Gümüşün antimikrobiyal etkisini ve titanyum nitrürün mekanik ve kimyasal özelliklerini birleştirmek için, ortopedik implant malzemesi olarak kullanılan Ti6Al4V alaşımının yüzeyine N2 plazmasında plazma daldırma iyon implantasyonu ve biriktirme (PIII&D) yöntemi ile gümüş biriktirilmiştir. Kaplamalar, PIII&D prosesinin farklı negatif darbeli yüksek gerilim ve magnetron sıçratma gerilimi değerleri kullanılarak elde edilmiştir. Kaplamaların yüzey morfolojileri atomik kuvvet mikroskobu (AFM) ve kesit morfolojileri ise alan emisyonlu taramalı elektron mikroskobu (FE-SEM) kullanılarak tespit edilmiştir. Kaplamaların fazları ve kimyasal kompozisyonları sırasıyla X-ışını difraksiyonu (XRD) ve X-ışını fotoelektron spektroskopisi (XPS) kullanılarak tespit edilmiştir. Buna ek olarak, numunelerin yüzeyinden içeri Ag+ iyonlarının nüfuziyetini belirlemek için XPS derinlik profili (depth profile) analizi yapılmıştır. In vitro anti-bakteriyel etkinlik deneylerinde numunelerin yüzeylerine 99mTc radyoizotopu ile işaretlenmiş E. coli (Escherichia coli) mikroorganizmasının adezyonu araştırılmıştır. Çalışmada, 8 kV'luk negatif darbeli gerilimin kullanıldığı deneylerle elde edilen kaplamalarda gümüşün dominant olduğu; bunun yanında, negatif darbeli yüksek gerilimin artmasıyla yüzeyde gümüş miktarının azaldığı gözlemlenmiştir., In order to combine the antimicrobial effect of silver with mechanical and chemical properties of titaniumnitrate, Ag was deposited on the surface of Ti6Al4V (orthopedic implant material) in N2 plasma by using plasma immersion ion implantation and deposition (PIII&D) method. The coatings were formed by usingvaried negative pulsed high voltage and magnetron sputtering voltage values of PIII&D process. Surfacemorphologies of coatings were determined with atomic force microscopy (AFM), while cross-section morphologies were determined with field emission scanning electron microscopy (FE-SEM). The phases and chemical compositions of coatings were investigated by using X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS), respectively. XPS depth profile analysis were done to reveal thepenetration of Ag+ ions into the surface of specimen. In in vitro anti-bacterial effectiveness experiments,the adhesion of E. coli (Escherichia coli) microorganism, which were marked with 99mTc radioisotope, on surfaces of specimens was investigated. It was consequently determined that Ag was dominant in thecoatings obtained by applying 8 kV negative pulsed high voltage, while Ag decreased on the surfacedepending on the increase in the voltage.

Bu çalışma normal ve hiperplastik endometriumda damar yüzey .dansitesini ölçmek ve hiperplastik endometriumda bu parametrenin sitolojik atipi ile ilişkisini değerlendirmek için planlandı. Histopatolojik tanıları sekretuvar endometrium, proliferativ endometrium, basit atipisiz .hiperplazi, basit atipili hiperplazi, kompleks atipisiz hiperplazi ve kompleks atipili hiperplazi olan 64 premenopozal kadından alınan endometrial örnekler çalışmaya alındı. Kesitler immünperoksidaz yöntemi ile faktor-8 ile boyandı ve damar yüzey dansitesini hesaplamak için stereolojik yöntemle damarlar sayıldı. Kompleks hiperplazi, basit ve kompleks atipili hiperplazilerin stroması anlamlı düzeyde proliferativ endometriumdan daha fazla damarlıydı (p=0.03, p=0.004 and p=0.01 sırasıyla). Bununla birlikte, damar yüzey dansitesi yönünden kompleks atipik ve basit atipisiz hiperplazi (p=0.004), basit atipili ve basit atipisiz hiperplazi (p=0.04) ve kompleks atipili ve kompleks atipisiz hiperplazi arasında anlamlı ilişki mevcuttu (p=0.035). Atipili basit ve kompleks hiperplaziler arasında damar yüzey dansitesi yönünden anlamlı fark yoktu. Kompleks hiperplazide damar yüzey dansitesi hafice artmış olmasına karşın atipisiz kompleks ve basit hiperplaziler arasında anlamlı ilişki saptanmadı. Bulgularımız basit ve kompleks hiperplazide sitolojik atipi ile anjiogenezis arasında yakın ilişkinin olduğunu göstermektedir. Sitolojik atipi ve yoğun damarlanması olan lezyona sahip kadınlarda lezyonlar tekrarlama ve malignite riski taşıma eğiliminde olabilir., OBJECTIVE: This study was designed to quantify the vascular surface density of normal and hyperplastic endometria and to evaluate the relation of this parameter with cytologic atypia in the hyperplastic endometrium. STUDY DESIGN: Endometrial samples from 64 premenopausal women with the histopathologic diagnosis of proliferative endometrium, secretory endometrium, simple endometrial hyperplasia without cellular atypia, complex endometrial hyperplasia without cellular atypia, simple endometrial hyperplasia with cellular atypia and complex endometrial hyperplasia with cellular atypia were included in the study. The sections were stained with Factor 8' related antigen with standard streptavidin biotin immunoperoxidase method and the microvessels were quantified by a stereological method described previously, for the assessment of vascular surface density. RESULTS: The stroma of complex hyperplasia, simple and complex hyperplasia with atypia were significantly more vascular than that of proliferative endometrium (p=0.03, p=0.004 and p=0.01 respectively). On the other hand, statistically significant difference was found between vascular surface density values of the complex hyperplasia with atypia and simple hyperplasia (p=0.004), simple hyperplasia with atypia and simple hyperplasia (p=0.04) as well as between complex hyperplasia with atypia and complex hyperplasia without atypia (p=0.035). There was no difference between the vascular surface density of simple hyperplasia with atypia and complex hyperplasia with atypia. Vascular surface density of simple hyperplasia and complex hyperplasia showed no statistically significant difference although we found slight increase in the vascular surface density of complex hyperplasia. CONCLUSION: Our results suggest that there is a close relationship between cytologic atypia and angiogenesis in both simple and complex hyperplastic endometrium. Women with cytologically atypical and more vascularized lesions may tend to have persistence and carry the risk for malignancy.
