş ve bazı değişkenler arasında istatistiksel olarak anlamlı ilişkilerin olduğu bulunmuştur. PAH’lar ve iz elementler için örnekleme noktalarına göre birikim yoğunluk haritaları oluşturulmuş ve söz konusu kirleticilerin termik santrallere yakın noktalarda daha yüksek seviyelerde olduğu gözlenmiştir., This research was carried out in the Çatalağzı region (Zonguldak Province), where two coal-fired thermal power plants are located. In this study, biomonitor moss samples (Hypnum cupressiforme Hedw.) were collected from 39 points, and the atmospheric accumulations of some polycyclic aromatic hydrocarbons (PAHs) and trace elements were investigated in these samples. PAHs in the moss samples were extracted with dichloromethane using an ultrasonic bath, and the samples obtained after a cleaning process were analyzed by high performance liquid chromatography with ultraviolet detector (HPLC-UV). Total PAH concentrations determined in moss samples were ranged from 53.7 to 1707.9 ng/g. The predominant PAHs were phenanthrene, pyrene and fluoranthene. Regarding trace elements, moss samples were analyzed by inductively coupled plasma-mass spectrometry (ICP-MS) after microwave digestion process. The concentration order was found for trace elements in moss samples was Zn> Pb> Cu> Cr> Ni> As> Cd. Possible relationships between PAHs and metals, both within themselves and with each other, were investigated using Pearson correlation analysis, and it was found that there were statistically significant relationships between some variables. Deposition density maps were created according to the sampling points for PAH and trace elements, and it was observed that the pollutants were at higher levels near the thermal power plants.

Amaç: Kanser tahrip edici, ölümcül bir hastalıktır ve dünya çapındaki ölüm nedenleri arasında ikinci sırada yer almaktadır. Mide kanseri gibi kanser türleri için farklı tedavi yöntemleri mevcut olsa da çoğu tedavi yöntemleri birçok yan etkiye sahiptir. Çeşitli iyileştirici özelliklere sahip Cuscuta campestris' in kanser hücreleri üzerindeki etkilerinin araştırılması yeni bir alandır ve etkileri tam olarak çalışılmamış ve aydınlatılmamıştır. Bu çalışmada, C. campestris özütünün farklı konsantrasyonlarının mide kanseri hücreleri üzerindeki sitotoksik, genotoksik, apoptotik ve reaktif oksijen türlerinin üretimindeki etkilerini araştırdık. Gereç ve yöntem: Araştırmamızda, C. campestris için antioksidan, pro-oksidan ve radikal temizleyici aktiviteleri değerlendirildi ve miktar tayini LC-MS / MS yöntemi ile analiz edildi. C. Campestris’ in normal hücrelere kıyasla mide kanseri (AGC) hücreleri üzerindeki seçiciliğini göstermek için insan normal deri fibroblastik (CCD-1079Sk) hücre hattı kullanıldı. Apoptoz belirteçlerinin tayininde akridin oranj / etidyum bromür çift boyama, akış sitometrisi ve western blot metotlarından faydalanıldı. Genotoksik aktivite tayini, Comet analizi ile gerçekleştirildi. Bulgular: Sonuçlarımız, C. Campestris etanolik özütünün doza bağlı bir şekilde, mide kanseri hücre hattı üzerinde normal hücrelere kıyasla önemli ölçüde daha yüksek sitotoksik etkilere sahip olduğunu göstermiştir. Ayrıca, elde ettiğimiz veriler C. campestris'in mide kanseri hücre hatlarında reaktif oksijen türlerinin oluşumunu arttırdığını ve DNA hasarına neden olduğunu açıkça göstermiştir. Yapılan apoptoz tayin ölçümleri C. campestris' in bölünmüş kaspaz-3, bölünmüş kaspaz-9, bölünmüş PARP ve P-53'ü aktive etmesiyle kanser hücreleri üzerinde apoptotik bir etkiye sahip olduğunu da doğrulamıştır. Sonuç: Bu çalışmanın sonucu olarak, C. Campestris’in mide karsinomu tedavisinde umut vadeden bir antikanser ajanı olabileceği düşünülmektedir. Cancer is a destructive disease and second most common cause of death worldwide. Although there are different therapeutic methods for cancers such as gastric cancer, most of them possess many adverse effects. Investigating the effects of Cuscuta campestris, which has various medicinal properties, on cancer cells is a new field and has not been fully elucidated. In this study, we investigated the effects of cytotoxic, genotoxic, apoptotic and reactive oxygen species production on gastric cancer cells with various concentrations of C. campestris extract.MATERIAL AND METHOD: Content analysis of C. campestris was measured by LC-MS/MS method. Selectivity on gastric cancer cell line (AGC cell line) was compared to normal cells, human normal skin fibroblastic cells (CCD-1079Sk) for all experiments. Antioxidant, pro-oxidant and radical scavenging activities were evaluated, and apoptosis indicators were measured via acridine orange/ ethidium bromide double staining, flow cytometer and western blotting assays. Genotoxic activity assay was carried out by using Comet assay. RESULTS: Our results demonstrated that ethanolic extract of C. campestris has significantly higher cytotoxic effects on gastric cancer cell line than normal cells in a dose dependent manner. We also observed that C. campestris increased the generation of reactive oxygen species in gastric cancer cell lines and caused DNA damage. Apoptosis detection assays further confirmed that C. campestris has an apoptotic effect by activated of cleaved caspase-3, cleaved caspase-9, cleaved PARP and P-53. CONCLUSION: Hence, C. campestris might represent a promising anticancer agent for gastric carcinoma treatment.

In this study, a total of 248 moss samples were collected from 25 stations as a result of the field studies carried out in the Acarlar Lake Longoz Forests during the different vegetation periods of 2016 and 2017. According to the identification studies of the collected samples, 23 families, 58 genera and a total of 93 taxa belonging to these genera were determined. Of the determined taxa, 29 are new records for Sakarya province bryophyte flora and 6 taxa are new records for A2 square. Also, registered from this region; Atrichum crispum, Pseudephemerum nitidum and Dicranella staphylina are also important species for bryophyte flora of Turkey., Bu çalışmada, 2016-2017 yıllarının farklı vejetasyon dönemlerinde Acarlar Gölü Longoz Ormanlarında yapılan arazi çalışmaları sonucunda 25 istasyondan toplam 248 karayosunu örneği toplanmıştır. Toplanan örneklerin teşhis çalışmaları yapılmış olup 23 familya, 58 cins ve bu cinslere ait toplam 93 takson tespit edilmiştir. Bu taksonlardan; 29’u Sakarya ili briyofit florası için, 6 takson ise A2 karesi için yeni kayıtır. Ayrıca, bu bölgeden kaydı verilen; Atrichum crispum, Pseudephemerum nitidum ve Dicranella staphylina Türkiye briyofit florası için önemli taksonlardandır.

Hyalomma marginatum’un ovaryumu ve ovogenez süreci ilk kez bu çalışmada ışık mikroskobu düzeyinde histolojik ve histokimyasal olarak incelenmiştir. Ovaryum nurse ve folikül hücrelerinden yoksundur ve bu nedenle panoistik olarak tanımlanmıştır. İçi boş tübül biçimindeki ovaryumun duvarı tek sıra epitel hücrelerinden ve gelişmekte olan ovositlerden oluşur. Ovositler bir pedisel ile hemosöle bakacak şekilde ovaryum duvarına bağlıdır. Sitoplazmanın görünüşü, germinal vesikülün varlığı, yumurta sarısı granüllerinin bileşimi ve koryonun oluşumuna göre gelişmekte olan ovositler sınıflandırılmış ve dört evre tanımlanmıştır. Olgun ovositler küresele yakın eliptik şekilli olup, en büyük ovosit 315x295 µm çapındadır. Protein ve karbonhidratların varlığını gösteren boyama teknikleri bu kene türünde, yumurta sarısı granüllerinde hem proteinlerin hem de karbonhidratların bulunduğunu, vitellogenezde karbonhidratların proteinlerden önce sentezlendiğini ve koryonda karbonhidratların baskın olduğunu göstermiştir. Çok yaygın olmasa da ovaryumda ovosit yozlaşması gözlenmiştir. With this study, the ovary and oogenesis in Hyalomma marginatum were histologically and histochemically examined at light microscope level for the first time. The ovary lacks nurse and follicular cells, thus defined as panoistic type. Wall of the ovary, a hollow tubular structure, is composed of a layer of epithelial cells and developing oocytes. Facing hemocoel, oocytes are attached to the ovarian wall through a pedicel. The developing oocytes were classified according to cytoplasm appearance, presence of germinal vesicle, constitution of yolk granules, and formation of chorion, and accordingly four developmental stages were described. Mature oocytes were oval to rounded in shape, the largest one being 315x295 µm in diameter. Staining techniques revealing the presence of proteins and carbohydrates indicate in this tick species that yolk granules contain both proteins and carbohydrates, that carbohydrates are first to be synthesized prior to proteins during vitellogenesis, and that carbohydrates predominate in chorion. Oocyte degeneration was observed although it was not so common in the ovary.

Bu çalışma, örtüaltı hıyar yetiştiriciliğinde sorun olan kök-ur nematodları (Meloidogyne spp.)’na karşı kimyasal mücadeleye alternatif mücadele olanaklarını araştırmak amacıyla 2017-2018 yıllarında Kocaeli ilinde yürütülmüştür. Denemeler, saksı denemesi şeklinde tesadüf parselleri deneme desenine göre 8 karakter ve 10 tekerrürlü olarak gerçekleştirilmiştir. Deneme yerleri Başiskele ilçesi Kullar mahallesinde yer alan üreticiye ve Arslanbey Meslek Yüksek Okulu’na ait ısıtmasız seralardır. Domates saf kültürden elde edilen ikinci dönem kök-ur nematodu (Meloidogyne incognita) larvaları, Ceylan F1 hıyar çeşidi bitkileri, Hint hardalı (Barassicae Juncae), Kadife çiçeği (Tagetes erecta) bitki aksamları, Susam yağı (Nemax®), Trichoderma herzianum Rifai KRL-AG2 4x106 spor/g (T-22 Planter Box®), Arbüskülar Mikorizal Fungus (Endo root®, %23,5 canlı Glomus spp.) ve karşılaştırma ilacı Fenamiphos etken maddeli (Nemasat®) denemenin materyallerini oluşturmuştur. Çalışma sonucunda, uygulama yapılan tüm karakterlerin pozitif kontrolle göre daha düşük ur skala değerlerine sahip oldukları ve istatistiki olarak (P ≤ 0,05) farklı gruplarda yer aldıkları belirlenmiştir. Elde edilen bulgulara göre; kök-ur nematodlarının hıyar bitkisinde oluşturduğu urların engellenmesi, topraktaki nematod sayısının azaltılması, daha az yumurta kümesi oluşumu, bitki boyunu artırıcı etkisi, bitki yaş ve kuru ağırlığına, kök yaş ve kuru ağırlığına etkileri bakımından her bir saksıya 50 g Hint hardalı yeşil aksam uygulamalarının etkisi diğer karakterlerdeki uygulamalara göre daha yüksek bulunmuştur. Hint hardalından sonra Trichoderma harzianum ve Susam yağı karakterleri etkili bulunmuştur. Pozitif kontrole göre, tüm uygulama karakterlerinde yumurta kümesi miktarı ve urlanmanın azaldığı, bitki gelişim parametrelerinin arttığı belirlenmiştir. Elde edilen bulgular ışığında tüm uygulamaların hıyar yetiştiriciliğinde kök-ur nematodlarının alternatif mücadelesinde kullanılabileceği kanısına varılmıştır., This study was conducted in order to investigate the alternative control to chemical control against root nematodes (Meloidogyne spp.) which was a problem in greenhouse cucumber growing in Kocaeli province during 2017-2018. The experiments were carried out according to randomized plot design with 8 characters and 10 replications as pot trials. The experimental locations were unheated greenhouses belonging to Arslanbey Vocational High School and the producer inhabited in the district of Kullar. The second stage Root nematode (Meloidogyne incognita) larvae obtained from the pure culture of tomatoes, Ceylan F1 cucumber variety plants, Indian mustard (Barassicae Juncae), Marigold (Tagetes erecta) plant components, Sesame oil (Nemax), Trichoderma herzianum Rifai KRL-AG2 4x106 spore/g (T-22 Planter Box®), Arbaceous mycorrhizal Fungus (Endo root®, 23,5% live Glomus spp.) and the comparative nematicides Fenamiphos (Nemasat®) composed of the experiment materials. As a result of the study, it was determined that all the characters in which the plant parts were applied had lower root-knot scale values than the positive control and they were included in different groups statistically (P ≤ 0,05). According to the findings obtained; it was been found more effective effect of 50 g Indian mustard green component applications in each pot than other characters, in terms of prevention of knots created by root-knot nematodes in cucumber plants, reduction of the number of nematodes in the soil, formation of less egg-cluster, increasing effect of plant height, effects of plant wet and dry weight and root wet and dry weight. After the Indian mustard, Trichoderma harzianum and Sesame oil characters were found to be effective. According to the positive control, it was determined that egg-cluster and knotting amounts decreased and plant development parameters increased in all application characters. In the light of the findings, it is concluded that all applications can be used in the alternative conrol of root-knot nematodes in cucumber cultivation.

Farklı toprak habitat ve sub-habitaların fungus oluşumuna ve dağılımına etkisini incelemek için Ki-kare testi yapılmıştır. Toprak habitatı ile B. bassiana’nın oluşumu ve dağılımı arasında istatiksel olarak anlamlı bir fark olmadığı (x2=,000, p>.005), M. anisopliae’nin oluşumu ve dağılımı ile toprak habitatının arasında istatiksel olarak anlamlı bir fark olduğu belirlenmiştir (x2=7,680, p, In this study, it was investigated the effect of soil factors on the occurrence and distribution of entomopathogenic fungi (Epf) isolated from Erzincan soils. For this purpose, 60 soil samples were collected from cultivated and natural areas. Two epf species, Beauveria bassiana (balsamo) Vuillemin and Metarhizium anisopliae (Metschnikoff) Sorokin, were isolated from the soils. Chi-square test was conducted to examine the effects of different soil habitats and sub-habitats on the occurrence and distribution of fungi. Bu çalışmada, 4-metoksiasetofenonun enantiyoseçici indirgenmesinde yedi biyokatalizörün indirgeme kapasitesi tam hücre biyokatalizör olarak araştırılmıştır. Lactobacillus paracasei BD28'in en iyi indirgeme kapasitesine sahip olduğu bulunmuştur. pH, inkübasyon süresi, çalkalama hızı ve sıcaklık gibi farklı parametrelerin enantiyomerik aşırılık ve dönüşüm üzerindeki etkileri araştırıldı. Tam hücre biyokatalizörü Lactobacillus paracasei BD28 kullanılarak, genel alerjik yanıt için tedavi fonksiyonuna sahip sikloalkil [b] indollerin sentezinde kullanılabilen (S)-1-(4-metoksifenil) etanol, gram ölçeğinde, yüksek verimli ve enantiyomerik olarak saf halde üretilmiştir. Gram ölçekli üretim gerçekleştirildi ve % 95 verimle optik olarak saf formda 9,69 g (S)-1-(4-metoksifenil) etanol üretildi. Bu, kimyasal işlemlere kıyasla (S)-1-(4-metoksifenil) etanol üretimi için ucuz, temiz ve çevre dostu bir işlemdir., In this study, the reductive capacity of seven biocatalysts were investigated as whole-cell biocatalyst in the enantioselective reduction of 4-metoxyacetofhenone. Lactobacillus paracasei BD28 was found to have the best reductive capacity. Effects of different parameters such as pH, incubation time, agitation speed and temperature, on enantiomeric excess and conversion were investigated in a bioconversion. (S)-1-(4-methoxyphenyl) ethanol which, can be employed for the synthesis of cycloalkyl [b] indoles which have the treatment function for general allergic response, was produced in gram-scale, high yield and enantiomerically pure form using whole-cell biocatalyst of Lactobacillus paracasei BD28. The gram-scale production was carried out, and 9.69 g of (S)-1-(4-methoxyphenyl) ethanol in optically pure form was produced with 95 % yield. This is a cheap, clean and eco-friendly process for production of (S)-1-(4-methoxyphenyl) ethanol compared to chemical processes.

In the study, the micromorphology of labellum in samples belonging to 10 different Ophrys taxa was examined by Scanning Electron Microscopy (SEM) method and the results were presented in comparative tables. While the samples were collected from the field, they were taken to 70% alcohol, some of them were dried according to the herbarium rules and identified. The labellum samples extracted from alcohol were dissected under the stereomicroscope with scalpel and needles and placed directly into the iron apparatus of the SEM device with double-sided adhesive tapes. These samples, whose alcohol was completely evaporated by waiting for one night, were examined directly with Scanning Electron Microscope (SEM) and labellum microphotographs were taken. According to our findings, it was determined that the speculum structures are very different from each other, and the shape and distribution of the hair / papilla in the basal, median and lateral regions differ from species to species.It was also determined that the apex region differs in O. fusca, O. lutea, O. speculum, O. mammosa, O. pseudomammosa, O. heldreichii, O. umbilicata, and O. holoserica species. In summary; It was concluded that the labellum speculum and apex structure of the 1st degree and the basal, median and lateral region surface characteristics of the 2nd degree differ widely between the species., Çalışmada 10 değişik Ophrys taksonuna ait örneklerde labellum mikromorfolojisi Taramalı Elektron mikroskobu (SEM) yöntemi ile incelenmiş ve sonuçlar karşılaştırmalı tablolar halinde sunulmuştur. Örnekler, araziden toplandığı sırada %70 alkole alınmış, bir kısmı da herbaryum kurallarına göre kurutularak teşhisleriyapılmıştır. Alkolden çıkarılan labellum örnekleri, bisturi ve iğnelerle stereomikroskop altında dissekte edilerekdoğrudan _üzeri çift taraflı yapışkan bantlar bulunan_ ve SEM cihazına ait çelik aparatlara yerleştirilmiştir. Bir gece kadar bekletilerek alkolü tamamıyla uçurulan bu örnekler, doğrudan Taramalı Elektron Mikroskobu (SEM) ile incelenmiş ve labellum mikrofotoğrafları çekilmiştir. Bulgularımıza göre spekulum yapılarının birbirinden çok farklı olduğu, bazal, medyan, lateral bölgelerde tüy / papilla şekli ve dağılımının türden türe farklılık gösterdiği tespit edilmiştir. Ayrıca apeks bölgesinin O. fusca, O. lutea var. minor, O. speculum, O. mammosa, O. pseudomammosa, O. heldreichii, O. umbilicata, O. holoserica türlerinde de farklılık gösterdiği tespit edilmiştir. Özetle; 1. derecede labellum spekulum ve apeks yapısının, 2. derecede de bazal, medyan ve lateral bölge yüzey karakterlerinin türler arasında çok farklılık gösterdiği sonucuna varılmıştır.

Üç tarafı denizlerle çevrili ve yüzlerce tatlı su kaynağı bulunan Türkiye'de tatlı su ve deniz türleri dahil olmak üzere toplam dokuz yüz üç balık türü olduğu bildirilmiştir. Türkiye balık sitogenetiğinde son otuz beş yılda, toplam balık türlerinin yaklaşık%11'ine karşılık gelen, 38'i endemik tür olan ve 17'den fazla takımda yer alan 103 tür/alttür kromozom özellikleri incelenmiştir. Tüm bu çalışmalarda geleneksel sitogenetik yöntemler kullanılarak (Giemsa, NOR-, C-, GTG-, Q- ve RE boyama-bantlama) balıkların karyotipleri, kromozom sayıları ve yapıları, kol sayıları (NF) ve bazı gen bölgeleri (NOR, heterokromatin vb.) ortaya çıkarılmıştır. Bununla birlikte, Türkiye’de ilk defa üç endemik cyprinid türünün kromozomlarında CMA3 ve DAPI boyama da yapılmıştır. 48 balık türünün kromozomlarında tekrarlayan DNA ve heterokromatik bölgeler NOR ve C-bantlama ile belirlendi. Türkiye'deki diğer balık gruplarına kıyasla en çok görülen Cypriniformes takımının üyesi (Cobitidae, Nemacheilidae, Cyprinidae ve Leuciscidae familyalarından) 69 türün sitotaksonomik ve sitogenetik analizi yapılmıştır. Bu sonuçlara göre sitogenetiksel incelenen balıklarda çoğunlukla kromozom sayısı 2n=50 olduğu tespit edilmiştir. En yüksek kromozom sayısının 2n = 150 olduğu ve en düşük kromozom sayısının iç sularda doğal olarak bulunan bir Bagridae türüne (2n = 32) ait olduğu da kaydedildi., In Turkey surrounded by sea on three sides and hundreds of freshwater resources, it was reported that there is a total of 903 species of fish including freshwater and marine species. In the last thirty-five years in Turkish fish cytogenetics, 103 species/subspecies, 38 of which are endemic species and belonging to more than seventeen orders, corresponding to approximately 11% of the total fish species, had been studied their chromosomes. In all studies, their karyotypes, chromosome number and structure, the arm number (NF), and some specific gene regions (NOR, heterochromatin, etc.) on chromosomes of fish had been revealed by using conventional cytogenetic methods (Giemsa, AgNOR-, C-, GTG-, Q- and RE staining/banding). However, CMA3 and DAPI fluorescence staining in the chromosomes of three species of endemic cyprinids is made for the first time in Turkey. Repetitive DNA and heterochromatic regions in the chromosomes of 48 fish species were determined by NOR and C-banding. Compared to other fish groups in Turkey, the most widely seen Cypriniformes order's member (from families of Cobitidae, Nemacheilidae, Cyprinidae, and Leuciscidae) is made the cytotaxonomic and cytogenetic analysis of 69 species. According to these results, it was found that mostly chromosome number was 2n = 50 in fishes which were examined cytogenetically. It had also been noted that the highest chromosome number is 2n = 150, and the lowest chromosome number was belonging to a Bagridae species (2n = 32) naturally found in inland waters.

In this study, the chemical structure of the wood and bark of willow (Salix alba L.), grown naturally in the Lakes Region, was investigated. The main components of the cell wall, the solubility values, the amount of lipophilic and hydrophilic substances obtained by hexane and acetone:water (95:5, v:v) extraction of willow wood and bark, and also fatty acid composition and suberin analysis were studied in the bark. Holocellulose amount was determined in wood and bark, respectively, as 81.70% and 70.18%, α-cellulose 48.42% and 40.32%, klason lignin 24.01% and 33.81%, hot water solubility 4.09% and 21.51%, 1% NaOH solubility 13.88% and 43.48%, hexane solubility 8.24 mg/g and 10.64 mg/g, acetone:water solubility 31.26 mg/g and 30.72 mg/g. Linoleic acid (25.89%) in wood and N-ethylacetamide (32.94 %) in bark were determined as the major lipophilic substances after hexane extraction. In additon, after hexane extraction, the amount of fatty acids was investigated in the bark samples according to the Supelco 37 fatty acid mixture, 9,12-octadecadienoic acid (55.71 %) was found to be predominant fatty acid. 2-methyl-4-keto-pentan-2-ol (17.00%) in the wood and sucrose (32.24%) in the bark were seen as the most common components as hydrophilic substances after acetone:water extraction., Bu çalışmada Göller Bölgesi’nde doğal olarak yetişen Söğüt (Salix alba L.) ağacının odun ve kabuğunun kimyasal yapısı araştırılmıştır. Söğüt ağacının odun ve kabuk örnekleri üzerinde hücre çeperi ana bileşenleri, çözünürlük değerleri, heksan ve aseton:su (95:5, v:v) ekstraksiyonu ile elde edilen lipofilik ve hidrofilik madde miktarı ve bileşenleri araştırılmış olup ayrıca kabukta yağ asidi tayini ve suberin analizi yapılmıştır. Holoselüloz oranı odun ve kabukta %81.70 ve % 70.18, α-selüloz %48.42 ve %40.32, lignin miktarı %24.01 ve %33.81, sıcak su çözünürlüğü %4.09 ve %21.51, %1’lik NaOH çözünürlüğü %13.88 ve %43.48, heksan çözünürlüğü 8.24 mg/g ve 10.64 mg/g, aseton:su çözünürlüğü 31.26 mg/g ve 30.72 mg/g olarak tespit edilmiştir. Heksan ektraksiyonu sonucu elde edilen lipofilik madde miktarları ve bileşenleri arasında, odunda linoleic asit %25.89, kabukta ise N-ethylacetamide %32.94 olarak en yüksek oranda tespit edilen bileşenlerdir. Heksan ekstraksiyonu sonrası kabuk örneklerinde Supelco 37’li yağ asidi karışımına göre yağ asitleri miktarları araştırılmış ve 55.71 mg/g ile 9,12-octadecadienoic acid en baskın bulunan yağ asidi olarak tespit edilmiştir. Aseton:su ekstraksiyonu sonucu elde edilen hidrofilik madde miktarları ve bileşenleri incelendiğinde ise odunda 2-methyl-4-keto-pentan-2-ol (%17.00) ve kabukta sucrose (%32.24) en fazla bulunan bileşenlerdir.

Amaç: Özellikle son 10 yılda Enterobacterales üyeleri arasında karbapenem direnci artan sıklıkta rapor edilmeye başlanmıştır. Karbapenemaz üreten izolatların taranması ve saptanması, hem tedavinin doğru yönlendirilebilmesi hem de yayılımın önüne geçilebilmesi açısından önem taşımaktadır. Çalışmamızda hastanemiz Mikrobiyoloji laboratuvarına gönderilen örneklerden ardışık olarak soyutlanan karbapenem dirençli Klebsiella pneumoniae izolatlarının karbapenemaz tipleri ve moleküler epidemiyolojik ilişkilerinin saptanması amaçlanmıştır.Yöntem: Temmuz- Eylül 2014 tarihleri arasında Mikrobiyoloji laboratuvarına gönderilen örneklerden soyutlanan toplam 32 karbapeneme dirençli K. pneumoniae izolatı çalışmaya alındı. İzolatların tür düzeyinde tanımlanması klasik yöntemlere ek olarak BD Phoenix ID/AST otomatize sistemi ile yapıldı. İzolatların karbapenemaz tipleri (blaOXA-48, blaNDM, blaIMP, blaKPC, blaVIM ve blaGES) PCR ile araştırıldı. İzolatlar arasındaki klonal ilişki PFGE ile değerlendirildi. Bulgular: Onsekiz izolatın yoğun bakım ünitelerinden, dokuz izolatın servislerden ve beş izolatın polikliniklerden gönderilen örneklerden izole edildikleri görülmüştür. İzolatların tümünde blaOXA48 geni pozitif olarak saptanmış, diğer karbapenemaz genleri bulunmamıştır. Hastanemizde 32 farklı hastadan üretilen izolatların A-L olarak adlandırılan 12 farklı PFGE pulsotipine sahip olduğu belirlendi. Bunlar arasında en fazla görülenler B (n=18) ve bununla yakın ilişkili B1 paterni (n=2) idi. Geriye kalan izolatlar, birbirinden farklı olan 11 tiple temsil edildi. Salgından sorumlu B pulsotipine sahip ilk izolatın Genel Yoğun Bakım ünitesinden kaynaklanarak yayılım göstermiş olduğu görüldü. Sonuç: Karbapeneme dirençli K. pneumoniae izolatlarının hastanede yayılımının gastrointestinal kolonizasyonu olan hastalardan, hastane personelleri aracılığıyla diğer hastalara izolatların transferi yoluyla gerçekleşmiş olduğu düşünülmüştür. Bu nedenle aktif sürveyans programları ile kolonizasyonun saptanarak temas izolasyonu ve etkin enfeksiyon kontrol önlemlerinin uygulanması yoluyla hastanelerde izolatların yayılımı sınırlandırılabilir Objective: Carbapenem resistance has been reported with increasing frequency among members of Enterobacterales, especially in the last 10 years. Screening and detection of carbapenemase-producing isolates is important in terms of both directing the treatment and preventing its spread. In our study, it was aimed to determine the carbapenemase types and molecular epidemiological relationships of carbapenem resistant Klebsiella pneumoniae isolates, which were isolated sequentially from the samples sent to microbiology laboratory of our hospital.Method: A total of 32 carbapenem-resistant K. pneumoniae isolates of the samples sent to microbiology laboratory between July and September 2014, were included in the study. In addition to classical methods, identification of isolates at species level was made with BD Phoenix ID/AST automated system. Carbapenemase types (blaOXA-48, blaNDM, blaIMP, blaKPC, blaVIM and blaGES) of the isolates were investigated by PCR. The clonal relationship between the isolates was assessed with PFGE.Results: It was noted that 18 isolates were obtained from intensive care units, 9 from inpatient and 5 from outpatient departments. The blaOXA48 gene was found in all isolates while the other carbapenemase genes were not found. It was determined that strains were isolated from 32 patients in our hospital had 12 different PFGE pulsotypes, named as A-L. Among these, the most common ones were B (n=18) and closely related B1 pattern (n=2). The remaining isolates were represented by 11 different types. It was observed that the first isolate with B pulsotype was responsible for the spread of the outbreak from General Intensive Care Unit.Conclusion: It has been thought that the spread of carbapenem- resistant K. pneumoniae isolates in the hospital was probably occurred through the transfer of isolates from patients with gastrointestinal colonization to other patients through hospital staff. Therefore, the spread of the isolates in hospitals can be limited by detecting colonization with active surveillance programs and by applying contact isolation and effective infection control measures.

Background: Hypericum species have chemopreventive and antioxidant effects which have been described in the past. However, Hypericum perforatum L. extract has not been the subject of any in vivo chemoprevention studies. The purpose of this study was to investigate the effects of H. perforatum L. extract on the oxidant-antioxidant system in 7,12 dimethylbenzantracene applied tongue tissues of rats.Methods: Thirty male Wistar rats were divided into four groups. Control group: only paraffin, DMBA group: only 7,12-dimethylbenzantracene, HP+DMBA group: 7,12 dimethylbenzantracene and H. perforatum L. extract and HP group: only H. perforatum L. extract application to the oral mucosa. After a 16-week study period, animals were sacrificed and tongue samples were taken. Superoxide dismutase, catalase, glutathione peroxidase enzyme activities, and malondialdehyde and total antioxidant status levels were measured.Results: HP+DMBA group revealed significant differences with regard to catalase and superoxide dismutase enzymes compared to control, DMBA and HP groups (Catalase: p = 0.019, 0.019, 0.000, respectively; Superoxide dismutase: p = 0.001, 0.012, 0.009, respectively). Parallel to this data, total antioxidant status value in the same group was decreased with regard to other groups. Glutathione peroxidase enzyme activity and malondialdehyde levels did not demonstrate any significant differences among groups.Conclusions: H. perforatum L. extract did not reveal any significant evidence that indicates an antioxidant effect. Moreover, antioxidant enzymes (catalase and superoxide dismutase) were believed to be suppressed by a by-product of H. perforatum L. and 7,12 dimethylbenzantracene reaction in the HP+DMBA group.

Amaç: İnfantil kolik, aile ve hekim için oldukça kaygı verici 4 aylıktan küçük bebeklerde sık olarak acile başvuru nedeni olan ve etiyolojisi net bilinmeyen ağlama krizleridir. Yoğun ağlama krizleri sırasında çaresiz kalan aileler, çevredekilerin de etkisi ile farklı tedavi yöntemleri arama eğilimindedirler. 28 yaşındaki annenin 1. gebeliğinden normal spontan vaginal yol ile 39 gebelik haftasında doğan iki aylık erkek bebeğin, son 24 saattir uyandırılamaması şikayeti, acil servise başvuru nedenidir. Acil servise başvuru öyküsünde ailenin, bebeğin yoğun ağlama nöbetleri nedeniyle eczaneden alınan farklı farmakolojik ürünleri ardı ardına denediği, ancak bebeğin şikayetlerinin devam etmesi nedeniyle, çevredekilerinin de önerisi ile “elma yağı” adı ile satın aldıkları ve içeriğini bilmedikleri bir ürünü bebeğin göbek bölgesine masaj yoluyla uygulandığı ve sonrasında bebeğin ağlamalarının azaldığı ve uykuya meyilli olduğu bilgisi yer almıştır. Hastanedeki tedavi sırasında bebeğin sadece uyarılınca uyandığının belirlenmesi sonucu aileden kullandıkları ürün istenmiştir.Gereç ve Yöntem: Piyasadan satın alınan ve bebeğe masaj yoluyla uygulanan “elma yağı” adı ile satışa sunulmuş ürünün, GC-FID ve GC/MS yöntemleri ile içerik analizi yapılmıştır. Uçucu bileşiklerin teşhisi için çeşitli bilimsel veri kütüphaneleri kullanılmıştır.Sonuç ve Tartışma: Bu çalışma, son yıllarda doğala yönelme kavramının bilinçsiz ellerde geri dönüşümü olmayacak kadar olumsuz sonuçlara neden olabileceğini göstermekte ve aynı zamanda ilaçlar ve tıbbi doğal ürünler alanında yetkin olan eczacılık mesleğinin önemini göz önüne sermektedir., Objective: Infantile colic is crying crises that a cause of frequently emergency application in infants younger than 4 months and whose aetiology is unknown. Families who are desperate during intense crying crises tend to seek different treatment methods with the influence of those around. The complaint of not waking up for the last 24 hours of a two-month-old male baby born at 39 weeks of gestation with normal spontaneous vaginal delivery from the first pregnancy of a 28-year-old mother is the reason for admitting to the emergency department. In the history of applying to the emergency department, the family tried different pharmacological products purchased from the pharmacy one after another due to the intense crying attacks of the baby, but the complaints of the baby continued, the family bought a product called "apple oil" and applied the product that they did not know the content to the baby by massaging the oil to the umblicus area with the suggestion of the people around. It was reported that the baby's cries were reduced and he was susceptible to sleep. As a result of the detection that the baby woke only when stimulated during the hospital treatment, the product they used was requested from the family.Material and Method: The content analysis of the product, which was bought from the market and put up for sale under the name of "apple oil" applied to the baby by massaging, was performed using GC-FID and GC/MS methods. Various scientific data libraries have been used to identify volatile compounds.Result and Discussion: This study shows that the concept of turning to nature in recent years can cause irreversible negative consequences in unconscious hands.and also highlights the importance of the pharmacy profession, which is highly competent in the field of medicines and medicinal natural products.

OBJECTIVE Klebsiella pneumoniae, a Gram-negative pathogen, especially which produces carbapenemase, is seen as a major threat to public health due to rapid plasmid-mediated spread of resistance and limited therapeutic options available for treatment. Although colistin has been recognized as a "last resort" antimicrobial for multidrug-resistant K. pneumoniae infections, these isolates have developed resistance to colistin as a result of its intensive use. The aim of this study was to evaluate the efficacy of double-carbapenem treatment of colistin-resistant K. pneumoniae experimental sepsis in mice. METHODS In the study, 8-10-week-old Balb-c mice were divided as control groups (positive and negative) and treatment groups (colistin, ertapenem+meropenem, and ertapenem+meropenem+colistin). Sepsis was developed in mice by an intraperitoneal injection of colistin resistant K. pneumoniae. Antibiotics were given intraperitoneally 3 h after bacterial inoculation. Mice in each subgroup were sacrificed with overdose anesthetic at the end of 24-48 h and cultures were made from the heart, lung, liver, and spleen. Furthermore, homogenates of lung and liver were used to detect the number of colony-forming units per gram. Bacterial clearance was evaluated in lung and liver at different time points. RESULTS When the quantitative bacterial loads in the lung and liver tissues are evaluated, no statistically significant difference was observed between different antibiotic treatments (p>0.05). All three treatment options were not effective, especially in 24 h. Objectives: S.maltophilia despite its low pathogenicity, can cause colonization in immunocompetent and long-term inpatients in the intensive care unit, and can cause pneumonia and bacteremia, often selected from the respiratory and gastrointestinal tract. It is known that especially cystic fibrosis patients are at risk. The aim of this study was to obtain local epidemiological data by examining S. maltophilia isolates and trimetoprim-sulfometaxazole susceptibility. Methods: Ninety six S. maltophilia isolates, isolated from culture samples sent to Kahramanmaras Sutcu Imam University Medical Faculty Hospital Microbiology Laboratory between January 1, 2018 and May 31, 2019 were analyzed retrospectively. Conventional culture and Gram staining characteristics of the isolates were examined. BD Phoenix (BD, USA) automated identification system was used for species level identification and antibiotic susceptibility testing. Results: Microorganism was mostly isolated from the respiratory tract samples (50%) and clinically from the anesthesiology and reanimation intensive care unit with a rate of 28.1%. The trimethoprim-sulfomethaxazole resistance of the isolates was found to be 1.04% (n = 1). Conclusion: Stenotrophomonas maltophilia is resistant to many antibiotics, causing treatment difficulties. Trimethoprim-sulfamethoxazole is the most effective antibiotic that can still be used in Stenotrophomonas maltophilia infections. Especially when S. maltophilia is isolated from cultural examples, pathogen/ colonization should be differentiated and resistance should be prevented by avoiding unnecessary antibiotic use., Amaç: S.maltophilia düşük patojenite özelliklerine rağmen, immün düşkün ve yoğun bakım ünitelerinde uzun süre yatan hastalarda kolonize olabilen, solunum ve gastrointestinal sistemden seçilerek sıklıkla pnömoni ve bakteriyemi gibi enfeksiyonlara neden olabilen bir mikroorganizmadır. Özellikle kistik fibrozisli hastaların risk altında olduğu bilinmektedir. Çalışmamızın amacı, S. maltophilia izolatlarını ve trimetoprim-sülfometaksazol duyarlılık durumunu inceleyerek lokal epidemiyolojik veri elde etmektir. Gereç ve Yöntem: 1 Ocak 2018-31 Mayıs 2019 tarihleri arasında Kahramanmaraş Sütçü İmam Üniversitesi Tıp Fakültesi Hastanesi’ne gelen hastaların kültür örneklerinden izole edilmiş 96 S. maltophilia izolatı retrospektif olarak incelenmiştir. İzolatların konvansiyonel kültürleri yapılmış ve Gram boyanma özellikleri incelenmiş. Tür düzeyinde tanımlama ve antibiyotik duyarlılık testi için BD Phoenix (BD, USA) otomatize identifikasyon test sistemi kullanılmıştır. Bulgular: Mikroorganizma en çok (%50) solunum yolu örneklerinden ve klinik olarak % 28.1 oranıyla anesteziyoloji ve reanimasyon yoğun bakım ünitesinden izole edilmiştir. İzolatların trimetoprim-sülfometaksazol direnci %1,04 (n=1) olarak bulunmuştur. Tartışma ve Sonuç: Stenotrophomonas maltophilia ‘nın birçok antibiyotiğe dirençli olması tedavide güçlüklere neden olmaktadır. Trimetoprim-sülfometaksazol, Stenotrophomonas maltophilia enfeksiyonlarında hala kullanılabilecek en etkili antibiyotiktir. Özellikle kültür örneklerinden S. maltophilia izole edildiğinde etken –kolonizasyon ayırımı yapılmalı ve gereksiz antibiyotik kullanımından kaçınılarak direnç gelişimi önlenmelidir.

Parazitintürlerine ve konakçı immün tepkisine bağlı olarak, hastalıkların üç temel klinik formu; kutanöz, mukokutanözve viseral leyşmanyazis olarak adlandırılmaktadır. Kutanöz leyşmanyazis kronik bir hastalık olup ülserleşmişlezyonların varlığı ile karakterizedir. Hastalık sırasında oluşan cilt patolojisi, kısmen enfekte edenLeishmania türleriyle, aynı zamanda çeşitli klinik sonuçlara neden olan enflamatuvar ve anti-enflamatuvarkonak immün yanıt faktörlerinin kombinasyonu ile belirlenmektedir. Bu çalışmada, Leishmania major veLeishmania braziliensis’e karşı doğal dirençli ve duyarlı olan farelerde hastalığın patogenezi ile bağışıklıkyanıtında rolü olan ve konak-parazit etkileşimlerini belirleyen genleri, moleküler sinyal mekanizmalarınıve moleküllerin biyolojik fonksiyonlarını belirlemek amaçlanmıştır. Bunun için, L.major/L.braziliensis ileenfekte ve sağlıklı farelerden elde edilmiş yirmi dört doku örneğinin ekspresyon profilini içeren GSE56029kodlu transkriptomik veri seti “Gene Expression Omnibus” (GEO) veri tabanından elde edilmiştir. Dahasonra, R betiğinde limma paketi ile diferansiyel olarak eksprese edilmiş genler tanımlanmıştır. AnalizlerdeFDR q< 0.05 ve mutlak log2FC> 2 eşik değer olarak kabul edilmiştir. Ardından diferansiyel olarak ekspreseedilmiş genler için fonksiyonel ve yolak zenginleştirme analizleri “Ingenuity Pathway Analysis” (IPA) ilegerçekleştirilmiştir. Anlamlı derecede farklı seviyede eksprese edilen genlerin her biri için p< 0.01, FDR qdeğeri< 0.01 ve mutlak log2FC> 1 kullanılarak IPA 8.0 yazılım programı ile analiz edilmiştir. Bu analizlerile en yüksek oranda zenginleştirilmiş yolakların enflamasyon, dendritik hücre olgunlaşması ve TREM-1“Triggering Receptor Expressed on Myeloid Cells 1” (TREM-1) sinyal mekanizmaları olduğu ve bağışıklık sisteminin düzenlenmesiyle ilgili diferansiyel olarak eksprese edilmiş genlerin kutanöz leyşmanyazis seyriile yakından ilişkili olduğu belirlenmiştir. Akım-yukarı regülatör analizi ile dokularda biyolojik aktivitelereneden olan gen ekspresyon değişikliklerini açıklayan regülatörün TNF-?, IFNy, IL-1 ?, IL-10RA ve “SignalTransducer and Activator of Transcription-1” (STAT-1) olduğu öngörülmüştür. Çalışmada ayrıca, antileishmanialetkiye sahip olabilecek kimyasal bileşikler de tanımlanmıştır. Bu çalışma ile direnç/duyarlılıkfenotipini belirleyen parazit türlerine ve konağa ait mekanizmalar aydınlatılmaya çalışılmıştır. L.major veL.braziliensis ile enfekte olan BALB/c ve C57BL/6 farelerinde farklılık gösteren gen ekspresyon paternleri,sitokin/kemokinler ve sinyal yolaklarının değerlendirilmesi, enfeksiyonun altında yatan potansiyel mekanizmalarıgenetik açıdan daha iyi anlamamızı sağlayacaktır. Bu sonuçların kutanöz leyşmanyazisin tanıve prognoz tahmini için potansiyel biyobelirteçlerin geliştirilmesi ve yeni tedavi hedefleri hakkında bilgisağlaması açısından gelecek çalışmalar için yol gösterici nitelikte olabileceği düşünülmektedir.

Anthemis ekicii (Asteraceae) is described and illustrated as a new species from Tuz Lake in Aksaray Province in CentralAnatolia. The new species belongs to the section Anthemis, and it is closely related to A. pauciloba var. pauciloba, from which it differs instem, leaves, and capitulum features. Diagnostic characters, description, detailed illustrations, ecology, IUCN conservation status, andpollen and seed morphology are presented in this study.&nbsp

Habitatlar üzerinde devam eden baskı ile artan nüfus, endüstriyel ve kentsel gelişim, artan tarım uygulamaları, istilacı türlerin artan tehditleri ve iklim değişikliğinin etkileri ile birlikte bugün tehlike altında olmayan birçok tür, gelecekte büyük olasılıkla tehdit altına girecektir. Bu nedenle tehdit altında olan ya da olmayan tüm türler için yaşayabilir popülasyonların sürdürülmesini sağlamak amacı ile ortaya koyulan eylemler, türlerin yerinde korunmasını esas almalı ve bu eylem planları gerekli müdahale derecesine bağlı olarak tür yönetimi, eylem, koruma veya kurtarma planları olarak sınıflandırılmalıdır. Bu çalışmada, gelecekte gerçekleştirilmesi önerilen tür yönetimi planlarındaki ilk adım olması amacı ile Dünya Doğa ve Doğal Kaynakları Koruma Birliği (IUCN) Kırmızı Listesi’nde yer alan ve ülkemizde soyu tükenme tehlikesi altında bulunan Fabaceae familyasına ait türlerin (Lathyrus, Astragalus, Vicia, Lotus, Thermopsis) detaylı bir araştırması gerçekleştirilmiştir. Çalışmada, (1) Fabaceae familyasına ait türlerin yayılış gösterdiği biyocoğrafik bölgelere, (2) yetiştiği habitatlara, (3) ekolojik özelliklerine, (5) karşı karşıya oldukları tehditlere, (6) ekonomik önemlerine, (7) etnobotanik özelliklerine ve halk arasındaki kullanım olanaklarına ilişkin detaylara yer verilmiştir. Ayrıca bu çalışma ile, türlerin lokasyonlarına ait güncel kayıtlar da değerlendirilmiştir. Yapılan değerlendirmeler sonucunda familyaya ait türlerin doğal yaşam alanlarına yönelik en önemli tehdit unsurlarının iklim değişikliği, aşırı otlatma, arazi ıslahı, saman yapma ve erozyon, baraj yapımı ve kentleşmenin neden olduğu bozulmalar olduğu görülmüştür. Tarımsal faaliyetler ve insan kaynaklı habitat kaybı ve bozulmaları da önemli tehdit unsurlarındandır. Bu çalışmaya göre ülkemizin önemli genetik zenginliklerinden olan ve IUCN Kırmızı Liste’sindeki Fabaceae familyasına ait türlerin nesillerini devam ettirebilmeleri amacı ile gerekli koruma önlemlerinin alınması ve koruma bilincinin geliştirilmesi gerekmektedir. Ayrıca, bu türlerin kullanım alanlarının belirlenmesi, ekolojik ve ekonomik önemlerinin saptanması son derece önemlidir. Türlerin korunmasına yönelik eylem planlarının hazırlanması ve genetik çeşitliliğin de içinde yer aldığı sürdürülebilir koruma ve kullanım stratejilerinin geliştirilmesi ise bu türlerin tüm yönleriyle araştırılmasına bağlıdır Many species that are not endangered today will likely be threatened in the future because of the growing population, the rising need for industrial and urban development and agricultural practices, the increasing threats of invasive species, and the effects of climate change. Therefore, actions aiming to maintain viable populations for all threatened or non-threatened species should be based on in-situ conservation of the species. These action plans should be classified as species management, action, conservation, or rescue plans depending on the degree of intervention required. For the future species management plans, in this study, the species belonging to the Fabaceae family (Lathyrus, Astragalus, Vicia, Lotus, Thermopsis), which are in the International Union for Conservation of Nature (IUCN) Red List and under threat of extinction in Turkey, has been reviewed based on the details about (1) their biogeographic regions, (2) habitats, (3) ecological characteristics, (4) potential threats, (5) economic importance, (6) ethnobotanical features and (7) usage opportunities among the people. Additionally, this study evaluates up-to-date records of the species' locations. As a result of the evaluations, the most critical threats to the natural habitats of the species belonging to the Fabaceae family are climate change, overgrazing, land reclamation, haymaking and erosion, deterioration caused by dam construction, and urbanization. Agriculture and anthropogenic habitat loss and degradation are also important threats. According to this study, it is vital to take the necessary conservation measures and develop conservation awareness for the future. Additionally, it is essential to determine these species' usage areas and determine their ecological and economic importance. The preparation of action plans for the conservation of species and the development of sustainable conservation and usage strategies, including genetic diversity, depend on the investigation of these species in all aspects

Invasive insect species, brown marmorated stink bug [Halyomorpha halys (Stål, 1855) (Hemiptera: Pentatomidae)], is an indigenous species to China, Japan, Korea and Taiwan, which is one of the most dangerous pest for many agricultural plants. It is polyphagous and cause significant damages to nearly 300 plant species, including ornamental plants, agricultural crops, tree nuts and vegetables In Europe, it was first reported in 2004 and has extended its range to many European countries. H. halys was first reported faunistically from Istanbul, Turkey in 2017 and afterwards from Artvin province in the same year. Turkey is the largest producer and exporter of hazelnut in the world and recent occurrence of H. halys in Turkey reveals a real potential threat to the hazelnut production as well as other crops in the country. In this study, the insect has been shown parts to be considered in comparison with the most similar species, Rhaphigaster nebulosa. Furthermore, other stink bug species found in Turkish fauna was given to avoid confusion and the current status and potential threat of the insect were discussed. As a result, the insect should seriously be considered in Turkey and pest management programs should be initiated to take under control before it spreads more. On the other hand, the studies on the biology of the insect and suitable management strategies including especially classical biological control are urgently needed in the country., İstilacı bir böcek türü olan kahverengi kokarca, [Halyomorpha halys (Stål, 1855) (Hemiptera: Pentatomidae)], birçok kültür bitkisi için en tehlikeli zararlılardan biri olup, Çin, Japonya, Kore ve Tayvan orijinli bir türdür. Bu tür polifag olup, tarımsal ürünler, kabuklu yemişler, sebzeler ve süs bitkileri dahil olmak üzere yaklaşık 300 bitkide önemli zararlara sebep olmaktadır. Avrupa'da, ilk olarak 2004 yılında rapor edilmiş ve pek çok Avrupa ülkesine yayılmıştır. H. halys ilk olarak 2017 yılında İstanbul'dan, daha sonra aynı yıl Artvin ilinden Türkiye'ye girişi bildirilmiştir. Türkiye, dünyanın en büyük fındık üreticisi ve ihracatçısıdır ve Türkiye'de H. halys'in son zamanlarda ortaya çıkması ve yayılmaya devam etmesi, fındık üretimi ve ülkedeki diğer ürünler için gerçek bir potansiyel tehdit oluşturmaktadır. Bu çalışmada, bu zararlıya en benzer tür olan Rhaphigaster nebulosa ile karşılaştırılarak teşhiste dikkat edilmesi gereken kısımlar gösterilmiştir. Ayrıca Türkiye faunasında bulunan ve bu zararlı ile karıştırılabilecek diğer pis kokulu böcek türleri verilmiş ve zararlının mevcut durumu ve potansiyel tehdidi tartışılmıştır. Sonuç olarak, zararlı Türkiye'de ciddi bir şekilde ele alınmalı ve daha fazla yayılmadan mücadele programı başlatılarak kontrol altına alınmaya başlanmalıdır. Ayrıca, böceğin biyolojisi ve özellikle klasik biyolojik mücadeleyi de içeren uygun mücadele stratejileri üzerine çalışmalara acilen ihtiyaç duyulmaktadır.

Glandula uropygialis (uropygial bez), kuşlarda derinin holokrin yapıda bir yağ bezidir. Bu çalışma erkek peçeli baykuşlarda (Tyto alba) uropygial bezin morfolojik ve histolojik yapısını belirlemek amacıyla yapıldı. Çalışmada erişkin yaşta 4 adet erkek peçeli baykuş materyali kullanıldı. Kuşların kuyruk bölgesinde yer alan bezler diseke edilerek vücuttan ayrıldı. Bezlerin morfometrik incelemeleri yapıldı. Bezin histolo- jik yapısını ortaya koyabilmek amacıyla doku kesitleri Hematoksilen & Eozin (H&E) ve Masson Trichrome yöntemleri ile boyandı. Boyanan kesitler ışık mikroskobu ile incelendi. Topografik incelemede, bezin synsakrumun kaudalinde, kuyruk omurlarının üzerinde yerleştiği belirlendi. Bezin ortalama ağırlığı 0.493±0.02 g, relatif indeks değeri (bez ağırlığının vücut ağırlığına oranı) ise 0.10±0.01 olarak bulundu. Histomorfolojik incelemede her bir lobun dışarıdan bağ doku ile sarıldığı görüldü. Loblar sıralı olarak dizilmiş tubulo-alveolar bezlerden oluşmaktaydı. Bezler ince bağ doku ile sınırlandırılarak birbirinden ayrılmışlardı. Bezler arasındaki bağ doku içerisinde kan damarları yer almaktaydı. Bezler kranialden kaudal uca doğru gidildikçe şekil ve hücre dizilişi bakımından farklılık gösteren 3 farklı bölgeden oluşmaktaydı. Sonuç olarak peçeli baykuş uropygial bezinin gerek morfolojik gerek histolojik yapısı bir takım yapısal farklılıklara rağmen literatürlerde bildirilen diğer birçok kanatlı hayvanlardaki ile benzerdi. The glandula uropygialis (uropygial gland) is a holocrine sebaceous gland of the skin in birds. This study was carried out to determine the morphological and histological structure of the uropygial gland in male barn owls (Tyto alba). In this study, four adult male barn owl materials were used. The glands in the tail area of the birds were dissected and separated from the body. Morphometric examinations were made of the glands. Tissue sections were stained with Hematoxylin & Eosin (H&E) and Masson Trichrome methods to reveal the histological structure of the gland. The stained sections were examined with by light microscope. In topographical examination, it was determined that the gland was located in the caudal of the synsacrum, above the tail vertebrae. The average weight of the gland was found 0.493 ± 0.02 g, and the relative index value (ratio of gland weight to body weight) was found 0.10 ± 0.01. In histomorphological examination, it was seen that each lobe was externally surrounded by connective tissue. The lobes consisted of sequentially lined tubulo-alveolar glands. The glands were separated by a thin connective tissue. There were blood vessels in the connective tissue between the glands. The glands consisted of 3 different regions that differ in shape and cell sequence to throught from the cranial to the caudal end. As a result, the morphological and histological structure of the barn owl uropygial gland was similar to many other poultry animals reported in the literature, despite some structural differences.

Bir Lydia kenti olan Thyateira, günümüzde Manisa ili, Akhisar ilçe sınırları içerisinde yer almaktadır. Antik kentte 2011 yılında başlayan yeni dönem kazıları yoğun olarak Hastane Höyüğü alanında devam etmektedir. Söz konusu alanın Hellenistik Dönem’den itibaren Thyateira antik kentinin akropolü olduğu düşünülmektedir. Höyüğün zirve noktası olarak tanımlayabileceğimiz alanda gerçekleştirilen kazı çalışmalarında Hellenistik Dönem’de inşasına başlanıldığı düşünülen Ion düzenli bir tapınak yapısının kalıntıları ortaya çıkarılmaktadır. Kazı çalışmaları günümüzde yapının doğu girişi ve merkezi üzerinde yoğunlaşmaktadır. Tapınağın üzerinde geç dönem kullanımlarının bir hayli yoğun olması tahribatı arttırmıştır. Özellikle tapınağın güney ve kuzey yönünde yer alan nekropol alanlarının kullanımı sırasında yapının mimari parçalarının büyük oranda tahrip edildiği tespit edilmiştir. Tahribat tabakası olarak adlandırdığımız bu seviye içerisinde tapınağın bezemeli kısımlarına ait çok sayıda kırık parça bulunmuştur. Yapının ilk inşasının tespiti açısından mimari bezeme parçaları oldukça önemlidir. Benzer mimari bezemeler yardımıyla tapınağın inşasına MÖ II. yüzyılın ikinci ve üçüncü çeyreği arasında başlandığı anlaşılmaktadır. Thyateira, Hastane Höyüğü alanında açığa çıkarılmaya başlanılan tapınak yapısı, altyapı ve bezeme özellikleri açısından en çok Chryse Apollon Smintheios Tapınağı ile benzerlik göstermektedir., Thyateira, a Lydian city, is located within the borders of Akhisar district of Manisa province today. New excavations contuining since 2011 go on intensively in Hastane Hoyuk. The site in question is estimated to be the acropolis of Thyateira from the Hellenistic period onwards. During the excavations on the area that can be defined as the top of the mound, ruins of an Ionic ordered temple; construction of which is believed to had been started in the Hellenistic era, have been brought to daylight. At present, the excavations are concentrated on the eastern entry and the center of this structure. The late period usages on the temple have been extensive and this amplified its destruction. It has been confirmed that the architectural pieces of the structure had been destroyed to a large extend due to the usages of the areas of the necropolis located on south and north of the temple. At this level which we call as the layer of destruction; a large number of broken pieces belonging to ornamental sections of the temple, had been found. With respect to the determination of the initial construction date; these ornamental archeological pieces are of great significance. In the light of the evidence provided by the similar architectural ornaments; it has been realized that the construction of the temple dates back to a period between second and third quarters of the second century BC. The temple in Thyateira, Hastane Hoyuk, whose structure has been started to be revealed, is at most similarity with the temple of Chryse Apollon Smintheios in terms of its substructure and ornamental features.

Introduction: This study aims to assess the in vitro activity of ceftazidime-avibactam (CZA) and ceftolozane-tazobactam (C/T) against the multi-drug resistant (MDR) Klebsiella pneumoniae. Furthermore, we aimed to determine the types of carbapenemase enzyme responsible for carbapenem resistance and compare the activity of ceftolozane-tazobactam and CZA according to the types of carbapenemase enzymes produced. Materials and Methods: Twenty-two MDR isolates were investigated in the study. The identification and antimicrobial susceptibilities of the isolates were performed by VITEK 2 (BioMérieux, France) automated system. The activity of C/T and CZA was determined by the gradient strip test (Liofilchem MIC strip test, Italy). In all K. pneumoniae isolates, blaIMP-1, blaKPC, blaNDM-1, blaOXA-48 and blaVIM gene regions encoding the carbapenemase enzyme were investigated by using Xpert CARBA-R test kits (Cepheid, Sunnyvale, CA, USA) of the Gene-Xpert® system. Results: Ceftolozane-tazobactam resistance was detected in 21 (95%) isolates, whereas CZA resistance was detected in six (27%) isolates. Among the 22 K. pneumoniae isolates, NDM-1 gene region was detected in three, NDM-1+OXA-48 gene region was detected in two, OXA-48 was detected in eight, and KPC gene region was detected in nine isolates. VIM and IMP-1 gene regions were not detected. Ceftolozane-tazobactam resistance was detected in seven isolates, whereas CZA resistance was not detected among the eight K. pneumoniae isolates producing only OXA-48. NDM-1 and OXA-48 co-producing isolates were detected resistant to C/T and CZA with high minimum inhibitory concentration (MIC) levels (MIC ≥256 µg/ml). All NDM-1 producing isolates had high MIC levels (MIC ≥256 µg/ml) to both C/T and CZA. Ceftazidime-avibactam resistance was detected in only one isolate, whereas C/T resistance was detected in all K. pneumoniae isolates producing KPC. Ceftolozane-tazobactam was detected inefficient whereas CZA was found very efficient in MDR K. pneumoniae isolates producing KPC. Conclusion: According to the obtained data, we detected the in vitro antibacterial activity of CZA against the MDR K. pneumoniae isolates was superior to that of C/T. Ceftolozane-tazobactam was found to be weakly efficient, whereas CZA was found to be highly efficient against the MDR K. pneumoniae isolates producing OXA-48 and KPC. Because avibactam can inhibit the activity of KPC-type carbapenemase, the combination drug CZA should be considered to be effective in the treatment of KPC-type carbapenemase-producing strains but noneffective against NDM-type carbapenemase-producing strains.

Bu araştırmada Yağlı Güreşleri’ne katılan Yağlı güreş sporcularının serbest zaman doyumları ile mutluluk düzeyleri ilişkisinin incelenmesi amaçlanmıştır. Araştırmaya, 2019 yılı “658. Tarihi Kırkpınar Yağlı Güreşleri’nde Türkiye Güreş Federasyonu’nun “baş”, “başaltı”, “büyük orta” ve “küçük orta büyük” yağlı güreşlerinde lisanslı olarak aktif olan 204 yağlı güreş sporcusu gönüllü olarak katılmıştır. Katılımcıların demografik özelliklerini belirlemek amacıyla araştırmacı tarafından hazırlanan kişisel bilgi formu ile birlikte, serbest doyum düzeylerini belirlemede Beard ve Ragheb (1980) tarafından geliştirilen, Gökçe ve Orhan (2011) tarafından Türkçe uyarlama çalışması yapılan “Serbest Zaman Doyum Ölçeği (Leisure Satisfaction Scale)” ve katılımcıların mutluluk düzeylerini belirlemeye yönelik kullanılan diğer ölçek Hills ve Argyle (2002) tarafından geliştirilen, Doğan ve Çötok (2011) tarafından Türkçe uyarlama çalışması yapılan “Oxford Mutluluk Ölçeği Kısa Formu” kullanılmıştır. Elde edilen veriler SPSS 25.0 paket programı ile analiz edilmiş olup istatistiksel açıdan güvenirlik katsayısı hesaplamaları yapılarak veriler arasındaki ilişkiyi belirlemede Pearson Korelasyon analizi ve katılımcılarda serbest zaman doyumunun mutluluk düzeylerinin tespitinde doğrusal regresyon analizi kullanılmıştır. Araştırmanın sonucunda yağlı güreş sporcularının serbest zaman doyum düzeylerindeki artışın mutluluğu seviyelerini de arttıracağını göstermektedir. Bu bağlamda dünyanın en zor sporlarından biri olan yağlı güreş sporunu yapan sporcuların serbest zaman doyumlarını arttırmaya yönelik çeşitli aktivitelere yönelmeleri ve rekreasyonel anlamda yapılacak farklı araştırmalara da yer verilmesi tavsiye edilmektedir., In this study, it is aimed to examine the relationship between leisure time satisfaction and happiness levels of Oil wrestling athletes participating in Oil Wrestling. 206 oil wrestling athletes voluntarily participated in the research from Turkey Wrestling Federation licensed wrestling in different categories in Historic Kırkpınar 658. Oil Wrestling held in 2019. personal information form was prepared by the researcher in order to determine the demographic characteristics of the participants. “Leisure Time Satisfaction Scale” developed by Beard and Ragheb (1980) and adapted into Turkish by Gökçe and Orhan (2011) was used to determine leisure time satisfaction levels and The “Oxford Happiness Scale Short Form” developed by Hills and Argyle (2002) and adapted to Turkish by Doğan and Çötok (2011) was used to determine the happiness levels of the participants. The obtained data were analyzed with the SPSS 25.0 package program, and Pearson Correlation analysis was used to determine the relationship between the data by making statistical reliability coefficient calculations and linear regression analysis was used to determine the happiness levels of leisure, the increase in the leisure time satisfaction levels of oil wrestling athletes will also increase their happiness level. Thus, it is recommended that athletes engaged in oil wrestling, which is one of the most difficult sports in the world, should participate to various activities aimed at increasing their leisure time satisfaction and to include different recreational researches.

Introduction: Infections with extended-spectrum beta-lactamase (ESBL)-producing strains are seen at increasing rates, resulting in increased morbidity, mortality, length of hospitalization, and cost. Effective and safe treatment of these infections is important. The aim of this study was to compare the effectiveness of piperacillin-tazobactam (PTZ) and carbapenems in nosocomial infections with ESBL-producing Escherichia coli and Klebsiella pneumoniae in hospitalized patients. Materials and Methods: This study was performed retrospectively on patients who were infected with ESBL-producing E. coli and K. pneumoniae while hospitalized in Ankara Atatürk Tranining and Research Hospital between 2014 and 2017. Both PTZ- and carbapenem-susceptible isolates were included in the study. Patients with signs of infection who were older than 18 years, hospitalized for at least 48 hours, and received PTZ or carbapenem therapy were included in the study. Patients who received PTZ and carbapenem antibiotics consecutively in same infectious episode, received another antibiotic active against ESBL-producing microorganisms, received the selected antibiotic for less than 48 hours, or had polymicrobial infection were excluded from the study. Demographic characteristics, comorbid diseases, antibiotics used and their dose/duration, mechanical ventilation, presence of central catheter, clinical and microbiological responses to treatment, and end-of-treatment and 30-day mortality were recorded and compared. Results: Of the total 113 patients, 60 (53.1%) were male and the mean age was 66.14±18.2 years. Of the patients, 73.5% had E. coli and 26.5% had K. pneumoniae. Seventy patients (61.9%) received carbapenem and 43 patients (38.1%) received PTZ treatment. Microbiological eradication rate was 71.7% (43/60) in patients with control cultures. Clinical response was observed in 72.6% of the cases. The overall mortality rate was 31.0% (n=35). There were no significant differences between the groups in clinical and microbiological response (p=0.055; p=0.303) or end-of-treatment and 30-day mortality (p=0.180, p=0.288). Age and bacteremia was found to be independent risk factors for mortality (p=0.006 for both). Conclusion: PTZ is an important alternative for infections caused by ESBL-producing microorganisms. In recent years, the need for rational antibiotic use has increased and carbapenem-sparing treatments have became more important. Piperacillin-tazobactam is one of the best alternatives and has comparable effectiveness in infections caused by ESBL-producing E. coli and Klebsiella spp.

In this study, anatomy and histology of the male reproductive system in adults of Capnodis tenebrionis, which are called the Mediterranean flat-headed root-borer, are described and illustrated by using light and scanning electron microscopies. Data from the male gonad of this species provide more extended and precise knowledge regarding to histoanatomical structure of the testis in Buprestidae. In this study, male reproductive system of C. tenebrionis consisted of two testes, two vas deferens and two accessory glands which are long tube-like, a pair of spermatophoral glands, an ejaculatory duct, and an aedeagus. Each testis was subdivided into a lot of tubular testicular follicles. Vas deferens was a tubular structure and located just below the testis. Its wall was surrounded by a monolayer cylindrical epithelium and a thin muscle layer. In its lumen, it was observed that mature sperm stacks were stored. Vas deferentia were immediately opened ejaculatory duct. It was surrounded by a single layered epithelium and a very thick muscle layer. Long, tubular, convoluted a pair of accessory glands were seen which were connected to the ejaculatory duct. In the accessory glands, it was seen that the wall was surrounded by monolayer cubic epithelium and sheath. In addition, it was seen that oval, sheet-shaped, paired spermatophoral glands were opened into the ejaculator canal. Its histological examinations revealed that the wall was surrounded by multilayered epithelial cells and muscle layer. The ejaculatory canal was connected to the aedeagus with very dense striated muscle bundles. The aim of the present study is to contribute information on the histomorphology of the male reproductive system of the insects.

Amaç: Pelargonium endlicherianum uçucu yağının enfeksiyon tedavilerinde sıklıkla kullanılan benzilpenisilin ile kombine edilerek in vitro deney modeli içerisinde Streptoccus pneumoniae’ye karşı antimikrobiyal etkisinin ortaya çıkarılması ve insan lökosit hücreleri üzerindeki fonksiyonun tespit edilmesi amaçlandı. Yöntem: P. endlicherianum uçucu yağının benzilpenisilin ile kombine kullanımının Streptococcus pneumoniae’ye karşı antimikrobiyal etkisi agar difüzyon tekniği kullanılarak ve lökosit hücreleri üzerindeki fagositik fonksiyonları in vitro hücre deney modeli içinde insan lökosit hücresi WBC 264-9C ATCC HB-8902 kullanılarak ortaya çıkarılmıştır. P. endlicherianum uçucu yağ ve benzilpenisilinin bakterisidal etkisi zamana bağlı öldürme yöntemi ile zamana ve antibiyotik yoğunluğuna bağlı olarak dinamik olarak ortaya konuldu. Uçucu yağın tek başına ve antibiyotik ile kombine kullanıldığında bakteriyel dış membrandaki geçirgenliği U.V. spektrofotometre ve morfolojik görüntüsü taramalı elektron mikroskobu ile analiz edildi. Bulgular: Tedavi sonrası 24. saatte, ilaç kombinasyonu sadece uçucu yağ tedavisi ile karşılaştırıldığında, yaşayabilir hücre sayısında azalma olduğu gözlendi. Uçucu yağ+antibiyotik kombinasyonlarında uçucu yağ ve antibiyotik arasında sinerjizm gözlenmiş olup, lökosit hücrelerinin fagositik aktivitesi artarak canlı bakteri sayısında belirgin bir düşüş belirlendi. Sonuç: Çalışmanın sonuçlarına göre S. pneumoniae’ye karşı kullanılan benzilpenisilinin P. endlicherianum uçucu yağı ile kombine kullanımı sonucu antibiyotik etkinliğinin arttığı ve böylece bakterilerin antibiyotiklere karşı direnç geliştirmesi engellenerek etkin bir tedavi sağlanabileceği gösterilmiştir.

Bu çalışmada, Artvin ili Merkez İlçesi Genya Dağı Ceravent mevkiinde 1500 m rakımda, normal kapalılıktaki saf Doğu Ladini meşçerelerinde toprak üstü biyokütle miktarı belirlenmiştir. Tespit edilen normal kapalılıktaki saf Doğu Ladini meşçeresinde, her çap sınıfını temsil edecek şekilde farklı çaplarda, canlı, tepesi sağlam, tek gövdeli, sağlıklı ve dal budanmasını tamamlamış 30 adet deneme ağacı tespit edilmiştir. Deneme alanında tespit edilen bireylerin göğüs çaplarını belirlendikten sonra topraktan 20 cm yukarıdan kesilmişlerdir. Bütün bireylerin boyu ölçülerek, ibreli ve ibresiz dallar gövdeden ayrılmıştır. Gövde dipten tepeye doğru, 5 m lik kesitlere ayrılmış ve yaş ağırlıkları belirlenmiştir. Daha sonra her bir gövde kesitinin kalın uçundan 5 cm kalınlığında örnek diskler alınmıştır. Dal ve ibrelerin yaş ağırlığı birlikte belirlenmiş, her bir birey için tartılan ibreli dallardan deneme ağacını temsil edecek şekilde alt, orta ve üst bölümlerden yeteri kadar örnek alınmıştır. Her bir gövde kesitinden alınan örnek disklerle ibreli dal örnekleri polietilen torbalara konularak laboratuvara getirilmiştir. Kesitlerin 70 oC lik fırındaki nem kayıp oranlarından yararlanılarak her bir ağacın kuru ağırlığı belirlenmiştir. Elde edilen verilere göre, incelenen tek ağaç bileşenleri için çap, toprak üstü biyokütledeki varyasyonu, boya göre daha iyi tanımlamaktadır. Çapa dayalı denklemlerin R2 değerleri (en yüksek % 95) boya dayalı denklemlerden (en yüksek % 85) daha büyüktür.

Bu çalışma, kekik (Origanum onites L.) ve biberiye (Rosmarinus officinalis L.) bitkilerinden elde edilen uçucu yağların Fusarium solani‘ye karşı antifungal aktivitesini tespit etmek amacıyla laboratuvar ortamında yürütülmüştür. Bitkilerden elde edilen uçucu yağlar 1, 2 ve 4 μl dozunda otoklav edilen Patates Dekstroz Agar (PDA) besi yerine ilave edilmiştir. Patojenlere ait 8 mm çapında miselyum diskleri PDA besi yerlerine ekilmiştir. Kontrol grubu olarak uçucu yağlardan ari PDA besi yeri kullanılmıştır. PDA’lı petriler 24±1oC’da 7 gün inkübasyona bırakılmıştır. İnkübasyon sonrasında fungusların koloni çapları ölçülerek, kontrol grubuna göre bitki uçucu yağlarının % engelleme oranları hesaplanmıştır. Çalışmadan elde edilen verilere göre, Rosmarinus officinalis L. uçucu yağının yüksek dozu (4 μl) test edilen Fusarium solani’ye karşı %71,72 etki göstermiş ve doz seviyesi arttıkça antifungal etkinin de arttığı belirlenmiştir. Origanum onites L. uçucu yağı, uygulanan üç dozda Fusarium solani’nin misel gelişiminde %78,91-%82,70 oranları arasında engelleme göstermiştir. Çalışmada kekik ve biberiye uçucu yağları kontrol grubu ile karşılaştırıldığında Fusarium solani’ye karşı farklı seviyelerde engelleyici etkiler gösterdikleri tespit edilmiş ve patojenlerin koloni gelişimine en yüksek antifungal etki (%82,70) kekik uçucu yağının 2 μl ve 4 μl doz uygulamalarında saptanmıştır. Bu sonuçlar bitkilerden elde edilen uçucu yağların sentetik pestisitlere alternatif olabileceğini göstermiştir.

Karolenj reformlarının odağı devlet yönetiminde standarda ulaşabilmekti. Manevi çöküntü içindeki ruhban sınıfını bilge keşişler; prensleri ise donanımlı devlet adamları olarak yetiştirebilecek entellektüel eğitimi sağlamak ise Karolenj Rönesansı’nın sosyal alana yönelik düzenlemelerinin saç ayaklarından birini teşkil ediyordu. Dini eğitimin merkezi manastırların en önemli yan ürünü ise yazıhanelerde istinsah edilen klasik dönem tezhip eserleriydi. Bu bilgilerden hareketle, çalışma 16. yüzyılda Avrupa’yı etkisi altına alan büyük İtalyan Rönesansı’ndan önce ortaya çıkan ve muhtemelen öncülü olan bir dönem olan Karolenj dönemin entellektüel boyutunun Kilise olan bağlantısını ortaya koymaya çalışmakta, aynı zamanda Klasik dönem el yazmalarının Karolenj manastır ve saray kütüphanelerindeki kitap istinsahını ve bu el yazmalarının Karolenj Rönesansı açısından öneminin altını çizmektedir., The focus of the Carolingian reform was to achieve a standard in state administration and provide an intellectual education that could train the clergy as wise monks and princes as well-equipped statesmen. That would constitute the so-called Carolingian reform’s other two pillars: intellect and religion, the first of which were the military and the politics. Those who pray on behalf of the people were the clergy, and how they did it was important. With Charlemagne becoming the Holy Roman emperor, the church-state relationship became more unitary than ever in the Frankish Empire. The most important essence of the monasteries as the center of religious education was the production of illuminated manuscripts of the classical period copied in monastic scriptoria. Based on this information, the study attempts to reveal the connection between the Church and the intellectual dimension of the Carolingian period, a period that emerged before the great Italian Renaissance in the 16th century and probably preceded it. Besides, the study also underlines the copying of Classical period manuscripts in the libraries of Carolingian monasteries, palaces, and the importance of these manuscripts for the Carolingian Renaissance.

ÖZET Helicobacter pylori (H. pylori), insanlarda, gastrit, peptik ülser, gastrik kanser ve mukoza ilintili lenfoid doku lenfoması gibi ciddi mide hastalıklarına yol açan önemli bir hastalık etkenidir. H. pylori, ürettiği üreaz enzimleri sayesinde üreyi, karbondioksit ve amonyağa parçalayarak mide çeperinin asidik koşullarını normal pH’ya getirip hayatta kalabildiği için bu bakterilerle vücudun savaşımı kolay değildir. H. pylori enfeksiyonları için antibiyotikler mevcut olsa da antibiyotik direnci gelişimi nedeniyle bu tedaviler sonuçsuz kalabilmekte ve yeni antibiyotiklere gereksinim her geçen gün artmaktadır. Hidroksisinamik asit türevleri basit fenolik asitler olup meyvelerde, meyve çekirdeklerinde ve sebzelerde bulunmaktadır. Ferulik asit, kafeik asit, p-kumarik asit, sinapik asit, sözü edilen bu fenolik asit grubuna ait olup antioksidan, anti-inflamatuvar, antimikrobiyal özelliklere sahiptir ve bu nedenle bazı bakteri enfeksiyonlarının tedavisinde, ilaçlara seçenek olarak kullanılmaktadır. Bu çalışmamızda, bu üç fenolik asidin H. pylori üzerindeki antimikrobiyal etkinliği ve ürez enzimini engelleme etkisi araştırıldı. Hidroksisinamik asit türevlerinin anti-H. pylori etkisi H. pylori G27 standart suşu üzerinde test edildi. Minimum inhibisyon konsantrasyonu (MİK), değerleri 512 ila 0,5 ug/mL arasında değiştiği seri tüp seyreltme yöntemiyle, minimum bakterisidal konsantrasyon (MBK) değerleri ise MİK içinde kullanılan aynı konsantrasyonlarda canlı ve ölü bakterilerin nispi oranının hesaplanması ile belirlendi. MİK için CLSI M07-A9, MBC için CLSI M26-A protokolleri kullanıldı. Ureaz inhibisyon aktivitesi Helicheck, üreaz aktivitesine özgü indikatörlü besiyerinde ölçülmüştür. H. pylori’ye karşı test edilen tüm bileşikler için MİK 64 ug/mL ve MBK 128 ug/mL idi. Test edilen bileşiklerin H. pylori tarafından salgılanan üreaz enzimi üzerinde hiçbir inhibisyonu saptanmadı. Nükleotid salma deneyi sonuçlarına göre, hidroksisinamik asit türevlerinin bakteri zarında hasara sebep olması ile zarda oluşan deliklerden dışarı salınma eğiliminde olması beklenen herhangi bir nüklotit miktarı ölçülememiştir. Gerçekleştirilen bu çalışma, literatür taramalarımız doğrulutusunda, hidroksisinamik asit türevlerinin anti-H. pylori aktivitesini gösteren ilk çalışmadır. Bu bileşiklerin anti-H. pylori üzerindeki etki mekanizmasını anlamak için daha ileri analizlere ihtiyaç vardır.

Böcek büyüme düzenleyicileri ve mikrobiyal insektisitler zararlılarla mücadelede tarımsal alanlarda sıklıkla kullanılmaktadır. Bu nedenle, her iki insektisit türüne karşı zararlılarda direnç gelişimi meydana gelmektedir. Son yıllarda, direnç gelişimine karşı farklı etki mekanizmalı insektisitlerin karışım halinde uygulanması sıklıkla tercih edilen bir yöntem olarak kabul edilmektedir. İmmun sistem parametreleri, insektisitlerin zararlı böcek türleriyle mücadelede toksik etkilerinin belirlenmesinde biyobelirteç olarak kullanılmaktadır. Bu amaç kapsamında yapılan çalışmada juvenil hormon analoğu pyriproxyfen ve mikrobiyal insektisit B. thuringiensis’ in 50 ve 100 µg/mL derişimlerinin tek başına ve karışım halindeki (25+25 ve 50+50 µg/mL) uygulamalarının Galleria mellonella larvalarında total hemosit sayısı ve diferansiyel hemosit sayısı üzerine etkileri araştırılmıştır. Elde edilen bulgulara göre, pyriproxyfenin, G. mellonella larvalarında bağışıklık sistemini baskılayarak, B. thuringiensis biyopreparatının toksik etkisini arttırdığı belirlenmiştir., Insect growth regulators and microbial insecticides are frequently used in agricultural fields for pest management. Therefore, resistance development occurs in pests against both insecticide types. In recent years, the use of insecticides with different mode of action in mixture against resistance development has been accepted as a frequently preferred method. Immune system parameters are used as biomarkers to determine the toxic effects of insecticides in pest management. In this study, the effects of 50 and 100 µg/mL concentrations of the juvenile hormone analogue pyriproxyfen and the microbial insecticide Bacillus thuringiensis alone and in mixture (25+25 and 50+50 µg/mL) on total hemocyte count and differential hemocyte count in Galleria mellonella larvae were investigated. According to the findings, it was determined that pyriproxyfen increased the toxic effect of B. thuringiensis biopreparate by suppressing the immune system in G. mellonella larvae.
