IV ÖZET Mttftüoğlu, M., Cockayne sendromu B grubu geni helikaz domainlerindeki bölge-yönelimli mutasyonların sonuçları, Hacettepe Üniversitesi Sağlık Bilimleri Enstitüsü Biyokimya Programı Doktora Tezi, Ankara, 2001. Cockayne sendromu çeşitli nörolojik ve gelişim anamolileri ile karakterize kalıtsal bir hastalıktır. Cockayne sendromunun CS-A and CS-B olmak üzere iki genetik komplementasyon grubu tanımlanmıştır. CSB proteini helikaz protein ailesinin 2. grubuna ve spesifik olarak da SWI/SNF protein ailesine aittir. CSB poteininin transkripsiyona kenetlenmiş tamirde, transkripsiyonda, kromozomun yeniden yapılanmasında ve apoptozda rolü olduğu bildirilmiştir. Fakat CSB proteininin spesifik motiflerinin bu biyolojik yollardaki rolü henüz kesinlik kazanmamıştır. Bu çalışmada, CSB proteininin UV ile muameleden sonra canlılık, RNA sentezi ve yeniden başlaması ve apoptozdaki genetik fonksiyonunu araştırmak üzere, proteinin la, III, V ve Vl'ncı helikaz motiflerinin ve ikinci putatif nükleotit bağlanma motifinin korunmuş rezidularında bölge yönelimli mutagenez ile tek amino asit değişiklikleri oluşturuldu. UV ışığının indüklediği lezyonlar (siklobütan primidin dimerleri) transkripsiyona-kenetlenmiş tamir mekanizması ile tamir edilirken kimyasal karsinojen 4-Nitrokinolin 1 -oksit (4-NQO)'in indüklediği lezyonlar (büyük adüktler) global genom tamiri ile tamir edilmektedir. Bu nedenle, CSB mutant hücre hatlarının canlılığı 4-NQO ile muameleden sonra da analiz edilmiştir. Bu çalışma sonucunda, helikaz motiflerinin (motif la, III, V and VI) korunmuş rezidularında oluşturulan nokta mutasyonlan, UV ile muameleden sonra, CSB proteininin canlılıkta, RNA sentezi ve yeniden başlamasında ve apoptozdaki genetik fonksiyonunu engellediği gösterilmiştir. Benzer bir şekilde, aynı mutantlann 4- NQO'ya da duyarlılığı belirlenmiştir. Sonuç olarak, bu helikaz motiflerinin bütünlüğü CSB proteininin biyolojik fonksiyonu için önem taşımaktadır. Öte yandan CSB proteinin ikinci putatif nükleotit bağlanma motifi proteininin tamir fonksiyonunda rol oynamamaktadır. Cockayne syndrome (CS) is a human genetic disorder characterized by several neurological and developmental abnormalities. Two genetic complementation groups, CS-A and CS-B, have been identified. The CSB protein belongs to helicase superfamily 2, and more specifically, the SWI/SNF family of proteins. The CSB protein is implicated in transcription-coupled repair (TCR), transcription, chromosome remodeling, and apoptosis. However, the role of specific domains of the CSB protein in these biological pathways has remained unclear. In this study, single amino acid changes in highly conserved residues of helicase motifs la, III, V, VI and a second putative nucleotide-binding motif of the CSB protein were generated by site-directed mutagenesis to analyze the genetic function of the CSB protein in survival, RNA synthesis recovery and apoptosis after UV treatment. The lesions induced by UV light, cyclobutane pyrimidine dimers, are known to be repaired by transcription coupled repair whereas the lesions (bulky adducts) induced by the chemical carcinogen 4-nitroquinoline-l -oxide (4-NQO) are repaired by global genome repair. Therefore, the survival analysis of these CS-B mutant cell lines was also performed after treatment with 4-NQO. The results of this study demonstrated that the point mutations in highly conserved residues of helicase motifs la, III, V and VI abolished the genetic function of the CSB protein in survival, RNA synthesis recovery and apoptosis after UV treatment. Similarly, the same mutants failed to complement the sensitivity toward 4-NQO. Thus, the integrity of these helicase motifs is important for the biological function of the CSB protein. On the contrary, the second putative nucleotide-binding motif of CSB protein does not play a role in the repair function.
