For this, strains with several gene deletions and genetic constructions for control of SulA level were created. Standard methods of molecular biology, conventional fluorescence microscopy, super-resolution microscopy, and semi-quantitative Western blotting for concentration measurements were used.During the work an indirect method was used to estimate the concentration of the SulA protein in cells during the SOS response. The results indicate that the SulA concentration was several times lower than the FtsZ concentration in the same cells. In addition, the role of Z-ring positioning systems in the cell division recovery was studied and it was shown that the Min-system plays a key role in this process. These results indicate that the sequestration model based on in vitro experiments does not explain the process of division inhibition by SulA in vivo. Also, new knowledge about the role of positioning systems in the cell division recovery could help to understand how bacteria are able to survive under adverse conditions, such as antibiotic therapy.
