Desde mediados del siglo XX numerosas especies, muy diferentes entre sí y ubicadas en todas las áreas y rincones del planeta, comenzaron a presentar diversas alteraciones, muchas de las cuales sugerían estar relacionadas con trastornos del sistema endocrino. Las investigaciones demostraron que tales alteraciones eran producidas por la exposición a diferentes sustancias químicas contaminantes, las cuales podían alterar la salud y producir graves enfermedades. Dentro de ellas destacó un grupo heterogéneo de compuestos con estructuras químicas muy diferentes, capaces de actuar a dosis muy bajas, mostrar distintos mecanismos de acción y ser capaces de alterar el equilibrio hormonal, por lo que se les denominó “disruptores endocrinos químicos”. Estas sustancias, al ser liberadas al medioambiente o formar parte de objetos, alimentos o medicinas, constituyen un gran riesgo para los seres humanos y toda la vida del planeta, produciendo no solo disfunciones endocrinas sino también diferentes tipos de cáncer, destacando los más frecuentes. A pesar de la trascendencia y significado del impacto de estos compuestos, ellos no son suficientemente conocidos ni entendidos, por lo que el objetivo de esta revisión es mostrar su origen e impacto en la salud humana, resaltando su papel como inductores de cáncer, lo cual ha motivado múltiples investigaciones clínicas y biológicas. Since the mid-twentieth century, many species, very different from each other and located in all areas and corners of the planet, began presenting various alterations, many of which suggested to be related to endocrine disorders. Research has shown that such alterations were caused by exposure to various chemical contaminants that could affect the health and cause serious illnesses. Among them stands a diverse and large group of compounds, with very different chemical structures, capable of altering the hormonal balance, act at very low doses and with different mechanisms of action, that are called “endocrine disrupting chemicals”. When released into the environment or as part of objects, food or medicines, constitute a major risk to animals and humans, which produces not only endocrine dysfunctions but also different cancers, which include the most common types. Despite the importance and significance of the impact of these compounds, they are not sufficiently known or understood, so the aim of this review is to show their origin and impact in the field of human health, highlighting their role as inducers of cancer, which has led to multiple clinical and biological investigations.

Metallothioneins (MTs) are very particular proteins with a molar mass between 6-7 kDa, rich in cysteine (30%) and absence of aromatic amino acids. They undertake the maintenance of homeostasis of Cu and Zn and the detoxification of non-bioessential metals as Cd and Hg, and they are scavengers of free radicals, mainly of free hydroxyl and superoxides. These proteins are involved in a large number of physiological processes that have been studied in detail in vertebrate organisms. However, in invertebrates the study has not advanced at the same rate. Studies of MTs in bivalve mollusks have increased as a physiological response to heavy metal exposure. These invertebrates have been used widely as biomarkers of heavy metals, which supported their use as a tool to determine contaminated environments. More recently, isoforms of the MTs on at different stages of the life cycle. Additionally environmental and xenobiotic factors modulate the expression and function of these proteins. The studies have demonstrated that there is a great polymorphism of MTs in bivalves, Some MTs can be induced by heavy metals, other are only associated with physiological processes. This revision deals about the characteristics and function of MTs, starting from the wide available knowledge in vertebrates and less knowledge in invertebrates. Las metalotioneínas (MTs) son proteínas muy particulares con masa molar entre 6-7 kDa, alto contenido de cisteínas (30%) y ausencia de aminoácidos aromáticos. Se encargan de mantener la homeostasis del Zn y Cu e intervienen en la desintoxicación de metales no esenciales, como Cd y Hg. Son barredores de radicales libres, principalmente de hidroxilos libres y superóxidos. Estas proteínas participan en una gran cantidad de procesos fisiológicos que han sido estudiados detalladamente en organismos vertebrados. Sin embargo, en invertebrados el estudio no ha avanzado a la misma velocidad. En los moluscos bivalvos, los estudios de estas proteínas surgen como una respuesta fisiológica ante la exposición a metales pesados, por lo que han sido utilizados ampliamente como biomarcadores de metales pesados, lo cual sustentó su validación como herramienta para determinar ambientes contaminados. Más recientemente, se han aislado y caracterizado isoformas de las MTs en moluscos bivalvos. En algunas especies se ha determinado la función de cada una de ellas en diferentes etapas del ciclo de vida y se han establecido asociaciones entre la inducción de MTs y factores ambientales. Los estudios han demostrado que en bivalvos existe gran polimorfismo de las MTs y mientras algunas pueden ser inducidas por metales pesados, otras sólo se asocian a procesos fisiológicos. Esta revisión trata las características y funciones de las MTs partiendo del amplio conocimiento que se tiene en vertebrados y los alcances en los estudios realizados en bivalvos. Los lubricantes usados de motores de automóviles son fuente potencial de contaminantes que pueden afectar adversamente a la biota en áreas costeras. Los bivalvos que habitan asociados a arrecifes coralinos han sido reconocidos como organismos sensores para evaluar el impacto de sustancias químicas estresantes mediante el uso de biomarcadores, en especial cambios en su composición bioquímica. Con el objetivo de determinar los niveles de los sustratos energéticos (proteínas, lípidos y carbohidratos) e índices moleculares de condición (ARN/ADN y Proteína/ADN) en el manto de la almeja flama Lima (Ctenoides) scabra expuesta a una fracción acuosa de lubricantes usados de motores de vehículos (FALUMV), se realizó un bioensayo estático con concentraciones subletales de 0, 10-3%, 10-2% y 10-1%, durante 7 y 13 días. Los individuos fueron colectados en el golfo de Santa Fe, estado Sucre, Venezuela, y mantenidos en un sistema de acuarios, con renovaciones interdiarias de contaminantes y alimentos (microalgas), bajo condiciones controladas. El contenido de carbohidratos y proteínas, y los coeficientes moleculares de condición mostraron incrementos en los organismos expuestos a la FALUMV durante algunos de los períodos de exposición, sin embargo los niveles de lípidos totales no variaron en relación a los tratamientos evaluados. En el manto de L. scabra se evidenciaron ciertos ajustes bioquímicos de importancia, tales como la acumulación de macromoléculas energéticas e incremento en los niveles de los índices moleculares de condición, lo que es indicativo de una posible producción de proteínas vinculadas con la defensa bioquímica en presencia de mezclas complejas de xenobióticos. Used crankcase oil of automobiles is a potential source of hazardous contaminants, which upon reaching coastal ecosystems may affect adversely the biota. The bivalve associated to coralline reefs have been recognized as biological sensors to assessing the impact of stressing chemical substances in these marine ecosystems by means of use of biomarkers, especially changes in chemistry composition. In order to determine energetic substrate levels (protein, lipid and carbohydrate) and molecular ratios of condition (RNA/DNA and Protein/DNA) in the mantle of fire scallop Lima (Ctenoides) scabra exposed to water-soluble fraction (WSF), obtained from a mixture of used lubricants of automobile engines, a static bioassay was realized with sublethal concentrations of 0, 10-3%, 10-2% y 10-1%, during 7 and 13 days. Individuals were collected from Gulf of Santa Fe, Sucre state, Venezuela; and maintained in seawater aquaria system with changes of WSF and food (microalgae) inter-daily, under controlled conditions. Carbohydrate and protein levels, molecular ratios of condition showed increase in organisms exposed to WSF, during some exposure periods, however lipid levels did not differ in relation to the treatments. In the mantle of L. scabra certain biochemical adjustments of importance were evidenced, such as accumulation of energetic macromolecules and molecular ratios increased, indicating that production of protein may be associated to molecular defense against complex mixtures of xenobiotics.

Los cambios de nuestro hábitat en los últimos 40 años han generado contactos frecuentes con sustancias químicas tanto naturales como sintéticas que funcionan como disruptores endocrinos; son ejemplos los pesticidas ampliamente usados tras la introducción del DDT, los derivados petroquímicos en la industria del plástico, las medicinas sintéticas, y el volumen creciente de artículos de vestuario, aseo, cosmética y transporte. En cada uno de los grupos anotados hay señales de actividad de algún disruptor endocrino sea directamente o a través de sus derivados. La controversia inicial se centró principalmente en los compuestos químicamente similares a los estrógenos y en todos aquellos con actividad antiandrógenos o antitiroidea, debido a que se sospechaba que dañaban el ADN; recientemente se han estudiado otros compuestos y condiciones de vida que no lesionan el genoma pero sí interactúan con enzimas y sus metabolitos. Entre los efectos potenciales de estas sustancias cabe mencionar los siguientes: anomalías intrauterinas por daños del tejido fetal en formación; cambios del fenotipo genital al nacer y diversas manifestaciones clínicas durante la adolescencia; además, no pueden perderse de vista los efectos sobre las generaciones futuras. Los disruptores endocrinos son compuestos que pueden modular tanto la expresión endocrina e inmune del afectado como la homeostasis, la reproducción, el desarrollo y el comportamiento y tienen efectos sobre los tejidos reproductivos masculino y femenino, la fertilidad, la función tiroidea y el sistema nervioso central. Initial controversy dealt with products chemically similar to estrogens and with those with antiandrogenic and antithyroid activity because of their suspected harmful activity on DNA; more recently, new substances and life conditions have been introduced in the list of endocrine disruptors; although such substances do not damage the genome, they interact in inappropriate ways with enzymes and their metabolic pathways. Among the potential harmful effects of these substances the following should be emphasized: intrauterine abnormalities due to damage of fetal tissues, changes of genital phenotype, several clinical manifestations during adolescence and potential effects on future generations. Finally, there are substances that can modulate the endocrine and immune expressions of individuals as well as homeostasis, reproduction, development and behavior; they have effects on reproductive tissues, both male and female, fertility, thyroid function and the central nervous system.

Resumen: En la actualidad existe evidencia de los efectos deletéreos de la exposición de diferentes especies animales a xenobióticos, incluyendo herbicidas, pesticidas y plaguicidas.. En la literatura se han descrito los efectos de herbicidas tales como el glifosato en sistemas orgánicos como el respiratorio y el tegumentario en peces, sin embargo, reportes acerca del efecto de dichos productos en el sistema inmune de animales acuáticos son tan escasos como necesarios. La alteración de la dinámica y equilibrio de la respuesta inmune pudiera estar generando efectos importantes sobre la capacidad del pez para sobrevivir en el medio. El presente trabajo evaluó el efecto del glifosato (Roundup®) y surfactantes acompañantes (Cosmoflux® 411F), sobre el estado oxidativode la cachama blanca (Piaractus brachypomus) en un periodo de 14 días de exposición crónica subletal. Se evidenció que el glifosato(Roundup®) así como la mezcla de este con el surfactante induce aumento de la actividad oxidativa (estrés oxidativo) en los animales expuestos. Estos efectos fueron mayores cuando se expusieron a la mezcla atribuyendo un efecto importante al surfactante. These effects were higher in animal exposed to the mixture than Roundup® alone, giving an important role of surfactant on the effect. El presente trabajo determina el efecto tóxico de los metales cadmio (Cd), cobre (Cu), mercurio (Hg) y plomo (Pb) en la microalga tropical Tetraselmis chuii (Butcher, 1959). Se expuso, por triplicado, 50 ml de cultivo (f/2 Guillard) de la microalga en fase de crecimiento logarítmica ante las concentraciones de 0 (control); 0.1; 1.0; 5.0; 10.0 y 20.0 mg· l-1 durante 96 hr. La evaluación del efecto letal se realizó diariamente, mediante recuento celular con una cámara de Neubauer. En el tratamiento control, sin exposición al metal, se observó un incremento de la densidad celular, en contraste con un decrecimiento en los tratamientos con exposición a los metales, los cuales fueron acelerados hasta las 48 hr, a partir de cuando el decrecimiento se hizo menos pronunciado. Una excepción se produjo con el Cd y el Cu a las 24 h, donde no se determinó decrecimiento significativo, probablemente debido a su capacidad de actuar como micronutriente a bajas concentraciones. El metal que produjo mayor efecto fue el Pb, produciendo una letalidad al 50% de la población microalgal a 0.40 mg· l-1, la cual fue casi tres veces menor que la establecida para el Hg y más de 13 veces menor que la del Cd y Cu. Se recomienda la microalga Tetraselmis chuii como especie modelo para la utilización en pruebas en función estimar efectos tóxicos por xenobióticos en el ambiente acuático marino tropical.Efect of heavy metals on the growth of the tropical microalga Tetrasermis chuii (Prasinophyceae). We determined the toxic effect of four metals, cadmium (Cd), copper (Cu), mercury (Hg) and lead (Pb), on the tropical microalga Tetraselmis Chuii (Butcher, 1959). We exposed 50 ml of cultivated microalgae (f/2 Guillard) in the exponential growth phase, with three replicates, to concentrations of 0 (control), 0.1, 1.0, 5.0, 10.0 and 20.0 mg· l-1 with each metal for 96 hr. We evaluated the lethal effect daily, through the cellular count. In the control treatment (not exposed to any metal) we observed an increase in cellular density. In all treatments exposed to metals, we observed a decrease in cellular density, which accelerated in 48 h, after which it became less pronounced. There were exceptions with low concentrations of Cd and Cu at 24 h, as there was no significant decrease, probably due to their use as micronutrients at these low concentrations. The metal that caused the most lethal effect was Pb, which killed 50% of the microalgal population at a concentration of 0.40 mg· l-1. This concentration was 3 times lower than that of mercury and 13 times lower than those of cadmium and copper. Bracken adaptation mechanisms and xenobiotic chemistry (Alonso Amelot, Miguel; Oliveros, Alberto; Calcagno Pisarelli, María Pía and Arellano, Elida) Abstract As opposed to animals, plants have to cope with the resources, environmental restrictions, herbivores, and pathogens they find in the particular spot where they are bound to grow. Hence, resource sequestration, predation and competition relationships, and adaptation to various sources of other environmental stresses and their seasonal variation must be flexible enough to ensure survival and successful reproduction. Plants express this fitness by a combination of biological traits and chemical arsenals which operate under the reign of a genome of considerable plasticity. For the great majority of plants it is either the biological characters or the chemical composition that are explored independently to understand their fitness. But only in a few instances is the combination of these two avenues examined jointly. The extensive studies on the ecology, chemistry, and toxicology of bracken (Pteridium aquilinum) make this fern one of the few examples where a reasonable explanation for its extraordinary success is beginning to emerge by the combined perception of these two most important aspects of plant life. It is the purpose of this article to briefly review how the sum of biological and chemical traits cooperates to make of bracken one of the five most pernicious weeds in the world today. © 2001 IUPAC Pure Appl. Cytochrome P450 (CYP) enzymes participate in the metabolism of a variety of naturally occurring and foreign compounds by reactions requiring NADPH and O2. The diversity of reactions catalyzed and its extensive substrate specificity render CYP enzymes as one of the most versatile known catalysts. Individual members of the CYP superfamily are expressed in almost every cell type in the body. As compared to hepatic enzymes, the regulation of human extrahepatic CYPs has not been so well studied. In general, the levels of some hepatic CYP enzymes are depressed by diseases, causing potential and documented impairment of drug clearence and clinical drug toxicity. However, modulation of CYPs is enzyme selective and this selectivity differs in different diseases.
