Hyperprolactinemia has multiple etiologies and is the most common endocrine manifestation of pathology of the hypo­thalamic-pituitary axis. Hyperprolactinemic hypogonadism is of great scientific interest due to the prevalence of endocrine pathology in the reproductive period and its effect on metabolic processes. Hyperprolactinemia is one of the serious obstacles to the implementation of reproductive function in both women and men. The article discusses current issues of diagnosis and management tactics for patients with hyperprolactinemia of various etiologies within the framework of preconception preparation.

Rapid changes in the morphological characteristics of the population, which began in the middle of the last century, are due to changes in the habitat. The “obesogenic” environment that is formed in urbanized spaces contributes to physical inactivity and a hypercaloric diet, and the processes of globalization contribute to obesity in the population in all countries of the world. From the obesity epidemic declared by WHO in 1988, humanity has moved to an obesity pandemic. The presented review analyzes global and Russian trends in the development of obesity. Data on the prevalence of latent obesity and diagnostic criteria for this condition are presented. An analysis of data on the dynamics of BMI in the world and in the Russian Federation was carried out. Various options for the topography of fat deposition and their relationship with comorbid diseases are considered. The structure of the subcutaneous fat depot in the abdominal area and the influence of the outer (superficial subcutaneous abdominal adipose tissue, SSAT) and deep (deep subcutaneous abdominal adipose tissue, DSAT) layers of subcutaneous fatty tissue in the abdominal area on the risks of developing diseases are discussed. The results of assessing the diagnostic value of the T/A substitution in the FTO gene (rs9939609) for predicting the proportion of fat mass and abdominal fat deposition in adults are presented.

It has now been shown that overweight and obesity are associated with the development of at least 13 types of cancer, including cancer of the breast, colon and rectum, endometrium, esophagus (adenocarcinoma), gallbladder, stomach, kidney (renal cell), liver, ovaries, pancreas and thyroid glands, as well as multiple myeloma. Obesity-related cancers account for about 40% of all cancers. Thus, obesity is ahead of smoking as the most common modifiable risk factor for carcinogenesis. This review collects the latest data on the impact of obesity phenotypes on the risk of developing cancer, including a discussion of the contribution of the metabolically healthy obesity phenotype (MHO) to tumorigenesis. Possible mechanisms contributing to the emergence and progression of cancer are analyzed, for example: stimulation of cell proliferation, evasion of growth suppressors, inhibition of apoptosis and provision of replicative immortality, induction of angiogenesis, activation of invasion and metastasis, influence on genome instability, as well as chronic low-grade inflammation that contributes to the development of cancer. Understanding how obesity influences the above mechanisms may facilitate the development of new cancer prevention and treatment strategies for obese populations.

We recommend acromegaly to be ruled in all patients with characteristic changes in appearance (A3). In all patients without characteristic changes in appearance, we recommend to rule out acromegaly, if several clinical signs suspicious for acromegaly are present (B3). We recommend acromegaly to be ruled out in all patients with pituitary adenoma (B3). We recommend to rule out acromegaly in all patients with prolactin-secreting adenomas (C4). For laboratory confirmation of acromegaly, we recommend to measure serum insulin-like growth factor-1 (IGF-1, somatomedin C) level (A3). In patients without obvious clinical signs of acromegaly and/or with a moderate increase in IGF-1 levels (IGF-1 index less than 1.3), we recommend to assess the response of somatotropic hormone (GH) to hyperglycemia (GH during an oral glucose tolerance test) (B3). In all patients with biochemically confirmed acromegaly, magnetic resonance imaging of the pituitary gland with intravenous contrast is recommended to assess pituitary adenoma size, location and growth pattern (A3). We recommend to measure blood glucose and glycated hemoglobin in all patients with confirmed acromegaly (B3). We recommend to measure serum prolactin levels in all patients with acromegaly (B3).In all patients with acromegaly and pituitary adenoma, transnasal transsphenoidal adenomectomy is recommended as first-line treatment if the patient’s consent is given and there are no contraindications (A1). For all patients with acromegaly for whom neurosurgical treatment is indicated, surgical intervention is recommended in medical centers, specializing in pituitary diseases, by neurosurgeons who perform at least 50 such operations per year (A3). We recommend that morphological and immunohistochemical examination of the removed pituitary adenoma is performed in all patients with acromegaly (A3). We do not recommend routine medical therapy for acromegaly before surgery as a mean to reduce tumor size (B2).For patients with acromegaly in whom neurosurgical treatment is not indicated or ineffective or while patient awaiting for the effect of radiation therapy, we recommend medical therapy (A1). Long-acting first-generation somatostatin analogues are recommended as first-line drug therapy for acromegaly. In case of partial/complete resistance to or intolerance to somatostatin analogs, pegvisomant is recommended as a second-line medical therapy for acromegaly (A1). We recommend starting pegvisomant from a single dose of 40–80 mg per day subcutaneously, then continue injections at a starting dose of 10 or 15 mg per day with IGF-1 monitoring after 4–6 weeks and dose adjustment if necessary (B2). In patients with acromegaly and a moderate increase in IGF-1 levels, we recommend to initiate cabergoline as medical monotherapy or, if somatostatin analogs are ineffective, in combination with them (B2).In patients with acromegaly, if the disease remains active after surgical treatment, if drug treatment is unavailable, intolerable or ineffective, and repeated neurosurgical intervention is inappropriate, it is we recommend radiation therapy (A1). A multidisciplinary approach is recommended in the management of acromegaly complications (C3).

BACKGROUND: Obesity is a pathology that highly contributes to the development of type 2 diabetes mellitus and cardiovascular diseases. Meanwhile, different obesity phenotypes have varying levels of risk, and the indication of early predictors that can help determine the likelihood of developing cardiometabolic disorders is an important task. Since obesity is characterized by excessive energy intake, hormones involved in energy homeostasis (leptin and other adipokines, ghrelin, incretins) can be considered as serious candidates for the role of such predictors. The profile of changes in their level in the post-­nutrition status testifies in favor of the development of resistance not only to insulin, but also to leptin, and to ghrelin, and, possibly, to incretins.AIM: This study aims to investigate the relationship between the dynamics of the level of hormones involved in energy homeostasis in a sample with a standard carbohydrate breakfast and metabolic health.MATERIALS AND METHODS: Metabolic parameters (including НОМА-IR and НОМА-B, lipid profile, glucose level, blood pressure) were assessed in 90 obese patients (BMI 37.31±4.83 kg/m2). Additionally, biomarkers such as adiponectin, CRP were measured. Levels of leptin, ghrelin, GLP1 and GIP were assessed before and 60 minutes after a standard carbohydrate breakfast. These hormone levels were classified depending on the nature of the changes, considering the physiological dynamics described in the literature in healthy people or not. Patients were divided into groups of metabolically healthy and ­metabolically unhealthy obesity, first using the most commonly known criteria (Meigs et al.), then based on the criteria discussed in the literature for perfectly healthy obesity, in which there are no metabolic disorders.RESULTS: The analysis revealed that patients without metabolic disorders had a ghrelin level profile similar to that in healthy people without obesity, while patients with at least one of the manifestations of the metabolic syndrome showed evidence of ghrelin resistance (the level of ghrelin on an empty stomach was reduced, and after food it was not adequately reduced). Adiponectin levels were normal in metabolically healthy patients. Incretin levels showed unidirectional changes in obesity, regardless of metabolic status. The level of CRP did not differ between the groups. The glycemic level was significantly lower in metabolically healthy patients (the group was characterized by normal insulin levels and had a lower degree of insulin resistance).CONCLUSION: The data obtained allow us to consider ghrelin resistance as an early and rather sensitive indicator of the development of metabolic disorders in obesity.

BACKGROUND: Recent clinical descriptions have shown that in adult patients, the cause of nondiabetic hypoglycemia (NDH) may be various genetically determined disorders of glucose metabolism or insulin synthesis/bioavailability. In this connection, in adult patients with NDH of unclear genesis, it is important to conduct a genetic study in order to search for mutations in genes associated with congenital disorders of glucose metabolism (CDGM).AIM: To evaluate the effectiveness of genetic testing to exclude CDGM in adult patients with idiopathic NDH.MATERIALS AND METHODS: Based on the analysis of the literature, a targeted panel has been developed, including 30 genes, mutations in which are associated with the following groups of diseases: 1) congenital hyperinsulinism (KCNJ11, ABCC8, GLUD1, HADH, UCP2, HNF4A, HNF1A, GCK, INSR, SLC16A1); 2) glycogen storage diseases (AGL); 3) other carbohydrate metabolism disorders (ALDOB, FBP1); 4) glycosylation defects (PMM2, ALG3, PGM1, MPI); 4) defects in fatty acid oxidation (ACADM, ETFA, ETFB, ETFDH, FLAD1, SLC25A32, SLC52A1, SLC52A2, SLC52A3); 5) disorders of ketone body metabolism (CPT1A, CPT2, HMGCL); 6) mitochondrial disorders (DLD). Twenty nine patients (n=29: with idiopathic NDH n=17 and with insulinoma n=12) aged 19 to 66 years underwent a genetic study using this custom panel.RESULTS: As a result of the examination 12 genetic variants (all heterozygous) were identified in 8 patients with idiopathic NDH (47%, 95% CI (23%; 72%)), at that two mutations were detected in three patients: in the genes AGL and HMGCL; ACADM and FLAD1, respectively; and one patient had three mutations: one mutation in the ETFA gene and two mutations in the ABCC8 gene. Frequencies of genetic variants: AGL — 18%, 95% CI (4%; 43%), ETFA — 12% (1%; 36%), HMGCL — 6% (0%; 29%), ALDOB — 6% (0%; 29%), CPT1A — 6% (0%; 29%), ABCC8 — 6% (0%; 29%), ACADM — 6% (0%; 29%), FLAD1 — 6% (0%; 29%). 5 genetic variants (all heterozygous) were identified in 5 patients with insulinoma (42%, 95% ДИ (15%; 72%)). Frequencies of genetic variants: ABCC8 — 17%, 95% CI (2%; 48%), HNF1A — 8% (0%; 38%), ETFDH — 8% (0%; 38%), MPI — 8% (0%; 38%). We did not include benign variants in this analysis. At the same time, clinically significant variants were identified only in one patient from the group with idiopathic NDH (6%, 95% CI (0%; 29%)) in the ABCC8 gene and in one patient from the group with insulinoma (8%, 95% CI (0%; 38%)) in the same gene congenital hyperinsulinism (CHI).CONCLUSION: A panel of 30 genes has been developed, variants of which are associated with a CDGM. The results of our study confirm the possibility of detecting CDGM in adulthood, in particular CHI, and indicate the need for genetic testing, primarily in patients with idiopathic NDH.

AIM: The purpose of this study was to analyze the characteristics of management, diagnosis and treatment of patients with vitamin D deficiency/ by endocrinologists in the regions of Russian Federation using a sociological survey.MATERIALS AND METHODS: A cross-sectional sociological uncontrolled study was carried out. To implement this work, we used an online questionnaire “Questioning doctors on vitamin D” specially developed on the basis of the Google forms platform. The study was conducted in January 2021.All the data obtained were entered into an electronic database in MS Excel. Statistical processing was performed using the IBM SPSS Statistics 25 software.RESULTS: The survey involved 707 physicians from 86 regions of the Russian Federation. Our study revealed that not all doctors strictly adhere to clinical recommendations in patient management. Issues identified include unjustifiably widespread ordering of 25(OH)D blood tests (58.5%), lack of consideration for individual patient characteristics and clinical situations in dose selection (99.2%) and prescription of active metabolite or analogs of vitamin D to correct low 25(OH)D levels in blood (14.7%). Furthermore, recommendations for improving clinical guidelines focused on the lack of illustrative material (21.1%), expanding patient information sections (20.7%), and insufficient coverage of issues arising in clinical practice (14.6%). Our study also highlighted limited capabilities of laboratory diagnostics for assessing vitamin D status in patients in Russia. The survey revealed that only 9.2% of respondents have the ability to measure 1,25(OH)2D concentrations, and only 1.4% can order tests for 24,25(OH)2D. About 8.3% of participants did not specify which tests for assessing vitamin D status are available for prescription. Technical enhancement of laboratories and the inclusion of all recommended laboratory study requirements in the compulsory health insurance system could address this.Regarding vitamin D toxicity, 25% of surveyed doctors encountered it. Main causes included self-administration of elevated doses of cholecalciferol by patients (62%) or prescribed by physicians (40%), the use of active metabolites or analogs of vitamin D (21%), incorrect dosing of cholecalciferol preparations as indicated by the manufacturer (18%), and defects in CYP24A1 (13%). Rare causes included granulomatous and lymphoproliferative diseases (11.5%).CONCLUSION: The current clinical guidelines of the Russian Association of Endocrinologists for «Vitamin D Deficiency in Adults» are generally effective and widely used by clinicians. However, they do not entirely meet the needs of specialists and require refinement in terms of improving structure and clarity of presentation, expanding illustrative material, and justifying the frequency of laboratory diagnostics. Cases of vitamin D toxicity observed in clinical practice were predominantly associated with incorrect administration of vitamin D supplements. The identified high frequency of vitamin D toxicity in real clinical practice necessitates additional attention to this issue during the update of clinical recommendations.

Acromegaly is a rare neuroendocrine disease caused by excessive production of growth hormone (GH), which acts as a trigger for cartilage tissue destruction leading to joint damage.Patients with acromegaly, especially in the active stage, often complain of joint pain in various locations. Joint pain can be one of the first symptoms of the disease, the intensity of which worsens without proper treatment. Increased production of GH leads to configuration changes in the joints, which in turn trigger destructive processes typical of degenerative diseases such as osteoarthritis. Despite successful treatment of acromegaly, joint-related issues can persist and significantly worsen the quality of life for patients. In this regard, the search for potential markers of early joint involvement in acromegaly is relevant for use in predicting the severity of arthropathy progression and monitoring this cohort of patients.This review provides a general overview of the effects of growth hormone on cartilage tissue, the characteristics of musculoskeletal pathology in patients with acromegaly and possible markers associated with early joint damage.

AIM: to identify the features of immune reactions in practically healthy individuals in response to short-term cold exposure, depending on the level of total cholesterol in the blood.MATERIALS AND METHODS: On the basis of the Laboratory of Ecological Immunology of the Institute of Physiology of Natural Adaptations of the Federal Research Center for Integrated Arctic Studies named after Academician N.P. Laverov of the Ural Branch of the Russian Academy of Sciences, 108 practically healthy women living in Arkhangelsk aged 21 to 50 years were examined. Blood was taken from the ulnar vein on an empty stomach in the morning before and immediately after general cooling for 5 minutes at a temperature of -25 °C. The complex of immunological studies included the study of venous blood hemograms: the number of platelets, erythrocytes, leukocytes, total hemoglobin in the blood, leukograms, phagocytic activity of neutrophils and blood monocytes, determination of cytokines IL-1β, IL-6, TNF-α. The study of the lipid profile included the determination of total cholesterol, glucose, triglycerides, phospholipids.RESULTS: In the Q1 group, no cases of hemoglobin deficiency were recorded, while in the Q4 group, hemoglobin deficiency was recorded in 14.29% of the surveyed, and after a short–term general cooling — in 21.43%. The total number of platelets, as well as the thrombocrit index in the Q4 group was significantly higher (259.07×10^3 cells/µl and 0.27%, respectively). BACKGROUND: Obesity is a progressive health and economic problem of modern society. Endocrine-metabolic changes in childhood and adolescence have an impact on cognitive status, which is accompanied by changes in the perfusion of gray and white matter of the brain, as well as its architectonics.AIM: Verification of fatty acid (FA) levels in relation to cognitive function and brain neuroimaging data in adolescents with obesity.MATERIALS AND METHODS: The study is a single-center observational one-stage comparative. 60 teenagers aged 10–16 years were selected to participate. The main study group was formed of 34 adolescents with obesity (SDS BMI ≥+2.0), 26 subjects with normal weight made up the control group. The calculation of SDS BMI was performed on the WHO «Anthroplus» calculator. To obtain anthropometric data, a vertical height meter MSK — 233, scales and a soft centimeter tape were used. The levels of free fatty acids in the blood serum were determined by chromatomass spectrometry on the Agilent 7000B detector. For neurocognitive testing, the Montreal Cognitive Function Assessment Scale and the Ray test were used. Contrast-free perfusion magnetic resonance imaging of the brain was performed on a Signa Creator “E” MR tomograph by GE Healthcare, 1.5 Tl, China, by Arterial Spin Labeling. For MR tractography, the GE Sygna Creaton 1.5 Tesla apparatus was used using the method of diffusion imaging with high angular resolution HARDI (High Angular Resolution Diffusion Imaging). BACKGROUND: Cytokines secrete all cells when there is a threat of damage to it. Leptin, having all the properties of a cytokine, produces fat cells. It was of interest to identify the ratio of leptin concentrations and cytokines activating preventive inflammatory reactions in the blood of practically healthy Northerners with hyperlipoproteidemia.AIM: To identify the ratio of concentrations of leptin and IL-1ß, TNF-α, IL-6 and IL-10 in the blood of practically healthy Northerners with hyperlipoproteidemia.MATERIAL AND METHODS: A survey of 286 practically healthy people aged 35-55 years living in the Arkhangelsk region was conducted. The body mass index in persons of the first group (n=211) was in the range of 19.5-26.8 kg/m2 and corresponded to the norm; and 75 people of the second group (body mass index 31-40 kg/m2). Objective: The present study is aimed to evaluate the curative effect of herbal ointment containing Peristrophe paniculata Forssk. against incision wound. Methods: Animals weighing 150 -200g were divided into five groups each comprising of six rats: Group I served normal control, Group II served as incision wounded control and Group III, IV served as incision wounded animals were treated with 10% and 20% of herbal ointment (HO) containing Peristrophe paniculata Forssk. applied topically for 14 days respectively and group V served as incision wounded animals treated with reference ointment soframycin. The healing of wound was assessed by breaking strength, level of hydroxyproline, hexosamine, tissue and serum protein, ascorbic acid, Deoxyribonucleic Acid (DNA), Ribonucleic Acid (RNA), Superoxide Dismutase (SOD), Lipid Peroxidation (LPO). Results: The topical application of herbal ointment treated groups showed increase in hydroxyproline, hexosamine, protein, breaking strength, DNA, RNA and SOD, Ascorbic acid and significant decrease in the LPO. Conclusion: The results concluded that the Peristrophe paniculata Forssk. has the efficacy in the management of wound healing. Further in-depth studies will be carried out by the isolation of active compounds for drug discovery.

Background and Aim: Laboratory assessment of sialic acid as a marker of microvascular complications can be a diagnostic tool for diabetes mellitus. Microalbumin is an established predictor of diabetic nephropathy. The study was undertaken to evaluate serum sialic acid and microalbumin levels and to correlate it with glycemic control in diabetic nephropathy patients. Material and Methods: Clinically diagnosed diabetic nephropathy patientsvisiting the medicine OPD (outdoor patient department) and NCD (Non-Communicable Disease) were selected for the study. This study included 75 subjectswith diabetes types 2 of which 44 were males and 31were females. Control includes a total of 75 out of which 49 Males and 26 females. Blood samples were analysed for the glucose, C-reactive protein, and sialic acid. Urine sample was analyzed for microalbumin and sialic acid. Results: The mean value of FBS was 161±53.2 mg/dl which was significantlyhigher than controls 81±5.2. The Sialic acid in subjects was 105±21 mg/dl which is higher than controls 51±6 mg/dl. The urine microalbumin level was found to be 96.5±17.2 in subjects, whereas 6.4±1.6 mg/dl was in controls. Serum Creatinine level in subjects was 8.75±2.05 mg/dl which was higher than subjects 0.8±0.3mg/dl. Serum Urea in diabetic nephropathy subjects was 93.5±15.4 mg/dl, which is positively higher than controls 30.8±7.1 mg/dl. Conclusion: Urine microalbumin is established as a potential marker for diabetic nephropathy. The correlation of serum sialic acid, urine microalbumin and serum creatinine proposed serum sialic acid as an accessory diagnostic marker for diabetic nephropathy. Estimation of sialic acid before microalbumin in diabetic patients helps assess glycemic control and identify the risk for nephropathy and other secondary complications of diabetes mellitus, which are the main causes of mortality and morbidity among type-2 diabetes mellitus patients.

Physiological and molecular response of Padina pavonica (phaeophyta) marine alga exposed to different cadmium (0, 2.5, 5 and 10 mg/L) concentrations after 4 days of exposure has been investigated. Physiological data revealed decrease in specific growth rate (SGR%), pigments (Chlorophyll a & b, total chlorophyll and total carotendois) content, osmotic potential with increased electric conductivity (EC) under Cd applied concentrations in the studied alga species. Whereas, carotendois pigments content increased in P. pavonica alga as Cd applied concentrations increased. As for molecular test, RAPD marker has been applied, a decrease in polymorphic bands (PB) from 95 to 87 has been recorded when applied Cd concentration increased from 2.5 to 10 mg/L. Whereas, genomic template stability (GTS%) as a qualitative measurement reflect DNA changes induced by Cd treatment was displayed by RAPD marker. Molecular study revealed increased GTS% value from 30.7 to 42.7% when applied Cd concentration increased from 2.5 to 10 mg/L. Based upon observed physiological (significant increase in carotendois content) and molecular data (decrease in the new induced bands number and increase in disappeared bands as Cd concentration increased from 2.5 to 10 mg/L) in P. pavonica alga, the current investigation could be assumed that P. pavonica alga adopted certain mechanism to minimize Cd stress damages.

Polyol is an organic compound containing multiple hydroxyl groups. This study looked at the possibility of using an edible oil extract from Salvia hispanica seeds as a sustainable source for polyols and, eventually, biodiesel or polyurethane. For this, a combination of hydrogen peroxide and acetic acid was used to create new polyol from the aforementioned oil in one-step synthesis. Standard techniques such as physicochemical analysis, phytochemical and basic radical identification, FTIR and NMR were used to characterize the polyol derivative that was extracted from the oil. Antimicrobial activity of both oil and polyol were tested against certain bacteria and fungi. Spectral analysis demonstrated the formation of polyol and this result indicated the possible of using Salvia hispanica polyol as a raw material for the preparation of bio-polymers.

Salinity stress is a major abiotic stress factor that affects plant growth, physiological activities and developmental processes. This study involves establishing efficient somatic embryogenesis and plantlet regeneration system using immature inflorescence of kodo millet (Paspalum scrobiculatum L. cv. TNAU86) under NaCl-salinity stress conditions. To begin with, the immature inflorescence (0.5cm) of kodo millet was excised from the 40-45 days old field growing plants followed by surface sterilization and inoculation in Murashige and Skoog (MS) medium supplemented with the various concentrations of NaCl (10mM, 25mM, 50mM, 75mM, 100mM, 150mM, and 200mM) salt along with 2,4-Dichlorophenoxyacetic acid (2,4-D) (1.5 mg/L). The results reveal that the maximum mean frequency (89.3±0.3%) of somatic embryogenesis was obtained from embryogenic callus that was growing with medium added with NaCl (50mM) while it was found to be the least mean frequency (12±4.1%) with 150mM of NaCl-salt treatments. Moreover, the highest concentration of NaCl (200mM) salinity was found to be lethal and explants were observed to get gradually necrosed. Later, embryogenic calli showing differentiation of somatic embryos were sub-cultured on basal medium supplemented with 0.5mg/L of 6-Benzylaminopurine (BAP) along with respective concentrations of NaCl-salt for the germination of somatic embryos into plantlets. Significantly, 100mM of NaCl-treatment was proved to show strong inhibitions and thus minimum salt tolerant plantlets regeneration (4±2.21%) was recorded. Further, in vitro grown salinity stress tolerant plantlets were transferred to plastic cups and gradually acclimatized under greenhouse conditions.

A commonly used herbicide in agriculture, paraquat dichloride (PD) has caused a great deal of concern due to its high toxicity and potential impact on the environment. The present study aims to study the effect of paraquat dichloride’s (PD) acute toxicity, behavioural and morphological changes on Anabas testudineus. The species Anabas testudineus, also known as the climbing Perch. Fish were treated with five different doses of PD concentrations in a Fishery Science lab to estimate the LC50 value. The probit analysis method was used to calculate the LC50 value for PD exposure. Fish exposed to (PD) exhibited behavioral abnormalities such as altered nervous behavior, elevated stress response, and respiratory distress. When exposed fish were examined morphologically, several abnormalities were found, such as Sclerosis in the head and tail region, Mucous layer on the whole body, Blood from gills, Redness in eyes, Belly swelling, Red color appearing in the head and tail region, pelvic fin, and anal fin destroyed. The finding of the study shows, Anabas testudineus exposed to PD had an LC50 value of 116.94 mgL-1, which implies the level of toxicity concentration. These results indicate that exposure to (PD) influences the behavior and external morphology of Anabas testudineus. This study emphasizes how PD affects freshwater fish, specifically Anabas testudineus, in an acute toxicological way. In addition, observed morphological and behavioral changes highlight the significance of tracking and controlling the use of PD in agricultural practices to minimize any potential adverse environmental impacts and protect the aquatic ecosystem.

Background: Cascabela thevetia (L.) Lippold, a popular member of family Apocynaceae, (Yellow Oleander), contains pharmacologically active constituents include terpenoids, fla-vonoid, steroids and glycosides in seeds. This plant is being the preferred tool for suicides in villages of India. Purpose: To identify the potency of cytotoxicity of the aqueous decoction in different eukaryotic cellular systems (in vivo testing). To fulfil the purpose germinating root tips of Lathyrus sativus root tip cell nucleolus, Chana punctatus and Gallus gallus RBC were chosen as biological models to evaluate the extent of cytotoxicity of the aqueous decoction of Cascabela thevetia (L.) Lippold dried Seed powder. Results: The presence of Carbohydrates, proteins, tannins, phenolics, terpenoids Some important alkaloids (viz., Hordenine, Ismine, Trisphaeridine, Crinine, Galanthamine, Anhydrolycorine, Assoanine, Galanthine, Incartine, Lycorine and Galwesine), have been already identified in the dried seed powder of Cascabela thevetia (L.) Lippold. The Cascabela thevetia seed aqueous extract (CTSAE) when incubated with 2% RBC produced hemolysis of chicken RBC (Gallus gallus) and found to produce significant changes in the nucleolar frequency and volume in pre-treated germinating Lathyrus sativus L. seeds with serially diluted concentrations (5mg, 10 mg, 20 mg, 30 mg and 40 mg/ ml respectively). There was significant increase in electrolyte leakage and decrease in root oxidizability in the aforementioned doses of seed priming in germinating Lathyrus root tip cells. The said pre-treatments induced significant increase in nucleolar breakage and fragmentations followed by the formationof micronucleoli and karyolysis. There was high frequency of occurrence of giant cells and apoptotic cells. In Chana punctatus RBCs the doses produced significant induction of micronuclei. Conclusions: So it might be conclusively inferred that the seeds of Cascabela thevetia (L.) Lippold, is highly toxic not only for plant cells but can evoke elicitation of variable degree of in vitro and in vivo cytotoxicity in animal (Gallus gallus and Chana punctatus) tissues in addition to plant genomes. Implications: Strict regulation and monitoring is highly needed to keep this ornamental plant from the reach and vicinity of common man and aquatic vis-a-vis domestic animals for better safety and prevent genotoxicity.

Diabetes mellitus is a chronic metabolic disorder caused by inherited or acquired deficiency in production of insulin by the pancreas, or by the ineffectiveness of the insulin produced. It is widely present in all parts of the world affecting nearly 15 % of the population and considered as one of the leading cause of death in humans. In recent years alternative treatment methods are being followed by the diabetic patients in order to avoid side effects of allopathic medicines. Many plant sources are being used to treat diabetes mellitus from time immemorial but all are not validated scientifically. Hence in the present study a commonly available drug source – Vetiveria zizanioides L. (Nash) belonging to the family Poaceae has been chosen and tested for its in vitro antidiabetic potential. Aqueous and ethanolic root extract of Vetiveria zizanioides L. (Nash) were prepared and evaluated for its in vitro antidiabetic potential. Preliminary phytochemical screening of plant extracts revealed the presence of saponin, terpenoid, alkaloid, coumarin, flavanoids, lignins, glycosides and Phenol. The extracts were studied for in vitro antidiabetic activity and its effect on inhibition of glucose diffusion, glycosylation of hemoglobin and glucose transport across yeast cells. From the extracts, ethanol extract exhibited potent inhibition of glucose diffusion, glycosylation of hemoglobin, glucose uptake by yeast cells, significant glucose adsorption capacity, uptake of glucose by rat hemi-diaphragm were found to be in a dose dependent manner. From the results of the study, it is inferred that, Vetiveria zizanioides L. (Nash) root ethanol extract possesses excellent antidiabetic activity compared to aqueous extract. However, these effects need to be confirmed using in vivo models and clinical trials for its effective utilization as therapeutic agent.

The fall army worm (FAW) (Spodoptera frugiperda) is a polyphagous pest which causes damage to commercially important cultivated crops such as maize, rice, sorghum, cotton and also different vegetable crops. Carboxyl esterases (CarE, EC.3.1.1.1) or esterases are enzymes in the carboxyl / cholinesterase gene family that catalyze the hydrolysis of carboxyl esters. The carboxylesterases enzyme are the detoxification enzymes in FAW. Therefore, the inhibition of carboxylesterases from FAW would help in pest management. In this scenario, Isolation, purification and inhibition studies were carried out on the midgut carboxylesterase enzyme of FAW. Through a combination of steps including centrifugation, ammonium sulfate gradient precipitation, DEAE-Cellulose ion exchange chromatography, the enzyme was purified from fifth instar larvae of FAW. The final purified carboxylesterase after ion exchange chromatography had a specific activity of 7282.22 units / mg protein, 5.6 – fold of crude homogenate, and a yield of 25%. The purity of esterase was established by PAGE and SDS-PAGE. The SDS-PAGE revealed a molecular weight of approximately 45kDa to 66kDa. Our studies on the purified midgut carboxylesterase showed complete inhibition by organophosphorous inhibitor (10-4 M). The enzyme was also inhibited by 1x concentration of Lizol (disinfectant) and by different natural extracts (1x) as well.

Breast cancer is one of the leading causes of cancer-related deaths among females and accounts for around 25% of all female melanomas worldwide with increasing incidence every day. Among various natural phytochemicals studied against breast cancer, curcumin has been found to possess antioxidative properties with established safety record. Curcumin is a pleiotropic molecule possessing chemopreventive and chemotherapeutic properties. Curcumin arrests cancer formation at various stages ranging from transformation, proliferation as well as invasion. Transformation is arrested by constitutive de-activation of transcription factors like STAT 3, AP-1, NF-kB. Curcumin controls the expressions of oncogenes, growth factor like EGF, PGDF, FGF, decoy receptors, cyclin D1, survival factors that are important for tumoral cell proliferation. Curcumin successfully inhibits tumor metastasis by supressing the expression of matrix metalloproteinase, COX 2, adhesion molecules, Chemokines and TNF. Curcumin has revealed anticancerous effects in synergism with various compounds like piperine, genistein, mitomycin C, phosphatidicholine, monomethoxy polyethylene glycol, epigallocatechin gallate and metformin. Recent studies have attempted to study the efficacy of curcumin loaded nanoparticles as drug delivery system. This paper attempts to review the work on antitumoral effects of curcumin against breast cancer, underlying mechanisms and proposes further investigations that are needed for rational cancer therapy.

Topoisomerase is an essential enzyme which regulates the topological state of DNA supercoils during replication and transcription. Topoisomerases cleave either one or two DNA strands and then re-join the cleaved strands after passing the intact strand or a double strand through the gap respectively. During relaxation of supercoiled DNA, if topoisomerase is trapped by drugs or alteration of DNA structure, they stabilize topoisomerase-DNA cleavage complex which leads to DNA damage. If Topoisomerase cleavage complex is trapped by any anticancer or others drug, exogenous and endogenous DNA lesion involving mismatches, abasic sites, oxidative damage etc. it may cause DNA damage. DNA damages leads to several diseases such as tumorigenesis, autoimmune disease, Angelman syndrome, SCAN1, SCAR23, Papillary Thyroid Cancer (PTC), cancer therapy-related acute myeloid leukemia. Topoisomerase uses as a potential drug target to manage infectious diseases like leishmaniasis, Chagas disease, pneumococcal, dengue, yellow fever, corona virus, gastrointestinal infection. Here we review the recent information about the topoisomerase mediated DNA damage, related diseases, role of topoisomerase in heterochromatin structure and uses of topoisomerase as drug target in many diseases.

Diabetes Mellitus (DM) presents a significant global health challenge, with escalating prevalence rates and associated morbidity and mortality. Despite medical advancements, the mortality rates continue to rise, necessitating a deeper understanding of the pathophysiology and potential therapeutic targets. Copper, a vital trace element, plays an important role in various metabolic processes and homeostatic functions within the human body. Dysregulation of copper metabolism has been implicated in the pathogenesis and progression of DM, contributing to oxidative stress, insulin resistance, altered glycemic control, and metabolic abnormalities. The present review provides an illustrative overview of the role of copper in DM, elucidating its impact on oxidative stress, insulin resistance, glycemic control, and metabolic disturbances.

This study was conducted to establish an efficient in vitro regeneration technique for the induction of somatic embryo and plantlet regeneration under NaCl-salt stress conditions in black rice (Oryza sativa. L., cv Co57), a traditional Indian cereal food. Embryogenic calli were obtained from mature caryopsis culture on Murashige and Skoog (1962) medium fortified with 2,4-dichlorophenoxyacetic acid (2.0mg/L) either alone or in combination with various concentrations of NaCl (10mM, 25mM, 50mM, and 100mM) in order to induce the salt tolerant somatic embryo differentiation. Furthermore, embryogenic calli were found to show shoot regeneration on MS-medium fortified with 2.0mg/L of 6-benzylaminopurine (BAP) in combination with 0.5mg/L of α-naphthaleneacetic acid (NAA) and in presence of NaCl (10mM, 25mM, 50mM, and 100mM). Significantly, the high concentration (150mM) of NaCl was proved to be lethal for both somatic embryogenesis as well as plantlets regeneration. Moreover, the low frequency (40.3%) of somatic embryogenesis and minimum number of salt tolerant somatic embryos per callus (3.2±0.1) was recorded with the caryopsis explants that were treated with the high concentration (100mM) of NaCl-salt added embryo induction medium. Significantly, low frequency of salt-tolerant plantlets regeneration (25%) and minimum number of plantlets per embryogenic callus (1.5±0.1) was recorded in the embryogenic callus that was treated with 100mM of NaCl in regeneration medium containing BAP (2.0mg/L) in combination with NAA (0.5mg/L). Further, salinity tolerant plantlets were transferred to soil and gradually acclimatized under growth chamber conditions. This study thus offers a suitable technique for production of salt tolerant black rice, an alternative approach for the traditional Indian black rice crop improvements.

Water deficit stress is one major environmental constraint having a devastating impact on crop productivity. Improving plant tolerance to drought is imperative to ensure food security. Drought stress during maize seedling establishment stage lowers the seedling survival rate and increases post pollination embryo abortion rate. Thus, an experiment was carried out as factorial in randomized complete design with three replicates to evaluate the tolerance mechanism of five local maize cultivars at Department of Life Sciences (Botany), Manipur University, Manipur during February to April, 2022. The maize seeds were allowed to germinate under control environment conditions: 14h light/10h dark, 25±2oC temperature and 60% relative humidity using a growth chamber (Tanco PLT-149 Plant Growth Chamber). At seven days after emergence, different concentrations of PEG-6000 were used to impose water deficit stress to maize seedlings. It was observed that drought stress substantially inhibited growth and development across all cultivars. Relative water content (RWC) as well as chlorophyll pigment concentration showed gradual decline under different drought stress levels with cultivar M002 being the least affected. Drought stress also triggered significant augmentation of osmolytes and antioxidant activity with maximum accumulation in cultivar M002. Overall findings from this study indicate that cultivar M002 possess promising drought tolerance characteristics and can perform successfully in water scarce regions.

Background: Phytochemicals are the physiologically active substances found in plants. These phytochemicals which can be found in a variety of plant parts including leaves, flowers, seeds, bark, roots, and pulps, have been employed as direct therapeutic agents. The perennial herb Oenanthe javanica, sometimes called Java water dropwort, water celery, water dropwort, or Indian pennywort, is endemic to East Asia and is specifically utilized in traditional medicine and culinary arts. Purpose: The goal of this plant study is to determine Oenanthe javanica phytochemical composition, extractive values, and antibacterial activity. Results: The extracts contained alkaloids, terpenoids, phenols, flavonoids, coumarins, and saponins, according to the results of the initial phytochemical analyses. Petroleum ether has a lower percentage yield extract than ethanol solvent. The agar-well diffusion technique has been utilized to ascertain Oenanthe javanica antibacterial activity. Both gram-positive and gram-negative bacteria were inhibited by the extracts. In comparison to petroleum ether, ethanol extracts demonstrated the highest level of susceptibility to the pathogens under study. Conclusion: According to these results, Oenanthe javanica is a useful source of phytochemicals that appear to have promising antibacterial properties. Oenanthe javanica could be further investigated for toxicity and to obtain some novel antibacterial compounds in light of this bioactivity.

Background: Drug addiction is a significant global concern. Individuals suffering from drug addiction often experience high levels of Depression, Anxiety, and Stress (DAS). These psychological conditions can intensify the cycle of substance abuse and pose significant barriers to recovery. Although yoga is widely recognised for its benefits to physical and mental health, its specific effects on males with Substance Use Disorders (SUDs) during rehabilitation— particularly in managing DAS — remain underexplored. Conclusion: An 8-week yoga program significantly reduced DAS among male drug addicts as compared to the control group. Material and Methods: A randomised controlled trial was conducted with 80 male participants from a drug addiction rehabilitation centre. The participants were divided into two groups: a control group with 40 subjects and an experimental group with 40 subjects. This study used a pre-test-and-post-test design to evaluate participants' psychological states, measuring Depression, Anxiety, Stress levels using the DASS-21 scale both before and after the intervention. The experimental group engaged in an 8-week yoga program for 60 minutes daily, excluding Sundays, while the control group continued with other rehabilitation routine activities without yoga intervention. Results: After the intervention, significant improvements were observed in the experimental group (p < 0.001), where 97.5% of participants exhibited normal stress levels, 95% showed reductions in anxiety, and 85% experienced decreased symptoms of depression. These percentages were markedly greater than those recorded in the control group.

Suaeda maritima (L.) Dumort. of the family Chenopodiaceae, is an annual succulent herb growing on salty marshy habitat as one of the dominant mangrove associate species and also as pure vegetation of that. It is regularly harnessed by the local people for use as food as well as for alleviating different maladies. Alpha-amyrin, a triterpenoid, is a remarkable biomolecule available in S. maritima. It is reported to have cardioprotective, anti-inflammatory and anti-oxidant properties. A survey of the amount of alpha-amyrin content available in the individuals of S. maritima, collected from eight different regions of the sea coast of the bay of Bengal like, Digha, Sankarpur, Tajpur, Dadanpatrabarh, Shoula, Bankiput and Petuaghat, was conducted with normal phase high performance thin layer chromatography (HPTLC) in this study. It also portrayed variation in the amount of alpha-amyrin among the plant individuals of the species growing in the said zones. The existence of variation in the amount of alpha-amyrin seems to be prospective for selecting the best producer out of them.

The emerging threat of antibiotic resistance and the formation of resilient biofilms pose a challenge to contemporary healthcare systems. This review dives into the interplay between antibiotic resistance mechanisms and biofilm production. Many pathogenic bacteria have an inherent ability of adhering tightly to a surface forming a complex matrix of extracellular polymeric substances (EPS) surrounding their cells. This is called a biofilm which allows pathogenic bacteria to survive in unsuitable environment. The adaptive nature of biofilms provides a protective shield against conventional antimicrobial agents, promoting chronic infections and complicating medical interventions. This phenomenon further adds to the ever-increasing problem of antibiotic resistance. Thus, there is an immediate necessity in developing novel strategies to deal with bacterial biofilms. In terms of human health, biofilms can be formed on mucosal surfaces and on surfaces of medical equipment. They are also a major reason for causing ‘biofouling’. Different approaches have been undertaken to counteract the menace of biofilms encompassing physical, chemical as well as biological methods. However, recent studies have shown that natural bioactive compounds found in fungal extracts, which has already been gaining attention due to their various properties like immunomodulatory activity, anti-tumor activity, antimicrobial activity etc., have the ability to prevent the formation as well as viability of biofilms through numerous mechanisms. This article thus explores the nuances of biofilm formation and its effects, and further delves deep into the convincing potential of the different components in fungal extracts against bacterial biofilms.

In agro-ecosystems, abiotic stress effects can create havoc diminishing productivity and severely deteriorating yield and yield quality. To meet the rising food and feed crisis, rigorous control of productivity and yield losses in agriculture due to environmental stress must be executed. Coupled with climate change, abiotic stress is causing havoc to crop physiology, productivity and quality of agricultural produce. With the SDG 2 target of Zero Hunger to an unprecedented 8.5 billion by 2030, such massive loss of agricultural yield and economy poses a huge challenge. Stress mediated responses of plants are multiple and varied. Nanotechnology is an emerging field which could profoundly affect crop stress physiology through biotechnological and biochemical interventions to usher in a new era of Sustainable Agriculture and Zero Hunger. Such interventions may be suggested for a paradigm shift towards sustainable agriculture and to meet the massive global challenge of 70% rise in crop output for the rising millions of 2050.

Wheat and barley can be massively invaded with Fusarium head blight (FHB) which results in decrease in grain weight and quality. Accurate evaluation of the stress-physiological status including quantitative resistance (QR) and aggressiveness in breeding programs is a key to selecting resistant cultivars. Out of several analyzed components, one or two metric components composing a disease index are commonly employed as a typical methodology to score FHB; however, this resistance index can not describe correctly and accurately the whole resistance in host plants. We have therefore assessed a novel index to estimate the physiological and pathogenic status in a set of diverse bread and durum wheat and barley cultivars under artificial infection with four Fusarium species. Eighteen QR components obtained under in vitro, growth camber and field conditions were evaluated and nine components at the earliest and latest growth stages were selected and combined in one index to score QR. This novel index was further investigated for its stability in the same fungi-seedling and -adult plants treated with silicon that reduced these nine pathogenic components forming this index. The scores of our index were highly significant and correlated with data of reductions resulted in silicon treatments across the bread and durum wheat and barley cultivars, thus it is suitable to be used for quantification of resistance in host plants and aggressiveness of Fusarium isolates. Findings show that the novel index analyzed in the present report captured key components of quantitative wheat and barley resistance. This index can be applied for screening the most aggressive isolates of different FHB species for FHB resistance breeding of wheat and barley. The mean of nine disease components is introduced in this paper as a new index for QR in plants and aggressiveness of fungi. Moreover, the novel index could be a starting point for more correct techniques for assessing on of the most damaging biotic stresses on cereals, FHB.

At elevated temperatures and humidity, cattle experience heat stress which, depending on its intensity, can lead to reduced productivity, health problems and even mortality. At the same time, some animals are currently kept in free-range conditions and this practice is increasing as it is believed to be more beneficial for the socialisation of the animals. It is therefore essential to monitor the health of animals during periods of high temperatures and to take appropriate measures to ensure their well-being. The temperature-humidity index (THI) is a threshold heat index suitable for the physiology of dairy cattle. In Russia, it is generally accepted that the threshold for stress begins at a THI of 75 or higher. Accordingly, this level was used as the boundary at which moderate heat stress develops. Analysing the number of days on which the THI reaches stress levels over time is important in determining the extent of the impact of climate change on cow health. This can help to identify factors that contribute to an increase or decrease in these days and to develop strategies to adapt to changing climate conditions. Therefore, the aim of this study was to determine the number of stressful days over a relatively long period of time in the Central Region of Russia and to attempt to establish a correlation with external factors influencing the climate, such as solar activity. This study shows that the number of days with temperature indices (THI) greater than or equal to 75 is likely to be related to solar activity, albeit with a one-year lag, which most likely indicates an indirect mechanism of this influence. At the same time, another factor has been identified, occurring on average every five years, which significantly influences the number of THI≥75 days, but does not seem to be related to solar activity.

BACKGROUND. Due to its role as a risk factor for the emergence of metabolic illnesses including type 2 diabetes, cardiovascular disease, and certain cancers with pandemic evolution, obesity is a serious public health concern. Diabetes mellitus type 2 (T2DM) poses a major risk to human health. The byproducts of the breakdown of cholesterol are bile acids, which are crucial for preserving cholesterol homeostasis. Research indicates that bile acids might control insulin sensitivity, energy metabolism, and glucose tolerance. Farnesoid X receptors (FXRs) are crucial for controlling bile acid production and hepatic glucose metabolism. The ligand for FXR The semisynthetic derivative of chenodeoxycholic acid, a bile acid, is obeticholic acid (OCA). Research indicates that bile acids may be a viable therapeutic target for type 2 diabetes (T2DM) given that therapy with oleic acid (OCA) enhanced insulin sensitivity and decreased indicators of liver inflammation and fibrosis in individuals with T2DM and nonalcoholic steatohepatitis (NASH).AIM. To assess Obeticholic acid’s effectiveness in obese individuals with prediabetes.MATERIALS AND METHODS. Over the course of three months, we performed a randomized single blind placebo controlled trial on eighty-two overweight and obese patients with prediabetes in the outpatient clinic at Ain Shams University Hospital. Through block randomization, patients were split into two groups (Group A received daily oral tablets containing 5 mg of obeticholic acid, while Group B received non-sweet capsules as a placebo). Three follow-up visits were conducted to ensure adherence and monitor for any emergence of side effects.RESULTS. 82 patients of matched age and sex criteria who underwent block randomization into 2 equal groups, group (A) representing cases and group (B) the placebo controlled group, with 3 months’ regular follow up showed at end of treatment statistically significant difference in weight being lower in group (A) with p-value 0.004 with decreased parameters of glycemic profile (Fasting insulin, FPG, HOMA_IR, 2h PP, HbA1c) in group (A) with p-value

Background: Anxiety, depression and stress (ADS) are prevalent mental health disorders among university students due to the demanding nature of their academic pursuits. Pranayama yogic breathing (PYB), a controlled breathing technique has been suggested as a potential intervention to alleviate these psychological maladies. Aim: This study aimed to investigate the effect of PYB practices on ADS levels of post-graduate students (PGSs) while exploring the potential relationships with serum cortisol (SC) and hemoglobin (Hb) levels. Methods: 60 university students aged between 21 to 24 years were randomly assigned into two groups: an experimental group participant (n=30) who practiced PYB for 45 minutes daily for three months and a control group participant (n=30) who did not receive any intervention. ADS levels were assessed using a standardized questionnaire ADSS, while SC levels and Hb were measured using laboratory tests. Data were collected at three states pre (before intervention), mid (after 45 days of intervention) and post (after 90 days of intervention). Data were analyzed using SPSS version 2.5.11 (110) Results: The results demonstrated that PGSs who underwent the PYB intervention exhibited significantly reduced anxiety, depression and stress compared to the control group. Furthermore, the experimental group showed a significant decrease in SC levels and a significant increase in Hb levels after the intervention. Conclusion: PYB practice has been shown to significantly reduced ADS and positively influenced SC and Hb levels in PGSs. Further research is warranted to explore the long-term effects and broader applicability of PYB on diverse populations.

Late blight, caused by the pathogen Phytophthora infestans is an economical disease of potato worldwide. To better understand mechanisms of potato to resist this fungus, pathway signaling of TUB and PR5 were monitored using quantitative real-time PCR (qPCR) across four time points post infection. Results demonstrated that significant variance in the expression profiles of both genes in infected potato plants as compared to the non-infected controls. It is also notable that TUB and PR5 genes have a higher and faster expression in the resistant cultivar ‘Spunta’ as compared to the susceptible one ‘Draga’ with a maximum expression for TUB (1.8 and 1.4-fold) and PR5 (3.5 and 1.2-fold) respectively, at 48 hours post infection. Our data suggest that TUB and PR5 genes, positively regulate P. infestans—resistance in potato plants during disease progress, which can provide testable hypotheses that will need direct future experiments to define how these pathway signaling of both genes may be specified in potato defense system.

Article deals γ-radiation doses of 5, 10, 50, 100, 200, 300 Gy, sulfate (Na2SO4, ZnSO4) concentrations of 5, 10, 50, 100, 200, 300 mM of cotton Gossypium hirsutum L. The dynamics of changes in the activity of carbonic anhydrase (CA, carbonate hydrolyase, EC 4.2.1.1) enzyme were studied in the ontogeny of the Ganja-182 cultivar of the species Gossypium hirsutum L. in type of true leaf emergence (LP), budding (BP), flowering (FP) and opening of seed boll phase (OBP). It was determined that chloride and sulfate salts have different effects on CA activity. Thus, CA activity increases at 200 Gy dose of γ-radiation, 100 mM of NaCl, and 200 mM of ZnSO4. It seems that the increase in CA activity in the medium containing ZnSO4 is related to the increase in the demand for CO2 under stress. The obtained results show that radiation and sulfate salts have a more regulatory effect on salt adaptation than other salts in cotton plant.

Salt stress has a significant effect on plants, causing a whole range of changes in metabolism. Unfortunately, the mechanisms that ensure the adaptive response of cells in response to salinity have not been sufficiently studied. The study examined changes in the functioning of one of the enzymes of the anaplerotic pathway of the Krebs cycle - GABA shunt - GABA transaminase (GABA-T, EC 2.6.1.19). It has been shown that salt stress caused by incubation of corn seedlings in a 150 mM sodium chloride solution causes activation of GABA transaminase (GABA-T, EC 2.6.1.19). The established increase in GABA-T enzymatic activity in the first hours of incubation in saline solution reaches a maximum at 3 hours of incubation. At the same time, differences are observed in the expression profilees of the GTA-1 and GTA-2 genes, which encode this enzyme in the maize genome. Salinity in the first three hours induces an increase in the expression of the GTA-1 GABA-T gene, while the GTA-2 gene demonstrates an increase in transcriptional activity from 6 to 12 hours of the experiment. An increase in GABA-T enzymatic activity under salinity indicates activation of the GABA shunt to maintain the energy metabolism of the plant cell under stress conditions.

A study is made of the distribution of 674 species of flowering plants in the phylogenetic system according to the specific applications of their active materials in traditional medicine in the prophylaxis or treatment of 21 virus diseases of humans and animals. The analysis revealed statistically reliable connections between the particular applications and the taxonomic position of plant species at the level of a family or a subclass, and connections between virus diseases according to the similarity of the sets of plant species that were used in their prophylaxis or treatment. We suggest the model correlating the taxonomic position with medical applications of plant species. We identified 62 plant species holding the greatest promise as regards the detection, further investigation and antiviral activity. These virus diseases at study are divided into 7 groups such as smallpox, rabies, respiratory infection, jaundice, warts, measles and others. Class Magnoliopsida is superior to class Liliopsida in occurrence of species used in various viral infections. The proportion of these species is most large in evolutionarily later subclasses out of class Magnoliopsida. The families from division Magnoliophyta have reliably increased occurrence of species used at least against one group of diseases such as Ranunculaceae (measles, smallpox), Fumariaceae (jaundice), Euphorbiaceae (rabies, warts), Tamaricaceae (jaundice), Malvaceae (respiratory infection), Dipsacaceae (respiratory infection), Sambucacea (rabies), Viburnaceae (respiratory infection), Trapaceae (rabies), Gentianaceae (rabies, jaundice), Solanaceae (respiratory infection, jaundice, rabies), Cuscutaceae (rabies), Lamiaceae (respiratory infection), Asteraceae (jaundice), Alismataceae (rabies). Closely related families are often similar on sets of infections in which species of these families are applied. Viral diseases are reliably interconnected with similarity of sets of plant species used in the prophylaxis or treatment. Particularly strong connections are "respiratory infections-jaundice", "respiratory infections–rabies", "measles–smallpox". The 62 plant species are recognized as the most promising for discovery, further study and use of their antiviral activity accounting established relations "family – disease" and "disease – disease ". The taxonomic status of plant species is associated with their medical applications using mathematical model. The model is constructed as the equations of the theory of reliability (effectiveness). It describes the changing the frequency of the usage of plant species along the parameter of level of evolutionary development of taxons on rank of subclass. The data on the frequency of application demonstrate good correlation with calculated values (R2 = 0.91) what allows to use the equations for forecasting and valuation.

Silicon (Si) is recognized for its protective role in decreasing disease damage when absorbed by barley plants and has been proposed as a possible solution against Fusarium head blight, associated with devastating agronomic effects on overall yield and grain quality. However, root treatment of exogenous Si irrigating to enhance host resistance to Fusarium infection is unknown. For this purpose, a series of greenhouse experiments was conducted to examine the effects of Si irrigation at 1.7 mM to roots on pathogen development in barley heads. Two barley cultivars with contrasting FHB resistance (moderately resistant Arabi Aswad, AS, and moderately susceptible Arabi Abiad, AB) and infected with four Fusarium species with diverse pathogenicity were used. The quantification of the disease was through the determination of the disease incidence (DI, Type I resistance), disease severity (DS, Type II) and area under disease progress curve (AUDPC) calculated on the basis of DI and DS. Si absorption in barley enhanced the defense system in head tissues to pathogen invasion; FHB developed more severely on AS and AB plants grown without Si irrigation than on plants supplied with Si. Barley plants treated with exogenous Si irrigating were associated with a reduction of up to 19.3%, 19.8%, 18.7%, and 20.0%, respectively, in DI, DS and AUDPC calculated on the basis of DI and DS. Si contributed to the reduction of FHB in barley, especially for the moderately resistant cultivar; however, Si reduced the intensity of FHB in AB to a level comparable with AS. Importantly, Si treatment at 1.7 mM decreased disease damage FHB in previous bio-trials conducted on AS and AB under in vitro and field environments, showing that Si enhanced the expression of resistance to FHB infection in seedlings and adult barley plants. Taken together, the link of Si and host resistance provided a greater decrease in head blight in which both cultivars had augmented performances upon exogenous Si irrigating to roots; highlighting that Si is a potential safe and efficient policy to defend barley when invaded by Fusarium.

Thus, our research seeks to establish through a tryptophan-derivative plant molecule known as phytomelatonin, which may play a significant function in plant responses to various environmental stresses. Through the pieces of literature analysis, we reviewed the exogenous melatonin application and its influence on oxidative stress such as ROS and RNS generated in plant tissues under different abiotic stimuli. Our investigations also concern how phytomelatonin impacts the level of antioxidative proteins such as Superoxide dismutase (SOD), Catalase, and Glutathione Peroxidase (GPx) under these stimuli. After our deep investigation through the literature survey, we found that phytomelatonin acts as a powerful scavenging agent to detoxify ROS and RNS under abiotic threats. Additionally, it also significantly enhanced the level of oxidant proteins to minimize the negative impact of reactive species under these threats. In this way, phytomelatonin exhibits multiple crucial capabilities including root growth, leaf senescence, photosynthetic rate, and increased biomass. Moreover, we discussed in brief how phytomelatonin acts as an emerging regulator of oxidative imbalance between oxidative stress and antioxidative proteins induced by abiotic stresses, generated primarily in cell organelles, nuclei, plasma membrane, cytosol, and apoplast. Thus, it may be concluded that the phytomelatonin molecule might be improving the balance of these stressful conditions in plants for its better-surviving capacities under different threatful situations.

Lysimeters have been used to obtain accurate information for developing calibrating, and validating crop evapotranspiration and crop coefficients for many plants and crops. Lysimeters are also an unique equipment for studying the transport of solutes when saline waters are used, and therefore, for assessing alkalization and salinization hazards. Three main types of lysimeters are used: constatnt water-table lysimeters, drainage lysimeters, and weighing lysimeters. The weighing lysimeters provide scientist the basic information for research related to the evapotranspiration, and they are commonly divided into two types, continuous weighing and intermittent weighing. Lysimeters are foremost devices, typically tanks or containers, that define a specific boundary to contain soil water and permit measurement of either the soil-water balance or the volume of water percolating vertically and its quality. The limitations are expense which depends on design, variable experimental conditions such as climatic/environmental factors which are usually not controlled, the spatial variability is normally less, they are not appropriate for every plant species and even every soil type. The main goal of lysimeter is defining the crop coefficient (Kc) which used to convert Etr to equivalent crop ET (Etc) values, and determining agronomical parameters of crops which are planted on the field of lysimeter. All weather data like air temperature, humidity, solar radiation, and potential evaporation should be obtained onsite, and the frequency and time of measurements should be at least daily. For crop products, the management such as fertilization, sowing tillage, seed bed preparation, and harvest of the lysimeter including its surrounding area is carried out on the basis of good agricultural practice. it may be required to complement natural precipitation by irrigation. The main purpose of this literature review is to give a brief summary about lysimeters, and survey the impacts of lysimeter studies and crops production. Under model conditions, the effect of potassium humate on some indicators in corn seedlings obtained from gamma-irradiated seeds was determined. A physicochemical analysis of potassium humate obtained from pine wood waste was carried out. In field and laboratory experiments, we studied the effect of potassium humate solutions on the morphological and biochemical parameters of corn seedlings obtained from irradiated seeds at doses of 50 and 100 Gy. In all experiments, solutions of potassium humate had a positive effect on the dynamics of growth and development of corn plants grown from irradiated seeds. The effect of potassium humate solutions on chlorophyll and carotenoid pigments, and on the photosynthetic activity (maximum quantum yield of PSII) of plants was studied. It has been established that the treatment of seeds with a 0.1% solution of potassium humate before irradiation reduces the amount of malondialdehyde, a product of lipid peroxidation. Seed treatment with 0.1% and 0.01% solutions of potassium humate before irradiation caused a significant reduction in the harmful effects of ionizing radiation on seedlings of corn plants.

A dielectric barrier discharge plasma device (DBD) was built, characterized and operated. Ten species of gram-positive and gram-negative bacteria, as well as fungus and spores of Bacillus cereus and its vegetative cells, in addition to eggs and pupae of the potato tuber moth (PTM) were exposed to DBD plasma. A strong detrimental effect on the exposed species was observed in function to the exposure time at helium: air ratio 98:2, electrode gap 1.8 cm, amplitude of discharge voltage 6 kV and an effective power density 208 mW/cm3. The outcome of this work provides valuable data on the use of DBD plasma as an alternative non-heating sterilization method to kill or inhibit microbial growth and protecting potatoes from PTM infestation.

One of the main environmental factors that limits plant performance and lowers production is soil salinity, such as NaCl stress. Therefore, to cultivate salt-tolerant crops, it is essential to understand the physiological aspects of plant to NaCl stress and use the natural genetic resource linked with salt tolerance. Rice is relatively a salt-sensitive crop that shows a significant variation in metabolic activities towards NaCl salt stress. In this view, the present investigation was carried out to find the NaCl stress tolerance of rice (ASD-16) variety. The whole experiment was accomplished in pot culture under varying NaCl concentrations (0, 25, 50, 75, and 100mM). The salt stress was imposed on the 13th days after sowing and sampling were done after five days of treatment i.e, 18th DAS. Salt stress results in a decline in growth traits, biomass production, and also reduction in carotenoids and soluble protein contents were observed with hike in NaCl treatments. However, Chlorophyll a, b, (a+b), proline contents, were found to be increased under increasing NaCl concentrations. Thus, enhancing pigment constituents and osmolytes accumulation provides salinity stress tolerance to rice seedlings.

Species of medicinal plants are grouped into sets, each of which was used in folk or scientific medicine for a specific infectious disease. A total of 59 infections and species sets were taken into account. Increases in the occurrence, in such sets, of species belonging to certain families and species containing chemical compounds of certain groups (flavonoids, terpenoids, alkaloids, phenols, lactones, coumarins, iridoids, xanthones) were studied. The mathematical-statistical Student's t-test was used. As a rule, in the set of species used for infectious diseases, the occurrence of some (from 1 to 12) families is significantly increased. 67 families are characterized by a significantly increased occurrence in at least one such set. Significant differences were noted between families in the number of infections with which they are associated through their affinity for the sets of plants used. It was also revealed that, as a rule, in the set of species used for infectious diseases, the occurrence of certain (from 1 to 8) groups of compounds was significantly increased. Significant differences were noted between infections in combinations of groups of compounds that were reliably related to the plant species used. Information about the considered patterns can be used to assess the prospects of plant species, families, and chemical compounds in the study of their antimicrobial and antiviral activity.

They have an impact on functions like cellular growth, hormone production, reproduction, and tissue regeneration. Melatonin, cortisol, vasopressin, acetylcholine, insulin, leptin, and body temperature all have an impact on the circadian rhythm. The transcriptional and translational feedback loops formed by a network of genes and proteins are thought to represent the biochemical basis of circadian rhythms. Retinal light exposure is the main factor in the circadian clock's regulation. Background: Therapeutic plants are an important source of medicine, and various active compounds were isolated from the plants. As per the Indian traditional system of medicine, many medicinal plants have been used for the management of various health disorders, including diabetes mellitus. Diabetes mellitus is a non-communicable disease that is also referred to as a lifestyle disorder that requires modifications in diet, exercise, and behavior along with medication. The present research work was designed to evaluate the impact of multiherbal formulations on blood sugar and body weight in albino rats. Three different formulations were prepared from five plant extracts (Allium sativum, Azadirachta indica, Phyllanthus emblica, Tamarindus indica, and Zingiber officinale). After daily administration of multiherbal formulations at a dose of 300 mg/kg b.wt. and the standard drug (Glibenclamide) at a dose of 5 mg/kg b.wt. for 15, 30, and 45 days, respectively, blood samples were collected from each rat and analyzed according to standard techniques (parameters).
