Данная работа посвящена изучению гетеротетрамерного стрептавидина. Целью работы является продемонстрировать возможность получения рекомбинантного гетеротетрамерного белка в E.coli при помощи котрансфекции бактерий двумя плазмидами. При этом одна из плазмид отвечает за синтез мономера белка с его преимущественным накоплением в тельцах включения, а вторая плазмида отвечает за синтез мономера белка и его транспорт в периплазму. В данной работе проработаны методики экспериментальных исследований: создание экспрессионной генетической конструкции белка, изучение оптимальных условий роста клеточной культуры для синтеза целевого белка, очистка целевого белка включающие электрофорез белков в ПААГ, выделение ДНК, рестрикция, лигирование, выделение белка из питательной среды культивирования, растворимой фракции и из телец включения и периплазмы. В качестве вектора для экспрессии стрептавидина использовалась плазмиды pET22b(+) и pTrc 99A, для наработки бактериальной массы использовались штаммы E.coli BL21(DE3). Гетеротетрамерный стрептавидин, локализируется в различных фракциях E.coli в зависимости от температурного режима индукции синтеза белка и IPTG. При температуре индукции 26°С гетеротетрамерный стрептавидин локализируется в значимом количестве в культуральной среде и периплазме. При температуре индукции 16°С гетеротетрамерный стрептавидин локализируется в значимом количестве в растворимой фракции клетки. Результаты проведенных исследований могут быть положены в основу разработки новых более эффективных методов наработки моновалентноо стрептавидина для его последующего применения в медицине и науке, также могут быть положены в основу разработки для обнаружения у пациента мутации в гене TTR при транстиретином амилоидозе., This work is devoted to the study of heterotetrameric streptavidine. The purpose of this work is to demonstrate the possibility of obtaining recombinant heterotetrameric protein in E. coli by simultaneous transfusion of bacteria by two plasmids. In this case, one of the plasmids is responsible for the synthesis of a protein monomer with its predominant accumulation in the inclusion bodies, and the second plasmid is responsible for the synthesis of a protein monomer and its transport into the periplasm. In this book methods of experimental research have been developed: creation of a protein expression of the genetic structure, the study of optimal growth conditions of cell culture for the synthesis of the target protein, the purification of the target protein, including protein electrophoresis in PAAG, DNA isolation, restriction, ligation, protein isolation from culture media, a soluble fraction and inclusion bodies and the periplasm. As vectors for the expression of streptavidine, pet22b(+) and ptrc 99A plasmids and E. strains were used. coli BL21 (DE3) were used to produce bacterial mass. Heterotetrameric streptavin is localized in different fractions of E. coli depending on the temperature regime of induction of protein synthesis and IPTG. At a Injection temperature of 26°С, heterotetrameric streptavidine is localized in a significant amount in the culture medium and periplasma. At an induction temperature of 16os, heteroteramic streptavidine is localized in a significant amount in the soluble fraction of the cell. The results of the research can be used as a basis for the development of new more effective methods for the development of monovalent streptavidin for its subsequent use in medicine and science, and can also be used as a basis for the development of a mutation in the TTR gene for patients with transtyretin amyloidosis.
