RESUMEN El diseño de cebadores es fundamental para amplificar regiones de genes debido a que la especificidad que mantienen cebador-secuencia de interés puede causar el éxito o fracaso en la reacción de PCR. En relación a Potamotrygon magdalenae (especie de interés de acuerdo al PAN Tiburones-Colombia), existe poca información disponible de aspectos relacionados con la genética poblacional de esta Raya. El objetivo del presente trabajo consistió en diseñar cebadores bajo los criterios del Modelo de Pérdida de ADN (DNA-LM), que permitan evaluar el estado genético de las poblaciones de P. magdalenae. Alineamos secuencias de la superfamilia Dayastoidea, disponibles en el NCBI, de los genes mitocondriales Citocromo C Oxidasa 1 (MT-CO1) y Citocromo b (MT-CYB). Seguimos los parámetros Gap open penalty (5), Gap extension penalti (0,2) y Terminalgap penalties (0,1) y seleccionamos dos pares de cebadores de acuerdo con el DNA-LM. Estimamos el producto amplificado del gen MT-CO1 en 916 pb y del gen MT-CYB en 774 pb, en muestras de P. magdalenae procedentes de diferentes ciénagas del Magdalena medio. Discutimos los resultados desde la perspectiva de validar la especificidad de los cebadores diseñados, teniendo en cuenta la correspondencia e identidad de las secuencias de los genes considerados. Los cebadores aquí reportados pueden contribuir a ampliar el conocimiento de la genética poblacional, biogeografía y filogenética de la raya de agua dulce P. magdalenae. ABSTRACT The primer design is critical for amplifying gene regions because the specificity that primer-sequence maintain can cause success or failure in the PCR reaction. Concerning Potamotrygon magdalenae (a species of interest according to the PAN Tiburones-Colombia), there is little information available about aspects related to population genetics of this river stingray. This study aimed to design primers under DNA Loss Model (DNA-LM) criteria, which will allow assessing the genetics status of populations of P. magdalenae. We aligned sequences from the superfamily Dayastoidea, available in the NCBI, of the mitochondrial genes Cytochrome C Oxidase 1 (MT-CO1) and Cytochromeb (MT-CYB). We followed the parameters Gap open penalty (5), Gap extension penalty (0.2) and Terminal gap penalties (0.1) and selected two pairs of primers according to the DNA-LM. We calculated the amplified product of the MT-CO1 gene in 916 pb and the MT-CYB gene in 774 pb in samples from different swamps of the middle Magdalena. We discussed the results from the perspective of validating the specificity of the primers designed, considering the correspondence and identity of gene sequences considered. The primers reported here will contribute to broadening the knowledge of population genetics, biogeography and phylogenetic of the river stingray P. magdalenae.

La taxonomía es una de las principales herramientas de la conservación pero también de la biología en general: es imposible pensar en llevar a cabo una investigación sin conocer los organismos que se están estudiando. Sin embargo, Garnett y Christidis (2017) generaron una gran controversia al postular que La anarquía taxonómica obstaculiza la conservación, publicado en la prestigiosa y ampliamente leída revista Nature. En conjunto con este tan agudo y controvertido título, señalaron algunas de las principales falencias del quehacer taxonómico. Las críticas no se hicieron esperar: por un lado quienes se opusieron fuertemente a esta crítica, respondieron cuestionando rudamente y hasta ridiculizando a estos autores; por otro lado, quienes vieron la crítica de manera constructiva, tomaron los puntos fuertes y comenzaron a elaborar posibles soluciones, principalmente abordando las implicancias del albedrío y su relación con las conceptualizaciones de especie. En la presente tesis, tomo las principales críticas generadas en este debate entre taxonomistas, con el objetivo de generar una propuesta para el trabajo taxonómico, la cual denominé coherencia taxonómica. La puesta a prueba de este nuevo marco teórico resulta en una amalgama entre filosofía, diseño metodológico, estadística y taxonomía, cada una de ellas requerida para un abordaje coherentista. En primera instancia, y consecuentemente analizado desde la biología evolutiva, realizo una síntesis histórica de las principales disciplinas, conceptos y nociones que abordaré en el resto de la tesis. Seguidamente, planteo un marco ontológico novedoso sobre la noción de especie basado en la complejidad y la individualidad, y cómo llevar a cabo una empresa taxonómica dentro de este marco. El carácter individual y complejo de todo este marco teórico requiere ser puesto a prueba por fuera de la trivialidad de los organismos reconocidos como modelos. Es en este sentido que el grupo de lagartijas Liolaemus elongatus plantea un adecuado desafío para ser evaluado: posee una intrincada genealogía histórica y una morfología muy variable. Previo a hacer un análisis sobre las especies de este grupo, puse a prueba la repetibilidad, replicabilidad y el error de medición en la cuantificación de la morfología de la cabeza utilizando morfometría geométrica con el fin de explicitar la credibilidad de los resultados. Conociendo esto, junto con la potencialidad de esta técnica, la ulterior comparación entre taxones resulta más clara y genuina; así, pongo a prueba las diferencias entre los taxones de este grupo considerando distintas formas de clasificarlos. Finalmente, concluyo sobre el quehacer taxonómico en un marco coherentista. Taxonomy is one of the main tools of conservation but also of biology in general: it is impossible to think of research without knowing the organisms under study. However, Garnett & Christidis (2017) generated quite a controversy by postulating that Taxonomy anarchy hampers conservation, published in the prestigious and widely read journal Nature. With this very sharp and controversial title, they pointed out some of the main shortcomings of the taxonomic task. The critics did not wait: on the one hand, those who strongly opposed this criticism replied by rudely questioning and even ridiculing these authors; on other hand, those who took this criticism as constructive, took the strengths, and began to elaborate possible solutions, mainly addressing the implications of free will and its relationship with the conceptualization of species. In this thesis, I echo the main criticisms generated in this debate between taxonomists to build a proposal for taxonomic task, which I call taxonomic coherence. Testing this new theoretical framework results in an amalgam between philosophy, methodological design, statistics, and taxonomy, each of them required for a coherent approach. Firstly, from an evolutionary biology perspective, I make a historical analysis of the main disciplines, concepts, and notions that I will later use in the thesis. Next, I propose a novel ontological framework on the notion of species based on complexity and individuality and how to carry out a taxonomic enterprise within this framework. The individual and complex character of this ontological framework needed to be tested outside the ―model organisms‖ triviality. The Liolaemus elongatus group poses a suitable challenge to evaluate because it has an intricate historical genealogy and a highly variable morphology. Before analyzing species, I tested the repeatability, replicability, and measurement error in the quantification of the head morphology using geometric morphometrics to explain the results‘ reliability. Knowing the potentiality and reliability of this technique, the subsequent comparisons between taxa are clearer and more genuine; thus, I test the differences between the taxa of the Liolaemus elongatus group according to different classifications. RESUMEN La estructura genética de poblaciones de mariposas con distribución en islas y sus pares continentales ha sido poco documentada para el neotrópico. Este estudio presenta la caracterización de una población de Heliconius sara con distribución en la Isla Gorgona, ubicada en la región del Pacífico Oriental Colombiano. Para esto se examinaron secuencias parciales de un marcador mitocondrial incluyendo información obtenida del GenBank. Se comparó la diversidad y estructura genética con sus conespecíficos continentales y también con congéneres, con los que comparte un ancestro común cercano en el clado Sapho-Sara. Para el análisis de diversidad y estructura genética se realizó un análisis de varianza molecular. Este análisis muestra que la distancia entre la población de la isla y sus pares en el continente es consistente con la variación intraespecífica observada en otras especies del género Heliconius. Para la reconstrucción de la genealogía y datación reciente en el Pleistoceno superior del grupo monofilético de secuencias de H. sara, se realizó un análisis de inferencia bayesiana, así como una de máxima verosimilitud. Del análisis demográfico se seleccionó un modelo histórico de flujo asimétrico desde la isla hacia el continente que sugiere baja resistencia de la discontinuidad geográfica a la dispersión de esta mariposa diurna desde la isla. Este es el primer estudio en examinar un posible evento de aislamiento de una población insular de mariposas en Colombia. ABSTRACT The genetic structure of butterfly populations among islands and mainland has been poorly documented for the neotropics. This study shows a characterization of the Heliconius sara population with distribution on Gorgona Island in the Colombian Eastern Tropical Pacific region. We obtained partial sequences of a mitochondrial DNA, including information obtained from GenBank. The genetic diversity and structure were compared among the island population and their mainland conspecific, but also with congenerics, with those shared by a recent common ancestor within the Sapho/Sara clade. For the analysis of diversity and genetic structure, an analysis of molecular variance was performed. This analysis shows that the genetic distance between the island's population and that of the mainland is consistent with the intraspecific variation observed in other species of the Heliconious genus. For the reconstruction of the genealogy and the recent dating calibration in the upper Pleistocene of the monophyletic group of H. sara, a Bayesian inference was carried out as well as one of maximum likelihood. From the demographic analysis, an asymmetric gene flow model from the island to the mainland was selected. This model suggests low historical resistance of the geographic discontinuity to dispersal of this small and diurnal butterfly from the island. This is the first study to examine a possible event of local isolation of an island population of a butterfly in Colombia.

espanolEn los escenarios academicos e investigativos ha venido abriendose paso la transdisciplinariedad con las dificultades propias que trae la emersion de un paradigma revolucionario, trayendo consigo escollos de orden conceptual, epistemologico, metodologico y organizacional. Tratando de posicionarse en medio de un sistema cientifico jerarquizado, fragmentado, lineal, binomial y reduccionista; sin contar los influjos politico, economico y cultural que afectan toda empresa. Pero este joven modelo de comprension presenta en sus dinamicas intelectuales y operativas un problema de hondo calado: un hecho de caracter comportamental que ralentiza su progreso y que se situa en un nivel de realidad biologico primitivo en el que las comunidades educativas e investigativas poco reparan en sus dinamicas. Por tanto, se trata de un factor evolutivo que puede resultar muy sensible en la transdisciplinariedad, dado que esta nueva apuesta cientifica tiene como uno de sus mayores objetivos superar dicotomias y antagonismos en el conocimiento disciplinal para tener una comprension mas integral de la realidad, lo que amerita una logica subyacente complementaria como la subdisciplinariedad. Razones por la cuales se ha dispuesto discurrir de la mano del pensamiento complejo, fundamentalmente desde la perspectiva de Edgar Morin y Basarab Nicolescu. EnglishIn the academic and scientific research scenarios, transdisciplinarity has been opening up among the inherent difficulties that come from the emergence of a revolutionary paradigm, bringing conceptual, epistemological, methodological and organizational obstacles with it. Transdisciplinarity has been trying to harmonize with a hierarchical, fragmented, linear, binomial and reductionist scientific system. In addition, it is dealing with the political, economic and cultural influences that affects such emergence. However, this young comprehensive model presents in its intellectual and operational dynamics a deep-seated problem: a behavioral fact located on a level of primitive biological reality that slows down its progress and which the educational and research communities do not pay much attention to. We are dealing with an evolutionary factor that can be very sensitive in transdisciplinarity, since this new scientific trend has as one of its major objectives to overcome dichotomies and antagonisms in disciplinary knowledge in order to have a more comprehensive understanding of reality. This would demand a complementary underlying logic such as subdisciplinarity. Consequently, we have relied on complex thought, mainly from the perspective of Edgar Morin and Basarab Nicolescu.

The intrageneric relationships of the pampatheriid genus Scirrotherium and its affinities with supposedly related genera, i.e., Kraglievichia and Holmesina, are revised through new comparative morphological descriptions and parsimony phylogenetic analyses. For this work, unpublished material of pampatheriids (numerous osteoderms, one partial skull and a few postcranial bones) from Neogene formations of Colombia was analyzed. The results show that Scirrotherium is paraphyletic if we include all its referred species, i.e., Scirrotherium hondaensis, S. carinatum and S. antelucanus. The species S. carinatum is closer to Kraglievichia paranensis than to S. hondaensis or S. antelucanus, therefore the new name K. carinatum nov. comb. is proposed. The relationship among S. hondaensis and S. antelucanus could not be resolved, so these species should be designated in aphyly. In spite of failing to recover S. hondaensis and S. antelucanus as one single clade, here is preferred to maintain the generic name Scirrotherium in both species based on diagnostic evidence. New emended diagnoses for Scirrotherium, S. hondaensis and Kraglievichia are provided. The genus Holmesina was found to be monophyletic and positioned as the sister clade of Scirrotherium + Kraglievichia. The evolutionary and biogeographic implications of the taxonomic re-arrangements and the new phylogeny are discussed. A possible geographic origin of the family Pampatheriidae and Scirrotherium in low latitudes of South America as early as Early Miocene times is claimed.
