Introduction. Search for new plant species containing biologically active substances (hereinafter – BAS) is one of the leading tasks of pharmacognosy as a science. The search for flavonoid glycosides in plant raw materials is especially relevant, since they have anti-inflammatory, antioxidant, immunostimulating, as well as weak fungicidal and bacteriostatic action. Staphylea pinnata L. is an endemic plant of the Caucasus, cultivated not only in Georgia, but also in the Russian Federation in the Northern and Northwestern Caucasus. In foreign literature there are studies of antioxidant, antiproliferative and cytotoxic activity of leaf extracts of several species of S. pinnata L., as well as inhibitory activity of COX-1, COX-2 and LTB4 formation. Meanwhile, no serious Russian-language scientific studies on either the chemical composition or pharmacological action of generative organs of S. pinnata were found in the literature. This work is part of a comprehensive phytochemical study of S. pinnata. The aim of the work is to study the qualitative and quantitative composition of flavonoids in the studied object.Aim. To isolate, identify and quantify flavonoids in flowers and buds of Staphylea pinnata L.Materials and methods. Alcohol-water extracts from dried generative organs of the studied plant were used as analyzed solutions. Solutions were analyzed on a spectrophotometer SF-2000 (LLC "OKB Spectr", Russia) after sample preparation with aluminum chloride and on an HPLC Nexera-i LC-2040 (Shimadzu Corporation, Japan) equipped with a column and sample thermostat, degasser and autosampler using an individually selected elution gradient of the mobile phase (0.1 % orthophosphoric acid/acetonitrile solution). The primary data were processed using LabSolutions Single LC software (Shimadzu Corporation, Japan). Compounds from the flavonoid group were identified by retention times. Detection was carried out using a UV detector with an absorption wavelength of 365 ± 2 nm.Result and discussion. Alcohol-water extracts from flowers and buds of S. pinnata L. were obtained. Quantitative evaluation by spectrophotometry for flavonoid content was carried out. A gradient elution mode for HPLC was selected for simultaneous determination of 7 flavonoid glycosides. These chromatographic conditions allowed the identification and quantification of astragaline, cynaroside, cosmosiin, narcissin and rutin in flowers and buds of Staphylea pinnata L. Flavonoid glycosides: raponticin and kaempferol were not detected.Conclusion. Flavonoid glycosides were isolated from the generative organs of S. pinnata L., a technique for quantitative determination of flavonoid glycosides in alcohol-water extracts was developed, astragalin, cynaroside, cosmosiin, narcissin and rutin were detected and quantified.

The Thymus L. genus species are extremely widespread in temperate countries and number up to 200 main wild plants, of which up to 50 have been identified in the modern Ukraine flora. Extracts from the official species of the Thymus L. genus are part of effective herbal preparations with pronounced anti-inflammatory, antimicrobial, and antioxidant activities. A promising direction of modern phytochemical research is the determination of the presence and accumulation of biologically active ascorbic acid in the herbs of widespread wild and cultivated species of the Thymus L. genus. The aim of the work is to determine the presence and accumulation of biologically active L-ascorbic acid in the herbs of four widespread wild and cultivated species of the Thymus L. genus of Ukraine flora using thin layer chromatography (TLC) and spectrophotometry methods during vegetation season. Materials and methods. The research used herbs of four widespread wild and cultivated species of the Thymus L. genus of Ukraine flora during the growing season (June – August 2023). The presence and quantitative content of ascorbic acid were determined by TLC on a “Biostep” CD 60 densitometer (Germany) and spectrophotometry on a “Lambda 365’ device (USA). Results. TLC and spectrophotometry methods were used to determine the presence and quantitative content of ascorbic acid in the herbs of four widespread Thymus L. genus species of Ukraine flora during the growing season. Accumulation of the compound was higher during flowering of cultivated species than wild plants. For the herbs Thymus vulgaris L. up to 39.10 ± 3.88 mg%; Th. x citriodorus (Pers.) Schreb. var. “Silver Queen” up to 36.19 ± 3.59 mg%. Conclusions. Considering the results obtained by TLC and spectrophotometry methods, it can be concluded that the determination of ascorbic acid presence and content in the Thymus L. species genus herbs is appropriate for obtaining cosmetology preparations with pronounced anti-inflammatory, antioxidant, and regenerating activity for normal and problem skin.

Lignosulfonates are the most common commercial lignin-based product due to their unique properties. Various methods are known for modifying lignosulfonates and lignosulfonic acids. This article presents the results of the development of a new approach to the production of nitrosated lignosulfonic acids. The method is based on a reaction catalyzed by cation exchange resins in the H-form: KU-2-8 cation exchanger and wofatite. The influence of reagent consumption and reaction duration on the course of nitrosation has been studied. The dynamics of the proposed nitrosation practically coincides with the dynamics of a similar reaction using sulfuric acid by the known method. The optimal consumption of sodium nitrite equals 15 %, and the optimal consumption of cation exchanger equals 100 % by weight of lignosulfonates. During the nitrosation of lignosulfonates, significant changes in the electronic spectra occur in the region of 280...500 nm. Two overlapping absorption bands appear with maxima at 300 and 330 nm, as well as an intense absorption band at 430 nm, due to nitroso groups conjugated with the aromatic nuclei of phenylpropane units. To analyze the ionization spectra, they have been deconvoluted. The resulting spectra are well approximated by 5 Gaussians with an error of no more than 5 %. Two options for carrying out the nitrosation reaction of lignosulfonates have been proposed: under static and dynamic conditions. It has been established that under dynamic conditions, nitroso derivatives of lignosulfonic acids are formed that do not contain metal cations, and the pH of the resulting solutions does not exceed 1.4. The elemental compositions of the isolated initial and nitrosated lignosulfonic acids have been determined. The nitrogen content of lignosulfonic acids has increased from 0.32 (initial) to 2.17 % (nitrosated). In addition, under dynamic conditions, an additional stage of separating the cation exchanger from the reaction medium by filtration is not required. New bands have appeared in the IR spectrum of nitrosated lignosulfonic acids: at 1540 cm–1, which is due to the presence of nitroso groups, and a wide absorption band at 1700...1715 cm–1, which can be caused by vibrations of the carboxyl group or the quinone monooxime tautomeric form of the guaiacyl structures of lignosulfonic acids.

INTRODUCTION. Polyphenolic compounds of various classes have pronounced absorption bands overlapping in almost the same ultraviolet region. This superposition of bands complicates the quantification of total hydroxycinnamic acids in herbal drugs by direct spectrophotometry without additional sample preparation.AIM. This study aimed to establish the possibility of quantifying hydroxycinnamic acids in herbal drugs by spectrophotometry in the presence of other polyphenolic compounds.DISCUSSION. This review analyses publications on analytical procedures for the spectrophotometric quantification of hydroxycinnamic acids that are based on the preliminary separation or chemical modification of polyphenolic compounds (combining the use of spectrophotometry, chromatography, extraction, Arnow’s reagent, etc.). The described sample preparation can help to resolve absorption bands in the spectra of polyphenolic compounds. The results of assessing the characteristic parameters of these analytical procedures (selectivity, sensitivity, labour intensity, etc.) can serve as a basis for searching novel approaches to the quantitative determination of hydroxycinnamic acids in plants.CONCLUSIONS. To determine the content of hydroxycinnamic acids in mixtures of polyphenolic compounds, analysts may use special sample preparation (combining spectrophotometry with chromatography and extraction) or chemical modification of the initial compounds. The acquired spectra could benefit from specific mathematical processing (derivative photometry and Vierordt’s method). However, even with the abovementioned approaches, it may not always be possible to increase the selectivity of an analytical procedure. Therefore, there is a need to further improve existing analytical procedures for the quantification of hydroxycinnamic acids and develop new ones.

Background. The increasing pollution of natural waters by heavy metals has become a major problem in recent years, so their determination requires separation and pre-concentration techniques. Purpose. Development of a hybrid, sensitive sorption-spectrophotometric method for determining the Zn(II) ion using a polyethylene polyamine polyacrylonitrile (PPF-1) matrix immobilized in alizarin red C. Methodology. Research was carried out using a spectrophotometer, diffuse reflectance spectroscopy, IR spectroscopy and methods of mathematical statistics. Originality. For the first time, alizarin red C (AKS) was immobilized on a modified PPF-1 and a new matrix was used for the effective determination of Zn(II) Findings. The sensitivity of Zn(II) determination by selective immobilized ACS was maximum at 522 nm. The optimal pH for determining Zn(II) is 5.0 at room temperature. The linear range was found to be 6.5-40 mg/L, the detection limit was 0.6 mg/L, and the adsorption percentage was about 97%. [ABSTRACT FROM AUTHOR]

Background. Due to the increasing demand for hepatoprotectors based on silymarin, it is very important in Turkmenistan to work out the quality indicators of the raw materials used to obtain extracts from local medicinal plants. Purpose: Determination of quality indicators of milk thistle seeds through pharmacognostic studies. Methodology. The phytochemical composition and quality indicators of milk thistle seeds were studied according to the methods of the State Pharmacopoeia of the Russian Federation-XIII. Originality. Increased polymorphism in milk thistle seed coats was established, as well as a change in their color from light brown to brown and black. Microscopic examination of longitudinal and transverse sections of milk thistle achenes reveals cotyledons surrounded by a thick layer of tightly fused sclereids, noticeable by their natural yellow color. Findings. The phytochemical composition and quality indicators of milk thistle seeds have proven compliance with Global Fund standards. [ABSTRACT FROM AUTHOR]

The method has been proposed for the spectrophotometric determination of antibiotics of the cephalosporin (cefuroxime, ceftriaxone, cefotaxime, cefazolin) and penicillin (amoxicillin) groups using chemometric data processing (method of partial least squares, PLS). The absorption spectra of 24 binary mixtures of “cephalosporin – cephalosporin” and 16 binary mixtures of “cephalosporin – amoxicillin” with various concentrations in the range of 3–27 and 5–140 μg/ml have been recorded. The optimal number of latent variables for each chemometric method (PLS-1 and PLS-2) has been selected, which ranged from 2 to 7 depending on the type of model and the nature of the antibiotics. The smallest root mean square calibration errors (RMSEC) have been found for the ceftriaxone–cefazolin system, which have been 0.09 and 0.08 for PLS-1 and PLS-2, respectively. The smallest root mean square errors of prediction (RMSEP) for the mixture “cefuroxime – amoxicillin” have been 0.07 for both methods (PLS-1 and PLS-2). It has been established that equations of linear relationships “measured-predicted” are characterized by a regression coeffi cient and the square of the approximation coeffi cient close to 1; which indicates the high quality of the models. The relative error of determination in test mixtures has been 0.4–6.2% by the PLS-1 method for “cephalosporin – cephalosporin” mixtures, and 0.07–6.0% by the PLS-2 method. At the same time, there is no signifi cant diff erence in the error in determining the components of the mixture, in contrast to the “cefuroxime – amoxicillin” system, for which the error is 0.1–1.1% for the fi rst component and 2.6–14.7% for the second component using the PLS-1 method and 0.2–1.2% for the fi rst component and 4.3–13.3% for the second component using the PLS-2 method. Thus, it has been shown that the use of chemometric methods ensures high accuracy of the spectrophotometric determination of the studied antibiotics with overlapping absorption bands in their mixtures.

Scientific relevance. When harmonising conditions for qualitative and quantitative analysis of bioactive compounds in herbal medicines, the Institute of Pharmacopoeia and Medicinal Product Standardisation has discovered that horse-chestnut products lack national quality standards.Aim. This study aimed to review international quality standards in order to identify the most promising testing methods for the main groups of bioactive compounds in horse-chestnut seeds and draw upon these methods when drafting national pharmacopoeial monographs for horse-chestnut seed products.Discussion. The authors compared the requirements established by leading world pharmacopoeias for the identification and assay of herbal drugs and herbal drug preparations derived from horse-chestnut seeds.Conclusions. The study results show that, predominantly, identification tests are based on thin-layer chromatography, assays rely on spectrophotometry, and aescin is used as a pharmacopoeial reference standard. The authors drafted a pharmacopoeial monograph for the medicinal product Horse-Chestnut Seed Dry Extract + Thiamine Hydrochloride, Oral Solution, which requires using thin-layer chromatography and spectrophotometry for qualitative and quantitative testing.

Licorice herb (Glycyrrhiza glabra L.) is a promising herbal raw material, which can be comprehensively used to develop drugs with an anti-inflammatory action.The aim of the article was to development a quantitative determination method of total flavonoids in Glycyrrhiza glabra L. herbs.Materials and methods. The subjects of research were 5 samples of licorice herb harvested in summer in various places of growing and cultivation. Pinostrobin was used as a standard sample. The registration of the electronic spectra was carried out with a spectrophotometer (Analytik Jena AG, Germany) by differential spectrophotometry, 96% ethanol was used as a solvent.Results. The methods for quantitative determination of total flavonoids in Glycyrrhiza glabra L. was carried out at an analytical wavelength of 310 nm equivalent to pinocembrin. The optimum parameters for the extraction of total flavonoids from Glycyrrhiza glabra L. were as follows: the extractant – 90% ethanol; the «raw material-extractant» ratio was 1:50; the extraction time was 60 min; the degree of atomization was 2 mm. The content of total flavonoids for the Glyccyrhiza glabra L. herb has been determined, it varies from 0.39±0.002 to 3.41±0.015% with the humidity of the vegetative raw material from 9.97±0.003 to 10.03±0.003% depending on the place of the vegetation, cultivation and year of the raw material collection. The error of the single determination with a 95% confidence level was ±0.73%.Conclusion. The developed methods for the quantitative determination of total flavonoids in Glycyrrhiza glabra L. herbs can be used to solve the issues of standardization of these medicinal plant raw materials.

Purpose: to compare the regenerative activity of benign and malignant conjunctival tumors.Material and methods. The study was performed on 86 tumor biopsies from patients with a clinically diagnosed conjunctival neoplasm. Healthy tissues from the same eye were taken as a control sample. The optical density of the reaction mixture containing biogenic nanoparticles formed in the presence of conjunctival tumors and the corresponding paired healthy tissue samples from the control group was measured by spectrophotometry. The data obtained were later verified by a pathohistological analysis.Results. A significantly higher level of in situ formation of biogenic silver nanoparticles was recorded in the reaction mixture with malignant tumors of the conjunctiva (3.0 ± 1.1, (n = 32) compared to benign tumors (1.3 ± 0.2, n = 54). No differences were found between various types of benign tumors. In samples of malignant tumors, the indicators of recovery activity in melanoma (3.4 ± 1.0, n = 14) and lymphoma (2.8 ± 1.0, n = 7) were significantly higher than in squamous cell carcinoma (2.0 ± 0.6, n = 11), but no significant differences were found between the two types of tumors.Conclusion. The method of spectrophotometric measurement of the regenerative activity of conjunctival tumors can be used in preoperative or intraoperative diagnostics thanks to the fact that the results can be obtained rapidly, which will help to quickly determine the extent of surgical intervention needed and optimize the treatment tactics.

Aim. To develop and validate a spectrophotometric technique for quantitative determination of glibenclamide in tablets by reaction with 2,3-dichloro-1,4-naphthoquinone. Materials and methods. In the study, the substance glibenclamide of pharmacopoeial purity was used, tablets – “Maninil” 5 mg and “Glibenclamid-Zdorovye” 5 mg, 2,3-dichloro-1,4-naphthoquinone of the “CFA” qualification was chosen as a reagent, as a solvent – dimethylformamide (DMF) “CFA”. The following analytical equipment was used for the research: spectrophotometer “Specord-200” (Analytic Jena AG, Germany), water bath “MEMMERT WNB7”, laboratory electronic scales RADWAG XA 210. 4Y, measuring laboratory vessels of class A. Results. A new, simple spectrophotometric method of quantitative determination of glibenclamide in tablets by reaction with 2,3-dichloro-1,4-naphthoquinone in DMF medium was developed. The absorption maximum was at 489–491 nm. The value of the detection limit is 10.9 μg/ml, which indicates sufficient sensitivity of the reaction. Subordination to the basic law of light absorption is within the limits of concentrations of 13.7–27.4 mg/100 ml. In the process of developing the methodology, the following validation characteristics were determined: specificity, linearity, precision, correctness, and robustness. Conclusions. The methodology for the quantitative determination of glibenclamide was developed and validated according to the requirements of the State Pharmacopoeia of Ukraine. It has been proven that the method is simple, accessible, and validated for such characteristics as linearity, convergence and robustness and can be used for application in laboratories for quality control of medicinal products.

According to the authors’ analysis of regulatory documents for homoeopathic medicines of Virginian witch hazel (Hamamelis virginiana L.), there is a need to transform quality assessment approaches and to unify analytical procedures for identification and assay tests.The aim of the study was to determine approaches to identification and assay of homoeopathic mother tinctures of Virginian witch hazel leaves and bark and medicinal products based on the tinctures.Materials and methods: in order to select quality evaluation procedures, the authors analysed scientific literature, Russian and foreign pharmacopoeias and regulatory documentation regarding homoeopathic medicines of Witch hazel. The authors performed practical evaluation of the selected procedures based on thin-layer chromatography, spectrophotometry, qualitative reactions, and titrimetry, using samples of homoeopathic mother tinctures of Witch hazel leaves and bark, as well as corresponding homoeopathic medicinal products in various dosage forms.Results: the authors proposed identification and assay procedures for the studied active pharmaceutical ingredients (APIs) and homoeopathic medicinal products and prepared the drafts of pharmacopoeial monographs Hamamelis virginiana e foliis (Hamamelis, Folium) homoeopathic mother tincture and Hamamelis virginiana (Hamamelis) homoeopathic mother tincture. The unified procedures were included in the draft monographs for homoeopathic medicinal products Hamamelis, homoeopathic drops; Hamamelis D1, homoeopathic ointment; Hamamelis, folium D1, homoeopathic ointment; Hamamelis virginiana e foliis D1, rectal homoeopathic suppositories.Conclusions: the developed procedures are compatible with the principle of holistic, end-to-end standardisation, as they make it possible to carry out identification tests and assays using the same class of biologically active compounds throughout the whole range of homoeopathic medicines from APIs to finished products. The authors proposed the methodology for assessing homoeopathic medicinal products’ quality, taking into account the degree of dilution of the homoeopathic tinctures used.

Introduction. Golden dock (Rumex maritimus L., Polygonaceae) is used as a medicinal and food plant in Asian countries. The plant contains phytochemicals of various classes: flavonoids, tannins, anthraquinones etc. Plant extracts exhibit antibacterial, antioxidant, anti-inflammatory, astringent activity, and have antidiabetic potential. The plant is annual, and most of the biologically active substances accumulate in its aboveground organs. An important problem is the standardization of Rumex maritimus and the development of regulatory documentation for its the introduction to medical practice.Aim. To conduct phytochemical analysis of the aerial part of golden dock.Materials and methods. Air-dried aerial part of Rumex maritimus collected at flowering and beginning of fruiting stage, as well as individual above-ground organs (leaves, flowers, fruits, stems), were used for obtaining the extracts. Qualitative analysis of the extracts was carried out using reverse phase HPLC. The relative content of the components in the mixture was calculated by the method of simple normalization. Total content of free anthraquinones and anthraglycosides in terms of chrysophanic acid was determined using spectrophotometric method after acid hydrolysis. Total tannin content was calculated by titrimetric method.Results and discussion. Flavonoids isoquercetin and avicularin were first discovered in the aerial part of Rumex maritimus. The dominant component of the plant is rutin. Chrysophanol predominates among anthraquinones. The highest concentration of anthraquinones (2.80 ± 0.04 %) was found in flowers. Tannins accumulate mainly in leaves (9.97 ± 0.02 %). A significant amount of tannins (6.60 ± 0.03 %) and anthracene derivatives (1.96 ± 0.03 %) is contained in the whole aerial part.Conclusion. Phytochemical analysis of the aerial part of Rumex maritimus showed the presence of a significant amount of anthraquinones. As a plant raw material it is proposed to use the herb of Rumex maritimus. Standardization is recommended for anthraquinones in terms of chrysophanic acid (at least 1.5 %).

Background: Sepsis is a serious clinical condition caused by a dysregulated immune response to infection resulting in multiple organ failure. In the pathogenesis of sepsis, especially that of septic shock, great importance is given to the endothelial marker of vascular regulation, nitric oxide (NO). In septic shock, dysregulation of the vascular tone plays a key role in the development of hypotension. Therefore, the control of the level of nitric oxide and its stable metabolites in critically ill patients is a very important task. Aim: the aim of this study was to evaluate the potential of the electrochemical nitrite detection in the patients blood serum. Methods: The levels of nitric oxide stable metabolites in the blood serum of healthy individuals (n=20) and septic patients (n=25) were studied by the electrochemical method using a composite electrode and by the spectrophotometric method using the Griess reagent. Results: The data in the groups of healthy people and patients with sepsis differ significantly (p 0.00001) both when measured using electrochemical and spectrophotometric methods. The median value of the current response in healthy people was 0.41 A (0.33; 0.55), and the total content of nitric oxide metabolites (NOx) was 26.8 mol/L (20.8; 31.0), while in patients with sepsis, these values were 0.79 A (0.61; 1.28) and 38.89 mol/L (29.64; 57.45), respectively. A negative correlation was found between the data obtained for practically healthy persons (r=-0.696, p=0.0007). Conclusion: The obtained results allow us to conclude that the nitrite measurement in the blood serum by amperometry using a composite electrode is promising as a diagnostic technique.

According to statistics, craniocerebral injuries are often the cause of disability among the population. The choice of medicines for traumatic brain injuries is one of the most difficult problems in the complex treatment of such patients. Ademol (1-adamantylethyloxy-3-morpholino-2-propanol hydrochloride) is a promising medicinal substance that has a huge positive therapeutic effect of a protective act on the damaged brain and can be presented in medicinal preparations in different dosages. Therefore, the urgent task is the development of highly accurate, reliable, affordable, and fast methods of quantitative determination of ademol. The aim of the work is to study the optimal conditions for the “ademol – bromocresol green” reaction and to develop an express, sensitive and easy-to-implement method for the quantitative determination of ademol. Materials and methods. Bromocresol green (BCG) grade “chemically pure” and acetone grade “pure for analysis” were used as reagents and solvents. Analytical equipment: spectrophotometer Specord 200, electronic scales AVT-120-5DM, ultrasonic bath Elmasonic E 60H, class A measuring vessels. Results. A new method of quantitative determination of ademol by the spectrophotometric method was developed. The optimal conditions of the flow between the substance to be determined and the reagent have been studied, the concentration limits have been established, in which subordination to the basic law of light absorption is observed. A forecast of the complete uncertainty of the results of the specified method of quantitative determination was made to assess the correctness of the reproduction of the developed method in other laboratories. The proposed method is relevant according to the requirements of the State Pharmacopoeia of Ukraine.

The brackish microalgae Dunaliella salina, being an extremophilic halophyte, is a promising object for biotechnological production. The aim of this work is to develop a methodology for non-destructive control of the development of a microalgae culture under conditions of balanced growth during periodic cultivation on plates. Before the start of the experiment, the microalgae culture was synchronized. The quantitative content of chlorophylls a and b, as well as carotenoids, was determined spectrophotometrically in alcohol extracts. During cultivation, time-lapse images were recorded on a smartphone camera. The basis of the colorimetric evaluation is the analysis of the time series of images in the RGB color model. It is shown that the ratio of colors correlates to a high degree with the content of the determined main plant pigments – chlorophylls and carotenoids, and with the data of spectrophotometric measurements of live suspensions. The changes in the blue channel are the most pronounced, the least being in the green channel. The logarithm of color intensity is linearly dependent on the degree of dilution of the culture. The developed method for real-time monitoring of the development dynamics of the D. salina microalgae culture makes it possible to build growth curves and solve multiparametric problems to optimize the cultivation of microalgae, including when working with large arrays of samples.

Ensuring food safety of the quality of sold products is one of the urgent problems of the wine industry. The trend of falsification of well-known brands of domestic and imported wines is a characteristic feature of the modern market of wine products. Known methodological bases for assessing the quality of wine products in most cases do not have the potential to recognize many modern «technological solutions». To solve this problem an analytical method can be proposed on modern equipment "FTIR BACCHUS 3". This analyzer allows you to determine the chemical substances and the range characteristics of these substances. This method has been chosen for the identification of wine samples and not by chance, when using it, the analysis time is significantly reduced, and the range of chemical elements determined by the analyzer is quite wide, and reagents are not needed for this equipment. The method does not require preliminary preparation of samples; it is also quite new, not popular in Russia. The article presents a comparative review of classical wine identification methods. The presented material indicates the relevance of creating a database on the chemical composition of wines that determine their authenticity using new instrumental techniques. The proposed multi-parameter wine analyzer FTIR BACCHUS 3 is a system for the analysis of worts and wines using chemometric methods, based on the acquisition of the mid-IR spectrum (FTIR). The method is based on chemometrics, i.e. obtaining chemical data using mathematical methods of data processing and extraction, which will increase the reliability of assessing the authenticity and place of origin of wine products.

Interferons (IFN) have antiviral activity against many viruses, including SARS-CoV-2. A combination of IFN-a2b and the antioxidant taurine is widely used in the Russian Federation, and its antiviral activity has not been studied before. The aim of this study was to determine the antiviral activity of interferon drugs, in combination with taurine and without it. The study included cytotoxicity and antiviral activity assays of IFN-a2b preparations, when stored according to the instructions at 2–8°C, and after 1 month storage at the temperature of 20–26°C in a pre-opened state. The combination of IFN alpha-2b with taurine has a higher antiviral activity compared to IFN alpha-2b mono-preparation by more than 25% at a «low» and 85% at a «high» multiplicity of infection. Selectivity index for combinations of IFN-a2b (50,000 IU/dose) + taurine (1 mg/ml) and IFN-a2b (10,000 IU/ml) + taurine (0.8 mg/ml) was more than 600 units, whereas for the IFN-a2b (10,000 IU/ml) it was 200 units. Antiviral activity does not change after one month at room temperature. The combination of interferon with taurine at high concentrations was less toxic than interferon. The results obtained demonstrate practicability of interferon alpha-2b and taurine combination use for treatment and prevention of COVID-19.

Introduction. The features of the chemical composition and the identified pharmacological activities of substances from fresh raw materials of the Alchemilla vulgaris allow us to predict the prospects for the use of juice from the herb Alchemilla vulgaris in medical practice. The creation of drugs based on juices from fresh herb Alchemilla involves the development of methods for their standardization.Aim. To determine the methods of qualitative analysis for the identification and quantitative assessment of the main group of biologically active substances in Alchemilla vulgaris juice.Materials and methods. The herb Alchemilla vulgaris for juice production was harvested during the flowering period in various habitats. The qualitative composition of the phenolic compounds of the juice was studied by paper chromatography and thin layer chromatography (TLC). The quantitative determination procedure was developed and validated on an SF-2000 spectrophotometer.Results and discussion. Paper chromatography (CP) in the juice revealed derivatives of cinnamic and phenolcarboxylic acids, a group of flavonoids with a predominance of flavones and flavonols, rutin was identified among the dominant ones. The TLC in the butanol : acetic acid : water system (4 : 1 : 2), we achieved optimal separation of substances and identified rutin, which was proposed as the main substance in juice standardization. Correspondence of the configuration of the differential spectrum of the complex of cuff juice flavonoids with aluminum chloride to the spectrum of a similar complex of rutin SS and the results of TLC give grounds to use rutin as a standard. The best option when choosing an aliquot for quantitative determination of reliable results showed the use of 0.2 ml of juice. The content of flavonoids in the studied juice samples ranged from 2.15 ± 0.24 % to 7.34 ± 0.16 %.Conclusion. TLC in the butanol-acetic acid-water system (4 : 1 : 2) can be used to authenticate the juice from the herb Alchemilla vulgaris. The parameters of the method of quantitative determination are developed taking into account the variation in the ratio of the water component and the dry residue in the juice. The method is valid and can be used to standardize juice from the herb Alchemilla vulgaris. The taxation of Alchemilla vulgaris juice samples showed the variability of the content of flavonoids in the juice depending on the growing conditions.

Introduction. Alchemilla herb is proposed for introduction into pharmaceutical practice, for obtaining extracts with various pharmacological activity. To expand, resource base of Alchemilla it has been proposed to use a high biomass cultivated plant – Alchemilla mollis (Buuser) Rothm. In terms of qualitative and quantitative characteristics of the chemical composition Alchemilla mollis herb is comparable to the samples of raw materials of wild-growing Alchemilla. The presence of chemotaxonomic features necessitates the preparation of regulatory documentation for the introduction of this plant as a source of medicinal raw materials.Aim. Development of a methodology for assessing the main group of biologically active substances and the quantitative determination of flavonoids in Alchemilla mollis herb.Materials and methods. As objects of study, we used Alchemilla mollis herb harvested from plants cultivated in the Perm Krai. The chromatographic parameters of raw material authenticity were determined by thin layer chromatography (TLC). To develop the parameters for the quantitative determination of flavonoids in Alchemilla mollis herb, a modification of the method proposed for Alchemilla herb was carried out.Results and discussion. During chromatographic study of Alchemilla mollis herb were identified cinaroside, rutin, and quercetin. Cynaroside was referred to the marker substances. Ethyl acetate : acetic acid (85 : 15) was chosen as the optimal chromatographic system. A modification and validation of the method for the quantitative determination of flavonoids in Alchemilla herb was carried out. The change of the extractant, particle size, time and frequency of extraction for sample preparation and the optimal use of cynaroside as a standard substance are substantiated. The optimal conditions for the reaction of complex formation with aluminum chloride are established.Conclusion. To determine the authenticity of Alchemilla mollis herb, it was proposed to use the identification of cynaroside by TLC. The modified method for the quantitative determination of flavonoids in terms of cynaroside for the Alchemilla mollis herb, reproducible, correct and can be used for standardization. When testing the methodology on samples of raw materials harvested in the Perm region, a range of values for the content of flavonoids was 3.14–4.84 %, with an average level of variability.

Introduction. According to the World Health Organization, the number of patients with diseases of the nervous system in the world is increasing every year. The use of phytopreparations is a promising direction of pharmacotherapy; preparations from medicinal plant materials have a number of advantages in the treatment of chronic diseases.Aim. Qualitative analysis and quantitative determination of the main groups of biologically active substances collected from medicinal plant materials.Materials and methods. As an object of study, we used a collection of medicinal plant materials developed at the Department of Pharmaceutical Technology of the PSFA together with the Department of Pharmacognosy. Quantitative determination of biologically active substances of the collection was carried out by spectrophotometric method [Jenway PortLab 511 UV spectrophotometer PortLab Int., UK)] and by visual titration.Results and discussion. In this work, a qualitative analysis and quantitative determination of the content of flavonoids, phenolic compounds, sesquiterpenic acids and ascorbic acid was carried out, and the water absorption coefficient of the collection was determined.Conclusion. As a result of the work, the content of flavonoids, phenolic compounds, sesquiterpenic acids and ascorbic acid in the developed collection was confirmed, their amount was determined, which directly affects the pharmacological effect of the collection.

4-Aminophenol (APh) is a toxic chemical that may be present as an impurity in drugs based on paracetamol, one of the main antipyretic analgesics. Thus, its content must be controlled to curb potential negative effects. Here, we explored the use of the response surface method and the three-level Box–Behnken designs to obtain an optimal premicellar matrix for the spectrophotometric determination of the APh impurities in drugs. For this purpose, the spectrochemical reaction of APh with 5,7-dichloro-4,6-dinitrobenzofuroxan (DCDNBF) was carried out, and the UV spectrum of the reaction product in the premicellar matrix composed of ethoxylated nonylphenol (neonol, APh 9-10) + dimethyl sulfoxide (DMSO) (20%) + Н2О (80%) before and after the formation of micelles was analyzed. The response surfaces constructed with the help of the Statistica 10 software package turned out to be of prognostic value and were used to calculate the composition of the optimal matrix. An increase in the intensity of the absorption band in the premicellar region and a linear dependence in the concentration range of 5∙10–7–2∙10–4 mol/L APh were shown. The express and environmentally safe spectrophotometric method developed and tested in this study makes it possible to determine the APh content in dosage forms at the level of 0.0027%, which exceeds the maximum allowable one in paracetamol (no more than 0.01%).

Much interest is taken in identification of Aloe arborescens Mill. medicines by a major class of bioactive compounds (anthracene derivatives), as well as in quantitative determination of the compounds belonging to the class, with the same unified method for the entire spectrum from herbal drugs to herbal medicinal products. The aim of the study was to develop a methodological approach and select unified conditions for the identification and assay of anthracene-derived compounds in herbal drugs, herbal drug preparations and herbal medicinal products of Aloe arborescens Mill., observing the principle of holistic, end-to-end standardisation. Materials and methods: analytical conditions were unified using literature data and a comparison of reference values and requirements of the Russian and other pharmacopoeias. The subsequent practical evaluation of the selected conditions used three batches of the herbal medicinal product “Aloe Juice” by Vifitech (batches 131217, 030318 and 070618). The identification was carried out using a silica gel plate for high-performance thin-layer chromatography (HPTLC Silica Gel 60 HX85224933 1056330001 by Merck). The results were examined in daylight before and after spraying with a 2% alcohol solution of sodium hydroxide. The quantification was carried out using a Cary-100 spectrophotometer by Agilent at 512 nm and 0.5% magnesium acetate in methanol as a developing solvent. Results: the authors developed a unified approach to identification and quantification of anthracene-derived compounds in herbal drugs, herbal drug preparations and herbal medicinal products of Aloe arborescens Mill. and suggested using aloe-emodin as a reference standard. The authors established reference values for anthracene-derived compounds in herbal drugs, herbal drug preparations and herbal medicinal products of Aloe arborescens Mill. and drafted the corresponding monographs for the State Pharmacopoeia of the Russian Federation. Conclusions: the developed procedures allow for comprehensive identification and quantification of anthracene-derived compounds in herbal drugs, herbal drug preparations and herbal medicinal products of Aloe arborescens Mill. by the same class of biologically active compounds.

The principle of consistent, end-to-end standardisation builds upon the fact that qualitative and quantitative analysis of bioactive compounds in herbal drugs, herbal drug preparations, and corresponding herbal medicinal products involves the same methods of analysis. The possibility of such standardisation should be considered at the stage of specification development. The aim of the study was to explore approaches to analysing of Eucalyptus viminalis leaves and chlorophyllipt medicinal products, observing the principle of end-to-end standardisation. Materials and methods: the authors analysed Eucalyptus viminalis as a herbal drug (leaves), a herbal drug preparation (chlorophyllipt soft extract), and herbal medicinal products (Chlorophyllipt®, 1% alcohol solution for oral, cutaneous, rectal, and vaginal use; 2% oil solution for cutaneous and vaginal use; and 25 mg orodispersible tablets) by direct UV spectrophotometry, thin-layer chromatography (TLC), and qualitative reactions. Results: the study demonstrated the possibility of using a herbal reference standard of eucalimin to analyse the Eucalyptus viminalis herbal drug and chlorophyllipt preparations and medicinal products in different dosage forms by TLC. Conclusions: procedures for determination of total phenolic aldehydes, if included into specifications and production technology of chlorophyllipt preparations and products, ensure compliance with the principle of end-to-end standardisation and allow for quantitative determination and control of active ingredients throughout the product lifecycle: from the acceptance of raw materials, through in-process controls, to testing and monitoring of finished products’ quality.

Introduction: Anthocyanins, the polyphenolic plant pigments, have high antioxidant activity (AOA), reduce the risks of many pathological conditions in the human body. However, the wide medical and preventive use of anthocyanins is limited by their degradation during processing of plant raw materials. The objective of the work was to study the anthocyanins’ stability by spectrophotometric method during heat treatment and storage of vegetable and berry plants.Study objects and methods: Purple potatoes, eggplants, red cabbage, purple carrots, blue onions, red raspberries and blue honeysuckle were used in the study. Anthocyanins’ stability was determined by numerical values of extracts’ absorption spectra from the studied plants.Results and discussion: High stability indices of 0.623–0.986 were obtained for the anthocyanins of purple carrots, blue onion bulbs or red cabbage leaves whose main component is the antioxidant cyanidin with a relative AOA equal to 3.49. Low stability indices of 0.229-0.23 were obtained for anthocyanins of red raspberry berries and purple potato tubers containing pelargonidin or malvidin with 2.49–3.36 times lower relative AOA than for cyanidin. Within the framework of this work, the results are presented of a study of the effect of the concentrations of the stabilizer and precursor on the synthesis of silver nanoparticles. Silver nitrate was used as a precursor, and polyethylene glycol with a molecular weight from 200 to 600 Da was used as a stabilizer. The synthesis was carried out by the method of chemical reduction in an aqueous medium. The obtained samples were investigated by photon correlation spectroscopy and spectrophotometry. The presence of a characteristic absorption band at 400 nm in the absorption spectra of all samples of nanosized silver stabilized with polyethylene glycol was established, which is due to the appearance of surface plasmon resonance in metallic silver nanoparticles. It was also found that at the highest and lowest concentration of the stabilizer, equal, respectively, 0,005 and 0,1 mass. %, the formation of large silver particles with an average hydrodynamic radius from 132 to 1900 nm is observed in the reaction system. As a result, the optimal parameters for the synthesis of aggregatively stable silver nanoparticles were determined: the concentration of silver nitrate CM(AgNO3)=0,05 M and the concentration of polyethylene glycol equal to 0,01 – 0,05 %. Computer quantum-chemical modeling is carried out. It is found that the interaction of the silver atom with the terminal hydroxogroup in the polyethylene glycol molecule in the elementary act of interaction during the stabilization of silver nanoparticles by this polymer is energetically advantageous. This type of interaction is characterized by an absolute chemical hardness equal to η=0,146, and an internal energy of E=–2048,34 kcal / mol.

Today, the important problem is the creation of new decamethoxine and thiotriazoline based drugs for the treatment of stomatitis. The aim of the work is to develop a method for quantitative determination of decamethoxine and thiotriazoline in the model mixture (1:25) by spectrophotometry. Materials and methods. Series 6 model mixtures were made at the ratio of decamethoxine and thiotriazoline 1:25. We used certified substances: thiotriazoline series GTT 3460911 (manufacturer: State Enterprise Chemical Reagents Plant, Kharkiv), decamethoxine series № 010915 (manufacturer: LLC “PHARMCHIM”). Optizen POP spectrophotometer, polyvinyl alcohol, hydrochloric acid, and eosin were used. Results. A method for quantitative determination of decamethoxine and thiotriazoline in MS was developed. It was established that the content of active substances in MS is thiotriazoline from 0.5021 to 0.5096, decamethoxine from 0.0207 to 0.0211. Conclusions. A method for quantitative determination of decamethoxine and thiotriazoline in MS has been developed. The method of quantitative determination of decamethoxine and thiotriazoline in MS is reproducible.

To intensify the sorption concentration of humic substances from sodium chloride brines, the use of high-frequency ultrasound has been proposed. The optimal parameters of ultrasound were experimentally established as follows: frequency of 11.0-12.0 MHz, sound intensity of 7.5-8.5 W/cm², and duration of action not less than 2.0 min. Under the action of high-frequency ultrasound as compared to the use of low-frequency ultrasound, the degree of extraction of humic substances from brines with a concentration of sodium chloride of 150 g/l increases from 91.39-3.2% to 97.3-98.2% and from 93.2-95.0% to 98.1-99.0% on wofatit sorbent and diethylaminoethyl cellulose sorbent, respectively. In addition, the effect of high-frequency ultrasound allows quantitatively sorbing humic substances from more concentrated brines (degree of extraction ≥90%). Moreover, quantitative sorption of humic substances is possible up to a concentration of sodium chloride of 200 g/l and 250 g/l on the wofatit sorbent and diethylaminoethyl cellulose sorbent, respectively. We developed a method for spectrophotometry: determination of the content of humic substances in brines with the lower limits of determination of 0.002 mg/l and 0.001 mg/l for humic acids and fulvic acids, respectively. Introduction. The creation of new effective antibacterial drugs for the treatment and prevention of purulent-inflammatory diseases is an urgent task of modern pharmacy. Active use in the treatment of purulent infection is found by chemotherapeutic agents from the class of fluoroquinolones, which include ofloxacin.Aim. Development of a method for the quantitative determination of ofloxacin in the complex preparation "Ofloxazol".Materials and methods. For the analysis, the substance ofloxacin, titanium-containing gel "Tizol", solutions of ofloxacin on 95 % ethanol, hydrochloric acid 0.01 mol/l, ointment under the conditional name "Ofloxazol" containing 0.5 % of the drug in the gel "Tizol" were used. The study was carried out by near-UV spectrophotometry.Results and discussion. When studying the absorption spectra, it was found that for the quantitative spectrophotometric analysis of ofloxacin, it is rational to use the wavelength range of 275-320 nm (λmax = 294 nm). Statistical processing of the analysis results showed that the relative error of quantitative determination does not exceed ±1.66 %. The sensitivity of the determination of ofloxacin is 0.245 mcg/ml at A(min) = 0.02. The developed method is validated. Its specificity, linearity, correctness and precision are confirmed. According to the calibration schedule, the content of ofloxacin in the soft dosage form is determined, it is in the range of 0.0483-0.0562 g, which corresponds to the permissible deviations.Conclusion. The conducted studies allowed us to develop and propose a method for the quantitative determination of ofloxacin in the ointment "Ofloxazol", obtained on a titanium-containing basis. The method allows you to evaluate the quality of manufacturing the dosage form, including setting the content of the drug with an error that does not exceed the standard deviations.

Introduction. Indolocarbazole derivatives are of increasing scientific interest for practical oncology. A number of N-glycosides, indolo[2,3-a] carbazole under the laboratory code LCS, were synthesized in the laboratory of chemical synthesis of the National Medical Center of Oncology named after N.N. Blokhin. Currently, one of the most promising compounds in this class is LCS-1208, a representative of the arabinoside class of indolo [2,3-a]pyrrolo[3,4-c]carbazole-5,7-dione. According to the mechanism of biological action, LCS-1208 is a protein kinase C inhibitor and is of great interest for the treatment of malignant neoplasms.Aim. Chemical and pharmaceutical standardization of the pharmaceutical substance LCS-1208.Materials and methods. Laboratory samples of pharmaceutical substance LCS-1208. Methods of investigation: gravimetry, spectrophotometry, polarimetry, high-performance liquid chromatography (HPLC), high-resolution nuclear magnetic resonance (NMR) spectroscopy and infrared (IR) spectroscopy.Results and discussion. The quality assessment of LCS-1208 was carried out according to the indicators adopted in the XIV edition of the State Pharmacopoeia of the Russian Federation for quality control of pharmaceutical substances. LCS-1208 - orange amorphous powder, odorless; soluble in dimethylsulfoxide (DMSO) and dimethylformamide (DMF); very slightly soluble in 95 % ethyl alcohol and practically insoluble in water. The authenticity of the substance is confirmed by NMR and IR spectra, as well as electronic absorption spectra. The values of the specific optical rotation of LCS-1208 (1 % solution in DMF) are placed in the range from +58° to +61°. All the studied samples of the substance were free of inorganic impurities, sulphate ash, heavy metals and contained no more than 1.0 % water, determined by the K. Fischer titration method. The content of possible related impurities in the substance LCS-1208 and the content of the main active substance were determined by HPLC. The studied laboratory series of the pharmaceutical substance LCS-1208 contained no more than 1.0 % of any single and no more than 3 % of the total unidentified impurities. The content of the main active substance was more than 97 %.Conclusion. As a result of the work carried out, quality criteria and parameters were selected and methods for their determination were developed, which allow to adequately assess the quality and standardness of the pharmaceutical substance LCS-1208.

Arbutin is the main active ingredient in many herbal medicinal products that have diuretic, antimicrobial, bactericidal, and antioxidant effects. Many of these products are mixtures of different herbal substances. Therefore, the approaches to quality control of HMPs can vary significantly. The aim of the study was to compare arbutin assay procedures used in quality control of arbutin-containing products. Materials and methods. Samples of the following HMPs were used in the study: monocomponent Bearberry Leaf and multicomponent Brusniver® (herbal mixture, powder). The test methods used were titrimetry, ultraviolet-visible spectrophotometry, and high-performance liquid chromatography (HPLC). Results. The authors compared five arbutin assay procedures described in the monographs and product files for arbutin-containing HMPs. Conclusions. It has been established that the analysed procedures cannot be used interchangeably as equivalent test methods; the limits for arbutin have to be established for each specific procedure. Iodometric titration is the most labour- and time-consuming method, and the determined titration endpoint is a subjective assessment. Spectrophotometric methods do not require the use of an arbutin reference standard, but they can give overestimated results as compared to the HPLC and titrimetry methods. HPLC methods are more selective, but they require the use of reference standards. The recommended test methods for HMP quality control are HPLC and visible spectrophotometry; the titrimetric method is recommended for replacement.

Relevance. If we reduce the treatment exclusively to the eradication of Helicobacter pylori (Hp), the question of monitoring the results of anti-Hp therapy is controversial, given the widely known data on the detection of Hp, including virulent strains of Hp, in most healthy individuals.The aim of the study: to track the dynamics of stomach colonization with various Hp strains in patients with gastric ulcer and duodenal ulcer (PUD) immediately after the use of standard three-component anti-Hp therapy (AHT) and 1.5–2 months after AHT.Methods. Genotyping of Hp strains was carried out by the VNTR method together with the determination of the cagA gene.Results. Assessment of the results of AHT in the form of «eradication of Hp — re-detection of Hp» (i. e., without taking the determination of Hp strains into account) showed less reliability in the differences than the «eradication — preservation of the strain or change of the strain» score in patients with PUD; in addition, the differences between the initial bacteriological picture and that observed immediately after AHT were on the verge of reliability, while the differences between the initial bacteriological picture and that observed after 1.5–2 months had a high degree of reliability. The number of eradications increased (eradication achieved immediately after AHT was not preserved, except for one) and the number of cagA-containing strains decreased (due to new eradication detected at late follow-up periods and due to a change in strains) in long-term follow-up in patients with PUD.Conclusion. Since all patients with PUD achieved clinical remission, which lasted for the next 1.5–2 months, the success of AHT should not be unambiguously associated with the eradication of Hp; the restoration of the organism's colonization resistance to Hp after a course of therapy is more likely.

Introduction. Natural dye beta-carotene E160a (pro-vitamin A) is part of many functional foods. Therefore, the total intake of beta-carotene must not exceed the tolerable upper intake level, i.e. 10 mg/day. E160a is also used in formulations of complex food additives. These products contain antioxidants, emulsifiers, preservatives, carriers, stabilizers, and thickeners, which makes it make difficult to isolate and determine the amount of beta-carotene. Complex food additives vary in composition, which, together with incomplete extraction and degradation of beta-carotene, results in inaccuracy of its determination. The present research reviewed various techniques of beta-carotene extraction from liquid and dry complex food additives. Study objects and methods. The study featured commercial samples of complex food additives containing beta-carotene. It was isolated from the samples by liquid extraction methods using organic solvents and their binary mixtures. The extraction was performed in a stirring device at 120 rpm and in a Bandelin Sonorex ultrasonic bath at 128 W and 35 kHz. The extraction continued until the samples were completely discolored. The list of solvents included chloroform, petroleum ether, hexane, acetone, ethanol, and their mixes. The experiment was based on high performance thin layer chromatography with Sorbfil sheet plates. The optical density was determined by spectrophotometry using a SHIMADZU UV-1800 device. Results and discussion. The extraction of beta-carotene from liquid samples with mechanical stirring at 120 rpm led to the formation of stable opaque emulsions, which made it difficult to determine the amount of beta-carotene. The emulsions were destroyed by ultrasound exposure for 30 min. The degree of extraction of beta-carotene from liquid emulsion systems (≥ 95%) was achieved by ultrasonic extraction with chloroform or a binary extractant based on mixes of acetone with chloroform, hexane, or petroleum ether in volumetric ratios of 1:1. When extracted from dry samples using chloroform or binary extractants, beta-carotene was not isolated or was not completely isolated. As a result, pre-dispersion of dry samples in water with stirring at 120 rpm was proposed. Ultrasonic treatment did not contribute to the dispersion of dry samples due to insufficient cavitation action. Ethanol, which is a solubilizer, increased the extractability of beta-carotene up to 98 %. The optimal results were achieved by adding a binary extractant (ethanol:chloroform in a 1:2 ratio) to an aqueous solution of the dry sample (1:3). The optimal elution systems were hexane:benzene (9.7:0.3), butanol:methyl ethyl ketone:ammonia 10 % (9:6:6), and petroleum ether:benzene:acetone:acetic acid (23.3:5.8:0.6:0.3). Conclusion. The article offers new methods of beta-carotene isolation from liquid and dry complex food additives of various composition and elution systems for carotene identification. The methods caused no degradation of beta-carotene during extraction and can be used to prepare samples of complex food additives for beta-carotene identification and quantitative analysis.

Centre of Isotopic and Geochemical Research based on the Analytical Department of Vinogradov Institute of Geochemistry SB RAS (Irkutsk) performs a wide range of analytical studies to solve mineralogical and petrological, geochemical, prospecting, ecological, paleoclimatic and applied problems. The studies are supported by the modern equipment for electron microprobe, X-ray diffraction, X-ray fluorescence, atomic emission and mass spectrometric (including isotope) analyses, as well as the necessary international certified reference materials (SRM) and a collection of SRM of the natural and technogenic composition of our production.

Three unified procedures have been developed. The possibility of quantitative determination using the methods of kinetiс spectrophotometry, voltammetry, and redox titration of ampicillin (Amp) as a pure substance and in a drug using potassium hydrogen peroxomonosulfate (KHSO5) as an analytical reagent has been shown.

Complex formation in the ternary systems copper(II)/nickel(II)/cobalt(II) – isonicotinic acid hydrazide – L-histidine was studied by the methods of spectrophotometry and mathematical modeling in an aqueous solution with 0.1 mol dm–3 KNO3 as a background electrolyte. The compositions, formation constants, and spectral parameters of the heteroligand complexes with a metal/hydrazide/amino acid ratio of 1:1:1 were determined. It was found that the heteroligand complexes with the neutral form of isonicotinic acid hydrazide have higher stability values than those with the protonated form. The stability of bis- and tris-complexes of the same composition in the series copper(II) – nickel(II) – cobalt(II) is in agreement with the Irving–Williams order. Three isomers of the heteroligand complex of nickel(II) with the protonated form of isonicotinic acid hydrazide and histidine zwitterion were optimized by the method of molecular mechanics. In the system with cobalt(II), a reversible interaction with atmospheric oxygen was revealed.

Abstract. Papaverine hydrochloride products are used as anticonvulsants in routine medical practice. Most of the approved product specification files include thin-layer chromatography for assessment of product-related impurities and UV spectrophotometry for determination of active pharmaceutical ingredients. An HPLC assay is not used for determination of papaverine hydrochloride in drug dosage forms.The aim of the study was to develop an HPLC test method for determination of product-related impurities and for quantification of papaverine hydrochloride in solutions for injection, tablets, and rectal suppositories.Materials and methods: samples of the following Russian-made papaverine products were used in the study: Papaverine, solution for injection, 20 mg/mL; Papaverine, rectal suppositories, 20 mg; Papaverine, tablets, 40 mg. The Agilent 1260 Infinity II DAD System was used for the HPLC assay, and the Agilent 8453Е UV-Vis System was used for recording UV spectra. The determination of product-related impurities and the assay of active ingredients were performed simultaneously by HPLC using a reversed-phase column Kromasil 100-5-C18, 250×4.6 mm, 5 μm, the gradient elution mode, and detection at 238 nm. Papaverine Hydrochloride USP RS, 99% purity, and Noscapine EP CRS were used as reference standards.Results: the study demonstrated that determination of product-related impurities and assay of active ingredients in papaverine products can be performed simultaneously using HPLC.Conclusions: the authors proposed an HPLC test method for determination of active ingredients in papaverine products, which is aligned with the “consistent standardisation” principle and can be recommended for inclusion into draft monographs for papaverine products.

Study of the qualitative composition and quantitative content of some groups of BAS in dietary supplements with green tea leaf extract Recently, there has been a high demand from the population for dietary supplements that have antioxidant activity. First of all, these are dietary supplements with green tea leaf extract, because green tea contains catechins in a fairly high amount. Nowadays, according to the current legislation of Ukraine the qualitative composition and quantitative content of biologically active substances in dietary supplements are not established, which to a certain extent affects the quality and safety of these products. The aim of the study was to determine the qualitative composition and quantitative content of some groups of BAS in dietary supplements with green tea leaf extract. Materials and methods. Three dietary supplements from different manufacturers “Green Tea Extract”, “Green Tea Extract”, “Green Tea” were chosen for the study. Qualitative analysis was performed by thin layer chromatography, for quantification spectrophotometry in the UV-region was used. Results. The presence of catechins was found in the studied dietary supplements, caffeine was found only in trace amounts. The total content of catechins is 146.80 ± 1.36 mg, 79.00 ± 0.88 mg, 28.00 ± 0.75 mg, and in terms of green tea extract, which is stated in dietary supplements for dietary supplements “Green Tea Extract”, “Green Tea Extract”, “Green Tea”, respectively. Conclusions. Qualitative and quantitative analysis of some groups of BAS of three dietary supplements with green tea leaf extract was performed. It was found that dietary supplements “Green Tea Extract” and “Green Tea” meet the requirements of the US Pharmacopoeia 38 in terms of catechins. Based on the study, it can be concluded that the problem of compliance with dietary supplements is relevant today and requires the introduction of regulatory documentation for the detection and determination of biologically active substances in dietary supplements.

In this work, for the first time the results of studying the microstructure of manganese dioxide nanoparticles stabilized with sulfur-containing amino acids are presented. According to the results of scanning electron microscopy, nanoparticles with a diameter of 15-30 nm were presented in the samples. The optimal mass ratios of precursors were established by means of spectrophotometry, to obtain stable sols of manganese dioxide nanoparticles. The optimal mass ratios were: KMnO4 : methionine – 1:1 and KMnO4 : cysteine – 3:1. Investigation of the effect of the medium active acidity on the stability of manganese dioxide nanoparticles showed that MnO2 samples obtained with methionine are stable at 4≤pH≤11, and with cysteine – at 5≤pH≤8.

The results of theoretical and experimental studies of surface properties of nanostructured films based on tin dioxide using Fourier-transform infrared spectroscopy, ellipsometry and the indicator method of the distribution of adsorption centers are presented. Etching in inductively coupled plasma was used for additional post-processing of nanostructured films in order to activate the adsorption centers.

Introduction. Currently, more than 600 thousand people a year are affected by malignant tumors on the territory of the Russian Federation. Despite the large selection of antitumor drugs and the variety of mechanisms of their action, the effectiveness of existing drugs continues to be insufficient. The main disadvantages of most antitumor drugs are the emergence of tolerance to them of tumor cells, a limited range of action and high toxicity. In this regard, the creation of effective original domestic antitumor drugs still remains relevant. Among the numerous natural and synthetic heterocyclic compounds that exhibit antitumor activity, indolo[2,3-a]carbazole derivatives that can initiate various pathways of tumor cell death are of increasing interest. The main targets for their action are topoisomerases and protein kinases, which play an important role in the processes of replication, transcription, repair or recombination of deoxyribonucleic acid (DNA). Due to this, in addition to antitumor activity, this group of compounds shows antibacterial, аntiprotozoal and immunomodulatory activity, which makes them a very promising class of candidates for the creation of new drugs.Text. The purpose of this review is to discuss instrumental methods for qualitative and quantitative analysis of indolo[2,3-a]carbazole derivatives used in the world's leading pharmacopoeias. These methods can be used both in pharmacokinetic studies, and in the standardization of these compounds, in the form of pharmaceutical substances, or as part of drug forms.Conclusion. To further introduce a new group of antitumor drugs based on indole[2,3-a]carbazole derivatives into medical practice, a deep and thorough study of their physical and chemical properties is necessary. Justification and development of analysis methods allow us to develop methods applicable to pharmacokinetic studies, as well as to create regulatory documents for quality control and standardization of indolo[2,3-a]carbazole derivatives as pharmaceutical substances. A study of the literature that describes methods for analyzing indolo[2,3-a]carbazole derivatives indicates that spectrometric (infrared and ultraviolet spectrometry) and chromatographic (thin-layer chromatography and high-performance liquid chromatography) methods are most often used to determine the authenticity and quantitative analysis of these compounds.

Objectives. The Monarda fistulosa herb of the Lamiaceae family is particularly interesting among essential oils from medicinal plants that have a wide spectrum of pharmacological activities. However, information regarding some of its flavonoids, which are found in the essential oil, is controversial. Inaccuracies in identification of the chemical composition of the herb have led to several different standardization approaches, which are cumbersome. To establish a uniform classification, here, we present confirmation for new approaches for the standardization of the Monarda fistulosa herb. Methods. Silica gel column chromatography was used to extract the flavonoids. Identification was based on ultraviolet spectroscopy, nuclear magnetic resonance spectroscopy, mass spectrometry, and acidic hydrolysis. The quality of the proposed quantitation methodology for total flavonoids was assessed by differential spectrophotometry at 394 nm, in isorhoifolin equivalent.Results. We have verified new approaches for the standardization of the Monarda fistulosa herb. The approaches can determine the authenticity of the herb by detecting monoterpene phenols and flavonoids that have diagnostic value. We also developed a technique for quantitation of the total flavonoids. Conclusions. We investigated the possibility of establishing the authenticity of the Monarda fistulosa herb based on the diagnostically significant flavonoids, isorhoifolin and linarin.

Lipid metabolism is crucial in physiology. In recent decades the model object Drosophila melanogaster has been actively used in the study of the fundamental issues of lipid metabolism and its disorders, including obesity, as well as in the search for therapeutic goals for the treatment of metabolic disorders in humans. Quick and accurate quantification of lipid content is an important step in solving these problems. For the first time the method of quantitative measurement of total lipids with the use of the sulfophosphovanillin (SPV) method was described by Zöllner and colleagues in 1962, and adapted for insects by Van Handel on females of the yellow fever mosquito Aedes aegypti. The advantages of this method compared to traditional gravimetric and chromatographic methods of analysis are the use of a small amount of biological material, lack of need for complex manipulations with the sample, its high sensitivity, reproducibility and simplicity of implementation with a minimum set of equipment. Here, a modification of the Van Handel protocol is described, which allows the method to be adapted for quantitative determination of total lipids for various organisms as exemplified a widely used model, D. melanogaster. To test the effectiveness of the modified method, we measured the content of total lipids in D. melanogaster females carrying hypomorphic mutations of the dilp6 and dfoxo insulin signaling pathway genes compared to the wild-type Canton-S line, and showed that dilp6 took part in the regulation of fat metabolism, while dfoxo did not. The results obtained emphasize the effectiveness of the colorimetric method with the use of SPV reaction and spectrophotometry for the quantitative analysis of total lipids.

Specialists in pharmacognosy are facing an important task of improving quality control methods for combination herbal medicinal products in order to enable reliable assessment of the content of active substances that are responsible for the drug’s pharmacological effect.The aim of the study was to investigate the possibility of using the European Pharmacopoeia method to determine the total content of anthracene derivatives in the antihaemorrhoidal medicinal herb mixture Proctophytol® and to propose limit values for the total content of anthracene derivatives.Materials and methods: individual mixture components containing anthracene derivatives, such as senna leaves and frangula bark, as well as model mixtures containing these individual components in a 1:1 ratio, and a model mixture imitating Proctophytol® were used in the study. The determination of the anthracene derivatives content was carried out using spectrophotometry.Results: it was demonstrated that the European Pharmacopoeia method could be used to assess the quality of Proctophytol® in terms of anthracene derivatives total content. The spectrophotometric method described in the European Pharmacopoeia has advantages over the methods described in manufacturer specifications for Russian products, because it allows for thorough extraction of the active substances and is standardized for anthracene derivatives.Conclusions: the European Pharmacopoeia method was adjusted to determine anthracene derivatives in the medicinal herb mixture Proctophytol®. It was proposed to use 70% ethanol instead of 70% methanol as extraction solvent. The authors identified optimum sample weights and test solution dilutions, and calculated and verified the limit for anthracene derivatives content—“Not less than 1.9%”. The adjusted method can be recommended for inclusion in the monographs of the State Pharmacopoeia of the Russian Federation for medicinal herb mixtures similar to Proctophytol®.

The behavior of cephalexin in pharmaceutical and biological media was studied by spectrophotometric method. The ranges of linearity and the limits of cephalexin detection were determined. The possibilities of spectrophotometric cephalexin determination in mixed saliva and in blood serum were shown. Optimal conditions of proteins precipitation were revealed. Pharmacokinetic parameters of cephalexin in oral fluid of patients with sinusitis were determined.

The purpose of the work was to develop of a method for the quantitative determination of piperidinium 2-[5-(2-furyl)-4-phenyl-1,2,4-triazol-3-ylthio] acetate (trifuzol) as part of a dosage form – an effervescent intrauterine suppository using spectrophotometry in the ultraviolet region spectrum and its validation according to State Pharmacopoeia of Ukraine.Materials and methods. The study was used a working standard sample of trifuzol, intrauterine effervescent veterinary suppositories, 1.0 g of trifuzol, as a solvent – purified water. Analytical equipment: Specord 200 spectrophotometer, electronic scales ABT-120-5DM, ultrasonic bath ELMASONICE 60 H, class A measuring dishes. Method of spectrophotometric analysis was used.Results. A spectrophotometric method has been developed and validated for the quantitative determination of trifuzol in a dosage form – an intrauterine effervescent veterinary suppository, based on measuring the absorption of an aqueous solution of the drug at 278 nm. The methodology tally the requirements of State Pharmacopoeia of Ukraine for such validation characteristics as specificity, linearity, precision, correctness, and robustness. The analysis of the predicted total uncertainty of the analysis was showed the reproducibility of the method and the possibility of its application in other laboratories.Conclusions. A method for the quantitative determination of trifuzol in the composition of the dosage form, an effervescent intrauterine suppository, according to the requirements of State Pharmacopoeia of Ukraine, was developed and validated. It was proved that according to such validation characteristics as linearity, specificity, precision, correctness, and robustness, the technique is correct.

The quality of impregnation of textile materials with additive preparations significantly affects their appearance and performance properties. The impregnating properties of aqueous dispersions of polymers depend on their physicochemical characteristics and particle size of the dispersed phase. The process of preparing the aqueous dispersion of styrene acrylate in the medium of ultrasonic vibrations has been studied. Research results have proved the effectiveness of applying sounding to impregnation solutions to improve the quality of impregnation of hydrophobic textile materials.

The aim of this research is the development and validation of spectrophotometric methods for the determination of acetylcysteine and streptomycin sulfate, suitable for the standardization of test substances and analysis dosage forms. Materials and methods. The substances of the acetylcysteine and the streptomycin sulfate, the quinone derivatives (the solutions of sodium 1,2-naphthoquinone-4-sulfonate and 2,3-dichloro-1,4-naphthoquinone), as the analytical color reagents, were used for the experiment. The spectrophotometer Specord 200 for measuring the optical density was applied. Results. According to experimental data, the optimal conditions between the test substances and the added reagents were established. The factors affecting the character of the absorbance spectrum and the value of absorbance: the nature of the solution medium, the amount of added reagents, pH of the reaction mixture, conduction period of the reaction and reaction product resistance in time have been studied for the development of a methods of quantitative determination of acetylcysteine and streptomycin sulfate on based reactions with quinone derivatives. It was experimentally established that the sodium 1,2-naphthoquinone-4-sulfonate with streptomycin sulfate in the alkaline medium to form a colored composition with maximum light absorbance at 560 nm. The solution of 2,3-dichloro-1,4-naphthoquinone with acetylcysteine in the DMFA medium to form a colored composition with maximum light absorbance at 425 nm. It was necessary to heat it on the water bath for 10 min at 95 °С for more complete conduction period of reaction. According to SPU, the procedure of validation was determined. It has been proved that the developed techniques of the quantitative determination based on such characteristics as linearity, accuracy, validity, application range and robustness. The obedience to the Beer’s Law is 4.48–8.40 mg/100 ml for acetylcysteine and 2.00–8.00 mg/100 ml for streptomycin sulfate. Stoichiometric ratios of the reaction mixture component have been established by means of the method of isomolar series and continuous change method. Conclusion. As a result of this work, the optimal reaction conditions for the reaction of acetylcysteine with 2,3-dichloro-1,4-naphthoquinone and streptomycin sulfate with sodium 1,2-naphthoquinone-4-sulfonate have been established. Based on the obtained data, new spectrophotometric methods for the quantitative determination of the investigated drugs were developed and tested on the dosage forms. It is proved that the developed methods are simple, reproducible and expressive.

Plants of Cannaceae family is a popular decorative and leafy plant with bright flowers and large leaves. According to the literature they contain amino acids, free carboxylic acids, flavonoids, anthocyanins, terpene and steroid compounds, cardioglycosides and alkaloids. They exhibit a wide range of biological activity, including antioxidant, antimicrobial, anti-inflammatory, cardioprotective, hemostatic, antihypertensive and anti-ischemic activity. The aim of the research was to study the qualitative composition and to determine the quantitative content of chlorophylls and carotenoids in herb of the Canna hybrida Suevia, Liberty, Livadia, Artek and Dark Night varieties. Materials and methods. The qualitative composition of photosynthetic pigments of herb of the Canna hybrida varieties Suevia, Liberty, Livadia, Artek and Dark Night differed not significantly by thin-layer chromatography method. In the analyzed objects the content of chlorophyll a, chlorophyll b, carotenoids and lycopene was determined by absorption spectrophotometry method. Results. At least 10 compounds appeared on the chromatograms of the studied samples, at least 4 of which were attributed to carotenoids and not less than 6 – to chlorophylls. It was established that the content of the total of chlorophylls in herb of the Canna hybrida Liberty, Artek, and Dark Night varieties exceeded the content of the carotenoids by approximately 5 times, in herb of the Canna hybrida Suevia and Livadia variety – almost 8 times. Approximately 65 % of the total content of chlorophyll was chlorophyll a, 35 % was chlorophyll b, which was characteristic of herb of the Canna hybrida Suevia, Liberty, Artek and Dark Night varieties. In herb of the Canna hybrida Livadia variety a fundamentally opposite situation was observed. The total content of chlorophyll b was in 2.3 times higher than the content of chlorophyll a. The highest content of chlorophyll a (1661.76 ± 41.54 mg/kg), chlorophyll b (943.79 ± 23.60 mg/kg) and lycopene (62.37 ± 1.56 mg/kg) was observe in herb of the Canna hybrida Suevia variety. The equally high content of carotenoids – 373.75 ± 9.34 mg/kg, 381.84 ± 9.55 mg/kg and 386.05 ± 9.65 mg/kg, was watched in herb of Canna hybrida Suyeviya, Liberty and Artek varieties respectively. Conclusions. The obtained results will be used in the development of quality control methods for herb of Canna hybrida and medicinal plant products on their basis.

Saturated enzymes are an integral part of the technology of soft cheeses, which means that the market requires a significant amount of enzyme preparation, as recent studies have shown that consumption of whey cheese in Ukraine is increasing. An important aspect is that almost all industrial milk processing companies use a microbial analogue of the sifted enzyme, since it is significantly cheaper than natural, but some producers use natural enzymes principally and this trend is increasing. The purpose of our work was to determine the rates of extraction of pygus enzymes from calf sichugas for the use of different ratios of organic (dairy) and inorganic (hydrochloric) acids in the extractant. he research was conducted in 2 stages. At the first stage, the extraction of enzymes from the homogenate of the sichugas was studied for the use of extractants with different content of hydrochloric acid. In the second stage of the research, the effect of the presence of a solution of lactic acid in the extract on the parameters of elimination of enzymes from the sichugum homogenate was detected. The highest concentration of protein 89,8 mg/l was the fifth experimental group, where 0,25 cm3 of lactic acid solution was added, which was 34,5 % higher than the similar group from the 1 st stage of the research.
