The treatment of colon cancer with a mesenchymal-like phenotype poses a great challenge. Purpose of the study: to research expression of FRMD6, ZEB1, HTR2B, CDX2 in the primary tumor tissue with relation of the clinical and pathological features of colon cancer. Material and methods. a combined analysis of the expression of FRMD6, ZEB1, HTR2B, CDX2 with pathological criteria was performed in 100 patients with T1–4bN0–2bM0 colon cancer. Results of the study show that tumors with mesenchymal signs are characterized by a large size of the primary tumor, a high grade of differentiation, regional metastases, and eventually advanced stage of the disease. The frequency and expression levels of FRMD6, ZEB1 and HTR2B are depended of grade of adenocarcinoma. in cases with positive expression and a high percentage of expression of these proteins in the cancer cells, the degree of tumor morphological anaplasia increases. Conclusion. The study made it possible to reveal the relationship of IHC markers (FRMD6, ZEB1, HTR2B, CDX2) with tumor characteristics that determine the outcome. The studied markers may have prognostic and predictive value in treatment approaches of colon cancer.

Purpose of the study. Evaluation of expression of the epithelial-mesenchymal transition markers E-cadherin and ZEB1 in patients with stage II-IV colorectal cancer (CRC).Materials and methods. The study included operational material obtained from 299 patients aged 42–86 years (mean age 64.2±1.7 years) with stage II-IV CRC treated at National Medical Research Centre for Oncology in 2013-2017. Stage II CRC (T3-4 N0 M0 ) was diagnosed in 110 patients, stage III (T1-4 N1-2 M0 ) – in 88 patients, stage IV (T1-4 N0-2 M1 ) – in 101 patients. Polyclonal rabbit antibodies to ZEB1 (Biorbyt Ltd., UK) and mouse monoclonal antibodies to E-cadherin (Diagnostic BioSystems, USA) were used for an IHC analysis. The intensity and degree of tumor cell staining, percentage of stained tumor cells in the sample and the number of patients with positive and negative marker expression were determined. Groups were compared using the Mann–Whitney U test and the Pearson's chi-square test.Results. Positive expression of E-cadherin was found in 64.5 % (193 of 299 patients), ZEB1 – in 80.6 % (241 of 299 patients). The study analyzes the expression of protein genes involved in intracellular signaling pathways associated with a profibrotic response, activation of epithelial-mesenchymal and endothelial-mesenchymal transitions, in modeling tuberculous granulomatosis and pharmaceutical effects. Material and methods. The study was performed on male BALB/c mice aged two months and weighing 18–22 g. Generalized tuberculous granulomatosis was simulated by a single intravenous (retro-orbital) injection of 0.5 mg BCG vaccine in 0.2 ml of isotonic aqueous NaCl solution (SS), after 4 months a part of mice started to receive treatment drugs, after 2 months the animals were sacrificed by decapitation (6 months after BCG vaccine administration) and their lung tissues were collected. Mice were divided into 6 groups, 5 males in each: intact, which were intravenously into the retro-orbital sinus injected with 0.2 ml SS (C); infected with BCG and receiving SS intraperitoneal injections (BS); infected with BCG and received intraperitoneal injections of isonicotinic acid hydrazide solution (INAH) (BI); infected with BCG and received intraperitoneal injections of dextrazide solution (conjugate of 40 kDa oxidized dextran and INAH) (BD); infected with BCG and received intraperitoneal or inhalation injections of solution of molecular-nanosomal pharmaceutical compositions of oxidized dextran (MNPC) (BMP and BMH, respectively). Finally, the expression of mRNA of matrix metalloproteinase 9, type III procollagen, TGF-β, and transcription factors ZEB1 and Snai1 was determined by real-time PCR in mice lung tissue. Results. The modeling of generalized tuberculous granulomatosis was found to be accompanied by the induction of epithelial-mesenchymal transition and the activation of profibrotic processes, which after 6 months was manifested in increase of the type III collagen α1-chain and TGF-β mRNA expression. Administration of traditional (INAH) and original (dextrazide, MNPC) preparations with anti-tuberculosis activity within two months has inhibitory activity of varying severity in relation to various markers of these processes.

Fuchs' corneal dystrophy (FCD) is a common bilateral non-inflammatory endothelial pathology. It is a multigenic disorder with various expressivity, penetrance and population prevalence. This review discusses corneal endothelium pump function, FCD pathogenesis and its known genetic factors.

Keratoconus is a chronic disorder of the cornea, characterized by its progressive thinning, stretching, and conical protrusion. Diagnostics of subclinical keratoconus, as well as its early stages (forme fruste), is a complex problem. The presence of these forms of keratoconus in a patient is one of the reasons for the development of keratectasia after laser refractive surgery. Currently, the role of genetic factors in keratoconus development has been proven. This indicates the possibility of diagnostics of subclinical and forme fruste keratoconus using genetic markers. Knowledge about the patient's genetic susceptibility to keratoconus would allow correcting the tactics of treatment of refractive anomalies and avoiding serious side effects. The studies of causal mutations indicate the genetic heterogeneity of keratoconus, which complicates the development of a diagnostic panel. Selection of candidate variants from the currently known ones based on clear criteria may be one of the approaches for diagnostic markers search. In this review, we have analyzed articles on keratoconus markers in order to form a list of candidate variants for genotyping in the Russian population. The selection criteria took into account the complexes of symptoms in which a marker was found, populations in which a particular marker was investigated, the presence and results of replication studies. The analysis included markers in VSX1, SOD1, ZEB1, LOX, CAST, DOCK9, TGFBI, HGF, MAP3K19, KCND3, COL4A3, COL4A4, COL5A1, FNDC3B, FOXO1, BANP-ZNF469, MPDZ-NF1B, WNT10A genes. Based on the results of the analysis, the following candidate variants were selected for genotyping in the Russian population of patients with keratoconus: rs1536482 and rs7044529 in the COL5A1 gene, rs5745752 and rs2286194 in the HGF gene, rs4954218 in the MAP3K19 gene, rs4839200 near the KCND3 gene, rs2721051 near the FOXO1 gene, rs1324183 between the MPDZ and the NF1B genes, and rs121908120 in the WNT10A gene.
