Bu çalışmada genisteinin TM3 Leydig hücreleri üzerindeki sitotoksititesinin ve bir antioksidan olan Vitamin E'nin bu hasardan hücreyi koruyucu etkisinin toplam hücre sayısı, hücre canlılığı, hücrelerdeki apoptotik indeks, lipid peroksidasyonu ve proteinlerdeki oksidatif hasar belirlenerek araştırılması amaçlanmıştır. Özellikle fast-food yiyecekler ve diğer işlenmiş gıdalarda düşük maliyet nedeniyle et ürünleri yerine fitoöstrojenleri içeren soya ürünlerinin kullanılması yaygındır. Fitoöstrojenler endojen östrojene benzer aktiviteler gösterebilen bitkisel kaynaklı kimyasallardır ve son yıllarda yapılan bazı epidemiyolojik çalışmalarla önem kazanmıştır. Endojen östrojen varlığında dışarıdan ilaç olarak ya da diyetle alınan fitoöstrojenler doza bağlı olarak infertiliteye neden olabilmektedir. Genistein soya ve soya ürünlerinde bulunan steroid olmayan bir fitoöstrojendir ve özellikle östrojen reseptör ß (ER-ß)'ya bağlanarak hem östrojenik hem de anti-östrojenik etki gösterebilir. Yapılan çalışmalarda, genisteinin timosit, lösemi ve testis hücrelerinde apoptozu teşvik ettiği ve oksidatif hasara yol açtığından toksik etkisi olduğu gösterilmiştir. Ayrıca, vitamin E'nin önemli bir antioksidan olması ve hücreyi oksidatif hasardan koruması nedeniyle, insan ve hayvanların beslenmesinde ek besin olarak önemi bilinmektedir. Yaptığımız çalışmada vitamin E'nin genisteinle beraber bir antioksidan olarak etkisi gösterilmiştir.Çalışmamızda, genisteinin tek başına ve vitamin E ile birlikte TM3 Leydig hücreleri üzerine olan etkisi araştırılarak, yüksek dozlarda apoptoza ve lipid peroksidasyonuna neden olduğu; vitamin E ile birlikte uygulamalar sonucunda ise vitamin E'nin bu toksisiteyi azalttığı sonucuna varılmıştır. Bu çalışmanın genisteinin Leydig hücrelerindeki çok fonksiyonlu etkisinin moleküler düzeyde yapılacak çalışmalara temel olabileceği düşünülmektedir. In this study, Genistein-induced cytotoxicity and the protective effect of Vitamine E which is an antioxidant supplementation from this damage on TM3 Leydig cells were purposed to research by determination of total cell number, cell viability, proportion of apoptosis, oxidative damage of protein and TBA reactive species. Soya and soybean product is used instead of meat products because of low cost price, especially in fast-food and other processed foods. In the past few years, phytoestrgens are being important with several epidemiological studies. In presence of endogen estrogen, phytoestrogens that take as a medicine or diet may cause infertility depend on dosage. Genistein especially binds to estrogen receptor ß (ER-ß), acting as estrogen agonist or antagonist Phytoestrogens which act as an endogen estrogen are phytochemical. Genistein has also been demonstrated to induce apoptosis in cells as thymocytes, leukemia cells and testis cells and it is toxic and causes oxidative damage.Antioxidant are spent up enough portion for being obviate oxidative stress which create free radicals and being decrease their effects to minimum. Vitamine E a dietary factor is essential for reproduction in humans and animals. It is an antioxidant in all mammalian cells. Vitamine E is a chain breaking antioxidant that prevents the propagation of free radical reaction and thus protects cells from oxidative damage.In our study, by the research of the effect of the Genistein and Genistein with Vitamine E on TM3 Leydig cells, it were come to the conclusion that genistein in higher doses cause apoptosis and lipid peroxidation; by reason of with Vitamine E application Vitamine E decrease this toxicity. This study is thought to be a base for molecular studies which are corcern with the multifunctional effect of genistein on Leydig cells. 82
